We evaluated the epidemiological, hematological, and pathological data of Leishmania spp., Toxoplasma gondii, Platynosomum illiciens, feline immunodeficiency virus (FIV), and feline leukemia virus (FeLV) infections and the coinfections in stray cats of an endemic area for leishmaniasis. The diagnosis was performed by serological tests and necropsy. We described gross lesions and histopathological findings. We used immunohistochemistry and chromogenic in situ hybridization for L. infantum detection. We found infection in 27 out of 50 sampled cats, among them, 14 presented coinfections. A strong correlation between splenomegaly and lymphadenomegaly with FeLV, and an association between hepatic lesions and cachexia with parasitism due to P. illiciens were observed. Moreover, we found a significant increase in the monocyte count in the FeLV-infected and a decrease in the red blood cell count in the FIV-infected animals. Amastigote forms of Leishmania spp. and tissue changes were detected in lymphoid organs of an animal coinfected with P. illiciens, T. gondii, and FIV. Polyparasitism recorded in stray cats of the Brazilian Midwest should be considered in effective control strategies for public health diseases. Moreover, stray cats of Campo Grande may be a source of infection of FIV, FeLV and P. illiciens for populations of domiciled cats. Resumo Foi avaliada a epidemiologia, hematologia e patologia das infecções por Leishmania spp., Toxoplasma gondii, Platynosomum illiciens, vírus da imunodeficiência felina (FIV) e vírus da leucemia felina (FeLV) e das coinfecções em gatos não domiciliados em uma área endêmica para leishmaniose. O diagnóstico foi realizado por exames sorológicos e necropsia. Foram descritas lesões macroscópicas e achados histopatológicos. Usaram-se imuno-histoquímica e hibridização cromogênica in situ para detecção de L. infantum. Encontrou-se infecção em 27 dos 50 gatos amostrados, dentre eles, 14 apresentavam coinfecções. Foi observada forte correlação entre esplenomegalia e linfadenomegalia com FeLV, e associação entre lesões hepáticas e caquexia com parasitismo por P. illiciens. Além disso, foi encontrado um aumento significativo na contagem de monócitos nos animais infectados pelo FeLV e uma diminuição na contagem de hemácias nos animais infectados pelo FIV. Formas amastigotas de Leishmania spp. e alterações teciduais foram detectadas em órgãos linfoides de um animal coinfectado com P. illiciens, T. gondii e FIV. O poliparasitismo registrado em gatos errantes do Centro-Oeste brasileiro deve ser considerado nas estratégias de controle de zoonoses. Além disso, gatos errantes de Campo Grande podem ser fontes de infecção de FIV, FeLV e P. illiciens para populações de gatos domiciliados.

Arthropod-borne pathogens are medically important because of their ability to cause diseases in their hosts. The purpose of this study was to detect the occurrence of Ehrlichia spp., piroplasmids and Hepatozoon spp. in dogs with anemia and thrombocytopenia in southern Brazil. EDTA-whole blood was collected from 75 domestic dogs presenting anemia or/and thrombocytopenia from Guarapuava, state of Paraná, Brazil. DNA samples were subjected to conventional PCR assays for Ehrlichia spp. (dsb), piroplasmids (18S rRNA) and Hepatozoon spp. (18S rRNA), followed by sequencing and phylogenetic analyses. Among the 75 dogs, one (1.33%) was positive for Hepatozoon sp. and six (8%) were positive for piroplasmids in 18S rRNA cPCR assays. None of the dogs showed positive results in Ehrlichia spp.-cPCR targeting dsb gene. The phylogenetic analyses revealed that three piroplasm sequences were clustered with Rangellia vitalii, while one sequence was grouped with B. vogeli. The only sequence obtained from Hepatozoon spp.-PCR protocol was pooled with H. canis. Therefore, there is urgent need for differential molecular diagnosis of the two piroplasm species cited as etiological agents in clinical cases of canine hemoparasitic diseases, given the higher pathogenic potential of R. vitalii than of B. vogeli.

Resumo Agentes transmitidos por artrópodes têm grande importância na medicina veterinária devido à sua capacidade de causar doenças graves em seus hospedeiros. O presente estudo objetivou investigar a ocorrência de três patógenos transmitidos por vetores, Ehrlichia canis, Rangelia vitalii e Hepatozoon canis, em cães na região sul do Brasil. Foram coletadas amostras de sangue total de 75 cães domésticos que apresentavam anemia e/ou trombocitopenia, em Guarapuava, Paraná, Brasil. As amostras de DNA foram submetidas à técnica de PCR convencional para E. canis (dsb), piroplasmídeos (18S rRNA) e Hepatozoon spp. (18S rRNA), seguida de sequenciamento e análises filogenéticas. Das 75 amostras, uma (1,33%) foi positiva para Hepatozoon spp. e seis (8%) foram positivas para Babesia spp. Nenhuma amostra mostrou resultados positivos para Ehrlichia spp. utilizando a detecção pelo gene dsb. As análises filogenéticas revelaram que três sequências obtidas foram agrupadas no mesmo clado que R. vitalii , enquanto uma foi agrupada juntamente com B. vogeli. A única sequência obtida pelo protocolo de PCR para Hepatozoon spp. foi agrupada juntamente com H. canis. Assim, é justificada necessidade de diferenciação das espécies de piroplasmas, através do diagnóstico molecular, como agentes etiológicos nos casos clínicos de hemoparasitose canina, considerando o potencial patogênico de R. vitalii quando comparado à B. vogeli.

In fact, with the exception of Bartonella spp. and hemoplasmas, FVBP in cats has not been clearly determined in Brazil yet. The present study aimed at identifying, by using molecular methods, the presence of FVBP in three cats showing non-specific clinical signs and inclusions suggestive of hemoparasites in blood smears. Cytauxzoon felis, ‘Candidatus Mycoplasma haemominutum’, Ehrlichia sp. closely related to Ehrlichia canis, and Anaplasma sp. closely related to Anaplasma phagocytophilum were detected in blood samples from two out of three sampled cats. Both cats positive for multiple FVBP did not show hematological and biochemical abnormalities. The present work emphasizes the need for molecular confirmation of co-infection by multiple FVBP in cats presenting non-specific clinical signs and inclusions resembling hemoparasites in blood smears. O presente estudo tem como objetivo investigar a ocorrência de agentes transmitidos por artrópodes (agentes Anaplasmataceae, Bartonella spp., mycoplasmas hemotróficos, Rickettsia spp., Hepatozoon spp. e piroplasmideos) em animais selvagens, cães domésticos e seus respectivos ectoparasitos, amostrados na região do Pantanal sul matogrossense, por meio de métodos sorológicos e moleculares. Para tal, 31 Nasua nasua, 78 Cerdocyon thous, sete L. pardalis, 42 cães, 110 roedores e 30 marsupiais foram capturados. Os carrapatos recolhidos (1582) dos animais pertenciam às espécies Amblyomma sculptum, Amblyomma parvum, Amblyomma ovale, Amblyomma tigrinum, Rhipicephalus microplus, Rhipicephalus sanguineus sensu lato e Amblyomma auricularium. Adicionalmente, 80 pulgas Polygenis (Polygenis) bohlsi bohlsi foram recolhidas. Quatorze (17,9%) C. thous, sete (16,6%) cães e um (3,2%) N. nasua mostraram-se soropositivos (títulos≥80) para Ehrlichia canis, com títulos de anticorpos variando de 80 a 1280. Nenhum animal mostrou-se soropositivo Anaplasma phagocytophilum. Nove cães, dois C. thous, um N. nasua, oito roedores, cinco marsupiais e um pool de pulgas P. (P.) b. bohlsi mostram-se positivos para Ehrlichia spp. Todos os cães e o pool de pulgas P. (P.) b. bohlsi positivos para Ehrlichia spp. foram também positivos em para o ensaio em tempo real quantitativo (qPCR) para E. canis, baseado no gene dsb. Sete N. nasua, dois C. thous, um L. pardalis, quatro roedores, três marsupiais, 15 carrapatos A. sculptum, dois A. ovale, dois A. parvum e um pool de larvas de Amblyomma spp. foram positivos para Anaplasma spp. Co-positividade ou co-soropositividade para Ehrlichia spp. e Anaplasma spp. foi observada em dois cães, um N. nasua, um C. thous e dois marsupiais. Trinta e cinco roedores selvagens e três pools de pulgas P. (P.) b. bohlsi, mostraram-se positivos nos ensaios moleculares para Bartonella spp. A análise filogética revelou que pelo menos dois genótipos diferentes estão circulando entre os roedores amostrados no bioma Pantanal. Os resultados parciais sugerem que a pulga P. (P.) b. bohlsi possa estar atuando como um possível vetor de Bartonella nesta região. Vinte e quatro N. nasua, três C. thous, dois cães e um roedor mostraram-se positivos para Mycoplasma spp. A análise filogenética apontou que os cães, C. thous e N. nasua aparentam estar sendo parasitados pela mesma espécie de Mycoplasma spp., filogenéticamente relacionada a M. haemocanis/M. haemofelis e que provavelmente um novo genótipo de Mycoplasma spp. está circulando entre os N. nasua e C. thous amostrados no bioma Pantanal. Vinte e sete (64,2%) cães, 59 (7,6%) C. thous e seis (85,7%) L. pardalis mostraram-se soropositivas para pelo menos uma espécie de Rickettsia. Para 17 (40,4%) cães, 33 (42,3%) C. thous e dois (33,3%) L. pardalis, foram observadas reações homólogas para Rickettsia amblyommatis. Cento e dezesseis carrapatos (93 A. parvum, 14 A. sculptum, três A. auricularim e seis pools de larvas de Amblyomma) e uma amostra de sangue obtida de C. thous, mostraram-se postivas nos ensaios para as espécies de Rickettsia do grupo da febre maculosa. As amostras seqüenciadas obtidas dos carrapatos A. parvum, mostraram-se filogenéticamente relacionadas com 'Candidatus Rickettsia andeanae'. Uma alta ocorrência de Hepatozoon foi encontrada nos carnívoros amostrados (C. thous [91,02%], cães [45,23%], N. nasua [41,9%] e L. pardalis [71,4%]), porém nenhum artrópode mostrou-se positivo. Adicionalmente, 24 roedores e um marsupial também mostraram-se positivos para Hepatozoon spp. Com base na análise filogenética, os C. thous, L. pardalis, N. nasua, cães, roedores e marsupiais aparentam não estar sendo parasitados pela mesma espécie de Hepatozoon spp. Sete cães, um C. thous, cinco L. pardalis, três N. nasua, seis roedores, oito carrapatos A. parvum, dois A. sculptum e um A. ovale mostraram-se positivos para os ensaios PCR de piroplasmídeos. Genótipos filogeneticamente relacionados com Babesia canis vogeli foram detectados em seis cães e cinco roedores. Genótipos filogeneticamente relacionados com Babesia caballi foram detectados em um C. thous, um cão, um carrapato A. ovale e um A. sculptum e genótipos relacionados com Babesia bigemina e Babesia bovis foram detectados em quatro carrapatos A. parvum. Quatro sequências obtidas de A. parvum, três N. nasua e um roedor mostraram-se filogeneticamente relacionadas com Theileria equi. Por fim, Cytauxzoon spp. foi detectado em quatro L. pardalis .O presente trabalho mostra, portanto, a ocorrência de agentes transmitidos por artrópodes vetores em animais selvagens e domésticos no Panatanal sul matogrossense. The diseases transmitted by arthropods have been recently studied in Brazilian wildlife, which can act as hosts for vectors and pathogens, many of which have zoonotic potential. The present work aimed to investigate the occurrence of tick-borne agents (Anaplasmataceae agents, Bartonella spp., hemotropic mycoplasmas, Rickettsia spp., Hepatozoon spp. and piroplasms) in wild animals, domestic dogs and their respective ectoparasites, in southern Pantanal region, central-western Brazil, by serological and molecular assays. For this reason, 31 Nasua nasua, 78 Cerdocyon thous, seven L. pardalis, 42 dogs, 110 rodents and 30 marsupials were captured. The ticks collected (1582) from animals belonged to the species Amblyomma sculptum, Amblyomma parvum, Amblyomma ovale, Amblyomma tigrinum, Rhipicephalus microplus, Rhipicephalus sanguineus sensu lato and Amblyomma auricularium. Additionally, 80 Polygenis (Polygenis) bohlsi bohlsi fleas were collected. Overall, 14 (17.9%) C. thous, seven (16.6%) dogs and one (3.2%) N. nasua were seroreactive (titer≥80) to Ehrlichia canis, with titers ranging from 80 to 1280. No animal showed to be seroreactive for A. phagocytophilum antigen. Nine dogs, two C. thous, one N. nasua, eight rodents, five marsupials and one P. (P.) b. bohlsi pool were positive for Ehrlichia spp. All positive dogs and the only P. (P.) b. bohlsi pool positive for Ehrlichia spp. were also positive in specific E. canis-qPCR based on dsb gene. Seven N. nasua, two dogs, one C. thous, one L. pardalis, four rodents, three marsupials, 15 A. sculptum, two A. ovale, two A. parvum and one Amblyomma larvae pool were positive for Anaplasma spp. Co-positivity or co-seropositivity for Ehrlichia spp. and Anaplasma spp. was observed in two dogs, one N. nasua, one C. thous and two marsupials. Thirty-five rodents and three P. (P.) b. bohlsi flea pools, showed to be positive to Bartonella spp. in the molecular assays. The phylogenetic analysis revealed that at least two different genotypes are circulating among the rodents sampled in the Pantanal biome. Partial results suggest that the flea P. (P.) b. bohlsi may be acting as a possible Bartonella vector in this region. Twenty-four N. nasua, three C. thous, two dogs and one rodent, were positive for Mycoplasma spp. Phylogenetic analysis indicated that dogs, C. thous and N. nasua appear to share the same Mycoplasma spp. species, closely related to M. haemocanis / M. haemofelis and probably a new genotype of Mycoplasma spp. is circulating among the N. nasua and C. thous sampled in the Pantanal biome. Overall, 27 (64.2%) dogs, 59 (75.6%) C. thous and six (85.7%) L. pardalis were seroreactive to at least one Rickettsia species. For 17 (40.4%) dogs, 33 (42.3%) C. thous, and two (33.3%) L. pardalis, homologous reactions to Rickettsia amblyommatis were suggested. One hundred and sixteen ticks (93 A. parvum, 14 A. sculptum, three A. auricularim and six Amblyomma larvae pools) and one blood sample obtained from C. thous showed positive results for Rickettsia spotted fever group species. The sequenced samples obtained from A. parvum ticks showed to be closely related to 'Candidatus Rickettsia andeanae'. A high occurrence of Hepatozoon was found in carnivores (C. thous [91.02%], dogs [45.23%], N. nasua [41.9%] and L. pardalis [71.4%]), however no arthropod showed positive results. Additionally, twenty-four rodents and a marsupial also showed positivity to Hepatozoon spp. Based on phylogenetic analysis, C. thous, L. pardalis, N . nasua, dogs, rodents and marsupials appear not to share the same of Hepatozoon species. Seven dogs, one C. thous, five L. pardalis, three N. nasua, six rodents, eight A. parvum, two A. sculptum and one A. ovale were positive for piroplasmids-PCR assays. Genotypes closely related to Babesia vogeli were detected in six dogs and five rodents. While genotypes closely related to Babesia caballi were detected in one C. thous, one dog, one A. ovale and one A. sculptum, genotypes closely related to Babesia bigemina and Babesia bovis were detected in four A. parvum ticks. Four sequences obtained from A. parvum, three N. nasua and one wild rodent were closely related to Theileria equi. Lastly, Cytauxzoon spp. was detected in four L. pardalis. Recently, an increasing number of Bartonella species have been emerged to cause human diseases. Among animal reservoirs for Bartonella spp., bats stand out due to their high mobility, wide distribution, social behaviour and long‐life span. Although studies on the role of vampire bats in the epidemiology of rabies have been extensively investigated in Latin America, information on the circulation and genetic diversity of Bartonella species in these bat species is scarce. In the present work, 208 vampire bats, namely Desmodus rotundus (the common vampire bat; n = 167), Diphylla ecaudata (the hairy‐legged vampire bat; n = 32) and Diaemus youngii (the white‐winged vampire bat; n = 9) from 15 different states in Brazil were sampled. DNA was extracted from liver tissue samples and submitted to real‐time PCR (qPCR) and conventional PCR (cPCR) assays for Bartonella spp. targeting five genetic loci, followed by phylogenetic and genotype network analyses. Fifty‐one out of 208 liver samples (24.51%) were positive for Bartonella DNA in the ITS real‐time PCR assay [40 (78.43%) of them were from D. rotundus from 11 states, and 11 (21.57%) samples from D. ecaudata from three states. Eleven genotypes were found for each gltA and rpoB genes. Several ITS sequences detected in the present study clustered within the lineage that includes B. bacilliformis and B. ancachensis. The Bayesian phylogenetic inference based on the gltA gene positioned the obtained sequences in six different clades, closely related to Bartonella genotypes previously detected in D. rotundus and associated ectoparasites sampled in Latin America. On the other hand, the Bartonella rpoB genotypes clustered together with the ruminant species, B. schoenbuchensis and B. chomelii. The present study describes for the first time the molecular detection of Bartonella spp. in D. ecaudata bats. It also indicates that Bartonella spp. of vampire bats are genetically diverse and geographically widespread in Brazil. [ABSTRACT FROM AUTHOR]

Swine hemoplasmosis is frequently associated with hemolytic anemia, disgalacty, infertility and immunosuppression, and it results in significant economic losses. This study investigates the occurrence of M. suis in non-technified swine herds in the northeastern region of Brazil using quantitative PCR (qPCR) based on the 16S rRNA gene. Between March and August 2013, blood samples from 147 swine were collected during slaughter in the city of Mossoró, state of Rio Grande do Norte, northeastern Brazil. One hundred and twelve samples (76.19%) were positive for M. suis by qPCR assays. The range of Cqs and quantification (copies of a M. suis-16S rRNAl gene fragment/μL) was 20.86-37.89 and 1.64×101-6.64×107, respectively. One can conclude that M. suis infection have high occurrence (76,19%) in non‑technified swine-rearing systems in Mossoró in the state of Rio Grande do Norte, Brazil. Different parasites that commonly occur concomitantly can influence one another, sometimes with unpredictable effects. We evaluated pathological aspects of dogs naturally co-infected with Leishmania infantum and Ehrlichia canis. The health status of the dogs was investigated based on histopathological, hematological and biochemical analyses of 21 animals infected solely with L. infantum and 22 dogs co- infected with L. infantum and E. canis. The skin of both groups showed chronic, predominantly lymphohistioplasmacytic inflammatory reaction. The plasmacytosis in the lymphoid tissues was likely related with the hypergammaglobulinemia detected in all the dogs. The disorganization of extracellular matrix found in the reticular dermis of the inguinal region and ear, characterized by the substitution of thick collagen fibers for thin fibers, was attributed to the degree of inflammatory reaction, irrespective of the presence of parasites. In addition, the histopathological analysis revealed that twice as many dogs in the co-infected group presented Leishmania amastigotes in the ear skin than those infected solely with Leishmania, increasing the possibility of becoming infected through sand fly vectors. Our findings highlight the fact that the health of dogs infected concomitantly with L. infantum and E. canis is severely compromised due to their high levels of total plasma protein, globulins, alkaline phosphatase and creatine kinase, and severe anemia. A infecção simultânea por parasitas de diferentes espécies pode resultar em alterações imprevisíveis. O presente estudo avaliou a patologia de cães naturalmente coinfectados por Leishmania infantum e Ehrlichia canis. A saúde dos cães foi investigada pelas análises histopatológicas, hematológicas e bioquímicas de 21 cães infectados somente por L. infantum e 22 cães coinfectados por L. infantum e E. canis. Observou-se uma reação inflamatória crônica, predominantemente linfohistioplasmocítica, na pele dos dois grupos. A plasmocitose, encontrada nos tecidos linfóides, provavelmente estava relacionada com a hipergamaglobulinemia observada em todos os cães amostrados. A desorganização da matriz extracelular da derme da região inguinal e da orelha, demonstrada pela substituição das fibras de colágeno espessas por fibras finas, foi relacionada com o grau de reação inflamatória, independente da presença de parasitas. Ainda, observamos duas vezes mais animais do grupo coinfectado apresentando formas amastigotas na pele de orelha pela histopatologia comparado ao número de cães infectados apenas por Leishmania, tornando-os desta forma mais infectivos aos vetores. Nossos resultados ressaltam que a saúde de cães coinfectados estava severamente comprometida devido aos altos níveis de proteína plasmática total, globulinas, fosfatase alcalina, creatina quinase e anemia acentuada.

An investigation was made into the occurrence of antibodies to Toxoplasma gondii, Leishmania infantum and Neospora caninum in 151 domestic cats, based on the indirect fluorescent antibody test (IFAT). There are two distinct lineages of ticks, Rhipicephalus sanguineus, in South America: tropical and temperate lineages. Only the tropical lineage is recognized as competent vector for Ehrlichia canis. The epidemiological data of canine monocytic ehrlichiosis is congruent with the distribution of the two lineages of R. sanguineus. Herein, we report the infection of R. sanguineus (tropical lineage) cell cultures with E. canis, after cryopreservation. R. sanguineus (tropical lineage) cell identity was confirmed by sequencing using a 16S rDNA gene fragment. Tick cell cultures were prepared in L-15B medium supplemented with 10%, 15%, and 20% Fetal Bovine Serum (FBS), and 10% of Tryptose Phosphate Broth (TPB). Cell cultures developed better at the concentration of 20% of FBS. Cultures in the fifth harvest (approximately 7 months later) were selected for the first infections. Optimal R. sanguineus cell growth and adhesion was observed (5.0 × 10
6 cells/mL, and the population doubling time every 57 h). Once infected with E. canis, the cultures were maintained in L-15B medium supplemented with 2% and 5% of FBS fortified with iron and 10% TPB. Infected cells were also cryopreserved. DNA was extracted from infected and noninfected cells and analyzed using quantitative real-time PCR targeting the E. canis-dsb gene. Primary culture of the fifth passage was infected by E. canis and it maintained the pathogen for at least 40 days before partial cell destruction. Subcultures of infected cells (fresh and cryopreserved cultures) onto new tick cell cultures were successful. The E. canis infection was confirmed by real-time PCR and light and transmission electron microscopy. The R. sanguineus (tropical lineage) cells infected with E. canis successfully infected new tick cell cultures, showing that these cells could be an alternative substrate for maintenance of this pathogen. O presente estudo teve como objetivo realizar o diagnóstico sorológico e molecular de parasitos das espécies Leishmania chagasi, Ehrlichia canis e Babesia canis em amostras de soro e de baço, respectivamente. Foram utilizados 60 cães naturalmente infectados, eutanasiados no Centro de Controle de Zoonoses do Município de Campo Grande, Mato Grosso do Sul. Pela Reação de Imunofluorescência Indireta (RIFI), 60 (100%), 39 (65%) e 49 (81%) dos 60 animais amostrados foram sororeagentes frente aos antígenos de L.chagasi, E.canis e B.canis respectivamente. Também foi realizado o ensaio Imunoenzimático Indireto (ELISA-teste) para detecção de anticorpos da classe IgG anti-L. chagasi, onde sessenta cães (100%) mostraram-se soropositividade. Cinquenta e quatro cães (90%) mostraram-se positivos na reação em cadeia pela polimerase (PCR) para o gênero Leishmania sp e destes cinquenta e três animais, 53 (88%) foram positivos para o complexo L.Donovani. Vinte e sete animais (45%) foram positivos para Ehrlichia e destes animais todos foram positivos para E.canis pelo nested PCR e 2 (3,33%) cães foram PCR positivos para Babesia sp. Os amplicons foram confirmados por seqüenciamento e os DNAs mostraram 99% de similaridade genética para L.chagasi, E.canis e B.canis. Este estudo mostrou que os cães residentes em Campo Grande/MS, podem albergar mais de um hemoparasita, As co-infecções por agentes patogênicos intracelulares de células do sistema monocítico fagocitário, como é o caso da Ehrlichia e Leishmania, associadas ou não a Babesia... Among emerging diseases caused by arthropods in dogs stand out infections by agents of the species Leishmania chagasi, transmitted by sandflies, Ehrlichia canis and Babesia canis transmitted by ticks of the species Rhipicephalus sanguineus. The present study aims to perform serological and molecular diagnosis of parasites of the species Leishmania chagasi, Ehrlichia canis and Babesia canis in samples of sera and spleen, respectively. A total of 60 naturally infected dogs were euthanized at the Zoonosis Control Center of the city of Campo Grande, Mato Grosso do Sul Through Immunofluorescence Assay (IFA), sixty (100%), 39 (65%) and 49 (81%) of the 60 animals sampled were reactive serum against the antigens L.chagasi, E.canis and B. canis, respectively. We also performed indirect immunosorbent assay (ELISA test) for detection of IgG antibodies anti-L. chagasi, where sixty dogs (100%) were seropositive. Fifty-four dogs (90%) were positive by PCR for the genus Leishmania, and of these fifty and three animals, 53 (88%) were positive for the complex L.Donovani. Twenty-seven dogs (45%) were positive for Ehrlichia sp and these animals were all positive for E.canis by nested PCR and two dogs (3%) were PCR positive for Babesia sp. The amplicons were confirmed by DNA sequencing and showed 99% genetic similarity to L.chagasi, E.canis and B. canis. This study showed that dogs living in Campo Grande / MS, can accommodate more than one haemoparasite. The co-infections by intracellular pathogens monocytic phagocyte system cells, as is the case with Ehrlichia and Leishmania, associated or not with Babesia (which multiplies in red blood cells), causing serious damage to the health of animals. OBJETIVO: Descrever o perfil clínico e epidemiológico e a prevalência da coinfecção tuberculose/HIV na Unidade Regional de Saúde do Tocantins, que envolve 14 municípios no estado do Maranhão. MÉTODOS: Estudo epidemiológico descritivo baseado em dados secundários das fichas individuais de tuberculose do Sistema Nacional de Informação de Agravos de Notificação. Foram incluídos todos os casos notificados de coinfecção tuberculose/HIV, por município de residência, no período entre janeiro de 2001 e dezembro de 2010. RESULTADOS: Foram notificados 1.746 casos de tuberculose no distrito. Dos pacientes testados para HIV, 100 eram coinfectados. equivalendo a uma prevalência de 39%. Dos coinfectados, 79% eram do sexo masculino, 42% eram de cor parda, 64% tinham idade entre 20 e 40 anos, 31% tinham até quatro anos de estudo, e 88% residiam em Imperatriz. A forma clínica predominante foi a pulmonar (87%), e 73% eram casos novos. Dos coinfectados, 27% apresentaram resultados positivos na baciloscopia de escarro e 89% tinham imagem sugestiva de tuberculose na radiografia do tórax. A cultura de escarro foi realizada em apenas 7% dos casos. CONCLUSÕES: Evidenciou-se que a situação clínica e epidemiológica da coinfecção tuberculose/HIV ainda é um grande problema de saúde pública no sudoeste do Maranhão e impõe uma maior articulação entre os programas de controle de tuberculose e de doenças sexualmente transmissíveis/AIDS. Além disso, são necessários o compromisso e o envolvimento político dos gestores e profissionais de saúde no planejamento de ações e serviços de saúde. OBJECTIVE: To describe the clinical and epidemiological profile, as well as the prevalence, of tuberculosis/HIV co-infection in the Regional Health District of Tocantins, which serves 14 cities in the state of Maranhão, Brazil. METHODS: This was a descriptive epidemiological study based on secondary data obtained from individual tuberculosis reporting forms in the Brazilian Case Registry Database. We included all reported cases of tuberculosis/HIV co-infection, by city, between January of 2001 and December of 2010. RESULTS: In the district, 1,746 cases of tuberculosis were reported. Of those tested for HIV, 100 had positive results, which corresponded to a tuberculosis/HIV co-infection prevalence of 39%. Of the co-infected patients, 79% were male, 42% were Mulatto, and 64% were in the 20- to 40-year age bracket, 31% had had < 4 years of schooling, and 88% resided in the city of Imperatriz. Cases of pulmonary tuberculosis and new cases of tuberculosis predominated (in 87% and 73%, respectively). Of the co-infected patients, 27% had positive sputum smear microscopy results, and 89% had chest X-ray findings suggestive of tuberculosis. Sputum culture was performed in only 7% of the cases. CONCLUSIONS: Our results show that, because of its clinical and epidemiological profile, tuberculosis/HIV co-infection is still a major public health problem in the southwestern region of Maranhão. This situation calls for better coordination between tuberculosis and sexually transmitted disease/AIDS control programs, as well as a political commitment and greater involvement on the part of administrators and health care professionals in the planning of interventions and the functioning of health care facilities.

- Abstract
The genus Rickettsiacomprises obligatory intracellular bacteria, well known to cause zoonotic diseases around the world. The present work aimed to investigate the occurrence of Rickettsiaspp. in wild animals, domestic dogs and their respective ectoparasites in southern Pantanal region, central-western Brazil, by molecular and serological techniques. Between August 2013 and March 2015, serum, whole blood and/or spleen samples were collected from 31 coatis, 78 crab-eating foxes, seven ocelots, 42 dogs, 110 wild rodents, and 30 marsupials. Serum samples from canids, felids, rodents and marsupials were individually tested by indirect fluorescent antibody test (IFAT) in order to detect IgG antibodies to Rickettsia rickettsii, Rickettsia parkeriand Rickettsia amblyommatis. DNA samples from mammals and ectoparasites were submitted to a multiplex qPCR assay in order to detect and quantify spotted fever group (SFG) and typhus group (TG) rickettsiae and Orientia tsutsugamushi. Positive samples in qPCR assays were submitted to conventional PCR assays targeting gltA, ompA, ompBand htrAgenes, followed by sequencing and phylogenetic analyses. The ticks collected (1582) from animals belonged to the species Amblyomma sculptum, Amblyomma parvum, Amblyomma ovale, Amblyomma tigrinum, Rhipicephalus (Boophilus) microplus, Rhipicephalus sanguineussensu lato and Amblyomma auricularium. Overall, 27 (64.2%) dogs, 59 (75.6%) crab-eating foxes and six (85.7%) ocelots were seroreactive (titer≥64) to at least one Rickettsiaspecies. For 17 (40.4%) dogs, 33 (42.3%) crab-eating foxes, and two (33.3%) ocelots, homologous reactions to R. amblyommatisor a closely related organism were suggested. One hundred and sixteen (23.5%) tick samples and one (1.2%) crab-eating fox blood sample showed positivity in qPCR assays for SFG Rickettsiaspp. Among SFG Rickettsia-positive ticks samples, 93 (80.2%) belonged to A. parvum, 14 (12%) belonged to A. sculptumspecies, three (2.5%) belonged to A. auricularim, and six (5.2%) were Amblyommalarval pools. Thirty samples out of 117 qPCR positive samples for SFG Rickettsiaspp. also showed positivity in cPCR assays based on gltA, htrAand/or ompBgenes. The Blast analyses showed 100% identity with ‘CandidatusRickettsia andeanae’ in all 30 sequences obtained from gltA, htrAand/or ompBgenes. The concatenated phylogenetic analysis based on gltAand 17-kDa htrAgenes grouped the Rickettsiasequences obtained from tick samples in the same clade of ‘CandidatusRickettsia andeanae’. The present study revealed that wild and domestic animals in southern Pantanal region, Brazil, are exposed to SFG rickettsiae agents. Future studies regarding the pathogenicity of these agents are necessary in order to prevent human cases of rickettsiosis in Brazilian southern Pantanal.
