Morphological and cytochemical characteristics of the blood cells of 14 free-living accipitrids seized or rescued in the Atlantic Forest biome of Rio de Janeiro were analyzed. This study was conducted using two cytochemical staining techniques, peroxidase (PER) and Sudan black (SBB). For morphological analysis, stained blood smears were evaluated under optical microscopy. To the best of the authors' knowledge, this was the first study on blood cell morphology in Parabuteo unicinctus and the first to cytochemically evaluate blood cells from this species and from Rupornis magnirostris. As for morphology, it was found that blood cells of the evaluated animals were similar to those of other acipitrids. Heterophils were the most prevalent leukocytes. Eosinophils and lymphocytes were also observed in expressive values. Only the eosinophils of P. unicinctus showed a positive reaction to PER e SBB. The specimens of R. magnirostris showed eosinophils with positive reaction only for SBB. The other leukocytes had negative results for both PER and SBB. It was concluded that the morphological and cytochemical characteristics of P. unicinctus and R. magnirostris blood cells are similar to those of other acipitrids, except for eosinophils, which differ biochemically. In this context, the present study of cytochemical techniques can provide an important database to be accessed in future research, including the influence of eosinophil composition on the function of these cells in predators. Se analizaron las características morfológicas y citoquímicas de las células sanguíneas de 14 aves rapaces silvestres, capturadas o rescatadas en el bioma de la Mata Atlántica de Río de Janeiro. Este estudio se realizó utilizando dos técnicas de tinción citoquímica, peroxidasa (PER) y Sudán negro B (SBB). Para la morfología, las muestras de sangre se tiñeron con colorante rápido y se evaluaron bajo microscopía de luz. Según el conocimiento de los autores, este fue el primer estudio sobre la morfología de las células sanguíneas en Parabuteo unicinctus y el primero en evaluar la citoquímica de las células sanguíneas de esta especie y de Rupornis magnirostris. Se encontró que las células sanguíneas de los animales evaluados eran similares a las de otras aves de la misma familia. Los heterófilos fueron los leucocitos más frecuentes, y los eosinófilos y linfocitos también se observaron en valores expresivos. Solo los eosinófilos de P. unicinctus mostraron una reacción positiva a PER y SBB. Rupornis magnirostris tuvieron eosinófilos positivos solo para SBB. El resto de leucocitos fueron negativos para PER y SBB. Nuestros resultados muestran que las características morfológicas y citoquímicas de las células sanguíneas de P. unicinctus y R. magnirostris son similares a las de otras aves de la misma familia; sin embargo, los eosinófilos difieren bioquímicamente. En este contexto, nuestros resultados de técnicas citoquímicas generaron datos importantes a la que acceder en futuras investigaciones, incluso sobre la influencia de la composición de eosinófilos en la función de estas células en los depredadores. Foram analisadas as características morfológicas e citoquímicas das células sanguíneas de 14 acipitrídeos de vida livre apreendidos ou resgatados no bioma Mata Atlântica do Rio de Janeiro. Este estudo foi conduzido utilizando duas técnicas de coloração citoquímica, a peroxidase (PER) e o sudan black B (SBB). Para análise morfológica, extensões sanguíneas coradas foram avaliadas em microscopia óptica. Até onde sabem os autores, este foi o primeiro estudo sobre morfologia de células sanguíneas em Parabuteo unicinctus e o primeiro a avaliar citoquimicamente as células sanguíneas dessa espécie e de Rupornis magnirostris. Quanto à morfologia constatou-se que as células sanguíneas dos animais avaliados foram similares às de outros acipitrídeos. Heterófilos foram os leucócitos mais prevalentes, sendo também observados eosinófilos e linfócitos em valores expressivos. Apenas os eosinófilos de P. unicinctus apresentaram reação positiva para PER e SBB. Os exemplares de R. magnirostris apresentaram eosinófilos com reação positiva somente para SBB. Os demais leucócitos tiveram resultados negativos para PER e SBB. Concluiu-se que as características morfológicas e citoquímicas das células sanguíneas de P. unicinctus e de R. magnirostris são semelhantes às de outros acipitrídeos, exceto os eosinófilos, que diferem bioquimicamente. Nesse contexto, o presente estudo das técnicas citoquímicas pode fornecer uma base de dados importante a ser acessada em futuras pesquisas, inclusive sobre a influência da composição dos eosinófilos na função dessas células em rapinantes.

The multivariate approaches, principal component analysis (PCA) and hierarchical cluster analysis (HCA), are applied to visualize these effects. Conventional energy-dispersive X-ray fluorescence equipment is used, where organic compounds produce intense scattering of the X-ray source. X-ray Raman processes, before obtained only for solid samples using synchrotron radiation, are indirectly visualized here through PCA scores and HCA cluster analysis, since they alter the Compton and Rayleigh scattering. As a result, their influences can be seen in known sample characteristics, as those associated with gender and melanin in dog hairs, and the differentiation in coconut varieties. Chemometrics has shown that, despite their complexity, natural samples can be easily classified.

Being considered a major public health problem, the canine reservoir derives from close and frequent contact between dogs and humans, and the fact that animals can present asymptomatic infection, despite a high degree of parasitism. The aim of the present study was to evaluate hematological, biochemical and clinical changes in dogs positive for Leishmania spp. by serological and molecular methods. Blood samples from 166 shelters dogs in the metropolitan region of Rio de Janeiro were collected. Hematological and serum biochemical analysis of the animals, as well as the detection of antibodies anti- Leishmania spp. by double-path immunochromatographic rapid test TR DDP - (Fiocruz?) and Indirect Immunofluorescence Reaction (RIFI) were performed. In addition, serologically positive animals were molecularly evaluated by polymerase chain reaction (PCR). The results showed that 10,84% (18/166) of the dogs presented seroreactivity by TR DDP, and these samples also underwent IFAT, in which 100% (18/18) of the animals were seroreagent. In PCR, based on the portion of the v7-v8 18S SSU rRNA gene, 16,66% (3/18) of shelter dogs were positive. No hematological, biochemical and clinical alterations were observed with the infection. No hematological, biochemical and clinical changes were observed with the infection. The results denote the importance of monitoring canine leishmnaniosis, especially in shelter dogs, so that control and prevention strategies can be implemented properly, minimizing the deleterious effects of this zoonosis and its spread to areas free of the disease Leishmaniose ? uma doen?a causada por protozo?rios do g?nero Leishmania e transmitida pelos vetores flebotom?neos hemat?fagos do g?nero Lutzomyia que infectam uma ampla variedade de hospedeiros mam?feros, incluindo c?es e humanos. The objective of the present study was to investigate the genetic diversity of geographically distinct isolates of 18S rRNA gene sequences of T. equi in naturally infected horses from the five Brazilian regions, characterize the hypervariable regions of the T. equi 18S rRNA sequences from this study and to elucidate the taxonomic relationships between T. equi isolates from Brazil and of the world. Equine blood samples were collected in the five regions of the country. The genomic DNA was extracted and the samples were submitted to real-time PCR for the selection of positive samples. Subsequently, the selected samples were submitted to a conventional PCR for amplification of the 18S rRNA gene (~1600 bp) of T equi, the products being sequenced. The sequences were aligned using ClustalW, and the alignment was submitted to the Maximum Likelihood reconstruction method with molecular distances estimated by the General Time Reversible Model, using 1000 replicates. From this alignment a similarity matrix was developed and the hypervariable regions were observed. Phylogenetic analyzes were performed in three different ways: using T. equi sequences from Brazil, using sequences from different species of Theileria and Babesia, and using T. equi sequences from all over the world. Twenty-three partial sequences of the 18S rRNA gene of T. equi were obtained, of which thirteen were unique variants of the gene in Brazil. The hypervariable gene regions V2, V4 and V8 were identified in the study samples, with V4 being the most variable. These hypervariable regions were confirmed by entropy analysis. Two distinct genotypes circulating in the Brazilian regions were identified. Only a single genotype was observed in the North region, while in the other regions both were observed. Samples were grouped into two clades (A and C), most of the study sequences being clustered in clade C. In another tree, all T. equi sequences grouped into group 1, different from the other theilerias and babesias, present in groups 2 and 3. In relation to the T. equi samples of the world, three clades were observed: A, B and C. This study adds important information on the genetic diversity of T. equi in horses from all regions of Brazil based on the 18S rRNA sequence. O objetivo do presente estudo foi investigar a diversidade gen?tica de isolados geograficamente distintos de sequ?ncias do gene 18S rRNA de T. equi em equinos naturalmente infectados provenientes das cinco regi?es do Brasil, caracterizar as regi?es hipervari?veis das sequ?ncias 18S rRNA de T. equi deste estudo e elucidar as rela??es taxon?micas entre os isolados de T. equi do Brasil e do mundo. As amostras de sangue dos equinos foram coletadas nas cinco regi?es do pa?s. O DNA gen?mico foi extra?do e as amostras foram submetidas a PCR em tempo real para a sele??o das amostras positivas. Posteriormente, as amostras selecionadas foram submetidas a PCR convencional para a amplifica??o do gene 18S rRNA (~1600 pb) de T. equi, sendo os produtos sequenciados. As sequ?ncias foram alinhadas utilizando o ClustalW, e o alinhamento foi submetido ao m?todo de reconstru??o da M?xima Verossimilhan?a com dist?ncias moleculares estimadas pelo General Time Reversible Model, utilizando 1000 repeti??es. Desse alinhamento foi desenvolvida uma matriz de similaridade e foram observadas as regi?es hipervari?veis. As an?lises filogen?ticas foram realizadas de tr?s formas distintas: utilizando sequ?ncias de T. equi do Brasil, utilizando sequ?ncias de diferentes esp?cies de Theileria e Babesia, e utilizando sequ?ncias de T. equi provenientes de todo o mundo. Foram obtidas vinte e tr?s sequ?ncias parciais do gene 18S rRNA de T. equi, sendo treze delas variantes ?nicas do gene no Brasil. As regi?es do gene hipervari?veis V2, V4 e V8 foram identificadas nas amostras do estudo, sendo a V4 a mais vari?vel. Essas regi?es hipervari?veis foram confirmadas pela an?lise de entropia. Foram identificados dois gen?tipos distintos circulantes nas regi?es do Brasil. Apenas na regi?o Norte foi observado um ?nico gen?tipo, enquanto que nas demais regi?es observaram-se ambos. As amostras se agruparam em dois clados (A e C), estando a maioria das sequ?ncias do estudo agrupadas no clado C. Em outra ?rvore, todas as sequ?ncias de T. equi se agrupam dentro do grupo 1, diferente das demais theilerias e babesias, presentes nos grupos 2 e 3. Em rela??o ?s amostras de T. equi do mundo, tr?s clados foram observados: A, B e C. Este estudo acrescenta importantes informa??es sobre a diversidade g?nica de T. equi em equinos de todas as regi?es do Brasil baseado na sequ?ncia 18S rRNA.

Pesquisa do Estado do Rio de Janeiro (FAPERJ) The aim of the present study was to characterize molecularly Ehrlichia canis using the membrane glycoprotein genes involved in the host parasite interaction (gp19, gp36, and p28), and also, the thermo unstable elongation factor (tuf) gene for characterization together with the 16S rDNA gene from dog blood samples, as well as to evaluate the distribution of E. canis in dogs from the mesoregions of Rio de Janeiro state. 267 blood samples from dogs from the state of Rio de Janeiro were obtained and tested by the Real-Time PCR (qPCR) targeting a fragment of 93 base pairs (bp) of 16S rDNA gene as a screening method for detection of the positive samples. Before evaluating the genetic diversity of E. canis, the samples were submitted to the molecular detection by 16S rDNA-qPCR. A total of 267 blood samples of dogs from the state of Rio de Janeiro were collected. Among the samples analyzed, 42.3% (n=113/267) were qPCR positive for 16S rDNA gene of E. canis. The average value of Cq observed in positive samples was 34,1 ? 5,1, ranging between 18 to 40 cycles. The detection limit of the qPCR was 10 copies of the plasmid per ?L containing a 16S rDNA gene from E. canis. The determination coefficient of the seven dilutions tested in the standard curve was 99.9% and efficiency de 95.7%. When performing gp19-PCR and gp36-PCR, 100% (n=113/113) and 5.3% (n=6/113) of the samples were positive, respectively. The six PCR positive samples for the 16S rDNA gene also amplified the p28 gene. Only one PCR positive sample for the three genes (gp19, gp36 and p28) in each of the six mesoregions were selected and subjected to analysis of amino acid and nucleotide sequences. The frequency in each mesoregion for the 113 animals positive for E. canis, by the 16S rDNA gene, was 59,29% (n=67/113) belonged to Metropolitan mesoregion, 13.27% (n=15/113) to South Fluminense, 15.04% (n=17/113) to Northern Fluminense, 5.3% (n=6/113) Center Fluminense, 4.42% (n=5/113) to Northwest Fluminense and 2.65% (n=3/113) to Coastal Baixada. The characterization based on the gp19 gene demonstrated that this gene is highly conserved and the samples showed 100% similarity with the Brazilian and worldwide isolates available in GenBank. Using the gp36 gene, it was possible to observe that there are polymorphic points between the sequences. Cluster analysis shows the formation of 3 clades, the sequences of the present study were allocated to the genogroup of the United States, suggesting a genetic similarity between the Brazilian and North American strains. In the protein analysis, all the samples presented repeated sequences (Tandem Repeat Sequences), with 11 replicates. Seven high entropy amino acid sites were observed, with variations of Hx: 0.5 to 0.67 generating mean entropy of 0.6 detected along the TRP36, suggesting a high degree of polymorphism. The ratio of non-silent to silent mutations using both genes showed significant genetic differences (p 0.05) with E. canis positivity. The investigation of gene differences in the isolates of E. canis becomes useful for understanding the parasite-host relationship; in addition, determining the distribution of this bacterium in the evaluated municipalities is advantageous for the updating of the epidemiological chain of canine ehrlichiosis. Os objetivos do presente estudo foram de caracterizar molecularmente Ehrlichia canis, utilizando os genes de glicoprote?nas de membrana (gp19, gp36, p28) e o gene Fator de Alongamento termo inst?vel (tuf) para caracteriza??o g?nica juntamente com o gene 16S rDNA das amostras de sangue de c?es naturalmente infectados, bem como avaliar a distribui??o de E. canis e fatores associados em c?es oriundos das mesorregi?es do estado do Rio de Janeiro. Foram testadas 267 amostras de sangue de c?es do estado do Rio de Janeiro, pela rea??o em cadeia da polimerase em tempo real (qPCR) com alvo em um fragmento de 93 pares de base (pb) do gene 16S rDNA para a detec??o de E. canis. Para realiza??o da caracteriza??o molecular, as amostras positivas nesta primeira an?lise foram testadas por PCRs convencionais para os genes gp19 (414 pb), gp36 (814 pb) e p28 (840 pb). Uma amostra de cada mesorregi?o amplificada para os genes gp19, gp36 e p28 foram purificadas e sequenciadas, utilizando-se o m?todo Sanger. Antes de avaliar a diversidade gen?tica de E. canis, as amostras foram submetidas ? detec??o molecular por 16S rDNA-qPCR. Foram coletadas 267 amostras de sangue de c?es do estado do Rio de Janeiro. Entre as amostras analisadas, 42,3% (n = 113/267) foram positivas para qPCR para o gene 16S rDNA de E. canis. O valor m?dio de Cq observado nas amostras positivas foi de 34,1 ? 5,1, variando entre 18 e 40 ciclos. O limite de detec??o do qPCR foi de 10 c?pias do plasm?deo por ?L contendo um gene 16S rDNA de E. canis. O coeficiente de determina??o das sete dilui??es testadas na curva padr?o foi de 99,9% e efici?ncia de 95,7%. Ao realizar PCR para os genes gp19 e gp36, 100% (n=113/113) e 5,3% (n=6/113) das amostras foram positivas, respectivamente. As seis amostras positivas para PCR para o gene 16S rDNA tamb?m amplificaram o gene p28. Apenas uma amostra PCR positiva para os tr?s genes (gp19, gp36 e p28) em cada uma das seis mesorregi?es foi selecionada e submetida ? an?lise de sequ?ncias de amino?cidos e nucleot?deos. A freq??ncia em cada mesorregi?o para os 113 animais positivos para E. canis, pelo gene 16S rDNA, foi de 59,29% (n=67/113) para mesorregi?o Metropolitana, 13,27% (n =15/113) para regi?o Sul Fluminense, 15,04% (n=17/113) para mesorregi?o Norte Fluminense, 5,3% (n= 6/113) para mesorregi?o Centro Fluminense, 4,42% (n=5/113) na mesorregi?o Noroeste Fluminense e 2,65% (n= 3/113) na Baixada Litor?nea. A caracteriza??o baseada no gene gp19 demonstrou que este gene ? altamente conservado e as amostras apresentaram 100% de similaridade com as sequ?ncias brasileiros e mundiais dispon?veis no GenBank. Utilizando o gene gp36, foi poss?vel observar que h? pontos polim?rficos entre as sequ?ncias. Na an?lise de agrupamentos nota-se a forma??o de 3 clados, as sequ?ncias do presente estudo alocaram-se no genogrupo dos Estados Unidos, sugerindo similaridade gen?tica entre as sequ?ncias brasileiras e norte-americanas. Na an?lise de prote?nas, todas as amostras apresentaram sequ?ncias repetidas (?Tandem Repeat Sequence?), com 11 repeti??es. Foram observados sete s?tios de amino?cidos de alta entropia, com varia??es de Hx: 0,5 a 0,67 gerando uma m?dia de entropia de 0,6 detectados ao longo de TRP36, o que sugere um alto grau de polimorfismo. A raz?o das muta??es n?o-silenciosas sobre as silenciosas, utilizando ambos genes mostrou diferen?as gen?ticas significativas (p0,05) com a positividade de Ehrlichia canis nas sequ?ncias analisadas. Pesquisa do Estado do Rio de Janeiro (FAPERJ, Brasil) Cats are the main reservoirs of Bartonella henselae (Bh), B. clarridgeiae (Bc) and B. koehlerae (Bk), which can be transmitted to humans through scratches or bites and cause disease. In Brazil there are very few studies, especially focusing on molecular detection of Bartonella species, as well as their impact in veterinary clinical practice and public health. The aims of this study were to detect Bartonella DNA in blood of shelter cats from metropolitan area of Rio de Janeiro State and their ectoparasites, and, in addition, to evaluate associated risk factors, clinical signs, and hematological abnormalities. Two-hundred and eight cats were sampled and divided into groups: G1, those in the Rio de Janeiro municipality (n=68) and G2, those in the Belford Roxo, Duque de Caxias, Guapimirim, Itagua?, Mesquita, Nova Igua?u, S?o Gon?alo, and Serop?dica municipalities (n=140). Cats were physically examined and inspected for ectoparasites presence. By appropriate restrainment, blood samples were aseptically obtained by cephalic phlebotomy, transferred into sterile tubes containing EDTA anticoagulant.Bartonella DNA screening in EDTA-blood samples and ectoparasites was based on the genes gltA and ITS (16S-23S). The association between bacterial infection, hematological data, clinical signs, and risk factors was assessed by Fisher's exact test or chi-square test. In the present study, eighty-three cats (39.9%) were positive for Bartonella species, of which 22.1% and 48.6% were from G1 and G2, respectively. Such discrepancy may be explained by differences in local hygiene conditions and preventive flea control. Bacterial DNA was detected in fleas and in the blood of cats infested by positive fleas, which were twice as likely to become infected (p>0.05). This finding highlights the importance of these ectoparasites in the transmission of pathogens among cats. Bh and Bc DNA were detected in cat fleas, while Bh, Bc and Bk in blood from bacteremic cats. Cats with blood collected between March and June months were significantly more frequently infected (p0,05). The transmission of these agents to man and animals occurs mainly by arthropod vectors. The participation of domestic felines and wild reservoirs has not yet been defined, with few studies on hemoparasitosis affecting these animals. The present study had as objective to investigate the presence of infection by Anaplasmataceae family agents in domestic felines and possums (Didelphis sp.) of the state of Rio de Janeiro, using molecular methods. We sought to sequence the PCR positive samples for validation of the same and to compare them with those deposited in Genbank, as well as to develop new diagnostic methods aiming at fast and high yield results, such as "deep sequencing", used in metagenomic assays. The aim of this study was to determine which hematological changes (hemogram, hematological morphology and hemoparasite inclusions) are more frequent and which associated factors are related to these in two distinct cat populations in the Metropolitan Region of Rio de Janeiro. Two populations of domestic cats, wandering cats (n = 105) and domiciled cats (n = 178) from the Metropolitan region of Rio de Janeiro were studied. In dogs, hemoplasma infection can cause hemolytic anemia in the acute phase, while in the chronic phase the signs are generally inapparent, and immunosuppression can trigger the acute disease. The present study aimed to detect molecularly the presence of hemotrophic mycoplasma agents in domestic dogs from the urban and rural areas of Aragua?na, Tocantins and, in addition, to correlate hematological alterations and factors associated with natural infection. In the analysis of the results, 2.9% (3/105) of the dogs sampled from the urban area and 24.49% (24/99) from the rural area were positive for Mycoplasma spp. by the Real Time Polymerase Chain Reaction (qPCR). The frequency of hemoplasmas species infection by PCR using species-specific oligonucleotides was 1.9% (2/105) and 13.26% (13/99) for Mycoplasma haemocanis in the urban and rural areas, respectively. No positive dogs were observed for ?Candidatus Mycoplasma turicensis? and ?Candidatus Mycoplasma haematoparvum?/?Candidatus Mycoplasma haemominutum? in both areas studied. Regarding hematological alterations, there was no statistical association with the infection by Mycoplasma spp. Similarly, among the analyzed variables (age, gender, racial pattern, site cleanliness and housing), no statistical correlation was observed in either urban or rural areas. This is the first report of the molecular detection of Mycoplasma haemocanis in dogs from Tocantins and opens up prospects for future studies in this group of hemotrophic agents.
