Abstract

For the planning of radiotherapy treatments for head and neck cancers, Computed Tomography (CT) scans of the patients are typically employed. However, in patients with head and neck cancer, the quality of standard CT scans generated using kilo-Voltage (kVCT) tube potentials is severely degraded by streak artifacts occurring in the presence of metallic implants such as dental fillings. Some radiotherapy devices offer the possibility of acquiring Mega-Voltage CT (MVCT) for daily patient setup verification, due to the higher energy of X-rays used, MVCT scans are almost entirely free from artifacts making them more suitable for radiotherapy treatment planning. In this study, we leverage the advantages of kVCT scans with those of MVCT scans (artifact-free). We propose a deep learning-based approach capable of generating artifact-free MVCT images from acquired kVCT images. The outcome offers the benefits of artifact-free MVCT images with enhanced soft tissue contrast, harnessing valuable information obtained through kVCT technology for precise therapy calibration. Our proposed method employs UNet-inspired model, and is compared with adversarial learning and transformer networks. This first and unique approach achieves remarkable success, with PSNR of 30.02 dB across the entire patient volume and 27.47 dB in artifact-affected regions exclusively. Abstract

From the spectroscopic analysis, using a portable commercial instrument, it is possible to obtain a diagnostic response for breast cancer, at different stages. The invention is constituted by these elements: 1. a nanostructured metallic substrate, made by gold or silver nanoparticles deposited on paper, used to amplify the spectroscopic signal; this type of SERS substrate is already known in literature, but ours has a better performance in terms of repeatability, with respect to those paper-based substrates already reported; it is easy to prepare, inexpensive, and can be prepared on large scale; innovation with respect to published procedures consists in modifying two parameters during the substrate synthesis, in particular in using concentrated (by centrifugation) nanoparticles and in using a paper with a 2 micrometers porosity; 2. the instrumental method of analysis, i.e. the use of paper-based SERS substrate to analyze biofluids such as serum. In fact, such substrate are not new (see above), but so far they have only been used with samples different from blood, serum of plasma. We were the first to find out their use with this type of samples (blood, serum and plasma). 3. a multivariate predictive model, built using a training set of spectral data from a group of patients and from a control group, capable of interpreting a spectrum in diagnostic terms. The algorithms used for such models (e.g. PCA-LDA) are already well known, but our model is unique in so far as it is built using our own data, and it has diagnostic performance parameters (sensitivity, specificity, accuracy) which are better than alternative methods (e.g. mammography).

Abstract

Abstract Background Colorectal cancer develops through two main genetic instability pathways characterized by distinct pathologic features and clinical outcome. Results We investigated colon cancer samples (23 characterized by microsatellite stability, MSS, and 16 by high microsatellite instability, MSI-H) for genome-wide expression of microRNA (miRNA) and mRNA. Based on combined miRNA and mRNA gene expression, a molecular signature consisting of twenty seven differentially expressed genes, inclusive of 8 miRNAs, could correctly distinguish MSI-H versus MSS colon cancer samples. Among the differentially expressed miRNAs, various members of the oncogenic miR-17-92 family were significantly up-regulated in MSS cancers. The majority of protein coding genes were also up-regulated in MSS cancers. Their functional classification revealed that they were most frequently associated with cell cycle, DNA replication, recombination, repair, gastrointestinal disease and immune response. Conclusion This is the first report that indicates the existence of differences in miRNA expression between MSS versus MSI-H colorectal cancers. In addition, the work suggests that the combination of mRNA/miRNA expression signatures may represent a general approach for improving bio-molecular classification of human cancer.

Abstract

Background: The TP53 tumor suppressor gene is the most frequently altered gene in tumors and mutant p53 gain-of-function isoforms actively promote cancer malignancy.Methods: A panel of wild-type and mutant p53 cancer cell lines of different tissues, including pancreas, breast, skin, and lung were used, as well as chronic lymphocytic leukemia (CLL) patients with different TP53 gene status. The effects of mutant p53 were evaluated by confocal microscopy, reactive oxygen species production assay, immunoblotting, and quantitative reverse transcription polymerase chain reaction after cellular transfection.Results: We demonstrate that oncogenic mutant p53 isoforms are able to inhibit SESN1 expression and consequently the amount of SESN1/AMPK complex, resulting in the downregulation of the AMPK/PGC-1α/UCP2 axis and mitochondrial O2-· production. We also show a correlation between the decrease of reduced thiols with a poorer clinical outcome of CLL patients bearing mutant TP53 gene. The restoration of the mitochondrial uncoupling protein 2 (UCP2) expression, as well as the addition of the radical scavenger N-acetyl-L-cysteine, reversed the oncogenic effects of mutant p53 as cellular hyper-proliferation, antiapoptotic effect, and resistance to drugs.Conclusions: The inhibition of the SESN1/AMPK/PGC-1α/UCP2 axis contributes to the pro-oxidant and oncogenic effects of mutant p53, suggesting pro-oxidant drugs as a therapeutic approach for cancer patients bearing mutant TP53 gene.
