Research has been conducted on nephric lineage. Results indicate that mouse embryos lacking transcription factors Pax2 and Pax8 cannot form pronephros and that these transcription factors are important regulators specifying nephric lineage.

Antibody-mediated cancer immunotherapy targets inhibitory surface molecules, such as PD1, PD-L1, and CTLA-4, aiming to re-invigorate dysfunctional T cells. We purified and characterized tumorinfiltrating lymphocytes (TILs) and their patient-matched non-tumor counterparts from treatmentnaïve NSCLC patient biopsies to evaluate the effect of PD1 expression on the functional and molecular profiles of tumor-resident T cells. We show that PD1+ CD8+ TILs have elevated expression of the transcriptional regulator ID3 and that the cytotoxic potential of CD8 T cells can be improved by knocking down ID3, defining it as a potential regulator of T cell effector function. PD1+ CD4 + memory TILs display transcriptional patterns consistent with both helper and regulator function, but can robustly facilitate B cell activation and expansion. Furthermore, we show that expanding ex vivo-prepared TILs in vitro broadly preserves their functionality with respect to tumor cell killing, B cell help, and TCR repertoire. Although purified PD1+ CD8+ TILs generally maintain an exhausted phenotype upon expansion in vitro, transcriptional analysis reveals a downregulation of markers of T-cell dysfunction, including the co-inhibitory molecules PD1 and CTLA-4 and transcription factors ID3, TOX and TOX2, while genes involved in cell cycle and DNA repair are upregulated. We find reduced expression of WNT signaling components to be a hallmark of PD1+ CD8+ exhausted T cells in vivo and in vitro and demonstrate that restoring WNT signaling, by pharmacological blockade of GSK3β, can improve effector function. These data unveil novel targets for tumor immunotherapy and have promising implications for the development of a personalized TIL-based cell therapy for lung cancer. [ABSTRACT FROM AUTHOR]

The ectopic Pax5 expression from the Ikaros promoter that induces proximal rather than distal V(sub H) -DJ(sub H) rearrangements in IK(super Pax5/+) is demonstrated. The conclusion states that Pax5 activates locus contraction and distal V(sub H) -DJ(sub H) rearrangements in collaboration with an unknown factor that is present in pro-B cells but absent in thymocytes.

The Th2 locus control region (LCR) has been shown to be important in efficient and coordinated cytokine gene regulation during Th2 cell differentiation. However, the molecular mechanism for this is poorly understood. To study the molecular mechanism of the Th2 LCR, we searched for proteins binding to it. We discovered that transcription factor YY1 bound to the LCR and the entire Th2 cytokine locus in a Th2-specific manner. Retroviral overexpression of YY1 induced Th2 cytokine expression. CD4-specific knockdown of YY1 in mice caused marked reduction in Th2 cytokine expression, repressed chromatin remodeling, decreased intrachromosomal interactions, and resistance in an animal model of asthma. YY1 physically associated with GATA-binding protein-3 (GATA3) and is required for GATA3 binding to the locus. YY1 bound to the regulatory elements in the locus before GATA3 binding. Thus, YY1 cooperates with GATA3 and is required for regulation of the Th2 cytokine locus and Th2 cell differentiation. [ABSTRACT FROM AUTHOR]

The transcription factor Pax8 is expressed in the developing thyroid gland, inner ear, kidney, and mid-hindbrain region. Pax8 mutant mice die only postnatally due to a thyroid gland defect. Here we report the generation and expression analysis of a Pax8(cre) allele. Cre recombinase activity was detected in known Pax8 expression domains of Pax8(cre/+) and Pax8(cre/cre) embryos carrying the Z/AP transgene, which expresses alkaline phosphatase only after Cre-mediated excision of lacZ sequences. Alkaline phosphatase expression was additionally detected in the adrenal gland and in the facial, vestibulocochlear, and cuneate nerves, which have so far not been associated with Pax8 expression.
