Bioregenerative food systems that routinely produce fresh, safe-to-eat crops onboard spacecraft can supplement the nutrition and variety of shelf-stable spaceflight food systems for use during future exploration missions (i.e., low earth orbit, Mars transit, lunar, and Martian habitats). However, current space crop production systems are not yet sustainable because they primarily utilize consumable granular media and, to date, operate like single crop cycle, space biology experiments where root modules are sanitized prior to launch and discarded after each grow-out. Moreover, real-time detection of the cleanliness of crops produced in spacecraft is not possible. A significant paradigm shift is needed in the design of future space crop production systems, as they transition from operating as single grow-out space biology experiments to becoming sustainable over multiple cropping cycles. Soilless nutrient delivery systems have been used to demonstrate post-harvest sanitization and inflight microbial monitoring technologies to enable sequential cropping cycles in spacecraft. Post-harvest cleaning and sanitization prevent the buildup of biofilms and ensure a favorable environment for seedling establishment of the next crop. Inflight microbial monitoring of food and watering systems ensures food safety in spaceflight food systems. A sanitization protocol, heat sterilization at 60°C for 1 h, and soaking for 12 h in 1% hydrogen peroxide, developed in this study, was compared against a standard hydroponic sanitization protocol during five consecutive crop cycles. Each cropping cycle included protocols for the cultivation of a crop to maturity, followed by post-harvest cleaning and inflight microbial monitoring. Microbial sampling of nutrient solution reservoirs, root modules, and plants demonstrated that the sanitization protocol could be used to grow safe-to-eat produce during multiple crop cycles. The cleanliness of the reservoir and root module surfaces measured with aerobic plate counts was verified in near real time using a qPCR-based inflight microbial monitoring protocol. Post-harvest sanitization and inflight microbial monitoring are expected to significantly transform the design of sustainable bioregenerative food and life support systems for future exploration missions beyond low earth orbit (LEO). AbstractPurposeMaterials and MethodsResults and ConclusionsThe study of mechanisms of plant responses to extreme conditions, particularly, microgravity and ionizing radiation, is crucial for space exploration. Modern space biology of plants focuses on increasing plant tolerance to harsh conditions of space environment. Given the limited access to the International Space Station, we designed and assembled the 3D clinostat for mimicking microgravity, which, in combination with proton irradiation, allows simulating space conditions. As a case study for testing the device, we studied the effect of clinostating on Arabidopsis thaliana accession originating from the Chernobyl exclusion zone.Using the combined clinostating and proton irradiation, we simulated the conditions of long-term space flight for Arabidopsis thaliana plants of the Chernobyl accession – progeny of chronically irradiated plants, grown from field-collected (Masa-0) and laboratory-cultivated (Masa-0-1) seeds, and for wild-type Col-8. The clinostating and irradiation of plants were also carried out separately. Plant responses were studied as photosynthetic and phenotypic endpoints of seedlings.Parameters of chlorophyll fluorescence estimated immediately after exposure showed that Masa-0-1 plants were resistant to the simulated space conditions, while Masa-0 demonstrated modulation of non-photochemical fluorescence quenching. Proton irradiation generally inhibited photosynthesis of Masa-0, Masa-0-1, and Col-8 seedlings. The combined effect of irradiation and clinostating modulated the photosynthetic activity of Col-8 seedlings. The leaf area of seedlings did not change after exposure to simulated conditions. Results: Here, we demonstrate that even short exposures to varying elevated relative humidity can facilitate rapid microbial growth and microbial community composition changes in dust from spacecraft. We modeled fungal growth in dust from the International Space Station using the time-of-wetness framework with activation and deactivation limited growth occurring at 85% and 100% relative humidity, respectively. Fungal concentrations ranged from an average of 4.4 × 106 spore equivalents per milligram of dust in original dust with no exposure to relative humidity to up to 2.1 × 1010 when exposed to 100% relative humidity for 2 weeks. As relative humidity and time-elevated increased, fungal diversity was significantly reduced for both alpha (Q < 0.05) and beta (R2 = 0.307, P = 0.001) diversity metrics. Bacteria were unable to be modeled using the time-of-wetness framework. However, bacterial communities did change based on constant relative humidity incubations for both beta (R2 = 0.22, P = 0.001) and alpha diversity decreasing with increasing moisture starting at 85% relative humidity (Q < 0.05). Conclusion: Our results demonstrate that moisture conditions can be used to develop and predict changes in fungal growth and composition onboard human-occupied spacecraft. The exploration of microgravity has garnered substantial scholarly attention due to its potential to offer unique insights into the behavior of biological systems. This study presents a preliminary investigation into the effects of simulated microgravity on esophageal cancer cells, examining various aspects such as morphology, growth behavior, adhesion, inhibition rate, and DNA damage. To achieve this, a novel microgravity simulator named "Gravity Challenge" was utilized for its effectiveness in minimizing external influences that could compromise microgravity conditions. The international cell line SK-GT-4 was utilized as the focal point of this investigation. Results revealed noticeable alterations in the growth behavior of cancer cells following exposure to simulated microgravity for 24 h, characterized by a loss of adhesion properties compared to control cells. Concurrently, cell viability exhibited a decline, as evidenced by cytotoxicity testing. Furthermore, the comet assay test demonstrated that cells subjected to microgravity simulation experienced a higher incidence of DNA damage compared to their control counterparts. In conclusion, this comprehensive examination of the impact of simulated microgravity on esophageal cancer cells extends beyond morphological changes, delving into genetic implications through observed DNA damage. In recent decades, the interest of the scientific community in plant space biology increased based on the certainty that cultivating higher plants in space is imperative for prolonged human missions. The cultivation of plants in space assumes great relevance moved by the consideration that there is a pivotal need to improve resource regeneration and onboard production of plant-based food, thus reducing the supply of resources from Earth. Therefore, the hereby paper provides an overview on the evolution of the interest in plant space biology from 2000 to 2023, basing the search on specific keyword combinations. The study encompasses (i) a proper quantitative search on the Scopus database; (ii) keyword frequencies and clustering analysis, (iii) a keyword network and time-gradient analysis and (iv) an elaboration of knowledge gaps comparing our results and recent reviews. Results highlighted an increase in publications in the last 10 years and a specific time-gradient related shifting on subtopics such as in situ resource utilization (ISRU) application and food production in a space mission context. Currently, knowledge gaps were identified in four research areas related to water dynamics in extra-terrestrial environments, plant physiology and biochemistry in response to growth on regolith, interactions with fertilizers, and plant/soil microbiome. Identifying these gaps will allow to canalize future efforts of space research where needed, facing the unsolved questions. As a model organism for space biology experiments, Caenorhabditis elegans (C. elegans) has low demand for life support and strong resistance to unfavorable environments, making experimentation with C. elegans relatively easy and cost-effective. Previously, C. elegans has been flown in several spaceflight investigations, but there is still an urgent need for analytical platforms enabling on-orbit automated monitoring of multiple phenotypes of worms, such as growth and development, movement, changes of biomarkers, etc. To solve this problem, we presented a fully integrated microfluidic system (WormSpace μ-TAS) with an arrayed microfluidic chip (WormChip-4.8.1) and a replaceable microfluidic module (WormChip cartridge), which was compatible with the experimental facility on the China Space Station (CSS). By adopting technologies of programmed fluid control based on liquid medium CeMM as well as multi-function imaging with a camera mounted on a three-dimensional (3D) transportation stage, automated and long-term experimentation can be performed for on-chip multi-strain culturing and bright-field and fluorescence imaging of C. elegans at the single-worm level. The presented WormSpace μ-TAS enabled its successful application on the CSS, achieving flight launch of the sample unit (WormChip cartridge) at low temperature (controlled by a passive thermal case at 12 °C), automated 30-day cultivation of 4 strains of C. elegans, on-orbit monitoring of multiple phenotypes (growth and development, movement, and changes of fluorescent protein expression) at the single worm-level, on-chip fixation of animals at the end of the experiment and returning the fixed samples to earth. In summary, this study presented a verified microfluidic system and experimental protocols for automated on-chip multi-strain culturing and multi-function imaging of C. elegans at the single-worm level on the CSS. The SpaceX Inspiration4 mission provided a unique opportunity to study the impact of spaceflight on the human body. Biospecimen samples were collected from four crew members longitudinally before (Launch: L-92, L-44, L-3 days), during (Flight Day: FD1, FD2, FD3), and after (Return: R + 1, R + 45, R + 82, R + 194 days) spaceflight, spanning a total of 289 days across 2021-2022. The collection process included venous whole blood, capillary dried blood spot cards, saliva, urine, stool, body swabs, capsule swabs, SpaceX Dragon capsule HEPA filter, and skin biopsies. Venous whole blood was further processed to obtain aliquots of serum, plasma, extracellular vesicles and particles, and peripheral blood mononuclear cells. In total, 2,911 sample aliquots were shipped to our central lab at Weill Cornell Medicine for downstream assays and biobanking. This paper provides an overview of the extensive biospecimen collection and highlights their processing procedures and long-term biobanking techniques, facilitating future molecular tests and evaluations.As such, this study details a robust framework for obtaining and preserving high-quality human, microbial, and environmental samples for aerospace medicine in the Space Omics and Medical Atlas (SOMA) initiative, which can aid future human spaceflight and space biology experiments. To study the effects of an increased MFE, the magnetic field typically experienced by life on Earth was amplified by an order of magnitude. In this study, six treatments of C. vulgaris with two repetitions for each treatment were exposed to a magnetic field of 5 Gauss (500 µT) about each axis, which was generated in a precision state-of-the-art calibrated Helmholtz cage. The treatments and the control were characterized by the duration of exposure, which was varied from 0 min to 120 min with a step increment of 20 min. The treatments were repeated for six days and twelve days in two separate experiments. For the parametric study of the outcomes, the treated species were analyzed for overall growth, protein, and beta-carotene content. It was largely observed that the increased MFE had a significant impact on the overall growth and to a lesser extent on the protein and beta-carotene production. Specifically, for the specimen treated with 40 min of the increased MFE, the growth on the 21st day was measured to be 38% higher than the control. For the specimen treated with 120 min of the increased MFE, the protein content on the 15th day was measured to be 15.6% higher than the control. For the specimen treated with 40 min of the increased MFE, the beta-carotene content on the 15th day was measured to be 20.4% higher than the control. For TR2 in general, the results were inferior compared to TR1 but showed higher production than the control specimen. Specifically, for the specimen treated with 80 min of the increased MFE, the protein content on the 21st day was measured to be 4.3% higher than the control. For the specimen treated with 100 min of the increased MFE, the beta-carotene content on the 15th day was measured to be 17.1% higher than the control. For the specimen treated with 100 min of the increased MFE, the growth on the 21st day was measured to be 5% higher than the control. The examination of the impact of microgravity on biological systems has gained considerable attention owing to its potential implications for health and disease. Simulated microgravity serves as a valuable methodology for elucidating the intricate cellular responses to altered gravitational conditions. This study investigates the effects of simulated microgravity on cellular DNA, employing four distinct cell lines—breast, brain, and esophageal cancer cells, in conjunction with normal cells for comparative analysis. The experiment utilized the comet assay test to quantitatively assess DNA damage. The results revealed a discernible disparity in the response to simulated microgravity, notably with cancer cells exhibiting a significant increase in DNA damage compared to the relatively minimal effects observed in both control and normal cells. Furthermore, within the cancer cell lines, significant variations in the extent of DNA damage were evident, implying a cell type-dependent response to simulated microgravity. These findings illuminate the potential differential susceptibility of cancerous and normal cells to microgravity-induced DNA damage. Consequently, this research substantially contributes to our comprehension of microgravity-induced cellular responses and unveils promising avenues for targeted interventions in cancer therapy. Exposure to ionizing radiation is considered by NASA to be a major health hazard for deep space exploration missions. Ionizing radiation sensitivity is modulated by both genomic and environmental factors. Understanding their contributions is crucial for designing experiments in model organisms, evaluating the risk of deep space (i.e. high-linear energy transfer, or LET, particle) radiation exposure in astronauts, and also selecting therapeutic irradiation regimes for cancer patients. We identified single nucleotide polymorphisms in 15 strains of mice, including 10 collaborative cross model strains and 5 founder strains, associated with spontaneous and ionizing radiation-induced in vitro DNA damage quantified based on immunofluorescent tumor protein p53 binding protein (53BP1) positive nuclear foci. Statistical analysis suggested an association with pathways primarily related to cellular signaling, metabolism, tumorigenesis and nervous system damage. We observed different genomic associations in early (4 and 8 h) responses to different LET radiation, while later (24 hour) DNA damage responses showed a stronger overlap across all LETs. Furthermore, a subset of pathways was associated with spontaneous DNA damage, suggesting 53BP1 positive foci as a potential biomarker for DNA integrity in mouse models. Our results suggest several mouse strains as new models to further study the impact of ionizing radiation and validate the identified genetic loci. We also highlight the importance of future human in vitro studies to refine the association of genes and pathways with the DNA damage response to ionizing radiation and identify targets for space travel countermeasures.

Periodically, the European Space Agency (ESA) updates scientific roadmaps in consultation with the scientific community. The ESA SciSpacE Science Community White Paper (SSCWP) 9, "Biology in Space and Analogue Environments", focusses in 5 main topic areas, aiming to address key community-identified knowledge gaps in Space Biology. Here we present one of the identified topic areas, which is also an unanswered question of life science research in Space: "How to Obtain an Integrated Picture of the Molecular Networks Involved in Adaptation to Microgravity in Different Biological Systems?" The manuscript reports the main gaps of knowledge which have been identified by the community in the above topic area as well as the approach the community indicates to address the gaps not yet bridged. Moreover, the relevance that these research activities might have for the space exploration programs and also for application in industrial and technological fields on Earth is briefly discussed. The in-depth analysis concerning application of microfluidic instruments for space biology research is presented. The article focuses on recently investigated key scientific fields, i.e., lab-on-chips applied to the biomedical studies performed in the (1) International Space Station and (2) CubeSat nanosatellites. The paper presents also the lab-on-chip devices that were fabricated with a view to future space biology research and to those that to date have been solely been tested under Earth laboratory conditions and/or simulated microgravity environments. All life on Earth has evolved under the influence of continuous gravity, and methods have been developed to balance this influence with the biological evolution of organisms at the cellular and system levels. However, when exposed to zero gravity in space, the balance between cell structure and external forces is destroyed, resulting in changes at the cellular level (e.g., cell morphology, adhesion, viability, apoptosis, etc.), and understanding the molecular mechanism of cell response to zero gravity will help to cope with diseases that rely on mechanical response. Therefore, biological research in space and zero gravity is a unique step in developing the best anti-cancer treatments, which is a great challenge to humanity. In this study, multicellular glioma cancer cells from a brain tumor in a 72-year-old Iraqi patient were subjected to simulated zero gravity for 24 h, and the results showed that most of the cells lost their adhesion, which is considered to be the first step toward cell apoptosis. In addition to the formation of multicellular spheroids, the results also showed that the inhibition rate for cell death was 32% in comparison to the control cells. Moreover, the cells showed a clear change in their cellular morphology and growth behavior. These results give new hope for fighting cancer distinctively, and such a treatment method has no side effects in comparison to traditional chemical and radiological ones. The intangible desire to explore the mysteries of the universe has driven numerous advancements for humanity for centuries. Extraterrestrial journeys are becoming more realistic as a result of human curiosity and endeavors. Over the years, space biology research has played a significant role in understanding the hazardous effects of the space environment on human health during long-term space travel. The inevitable consequence of a space voyage is space ionizing radiation, which has deadly aftereffects on the human body. The paramount objective of this study is to provide a robust platform for performing biological experiments within the Earth's stratosphere by utilizing high-altitude balloons. This platform allows the use of a biological payload to simulate spaceflight missions within the unique properties of space that cannot be replicated in terrestrial facilities. This paper describes the feasibility and demonstration of a biological balloon mission suitable for students and scientists to perform space biology experiments within the boundary of the stratosphere. In this study, a high-altitude balloon was launched into the upper atmosphere (∼29 km altitude), where living microorganisms were exposed to a hazardous combination of UV irradiation, ultralow pressure and cold shock. The balloon carried the budding yeast Saccharomyces cerevisiae to investigate microbial survival potential under extreme conditions. The results indicated a notable reduction in biosample mortality two orders of magnitude (2-log) after exposure to 164.9 kJ m−2 UV. Postflight experiments have shown strong evidence that the effect of UV irradiation on living organisms is stronger than that of other extreme conditions.

Studying microbes at the single-cell level in space can accelerate human space exploration both via the development of novel biotechnologies and via the understanding of cellular responses to space stressors and countermeasures. High-throughput technologies for screening natural and engineered cell populations can reveal cellular heterogeneity and identify high-performance cells. Here, we present a method to desiccate and preserve microbes in nanoliter-scale compartments, termed PicoShells, which are microparticles with a hollow inner cavity. In PicoShells, single cells are confined in an inner aqueous core by a porous hydrogel shell, allowing the diffusion of nutrients, wastes, and assay reagents for uninhibited cell growth and flexible assay protocols. Desiccated PicoShells offer analysis capabilities for single-cell derived colonies with a simple, low resource workflow, requiring only the addition of water to rehydrate hundreds of thousands of PicoShells and the single microbes encapsulated inside. Our desiccation method results in the recovery of desiccated microparticle morphology and porosity after a multi-week storage period and rehydration, with particle diameter and porosity metrics changing by less than 18% and 7%, respectively, compared to fresh microparticles. We also recorded the high viability of Saccharomyces cerevisiae yeast desiccated and rehydrated inside PicoShells, with only a 14% decrease in viability compared to non-desiccated yeast over 8.5 weeks, although we observed an 85% decrease in initial growth potential over the same duration. We show a proof-of-concept for a growth rate-based analysis of single-cell derived colonies in rehydrated PicoShells, where we identified 11% of the population that grows at an accelerated rate. Desiccated PicoShells thus provide a robust method for cell preservation before and during launch, promising a simple single-cell analysis method for studying heterogeneity in microbial populations in space.

Our ability to explore the cosmos by direct contact has been limited to a small number of lunar and interplanetary missions. However, the NASA Starlight program points a path forward to send small, relativistic spacecraft far outside our solar system via standoff directed-energy propulsion. These miniaturized spacecraft are capable of robotic exploration but can also transport seeds and organisms, marking a profound change in our ability to both characterize and expand the reach of known life. Here we explore the biological and technological challenges of interstellar space biology, focusing on radiation-tolerant microorganisms capable of cryptobiosis. Additionally, we discuss planetary protection concerns and other ethical considerations of sending life to the stars.

A recent study published in Experimental Physiology details the first Canadian study to investigate the physiological, psychological, and cognitive responses to 14 days of bed rest and the benefits of exercise during bed rest. The study involved 23 older male and female participants who completed 14 days of bed rest, followed by supervised rehabilitation. Half of the participants performed aerobic and resistance exercise during bed rest to test its effectiveness as a countermeasure. Background: Understanding and countering the well-established negative health consequences of spaceflight remains a primary challenge preventing safe deep space exploration. Targeted/personalized therapeutics are at the forefront of space medicine strategies, and cross-species molecular signatures now define the 'typical' spaceflight response. However, a lack of direct genotype–phenotype associations currently limits the robustness and, therefore, the therapeutic utility of putative mechanisms underpinning pathological changes in flight. Methods: We employed the worm Caenorhabditis elegans as a validated model of space biology, combined with 'NemaFlex-S' microfluidic devices for assessing animal strength production as one of the most reproducible physiological responses to spaceflight. Wild-type and dys-1 (BZ33) strains (a Duchenne muscular dystrophy (DMD) model for comparing predisposed muscle weak animals) were cultured on the International Space Station in chemically defined media before loading second-generation gravid adults into NemaFlex-S devices to assess individual animal strength. These same cultures were then frozen on orbit before returning to Earth for next-generation sequencing transcriptomic analysis. Results: Neuromuscular strength was lower in flight versus ground controls (16.6% decline, p < 0.05), with dys-1 significantly more (23% less strength, p < 0.01) affected than wild types. The transcriptional gene ontology signatures characterizing both strains of weaker animals in flight strongly corroborate previous results across species, enriched for upregulated stress response pathways and downregulated mitochondrial and cytoskeletal processes. Functional gene cluster analysis extended this to implicate decreased neuronal function, including abnormal calcium handling and acetylcholine signaling, in space-induced strength declines under the predicted control of UNC-89 and DAF-19 transcription factors. Finally, gene modules specifically altered in dys-1 animals in flight again cluster to neuronal/neuromuscular pathways, suggesting strength loss in DMD comprises a strong neuronal component that predisposes these animals to exacerbated strength loss in space. Conclusions: Highly reproducible gene signatures are strongly associated with space-induced neuromuscular strength loss across species and neuronal changes in calcium/acetylcholine signaling require further study. During the Advanced Plant Habitat experiment 2, radish plants were grown in two successive grow-outs on the International Space Station (ISS) for 27 days each. On days 10, 18, and 24, leaf punch (LP) samples were collected and frozen. At harvest, bulb tissue was sampled with oligo-dT functionalized Solid Phase Gene Extraction (SPGE) probes. The space samples were compared with samples from ground controls (GC) grown at the Kennedy Space Center (KSC) under the same conditions as on the ISS, with notably elevated CO2 (about 2500 ppm), and from lab plants grown under atmospheric CO2 but with light and temperature conditions similar to the KSC control. Genes corresponding to peroxidase (RPP), glucosinolate biosynthesis (GIS), protein binding (CBP), myrosinase (RMA), napin (RSN), and ubiquitin (UBQ) were measured by qPCR. LP from day 24 and bulb samples collected at harvest were compared with RNA-seq data from material that was harvested, frozen, and analyzed after return to Earth. The results showed stable transcription in LP samples in GC but decreasing values in ISS samples during both grow-outs, possibly indicative of stress. SPGE results were similar between GC and ISS samples. However, the RNA-seq analyses showed different transcription profiles than SPGE or LP results, possibly related to localized sampling. RNA-seq of leaf samples showed greater variety than LP data, possibly because of different sampling times. RSN and RPP showed the lowest transcription regardless of method. Temporal analyses showed relatively small changes during plant development in space and in ground controls. Simulated microgravity (SMG) is an environmental condition that affects bone density in vertebrates. Ground-based studies typically use a random positioning machine in either a 2D or a 3D mode to assess the effects of SMG, however the meaning of these results is difficult to compare between studies due to different experimental parameters. Here, we exposed larval Danio rerio at 3dpf to 23 h of SMG using a 2D and a 3D mode of rotation, using the same experimental setup. Zebrafish larvae were anaesthetized during the experiment. Our results showed that anesthesia (MS222) did not affect the amount of ossification while SMG-2D treatment slightly reduced the amount of ossification compared with the controls. On the other hand, SMG-3D treatment significantly reduced the overall ossification level of the skeleton. Specifically, the anterior end of the notochord, the ceratobranchial-5, the lower jaw articulation, the pharyngeal teeth, and the operculum were affected compared with control treatments. Overall, these results indicate that SMG-3D produced a more effective SMG effect compared with the SMG-2D. Mast cell (MC)-specific proteases are of particular interest for space biology and medicine due to their biological activity in regulating targets of a specific tissue microenvironment. MC tryptase and chymase obtain the ability to remodel connective tissue through direct and indirect mechanisms. Yet, MC-specific protease expression under space flight conditions has not been adequately investigated. Using immunohistochemical stainings, we analyzed in this study the protease profile of the jejunal, gastric, and hepatic MC populations in three groups of Mongolian gerbils—vivarium control, synchronous experiment, and 12-day orbital flight on the Foton-M3 spacecraft—and in two groups—vivarium control and anti-orthostatic suspension—included in the experiment simulating effects of weightlessness in the ground-based conditions. After a space flight, there was a decreased number of MCs in the studied organs combined with an increased proportion of chymase-positive MCs and MCs with a simultaneous content of tryptase and chymase; the secretion of specific proteases into the extracellular matrix increased. These changes in the expression of proteases were observed both in the mucosal and connective tissue MC subpopulations of the stomach and jejunum. Notably, the relative content of tryptase-positive MCs in the studied organs of the digestive system decreased. Space flight conditions simulated in the synchronous experiment caused no similar significant changes in the protease profile of MC populations. The space flight conditions resulted in an increased chymase expression combined with a decreased total number of protease-positive MCs, apparently due to participating in the processes of extracellular matrix remodeling and regulating the state of the cardiovascular system. Advancements in plant space biology are required for the realization of human space exploration missions, where the re-supply of resources from Earth is not feasible. Until a few decades ago, space life science was focused on the impact of the space environment on the human body. More recently, the interest in plant space biology has increased because plants are key organisms in Bioregenerative Life Support Systems (BLSS) for the regeneration of resources and fresh food production. Moreover, plants play an important role in psychological support for astronauts. The definition of cultivation requirements for the design, realization, and successful operation of BLSS must consider the effects of space factors on plants. Altered gravitational fields and radiation exposure are the main space factors inducing changes in gene expression, cell proliferation and differentiation, signalling and physiological processes with possible consequences on tissue organization and organogenesis, thus on the whole plant functioning. Interestingly, the changes at the cellular and molecular levels do not always result in organismic or developmental changes. This apparent paradox is a current research challenge. In this paper, the main findings of gravity- and radiation-related research on higher plants are summarized, highlighting the knowledge gaps that are still necessary to fill. Existing experimental facilities to simulate the effect of space factors, as well as requirements for future facilities for possible experiments to achieve fundamental biology goals are considered. Finally, the need for making synergies among disciplines and for establishing global standard operating procedures for analyses and data collection in space experiments is highlighted. Thus, gravity has shaped all biological functions, some examples include the growth of plants (e.g., gravitropism), the structure and morphology of biological parts in multicellular organisms, to its effects on our physiological function when humans travel into space. Moreover, from an evolutionary perspective, gravity has been a constant force on biology, and life, to our understanding, should have no reason to not experience the effects of gravity. Interestingly, there appear to be specific biological mechanisms that activate in the absence of gravity, with the space environment the only location to study the effects of a lack of gravity on biological systems. Thus, in this perspective piece, biological adaptations from the cellular to the whole organism levels to the presence and absence of gravity will be organized and described, as well as outlining future areas of research for gravitational biological investigations to address. Up to now, we have observed and shown how gravity effects biology at different levels, with a few examples including genetic (e.g., cell cycle, metabolism, signal transduction associated pathways, etc.), biochemically (e.g., cytoskeleton, NADPH oxidase, Yes-associated protein, etc.), and functionally (e.g., astronauts experiencing musculoskeletal and cardiovascular deconditioning, immune dysfunction, etc., when traveling into space). Based from these observations, there appear to be gravity-sensitive and specific pathways across biological organisms, though knowledge gaps of the effects of gravity on biology remain, such as similarities and differences across species, reproduction, development, and evolutionary adaptations, sex-differences, etc. Thus, here an overview of the literature is provided for context of gravitational biology research to-date and consideration for future studies, as we prepare for long-term occupation of low- Earth Orbit and cis-Lunar space, and missions to the Moon and Mars, experiencing the effects of Lunar and Martian gravity on biology, respectively, through our Artemis program. A single strain from the family Paenibacillaceae was isolated from the wall behind the Waste Hygiene Compartment aboard the International Space Station (ISS) in April 2018, as part of the Microbial Tracking mission series. This strain was identified as a gram-positive, rod-shaped, oxidase-positive, catalase-negative motile bacterium in the genus Cohnella, designated as F6_2S_P_1T. The 16S sequence of the F6_2S_P_1T strain places it in a clade with C. rhizosphaerae and C. ginsengisoli, which were originally isolated from plant tissue or rhizosphere environments. The closest 16S and gyrB matches to strain F6_2S_P_1T are to C. rhizosphaerae with 98.84 and 93.99% sequence similarity, while a core single-copy gene phylogeny from all publicly available Cohnella genomes places it as more closely related to C. ginsengisoli. Average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values to any described Cohnella species are <89 and <22%, respectively. The major fatty acids for strain F6_2S_P_1T are anteiso-C15: 0 (51.7%), iso-C16: 0 (23.1%), and iso-C15: 0 (10.5%), and it is able to metabolize a wide range of carbon compounds. Given the results of the ANI and dDDH analyses, this ISS strain is a novel species within the genus Cohnella for which we propose the name Cohnella hashimotonis, with the type strain F6_2S_P_1T (=NRRL B-65657T and DSMZ 115098T). Because no closely related Cohnella genomes were available, this study generated the whole-genome sequences (WGSs) of the type strains for C. rhizosphaerae and C. ginsengisoli. Phylogenetic and pangenomic analysis reveals that F6_2S_P_1T, C. rhizosphaerae, and C. ginsengisoli, along with two uncharacterized Cohnella strains, possess a shared set of 332 gene clusters which are not shared with any other WGS of Cohnella species, and form a distinct clade branching off from C. nanjingensis. Small satellite technologies, particularly CubeSats, are enabling breakthrough research in space. Over the past 15 years, NASA Ames Research Center has developed and flown half a dozen biological CubeSats in low Earth orbit (LEO) to conduct space biology and astrobiology research investigating the effects of the space environment on microbiological organisms. These studies of the impacts of radiation and reduced gravity on cellular processes include dose-dependent interactions with antimicrobial drugs, measurements of gene expression and signaling, and assessment of radiation damage. BioSentinel, the newest addition to this series, will be the first deep space biological CubeSat, its heliocentric orbit extending far beyond the radiation-shielded environment of low Earth orbit. BioSentinel's 4U biosensing payload, the first living biology space experiment ever conducted beyond the Earth-Moon system, will use a microbial bioassay to assess repair of radiation-induced DNA damage in eukaryotic cells over a duration of 6–12 months. Our understanding of the diversity of life on our planet and the possibility of finding or sustaining life elsewhere in the universe plays a central role in supporting human space settlement and exploration. Astrobiology and its outcomes require a multidisciplinary and comprehensive approach, in which the microbial, geological, chemical, astronomical, and physical domains of research are interlinked. An example of the applications of astrobiology and space microbiology is the use of extremophiles for in situ resource utilization through biomining and bioleaching. To better understand the multidisciplinary research landscape in this area, we quantitatively reviewed the global scientific literature on astrobiology, with a focus on biomining and bioleaching through bibliometric network analysis, investigating patterns and trends in its development over time. The network analysis of keyword co-occurrence highlights different connecting and overlapping clusters, illustrating the multidisciplinary character of astrobiology. Temporal analyses show a recent focus on topics related to microbiology and geomicrobiology, emphasizing the role that these fields will play in future astrobiology research. In conclusion, astrobiology, biomining, and bioleaching research are currently addressing the recognition of these techniques as valuable tools for biotechnological applications, expected to play a crucial role in long-term human space exploration. Introduction: RNA sequencing (RNA-seq) data from space biology experiments promise to yield invaluable insights into the effects of spaceflight on terrestrial biology. However, sample numbers from each study are low due to limited crew availability, hardware, and space. To increase statistical power, spaceflight RNA-seq datasets from different missions are often aggregated together. However, this can introduce technical variation or “batch effects”, often due to differences in sample handling, sample processing, and sequencing platforms. Several computational methods have been developed to correct for technical batch effects, thereby reducing their impact on true biological signals.Methods: In this study, we combined 7 mouse liver RNA-seq datasets from NASA GeneLab (part of the NASA Open Science Data Repository) to evaluate several common batch effect correction methods (ComBat and ComBat-seq from the sva R package, and Median Polish, Empirical Bayes, and ANOVA from the MBatch R package). Principal component analysis (PCA) was used to identify library preparation method and mission as the primary sources of batch effect among the technical variables in the combined dataset. We next quantitatively evaluated the ability of each of the indicated methods to correct for each identified technical batch variable using the following criteria: BatchQC, PCA, dispersion separability criterion, log fold change correlation, and differential gene expression analysis. Each batch variable/correction method combination was then assessed using a custom scoring approach to identify the optimal correction method for the combined dataset, by geometrically probing the space of all allowable scoring functions to yield an aggregate volume-based scoring measure.Results and Discussion: Using the method described for the combined dataset in this study, the library preparation variable/ComBat correction method pair out ranked the other candidate pairs, suggesting that this combined dataset should be corrected for library preparation using the ComBat correction method prior to downstream analysis. We describe the GeneLab multi-study analysis and visualization portal which will allow users to access the publicly available space biology ‘omics data, select multiple studies to combine for analysis, and examine the presence or absence of batch effects using multiple metrics. If the user chooses to perform batch effect correction, the scoring approach described here can be implemented to identify the optimal correction method to use for their specific combined dataset prior to analysis.

Space exploration and extraterrestrial civilization have fascinated humankind since the earliest days of human history. It was only in the last century that humankind finally began taking significant steps towards these goals by sending astronauts into space, landing on the moon, and building the International Space Station. However, space voyage is very challenging and dangerous, and astronauts are under constant space radiation and microgravity. It has been shown that astronauts are at a high risk of developing a broad range of diseases/disorders. Thus, it is critical to develop a rapid and effective assay to monitor astronauts' health in space. In this study, gene expression and correlation patterns were analyzed for 10 astronauts (8 male and 2 female) using the publicly available microarray dataset E-GEOD-74708. We identified 218 differentially expressed genes between In-flight and Pre-flight and noticed that space travel decreased genome regulation and gene correlations across the entire genome, as well as individual signaling pathways. Furthermore, we systematically developed a shortlist of 32 genes that could be used to monitor astronauts' health during space travel. Chizhevsky's ideas on periodicity as a major property of the organic and inorganic world and the links between rhythmic processes observed in living organisms and the rhythms of the cosmos are regarded as the fundamental basis of space biorhythmology, which was formulated in Russia in the second half of the 20th century. Space biorhythmology has become an independent scientific and practical area of space biology and medicine and seeks to study the rhythmic features of the functioning of living systems in both normal conditions and in space flight and to use the knowledge gained to ensure the health and performance of astronauts. A number of theoretical positions have been formulated in accordance with the principles of space biorhythmology: the most important of these are the definition of biological rhythms as a form of motion of living matter, expressing the unity and conflict between mutually exclusive and opposing principles within the immanent boundaries of any living system and ensuring its continuous self-reproduction; provisions relating to the integrating role of circadian rhythms in the system of rhythmic processes in the body; desynchronosis (desynchronization disorder) as an obligatory component of general adaptation syndrome; the wavelike nature of the adaptation process, as a general biological law, whereby this process, in any of its phases (alarm, resistance, exhaustion), operates in an oscillatory mode. Genes corresponding to peroxidase (RPP), glucosinolate biosynthesis (GIS), protein binding (CBP), myrosinase (RMA), napin (RSN), and ubiquitin (UBQ) were measured by qPCR. LP from day 24 and bulb samples collected at harvest were compared with RNA-seq data from material that was harvested, frozen, and analyzed after return to Earth. The results showed stable transcription in LP samples in GC but decreasing values in ISS samples during both grow-outs, possibly indicative of stress. SPGE results were similar between GC and ISS samples. However, the RNA-seq analyses showed different transcription profiles than SPGE or LP results, possibly related to localized sampling. RNA-seq of leaf samples showed greater variety than LP data, possibly because of different sampling times. RSN and RPP showed the lowest transcription regardless of method. Temporal analyses showed relatively small changes during plant development in space and in ground controls. This is the first study that compares developmental changes in space-grown plants with ground controls based on a comparison between RNA-seq and qPCR analyses.
