Abstract Background Bluetongue (BT), a vector‐borne disease of wild and domestic ruminants, is responsible for severe economic losses in flocks. To reduce this impact, a surveillance and control plan was implemented in Tunisia. However, the epidemiological situation of BT remains incompletely understood, especially for the circulating serotypes. Objective The aim of this survey was to determine the seroprevalence, to identify the circulating serotypes and to identify the associated risk factors for bluetongue virus (BTV) circulation in Tunisia using risk‐based sampling (RBS). Methods A total of 3314 blood samples were randomly collected from 67 sectors using risk‐based sampling and screened by competitive enzyme‐linked immunosorbent assays (c‐ELISAs). Out of the 1330 positive samples, 200 samples were analysed by serum neutralization test (SNT) to identify circulating BTV serotypes. Results Of 3314 sera, 1330 were c‐ELISA‐positive (40.1%) for antibodies against the BTV structural protein VP7. The result of SNT showed the presence of BTV‐1, BTV‐2, BTV‐3, BTV‐4 and, for the first time in Tunisia, BTV‐26. The logistic regression model revealed that older animals had nearly two times the odds of being infected with BTV compared to younger animals. Flocks with a history of BT were almost 1.5 times more likely to be at risk for contracting BTV infection. The flock size, housing indoors and intensive production system were significant protective factors. Conclusions High seroprevalence of BTV among sheep was highlighted in Tunisia. The neutralization test showed the presence of the following BTV serotypes: BTV‐1, BTV‐2, BTV‐3, BTV‐4 and, for the first time in Tunisia, BTV‐26. Age, production system and flock size were important variables associated with BTV infection in sheep. This finding is crucial, as it will allow the adjustment of the BT control programme in Tunisia.

Epizootic haemorrhagic disease (EHD) is a Culicoides-borne viral disease caused by the epizootic haemorrhagic disease virus (EHDV) associated with clinical manifestations in domestic and wild ruminants, primarily white-tailed deer (Odocoileus virginianus) and cattle (Bos taurus). In late September 2021, EHDV was reported in cattle farms in central/western Tunisia. It rapidly spread throughout the country with more than 200 confirmed outbreaks. We applied a combination of classical and molecular techniques to characterize the causative virus as a member of the serotype EHDV-8. This is the first evidence of EHDV- 8 circulation since 1982 when the prototype EHDV-8 strain was isolated in Australia. The epidemiological patterns of Bluetongue (BT) in North Africa and Mediterranean Basin (MB) dramatically changed by emergence of subsequent episodes of novel bluetongue virus (BTV) serotypes with highly pathogenic indexes and socio‐economic impacts. The objective of the study was to investigate the sero‐prevalence and serotype distribution of BTV in Libya. During 2015‐2016, a total of 826 serum samples were collected from domestic ruminants in Libya. All sera were assayed by competitive enzyme‐linked immunosorbent assays (c‐ELISA). C‐Elisa‐positive samples (43.3%; 173/400) were further analyzed by virus neutralization assay to identify BTV serotypes and determine the antibody titre of positive samples. An overall BTV sero‐prevalence was 48.4% (95% CI: 45.0%‐51.8%). Neutralizing antibodies were detected against the following BTV serotypes namely: BTV‐1, BTV‐2, BTV‐3, BTV‐4, BTV‐9 and BTV‐26. While BTV‐1, BTV‐2, BTV‐4 and BTV‐9 circulation was unsurprising as they have been responsible of the last year outbreaks in Northern African Countries, the detection of BTV‐3 and BTV‐26 was definitely new and concerning for the animal health of the countries facing the Mediterranean Basin. It is crucial that European and Northern African authorities collaborate in organizing common surveillance programmes to early detect novel strains or emerging serotypes in order to set up proper preventive measures, and, in case, develop specific vaccines and plan coordinated vaccination campaigns. This study is the first investigation on the sero‐prevalence and serotype distribution of BTV in Libya. In line with other Northern African countries, the obtained results (48.4%; 95% CI: 45.0–51.8%) in sheep, goats and cattle confirmed that, at least in the surveyed regions and provinces, BT is endemic in Libya. Abstract: Here we report and characterize a porcine epidemic diarrhea (PED) outbreak which occurred in a swine fattening farm in the province of Teramo, Abruzzi region (central Italy), in January 2016. PED virus (PEDV) identification was determined by real‐time RT‐PCR performed on RNAs purified from fecal samples collected from two symptomatic pigs. Whole genome sequence (PEDV 1842/2016) was also obtained by next generation sequencing straight from RNA purified from one fecal sample. Genome comparison with extant global PEDV strains revealed a high nucleotide identity with recently reported European and American S‐INDEL PEDVs. Efficient sequencing, share of genomic data combined with the implementation of epidemiological tools would be the ideal approach for study and analysis of transboundary infectious diseases as PED. In infants delivered by cesarian section, the immune system could be influenced by anesthetic drugs that may cross the placenta and reach the fetal circulation. Factors that determine placental transfer of anesthetics and their distribution into and ultimate removal from fetal tissue include physiochemical properties of the compound; the anatomic features of the maternal circulation, placenta, and fetus; and the hemodynamic and pharmacokinetic events that occur within them. Because the anesthetic agents can interfere with many neonatal function, the aim of this manuscript is to review the interference of regional or general anesthesia on neonatal immune response. Background. Energetic substrates and hormonal phase are important for uterine contractions. Etomoxir a muscle carnitine palmitoyltransferase-1 inhibitor, able to diverge uterine metabolic pathways towards glycolysis, facilitates glucose utilisation. The aim of this study was to evaluate its effect on uterine contractility in different hormonal situations. Methods. Uterine samples were collected from 60 cows during follicular phase, luteal phase and pregnancy. The cows were slaughtered at a local abattoir. Longitudinal strips were mounted vertically in a 30-ml organ bath connected to an isometric force transducer. Contractions were recorded with an ink-writing polygraph. After the equilibration period, etomoxir was added to the organ bath at different concentrations. The amplitude and frequency of contractions were registered before and after addition of etomoxir. Results. In 17 strips from pregnant cows, etomoxir increased the amplitude ( p < 0.05) of contractions but not the frequency in comparison with basal conditions. In 15 strips from cows in the luteal phase, etomoxir increased the amplitude ( p < 0.05) and frequency of contractions ( p < 0.05 at 5 μμM and p < 0.01 at 8 and 10 μμM). In 18 strips from cows in the follicular phase, etomoxir increased the frequency of contractions but not the amplitude ( p < 0.01 at 5 μμM and p < 0.05 at 8 and 10 μμM). Conclusions. This study confirms the importance of glucose for uterine contractility and, moreover, it underlines different patterns of contraction with regard to the hormonal status. [ABSTRACT FROM AUTHOR]

LH/hCG receptor has been found in extragonadal tissues in human and animals. The myometrium presents such receptors but their functional role is still not clear. Aim of our study was to test the activity of human chorionic gonadotropin (hCG) on bovine uterine contractility. Uterine strips from cows both during follicular and luteal phases were mounted in an organ bath and then exposed to increased doses of hCG. The amplitude of the myometrium contractions were significantly decreased in the follicular and luteal phase but the frequency was not affected. These findings prove a relaxing effect of hCG in the bovine uterus, as already shown in the sow and human, and its possible functional role in modulating uterine contractility. Bluetongue (BT), is one of the OIE-listed major diseases of ruminants. Following the official report of BT virus serotype 3 (BTV-3) in a sheep in Cap Bon (Tunisia), blood and serum samples of ruminants were collected from some areas of Tunisia to further investigate the presence of this virus in the country. A quantitative real time RT-PCR has been first developed for the detection and quantitation of BTV-3 RNA from field specimens. Out of 62 collected blood samples, 23 were shown to be positive for BTV-3 RNA. Isolation on cell cultures was also possible from six samples. Genome sequencing revealed the circulation of two unrelated western strains of BTV-3, one circulating in Cap Bon and neighboring areas, and the other circulating nearby the border with Libya. The presence of a putative novel BTV serotype (BTV-Y TUN2017) in sheep introduced from Libya to Tunisia, genomically related to the BTV strain contaminating a commercially-available sheep pox vaccine and to BTV-26, has been also demonstrated. This finding highlights the pressing need for a prompt production and release of a novel inactivated BTV-3 vaccine to be used in case of emergence or proactively in the areas of Southern Europe at major risk of BTV introduction. The virus neutralization test (VNT) is a functional immunoassay which detects the presence and quantity of neutralizing antibodies. It is a highly sensitive and specific test. As with most neutralization assays, the EHDV VNT does not react with all virus-targeting antibodies, but specifically with those antibodies that bind to VP2, the outermost capsid structural protein of the virus. The interaction between VP2 and neutralizing antibodies can block EHDV cell binding, neutralizing its infectivity. The detection and quantification of neutralizing antibodies are indicative of how protected an animal is against reinfection. The EHD VNT can therefore be a useful tool to monitor the efficacy of a vaccination campaign. VP2 is also the main determinant of EHDV serotype specificity, and so EHDV-neutralizing antibodies which target VP2 are also serotype-specific. Throughdetecting and quantifying neutralizing antibodies, the VNT can discriminate the EHDV serotype responsible for an infection and provides insights into the time of infection. It is considered the gold standard test for identifying and quantifying antibodies against EHDV serotypes present in test serum samples. The assay is performed in vitro and is based on inhibition of virus infectivity in the presence of neutralizing antibodies. A neutralizing antibody titer is determined through the presence or absence of cytopathic effect in a cell monolayer. The VNT is a relatively inexpensive assay using standard laboratory equipment; however, to perform the assay, cell cultures, significant time, intensive labor, and technical skill are required., (© 2024. Bluetongue (BT), caused by Bluetongue virus (BTV), is a disease that affects ruminants such as cattle, sheep, goats and deer. BTV is transmitted by female midges of the genus Culicoides. In Brazil, information on the prevalence of BTV in cattle is limited, so the objective of this work was to identify BTV serotypes in cattle. The State of São Paulo was divided into seven cattle-producing regions, and in each of them, 300 cattle farms were randomly selected. One animal from each farm (out of a total of 1,598 farms) was selected and its sera tested by virus neutralization technique against BTV serotypes (1-24 and 26) for determining antibody titre. Moreover, for each sampled farm, an epidemiological questionnaire was submitted to verify the type of cattle production and the zootechnical and sanitary practices carried out, which could be associated with a higher risk of BTV infection. In this study, antibodies (percentage, [95% confidence interval]) were identified against 11 serotypes: BTV-1 (22.15%, [15.72-27.92]), BTV-2 (31.03%, [26.65-37.98]), BTV-3 (18.96%, [12.42-24.90]), BTV-4 (24.90% [19.41-29.12]), BTV-9 (6.82%, [1.45-11.72]), BTV-12 (7.50%, [2.82-12.51]), BTV-17 (23.90%, [17.35-29.35]), BTV-19 (10.20%, [4.62-5.56]), BTV-21 (30.66%, [25.00-36.00]), BTV-22 (12.14%, [5.91-18.55]), BTV-26 (57.00%, [51.41-63.59]). In this study, for the first time in Brazil serological evidence of the presence of serotypes BTV-2, BTV-9, BTV-21 and BTV-26 is reported. Bovine follicular cysts are an ovarian disorder of dairy cows associated with abnormal estrous behaviour and infertility. The treatment of choice is intramuscular administration of a GnRH analogue, which acts by triggering pituitary release of LH. However, the presence of GnRH and GnRH receptors on spinal cord and ovary in some species, and the kind of innervation of the ovary, let us hypothesize that GnRH and its analogues may also act when administered by epidural route, as happens for other drugs. Therefore the aim of this study was to compare the effects of epidural vs intramuscular administration of lecirelin (a GnRH analogue) on FC regression, estrus detection and pregnancy outcomes. The study was conducted on 220 Friesian cows affected by follicular cysts, divided among 4 groups: Group L(epid) and Group L(im) received, respectively 50 μg of lecirelin in the epidural space and intramuscular; Group C(epid) and Group C(im) were used as control groups. In Group L(epid), estrus induction and pregnancy rates were significantly higher than in Group L(im). The results of this study show that the epidural administration of lecirelin promoted the remission of follicular cysts and an improvement of reproductive parameters compared to intramuscular administration. Thus, an alternative therapeutical approach is available for FC treatment, in order to obtain an easier restoration of the ovarian activity, especially in those cases refractory to classical therapeutic approaches., (Copyright © 2011 Elsevier B.V.
