The Gaga chicken is an indigenous Indonesian breed that is important to preserve using semen cryopreservation technology. The study was conducted to determine the effect of adding sorbitol and butylated hydroxytoluene (BHT) in the diluent on the structural and surface ultrastructure of cryopreserved Gaga chicken sperm during cryopreservation /frozen storage. The study aimed to assess how adding sorbitol and butylated hydroxytoluene (BHT) to the diluent affects the structure and surface ultrastructure of cryopreserved Gaga chicken sperm. A completely randomized design was employed with four treatments and 10 replications including egg yolk-lactate ringer diluent (EYLR) as the control group, EYLR diluent with 3 mM BHT, EYLR diluent with 2% sorbitol, and EYLR diluent with both 3 mM BHT and 2% sorbitol. Semen was collected using a massage technique from 4 male chickens aged approximately 10 months, pooled semen was diluted, packaged in 0.25 mL straws, equilibrated for 2 hours at 5 °C, pre-freeze for 10 minutes, frozen for 24 hours, and thawed for 30 seconds at 37 °C. The parameters evaluated were sperm plasma membrane integrity, acrosome integrity, DNA damage, mitochondrial functionality, and surface ultrastructure. The results showed that the treatment had a significant effect on plasma membrane integrity and post-thawing mitochondrial functionality compared to the control, but no effect was observed on acrosome integrity or DNA damage. The results showed that the combination treatment of BHT with sorbitol had a significant effect on plasma membrane integrity and post-thawing mitochondrial function, but did not affect acrosome integrity or DNA damage when compared to the control group. Ultrastructural observations indicated that cryopreservation caused damage to the head, middle, and tail of the sperm in the control groups. However, these changes were prevented by the diluent containing a combination of BHT and sorbitol. The addition of both components (BHT 3 mM + sorbitol 2%) effectively maintained plasma membrane integrity, mitochondrial functionality, and surface ultrastructure of Gaga chicken sperm during cryopreservation. Ninetinae is a group of small to tiny short-legged spiders largely restricted to arid habitats. Among daddy-long-legs spiders (Pholcidae) this is by far the least diverse subfamily but this may partly be a result of inadequate collecting, poor representation in collections or scientific neglect. We build on a large recent collection of the ninetine genus Papiamenta Huber, 2000 from the Leeward Antilles and use cytochrome oxidase 1 (COI) sequences, extensive scanning electron microscopy data, transmission electron microscopy data and karyotyping to analyse this geographically isolated and poorly known island genus. COI sequences support the split between the two morphologically distinct species on Curaçao but genetic distances between these are surprisingly low (7.4–9.8%; mean 8.6%). The type species P. levii (Gertsch, 1982) may include more than one species but COI and morphology suggest conflicting clade limits. A third species, P. bonay Huber sp. nov. is newly described from Bonaire. Our data on sperm ultrastructure and karyology are puzzling as these suggest different phylogenetic affinities of Papiamenta to other genera. Males transfer sperm as individual sperm (cleistosperm), agreeing with the putative closest relatives as suggested by molecular data, the North American genera Pholcophora and Tolteca. The sex chromosome system (X 1 X 2 X 3 Y) of P. levii , however, is as in the South American Ninetinae genera Gertschiola and Nerudia but different from the putative closest relatives. Vermetid worm-snails are sessile and irregularly coiled marine mollusks common in warmer nearshore and coral reef environments that are subject to high predation pressures by fish. Often cryptic, some have evolved sturdy shells or long columellar muscles allowing quick withdrawal into better protected parts of the shell tube, and most have variously developed opercula that protect and seal the shell aperture trapdoorlike. Members of Thylacodes (previously: Serpulorbis) lack such opercular protection. Its species often show polychromatic head-foot coloration, and some have aposematic coloration likely directed at fish predators. A new polychromatic species, Thylacodes bermudensis n. sp., is described from Bermuda and compared morphologically and by DNA barcode markers to the likewise polychromatic western Atlantic species T. decussatus (Gmelin, 1791). Operculum loss, previously assumed to be an autapomorphy of Thylacodes, is shown to have occurred convergently in a second clade of the family, for which a new genus Cayo n. gen. and four new western Atlantic species are introduced: C. margarita n. sp. (type species; with type locality in the Florida Keys), C. galbinus n. sp., C. refulgens n. sp., and C. brunneimaculatus n. sp. (the last three with type locality in the Belizean reef) (all new taxa authored by Bieler, Collins, Golding & Rawlings). Cayo n. gen. differs from Thylacodes in morphology (e.g., a protoconch that is wider than tall), behavior (including deep shell entrenchment into the substratum), reproductive biology (fewer egg capsules and eggs per female; an obliquely attached egg capsule stalk), and in some species, a luminous, ''neon-like'', head-foot coloration. Comparative investigation of the eusperm and parasperm ultrastructure also revealed differences, with a laterally flattened eusperm acrosome observed in two species of Cayo n. gen. and a spiral keel on the eusperm nucleus in one, the latter feature currently unique within the family. A molecular phylogenetic analysis based on mitochondrial and nuclear rRNA gene sequences (12SrRNA, trnV, 16SrRNA, 28SrRNA) strongly supports the independent evolution of the two non-operculate lineages of vermetids. Thylacodes forms a sister grouping to a clade comprising Petaloconchus, Eualetes, and Cupolaconcha, whereas Cayo n. gen is strongly allied with the small-operculate species Vermetus triquetrus and V. bieleri. COI barcode markers provide support for the specieslevel status of the new taxa. Aspects of predator avoidance/deterrence are discussed for these non-operculate vermetids, which appear to involve warning coloration, aggressive behavior when approached by fish, and deployment of mucous feeding nets that have been shown, for one vermetid in a prior study, to contain bioactive metabolites avoided by fish. Three sexually mature goshawks reared in captivity and imprinted on humans to express reproductive behavior according to the cooperative method were studied for three consecutive breeding seasons to assess the quality of their sperm. The following parameters were analyzed: ejaculate volume and sperm concentration, motility, viability, and morphology. Ejaculate volume, sperm concentration and motility fluctuated along the reproductive season, revealing the greatest quality of the reproductive material at full springtime (i.e., April). Motility of the sperm collected in March strongly reduced with age, contrary to samples collected in April or May. Sperm viability was not influenced by either age or month of collection within each season. Ultrastructural investigations provided information on normal sperm morphology for the first time in this species. The morphological categories of sperm defects in fresh semen, present at low percentages, are also described. Functional analyses (perivitelline membrane assay and artificial inseminations) confirmed the good quality of the semen obtained using the cooperative method. The reported data provide the basis for further studies aimed at developing protocols to improve the outcome of artificial insemination and semen cryopreservation in the goshawk as well as other bird of prey species. To date, little attention has been paid to identifying the effects of long-term cryopreservation on sperm quality for Piaractus orinoquensis. The object of this study was therefore to evaluate the effect of long-term cryopreservation (24 h, 1, 6 and 12 months) on sperm motility, viability, DNA integrity, ATP content, total antioxidant capacity (TAC), morphology and sperm ultrastructure in this species. Milt samples from six males were cryopreserved in a medium containing final concentrations of 7.5% Me2SO, 4.1% glucose and 9.0% egg yolk. The samples were frozen in liquid nitrogen (LN) vapor and stored in LN for periods of 24 h and 1, 6 and 12 months. After thawing, both the motility rate and the viability decreased significantly compared with fresh sperm; however, these parameters did not differ among the four cryopreservation times. The DNA integrity and ATP content decreased significantly after 6 months of cryopreservation. There were no significant differences in TAC values between fresh and cryopreserved sperm. The total sperm abnormalities in cryopreserved samples were about 5-fold higher than in fresh sperm; short tail was the most common defect occurring after cryostorage. The ultrastructural analysis reveals that P. orinoquensis spermatozoa consist of an ovoid head without acrosome, a cylindrical mid-piece, and a single flagellum. The nuclear fossa is located at the base of the nucleus and contains the centriolar complex. There are 1–2 ring-shaped mitochondria located in the mid-piece. The flagellum shows a 9 + 2 organization of microtubules in the axoneme. Post-thaw spermatozoa presented damage such as swelling and rupture of the plasma membrane, mitochondrial damage, loss of the electron-dense chromatin of the nucleus, and degeneration in the middle region.

The objective of this study was to assess testicular morphology and spermatozoal structure spatially within the reproductive tract and temporally among seasons in the sauger (Sander canadensis). The testis exists as two separate lobes joined at the urogenital pore and were characterised as unrestricted lobular with seminiferous tubules terminating at the ventral periphery and coalescing dorsally on the main sperm duct. Differences were observed between the pre‐breeding season (November) and breeding season (March), with every stage of spermatogenesis occurring in spermatocysts in pre‐breeding season in contrast to only spermatozoa being present in the tubules and main duct during the breeding season. Longitudinal folds in the main duct epithelium increased in number with increasing proximity to the urogenital pore, greatly increasing epithelial height regardless of season. Sauger spermatozoa consisted of an ovoid head, a midpiece containing 2 – 4 mitochondria incorporated into the head and a single flagellum containing an asymmetrical lateral ribbon. Motile spermatozoa were found throughout the testis during the breeding season. A decrease in sperm concentration was quantified moving proximally, suggesting a hydration effect by the main duct epithelium during the breeding season. Would the use of genome-wide genotyping be an advantageous strategy to identify the molecular aetiology of two brothers from a non-consanguineous family, clinically diagnosed with total globozoospermia? Two related Spanish globozoospermic patients were studied. Eight first- and second-degree family members were also included in the study. The clinical procedure included anamnesis, physical examination and semen analyses. Acrosome visualization was performed by fluorescein isothiocyanate–Pisum sativum agglutinin labelling and ultrastructural electron microscope sperm analysis. Sperm DNA fragmentation was determined by TUNEL and SCD. Molecular analysis included: the detection of deletion of the DPY19L2 gene by a BPa (break point "a") gap-polymerase chain reaction, and genotyping by using a high-throughput genome-wide genotyping platform and a genotype imputation strategy. The biological characteristics of the two globozoospermic siblings included round-headed spermatozoa without an acrosome; ultrastructural defects in spermatozoa; increased sperm fragmentation and aneuploidies, inability of spermatozoa to activate oocytes (correctable with artificial activation) and good developmental potential of embryos generated by IVF/intracytoplasmic sperm injection. This genetic study focused on a genome-wide compound heterozygote analysis that identified two deleterious rare coding variants in the DPY19L2 gene [rs771726551 (c.431T>A exon 3) and rs147579680 (c.869G>A exon 8)]. A genome-wide compound heterozygote analysis strategy should be considered for molecular screening in globozoospermia and other rare congenital diseases, particularly in cases from non-consanguineous families. The ultrastructure of sperm morphology and some aspects of acrosomal complex development were examined in Kulikovia alborostrata , K. torquata , Kulikovia sp., and Micrura bella using transmission and scanning electron microscopy. The studied Kulikovia species have spermatozoa with a bullet-shaped head consisting of an acrosomal complex with a single cup-like acrosomal vesicle, a cone-shaped nucleus, and a short midpiece that are characteristic of the ect-aquasperm type. Despite the general similarity in spermatozoon organization, an ultrastructural analysis has revealed some species-specific size differences. M. bella is distinguished by an elongated curved sperm head with an acrosomal complex containing one large electron-dense and a few small, moderately electron-dense acrosomal vesicles. As a rule, five mitochondria are located in the midpiece in all the studied species. According to data published in literature and the present findings, all Pilidiophora have spermatozoa with several (three, four or five) mitochondria, and they never contain a single mitochondrion. We suggest this trait to be a synapomorphy for the class Pilidiophora, while the posterior acrosomal ring component, an annular structure formed by the basal part of the acrosomal vesicle, to be a synapomorphy for Heteronemertea. Acrosomal vesicles develop from numerous proacrosomal vesicles that are synthesized in spermatocytes I and fuse (or partly fuse in M. bella) at the spermatid stage. The aim of this study was to describe sperm ultrastructure in Segmentinini F.C. Baker, 1945, a tribe of freshwater panpulmonates (Hygrophila), and identify sperm characters that may qualify as candidate synapomorphies of the group. Spermatozoa of seven species representing six genera of Segmentinini were examined by light microscopy and scanning (SEM) and transmission electron microscopy. Sperm ultrastructure is very similar in all species and is generally typical for the Hygrophila. The head consists of a nucleus and conical acrosomal pedestal tipped with an ovoid apical vesicle and is followed by a long midpiece filled with the mitochondrial derivative. The boundary between the midpiece and the next region, the glycogen piece, is marked by a two-partite cylindrical annulus. The glycogen piece is filled with homogeneous mass of glycogen and is followed by a short terminal region, the endpiece. The nucleus has an unusual shape consisting of a barrel-shaped basal region and a narrow apical stem bearing a disc-shaped nuclear keel. The axoneme arises from the centriolar derivative attached to the nucleus with nine wedge-shaped rods. The mitochondrial derivative contains two glycogen-filled helices that spiral apically in opposition to each other, but then become closely apposed and run in parallel. The comparison with other Hygrophila shows that at least three sperm characters (unusual nuclear shape, accessory centriolar rods and arrangement of glycogen helices) can represent potential synapomorphies of the tribe. Study Question: Are ICSI outcomes impaired in cases of severe asthenozoospermia with multiple morphological abnormalities of the flagellum (MMAF phenotype)?Summary Answer: Despite occasional technical difficulties, ICSI outcomes for couples with MMAF do not differ from those of other couples requiring ICSI, irrespective of the genetic defect.What Is Known Already: Severe asthenozoospermia, especially when associated with the MMAF phenotype, results in male infertility. Recent findings have confirmed that a genetic aetiology is frequently responsible for this phenotype. In such situations, pregnancies can be achieved using ICSI. However, few studies to date have provided detailed analyses regarding the flagellar ultrastructural defects underlying this phenotype, its genetic aetiologies, and the results of ICSI in such cases of male infertility.Study Design, Size, Duration: We performed a retrospective study of 25 infertile men exhibiting severe asthenozoospermia associated with the MMAF phenotype identified through standard semen analysis. They were recruited at an academic centre for assisted reproduction in Paris (France) between 2009 and 2017. Transmission electron microscopy (TEM) and whole exome sequencing (WES) were performed in order to determine the sperm ultrastructural phenotype and the causal mutations, respectively. Finally 20 couples with MMAF were treated by assisted reproductive technologies based on ICSI.Participants/materials, Setting, Methods: Patients with MMAF were recruited based on reduced sperm progressive motility and increased frequencies of absent, short, coiled or irregular flagella compared with those in sperm from fertile control men. A quantitative analysis of the several ultrastructural defects was performed for the MMAF patients and for fertile men. The ICSI results obtained for 20 couples with MMAF were compared to those of 378 men with oligoasthenoteratozoospermia but no MMAF as an ICSI control group.Main Results and the Role Of Chance: TEM analysis and categorisation of the flagellar anomalies found in these patients provided important information regarding the structural defects underlying asthenozoospermia and sperm tail abnormalities. In particular, the absence of the central pair of axonemal microtubules was the predominant anomaly observed more frequently than in control sperm (P < 0.01). Exome sequencing, performed for 24 of the 25 patients, identified homozygous or compound heterozygous pathogenic mutations in CFAP43, CFAP44, CFAP69, DNAH1, DNAH8, AK7, TTC29 and MAATS1 in 13 patients (54.2%) (11 affecting MMAF genes and 2 affecting primary ciliary dyskinesia (PCD)-associated genes). A total of 40 ICSI cycles were undertaken for 20 MMAF couples, including 13 cycles (for 5 couples) where a hypo-osmotic swelling (HOS) test was required due to absolute asthenozoospermia. The fertilisation rate was not statistically different between the MMAF (65.7%) and the non-MMAF (66.0%) couples and it did not differ according to the genotype or the flagellar phenotype of the subjects or use of the HOS test. The clinical pregnancy rate per embryo transfer did not differ significantly between the MMAF (23.3%) and the non-MMAF (37.1%) groups. To date, 7 of the 20 MMAF couples have achieved a live birth from the ICSI attempts, with 11 babies born without any birth defects.Limitations, Reasons For Caution: The ICSI procedure outcomes were assessed retrospectively on a small number of affected subjects and should be confirmed on a larger cohort. Moreover, TEM analysis could not be performed for all patients due to low sperm concentrations, and WES results are not yet available for all of the included men.Wider Implications Of the Findings: An early and extensive phenotypic and genetic investigation should be considered for all men requiring ICSI for severe asthenozoospermia. The study objective is to microscopically evaluate the morphology of sperm in patients with varicocele after surgical treatment and therapy with Prostatilen® AC.Materials and methods. The treatment group included 20 males between 26 and 45 years of age (mean age 31.6 ± 6.1 years) with various stages of varicocele and fertility problems; among them, 10 patients with subclinical stages of varicocele received conservative treatment with Prostatilen® AC (group 1) and were examined before and after the treatment; 10 males with infertility caused in part by varicocele of the spermatic cord veins were examined 6 months to 3 years after surgical treatment (group 2). Standard clinical and lab tests, sperm analysis, electron microscopy of the ejaculate were performed. The control group included 65 fertile males whose sperm samples were obtained from a bank of reproductive cells and tissues and used for comparison in microscopic examination.Results. In patients who received conservative treatment the number of sperm with immature chromatin decreased (p = 0.045) compared to the control group. This characteristic differed in patients after varicocelectomy and patients after conservative treatment (p = 0.037). Compared to control, the number of sperm with excess residual cytoplasm in the head and neck was higher in patients after varicocelectomy (p = 0.011). After conservative treatment, the number of sperm with excess residual cytoplasm was close to the control number and lower than in patients after varicocelectomy (р = 0.028).Conclusion. In patients with subclinical varicocele, conservative treatment with Prostatilen® AC leads to significant improvement in sperm ultrastructure compared to patients who underwent surgery to treat this pathology.

The testis morphology, spermatogenesis, and the spermatozoon structure in two palaeonemerteans, Callinera sp. and Parahubrechtia sp., have been examined by light, laser confocal, and electron microscopy. Early spermatogenesis is quite similar between the two species. The similarities include the formation of the initial proacrosomal vesicle at the primary spermatocyte stage, the presence of electron-dense non-membranous substance around the centrioles in spermatogonia and spermatocytes, and the occurrence of lipid droplets at those stages. However, some differences in the process of acrosome formation during spermiogenesis were also found. In Callinera sp., a spherical acrosomal vesicle (rounded in the sections) transforms into a convex disk structure (arc-shaped in the longitudinal sections) in the developing posterior part of the spermatid and then migrates anteriorly; in Parahubrechtia sp., a rounded acrosomal vesicle migrates towards the apex of the cell where it is transformed into the arc-shaped structure. Both species have spermatozoa of a primitive type, i.e., with a small acrosomal complex, a rounded nucleus, a short mid-piece, and a posteriorly oriented flagellum. Nevertheless, some modifications in the sperm organization such as the asymmetric location of the acrosomal complex and mid-piece were recorded. It is possible that such modifications are synapomorphic for palaeonemerteans. In Callinera sp., the mid-piece contains 2–4 mitochondria, while Parahubrechtia sp. has only one ring-shaped but unclosed mitochondrion. Studying biology of sperm provides valuable information to optimize artificial reproduction and is crucial for sustainable aquaculture. Here, we investigated morphology of spermatozoon in Atlantic cod (Gadus morhua) using transmission and scanning electron microscopy. Furthermore, spermatozoa motility kinetics at different osmolalities were studied using computer-assisted sperm analysis software. The spermatozoon lacked an acrosome and consisted of a head, midpiece, and flagellum. The head of spermatozoa was round, oval, and rather elongated in shape, showing high variations in dimensions. There were up to 6 mitochondria that encircled the proximal part of the flagellum. The proximal and distal centrioles were located within the nuclear notch and arranged orthogonal to each other. The axoneme had a typical 9 + 2 microtubule structure. The flagellar length of spermatozoon was 66.94 ± 0.46 μm. Spermatozoa were immotile in the seminal plasma. Dilution of sperm with natural seawater (1100 mOsmol/kg) resulted in initiation of motility for 91.0 ± 3.4% of spermatozoa with average velocity of 86.2 ± 2.3 μm/s and beating frequency of 52 Hz. The duration of spermatozoa motility was > 6 min; however, the percentage of motile spermatozoa decreased at 60 s post-activation. When osmolality of natural seawater was modified using distilled water or NaCl, spermatozoa motility was not initiated at ≤ 400 and ≥ 2500 mOsmol/kg, and the highest percentage of motility was observed at 730–1580 mOsmol/kg. In a sucrose solution, spermatozoa motility was initiated and suppressed at 600 and 1500 mOsmol/kg, respectively, and highest percentage of motility was observed at 800–1100 mOsmol/kg. Spermatozoon morphology comparisons within Gadiformes showed differences in dimensions of head and mitochondria, flagellar length, and number of mitochondria. The present study provides valuable data that can be used for phylogenetic implications based on spermatozoon morphology and for development of artificial fertilization and sperm cryopreservation protocols based on sperm motility. The Splinter bean, Entada abyssinica, is widely used in folk medicine. In the current work, we profiled the secondary metabolites from E. abyssinica bark extract using LC-MS and investigated its effect on cryopreserved ram semen. Twenty-eight compounds, including tannins and gallic acid derivatives that prevailed in the extract, were tentatively identified. Results showed that the quality of the post-thawed semen showed a significant improvement when the extract was added to the extender at a concentration of 375 μg/mL. The progressive motility and plasma membrane integrity of sperm cells were significantly increased in the post-thawed semen; however, the total antioxidant capacity (TAC) was insignificantly increased. A significant decrease in the concentration of hydrogen peroxide was detected as well. No significant changes were observed in activities of lactate dehydrogenase (LDH), alanine aminotransaminase (ALT), and aspartate transaminase (AST) within the treated samples. Intact sperm percentage was significantly increased, while apoptotic and necrotic sperm percentages were reduced significantly. Molecular docking of some individual components from the extract revealed their potential to interfere with the apoptosis cascade in which Bcl-2 is involved. In conclusion, Entada abyssinica appears to be useful for cryopreservation presumably owing to its polyphenol content that has potent antioxidant capacity scavenging reactive oxygen species (ROS), enhancing the endogenous antioxidant system and inhibiting lipid peroxidation.

Reproduction in snakes employs a wide range of mechanisms such as long-term sperm storage, mating aggregations and male–male combat. These varied mechanisms can be better understood when combined with the study of the ultrastructural morphological variability of spermatozoa. Within the coral snakes, genus Micrurus , there are differences in the reproductive strategies adopted between monads (BRM) and triads (BRT); the aim of this work was to relate these two strategies to morphological differences among spermatozoa within different species. The semen of six Micrurus was collected using a non-invasive technique, fixed and processed for scanning and transmission electron microscopy. Micrurus spermatozoa have the same ultrastructure as those of other snake species, comprising the head, divided into acrosome, nucleus and neck; the midpiece; and the two parts of the tail, the principal piece and the end piece. Micrurus corallinus and Micrurus surinamensis presented evident multilaminar membranes occurring along all the pieces, while Micrurus frontalis and Micrurus altirostris showed cytoplasmic droplets occurring mainly in the midpiece. The differences found in spermatozoa morphology may be related with sperm storage in these four coral snakes, since the development of multilaminar membranes in the midpiece and the maintenance of cytoplasmic droplets in the mature sperm are both features related to extra energy provision for the spermatozoa while stored. Due to the global decrease in jaguar population, conservation strategies are essential and the development of effective semen cryopreservation protocols would contribute to the formation of germplasm banks. Therefore, the objectives were to (1) evaluate the use of TRIS and ACP-117c extenders for jaguar semen freezing, (2) describe the ultrastructural changes in sperm after cryopreservation, and (3) evaluate the binding capacity of the thawed sperm. Eight ejaculates from five mature individuals were collected by electroejaculation, extended in TRIS or a coconut based-extender (ACP-117c), and frozen in liquid nitrogen. Samples were evaluated for sperm motility, vigor, membrane functionality, mitochondrial activity, morphology (using light microscopy, scanning electron microscopy - SEM and transmission electron microscopy - TEM), sperm kinetic parameters (by computerized analysis - CASA), and sperm binding capability using an egg yolk perivitelline membrane assay. Samples preserved in TRIS presented better post-thaw motility (46.0 ± 7.7%) and membrane functionality (60.5 ± 4.2%) and higher mitochondrial activity (21.5 ± 3.7%) than those preserved in ACP-117c (20.9 ± 5.4% motile sperm; 47.1 ± 2.5% functional membrane; 11.8 ± 1.7% mitochondrial activity). Regarding ultrastructural evaluations, SEM showed that both extenders were able to preserve the superficial membrane of the sperm, but TEM revealed the occurrence of nuclear electron lucent points, especially in samples extended in ACP-117c. Additionally, TRIS also provided a higher number of sperm bound to the perivitelline membrane (29.5 ± 3.3%) in comparison to samples diluted in ACP-117c (18.6 ± 1.5%). This study evaluated the efficacy of Turraea fischeri leaf extract for maintaining the viability of cryopreserved goat sperm. Ejaculated semen was collected from 5 mature Baladi bucks (50–60 kg, 2–4 years of age) and those samples with mass motility ≥ 70% and sperm concentration ≥ 2.5 × 109/mL were selected, pooled, and divided into 4 aliquots. Each aliquot was diluted in Tris-citric-soybean lecithin extender containing a different concentration of T. fischeri leaf extract (0, 125, 250, or 375 µg/mL). Treated semen samples were cooled to 5 °C, transferred to 0.25-mL French straws, and stored in liquid nitrogen (LN2) at −196 °C. After thawing, membrane integrity was examined by transmission electron microscopy, apoptotic activity by Annexin/propidium iodide staining and flow cytometry, and both enzyme activities and antioxidant capacity by spectroscopic assays. The leaf extract at 375 µg/mL significantly improved semen quality as indicated by enhanced total antioxidant capacity, reduced H2O2 concentration, a greater proportion of structurally intact motile sperm, and concomitant reductions in apoptosis and necrosis. The extract also significantly increased the proportion of sperm with a contiguous plasma membrane and intact acrosome (p < 0.05). Furthermore, LC-MS revealed numerous secondary metabolites in the extract that may contribute to sperm cryopreservation.

The male reproductive system, sperm structure, and spermatogenesis of Trypophloeusklimeschi (Coleoptera: Curculionidae: Scolytinae), which is one of the most destructive pests of Populus alba var. pyramidalis (Bunge), were investigated using light microscopy, scanning electron microscopy, and transmission electron microscopy. The male reproductive system of T.klimeschi is composed of testes, seminal vesicles, tubular accessory glands, multilobulated accessory glands, vasa deferentia, and a common ejaculatory duct. In spermatogenesis, two phenomena are apparent: The nuclear chromatin condenses into two different patterns, and an oval preacrosomal vesicle is present at the flank of the Golgi apparatus. The sperm are short, measuring 76.7 ± 1.8 μm in length, and are 508.1 ± 12.9 nm in width. The sperm are composed of a three-layer acrosomal complex, a cylindrical nucleus, two mitochondrial derivatives, a 9 + 9 + 2 axoneme, and two accessory bodies with a large “puff”-like expansion. Mature sperm are individually stored in seminal vesicles. During spermiogenesis, the similarities in the nuclear chromatin condensation characteristics of Curculioninae and Scolytinae are indicative of their close phylogenetic relationship. It appears that the preacrosomal vesicle being flanked by the Golgi apparatus is a characteristic of spermatogenesis in Curculionidae.

This study aims to analyze the functional anatomy of the male reproductive system in Neocaridina davidi, a very popular ornamental species of caridean shrimp. Mature males were cold‐anaesthetized and their reproductive systems were dissected for histological and histochemical analysis, while the spermatozoa and spermatophore wall ultrastructure were analyzed under transmission electron microscopy. The male reproductive system consisted of two coiled testes, which were continuous with the vasa deferentia. Testes were positioned on the dorsal side of the cephalothorax above the hepatopancreas, and comprised seminiferous tubules where spermatogenesis occurred. Each vas deferens (VD) was a long tube dorsolaterally positioned with respect to the hepatopancreas, and increased in diameter at the distal end. Three regions could be recognized in the VD: proximal, middle, and distal. The proximal region had a cylindrical epithelium with secretory cells. The middle region (or typhlosole) had a dorsal fold (or typhlosole) with a thick columnar epithelium and high secretory activity. The spermatophore was a continuous cord with three acellular layers, which were mainly characterized by the presence of neutral glycoconjugates and proteins. The sperm morphology was distinct from the inverted cup‐shaped spermatozoa observed in the majority of caridean shrimps. The spermatozoa in specimens of N. davidi were spherical in shape, with a cross‐striated, single, short spike, and arranged in clusters of three or four sperm cells. Abstract The ultrastructure of the complex organisation of the spermatozoa in Harmonia axyridis and Adalia decempunctata (Coccinellidae) was studied, with particular emphasis on the origin of the anterior shifting of the axonemal structure, which becomes parallel to the nucleus in the sperm flagellum. In studying the spermiogenesis, a centriolar remodelling was observed with the long centriole, present in the early spermatids, transformed in the spermatozoa into an exceptionally long and narrowed basal body (about 0.16 × 3.5–4.0 μm long) displaying a 9 + 0 microtubular pattern in the proximal part and a 9 + 2 pattern in the following part; this is a characteristic not observed in any other pterygotan insect. The sperm also have a very long acrosome surrounded by a dense layer of material extending along the whole basal body. These two uncommon features were discussed in the light of sperm movement. Vermetid worm-snails are sessile and irregularly coiled marine mollusks common in warmer nearshore and coral reef environments that are subject to high predation pressures by fish. Often cryptic, some have evolved sturdy shells or long columellar muscles allowing quick withdrawal into better protected parts of the shell tube, and most have variously developed opercula that protect and seal the shell aperture trapdoor-like. Members of Thylacodes (previously: Serpulorbis ) lack such opercular protection. Its species often show polychromatic head-foot coloration, and some have aposematic coloration likely directed at fish predators. A new polychromatic species, Thylacodes bermudensis n. sp., is described from Bermuda and compared morphologically and by DNA barcode markers to the likewise polychromatic western Atlantic species T. decussatus (Gmelin, 1791). Operculum loss, previously assumed to be an autapomorphy of Thylacodes , is shown to have occurred convergently in a second clade of the family, for which a new genus Cayo n. gen. and four new western Atlantic species are introduced: C. margarita n. sp. (type species; with type locality in the Florida Keys), C. galbinus n. sp., C. refulgens n. sp., and C. brunneimaculatus n. sp. (the last three with type locality in the Belizean reef) (all new taxa authored by Bieler, Collins, Golding & Rawlings). Cayo n. gen. differs from Thylacodes in morphology (e.g., a protoconch that is wider than tall), behavior (including deep shell entrenchment into the substratum), reproductive biology (fewer egg capsules and eggs per female; an obliquely attached egg capsule stalk), and in some species, a luminous, "neon-like", head-foot coloration. Comparative investigation of the eusperm and parasperm ultrastructure also revealed differences, with a laterally flattened eusperm acrosome observed in two species of Cayo n. gen. and a spiral keel on the eusperm nucleus in one, the latter feature currently unique within the family. A molecular phylogenetic analysis based on mitochondrial and nuclear rRNA gene sequences (12SrRNA, trnV, 16SrRNA, 28SrRNA) strongly supports the independent evolution of the two non-operculate lineages of vermetids. Thylacodes forms a sister grouping to a clade comprising Petaloconchus , Eualetes , and Cupolaconcha, whereas Cayo n. gen is strongly allied with the small-operculate species Vermetus triquetrus and V. bieleri . COI barcode markers provide support for the species-level status of the new taxa. Aspects of predator avoidance/deterrence are discussed for these non-operculate vermetids, which appear to involve warning coloration, aggressive behavior when approached by fish, and deployment of mucous feeding nets that have been shown, for one vermetid in a prior study, to contain bioactive metabolites avoided by fish. The traditional study of spermogram readings is a main method for examination of male infertility. However, the spermogram readings may be considerably covered in the group of fertile and infertile men and fail to give a full insight into sperm fertilizing ability. In recent years, tests determining the functional properties of sperm have been intensively developed. This review considers a quantitative electron microscopic technique of sperm (EMTS), which assesses the structure and function of the sperm penetration and motor apparatus. The detection of sperm chromatin structural disorders can understand the causes of early embryonic malformation. Indications for EMTS and interpretation of its results are given.

The corn leafhopper, Dalbulus maidis , which is one of the most important vectors of maize diseases, had the morphology of its spermatozoa described more than 40 years ago. However, the description is imprecise, requiring certain rectifications. Hence, we re-describe the sperm morphology of this species using light microscopy and transmission electron microscopy. Their spermatozoa measured from 118.1 to 128.5 μm in length, this value being about three times smaller than what was previously described. The head region is formed by a small acrosome and the nucleus. The acrosome is paracrystalline and has the bifurcated base that is docked in two cavities on one side of the anterior region of the nucleus. The nucleus, which is linear and thin, measured from 19.3 to 22.9 μm in length, and exhibited homogeneously compact chromatin. In cross-section, it has a thumbtack shape, except in the anterior and posterior regions, where it is oval. The flagellum consists of an axoneme with 9 + 9 + 2 microtubules, two similar mitochondrial derivatives, two accessory bodies each associated with a small sub-ellipsoidal structure, and a ‘center-flagellar material’. In all the spermatozoa observed, the final portion of the flagellum was branched into four filaments. Some of these morphological features are now being re-described, while others have been observed for the first time in the Cicadellidae family, as the center-flagellar material. We studied spermatogenesis and spermiogenesis in Adalia decempunctata (L), a beetle of the Coccinellidae family. The spermatocyte exhibits two centrioles which elongate to form a pair of primary cilia. A novel structure, appearing in cross sections as a dense droplet, is observed near the long centriole during spermiogenesis, and is soon accompanied by a procentriole (PCL). PCL structure consists of singlet microtubules, a central tubule and an incomplete cartwheel. The PCL persists until the end of spermiogenesis, when it vanishes together with the dense droplet. The sperm has an exceptionally long basal body and the nucleus is disposed parallel to the flagellar components, a peculiar trait shared by other species of the coccinellid group. The springtail Collembola are characterized by having rolled spermatozoa, with a long cylindrical extracellular structure adhering to the acrosome. This structure is produced by the secretory activity of the testes epithelial cells at almost the end of spermiogenesis. At the beginning of its formation, it is a thin extension with a helical wall and a dense axial region. Later the cylindrical structure shows an inner organization which is different in the several species examined: species of Entomobryidae contain material with a paracrystalline structure, whilst some of Symphypleona contain ovoid structures. The outer envelope of the extracellular structure consists of two overlapped layers orthogonally arranged, clearly identified by cryo-preparations. Immunoblot analysis and lectin stainings have indicated that the cylindrical structure has a glycoproteic composition. As the structure is no longer visible after the sperm transfer into the female spermatheca, it is suggested that it could contain enzymes able to activate the sperm unwinding process and possibly allowing the reacquisition of sperm motility. This study evaluated the semen of two penguin species from separate genera with reference to unique features in sperm structure using light microscopy and transmission electron microscopy. Ejaculates from African penguin (n = 51) and Rockhopper penguin (n = 9) contained on average more than 60% motile spermatozoa and a sperm concentration of 3274 × 10 6 /ml and 1423 × 10 6 /ml, respectively. The percentage progressive motility was similar for the two species as well as all the kinematics parameters. The sperm morphology of these two penguin species is almost identical and largely resembles that of non-passerine birds in terms of the filiform head, small acrosome and mid-piece containing 13 spherical mitochondria, arranged around the proximal and distal centrioles in a single helix. Apart from a shorter mid-piece, penguin sperm morphometrics were similar to other non-passerine birds. The ultrastructure of the sperm principal piece revealed the typical 9 + 2 microtubular arrangement without any outer dense fibres. An unusual feature in both African and Rockhopper penguin spermatozoa was the occurrence of multiple axonemes contained in one plasmalemma in 4% of spermatozoa. These double, triple and quadruple axonemal arrangements have not been described previously albeit multiple tails were reported in other bird species. It is unclear whether such a unique structural feature will be of any advantage for sperm motility and might rather be a result of the absence of sperm competition. We describe the sperm ultrastructure of six penaeid species, including at least one member of each tribe (Penaeini, Parapenaeini and Trachypenaeini). Fragments of the vas deferens of the Penaeidae Farfantepenaeus brasiliensis , Farfantepenaeus paulensis , Litopenaeus schmitti , Parapenaeus americanus , Rimapenaeus constrictus and Xiphopenaeus kroyeri were fixed and processed according to the routine for transmission electron microscopy. The morphological results were contextualized in an evolutionary perspective using molecular markers for the phylogenetic reconstruction of this group. A phylogram was proposed by Bayesian inference based on 1007 bp of 33 sequences of the combined genes (16S rDNA and COI mtDNA) from 27 dendrobranchiate specimens. Our findings show that morphological differences in the sperm ultrastructures of members among the tribes of Penaeidae can be used as a baseline to understand their evolutionary relationships. Individuals from the Penaeini tribe show plesiomorphic characteristics in the sperm ultrastructure compared to the Trachypenaeini tribe from which they were derived, such as shrimp from family Sicyoniidae. The morphological complexity of the sperm of the different penaeid members corroborated with the genetic phylogeny, which showed different clades for each tribe and the close relationship with Sicyoniidae. The sperm features of the selected species studied here reflected their evolutionary history. These features confirm the previous phylogenetic hypothesis and question the monophyly of Penaeidae, which should be verified in the future with a more complete set of representative members of each tribe. The testicular, spermatogenesis and sperm morphology of the backswimmer Martarega bentoi was described using light and transmission electron microscopy. In this species, a pair of testes, two deferent ducts, two different pairs of accessory glands, and an ejaculatory duct form the male reproductive system. Each testis consists of two testicular follicles, which are arranged side by side in snail shape. The follicles are filled with cysts at different stages of spermatogenesis, but in the same cyst the germ cells (up to 64) are in the same stage. At the end of spermatogenesis, the sperm cells are very long, with the flagellum measuring approximately 2500 μm in length, the nucleus only 19 μm, and the acrosome, with two distinct regions, 300 μm. The flagellum is composed of an axoneme, with a 9 + 9 + 2 microtubular pattern, and 2 asymmetric mitochondrial derivatives (MDs). These have the anterior ends inserted into two cavities at the nucleus base, exhibit two paracrystalline inclusions, and have bridges linking them to the axoneme. Few spermatozoa per cyst, asymmetry in size and shape of the MDs, as well as their insertion at the nuclear base are characteristics considered derived, and that differentiate the sperm of M. bentoi from those of the Nepomorpha, Belostomatidae and Nepidae. Spermatogenesis and the spermatozoon of Acanthostomum (Atrophocaecum) aswaninesis, were studied by transmission electron microscopy. Spermatogonia characterized by a large nucleo-cytoplasmic ratio, give rise to two generations of spermatocytes. The secondary spermatocytes divide forming early spermatids that fuse together into a common cytoplasmic mass. Spermiogenesis begins with the formation of the zone of differentiation in front of 2 centrioles associated by an inter-centriolar body. Each centeriole is linked to striated rootlet, and gives rise to a flagellum. It is characterized by an asynchronic flagellar rotation and subsequent proximodistal fusion with a median cytoplasmic process. The spermatozoon is formed after constriction of arched membranes. Mature spermatozoon of A. (A.) aswaninesis is filiform, tapering at both ends and it presents all the characteristic features found in the digenean gametes. The spermatozoon possesses 2 axonemes of the 9+ ‘‘1” pattern, a nucleus, mitochondria, glycogen and two bundles of parallel cortical microtubules. A major feature is the presence of external ornamentation in the anterior part of the mature spermatozoon, and the presence of two mitochondria rather than just (1) as in most of the digenean spermatozoon. (A.) aswaninesis match the general pattern found in the digenea, but certain peculiarities are characteristic and differentiate the sperm of this fluke from that of other digenetic trematodes.

The current taxonomy of most Atelopus species is based on morphological and color data only. Recent studies suggest that A. spumarius may represent a species complex assigned under the same name. Karyotypic data and description of sperm ultrastructure for 13 specimens of A. spumarius are presented here for the first time. A chromosomal analysis revealed 2n = 22 chromosomes, with centromeric heterochromatin in all pairs and a nucleolar organizer region (NOR) on the telomere of pair 7. The sperm was of the bufonoid type, presenting a filiform nucleus covered by an acrosomal complex and a mitochondrial collar in the neck region. The tail was composed of an axoneme, an undulating membrane and an axial rod. A karyotype analysis of A. spumarius showed the same chromosome number and similar chromosomal morphology as described for congeneric species, with slight differences probably resulting from pericentric inversions. The NOR location (on pair 7) was the same as that observed for species belonging to the genus Rhinella. The spermatological findings indicate a close relationship between Atelopus and the bufonoid lineage. The present data are useful for reference in future studies to determine whether more than one species are assigned to A. spumarius.

The sperm ultrastructure and spermatogenesis of the ectoparasitic polychaete Asetocalamyzas laonicola Tzetlin, 1985 (Calamyzidae) is investigated. The male cells are located freely in the coelom. The spermatocytes are large cells of irregular shape; their nuclei have condensed chromatin in the periphery. Spermatocyte cytoplasm is granular and electron-dense with several spherical mitochondria. During early developmental stages spermatids are aggregated into a rosette (four cells). The early spermatids have a tiny acrosomal vesicle at one side of the cell, a few round mitochondria at another, and electron dense nuclei. The late spermatids have elongated mitochondria, a well-developed acrosome and a flagellum. The mature sperm are threadlike with a round acrosomal vesicle, an electron-dense structure. The elongated nuclei have anterior and posterior depressions. The supporting root zone of the acrosome is located behind the acrosomal vesicle in the anterior invagination of the nuclei. Six elongated mitochondria surround the flagellum and form the midpiece of the sperm. A single centriole lies in the posterior depression of the nucleus. The middle part of the flagellum possesses a normal (9+2x2) pattern. Probably, the terminal part of flagellum is modified. The sperm structure suggests internal fertilization or another type of specialized sperm transfer in A. laonicola.

Testes morphology, spermatogenetic process and mature sperm ultrastructure were analysed in Hippocampus guttulatus, using both light and transmission electron microscopy. Both testes were organized in a single large germinal compartment, with a central lumen. Spermatocysts only contained spermatogonia and primary spermatocytes. Inside the testis lumen, together with mature sperm, two types of large mono-nucleate cells, flagellate and aflagellate, were present. Both types of cells were interpreted as developing germ cells precociously released inside the testis lumen, where their maturation was completed. According to the different morphological features of the nuclei, such as chromatin condensation degree, aspect of the nuclear fossa and others, the flagellate cells were unquestionably developing spermatids. On the contrary, the developmental stage of the aflagellate was more difficult to interpreted. They could be secondary spermatocytes or young spermatids. No dimorphic sperm were recognizable, the only sperm type observed have features typical of the intro-sperm reports in other syngnathids species. They had a cylindrical head, a short midpiece, characterized by two mitochondrial rings housed inside a cytoplasmic collar, and a long flagellum. Asthenozoospermia has been related to structural defects of the sperm flagellum. However, few reports have studied in detail the ultrastructure of sperm with total immotility. We present an ultrastructural study of sperm from five patients with total sperm immotility, four due to dysplasia of the fibrous sheath (DFS) and one with situs-inversus. Of the four patients with DFS, three cases presented a hypertrophic and hyperplastic fibrous sheath that invaded the midpiece space, absence of the annulus, and a short midpiece containing a few disorganized and pale mitochondria. Of these cases, two presented absence of the central complex and radial spokes; another additionally presented absence of dynein arms and nexin bridges; and the other patient presented an intact annulus with a dysplastic fibrous sheath restricted to the principal piece with disorganized microtubule doublets. The patient with situs-inversus presented severe respiratory symptoms, with absence of dynein arms and nexin bridges. In conclusion, we present three cases with DFS associated with total sperm immotility, abnormal mitochondria, and absence of the annulus, central pair complex and radial spokes, of which one had in addition absence of dynein arms and nexin bridges. We also describe a patient, with total sperm immotility and a different presentation of DFS, as the annulus was present and the dysplastic fibrous sheath was restricted to the principal piece. These findings thus confirm the heterogeneity of the DFS condition. The changes observed in the patient with situs-inversus also further support previous observations. [ABSTRACT FROM PUBLISHER]

In contrast to numerous studies on spermatozoa length, relatively little work focuses on the width of spermatozoa, and particularly the width of the midpiece and flagellum. In flagellated spermatozoa, the flagellum provides forward thrust while energy may be provided via mitochondria in the midpiece and/or through glycolysis along the flagellum itself. Longer flagella may be able to provide greater thrust but may also require stronger structural features and more or larger mitochondria to supply sufficient energy. Here, we use scanning electron microscopy to investigate the ultrastructure of spermatozoa from 55 passerine species in 26 taxonomic families in the Passerides infraorder. Our data confirm the qualitative observation that the flagellum tapers along its length, and we show that longer flagella are wider at the neck. This pattern is similar to mammals, and likely reflects the need for longer cells to be stronger against shearing forces. We further estimate the volume of the mitochondrial helix and show that it correlates well with midpiece length, supporting the use of midpiece length as a proxy for mitochondrial volume, at least in between-species studies where midpiece length is highly variable. To re-evaluate the relationships of the major bivalve lineages, we amassed detailed morpho-anatomical, ultrastructural and molecular sequence data for a targeted selection of exemplar bivalves spanning the phylogenetic diversity of the class. We included molecular data for 103 bivalve species (up to five markers) and also analysed a subset of taxa with four additional nuclear protein-encoding genes. Novel as well as historically employed morphological characters were explored, and we systematically disassembled widely used descriptors such as gill and stomach 'types'. Phylogenetic analyses, conducted using parsimony direct optimisation and probabilistic methods on static alignments (maximum likelihood and Bayesian inference) of the molecular data, both alone and in combination with morphological characters, offer a robust test of bivalve relationships. A calibrated phylogeny also provided insights into the tempo of bivalve evolution. Finally, an analysis of the informativeness of morphological characters showed that sperm ultrastructure characters are among the best morphological features to diagnose bivalve clades, followed by characters of the shell, including its microstructure. Our study found support for monophyly of most broadly recognised higher bivalve taxa, although support was not uniform for Protobranchia. However, monophyly of the bivalves with protobranchiate gills was the best-supported hypothesis with incremental morphological and/or molecular sequence data. Autobranchia, Pteriomorphia, Heteroconchia, Palaeoheterodonta, Archiheterodonta, Euheterodonta, Anomalodesmata and Imparidentia new clade (= Euheterodonta excluding Anomalodesmata) were recovered across analyses, irrespective of data treatment or analytical framework. Another clade supported by our analyses but not formally recognised in the literature includes Palaeoheterodonta and Archiheterodonta, which emerged under multiple analytical conditions. The origin and diversification of each of these major clades is Cambrian or Ordovician, except for Archiheterodonta, which diverged from Palaeoheterodonta during the Cambrian, but diversified during the Mesozoic. Although the radiation of some lineages was shifted towards the Palaeozoic (Pteriomorphia, Anomalodesmata), or presented a gap between origin and diversification (Archiheterodonta, Unionida), Imparidentia showed steady diversification through the Palaeozoic and Mesozoic. Finally, a classification system with six major monophyletic lineages is proposed to comprise modern Bivalvia: Protobranchia, Pteriomorphia, Palaeoheterodonta, Archiheterodonta, Anomalodesmata and Imparidentia. Sperm ultrastructure in two species of the marine snail family Olividae is examined. Euspermatozoa of both species are composed of a conical, membrane-bound acrosomal vesicle; an axial rod and a basal plate similar in both species; a solid and highly electron-dense nucleus; an elongate midpiece consisting of the axoneme sheathed by helical mitochondrial elements; an elongate glycogen piece; a double electron-dense ring at the junction of the midpiece and glycogen piece; and a free tail region. The slight narrowing in the acrosomal vesicle invagination is situated in different levels between Olivancillaria deshayesiana and Olivancillaria carcellesi. This morphology could be considered as a specific character. The length of the nucleus in O. carcellesi and in O. deshayesiana is shorter than that of other neogastropods, and could be diagnostic at family level. [ABSTRACT FROM PUBLISHER]

Abstract: Spermatogenesis and the spermatozoon of Acanthostomum (Atrophocaecum) aswaninesis, were studied by transmission electron microscopy. Spermatogonia characterized by a large nucleo-cytoplasmic ratio, give rise to two generations of spermatocytes. The secondary spermatocytes divide forming early spermatids that fuse together into a common cytoplasmic mass. Spermiogenesis begins with the formation of the zone of differentiation in front of 2 centrioles associated by an inter-centriolar body. Each centeriole is linked to striated rootlet, and gives rise to a flagellum. It is characterized by an asynchronic flagellar rotation and subsequent proximodistal fusion with a median cytoplasmic process. The spermatozoon is formed after constriction of arched membranes. Mature spermatozoon of A. (A.) aswaninesis is filiform, tapering at both ends and it presents all the characteristic features found in the digenean gametes. The spermatozoon possesses 2 axonemes of the 9+ ‘‘1” pattern, a nucleus, mitochondria, glycogen and two bundles of parallel cortical microtubules. A major feature is the presence of external ornamentation in the anterior part of the mature spermatozoon, and the presence of two mitochondria rather than just (1) as in most of the digenean spermatozoon. The spermiogenesis process and spermatozoon organization of A. (A.) aswaninesis match the general pattern found in the digenea, but certain peculiarities are characteristic and differentiate the sperm of this fluke from that of other digenetic trematodes. Abstract: In the management of asthenozoospermia, the spermogram-spermocytogram plays an important role during diagnosis. It is of major importance to distinguish between necrozoospermia and sperm vitality. An ultrastructural study of spermatozoa is processed in the case of primary infertility without female implication, severe, unexplained and irreversible asthenozoospermia, sperm vitality at least 50 % and normal concentration of spermatozoa. Ultrastructural flagellar abnormalities are numerous and involve most spermatozoa. ICSI provides a suitable solution for patients with sperm flagellar defects to conceive children with their own gametes but the rate of ICSI success may be influenced by the type of flagellar abnormality. Abstract: The structure of the reproductive organs of males and females of four species of Astroblepus were studied by light microscopy and TEM. In females, the gonadal and urinary orifices are separated. The urinary pore opens at the tip of a urinary papilla. In males, the testes conform to the anastomosing tubular type. Spermatogenesis is partially cystic, with most spermatid stages taking place within the lumen of the seminiferous tubules. The sperm ducts pass through seminal vesicles. The main sperm ducts fuse and join the urinary duct to form a urogenital duct that passes through to the tip of an long urogenital papilla. A circularly arranged skeletal muscle at the base of the urogenital papilla probably serves as a sphincter. The spermatozoon is an anacrosomal introsperm with an elongate, conical nucleus and an elongate midpiece. The midpiece contains numerous fused mitochondria that form rings within the cytoplasmic collar, but no vesicles were observed. The flagellum has a classic 9+2 axoneme with the A-tubules of the peripheral doublets being electron-dense. Two lateral fins on the posterior part of the flagellum have electron-dense termini. Nuclear chromatin is highly condensed, and the two centrioles are oriented perpendicularly to one another and are contained within a nuclear fossa. [Copyright &y& Elsevier]
