1. Small Angle Scattering and Zeta Potential of Liposomes Loaded with Octa(carboranyl)porphyrazine

Abstract

In this work the physicochemical characterization of liposomes loaded with a newly synthesised carboranyl porphyrazine (H2HECASPz) is described. This molecule represents a potential drug for different anticancer therapies, such as Boron Neutron Capture Therapy, Photodynamic Therapy and Photothermal Therapy. Different loading methods and different lipid mixtures were tested. The corresponding loaded vectors were studied by Small Angle Scattering (SANS and SAXS), light scattering and zeta potential. The combined analysis of structural data at various length scales and the measurement of the surface charge allowed to obtain a detailed characterization of the investigated systems. The mechanisms underlying the onset of differences in relevant physicochemical parameters (size, polydispersity and charge) were also critically discussed.

Abstract

International audience; Liquid state transmission electron microscopy is a recent te-chnique allowing researchers to visualize the nanoparticles dire-ctly in their liquid environment. Accurate visualization of growth in small nanoparticles is desired yet a challenging process due to limited resolution obtained due to surfactant present in the sys-tem. In this work, by using liquid environmental cell specimen holder (e-cell), developed for scanning transmission electron microscopy, we were able to monitor the formation of anisotropic gold nanoparticles from small precursors (< 7 nm) in surfactant assisted media, under electron beam. Technique offers the possi-bility to probe nanoparticles in-situ and to obtain detailed infor-mation on how nanoparticles grow in solution. However, under electron beam irradiation, due to radiolysis of water, some radicals are produced which can interact with the reactants and modify the chemical processes. Such phenomenon has to be considered during in-situ STEM analysis. In case of gold, this prior drawback can also be used to control the reaction. In our work, the incident electron beam was chosen to achieve two tasks simultaneously; first, as a radiation source to induce the radiolysis of water (whi-ch is responsible for the reduction of gold precursor) and the se-cond, it allows the real time imaging of particles of interest in a thin layer of liquid. Initially, we have described influences of the beam parameters and the chemical conditions on the final shape of nanoparticles. Between these parameters, in case of altering the magnification (i.e. the dose rate), we have observed major diffe-rences in the shape of final nanoparticles obtained. Regarding to these differences, following formation mechanism has been pro-posed; at low dose rate, few number of produced seeds are able to grow into branched nanoparticles due to slow kinetics, caused simultaneously by slow ascorbic acid reduction at the surface of the nanoparticles and low number of radicals induced by electron beam, while at higher dose rate, the concentration of strong redu-cing agent is so high that the alimentation of the seeds will certa-inly proceed initially by fast feeding by pre-reduced Au(0) due to aqueous electrons and strong reducing agent and the slow feeding due to ascorbic acid reduction at the surface of the particles. Additionally, the structure of final nanoparticles obtained by in-situ STEM and by gamma irradiation source (at similar chemical com-positions) is compared to demonstrate the crucial importance of beam parameters on nanoparticle morphology.

Abstract

International audience; Small-Angles X-Ray Scattering (SAXS) has been established as a metrological method for the determination of nanoparticles size and size distribution. Modern SAXS Laboratory experiments 1 , by involving synchrotron-based instrumentation at lower price and very stable X-ray source, are more and more used in nanomaterials metrology. In the frame of the EURAMET Innanopart project, we have developed a methodology for the size, size distribution and concentration determination of spherical nanoparticles. This protocol involves a precise sample preparation, a rigorous procedure for the data acquisition, and a set of homemade software tools for the data processing-from the acquisition, the absolute scaling, to the analysis. The SAXS technique is very accurate for the characterization of simple case using nanoparticles : monodisperses, spheric form, unique composition. But it is more complicated in the case of nanoparticles size mixtures and in case of nanoparticles in complex media. Recent works in the lab shows it is possible to achieve precise measurement on mixture of spheric nanoparticles and nanoparticles in complex media (food additives) using different software or methodology approaches. Abstract

International audience; Despite the long-lasting interest in the synthesis control of nanoparticles (NPs) in both fundamental and applied nanosciences, the driving mechanisms responsible for their size and shape selectivity in an environment (solution) are not completely understood, and a clear assessment of the respective roles of equilibrium thermodynamics and growth kinetics is still missing. In this study, relying on an efficient atomistic computational approach, we decipher the dependence of energetics, shapes and morphologies of gold NPs on the strength and nature of the metal–environment interaction. We highlight the conditions under which the energy difference between isotropic and elongated gold NPs is reduced, thus prompting their thermodynamic coexistence. The study encompasses both monocrystalline and multi-twinned particles and extends over size ranges particularly representative of the nucleation and early growth stages. Computational results are further rationalized with arguments involving the dependence of facet and edge energies on the metal–environment interactions. We argue that by determining the abundance and diversity of particles nucleated in solution, thermodynamics may constitute an important bias influencing their final shape. The present results provide firm grounds for kinetic simulations of particle growth.

Abstract

High resolution cryo-TEM and Small Angle X-ray scattering are used for understanding the mesoscale structure of Al-Ge imogolite-like nanotubes in two contrasted situations. SAXS data has shown that the two concentric tubes have an equal length and identical wall structure and both SAXS and cryo-TEM data has confirmed the formation of single-walled nanotube at higher concentration.

Abstract

International audience; Small Angle X-ray Scattering (SAXS) allows traceable measurements of size, size distribution , surface area, concentration and shape of nanomaterials in solutions, powders and in bulk materials. We present here a custom-made state-of-the-art SAXS laboratory instrument (see Figure 1) based on Mo X-ray generator. Its design has been thought and optimized for the wide angles (WAXS) for investigate nanostructured material in particular in the size range from below 1 nm to above 20 nm. The X-ray generator is a molybdenum (17 keV) rotating anode. A combination of a multilayer collimating mirror and a patented hybrid slits gives us a very sharp and high purity beam for a laboratory setup, with a size of 1x1 mm2 and a flux of 10 8 photons/s. The motorized sample holder can load 20 capil-laries, a temperature controlled system or a circulation environment system for kinetic studies. A vacuum chamber is placed behind whose output window diameter is designed for the 2D image plate Mar345 detector. The accessible q-range is qmin = 0.03 Å-1 to qmax = 3 Å-1. A special attention was given to the complete data treatment processing (absolute intensities, uncertainties, reference samples, calibration ,…), enable us to make traceable measurements. Combining molybdenum energy and the state-of-art experimental setup provide us with a very powerful tool for nanomaterial studies [2][3]. We present here some results obtained with this experiment for inter-laboratories characterization studies in particular of silver nanoparticles, and Si nanoparticles.

24. Surface charging process in colloidal suspensions: A new statistical approach.

Abstract

We introduce a new statistical-mechanical model of the surface charge process in colloidal suspensions. The chemical adsorption of ions on the surface is described with a narrow and deep well in the pair potential between the spherical colloids and the ions. This extends the Cummings and Stell model of bulk chemical reactions. In this simple case of association of spherically symmetric atoms into diatomic molecules, we verify that the model fulfils the law of mass action involving the activities of the reactant and product particles. In the case of surface reactions, we solve numerically the hypernetted-chain integral equation for the equilibrium properties of the system. From the pair distribution functions we deduce the implicit structural charge of the colloid as well as the electrostatic condensation of the counter ions, the ion–ion correlations inside the chemical shell and the colloid–colloid effective repulsion. Abstract

International audience; The development of functional materials by taking advantage of the physical properties of nanoparticles needs an optimal control over their size and crystal quality. In this context, the synthesis of crystalline oxide nanoparticles in water at room temperature is a versatile and industrially appealing process but lacks control especially for "large" nanoparticles (>30 nm), which commonly consist of agglomerates of smaller crystalline primary grains. Improvement of these syntheses is hampered by the lack of knowledge on possible intermediate, noncrystalline stages, although their critical importance has already been outlined in crystallization processes. Here, we show that during the synthesis of luminescent Eu-doped YVO4 nanoparticles a transient amorphous network forms with a two-level structuration. These two prestructuration scales constrain topologically the nucleation of the nanometer-sized crystalline primary grains and their aggregation in nanoparticles, respectively. This template effect not only clarifies why the crystal size is found independent of the nucleation rate, in contradiction with the classical nucleation models, but also supports the possibility to control the final nanostructure with the amorphous phase.

The European Commission's Joint Research Centre (JRC) provides scientific support to European Union policy including nanotechnology. Within this context, the JRC launched, in February 2011, a repository for Representative Test Materials (RTMs), based on preparatory work started in 2008. It supports both EU and international research projects, and especially the OECD Working Party on Manufactured Nanomaterials (WPMN). The WPMN leads an exploratory testing programme "Testing a Representative set of Manufactured Nanomaterials" for the development and collection of data on characterisation, toxicological and ecotoxicological properties, as well as risk assessment and safety evaluation of nanomaterials. The purpose is to understand the applicability of the OECD Test Guidelines for the testing of nanomaterials as well as end-points relevant for such materials. The Repository responds to a need for nanosafety research purposes: availability of nanomaterial from a single production batch to enhance the comparability of results between different research laboratories and projects. The availability of representative nanomaterials to the international scientific community furthermore enhances and enables development of safe materials and products. The present report presents the physico-chemical characterisation of the Titanium dioxide series from the JRC repository: NM-100, NM-101, NM-102, NM-103, NM-104 and NM-105. NM-105 was selected as principal material for the OECD test programme "Testing a representative set of manufactured nanomaterials". NM-100 is included in the series as a bulk comparator. Each of these NMs originates from one batch of commercially manufactured TiO2. The TiO2 NMs may be used as representative material in the measurement and testing with regard to hazard identification, risk and exposure assessment studies. The results for more than 15 endpoints are addressed in the present report, including physico-chemical properties, such as size and size distribution, crystallite size and electron microscopy images. Sample and test item preparation procedures are addressed. The results are based on studies by several European laboratories participating to the NANOGENOTOX Joint Action, as well as by the JRC., JRC.I.4-Nanobiosciences

Abstract

In 2011 the JRC launched a Repository for Representative Test Materials that supports both EU and international research projects, and especially the OECD Working Party on Manufactured Nanomaterials' (WPMN) exploratory testing programme "Testing a Representative set of Manufactured Nanomaterials" for the development and collection of data on characterisation, toxicological and ecotoxicological properties, as well as risk assessment and safety evaluation of nanomaterials. The JRC Repository responds to a need for availability of nanomaterial from a single production batch to enhance the comparability of results between different research laboratories and projects. The present report presents the physico-chemical characterisation of the multi-walled carbon nanotubes (MWCNT) from the JRC Repository: NM-400, NM-401, NM-402 and NM-403. NM-400 was selected as principal material for the OECD WPMN testing programme. They are produced by catalytic chemical vapour deposition. Each of these NMs originates from one respective batch of commercially manufactured MWCNT. They are nanostructured, i.e. they consist of more than one graphene layer stacked on each other and rolled together as concentric tubes. The MWCNT NMs may be used as a representative material in the measurement and testing with regard to hazard identification, risk and exposure assessment studies. Abstract

The size control of gold nanoparticles synthesized in surfactant free water with a continuous flow mode was elucidated and used to produce higher concentration (3 mM) of stabilized gold nanoparticles. The originality of the synthesis was to finely modulate the initial pH of the reducing agent instead of the gold precursor to modify the kinetic of the reaction. The acceleration of the kinetic (~1 s) prevents the modification of the gold precursors ensuring the control of the final size (from 3 to 25 nm) of the nanoparticles with a low polydispersity for aqueous surfactant free solution. The accurate measure of the size distribution by small angle X-ray scattering was combined to the use of a model based on the coupling of nucleation and growth equations together with a progressive injection of monomers. The results on the final state show that the size of the nanoparticles is indeed controlled by the kinetic of reduction of gold atoms. A millifluidic setup equipped with a homemade mixer offers a robust way of rapid mixing to obtain a reproducible production of large amounts of nanoparticles.

We investigated the responses of two different models of unicellular bacteria to cerium oxide (CeO2) nanoparticles. These organisms are: Synechocystis PCC6803 a representative of environmentally important cyanobacterial organisms (producer of biomass for aquatic food chains), and Escherichia coli a representative of intestine-colonizing bacteria. Coupling physico-chemical (adsorption isotherms and electrophoretic mobility), biological (survival tests), microscopical (SEM, TEM and EDS) and spectroscopic (XANES) methods, we enlightened two distinct mechanisms for the CeO2 nanoparticles toxicological impact: A 'direct' mechanism that requires a close contact between nanoparticles and cell membranes, and an 'indirect' influence elicited by the acidity of nanoparticles stabilizing agents. We showed that E. coli is sensitive to the 'direct' effects of nanoparticles, whereas Synechocystis being protected by extracellular polymeric substances preventing direct cellular contacts is sensitive only to the 'indirect' mechanism. Consequently, our findings demonstrate the importance of the 'direct/indirect' effects of nanoparticles on cell fitness, a phenomenon that should be systematically investigated with appropriate techniques and dose metrics to make meaningful environmental and/or health recommendations.

Abstract

A Belousov Zhabotinsky(BZ) reaction, which is carried out in the anisotropic environment of the 1,2-dipalmitoyl-sn-glycero-3-phosphocholine(DPPC)/water binary system that presents layered aqueous domains separated by lipid bilayers are studied. Experiments on BZ behavior were carried out at room temperature by mixing DPPC with all the reactants, except ferroin, in a beaker under stirring.
