1. Interdisciplinary Training for Future Leaders through the CREATE-REDEVELOP Graduate Student Program

Abstract

REDEVELOP is a graduate student training program funded by the NSERC-CREATE grant, starting in 2017. Its goal is to support the training of new professionals and researchers (> 100) who will be the next generation of science and engineering leaders and policymakers in Canada. The program has successfully developed a framework for operating almost completely virtually, well ahead of the world's transition to online learning due to the COVID-19 pandemic. Our psychology lab, The Individual and Team Performance (ITP) lab, has dedicated over a decade to researching and designing tools that enhance specific training and skill growth necessary for effective remote teamwork. In partnership with the REDEVELOP program, we support students in navigating the unique interpersonal and collaboration challenges posed by virtual team environments. We will discuss how a complex and multidisciplinary program succeeds in training graduate students to become stronger academics, practitioners, and communicators of knowledge. 2. Early Cold Stored Platelet Transfusion Following Severe Injury: A Randomized Clinical Trial.

Abstract

OBJECTIVE: To determine the feasibility, efficacy, and safety of early cold stored platelet transfusion compared with standard care resuscitation in patients with hemorrhagic shock. BACKGROUND: Data demonstrating the safety and efficacy of early cold stored platelet transfusion are lacking following severe injury. METHODS: A phase 2, multicenter, randomized, open label, clinical trial was performed at 5 US trauma centers. Injured patients at risk of large volume blood transfusion and the need for hemorrhage control procedures were enrolled and randomized. The intervention was the early transfusion of a single apheresis cold stored platelet unit, stored for up to 14 days versus standard care resuscitation. The primary outcome was feasibility and the principal clinical outcome for efficacy and safety was 24-hour mortality. RESULTS: Mortality at 24 hours was 5.9% in patients who were randomized to early cold stored platelet transfusion compared with 10.2% in the standard care arm (difference, -4.3%; 95% CI, -12.8% to 3.5%; P =0.26). No significant differences were found for any of the prespecified ancillary outcomes. Rates of arterial and/or venous thromboembolism and adverse events did not differ across treatment groups. CONCLUSIONS AND RELEVANCE: In severely injured patients, early cold stored platelet transfusion is feasible, safe and did not result in a significant lower rate of 24-hour mortality. Early cold stored platelet transfusion did not result in a higher incidence of arterial and/or venous thrombotic complications or adverse events. The storage age of the cold stored platelet product was not associated with significant outcome differences. TRIAL REGISTRATION: ClinicalTrials.gov identifier: NCT04667468.

3. Extension of graph-accelerated non-intrusive polynomial chaos to high-dimensional uncertainty quantification through the active subspace method

Abstract

The recently introduced graph-accelerated non-intrusive polynomial chaos (NIPC) method has shown effectiveness in solving a broad range of uncertainty quantification (UQ) problems with multidisciplinary systems. It uses integration-based NIPC to solve the UQ problem and generates the quadrature rule in a desired tensor structure, so that the model evaluations can be efficiently accelerated through the computational graph transformation method, Accelerated Model evaluations on Tensor grids using Computational graph transformations (AMTC). This method is efficient when the model's computational graph possesses a certain type of sparsity which is commonly the case in multidisciplinary problems. However, it faces limitations in high-dimensional cases due to the curse of dimensionality. To broaden its applicability in high-dimensional UQ problems, we propose AS-AMTC, which integrates the AMTC approach with the active subspace (AS) method, a widely-used dimension reduction technique. In developing this new method, we have also developed AS-NIPC, linking integration-based NIPC with the AS method for solving high-dimensional UQ problems. AS-AMTC incorporates rigorous approaches to generate orthogonal polynomial basis functions for lower-dimensional active variables and efficient quadrature rules to estimate their coefficients. The AS-AMTC method extends AS-NIPC by generating a quadrature rule with a desired tensor structure. This allows the AMTC method to exploit the computational graph sparsity, leading to efficient model evaluations. In an 81-dimensional UQ problem derived from an air-taxi trajectory optimization scenario, AS-NIPC demonstrates a 30% decrease in relative error compared to the existing methods, while AS-AMTC achieves an 80% reduction.

Abstract

This study is an exploration of collective patterns of conscious experience, as described by various psychological models, using a self-report questionnaire: The Consciousness Quotient Inventory (CQ-i). The CQ-i evaluates patterns of behaviors, attitudes, and attentional styles as well as the usage of conscious skills, awareness, and the capacity to “feel awake and alive,” providing a complex exploration of conscious experience. A set of 237 items covering major aspects of the subjective conscious experience was selected to detect the phenomenal patterns of subjective conscious experience. An exploratory factor analysis on a large sample (N = 2,360), combined with our previous meta-research on conceptual convergence of conscious experiences, revealed that these experiences appear to have 15 patterns common to all of us. A sample with a quasi-normal distribution (n = 2,266) was employed for standardization and classification of scores (M = 100; SD = 15). The study provides a conceptual framework for future in-depth studies on collective patterns of self-awareness, inner growth dynamics, and psychological maturity.

5. A gradient-enhanced univariate dimension reduction method for uncertainty propagation

Abstract

The univariate dimension reduction (UDR) method stands as a way to estimate the statistical moments of the output that is effective in a large class of uncertainty quantification (UQ) problems. UDR's fundamental strategy is to approximate the original function using univariate functions so that the UQ cost only scales linearly with the dimension of the problem. Nonetheless, UDR's effectiveness can diminish when uncertain inputs have high variance, particularly when assessing the output's second and higher-order statistical moments. This paper proposes a new method, gradient-enhanced univariate dimension reduction (GUDR), that enhances the accuracy of UDR by incorporating univariate gradient function terms into the UDR approximation function. Theoretical results indicate that the GUDR approximation is expected to be one order more accurate than UDR in approximating the original function, and it is expected to generate more accurate results in computing the output's second and higher-order statistical moments. Our proposed method uses a computational graph transformation strategy to efficiently evaluate the GUDR approximation function on tensor-grid quadrature inputs, and use the tensor-grid input-output data to compute the statistical moments of the output. With an efficient automatic differentiation method to compute the gradients, our method preserves UDR's linear scaling of computation time with problem dimension. Numerical results show that the GUDR is more accurate than UDR in estimating the standard deviation of the output and has a performance comparable to the method of moments using a third-order Taylor series expansion.

12. Accelerating model evaluations in uncertainty propagation on tensor grids using computational graph transformations

Abstract

Methods such as non-intrusive polynomial chaos (NIPC), and stochastic collocation are frequently used for uncertainty propagation problems. Particularly for low-dimensional problems, these methods often use a tensor-product grid for sampling the space of uncertain inputs. A limitation of this approach is that it encounters a significant challenge: the number of sample points grows exponentially with the increase of uncertain inputs. Current strategies to mitigate computational costs abandon the tensor structure of sampling points, with the aim of reducing their overall count. Contrastingly, our investigation reveals that preserving the tensor structure of sample points can offer distinct advantages in specific scenarios. Notably, by manipulating the computational graph of the targeted model, it is feasible to avoid redundant evaluations at the operation level to significantly reduce the model evaluation cost on tensor-grid inputs. This paper presents a pioneering method: Accelerated Model Evaluations on Tensor grids using Computational graph transformations (AMTC). The core premise of AMTC lies in the strategic modification of the computational graph of the target model to algorithmically remove the repeated evaluations on the operation level. We implemented the AMTC method within the compiler of a new modeling language called the Computational System Design Language (CSDL). We demonstrate the effectiveness of AMTC by using it with the full-grid NIPC method to solve four low-dimensional UQ problems involving an analytical piston model, a multidisciplinary unmanned aerial vehicle design model, a multi-point air taxi mission analysis model, and a single-disciplinary rotor model, respectively. For three of the four test problems, AMTC reduces the model evaluation cost by between 50% and 90%, making the full-grid NIPC the most efficacious method to use among the UQ methods implemented.

Abstract

The conversion of biomass into sugar platform compounds is very important for the biorefinery industry. Pretreatment is essential to the biomass of the sugar platform, however, the lignin obtained by pretreatment, as a key part of lignocellulose, generally has a passive effect on the enzymatic hydrolysis of cellulose into sugars. In this study, p-TsOH (p-toluenesulfonic acid), DES (Deep eutectic solvent) and CAOSA (cooking with active oxygen and solid alkali) pretreatment ways were used to fraction lignin from bamboo biomass. After CAOSA treatment, the hydrolysis efficiency of the pulp was 95.57%. Moreover, the effect of different treatment methods on lignin properties was studied and the promotion effect of lignin was investigated by adding it to the cellulose enzymatic hydrolysis system. In this work, the results showed that CAOSA-extracted lignin with lower Đ (1.31–1.25) had a better adsorption effect on the enzyme protein. p-TsOH-extracted lignin with a larger S/G ratio enhanced the inhibition of enzymatic hydrolysis. In addition, the presence of -COOHs in lignin could reduce its inhibitory effect on cellulose saccharification.

Abstract

Abstract Background Optimizing resuscitation to reduce inflammation and organ dysfunction following human trauma-associated hemorrhagic shock is a major clinical hurdle. This is limited by the short duration of pre-clinical studies and the sparsity of early data in the clinical setting. Methods We sought to bridge this gap by linking preclinical data in a porcine model with clinical data from patients from the Prospective, Observational, Multicenter, Major Trauma Transfusion (PROMMTT) study via a three-compartment ordinary differential equation model of inflammation and coagulation. Results The mathematical model accurately predicts physiologic, inflammatory, and laboratory measures in both the porcine model and patients, as well as the outcome and time of death in the PROMMTT cohort. Model simulation suggests that resuscitation with plasma and red blood cells outperformed resuscitation with crystalloid or plasma alone, and that earlier plasma resuscitation reduced injury severity and increased survival time. Conclusions This workflow may serve as a translational bridge from pre-clinical to clinical studies in trauma-associated hemorrhagic shock and other complex disease settings.

26. High-temperature thermal conductivity measurements of macro-porous graphite

Abstract

Graphite is a unique material for high temperature applications and will likely become increasingly important as we attempt to electrify industrial applications. However, high-quality graphite can be expensive, limiting the cost-competitiveness of high-quality graphite technologies. Here, we investigate the thermal properties of low-cost, low-quality, macro-porous graphite to determine the tradeoff between cost and thermal performance. We use laser flash analysis (LFA) to measure the thermal diffusivity of graphite at high temperatures. However, due to the large pores in the graphite samples preventing uniform laser flash heating, we must apply a thick coating to achieve the required flat, parallel surfaces for LFA measurements. The presence of the coating directly impacts the measured diffusivity, not only because of the added thickness but also because of the sample/coating interface profile generated. We therefore develop a methodology based on finite element modeling of a variety of sample/coating interface profiles to extract properties of the sample. Validating the methodology against a reference sample demonstrates a mean absolute percentage error of 8.5%, with potential improvement with better sample characterization. We show low-cost, low-quality graphite has a thermal conductivity of ~10 W/m/K up to 1000$^{\circ}$C, which is an order of magnitude lower than high-quality graphite, but contributions from photon conductivity may result in higher conductivities at higher temperatures. Overall, we demonstrate an approach for measuring thermal properties of macro-porous materials at high temperatures, and apply the approach to measuring thermal conductivity of porous graphite, which will aid in the design of high-temperature systems for cost-competitive decarbonization.

27. Accuracy Booster: Enabling 4-bit Fixed-point Arithmetic for DNN Training

Abstract

The unprecedented demand for computing resources to train DNN models has led to a search for minimal numerical encoding. Recent state-of-the-art (SOTA) proposals advocate for multi-level scaled narrow bitwidth numerical formats. In this paper, we show that single-level scaling is sufficient to maintain training accuracy while maximizing arithmetic density. We identify a previously proposed single-level scaled format for 8-bit training, Hybrid Block Floating Point (HBFP), as the optimal candidate to minimize. We perform a full-scale exploration of the HBFP design space using mathematical tools to study the interplay among various parameters and identify opportunities for even smaller encodings across layers and epochs. Based on our findings, we propose Accuracy Booster, a mixed-mantissa HBFP technique that uses 4-bit mantissas for over 99% of all arithmetic operations in training and 6-bit mantissas only in the last epoch and first/last layers. We show Accuracy Booster enables increasing arithmetic density over all other SOTA formats by at least 2.3x while achieving state-of-the-art accuracies in 4-bit training.

Abstract

Drugs that induce reversible slowing of metabolic and physiological processes would have great value for organ preservation, especially for organs with high susceptibility to hypoxia-reperfusion injury, such as the heart. Using whole-organism screening of metabolism, mobility, and development in Xenopus, we identified an existing drug, SNC80, that rapidly and reversibly slows biochemical and metabolic activities while preserving cell and tissue viability. Although SNC80 was developed as a delta opioid receptor activator, we discovered that its ability to slow metabolism is independent of its opioid modulating activity as a novel SNC80 analog (WB3) with almost 1000 times less delta opioid receptor binding activity is equally active. Metabolic suppression was also achieved using SNC80 in microfluidic human organs-on-chips, as well as in explanted whole porcine hearts and limbs, demonstrating the cross-species relevance of this approach and potential clinical relevance for surgical transplantation. Pharmacological induction of physiological slowing in combination with organ perfusion transport systems may offer a new therapeutic approach for tissue and organ preservation for transplantation, trauma management, and enhancing patient survival in remote and low-resource locations.

Abstract

OBJECTIVES:. Anemia has been associated with an increased risk of both cardiac arrest and stroke, frequent complications of COVID-19. The effect of hemoglobin level at ICU admission on a composite outcome of cardiac arrest or stroke in an international cohort of COVID-19 patients was investigated. DESIGN:. Retrospective analysis of prospectively collected database. SETTING:. A registry of COVID-19 patients admitted to ICUs at over 370 international sites was reviewed for patients diagnosed with cardiac arrest or stroke up to 30 days after ICU admission. Anemia was defined as: normal (hemoglobin ≥ 12.0 g/dL for women, ≥ 13.5 g/dL for men), mild (hemoglobin 10.0–11.9 g/dL for women, 10.0–13.4 g/dL for men), moderate (hemoglobin ≥ 8.0 and < 10.0 g/dL for women and men), and severe (hemoglobin < 8.0 g/dL for women and men). PATIENTS:. Patients older than 18 years with acute COVID-19 infection in the ICU. INTERVENTIONS:. None. MEASUREMENTS AND MAIN RESULTS:. Of 6926 patients (median age = 59 yr, male = 65%), 760 patients (11.0%) experienced stroke (2.0%) and/or cardiac arrest (9.4%). Cardiac arrest or stroke was more common in patients with low hemoglobin, occurring in 12.8% of patients with normal hemoglobin, 13.3% of patients with mild anemia, and 16.7% of patients with moderate/severe anemia. Time to stroke or cardiac arrest by anemia status was analyzed using Cox proportional hazards regression with death as a competing risk. Covariates selected through clinical knowledge were age, sex, comorbidities (diabetes, hypertension, obesity, and cardiac or neurologic conditions), pandemic era, country income, mechanical ventilation, and extracorporeal membrane oxygenation. Moderate/severe anemia was associated with a higher risk of cardiac arrest or stroke (hazard ratio, 1.32; 95% CI, 1.05–1.67). CONCLUSIONS:. In an international registry of ICU patients with COVID-19, moderate/severe anemia was associated with increased hazard of cardiac arrest or stroke.

Abstract

BackgroundExtremely preterm (EPT) birth has been related to dysregulation of stress responses and behavioral/learning problems at school age. Early adverse experiences can blunt HPA axis reactivity. We hypothesized that an attenuated cortisol awakening response would be associated with developmental and behavioral problems at school age in EPT children.MethodsThis secondary analysis of a sub-cohort of the SUPPORT study included children born between 24 and 27 weeks, evaluated at 6-7 years with a neurodevelopmental battery and cortisol measures. Differences were tested between EPT and a term-born group. Relationships of cortisol awakening response to test scores were analyzed.ResultsCortisol was measured in 110 EPT and 29 term-born 6-7 year olds. Unadjusted WISC-IV and NEPSY-II scores were significantly worse among EPT children only. Conners Parent Rating Scale behavior scores were significantly worse among EPT children. After adjusting for covariates, blunted cortisol awakening responses were found to be associated with poorer scores on memory tests and greater problems with inattention for the EPT group (p

36. Predictability of Existing IOL Formulas After Cataract Surgery in Patients with a Previous History of Radial Keratotomy: A Retrospective Cohort Study and Literature Review

Abstract

Abstract Introduction This study aims to evaluate the accuracy of 12 different intraocular lens (IOL) power calculation formulas for post-radial keratotomy (RK) eyes. The investigation utilizes recent advances in topography/tomography devices and artificial intelligence (AI)-based calculators, comparing the results to those reported in current literature to assess the efficacy and predictability of IOL calculations for this patient group. Methods In this retrospective study, 37 eyes from 24 individuals with a history of RK who underwent cataract surgery at Hoopes Vision Center were analyzed. Biometry and corneal topography measurements were taken preoperatively. Subjective refraction was obtained 6 months postoperatively. Twelve different IOL power calculations were used, including the American Society of Cataract and Refractive Surgery (ASCRS) post-RK online formula, and the Barrett True K, Double K modified-Holladay 1, Haigis-L, Panacea, Camellin-Calossi, Emmetropia Verifying Optical (EVO) 2.0, Kane, and Prediction Enhanced by Artificial Intelligence and output Linearization-Debellemanière, Gatinel, and Saad (PEARL-DGS) formulas. Outcome measures included median absolute error (MedAE), mean absolute error (MAE), arithmetic mean error (AME), and percentage of eyes achieving refractive prediction errors (RPE) within ± 0.50 D, ± 0.75 D, and ± 1 D for each formula. A search of the literature was also performed by two independent reviewers based on relevant formulas. Results Overall, the best performing IOL power calculations were the Camellin-Calossi (MedAE = 0.515 D), the ASCRS average (MedAE = 0.535 D), and the EVO (MedAE = 0.545 D) and Kane (MedAE = 0.555 D) AI-based formulas. The EVO and Kane formulas along with the ASCRS calculation performed similarly, with 48.65% of eyes scoring within ± 0.50 D of the target range, while the Equivalent Keratometry Reading (EKR) 65 Holladay formula achieved the greatest percentage of eyes scoring within ± 0.25 D of the target range (35.14%). Additionally, the EVO 2.0 formula achieved 64.86% of eyes scoring within the ± 0.75 D RPE category, while the Kane formula achieved 75.68% of eyes scoring within the ± 1 D RPE category. There was no significant difference in MAE between the established and newer generation formulas (P > 0.05). Abstract

Abstract Injury mechanism is an important consideration when conducting clinical trials in trauma. Mechanisms of injury may be associated with differences in mortality risk and immune response to injury, impacting the potential success of the trial. We sought to characterize clinical and endothelial cell damage marker differences across blunt and penetrating injured patients enrolled in three large, prehospital randomized trials which focused on hemorrhagic shock. In this secondary analysis, patients with systolic blood pressure 108 were included. In addition, patients with both blunt and penetrating injuries were excluded. The primary outcome was 30-day mortality. Mortality was characterized using Kaplan–Meier and Cox proportional-hazards models. Generalized linear models were used to compare biomarkers. Chi squared tests and Wilcoxon rank-sum were used to compare secondary outcomes. We characterized data of 696 enrolled patients that met all secondary analysis inclusion criteria. Blunt injured patients had significantly greater 24-h (18.6% vs. 10.7%, log rank p = 0.048) and 30-day mortality rates (29.7% vs. 14.0%, log rank p = 0.001) relative to penetrating injured patients with a different time course. After adjusting for confounders, blunt mechanism of injury was independently predictive of mortality at 30-days (HR 1.84, 95% CI 1.06–3.20, p = 0.029), but not 24-h (HR 1.65, 95% CI 0.86–3.18, p = 0.133). Abstract

Drug repurposing requires distinguishing established drug class targets from novel molecule-specific mechanisms and rapidly derisking their therapeutic potential in a time-critical manner, particularly in a pandemic scenario. In response to the challenge to rapidly identify treatment options for COVID-19, several studies reported that statins, as a drug class, reduce mortality in these patients. However, it is unknown if different statins exhibit consistent function or may have varying therapeutic benefit. A Bayesian network tool was used to predict drugs that shift the host transcriptomic response to SARS-CoV-2 infection towards a healthy state. Drugs were predicted using 14 RNA-sequencing datasets from 72 autopsy tissues and 465 COVID-19 patient samples or from cultured human cells and organoids infected with SARS-CoV-2. Top drug predictions included statins, which were then assessed using electronic medical records containing over 4,000 COVID-19 patients on statins to determine mortality risk in patients prescribed specific statins versus untreated matched controls. The same drugs were tested in Vero E6 cells infected with SARS-CoV-2 and human endothelial cells infected with a related OC43 coronavirus. Simvastatin was among the most highly predicted compounds (14/14 datasets) and five other statins, including atorvastatin, were predicted to be active in > 50% of analyses. Analysis of the clinical database revealed that reduced mortality risk was only observed in COVID-19 patients prescribed a subset of statins, including simvastatin and atorvastatin. In vitro testing of SARS-CoV-2 infected cells revealed simvastatin to be a potent direct inhibitor whereas most other statins were less effective. Simvastatin also inhibited OC43 infection and reduced cytokine production in endothelial cells. Statins may differ in their ability to sustain the lives of COVID-19 patients despite having a shared drug target and lipid-modifying mechanism of action. These findings highlight the value of target-agnostic drug prediction coupled with patient databases to identify and clinically evaluate non-obvious mechanisms and derisk and accelerate drug repurposing opportunities.

Abstract

Pubesamides A and B are structurally unusual monoterpene N-benzoyltyramides isolated from Casimiroa pubescens, a tropical fruiting tree with known psychotropic properties. Herein we report the structural affirmation of both natural products through a chemical synthesis that features a Lewis-acid assisted cross-metathesis reaction between a 1,1-disubstituted alkene and a dienone in the presence of the Hoveyda-Grubbs second generation catalyst. Stereochemically pure samples of pubesamide A and B isomerise upon standing, suggesting they are both genuine natural products.

Abstract

Introduction Given the increasing prevalence of both obesity and pre-diabetes in pregnant adults, there is growing interest in identifying hyperglycaemia in early pregnancy to optimise maternal and perinatal outcomes. Multiple organisations recommend first-trimester diabetes screening for individuals with risk factors; however, the benefits and drawbacks of detecting glucose abnormalities more mild than overt diabetes in early gestation and the best screening method to detect such abnormalities remain unclear.Methods and analysis The goal of the Glycemic Observation and Metabolic Outcomes in Mothers and Offspring study (GO MOMs) is to evaluate how early pregnancy glycaemia, measured using continuous glucose monitoring and oral glucose tolerance testing, relates to the diagnosis of gestational diabetes (GDM) at 24–28 weeks’ gestation (maternal primary outcome) and large-for-gestational-age birth weight (newborn primary outcome). Secondary objectives include relating early pregnancy glycaemia to other adverse pregnancy outcomes and comprehensively detailing longitudinal changes in glucose over the course of pregnancy. GO MOMs enrolment began in April 2021 and will continue for 3.5 years with a target sample size of 2150 participants.Ethics and dissemination GO MOMs is centrally overseen by Vanderbilt University’s Institutional Review Board and an Observational Study Monitoring Board appointed by National Institute of Diabetes and Digestive and Kidney Diseases. GO MOMs has potential to yield data that will improve understanding of hyperglycaemia in pregnancy, elucidate better approaches for early pregnancy GDM screening, and inform future clinical trials of early GDM treatment.Trial registration number NCT04860336.

Abstract Detecting environmental DNA (eDNA) of numerous organisms from the same samples has been revolutionized by metabarcoding. However, utilizing the vast amounts of data generated from metabarcoding to predict occupancy probabilities for co‐occurring species is currently rare. Here, we demonstrate how metabarcoding data can be used to advance community ecology research through a case study using replicate stream water samples and Bayesian occupancy models to test hypotheses of eDNA occurrence for a native fish (brook trout, Salvelinus fontinalis), its major ectoparasite (gill lice, Salmincola edwardsii), and an introduced potential competitor (brown trout, Salmo trutta). Gill lice DNA occupancy was positively associated with brook trout biomass determined via electrofishing conducted near eDNA sampling sites, suggesting gill lice occupancy is dependent on host density. Leveraging site‐specific molecular operational taxonomic units identified from metabarcoding, DNA occupancy of trout and gill lice was often positively predicted by species richness of aquatic insect orders trout commonly feed on, which are also environmental quality indicators. Thus, high‐quality habitats that environmentally sensitive salmonids and their primary prey rely on may promote higher fish occupancy rates, further facilitating the spread of fish parasites. An increasing amount of community‐level data is being generated from global metabarcoding efforts, and we suggest our framework could be broadly implemented to enhance understanding of factors impacting distributions of co‐occurring species, reveal new ecological phenomena, and support management and conservation efforts.

Abstract

Abstract Analysis of environmental DNA (eDNA) has been successfully used across freshwater ecological parasitology to inform management of ecologically and economically important species. However, most studies have used species‐specific quantitative polymerase chain reaction (qPCR) assays to detect target taxa. While generally effective, this approach limits the amount of community and management‐supporting data that can be obtained from eDNA samples. If eDNA metabarcoding could be conducted with the same accuracy of a single species approach, researchers could simultaneously detect a target species while obtaining vast community data from eDNA samples. We sampled 38 freshwater sites on Fort McCoy, Wisconsin and compared qPCR to metabarcoding for eDNA detection of the ectoparasitic gill louse Salmincola edwardsii, an obligate parasite of Salvelinus fishes (chars). We found no evidence to suggest S. edwardsii occupancy or detection probabilities differed between qPCR and metabarcoding. Further, we found that the number of S. edwardsii reads from metabarcoding were negatively predictive of CT values from qPCR (CT value indicates cycle a significant amount of target eDNA is detected, with lower CTs indicative of more DNA), demonstrating that our metabarcoding reads positively predicted qPCR DNA quantities. However, the number of reads was not predictive of overall qPCR score (number of positive qPCR replicates). In addition to S. edwardsii, metabarcoding led to the detection of a vast community of over 2600 invertebrate taxa. We underscore the necessity for conducting similar analyses across environments and target species, as the ecology of eDNA will vary on a per‐study basis. Our results suggest that eDNA metabarcoding provides a highly sensitive and accurate method for detecting parasitic gill lice while also illuminating the broader biological community and co‐occurrence of species in the environment.

Abstract

Background: COVID-19 is primarily known as a respiratory illness; however, many patients present to hospital without respiratory symptoms. The association between non-respiratory presentations of COVID-19 and outcomes remains unclear. We investigated risk factors and clinical outcomes in patients with no respiratory symptoms (NRS) and respiratory symptoms (RS) at hospital admission. Methods: This study describes clinical features, physiological parameters, and outcomes of hospitalised COVID-19 patients, stratified by the presence or absence of respiratory symptoms at hospital admission. RS patients had one or more of: cough, shortness of breath, sore throat, runny nose or wheezing; while NRS patients did not. Results: Of 178,640 patients in the study, 86.4 % presented with RS, while 13.6 % had NRS. NRS patients were older (median age: NRS: 74 vs RS: 65) and less likely to be admitted to the ICU (NRS: 36.7 % vs RS: 37.5 %). NRS patients had a higher crude in-hospital case-fatality ratio (NRS 41.1 % vs. RS 32.0 %), but a lower risk of death after adjusting for confounders (HR 0.88 [0.83–0.93]). Conclusion: Approximately one in seven COVID-19 patients presented at hospital admission without respiratory symptoms. These patients were older, had lower ICU admission rates, and had a lower risk of in-hospital mortality after adjusting for confounders.

Abstract

This article is about the Squamish Ocean Canoe Family and is based on stories shared by the Skwxwú7mesh Chiaxten ("Protocol Keeper"), Wes Nahanee, and the President of the Squamish Ocean Canoe Family, Larry (Shucks) Nahanee. The article tells the story of a revival of the Skwxwú7mesh ocean-going canoe and traditions, particularly through the annual Tribal Canoe Journeys. Through the stories, reflections, and teachings shared by Wes and Shucks, this paper looks at the "canoe as a teacher" and how Indigenous Pedagogies are attained through Canoe Journeys, particularly in the urban Indigenous context where this canoe resurgence means cultural continuity and healing.
