Abstract

A key feature of the innate antiviral immune response is a rapid nonspecific response to virus infection largely mediated by the induction and extracellular secretion of type I interferons (IFNs) that restrict virus replication. Cytoplasmic sensors such as RIG-I recognize viral RNA and trigger antiviral signaling pathways that upregulate IFN transcription. However, it remains largely unknown how antiviral signaling is negatively regulated to maintain homeostasis after the elimination of virus. In this report, we have identified the RING domain-containing protein RING finger 11 (RNF11) as a novel negative regulator of innate antiviral signaling. Overexpression of RNF11 downregulated IFN-β expression and enhanced viral replication whereas siRNA-mediated knockdown of RNF11 suppressed viral replication. RNF11 interacted with the noncanonical IKK kinases TBK1/IKKi and attenuated their Lys63-linked polyubiquitination by blocking interactions with the E3 ligase TRAF3. The inhibitory function of RNF11 was dependent on the ubiquitin-binding adaptor molecule TAX1BP1 which was required for RNF11 to target TBK1/IKKi. Collectively, these results indicate that RNF11 functions together with TAX1BP1 to restrict antiviral signaling and IFN-β production.

Abstract

Background: Despite advances in surgery and pharmacotherapy, there remains significant residual ischemic risk after coronary artery bypass grafting surgery. Methods: In REDUCE-IT (Reduction of Cardiovascular Events With Icosapent Ethyl–Intervention Trial), a multicenter, placebo-controlled, double-blind trial, statin-treated patients with controlled low-density lipoprotein cholesterol and mild to moderate hypertriglyceridemia were randomized to 4 g daily of icosapent ethyl or placebo. They experienced a 25% reduction in risk of a primary efficacy end point (composite of cardiovascular death, myocardial infarction, stroke, coronary revascularization, or hospitalization for unstable angina) and a 26% reduction in risk of a key secondary efficacy end point (composite of cardiovascular death, myocardial infarction, or stroke) when compared with placebo. The current analysis reports on the subgroup of patients from the trial with a history of coronary artery bypass grafting. Results: Of the 8179 patients randomized in REDUCE-IT, a total of 1837 (22.5%) had a history of coronary artery bypass grafting, with 897 patients randomized to icosapent ethyl and 940 to placebo. Baseline characteristics were similar between treatment groups. Randomization to icosapent ethyl was associated with a significant reduction in the primary end point (hazard ratio [HR], 0.76 [95% CI, 0.63–0.92]; P =0.004), in the key secondary end point (HR, 0.69 [95% CI, 0.56–0.87]; P =0.001), and in total (first plus subsequent or recurrent) ischemic events (rate ratio, 0.64 [95% CI, 0.50–0.81]; P =0.0002) compared with placebo. This yielded an absolute risk reduction of 6.2% (95% CI, 2.3%–10.2%) in first events, with a number needed to treat of 16 (95% CI, 10–44) during a median follow-up time of 4.8 years. Safety findings were similar to the overall study: beyond an increased rate of atrial fibrillation/flutter requiring hospitalization for at least 24 hours (5.0% vs 3.1%; P =0.03) and a nonsignificant increase in bleeding, occurrences of adverse events were comparable between groups. Conclusions: In REDUCE-IT patients with a history of coronary artery bypass grafting, treatment with icosapent ethyl was associated with significant reductions in first and recurrent ischemic events. Abstract

Ghrelin influences a variety of metabolic functions through a direct action at its receptor, the GhrR (GhrR-1a). Ghrelin knockout (KO) and GhrR KO mice are resistant to the negative effects of high-fat diet (HFD) feeding. We have generated several classes of small-molecule GhrR antagonists and evaluated whether pharmacologic blockade of ghrelin signaling can recapitulate the phenotype of ghrelin/GhrR KO mice. Antagonist treatment blocked ghrelin-induced and spontaneous food intake; however, the effects on spontaneous feeding were absent in GhrR KO mice, suggesting target-specific effects of the antagonists. Oral administration of antagonists to HFD-fed mice improved insulin sensitivity in both glucose tolerance and glycemic clamp tests. The insulin sensitivity observed was characterized by improved glucose disposal with dramatically decreased insulin secretion. It is noteworthy that these results mimic those obtained in similar tests of HFD-fed GhrR KO mice. HFD-fed mice treated for 56 days with antagonist experienced a transient decrease in food intake but a sustained body weight decrease resulting from decreased white adipose, but not lean tissue. They also had improved glucose disposal and a striking reduction in the amount of insulin needed to achieve this. These mice had reduced hepatic steatosis, improved liver function, and no evidence of systemic toxicity relative to controls. Furthermore, GhrR KO mice placed on low- or high-fat diets had lifespans similar to the wild type, emphasizing the long-term safety of ghrelin receptor blockade. We have therefore demonstrated that chronic pharmacologic blockade of the GhrR is an effective and safe strategy for treating metabolic syndrome.

Abstract

Stimulation of the ghrelin receptor (GhrR) by ghrelin results in a variety of metabolic changes including increased food intake, fat storage and insulin resistance. Loss of ghrelin signaling is protective against diet-induced obesity, suggesting that ghrelin plays a significant homeostatic role in conditions of metabolic stress. We examined glycemic control in GhrR −/− mice fed a high-fat diet, and used indirect calorimetry to assess fuel substrate usage and energy expenditure. GhrR −/− mice fed a high-fat diet had several measures of greater insulin sensitivity, including: lower fasted blood glucose and plasma insulin, lower %Hb A1c , lower insulin levels during glucose tolerance tests, and improved performance in hyperinsulinemic-euglycemic and hyperglycemic clamp studies. GhrR −/− mice fed a high-fat diet did not develop hepatic steatosis and had lower total cholesterol, relative to controls. Furthermore, GhrR −/− mice demonstrated a lower intestinal triglyceride secretion rate of dietary lipid. GhrR −/− mice have higher respiratory quotients (RQ), indicating a preference for carbohydrate as fuel. The range of RQ values was wider in GhrR −/− mice, indicating greater metabolic flexibility and insulin sensitivity in these animals. We therefore propose that loss of ghrelin signaling promotes insulin sensitivity and metabolic flexibility, and protects against several fatty diet-induced features of metabolic syndrome due to convergent changes in the intake, absorption and utilization of energy.

Abstract

The innate immune system provides the first line of defense for protection against pathogenic bacteria or viruses. The host ‘senses' pathogen infection by recognition of pathogen-associated molecular patterns via germ-line-encoded pattern-recognition receptors including Toll-like receptors, RIG-I-like receptors (RLRs), NOD-like receptors (NLRs) and DNA sensors. Upon receptor engagement, signaling cascade events culminate in the production of the type I interferons (IFNs) IFN-α and IFN-β that restrict virus replication. RIG-I and MDA5 are the key RLRs that recognize viral genomic RNAs and transmit downstream signals to the mitochondrial adaptor molecule MAVS (also known as IPS-1, VISA or Cardif).1 MAVS nucleates a signaling complex anchored to the mitochondria containing the E3 ubiquitin ligase TRAF3, the non-canonical IκB kinases (IKKs) TBK1 and IKKi (also known as IKK epsilon) and the adaptor molecules NEMO (also known as IKK gamma) and TANK.1 TBK1 and IKKi phosphorylate the interferon regulatory factor 3 (IRF3) transcription factor to trigger IRF3 dimerization and nuclear localization to activate type I IFN transcription.2 TBK1 and IKKi are both regulated by posttranslational modifications that occur during virus infection including lysine 63 (K63)-linked polyubiquitination by TRAF3 or other E3 ubiquitin ligases.3,4 TBK1/IKKi K63-linked polyubiquitination is linked to their activation and may be involved in recruiting signaling complexes containing ubiquitin-binding domains to transmit downstream signals. Dysregulation of type I IFN signaling, characterized by overproduction of IFN-α and IFN-β, has been implicated in numerous autoimmune diseases such as systemic lupus erythematosus.5 Therefore, the production of IFN is subject to tight regulatory control by numerous mechanisms to maintain immune homeostasis. In a recent article in Nature Immunology, Cui et al.6 have reported that the NLR protein NLRP4 functions as a negative regulator in the RLR pathway by targeting TBK1 for proteasomal degradation. NLRs represent a family of innate immune regulatory proteins that consist of a nucleotide-binding and oligomerization (NOD) domain, a leucine-rich repeat region and a variable amino-terminal effector domain. NLRs may function as either activators or negative regulators of the RLR signaling pathway.7 NLRP4, an NLR family member, has been reported to inhibit the nuclear factor-κB signaling pathway; however, its role in the RLR pathway was not previously examined.8 The authors initially performed an overexpression screen using vectors encoding various NLRs and identified NLRP4 as an inhibitor of RLR signaling.6 NLRP4 inhibited the induction of IFN-β and ISRE luciferase reporters by poly(I:C) (double-stranded RNA mimic), poly(dA:dT) (double-stranded DNA mimic) and vesicular stomatitis virus-GFP infection. Knockdown of NLRP4 by RNA interference potentiated the activation of IFN-β and ISRE luciferase reporters in 293T cells. Furthermore, the authors used more physiologically relevant immune cells including monocytic THP-1 cells and peripheral blood mononuclear cells to demonstrate that knockdown of NLRP4 enhanced the expression of interferon stimulated genes generated during virus infection. To identify the target(s) of NLRP4, the authors overexpressed key signaling molecules in the RLR/Toll-like receptor pathways (RIG-I, MDA5, MAVS, TRIF, TBK1 and IKKi) to activate IFN-β and ISRE luciferase reporters and found that NLRP4 inhibited activation by all of the expression vectors except for IKKi.6 Therefore, NLRP4 inhibited RLR signaling at a downstream step, likely at the level of TBK1. Indeed, NLRP4 interacted specifically with overexpressed TBK1, but not IKKi or IRF3, as shown by coimmunoprecipitation experiments. However, an interaction between endogenous NLRP4 and TBK1 proteins was signal-dependent and was observed only upon virus infection, suggesting that NLRP4 interacts specifically with activated TBK1. Consistent with these observations, NLRP4 interacted with the kinase domain, but not the ubiquitin-like domain or coiled-coil domains, of TBK1. Interestingly, NLRP4 was not recruited to TBK1 until 8–10 h after virus infection, suggesting that NLRP4 participates in the later phases of the termination of RLR signaling. It was previously demonstrated that the regulatory molecules A20, TAX1BP1 and ABIN1 inhibit RLR signaling by suppressing K63-linked TBK1 polyubiquitination at an early phase after virus infection (4 h).4,9 Thus, TBK1 appears to be targeted by multiple inhibitory proteins in distinct phases after virus infection. It has been previously reported that TBK1 is conjugated by K63-linked polyubiquitin chains by the E3 ubiquitin ligase TRAF3.10 The authors conducted a kinetic analysis of TBK1 ubiquitination during virus infection that revealed that K63-linked ubiquitination of TBK1 occurred early after virus infection (4 h), whereas K48-linked ubiquitination of TBK1 was observed at later times (8 h) after virus infection correlating with NLRP4 and TBK1 binding.6 Indeed, the authors found that NLRP4 promoted K48-linked polyubiquitination of TBK1 which triggered its degradation by the proteasome. NLRP4 required its NOD domain, but not the pyrin or leucine-rich repeat domains, to interact with and promote the degradation of TBK1. Taken together, it appears that TBK1 is inactivated by a ubiquitin-editing mechanism and NLRP4 is essential for the K48-linked polyubiquitin conjugation step. Since NLRP4 lacks E3 ligase activity, the authors hypothesized that NLRP4 was functioning as an adaptor molecule for an E3 ligase to trigger TBK1 ubiquitination and degradation. Using a sublibrary of RING domain E3 ligase shRNAs, the authors identified the E3 ubiquitin ligase DTX4 as an inhibitor of TBK1-mediated activation of an ISRE luciferase reporter.6 Furthermore, knockdown of DTX4 using shRNA prevented the inhibition and degradation of TBK1 by NLRP4. Since DTX4 was also dependent on NLRP4 to ubiquitinate and degrade TBK1, it was likely that NLRP4 served as an adaptor molecule for DTX4. Consistent with this notion, NLRP4 and DTX4 formed an inducible protein complex in response to virus infection and both proteins interacted with TBK1 at the precise time when TBK1 was conjugated with K48-linked polyubiquitin chains. These data suggest that both NLRP4 and DTX4 cooperatively induce the ubiquitination and degradation of TBK1 to restrict the induction of virus-induced type I IFN. The authors next used a bioinformatics strategy to identify lysine 670 (K670) within the coiled-coil domain of TBK1 as the residue that was ubiquitinated by DTX4.6 Mutation of K670 to arginine (K670R) prevented NLRP4-mediated ubiquitination and degradation of TBK1. In addition, DTX4 knockdown enhanced ISRE reporter activation in the presence of wild-type TBK1 but not with the TBK1 K670R mutant. Finally, the authors demonstrated that NLRP4 was unable to induce the ubiquitination of a TBK1 phosphorylation mutant, suggesting that the activated form of TBK1 was selectively targeted by NLRP4 and DTX4. In summary, Cui et al. have demonstrated that NLRP4 functions as an adaptor for the E3 ubiquitin ligase DTX4 to target TBK1 for K48-linked ubiquitination to trigger its degradation and restrict the induction of type I IFN. Although this study has provided new insight into the mechanisms involved in the inhibition of innate antiviral signaling, there are still key questions that should be addressed in future studies. What is the purpose of multiple inhibitors of TBK1 such as the previously reported A20-TAX1BP1-ABIN1 axis which disrupts the interactions between TRAF3 and TBK1-IKKi to suppress TBK1-IKKi K63-linked ubiquitination? There are likely temporal differences that allow these proteins to suppress TBK1 by ubiquitin editing in a coordinated and highly complementary manner. Whereas A20-TAX1BP1-ABIN1 inhibits TBK1 at an earlier phase after virus infection (4 h), NLRP4-DTX4 triggers TBK1 degradation at later times (Figure 1). This two-step mechanism of substrate inactivation was first described for A20 inhibition of RIP1 in the tumor necrosis factor receptor 1 pathway.11 In future studies, it will be interesting to determine if NLRP4-DTX4 inhibition of TBK1 is dependent on the prior suppression of TBK1 K63-linked polyubiquitination by the A20 complex. This can be addressed by examining the ubiquitination and degradation of TBK1 by NLRP4-DTX4 in A20, TAX1BP1 or ABIN1-deficient cells. Furthermore, a recent study has implicated the E3 ubiquitin ligase TRAF-interacting protein in the degradation of TBK1 to limit virus-induced IFN-β production.12 Does TRAF-interacting protein function together with NLRP4 and DTX4 in a protein complex to promote K48-linked ubiquitination and degradation of TBK1? If so, why are multiple E3 ubiquitin ligases involved in TBK1 degradation? The answers to these questions may elucidate the complexities of homeostatic control of innate immune signaling and provide new insight into the underlying causes of certain autoimmune diseases. Abstract

A key feature of the innate antiviral immune response is a rapid nonspecific response to virus infection largely mediated by the induction and extracellular secretion of type I interferons (IFNs) that restrict virus replication. Cytoplasmic sensors such as RIG-I recognize viral RNA and trigger antiviral signaling pathways that upregulate IFN transcription. However, it remains largely unknown how antiviral signaling is negatively regulated to maintain homeostasis after the elimination of virus. In this report, we have identified the RING domain-containing protein RING finger 11 (RNF11) as a novel negative regulator of innate antiviral signaling. Overexpression of RNF11 downregulated IFN-β expression and enhanced viral replication whereas siRNA-mediated knockdown of RNF11 suppressed viral replication. RNF11 interacted with the noncanonical IKK kinases TBK1/IKKi and attenuated their Lys63-linked polyubiquitination by blocking interactions with the E3 ligase TRAF3. The inhibitory function of RNF11 was dependent on the ubiquitin-binding adaptor molecule TAX1BP1 which was required for RNF11 to target TBK1/IKKi. Collectively, these results indicate that RNF11 functions together with TAX1BP1 to restrict antiviral signaling and IFN-β production.

Abstract

The human T cell leukemia virus type I (HTLV-I) inhibits host antiviral signaling pathways although the underlying mechanisms are unclear. Here we found that the HTLV-I Tax oncoprotein induced the expression of SOCS1, an inhibitor of interferon signaling. Tax required NF-κB, but not CREB, to induce the expression of SOCS1 in T cells. Furthermore, Tax interacted with SOCS1 in both transfected cells and in HTLV-I transformed cell lines. Although SOCS1 is normally a short-lived protein, in the presence of Tax, the stability of SOCS1 was greatly increased. Accordingly, Tax enhanced the replication of a heterologous virus, vesicular stomatitis virus (VSV), in a SOCS1-dependent manner. Surprisingly, Tax required SOCS1 to inhibit RIG-dependent antiviral signaling, but not the interferon-induced JAK/STAT pathway. Inhibition of SOCS1 by RNA-mediated interference in the HTLV-I transformed cell line MT-2 reduced HTLV-I replication and p19Gag levels. Taken together, our results reveal that Tax inhibits antiviral signaling, in part, by hijacking an interferon regulatory protein.

Human T cell leukemia virus type 1 (HTLV-1) inhibits host antiviral signaling pathways although the underlying mechanisms are unclear. Here we found that the HTLV-1 Tax oncoprotein induced the expression of SOCS1, an inhibitor of interferon signaling. Tax required NF-κB, but not CREB, to induce the expression of SOCS1 in T cells. Furthermore, Tax interacted with SOCS1 in both transfected cells and in HTLV-1-transformed cell lines. Although SOCS1 is normally a short-lived protein, in the presence of Tax, the stability of SOCS1 was greatly increased. Accordingly, Tax enhanced the replication of a heterologous virus, vesicular stomatitis virus (VSV), in a SOCS1-dependent manner. Surprisingly, Tax required SOCS1 to inhibit RIG-I-dependent antiviral signaling, but not the interferon-induced JAK/STAT pathway. Inhibition of SOCS1 by RNA-mediated interference in the HTLV-1-transformed cell line MT-2 resulted in increased IFN-β expression accompanied by reduced HTLV-1 replication and p19 Gag levels. Taken together, our results reveal that Tax inhibits antiviral signaling, in part, by hijacking an interferon regulatory protein.

Abstract

The goal of this study was to examine factors that contribute to energy balance in female GHR −/− mice. We measured energy intake, energy expenditure (EE), fuel utilization, body mass ( M b ) changes and physical activity in 17 month-old female GHR −/− mice and their age-matched wild type littermates. The GHR −/− mice were smaller, consumed more food per unit M b , had greater EE per unit M b and had an increase in 24-h EE/ M b that was similar to the increase in their surface-area-to-volume ratio. Locomotor activity (LMA) was reduced in the GHR −/− mice, but the energetic cost associated with their LMA was greater than in wild type controls. Furthermore, M b and LMA were independent explanatory covariates of most of the variance in EE, and when adjusted for M b and LMA, the GHR −/− mice had higher EE during both the light and dark phases of the daily cycle. Respiratory quotient was lower in GHR −/− mice during the light phase, which indicated a greater utilization of lipid relative to carbohydrate in these mice. Additionally, GHR −/− mice had higher ratios of caloric intake to EE at several intervals during the dark phase, and this effect was greater and more sustained in the final 3 h of the dark phase. Therefore, we conclude that GHR −/− mice are able to overcome the substantial energetic challenges of dwarfism through several mechanisms that promote stable M b . Relative to wild type mice, the GHR −/− mice consumed more calories per unit M b , which offset the disproportionate increase in their daily energy expenditure. While GHR −/− mice oxidized a greater proportion of lipid during the light phase in order to meet their energy requirements, they achieved greater energy efficiency and storage during the dark phase through a combination of higher energy consumption and lower LMA.

Abstract

The orexigenic peptide ghrelin has been shown to have prokinetic activity in the gastrointestinal (GI) system of several species, including humans. In this series of experiments, we have evaluated the prokinetic activity of novel, small-molecule ghrelin receptor (GhrR) agonists after parenteral and peroral dosing in mice and rats. Gastric emptying, small intestinal transport, and fecal output were determined after intraperitoneal and intracerebroventricular dosing of GhrR agonists, using ghrelin as a positive control. These same parameters were evaluated after oral gavage dosing of the synthetic agonists. Regardless of dose route, GhrR agonist treatment increased gastric emptying, small intestinal transit, and fecal output. However, fecal output was only increased by GhrR agonist treatment if mice were able to feed during the stimulatory period. Thus, GhrR agonists can stimulate upper GI motility, and the orexigenic action of the compounds can indirectly contribute to prokinetic activity along the entire GI tract. The orexigenic and prokinetic effects of either ghrelin or small-molecule GhrR agonists were selective for the GhrR because they were absent when evaluated in GhrR knockout mice. We next evaluated the efficacy of the synthetic GhrR agonists dosed in a model of opiate-induced bowel dysfunction induced by a single injection of morphine. Oral dosing of a GhrR agonist normalized GI motility in opiate-induced dysmotility. These data demonstrate the potential utility of GhrR agonists for treating gastrointestinal hypomotility disorders.

Abstract

Background We have used a mouse model based on overexpression of c-Myc in B cells genetically engineered to be self-reactive to test the hypothesis that farnesyl transferase inhibitors (FTIs) can effectively treat mature B cell lymphomas. FTIs are undergoing clinical trials to treat both lymphoid and non-lymphoid malignancies and we wished to obtain evidence to support the inclusion of B cell lymphomas in future trials. Results We report that two FTIs, L-744,832 and SCH66336, blocked the growth of mature B cell lymphoma cells in vitro and in vivo. The FTI treatment affected the proliferation and survival of the transformed B cells to a greater extent than naïve B cells stimulated with antigen. In syngeneic mice transplanted with the transgenic lymphoma cells, L-744,832 treatment prevented the growth of the tumor cells and the morbidity associated with the resulting lymphoma progression. Tumors that arose from transplantation of the lymphoma cells regressed with as little as three days of treatment with L-744,832 or SCH66336. Treatment of these established lymphomas with L-744,832 for seven days led to long-term remission of the disease in approximately 25% of animals. Conclusion FTI treatment can block the proliferation and survival of self-reactive transformed B cells that overexpress Myc. In mice transplanted with mature B cell lymphomas, we found that FTI treatment led to regression of disease. FTIs warrant further consideration as therapeutic agents for mature B cell lymphomas and other lymphoid tumors.
