3. Portable NIR Spectroscopy to Simultaneously Trace Honey Botanical and Geographical Origins and Detect Syrup Adulteration.

Abstract

Fraudulent practices concerning honey are growing fast and involve misrepresentation of origin and adulteration. Simple and feasible methods for honey authentication are needed to ascertain honey compliance and quality. Working on a robust dataset and simultaneously investigating honey traceability and adulterant detection, this study proposed a portable FTNIR fingerprinting approach combined with chemometrics. Multifloral and unifloral honey samples (n = 244) from Spain and Sardinia (Italy) were discriminated by botanical and geographical origin. Qualitative and quantitative methods were developed using linear discriminant analysis (LDA) and partial least squares (PLS) regression to detect adulterated honey with two syrups, consisting of glucose, fructose, and maltose. Botanical and geographical origins were predicted with 90% and 95% accuracy, respectively. LDA models discriminated pure and adulterated honey samples with an accuracy of over 92%, whereas PLS allows for the accurate quantification of over 10% of adulterants in unifloral and 20% in multifloral honey. Abstract

5-(hydroxymethyl)furan-2-carbaldehyde, better known as hydroxymethylfurfural (HMF), is a well-known freshness parameter of honey: although mostly absent in fresh samples, its concentration tends to increase naturally with aging. However, high quantities of HMF are also found in fresh but adulterated samples or honey subjected to thermal or photochemical stresses. In addition, HMF deserves further consideration due to its potential toxic effects on human health. The processes at the origin of HMF formation in honey and in other foods, containing saccharides and proteins—mainly non-enzymatic browning reactions—can also produce other furanic compounds. Among others, 2-furaldehyde (2F) and 2-furoic acid (2FA) are the most abundant in honey, but also their isomers (i.e., 3-furaldehyde, 3F, and 3-furoic acid, 3FA) have been found in it, although in small quantities. A preliminary characterization of HMF, 2F, 2FA, 3F, and 3FA by cyclic voltammetry (CV) led to hypothesizing the possibility of a comprehensive quantitative determination of all these compounds using a simple and accurate square wave voltammetry (SWV) method. Therefore, a new parameter able to provide indications on quality of honey, named “Furanic Index” (FI), was proposed in this contribution, which is based on the simultaneous reduction of all analytes on an Hg electrode to ca. 􀀀1.50 V vs. Saturated Calomel Electrode (SCE). The proposed method, validated, and tested on 10 samples of honeys of different botanical origin and age, is fast and accurate, and, in the case of strawberry tree honey (Arbutus unedo), it highlighted the contribution to the FI of the homogentisic acid (HA), i.e., the chemical marker of the floral origin of this honey, which was quantitatively reduced in the working conditions. Excellent agreement between the SWV and Reverse-Phase High-Performance Liquid Chromatography (RP-HPLC) data was observed in all samples considered.

35. Relationship between markers of botanical origin in nectar and honey of the strawberry tree (Arbutus unedo) throughout flowering periods in different years and in different geographical areas

Abstract

The amounts of four well-recognized markers of the floral origin of strawberry tree honey (i.e., 2,5-dihydroxyphenylacetic acid, homogentisic acid; (±)-2-cis,4-trans-abscisic acid, c,t-ABA; (±)-2-trans,4-trans-abscisic acid, t,t-ABA, and 2-(1,2-dihydroxypropyl)-4,8,8-trimethyl-1-oxaspiro[2.5]oct-4-en-6-one, unedone) were measured in the nectar of the strawberry tree throughout the flowering periods of two years and at various sampling sites. The concentration of these compounds was also measured in reference to strawberry tree honey samples from the same geographical origin in order to establish correlations and verify the possibility that one (or more) of them may act as marker(s) of the purity level, i.e., of the amount of nectar by strawberry tree flowers that has been transferred to the relevant honey. The data obtained show that despite the existence of a good correlation level between the markers, none of them at the moment can be proposed as a marker of purity of this honey, due to very complex interactions between contamination by foreign nectars in strawberry tree honey and spatial (geographical origin of the samples) and temporal (year and flowering period) parameters. Among these compounds, homogentisic acid appears to be the most reliable marker of botanical origin of strawberry tree honey due to its higher concentration with respect to those measured for isomers of abscisic acid and unedone.

45. Polyphenol-rich strawberry tree honey inhibits cell proliferation, promotes apoptosis and ROS production, and regulates EGFR signaling pathways in HCT-116 human colon carcinoma cells

Abstract

Objectives: To evaluate the chemopreventive activity of polyphenol-rich Strawberry tree honey, on the human colon cancer HCT-116 cell line. Methodology: 12 monofloral Strawberry tree honey (STH) samples were analyzed to determine their total phenolic (TP), flavonoid (TF) and protein contents as well as their total antioxidant capacity (TAC). HCT-116 cells were treated with different concentrations of the best STH sample and the cell viability was evaluated by MTT assay. It was also determined the intracellular ROS production and apoptosis rate by Tali® Image-Based Cytometer. In order to elucidate the pathway involved, the expression of different proteins related to the apoptosis (Caspase-3, p53 and PARP) and EGFR (p-AKT and p-EKR1/2) pathway was determined by western blotting. Results: Berchidda’s honey which showed the highest amount of TP, TF, protein and TAC was selected for treating HCT-116 cells. STH significantly reduced HCT-116 cells proliferation in a concentration-dependent manner and induced apoptosis and ROS production. After STH treatment, the Caspase-3, p-53 and PARP were significantly upregulated and EGFR, p-AKT and p-ERK1/2 signalling proteins were suppressed. Conclusion: STH could be a novel and promising chemotherapeutic agent in colon cancer treatment although more analysis is needed to confirm this hypothesis., Journal of International Society of Antioxidants in Nutrition & Health, Vol 3, No 4 (2016)
