Birds generally employ bipedal terrestrial locomotion; their wings flap synchronously to provide aerial propulsion. Penguins (Spheniscidae) are non-flyers that use their modified wings (flippers) to 'fly' underwater, again using synchronous action. The family is distributed from the equator to the Antarctic; the earliest penguins lived at temperate latitudes. On land, penguins usually walk bipedally and slowly (waddling) because their legs are short and their bodies long and fusiform. Their flippers are held out sideways and show little movement; they may function like balance poles. A few penguin species breed at high latitude and make substantial terrestrial migrations over snow and ice; they intermittently use an alternative terrestrial locomotory mode (tobogganing) in which they lie on their ventral surfaces and use the feet alternately to propel them more quickly. The emperor penguin Aptenodytes forsteri makes two-way trips (<250 km) to breed, encountering both ice and several types of snow. The topography varies from flat areas to steep slopes and ridges. In this study we analysed 13 publicly available video records and demonstrate that the flippers are also used for propulsion (particularly for climbing in deep snow). Gait analysis reveals that all four limbs are employed in a quadrupedal, ipsilaterally coordinated manner: the classic mode of tetrapod terrestrial locomotion. During climbing, tetrapod limb action is supplemented by use of the beak as an anchor. Abandoned rookeries belonging to any of the Pygoscelis species are very particular taphocoenosis, usually preserved due to the accumulations of bones, pebbles, and guano year after year in the same nesting area, that condense together with other biogenic material and form ornithogenic sediments. The results of the excavation, sieved and analyses of sediment and materials found in two rookeries formed during the last 8000 years, named Pingfo I and Pingfo II and found in the 25 de Mayo/King George Island (South Shetland Islands, West Antarctica) are presented here. The analyses of the bones and eggshells, taxonomic and ontogenetic composition, elements representation, transport degree, predation, and scavenging marks, together with the micropalaeontological content provided informative tools for the reconstruction of the nesting areas, suggesting that bones were transported and accumulated in Pingfo I that would represent high energy beach, whereas a breeding colony was settled in Pingfo II. The preservation of hexactinellid spicules and several arthropod remains are particularly informative for the levels at Pingfo I. The preservation of a Culicidae (Diptera) wing could be the first middle Holocene record of the group in Antarctica, the only continent without extant mosquitoes. [ABSTRACT FROM AUTHOR]

It has been postulated that ultraviolet reflectance is important in mate choice in King Penguins Aptenodytes patagonicus, although not in other penguin species that do not have body parts that reflect UV light. However, this theory has been challenged. Here we aimed to determine the transmission of the ocular media in the large King Penguin as well as the smallest penguin, the Little Penguin Eudyptula minor, and a medium-sized penguin, the Gentoo Penguin Pygoscelis papua, to determine if the penguin eye is capable of seeing ultraviolet light. In all species the cornea absorbed the most damaging rays at 300 nm or below but it was the lens that predominantly determined the transmission of light between 300 and 400 nm. The lenses of a young King Penguin absorbed almost all light less than 370 nm and had 50% transmission at 406 nm, thus ultraviolet perception in the King Penguin is very limited. In contrast, 50% lenticular transmission was 329 nm in the Little Penguin and 367 nm in the Gentoo. Therefore, we suspect that ultraviolet light may be more important in the behaviour of smaller penguins than in the King Penguin, where it is unlikely to play a significant role. Osteohistological studies allow us to obtain valuable information on different aspects related to the bone microstructure, physiology and ecology of organisms. Although the anatomy and morphology of penguin bones are well known, studies in osteohistology are still insufficient. In order to analyze the osteohistological variations between male and female adults of Pygoscelis adeliae (Aves, Spheniscidae), histological sections were prepared from various bones including appendicular (humerus, radius, ulna, carpometacarpus, femur, tibiotarsus, tarsometatarsus) and axial (vertebral and sternal ribs) elements. The results indicate that all sections showed compact tissue with reduced or absent medullary cavities and high bone density due to internal tissue compaction. Histologically, we identified three distinct tissue regions based on their bone matrix, vascular channels organization and compactions degree. Our results indicate osteohistological variations between sexes throughout the whole skeleton, including the first definitive evidence of medullary bone in the Sphenisciformes females. While the male specimen exhibited a medullary cavity characterized by an inner circumferential layer, the female displayed a medullary region without an inner circumferential layer, lower bone compaction, presence of medullary bone in all the sections, and greater development of intertrabecular spaces. Penguins are the only extant family of flightless diving birds. They currently comprise at least 18 species, distributed from polar to tropical environments in the Southern Hemisphere. The history of their diversification and adaptation to these diverse environments remains controversial. We used 22 new genomes from 18 penguin species to reconstruct the order, timing, and location of their diversification, to track changes in their thermal niches through time, and to test for associated adaptation across the genome. Our results indicate that the penguin crown-group originated during the Miocene in New Zealand and Australia, not in Antarctica as previously thought, and that Aptenodytes is the sister group to all other extant penguin species. We show that lineage diversification in penguins was largely driven by changing climatic conditions and by the opening of the Drake Passage and associated intensification of the Antarctic Circumpolar Current (ACC). Penguin species have introgressed throughout much of their evolutionary history, following the direction of the ACC, which might have promoted dispersal and admixture. Changes in thermal niches were accompanied by adaptations in genes that govern thermoregulation and oxygen metabolism. Estimates of ancestral effective population sizes (N e ) confirm that penguins are sensitive to climate shifts, as represented by three different demographic trajectories in deeper time, the most common (in 11 of 18 penguin species) being an increased N e between 40 and 70 kya, followed by a precipitous decline during the Last Glacial Maximum. The latter effect is most likely a consequence of the overall decline in marine productivity following the last glaciation.

Aspergillosis is a condition causing serious morbidity and mortality in captive penguins and other bird species. It can be treated with antifungal drugs, such as voriconazole. However, the pharmacokinetics of voriconazole are variable between different animal and bird species. Therefore, the pharmacokinetics of voriconazole were investigated in this study in Magellanic penguins. Pharmacokinetic models were constructed and applied to predict the pharmacokinetics of voriconazole during long-term treatment in Magellanic penguins, since the voriconazole treatment duration in chronic aspergillosis cases can last up to several months. Plasma voriconazole concentration-time data from adult Magellanic penguins (Spheniscus magellanicus; n = 15) following a single oral (PO) dose of either 2.5 mg/kg or 5 mg/kg in a herring in three separate study periods 7-12 months apart were collected. Mean plasma voriconazole concentrations were above the targeted MIC for Aspergillus fumigatus for 2 hr following a single 2.5 mg/kg voriconazole dose while the plasma concentrations exceeded the MIC for least 24 hr following a 5 mg/kg dose. Nonlinear mixed-effects modeling was used to fit two pharmacokinetic models, one with first-order and another with saturable elimination, to the single-dose data. Fits were good for both, as long as dose was included as a covariate for the first-order model so that clearance was lower and the half-life longer for animals receiving the 5 mg/kg dose. Although the single-dose data suggested saturated elimination at higher concentrations, the model with saturable elimination did not predict plasma voriconazole concentrations well for a clinical aspergillosis case receiving long-term treatment, possibly because of induction of metabolizing enzymes with chronic exposure. Pharmacokinetic models should accurately predict plasma drug concentrations for different dosage regimens in order to be applicable in the field. Future studies should focus on determining clearance at steady-state to be able to refine the pharmacokinetic models presented here and improve model performance for long-term oral voriconazole administration in Magellanic penguins.

Understanding the causes of disease in Antarctic wildlife is crucial, as many of these species are already threatened by environmental changes brought about by climate change. In recent years, Antarctic penguins have been showing signs of an unknown pathology: a feather disorder characterised by missing feathers, resulting in exposed skin. During the 2018-2019 austral summer breeding season at Cape Crozier colony on Ross Island, Antarctica, we observed for the first time an Adélie penguin chick missing down over most of its body. A guano sample was collected from the nest of the featherless chick, and using high-throughput sequencing, we identified a novel circovirus. Using abutting primers, we amplified the full genome, which we cloned and Sanger-sequenced to determine the complete genome of the circovirus. The Adélie penguin guano-associated circovirus genome shares 99% pairwise identity with the one identified in 2018-2019. This is the first report of a circovirus associated with a penguin species. This circovirus could be an etiological agent of the feather-loss disorder in Antarctic penguins.

Antemortem serodiagnosis of aspergillosis remains challenging in Sphenisciformes. Protein electrophoresis, serology (antibody, antigen) by ELISA, and gliotoxin detection provide variable diagnostic value. In the present study, a commercially available Western blot (WB) validated for use in humans and dolphins was adapted for use with penguin samples. Using the same method and reagents, samples were analyzed from multiple institutions in the United States and one facility in France. This was inclusive of normal juvenile African penguins ( Spheniscus demersus , n = 10) and various species of penguins in the United States with confirmed infection (n = 9) as well as 52 samples from Humboldt penguins ( Spheniscus humboldti ) in France. Cumulative WB scores (based on reactivity to different antigens) were found to be significantly higher in the group of penguins with confirmed infection (p < 0.0001). Significant differences were also observed between the clinically normal penguins in the two populations, with higher scores in the United States (median score 1.0, 95%CI [0-5], min 0, max 11) compared to France (median score 0,95%CI [0-0], min 0, max 5). The utilization of the WB as a diagnostic tool is inconclusive due to the use of samples from varying institutions, environmental background, age, and stages of infection. However, this tool may provide an overview of antigen reactivity in penguins infected with Aspergillus to help design a more robust serology assay and further understand the humoral immune response during infection.

The African penguin is currently experiencing a significant decline, with just over 10,000 breeding pairs left. A substantial body of research reflects the impacts of contaminants of emerging concern (CECs) on the marine environment, with wastewater treatment plants reported as one of the main sources of CEC release. In South Africa, CECs were identified contaminating the marine environment and bioaccumulating in several marine species. Approximately 70 % of all African penguin colonies breed in close proximity to cities and/or harbors in South Africa. Currently, the impact of CECs as a stressor upon the viability of African penguin populations is unknown. Based on the search results there was a clear lack of information on CECs' bioaccumulation and impact on the African penguin. Conserving landscapes used by multiple stakeholder groups requires understanding of what each stakeholder values. Here we employed a semi-structured, participatory approach to identify features of value in the terrestrial Antarctic Peninsula related to biodiversity, science and tourism. Stakeholders identified 115 features, ranging from Adélie penguin colonies to sites suitable for snowshoeing tourists. We split the features into seven broad categories: science, tourism, historic, biodiversity, geographic, habitat, and intrinsic features, finding that the biodiversity category contained the most features of any one category, while science stakeholders identified the most features of any stakeholder group. Stakeholders have overlapping interests in some features, particularly for seals and seabirds, indicating that thoughtful consideration of their inclusion in future management is required. Acknowledging the importance of tourism and other social features in Antarctica and ensuring their integration into conservation planning and assessment will increase the likelihood of implementing successful environmental management strategies into the future., (© 2024. The present work extends the scope of prior studies through analysis, modelling and simulation of the As, Cd, Co, Cu, Fe Mn, Mo, Ni and Zn release from Gentoo (Pygoscelis papua) and Chinstrap (Pygoscelis antarcticus) penguin guano to the Southern Ocean seawater and to Antarctic snow meltwater. Laboratory experimental results have been modelled considering kinetic processes between water and guano using two element pools in the guano compartment; its application allows us to interpret behaviours and predict release concentrations of dissolved trace elements from guano which are potentially useful for incorporation as elements source into biogeochemical models applied in the Southern Ocean. Variations in quantities and release patterns depending on the type of guano and aqueous medium in contact have been identified. The release percentages from the guano to the aqueous medium, once the steady state has been reached, vary depending on the water medium and guano type in the ranges of 100-2.9 % for Mo; 91.5-68.6 % for Ni; 81.8-22.8 % As; 52.0-43.9 % Cu; 26.9-7.4 % Mn; 24.9-5.4 for Co; 4.4-3.2 % for Zn and 0.94-0.51 % for Fe. Considering a penguin population of 774,000 Gentoo and 8,000,000 Chinstrap, the estimated annual mass released to the both seawater and freshwater would be ≈18,500 kg for Cu, ≈1710 kg for Zn, ≈1944 kg for Fe, ≈1640 kg for Mn, ≈499 kg for As, ≈289 kg for Ni, ≈155 kg for Mo, ≈36.7 kg for Cd and ≈8.1 kg for Co. The Ross Sea Marine Protected Area (RS-MPA) hosts endemic species that have to cope with multiple threats, including chemical contamination. Adèlie penguin is considered a good sentinel species for monitoring pollutants. Here, 23 unhatched eggs, collected from three colonies along the Ross Sea coasts, were analysed to provide updated results on legacy pollutants and establish a baseline for newer ones. Average sum of polychlorinated biphenyls (∑PCBs) at the three colonies ranged 20.9-24.3 ng/g lipid weight (lw) and included PCBs IUPAC nos. 28, 118, 153, 138, 180. PCBs were dominated by hexachlorinated congeners as previously reported. Hexachlorobenzene (HCB) and p,p'-dichlorodiphenyldichloroethylene (p,p'-DDE) ranged between 134 and 166 and 181-228 ng/g lw, respectively. Overall, ∑PCBs was exceeded by pesticides, contrary to previous studies from the Ross Sea. Sum of polybrominated diphenyl ethers (∑PBDEs) ranged between 0.90 and 1.18 ng/g lw and consisted of BDE-47 (that prevailed as expected, representing 60-80 % of the ∑PBDEs) and BDE-85. Sum of perfluoroalkyl substances (∑PFAS) ranged from 1.04 to 1.53 ng/g wet weight and comprised five long-chain perfluorinated carboxylic acids (PFCAs), perfluorooctane sulfonate (PFOS), perfluorohexane sulfonate (PFHxS) and perfluorooctanoic acid (PFOA); perfluorooctane sulfonamide (PFOSA) was also detected. The PFAS profile was dominated by PFCAs as already observed in Arctic seabirds. Mercury ranged from 0.07 to 0.15 mg/kg dry weight similarly to previous studies. Legacy pollutants confirmed their ongoing presence in Antarctic biota and their levels seemed mostly in line with the past, but with minor variations in some cases, likely due to continued input or release from past reservoirs. PFAS were reported for the first time in penguins from the Ross Sea, highlighting their ubiquity. Abstract Background The astonishing variety of sounds that birds can produce has been the subject of many studies aiming to identify the underlying anatomical and physical mechanisms of sound production. An interesting feature of some bird vocalisations is the simultaneous production of two different frequencies. While most work has been focusing on songbirds, much less is known about dual-sound production in non-passerines, although their sound production organ, the syrinx, would technically allow many of them to produce “two voices”. Here, we focus on the king penguin, a colonial seabird whose calls consist of two fundamental frequency bands and their respective harmonics. The calls are produced during courtship and for partner and offspring reunions and encode the birds’ identity. We dissected, μCT-scanned and analysed the vocal tracts of six adult king penguins from Possession Island, Crozet Archipelago. Results King penguins possess a bronchial type syrinx that, similarly to the songbird’s tracheobronchial syrinx, has two sets of vibratory tissues, and thus two separate sound sources. Left and right medial labium differ consistently in diameter between 0.5 and 3.2%, with no laterality between left and right side. The trachea has a conical shape, increasing in diameter from caudal to cranial by 16%. About 80% of the king penguins’ trachea is medially divided by a septum consisting of soft elastic tissue (septum trachealis medialis). Conclusions The king penguins’ vocal tract appears to be mainly adapted to the life in a noisy colony of a species that relies on individual vocal recognition. The extent between the two voices encoding for individuality seems morphologically dictated by the length difference between left and right medial labium. The septum trachealis medialis might support this extent and could therefore be an important anatomical feature that aids in the individual recognition process.

A 21-year-old captive female Humboldt penguin (Spheniscus humboldti) was in good health until the day before the animal died. However, the animal suddenly exhibited symptoms of vomiting and anorexia, and died in shortly thereafter. The autopsy revealed a blood clot in the abdominal air sac and 280 mL of dark red fluid in the body cavity. The ovary was twisted around a blood vessel. Based on the findings described, it was considered that the cause of death was hypovolemic shock due to massive intracoelomic hemorrhage by vascular collapse resulting from ovarian torsion. This is the first report of ovarian torsion in penguins.

Microplastic pollution threatens some of the world's most iconic locations for marine biodiversity, including the remote Galápagos Islands, Ecuador. Using the Galápagos penguin (Spheniscus mendiculus) as a sentinel species, the present study assessed microplastics and suspected anthropogenic cellulose concentrations in surface seawater and zooplankton near Santa Cruz and Galápagos penguin colonies (Floreana, Isabela, Santiago), as well as in penguin potential prey (anchovies, mullets, milkfish) and penguin scat. On average, 0.40 ± 0.32 microplastics L -1 were found in surface seawater (<10 μm; n = 63 L), while 0.003, 0.27, and 5.12 microplastics individual -1 were found in zooplankton (n = 3372), anchovies (n = 11), and mullets (n = 6), respectively. The highest concentration (27 microplastics individual -1 ) was observed in a single milkfish. Calculations based on microplastics per gram of prey, in a potential diet composition scenario, suggest that the Galápagos penguin may consume 2881 to 9602 microplastics daily from prey. Despite this, no microplastics or cellulose were identified in 3.40 g of guano collected from two penguins. Our study confirms microplastic exposure in the pelagic food web and endangered penguin species within the UNESCO World Heritage site Galápagos Islands, which can be used to inform regional and international policies to mitigate plastic pollution and conserve biodiversity in the global ocean. Aspergillosis is a major cause of morbidity and mortality in penguins, with triazole antifungal drugs being commonly used for prophylaxis and treatment. This report describes 15 cases of fatal hemolysis associated with liquid itraconazole and voriconazole formulations administered to African penguins ( Spheniscus demersus ) from four institutions. All penguins underwent stressful events (e.g. relocation, induced molt) and were administered commercial liquid itraconazole formulations or compounded voriconazole liquid suspension. Observed clinical signs in affected penguins prior to death included hyporexia, weight loss, lethargy, dyspnea, red-tinged droppings, and obtunded mentation. Intra- and extravascular hemolysis and hemoglobinuric nephrosis were the primary pathologic manifestations on postmortem examination. The concentration-dependent hemolytic potentials of itraconazole, voriconazole, and commercial and compounded vehicle suspensions were evaluated in vitro by exposing chicken whole blood as a surrogate for penguin blood. Hemoglobin content in blood plasma was then measured by spectrophotometry. Neither itraconazole nor voriconazole alone induced hemolysis in vitro. The vehicle ingredients sorbitol and hydromellose induced hemolysis, but not at predicted plasma levels in chicken erythrocytes, suggesting neither the azole antifungals nor their major vehicles alone were likely to contribute to hemolysis in vivo in these penguins. Potential mechanisms of toxicosis include generation of an unmeasured reactive metabolite causing hemolysis, preexisting erythrocyte fragility, or species-specific differences in hemolytic thresholds that were not assessed in the chicken erythrocyte model. More research is needed on the potential for toxicosis of azole antifungal drugs and carrier molecules in this and other avian species.

Marine pollution is becoming ubiquitous in the environment. Observations of pollution on beaches, in the coastal ocean, and in organisms in the Antarctic are becoming distressingly common. Increasing human activity, growing tourism, and an expanding krill fishing industry along the West Antarctic Peninsula all represent potential sources of plastic pollution and other debris (collectively referred to as debris) to the region. However, the sources of these pollutants from point (pollutants released from discrete sources) versus non-point (pollutants from a large area rather than a specific source) sources are poorly understood. We used buoyant simulated particles released in a high-resolution physical ocean model to quantify pollutant loads throughout the region. We considered non-point sources of debris from the Antarctic Circumpolar Current, Bellingshausen Sea, Weddell Sea, and point source pollution from human activities including tourism, research, and fishing. We also determined possible origins for observed debris based on data from the Southern Ocean Observing System and Palmer Long-Term Ecological Research program. Our results indicate that point source pollution released in the coastal Antarctic is more likely to serve as a source for observed debris than non-point sources, and that the dominant source of pollution is region-specific. Penguin colonies in the South Shetland and Elephant Islands had the greatest debris load from point sources whereas loads from non-point sources were greatest around the southernmost colonies. Penguin colonies at Cornwallis Island and Fort Point were exposed to the highest theoretical debris loads. Antarctica is the most remote and coldest regions of the planet, but the presence of REEs there has received little attention. This study assessed REE-contents in the feathers of adult gentoo penguins from Ardley Island, Kopaitic Island and Base O'Higgins. Field work was accomplished during 2011 (austral summer), and determination of elements was performed with ICP-MS. In general, REE-levels showed descending relations as follows: Ce > La > Y > Nd > Sc > Pr > Gd > Sm > Dy >Er > Yb > Eu > Ho > Tb > Tm > Lu. The data showed an increase of the levels of REEs from the lower part of the feather to the tip. This finding seems to be spatially dependent, but geochemical, anthropogenic conditions, feeding habits, sex, or even health status of birds should also be considered. Humboldt Penguin (Spheniscus humboldti) population declines are attributable to several multifaceted anthropogenic impacts. At present, the exposure of Humboldt Penguins to high concentrations of heavy metals in the marine environment is a preeminent concern, due to mining along the Peruvian coast near key rookery sites. Metal and selenium concentrations were determined in eggs collected from September 2020 to April 2021 from a managed-care penguin population at the Brookfield Zoo to establish reference values for health indices conducted on wild populations. Concentrations of 16 elements, with emphasis on those found in mine efflux-arsenic, cadmium, copper, lead, mercury, selenium, and zinc-were assessed via inductively coupled plasma mass spectrometry in yolk, albumen, and eggshell. Data analyses indicate a clear delineation between egg constituents, with lipid-rich yolk displaying notably higher concentrations (μg/g) of arsenic (0.20 ± 0.064), chromium (0.086 ± 0.03), cobalt (0.01 ± 0.003), iron (238.65 ± 54.72), lead (0.32 ± 0.97), manganese (2.71 ± 0.66), molybdenum (0.57 ± 0.14), tin (3.29 ± 0.99), and zinc (64.03 ± 13.01) than other components (albumen and eggshell). Wild penguins are facing increased threats to their populations and their welfare as a consequence of human activities. Understanding the perception of animal welfare is essential to identify ethical concerns related to the negative impact of anthropogenic factors on wild species and to guide conservation efforts that reflect societal values. Since penguin conservation is of general interest, we examined the human dimension of welfare assessment across a range of interest groups concerned with penguins, seabird biology and wildlife conservation. We provided participants with a Penguin Welfare Assessment Tool (PWAT) based on the five domains model. The PWAT supports consideration of the impact of four physical aspects on welfare-relevant mental states. Bibliometric analysis of keywords from 347 scientific articles indicated that penguins around the world face five main types (themes) of anthropogenic factors and we then developed five hypothetical scenarios, each related to one theme. Seventy-five participants scored the overall impact of the events described in the scenarios on penguin welfare as negative using the PWAT. Participants rated short-duration, high-intensity events (i.e., being trapped in a ghost fishing net) as having a significantly more severe impact on penguin welfare than low-intensity, long-duration events (P < 0.0001). Scores provided by participants for each domain for each scenario were largely as expected and we found good correlation (all P < 0.0001) between the physical domains and “mental state” for all scenarios, indicating that the tool was facilitating the participants' assessment of welfare. No evidence was found that experience of working or studying penguins, or indeed any other demographic factor investigated, influenced the assessments of welfare. We found little agreement between participants in the scores provided (unalike scores mostly between 0.7 and 0.8), and agreement between participants with experience of working with penguins was no better than between participants without such experience. We discuss the possibility that low agreement within different interest groups may be improved by providing more scientific information to support the evaluation of penguin welfare. We conclude that scientific knowledge of penguin biological responses to anthropogenic factors is vital to support the evaluation of wild penguin welfare by the public and other stakeholders.

We describe a new breeding behaviour discovered in emperor penguins; utilizing satellite and aerial-survey observations four emperor penguin breeding colonies have been recorded as existing on ice-shelves. Emperors have previously been considered as a sea-ice obligate species, with 44 of the 46 colonies located on sea-ice (the other two small colonies are on land). Of the colonies found on ice-shelves, two are newly discovered, and these have been recorded on shelves every season that they have been observed, the other two have been recorded both on ice-shelves and sea-ice in different breeding seasons. We conduct two analyses; the first using synthetic aperture radar data to assess why the largest of the four colonies, for which we have most data, locates sometimes on the shelf and sometimes on the sea-ice, and find that in years where the sea-ice forms late, the colony relocates onto the ice-shelf. The second analysis uses a number of environmental variables to test the habitat marginality of all emperor penguin breeding sites. We find that three of the four colonies reported in this study are in the most northerly, warmest conditions where sea-ice is often sub-optimal. The emperor penguin's reliance on sea-ice as a breeding platform coupled with recent concerns over changed sea-ice patterns consequent on regional warming, has led to their designation as "near threatened" in the IUCN red list. Current climate models predict that future loss of sea-ice around the Antarctic coastline will negatively impact emperor numbers; recent estimates suggest a halving of the population by 2052. The discovery of this new breeding behaviour at marginal sites could mitigate some of the consequences of sea-ice loss; potential benefits and whether these are permanent or temporary need to be considered and understood before further attempts are made to predict the population trajectory of this iconic species.

Global targets for area-based conservation and management must move beyond threshold-based targets alone and must account for the quality of such areas. In the Southern Ocean around Antarctica, a region where key biodiversity faces unprecedented risks from climate change and where there is a growing demand to extract resources, a number of marine areas have been afforded enhanced conservation or management measures through two adopted marine protected areas (MPAs). However, evidence suggests that additional high quality areas could benefit from a proposed network of MPAs. Penguins offer a particular opportunity to identify high quality areas because these birds, as highly visible central-place foragers, are considered indicator species whose populations reflect the state of the surrounding marine environment. We compiled a comprehensive dataset of the location of penguin colonies and their associated abundance estimates in Antarctica. We then estimated the at-sea distribution of birds based on information derived from tracking data and through the application of a modified foraging radius approach with a density decay function to identify some of the most important marine areas for chick-rearing adult penguins throughout waters surrounding Antarctica following the Important Bird and Biodiversity Area (IBA) framework. Additionally, we assessed how marine IBAs overlapped with the currently adopted and proposed network of key management areas (primarily MPAs), and how the krill fishery likely overlapped with marine IBAs over the past five decades. We identified 63 marine IBAs throughout Antarctic waters and found that were the proposed MPAs to be adopted, the permanent conservation of high quality areas for penguin species would increase by between 49 and 100% depending on the species. Furthermore, our data show that, despite a generally contracting range of operation by the krill fishery in Antarctica over the past five decades, a consistently disproportionate amount of krill is being harvested within marine IBAs compared to the total area in which the fishery operates. Our results support the designation of the proposed MPA network and offer additional guidance as to where decision-makers should act before further perturbation occurs in the Antarctic marine ecosystem.

Although considered one of the most pristine ecosystems, Antarctica has been largely influenced by human activities during the last 50 years, affecting its unique biodiversity. One of the major global threats to health is the emergence of antibiotic-resistant bacteria that may be actively transferred to wildlife. We cultured and tested for antibiotic resistance in 137 cloacal and fresh fecal samples of several avian and marine mammal species from the Antarctic Peninsula, the most impacted area in Antarctica. Alarmingly, 80 % of the isolates showed antibiotic resistance, either phenotypically or genotypically. Most of the resistant bacteria, such as Enterobacteriaceae and Enterococcus species, are part of local gastrointestinal microbiota. Penguins and pinnipeds harbored a great diversity of antibiotic resistance and must be eligible as sentinels for future studies. Background: Scalp cooling is an increasingly recognized non-pharmacologic approach to minimize chemotherapy-induced alopecia (CIA). Several commercially available machine-based and manual scalp cooling systems are available; however, literature reports of effectiveness are highly variable. The purpose of this study was to determine real-world tolerability and subjective effectiveness of a manual cold capping system in minimizing CIA across a variety of patient race and hair types. This study was a single-institution review of outcomes from manual cold capping., Methods: We identified retrospective cohort of adult patients who presented to discuss cold capping between January 14, 2019, and March 31, 2022. Data collected from medical records included demographics, decision to pursue/continue cold capping, diagnoses, chemotherapy regimens, hair characteristics (length, thickness, coarseness, type), and subjective perception of percentage of hair retained. Those with successful vs. unsuccessful cold capping (≥ 50% vs. < 50% of hair retained) were compared based on the patient-level factors of interest., Findings: A total of 100 patients initiated cold capping during the study period, and 95% of them completed cold capping. The majority of patients who started cold capping completed it. The median-reported percentage of hair maintained was 75%, and 82/89 (92.1% of patients) had favorable results, defined as ≥ 50% of hair retained. The only patient-level factor associated with favorable response was chemotherapy regimen, with fewer patients receiving doxorubicin-containing regimens having successful hair retention compared to other chemotherapy types (71.4% successful results vs. 95.7% for those receiving paclitaxel-containing regimens and 96.6% for those receiving docetaxel-containing regimens (p = 0.018). There was no difference in success based on patient race/ethnicity or hair characteristics., Interpretation: The overall effectiveness (92.1%) in this study is consistent to higher than many literature reports. Like many polar animals, emperor penguin populations are challenging to monitor because of the species' life history and remoteness. Consequently, it has been difficult to establish its global status, a subject important to resolve as polar environments change. To advance our understanding of emperor penguins, we combined remote sensing, validation surveys and using Bayesian modelling, we estimated a comprehensive population trajectory over a recent 10-year period, encompassing the entirety of the species' range. Reported as indices of abundance, our study indicates with 81% probability that there were fewer adult emperor penguins in 2018 than in 2009, with a posterior median decrease of 9.6% (95% credible interval (CI) -26.4% to +9.4%). The global population trend was -1.3% per year over this period (95% CI = -3.3% to +1.0%) and declines probably occurred in four of eight fast ice regions, irrespective of habitat conditions. Thus far, explanations have yet to be identified regarding trends, especially as we observed an apparent population uptick toward the end of time series. Our work potentially establishes a framework for monitoring other Antarctic coastal species detectable by satellite, while promoting a need for research to better understand factors driving biotic changes in the Southern Ocean ecosystem.

Herpesviruses are associated with disease in many penguin species. Herpesvirus-associated lesions can cause significant morbidity and mortality in penguins and have been identified in African penguins ( Spheniscus demersus ), Humboldt penguins ( Spheniscus humboldti ), and a little blue penguin ( Eudyptula minor ) infected with spheniscid alphaherpesvirus 1 (SpAHV1). Further investigation is necessary to understand the impact of herpesviruses on penguin health, but there are no rapid, sensitive, and specific methods for detecting and quantifying herpesviral load. We therefore developed a quantitative real-time PCR (qPCR) assay for the detection of SpAHV1 in penguins. TaqMan primer-probes targeting the DNA polymerase gene were designed using a commercial software program. Inter- and intra-assay variability, dynamic range, limit of detection, and analytical specificity were assessed. We used our assay to analyze previously collected field samples from Punta San Juan, Peru, in which conventional consensus PCR had detected one SpAHV1-positive penguin sample. Our qPCR assay was highly specific for SpAHV1. It had a dynamic range of 10 7 -10 1 target copies per reaction and performed with high efficiency and low intra- and inter-assay variability. Reaction efficiency was not impacted by penguin DNA from SpAHV1-negative tracheal swabs. We detected an additional field sample as positive with our newly developed qPCR assay, and although this likely represents detection of another infected penguin, the true disease status of this population is currently uncharacterized given that no gold-standard test exists for SpAHV1. Pulmonary aspergillosis occurring in captive penguins living in zoos and aquariums is a fatal disease owing to its high mortality rate. On conducting computed tomography, two gentoo penguins (Pygoscelis papua) with respiratory clinical signs that were housed at an aquarium in Hokkaido, Japan were found to have a certain amount of fluid in their air sacs. This fluid was collected and cultured for bacteria and fungi, the results of which both came back negative. These results enabled us to rule out bacterial infection or aspergillosis and supported our decision not to administer antibacterial and antifungal drugs. Overall, the combination of computed tomography and air sac fluid examination was useful for ruling out bacterial infection or aspergillosis in penguins with respiratory clinical signs.

A 26-y-old, male, captive Humboldt penguin ( Spheniscus humboldti ) was euthanized following a 3.5-mo history of weakened elimination mechanics, recurrent tenesmus, intermittent hemorrhagic droppings, and a cloacal mass. Blepharospasm, of unknown cause, of the right eye was present for ~3 mo before euthanasia. Autopsy revealed a cloacal adenocarcinoma with localized coelomic carcinomatosis and distant metastases to the liver and lungs. On histopathology, a 2.6 × 1.2 × 0.5-mm, well-demarcated mass was found surrounding the right optic nerve, expanding the subdural space and wrapping the leptomeninges. The mass was composed of neoplastic spindle-to-polygonal cells consistent with a meningioma, meningothelial subtype. No evidence of neoplasia was found in the optic chiasm or brain, indicating a primary retrobulbar meningioma. Immunohistochemistry for cytokeratin AE1/AE3, vimentin, and S100 revealed robust and consistent immunoreactivity to vimentin, and weak and variable immunoreactivity to cytokeratin and S100, supporting the diagnosis. Meningiomas have been described only rarely in avian species, and we found no reports of optic nerve meningiomas in any avian species to date. Per- and Polyfluoroalkyl substances (PFAS) are increasingly detected in wildlife and present concerning and unknown health risks. While there is a growing body of literature describing PFAS in seabird species, knowledge from temperate Southern Hemisphere regions is lacking. Little penguins (Eudyptula minor) can nest and forage within heavily urbanised coastal environments and hence may be at risk of exposure to pollutants. We analysed scat contaminated nesting soils (n = 50) from 17 colonies in lutruwita/Tasmania for 16 PFAS, plasma samples (n = 45) from nine colonies, and three eggs for 49 PFAS. We detected 14 PFAS across the sample types, with perfluorooctanesulfonic acid (PFOS) and perfluorohexanesulfonic acid (PFHxS) most commonly detected. Cu is a vital micronutrient, but free Cu ions (Cu 2+ ) in seawater, even at subnanomolar concentrations, can impede phytoplankton growth. Natural Cu complexation with organic ligands regulates Cu acquisition and, in most instances, reduces Cu 2+ concentrations below toxic thresholds. Along the Antarctic coast, the sources and sinks of Cu and its associated ligands remain poorly defined. Despite the high productivity in the area, there are no studies on the role of trophic transfer in Cu cycling. This study explores penguin guano release of Cu and Cu ligands and its potential in neutralizing copper toxicity along the Antarctic coast. We collected guano in a Chinstrap penguin nesting location in the West coast of Deception Island and extracted its components into aqueous solution imitating natural processes. Copper concentration in guano was 0.4 mg (dry weight g) -1 constituting a potential toxic threat and showed biomagnification with respect to krill. Surface seawater samples collected from various locations varying in penguin activity, were analyzed to assess the potential influence of guano on the area. Visual examination and elevated levels of Al suggested that a substantial portion of guano was lithogenic. Consequently, only a modest 16 % of the total Cu present in guano could be extracted using mechanical methods. Notably, the concentrations of the extracted organic ligands were approximately 23 times higher than the concentrations of the extracted Cu. This significant presence of ligands effectively nullifies any potential toxicity that could have arisen from free Cu 2+ ions. Guano ligands' conditional stability constants were lower than those in surface seawater suggesting phytoplankton exudation was the main ligand source in the area. The African penguin Spheniscus demersus, frequently housed in zoos, holds potential for future reintroduction efforts due to its declining wild population. This paper aims to explore various aspects of reproductive performance in African penguins within a large ex situ colony at Zoo Wrocław in Poland, covering 9 years of breeding behaviors. The analysis reveals parallels in colony growth and partner change patterns with those observed in the wild. Positive correlations were found between breeding success and pair-bond duration, with the increasing colony size influencing reproductive performance. Contrary to their wild counterparts, captive African penguins initiate breeding attempt and produce a fertilized egg at a younger age. However, successful breeding still requires gaining experience or forming pairs with more experienced partners. Our research indicates that providing captive African penguins with unlimited food resources and sufficient nesting space results in rapid colony growth. The increased colony size facilitates breeding behaviors that positively influence population dynamics, particularly through the maintenance of long-term pair bond relationships and the potential for partner changes when necessary or desirable to enhance breeding success. Despite the well-known potential health benefits of prebiotics and non-viable probiotics (paraprobiotics) in various animal species, research regarding their use in penguins is scarce. Our study aimed to investigate the impact of a combined administration of prebiotics and paraprobiotics (referred to here as "parasynbiotics") on the gut microbiome and overall health of Magellanic penguins (Spheniscus magellanicus). The parasynbiotics consisted of 1-kestose, which is a fructooligosaccharide comprising sucrose and fructose, and heat-killed Lactiplantibacillus plantarum FM8, isolated from pickled vegetables. It was administered to eight penguins aged <3 years (Young-group) and nine penguins aged >17 years (Adult-group) for 8 weeks. Results from 16S rRNA sequencing revealed that compared to baseline, parasynbiotic administration significantly decreased the relative abundance of intestinal Clostridiaceae&#95;222000 in both groups and significantly increased that of Lactobacillaceae in the Young-group. Quantitative real-time polymerase chain reaction revealed a significant decrease in the plc gene levels encoding alpha-toxin of Clostridium perfringens in the Young-group after parasynbiotic administration (P=0.0078). In the Young-group, parasynbiotic administration significantly increased the plasma levels of total alpha-globulin (P=0.0234), which is associated with inflammatory responses. Furthermore, exposure of dendritic cells to heat-killed L. plantarum FM8 promoted the secretion of interleukin 10, a major anti-inflammatory cytokine. Overall, parasynbiotic administration enhanced the activity of gut Lactobacillaceae, decreased the levels of C. perfringens and its toxin encoding plc gene, and reduced inflammatory response in penguins. These results provide novel insights into the potential benefits of parasynbiotics for improving penguin health.

Background: The identification of ophthalmic diseases that affect vision and/or cause discomfort should be a priority in captive penguins to improve their overall health and quality of life., Methods: A routine ophthalmological examination was performed on 80 clinically normal penguins (160 eyes), and any lesions observed were recorded., Results: Ocular lesions were frequent (56% of penguins), with cataracts being the most common (48.8% of penguins). Cortical cataracts (63% of eyes) and posterior subcapsular cataracts (27.4%) were the most commonly occurring. All maturation stages were found; incipient cataracts (52.1% of eyes with cataracts) were predominant, while Morgagnian cataracts (8.2%) were the least frequent. A correlation existed between lenticular changes and increasing age. Uveitis was present in 43.8% of eyes with cataracts, and ectropion uveae was the predominant clinical sign. Other ocular findings included blepharitis (3.8% of all eyes), corneal leukoma (5.6%) and posterior lens subluxation (7.5%)., Limitations: The small number of birds of some species prevented the comparison of ophthalmic findings between species., Conclusion: This study corroborates the high prevalence of ocular lesions in captive penguins. Cataracts were frequent and age related. Most cataracts were cortical, and the predominant maturation stage was incipient. Lens-induced uveitis was a common finding. Seasonal migration, driven by shifts in annual climate cycles and resources, is a key part of the life history and ecology of species across taxonomic groups. By influencing the amount of energy needed to move, external forces such as wind and ocean currents are often key drivers of migratory pathways exposing individuals to varying resources, environmental conditions, and competition pressures impacting individual fitness and population dynamics. Although wildlife movements in connection with wind and ocean currents are relatively well understood, movements within sea-ice fields have been much less studied, despite sea ice being an integral part of polar ecology. Adélie penguins (Pygoscelis adeliae) in the southern Ross Sea, Antarctica, currently exist at the southernmost edge of their range and undergo the longest (~12,000 km) winter migration known for the species. Within and north of the Ross Sea, the Ross Gyre drives ocean circulation and the large-scale movement of sea ice. We used remotely sensed sea-ice movement data together with geolocation-based penguin movement data to test the hypothesis that penguins use gyre-driven sea-ice movement to aid their migration. We found that penguins traveled greater distances when their movement vectors were aligned with those of sea ice (i.e., ice support) and the amount of ice support received depended on which route a penguin took. We also found that birds that took an eastern route traveled significantly further north in two of the 3 years we examined, coinciding with higher velocities of sea ice in those years. We compare our findings to patterns observed in migrating species that utilize air or water currents for their travel and with other studies showing the importance of ocean/sea-ice circulation patterns to wildlife movement and life history patterns within the Ross Sea. Aspergillosis is a major health problem in captive penguins due to the inhalation and the development of airborne spores of opportunistic environmental molds of the genus Aspergillus. Diagnosis is often delayed and treatments, based on the use of azole antifungals, are not fully effective. This study assesses the risk of exposure to Aspergillus sp. and determines the environmental reservoirs in the direct environment of a colony of Humboldt penguins (Spheniscus humboldti) in a zoological park in Paris, and the risk of contamination with resistant isolates. Every 15 days between February and May 2022, environmental samples (air and subtract from the nests, pond water, pigeon and penguin droppings) were carried out in the penguin enclosure as well as clinical samples (one-time non-invasive sampling on chicks), and screened for Aspergillus sp. conidia. From 191 environmental samples, 264 strains of Aspergillus including 221 strains of A. fumigatus were isolated, mostly from ambient air, in the nests, and pond water. No "at risk" areas in the penguin environment have been highlighted, nor an increased risk because of the proximity with urban wild birds. However, the load of airborne Aspergillus in the nests increased significantly with outdoor temperature. Of the 221 strains isolated, we identified only one azole-resistant strain, displaying the TR34/L98H mutation in the cyp51A gene. This low prevalence of resistant strains may probably be partly explained by the urban location of the zoological park, surrounded by kilometers of urban areas without agricultural activities., (© 2023. As China's "double carbon" goal continues to advance, logistics as a key area of carbon emissions and low-carbon logistics center site selection are key links in the process. However, existing studies on logistics center location often ignore the impact of demand uncertainty, which leads to a waste of resources in the planning and construction processes. We take logistics cost and carbon emission as the objectives, and the multi-objective site selection model established based on stochastic programming theory takes demand uncertainty as a stochastic constraint. We transform the stochastic constraint model into a 0-1 mixed integer multi-objective planning model by utilizing the idea of equivalence transformation. The Aptenodytes Forsteri Optimization (AFO) algorithm is combined with the Ideal Point Method to solve the model, and the algorithm is compared with the Particle Swarm Optimization (PSO), Differential Evolutionary (DE), Tabu Search (TS), Sparrow Search (SS) algorithms, and the exact solver Linear Interactive and General Optimizer (LINGO). The examples verify the validity of the models and algorithms, with an average reduction of 6.2% and 3.6% in logistics costs and carbon emissions in the case of demand determination, and at the confidence level of 0.9 under demand uncertainty, both logistics costs and carbon emissions are decreased to varying degrees. Bioaccumulation and biomagnification of anthropogenic particles are crucial factors in assessing microplastic impacts to marine ecosystems. Microplastic pollution poses a significant threat to iconic and often endangered species but examining their tissues and gut contents for contaminant analysis via lethal sampling is challenging due to ethical concerns and animal care restrictions. Incorporating empirical data from prey items and fecal matter into models can help trace microplastic movement through food webs. In this study, the Galápagos penguin food web served as an indicator species to assess microplastic bioaccumulation and biomagnification potential using trophodynamic Ecopath with Ecosim (EwE) modelling with Ecotracer. Empirical data collected from surface seawater near Galápagos penguin colonies, zooplankton, penguin prey, and penguin scat in October 2021 were used to inform the ecosystem model. Multiple scenarios, including a 99% elimination rate, were employed to assess model sensitivity. Model predictions revealed that microplastics can bioaccumulate in all predator-prey relationships, but biomagnification is highly dependent on the elimination rate. It establishes the need for more research into elimination rates of different plastics, which is a critical missing gap in current microplastic ecotoxicological and bioaccumulation science. Compared to empirical data, modelling efforts underpredicted microplastic concentrations in zooplankton and over-predicted concentrations in fish. Ultimately, the ecosystem modelling provides novel insights into potential microplastics' bioaccumulation and biomagnification risks. Background: High-pathogenicity avian influenza (HPAI) has become a conservation threat to wild birds. Therefore, suitable vaccine technology and practical application methods require investigation., Methods: Twenty-four African penguins (Spheniscus demersus) were vaccinated with either a conventional inactivated clade 2.3.4.4b H5N8 HPAI whole virus or a tobacco leaf-produced H5 haemagglutinin-based virus-like particle (VLP). Six birds received a second dose of the inactivated vaccine. Antibody responses were assessed and compared by employing haemagglutination inhibition tests., Results: A second dose of inactivated vaccine was required to induce antibody titres above the level required to suppress virus shedding, while a single dose of VLP vaccine produced these levels by day 14, and one bird still had antibodies on day 430., Limitations: Bacterial contamination of the VLP vaccine limited the monitoring period and sample size in that treatment group, and it was not possible to perform a challenge study with field virus., Conclusion: VLP vaccines offer a more practical option than inactivated whole viruses, especially in logistically challenging situations involving wild birds., (© 2023 The Authors. As charismatic and iconic species, penguins can act as "ambassadors" or flagship species to promote the conservation of marine habitats in the Southern Hemisphere. Unfortunately, there is a lack of reliable, comprehensive, and systematic analysis aimed at compiling spatially explicit assessments of the multiple impacts that the world's 18 species of penguin are facing. We provide such an assessment by combining the available penguin occurrence information from Global Biodiversity Information Facility (>800,000 occurrences) with three main stressors: climate-driven environmental changes at sea, industrial fisheries, and human disturbances on land. Our analyses provide a quantitative assessment of how these impacts are unevenly distributed spatially within species' distribution ranges. Consequently, contrasting pressures are expected among species, and populations within species. The areas coinciding with the greatest impacts for penguins are the coast of Perú, the Patagonian Shelf, the Benguela upwelling region, and the Australian and New Zealand coasts. When weighting these potential stressors with species-specific vulnerabilities, Humboldt (Spheniscus humboldti), African (Spheniscus demersus), and Chinstrap penguin (Pygoscelis antarcticus) emerge as the species under the most pressure. Our approach explicitly differentiates between climate and human stressors, since the more achievable management of local anthropogenic stressors (e.g., fisheries and land-based threats) may provide a suitable means for facilitating cumulative impacts on penguins, especially where they may remain resilient to global processes such as climate change. Moreover, our study highlights some poorly represented species such as the Northern Rockhopper (Eudyptes moseleyi), Snares (Eudyptes robustus), and Erect-crested penguin (Eudyptes sclateri) that need internationally coordinated efforts for data acquisition and data sharing to understand their spatial distribution properly., (© 2024 The Authors. The Humboldt penguin ( Spheniscus humboldti ) population at the Punta San Juan Marine Protected Area in Peru is considered critical to the long-term sustainability of this endangered species in Peru. Exposure of the rookery to environmental toxicants is a mounting concern because of regional growth of industries and human populations. Whole blood samples were collected from 30 free-ranging penguins in 2011 as part of a broader population health monitoring program. Dried blood spots (DBS) containing 50 µl of blood were prepared and analyzed to assess exposure to five groups of environmental contaminants. Concentrations of elements arsenic, cadmium, iron, lead, mercury, selenium, and thallium were analyzed using inductively coupled plasma mass spectrometry. Persistent organic pollutant concentrations were measured using gas chromatography-tandem mass spectrometry to analyze organochlorine pesticides (OCP; p,p'-DDT, p,p'-DDE, β-hexachlorocyclohexane, t-nonachlor, and oxychlordane), polychlorinated biphenyls (congeners 138 and 153), and polybrominated flame retardants (polybrominated biphenyl-153 and polybrominated diphenyl ether congeners 47 and 99). Per- and polyfluoroalkyl substances, including perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid were measured using liquid chromatography-tandem mass spectrometry. Results revealed low levels of exposure to these selected contaminants, at levels not considered to be of concern for wildlife health. DBS methodology was considered effective in a field-based setting for quantification of whole blood concentrations of environmental contaminants in penguins.

The taxonomic status of strain P5891 T , isolated from an Adélie penguin beak swab, was investigated. Based on the 16S rRNA gene sequence, the strain was identified as a potentially novel Corynebacterium species, with the highest sequence similarities to Corynebacterium rouxii FRC0190 T (96.7 %) and Corynebacterium epidermidicanis DSM 45586 T (96.6 %). The average nucleotide identity values between strain P5891 T and C. rouxii FRC0190 T and C. epidermidicanis DSM 45586 T were 68.2 and 69.2 %, respectively. The digital DNA-DNA hybridization values between strain P5891 T and C. rouxii FRC0190 T and C. epidermidicanis DSM 45586 T were 23.7 and 21.4 %, respectively. Phylogenetic trees based on the 16S rRNA sequence placed strain P5891 T in a separate branch with Corynebacterium canis 1170 T and Corynebacterium freiburgense 1045 T , while a phylogenomic tree based on the Corynebacterium species core genome placed the strain next to Corynebacterium choanae 200CH T . Extensive phenotyping and genomic analyses clearly confirmed that strain P5891 T represents a novel species of the genus Corynebacterium , for which the name Corynebacterium mendelii sp. nov. is proposed, with the type strain P5891 T (=CCM 8862 T =LMG 31627 T ).

Our aim was to estimate the population of emperor penguins (Aptenodytes fosteri) using a single synoptic survey. We examined the whole continental coastline of Antarctica using a combination of medium resolution and Very High Resolution (VHR) satellite imagery to identify emperor penguin colony locations. Where colonies were identified, VHR imagery was obtained in the 2009 breeding season. The remotely-sensed images were then analysed using a supervised classification method to separate penguins from snow, shadow and guano. Actual counts of penguins from eleven ground truthing sites were used to convert these classified areas into numbers of penguins using a robust regression algorithm.We found four new colonies and confirmed the location of three previously suspected sites giving a total number of emperor penguin breeding colonies of 46. We estimated the breeding population of emperor penguins at each colony during 2009 and provide a population estimate of ~238,000 breeding pairs (compared with the last previously published count of 135,000-175,000 pairs). Based on published values of the relationship between breeders and non-breeders, this translates to a total population of ~595,000 adult birds.There is a growing consensus in the literature that global and regional emperor penguin populations will be affected by changing climate, a driver thought to be critical to their future survival. However, a complete understanding is severely limited by the lack of detailed knowledge about much of their ecology, and importantly a poor understanding of their total breeding population. To address the second of these issues, our work now provides a comprehensive estimate of the total breeding population that can be used in future population models and will provide a baseline for long-term research.

Seabirds are highly vocal on land where acoustic communication plays a crucial role in reproduction. Yet, seabirds spend most of their life at sea. They have developed a number of morphological, physiological and behavioural adaptations to forage in the marine environment. The use of acoustic signals at sea could potentially enhance seabirds’ foraging success, but remains largely unexplored. Penguins emit vocalisations from the sea surface when commuting, a behaviour possibly associated with group formation at sea. Still, they are unique in their exceptional diving abilities and feed entirely underwater. Other air-breathing marine predators that feed under water, like cetaceans, pinnipeds and marine turtles, are known to emit sound underwater, but such behaviour has not yet been described in seabirds. We aimed to assess the potential prevalence and diversity of vocalisations emitted underwater by penguins. We chose three study species from three different genera, and equipped foraging adults with video cameras with built-in microphones. We recorded a total of 203 underwater vocalisation from all three species during 4 h 43 min of underwater footage. Vocalisations were very short in duration (0.06 s on average), with a frequency of maximum amplitude averaging 998 Hz, 1097 Hz and 680 Hz for King, Gentoo and Macaroni penguins, respectively. All vocalisations were emitted during feeding dives and more than 50% of them were directly associated with hunting behaviour, preceeded by an acceleration (by 2.2 s on average) and/or followed by a prey capture attempt (after 0.12 s on average). The function of these vocalisations remain speculative. Although it seems to be related to hunting behaviour, these novel observations warrant further investigation.

Fossil penguins from late Neogene sediments are important for understanding the origins of modern penguins. Recent advances in penguin taxonomy and geometric morphometrics allow us to revisit previously described Pliocene fossils and potentially gain new insight into the evolutionary relationships of early crown penguin species. Here we re-evaluate the holotype of Tereingaornis moisleyi, a Pliocene fossil penguin discovered near Te Reinga Falls in the North Island of New Zealand. The holotype (hereafter referred to as the Te Reinga Falls fossil) does not possess any apomorphies that justify recognition as a distinct genus or species, and cannot be distinguished from the living crested penguins (Eudyptes spp.) or yellow-eyed penguin (Megadyptes antipodes) using discrete osteological characters or morphometric analysis. We propose that Tereingaornis moisleyi be considered a nomen dubium. The Te Reinga Falls fossil indicates that a penguin approximately the size of the extant tawaki (Fiordland crested penguin, Eudyptes pachyrhynchus) was present in New Zealand during the middle of the Pliocene (Waipipian Stage, 3.7–3.0 Ma), but a more complete specimen would be needed to confidently resolve the evolutionary relationships between this fossil and other penguins. Information on the extent of genetic differentiation among seabird populations is essential for conservation planning. Here we used, for the first time in little penguins (Eudyptula minor), a next generation sequencing approach to investigate population genetic structure, gene flow and inbreeding among eight colonies in South Australia. We found that Troubridge Island individuals were genetically distinct from the other sampled colonies, and that colonies on all islands were connected by moderate levels of gene flow possibly due to migration from the Kangaroo Island colonies. While further genetic sampling is needed, these data support the idea that South Australia may be a zone of subtle and rapid genetic change for little penguins. [ABSTRACT FROM AUTHOR]

An 11-day-old little blue penguin (Eudyptula minor) died unexpectedly. Prior to hatching, the egg experienced trauma and resultant defects were repaired. The chick hatched without complication and was clinically normal prior to death. Necropsy revealed congested lungs. Histologic examination showed moderate nonsuppurative encephalitis with focally extensive neuronal necrosis and intranuclear inclusions in neurons within necrotic foci. Herpesvirus DNA was detected in brain tissue with a generic herpesvirus polymerase chain reaction. Sanger sequencing demonstrated 100% and 98% sequence homology to sphenicid alphaherpesvirus 1 and penguin herpesvirus 2, respectively. In situ hybridization demonstrated large amounts of herpesvirus nucleic acid in intranuclear inclusions and neuronal nuclei. Combined histology, polymerase chain reaction, Sanger sequencing, and in situ hybridization results were most consistent with herpesviral encephalitis, most likely caused by sphenicid alphaherpesvirus 1. To our knowledge, this is the first report of a herpesvirus infection causing encephalitis in a penguin and the first report of herpesvirus in this species. [ABSTRACT FROM AUTHOR]

Background: The astonishing variety of sounds that birds can produce has been the subject of many studies aiming to identify the underlying anatomical and physical mechanisms of sound production. An interesting feature of some bird vocalisations is the simultaneous production of two different frequencies. While most work has been focusing on songbirds, much less is known about dual-sound production in non-passerines, although their sound production organ, the syrinx, would technically allow many of them to produce "two voices". Here, we focus on the king penguin, a colonial seabird whose calls consist of two fundamental frequency bands and their respective harmonics. The calls are produced during courtship and for partner and offspring reunions and encode the birds' identity. We dissected, μCT-scanned and analysed the vocal tracts of six adult king penguins from Possession Island, Crozet Archipelago. Results: King penguins possess a bronchial type syrinx that, similarly to the songbird's tracheobronchial syrinx, has two sets of vibratory tissues, and thus two separate sound sources. Left and right medial labium differ consistently in diameter between 0.5 and 3.2%, with no laterality between left and right side. The trachea has a conical shape, increasing in diameter from caudal to cranial by 16%. About 80% of the king penguins' trachea is medially divided by a septum consisting of soft elastic tissue (septum trachealis medialis). Conclusions: The king penguins' vocal tract appears to be mainly adapted to the life in a noisy colony of a species that relies on individual vocal recognition. The extent between the two voices encoding for individuality seems morphologically dictated by the length difference between left and right medial labium. The septum trachealis medialis might support this extent and could therefore be an important anatomical feature that aids in the individual recognition process. [ABSTRACT FROM AUTHOR]
