Abstract

The European Committee for Future Accelerators (ECFA) Early-Career Researcher (ECR) panel, which represents the interests of the ECR community to ECFA, presents in this document its initiatives and activities in the year 2023. This report summarises the process of the first big turnover in the panel composition at the start of 2023 and reports on the activities of the active working groups - either pursued from before or newly established. 3. Improving the Design of Linear Controllers for Homogeneous Platooning under Disturbances

Abstract

This paper addresses the problem of longitudinal platooning control of homogeneous vehicles subject to external disturbances, such as wind gusts, road slopes, and parametric uncertainties. Our control objective is to maintain the relative distance of the cars regarding their nearby teammates in a decentralized manner. Therefore, we proposed a novel control law to compute the acceleration commands of each vehicle that includes the integral of the spacing error, which endows the controller with the capability to mitigate external disturbances in steady-state conditions. We adopt a constant distance spacing policy and employ generalized look-ahead and bidirectional network topologies. We provide formal conditions for the controller synthesis that ensure the internal stability of the platoon under the proposed control law in the presence of constant and bounded disturbances affecting multiple vehicles. Experiments considering nonlinear vehicle models in the high-fidelity CARLA simulator environment under different disturbances, parametric uncertainties, and several network topologies demonstrate the effectiveness of our approach.

Abstract

Leishmaniasis is a complex disease caused by infection with different Leishmania parasites. The number of medications used for its treatment is still limited and the discovery of new drugs is a valuable approach. In this context, here we describe the in vitro leishmanicidal activity and the in silico interaction between trypanothione reductase (TryR) and (-)-5-demethoxygrandisin B from the leaves of Virola surinamensis (Rol.) Warb. The compound (-)-5-demethoxygrandisin B was isolated from V. surinamensis leaves, a plant found in the Brazilian Amazon, and it was characterized as (7R,8S,7R,8S)-3,4,5,3,4-pentamethoxy-7,7-epoxylignan. In vitro antileishmanial activity was examined against Leishmania amazonensis, covering both promastigote and intracellular amastigote phases. Cytotoxicity and nitrite production were gauged using BALB/c peritoneal macrophages. Moreover, transmission electron microscopy was applied to probe ultrastructural alterations, and flow cytometry assessed the shifts in the mitochondrial membrane potential. In silico methods such as molecular docking and molecular dynamics assessed the interaction between the most stable configuration of (-)-5-demethoxygrandisin B and TryR from L. infantum (PDB ID 2JK6). As a result, the (-)-5-demethoxygrandisin B was active against promastigote (IC50 7.0 µM) and intracellular amastigote (IC50 26.04 µM) forms of L. amazonensis, with acceptable selectivity indexes. (-)-5-demethoxygrandisin B caused ultrastructural changes in promastigotes, including mitochondrial swelling, altered kDNA patterns, vacuoles, vesicular structures, autophagosomes, and enlarged flagellar pockets. It reduced the mitochondria membrane potential and formed bonds with important residues in the TryR enzyme. The molecular dynamics simulations showed stability and favorable interaction with TryR. The compound targets L. amazonensis mitochondria via TryR enzyme inhibition.

Abstract

Constraining Beyond the Standard Model theories usually involves scanning highly multi-dimensional parameter spaces and check observable predictions against experimental bounds and theoretical constraints. Such task is often timely and computationally expensive, especially when the model is severely constrained and thus leading to very low random sampling efficiency. In this work we tackled this challenge using Artificial Intelligence and Machine Learning search algorithms used for Black-Box optimisation problems. Using the cMSSM and the pMSSM parameter spaces, we consider both the Higgs mass and the Dark Matter Relic Density constraints to study their sampling efficiency and parameter space coverage. We find our methodology to produce orders of magnitude improvement of sampling efficiency whilst reasonably covering the parameter space., Comment: 46 pages, 17 figures, 6 tables. Abstract

We propose a simple clockwork model of flavor which successfully generates the Standard Model flavor hierarchies from random order-one couplings. With very few parameters we achieve distributions of models in excellent agreement with observation. We explain in some detail the interpretation of our mechanism as random localization of zero modes in theory space. The scale of the vectorlike fermions is mostly constrained by lepton flavor violation with secondary constraints arising from rare meson decays., Comment: 27 pages + appendices
