During adolescence, healthy eating habits are important, and regular meal intake has an especially positive effect on future health. However, the rate of skipping breakfast has gradually increased. Therefore, this study was conducted to evaluate the positive effects of a rice-based breakfast in Korean adolescents who usually skip breakfast. In this open parallel-group, randomized controlled trial, 105 middle and high school students aged 12–18 years who habitually skipped breakfast were recruited. They were randomly divided into three groups: the rice meal group (RMG, n = 35), wheat meal group (WMG, n = 35), and general meal group (GMG, n = 35). The RMG and WMG received a rice-based breakfast and wheat-based breakfast, respectively, for 12 weeks. After a 12-week intervention, the body fat mass (p &lt, 0.05) and body mass index (p &lt, 0.05) in the RMG were significantly lower than those in the other two groups, and the stress score was also significantly lower in the RMG (p &lt, 0.05). Moreover, after the intervention, in the RMG only, compared to baseline levels, the relative theta (RT) wave activity significantly decreased in eight electrode sites, and the relative alpha (RA) wave activity increased significantly. Eating a rice-based breakfast has positive effects on body fat accumulation and cognitive function in Korean adolescents. Furthermore, a rice-based breakfast plan that is preferred by adolescents should be developed to assist them in developing healthy eating habits.

Background/objectives Elevation of postprandial lipemia characterized by a rise in triglyceride (TG)-rich lipoproteins can increase the risk of atherogenesis. The objective of this study was to investigate postprandial lipemia response to a single dietary fat/sugar load test and monitor beneficial changes induced by the consumption of Platycodi radix (AP) beverage in healthy subjects. Subjects/methods A total of 52 subjects were randomly assigned to either placebo or AP beverage group with a high-fat shake in a randomized controlled crossover trial. Postprandial blood was collected at 0, 1, 2, 4, and 6 h and analyzed for TG and lipoprotein lipase mass. Inhibition of pancreatic lipase was determined in vitro. Results AP inhibited pancreatic lipase activity in vitro (IC50 = 5 mg/mL). Compared to placebo beverage, AP beverage consumption with a high-fat shake induced significant increase of plasma lipoprotein lipase mass (P = 0.0111, β estimate = 4.2948) with significant reduction in very low-density lipoprotein (VLDL) TG concentration (P = 0.038, β estimate = -52.69) at 6 h. Based on significant correlation between high-fat dietary scores MEDFICTS and postprandial TG responses in VLDL (P = 0.0395, r = 0.2127), subgroup analysis revealed that 6 h-postprandial VLDL TG response was significantly decreased by AP consumption in subjects with MEDFICTS ≥ 40 (P = 0.0291, β estimate = -7214). Conclusions AP beverage might have potential to alleviate postprandial lipemia through inhibiting pancreatic lipase activity and elevating lipoprotein lipase mass. Subgroup analysis revealed that subjects with high-fat dietary pattern could be classified as responders to AP beverage among all subjects.

In the present investigation, glucosinolate content was identified and quantified in 210 accessions of Korean leaf mustard (Brassica juncea var. integrifolia) by a liquid chromatography (LC) with electrospray ionization (ESI) coupled with a positive-mode ion trap tandem mass spectrometry (LC–ESI–MS/MS). Eleven individual GSLs including two new compounds were identified in the accessions using desulfo-glucosinolates (DS-GSLs) LC–MS library, and they were elucidated by interpreting the fragmentation data ([M + H-glucose]+) obtained. The newly identified GSLs contained aliphatic and hydroxyl functional group in their back bone were named as 1-ethyl-2-hydroxyethyl DS-GSLs (glucosisautricin) or 2-hydroxy-2-methylpropyl DS-GSLs (glucoconringiin) and 3,4,5-trimethoxybenzyl DS-GSLs, respectively. Among all tested accessions, the total DS-GSLs content was found to be varied between 5.3 and 23.2 mg/g dry weight (DW) with an average of 13.0 mg/g DW in the germplasm. Each individual DS-GSLs component was found in decreasing order of sinigrin (41.7 %) >glucoiberverin (21.7 %) >gluconasturtiin (12.6 %) >glucobarbarin (10.0 %) >glucoiberin (5.1 %) >glucocheirolin (3.6 %) >glucobrassicanapin (2.6 %) >gluconapin (2.1 %), and >glucobrassicin (0.6 %). Interestingly, sinigrin and gluconasturtiin were present in higher content, and progoitrin was not detected significantly in the germplasm of Korean leaf mustard. In particular, accession K046197 (purple) was found to show highest total DS-GSL content (23.2 mg/g DW).

Koreans have been consuming Petasites Japonicus (PJ) as food. Although the therapeutic effect of PJ on allergic or inflammatory reactions associated with asthma has been proven, its effect on obesity is unclear. Therefore, the present study was aimed to assess the obesity related anti-inflammatory and anti-adipogenic effects of ethanol extract PJ (EPJ) on the inflammatory response in RAW 264.7 macrophages and on differentiation in 3T3-L1 adipocytes. In addition, the polyphenolic compound was quantitatively characterized from the EPJ using ultra performance liquid chromatography coupled with diode array detector, quadrupole time-of-flight-mass spectrometry (UPLC-DAD-QToF-MS). In RAW 264.7 or 3T3-L1, reduction of nitric oxide (in macrophages) production as well as monocyte chemoattractant protein-1 and tumor necrosis factor-&alpha, were observed. Treatment of EPJ in adipocyte differentiation showed an improvement in adiponectin and lipid accumulation and a significant reduction of PPAR&gamma, and FABP-4 mRNA expression levels. On the other hand, mRNA expression of UCP-1, PPAR&alpha, and ACO increased in the EPJ treated group. In addition, a total of 26 polyphenolic compounds were detected and of which 12 are reported for the first time from PJ. The higher content of diverse polyphenolic compounds presented in EPJ might be responsible for the observed anti-inflammatory and anti-adipogenic effect. These results suggest that PJ is valuable in improving obesity-related inflammatory responses.

Oxidative stress has been implicated in the pathophysiology of chronic diseases and cigarette smoking is known to be one of the major sources of oxidants. In this study, we investigated whether Korean black raspberry (BR) supplementation could reduce smoke-induced oxidative stress in healthy male smokers. Thirty nine healthy male smokers received either 30 g of freeze-dried BR or placebo for four weeks. Anthropometric and dietary data, smoking history, blood lipid profiles, lipid peroxidation, DNA damage, and antioxidant enzyme activities were assessed at baseline and after supplementation. There was no difference of age, smoking history, anthropometry, and nutrient intake between groups. BR supplementation had no effect on plasma lipid profiles, LDL oxidation, and DNA damage. However, it significantly increased the activity of the antioxidant enzymes, glutathione peroxidase and catalase, and reduced plasma lipid peroxidation. In conclusion, BR supplementation may decrease cigarette smoke-induced oxidative stress through increase of endogenous antioxidant enzyme activities.

SummaryBackground An effective treatment strategy for acne vulgaris is the reduction of Propionibacterium acnes in the skin. The Helicobacter pylori-derived synthetic antimicrobial peptide HPA3NT3 is a customized α-helical cationic peptide with antibacterial and anti-inflammatory activity. Objectives To examine the role of HPA3NT3 as a treatment against P. acnes-induced skin inflammation. Methods Morphological alteration of individual P. acnes cells by HPA3NT3 was visualized by scanning electron microscopy. Modulation by HPA3NT3 of a number of P. acnes-induced innate immune responses was analysed in vitro using cultured normal human keratinocytes (HKs), and in vivo using the ICR mouse, a well-established model for P. acnes-induced skin inflammation. Results The minimum inhibitory concentration of HPA3NT3 against P. acnes was low (0·4 μmol L−1). HPA3NT3 showed no cytotoxicity to HK cells at the concentrations used in our in vitro and in vivo studies. Treatment with HPA3NT3 in vitro induced morphological disruptions in P. acnes cells suggestive of a bactericidal effect. HPA3NT3 significantly decreased P. acnes-induced interleukin-8 expression and intracellular calcium mobilization in HK cells by inhibiting P. acnes-activated Toll-like receptor 2-mediated nuclear factor-κB signalling pathways. Intradermal injection of HPA3NT3 in vivo effectively decreased viable P. acnes, as well as erythema, swelling and inflammatory-cell infiltration in ICR mouse ears inoculated with P. acnes. Conclusions Our data suggest that HPA3NT3 has potential as a therapeutic agent for acne vulgaris due to its antimicrobial effects on P. acnes and its ability to block P. acnes-induced inflammation.

The present investigation intends to evaluate the changes in the content of cis/trans carotene isomers as provitamin A carotenoids by steaming and roasting processes in the roots of four Korean sweet potato varieties viz. Shinzami, Younwhangmi, Chuwhangmi and Jinhongmi using a liquid chromatography with diode array detection and the negative ion atmospheric pressure chemical ionization mass spectrometric (LC-DAD-APCI/MS) method and UV spectral pattern library created from several reference data. Except Shinzami, the content of all trans β-carotenes was found to slightly decreased or remained constant when steamed or roasted. The content of cis α-/β-carotenes was potentially increased about 2-fold or greater when raw or steamed and the content was slightly decreased while roasted. In Chuwhangmi, the content of 13-cis α-carotene and all trans α-carotenes were rapidly increased when steamed and slightly decreased when roasted. Chuwhangmi exhibited 27.2 mg/100 g DW content of all trans β-carotenes when roasted and thus, it was considered as a relatively superior cultivar.

The effect of administration of Korean black raspberry (BR) to smokers was assessed by 1H nuclear magnetic resonance (NMR) coupled with multivariate statistical analysis. In this study, to investigate the effect of BR on the level of urinary 8-hydroxy-2′-deoxyguanosine (8-OHdG), biomarker for DNA damage, 38 male smokers between 20 and 30 years old were treated with BR or placebo for 4 weeks. After 4 weeks of administration, urinary level of 8-OHdG was measured, and the metabolic profiling of plasma samples of all volunteers were established using 1H NMR-based metabolomics techniques. The volunteers of BR administered group were divided into responder group (BR-R group) and nonresponder group (BR-NR group) based on the mean decrement of urinary 8-OHdG level of placebo group (12%). There was a clear separation among weeks 0 and 4 of seven volunteers at BR-R group in principal component analysis (PCA) derived score plots. The levels of 16 metabolites including amino acids, and organic acids changed significantly between weeks 0 and 4. Then the comparative metabolic profiling of the BR-R and BR-NR groups at week 0 was performed. There was also clear separation in PCA-derived score plot. The relative levels of glycine and trimethylamine N-oxide in plasma changed significantly between the two groups, indicating that these compounds could be used as screening biomarkers to predict the 8-OHdG modulating activity of BR in smokers.

The metabolome of Rubus coreanus Miquel fruit (bokbunja) was characterized by simultaneously analyzing 92 samples to elucidate the integrative metabolic factors associated with 4 different regions and 3 levels of maturity. The region- and maturity-specific metabolomic differences were characterized using ultra-performance liquid chromatography–quadrupole time-of-flight mass spectrometry (UPLC–qTOF-MS). Unsupervised multivariate proved capable of classifying the metabolite profiles associated with the different regions and maturity levels. The maturity levels induced major clusters in which the most distinctive metabolic phenotypes were observed in the immature and middle-mature bokbunja cultivated in the Gok-sung region. The resultant loading score plot revealed that tannins, proanthocyanidins, and anthocyanidins were the metabolic features that differentiated the maturity levels. Furthermore, applying a multivariate extension of a univariate analysis of variance (ANOVA) isolated the region-dependent metabolic markers that are independent of the ripening stage (e.g., procyanidin dimer B4 and galloyl-HHDP-glucose). These metabolites are important bioactive compounds as well as determinants of taste quality. Our results could be useful in monitoring and producing black raspberry-related foods with superior functional, flavor, and medicinal qualities.

Rubus coreanus Miquel (RCM) has been used as one of the Korean traditional medicines for prostate health. In addition, recent studies have reported that RCM reduced chronic inflammatory diseases such as cancer, and rheumatoid arthritis. Therefore, in this study, we investigated the effects of unripe and ripe RCM on inflammation- related gene expressions in LPS-stimulated mouse peritoneal macrophages. Mice were fed with 2% unripe RCM (U2), 10% unripe RCM (U10), 2% ripe RCM (R2), and 10% ripe RCM (R10) for 8 weeks. Peritoneal macrophages were isolated and stimulated with LPS then proinflammatory mediators (TNF-α, IL-1β, and IL-6), and prostaglandin E 2 (PGE 2 ) productions were assessed. Moreover, gene expression profiles were analyzed by cDNA microarray method. Unripe and ripe RCM significantly reduced TNF-α production but only unripe RCM decreased IL-1β and IL-6 production. RCM intake significantly reduced inflammatory-related gene expressions such as arachidonate 5-lip-oxygenase, interleukin 11, and nitric oxide synthase 2. Furthermore, unripe and ripe RCM significantly decreased ceruloplasmin, tissue plasminogen activator, thrombospondin 1, and vascular endothelial growth factor A expression which modulates symptoms of chronic inflammatory diseases. RCM intake also significantly increased hypoxia inducible factor 3, alpha which is the negative regulators of hypoxia-inducible gene expression. Furthermore, only unripe RCM reduced chemokine (C-C motif) ligand 8, chemokine (C-X-C motif) ligand 14, and phospholipase A2 expression. In this study, we showed that RCM had anti-inflammatory effects by suppression of pro-inflammatory mediator ex-pressions and may reduce chronic inflammatory disease progress through regulation of gene expressions. These findings suggest that RCM might be used as a potential functional material to reduce chronic inflammatory responses.

Acorn-derived foods are commonly marketed in Korea, but their beneficial health effects have not been extensively explored. The aim of this study was to investigate the anti-inflammatory and antiasthmatic effects of an ethanol extract (AEx) from acorn (Quercus acutissima CARR.) in murine macrophage RAW264.7 cells and ovalbumin (OVA)-induced asthma model mice. The AEx at concentrations less than 100 µg/mL significantly, dose dependently attenuated the production of nitric oxide and the expression of inducible nitric oxide synthase, cyclooxygenase-2 and interleukin-1β (IL-1β) in RAW264.7 cells. The AEx (100 or 300 mg/kg/day) also inhibited the production of tumor necrosis factor-α and T-helper 2 (Th2) type cytokines (IL-4 and IL-13) in both the lung tissues and bronchoalveolar lavage fluid of the OVA-induced asthma model mice. These results suggest that the AEx has suppressive effects on asthma through its anti-inflammatory action both in vitro and in vivo. PRACTICAL APPLICATIONS AEx may be useful for preventing inflammatory diseases, such as allergic asthma, and may thus be effectively used as a natural anti-inflammatory ingredient in health food or pharmaceuticals.

In this study, muffins were made with the addition of 50% acorn jelly powder and 0, 0.1, and 0.5% acorn ethanol extract powder. The moisture content of muffins decreased with an increase in the acorn ethanol extract powder concentration. Baking loss rate decreased with an increase in powder concentration. The lightness and yellowness of muffins decreased as the concentration of powder increased. Redness increased as the concentration of powder increased. The hardness of muffins increased, and cohesiveness decreased with an increase in powder concentration. Gumminess, and chewiness decreased by the addition of powder. The results of a sensory test showed that the texture quality of the muffin increased as the amount of added acorn increased. The taste of muffins increased with an increase in the powder concentration, but 1.0% acorn ethanol extract powder decreased. In terms of the overall acceptability, 0.1% acorn ethanol extract powder had the highest preference level.

A novel synthetic hexapeptide (SFKLRY-NH2) that displays angiogenic activity has been identified by positional scanning of a synthetic peptide combinatorial library (PS-SPCL). This study was carried out to investigate the irritation of the SFKLRY-NH2 on the skin. The tests were performed on the basis of Korea Food and Drug Administration (KFDA) guidelines. In results, cell toxicity is not appeared for SFKLRY-NH2 in HaCaT cells and B16F10 cells. SFKLRY-NH2 induced no skin irritation at low concentration (10 μM), mild irritation at high concentration (10mM). We consider that this result is helpful for saying about the safety of SFKLRY-NH2 in clinical use.

This study examined the effects of acorn (Quercus. acutissima CARR.) jelly powder (0%, 25%, 50%, and 75%) and acorn extract (0%, 0.1%, 0.5%, and 1.0%) addition on the quality characteristics of hardtack. In the sensory test, acorn jelly powder added group scored 50% higher than the other added group. Regarding hardtack color, L (lightness) and b (yellowness) values decreased with increasing acorn powder and acorn extract addition. Hardness of hardtack increased with added acorn jelly powder, but no significant difference was observed with acorn extract. The taste and texture of the hardtack 0.1% acorn extract added group significantly increased. Overall, preferences decreased with increasing acorn extract but not significantly. In conclusion, the results of this study suggest that addition of 50% acorn jelly powder in combination with addition of less than 0.1% acorn extract to hardtack was the most desirable.

As hydrophilic pigments, anthocyanins reduce the incidence of cancer and cardiovascular diseases. In this study, the anthocyanin content and composition following steaming and baking of the roots of the Korean purple-fleshed sweet potato variety “Shinzami” were evaluated using liquid chromatography with diode array detection and electrospray ionisation/mass spectrometry (LC-DAD-ESI/MS). Anthocyanins of Shinzami were composed of mono- or di-acylated forms of p-hydroxybenzoic acid, caffeic acid and ferulic acid with the basic structure of cyanidin 3-sophoroside-5-glucoside or peonidin 3-sophoroside-5-glucoside. A total of 15 individual anthocyanins were isolated and confirmed, one of which was presumed to be a newly identified compound, peonidin 3-feruloyl-p-hydroxybenzoyl sophoroside-5-glucoside. Additionally, the amounts of di-acylated cyanidin 3-caffeoyl-p-hydroxybenzoyl sophoroside-5-glucoside and peonidin 3-caffeoyl-p-hydroxybenzoyl sophoroside-5-glucoside were the highest (137.0 and 565.9 mg/100 g DW, respectively) among cyanidin and peonidin compounds. After steaming, the total anthocyanin content was reduced by nearly a half, while roasting only slightly reduced the total anthocyanin content.

Extract of Taraxacum platycarpum (AF-343) has been reported to have several biological properties such as skin hydration and anti-inflammatory effects. Although clinical evidences of skin hydration and antiinflammatory effect were proven in clinical trial, precise mechanism of skin hydration was not fully understood yet. In this study, we have focused skin hydration mechanism related filaggrin, collagen, and matrix metalloproteinase (MMP) in vitro and animal study. Herein, skin hydration mechanism of AF-343 is due to recovery of filaggrin in mice model and increased production of collagen with suppression of matrix MMP in vitro fibroblast cell line.

Nuclear magnetic resonance (NMR) techniques coupled with multivariate data analysis were used to conduct monitoring of biochemical changes of black raspberry fruits at different stages of maturation and under various extraction and NMR dissolution solvent conditions: extraction with 50% methanol and D 2 O as an NMR dissolution solvent, extraction with 50% methanol and 50% methanol- d 4 as an NMR dissolution solvent, and extraction with 100% ethyl acetate and 100% methanol- d 4 as an NMR dissolution solvent. Partial least-squares discriminant analysis reliably distinguished black raspberry fruits according to the maturation stage, whereby the relative levels of various compounds such as amino acids, organic acids, sugars and phenolic compounds were compared using analysis of variance. Sucrose and most of the amino acids, and organic acids decreased, whereas fructose, glucose, and cyanidins increased in relative concentration according to maturation of black raspberry fruits. The total number and kinds of assigned compounds of the three solvent systems were also compared. This research demonstrates that the metabolic profile of black raspberry fruits changes during maturation, and provides objective criteria for determining the stage of black raspberry fruit maturation via a 1 H NMR-based metabolomics technique using multiple solvent systems.

Phytochemical compositions of five varieties of black soybeans (Glycine max) and their stabilities at room temperature, 4 and −80 °C over 14 months were determined by HPLC systems with electrochemical (ECD) and UV detectors. Polyphenol profiling was carried out by a liquid chromatography–electrospray ionisation-mass spectrometry (LC–ESI-MS) with orbitrap as a mass analyser in both positive and negative ion modes, and polyphenols in aglycone forms were quantified by HPLC–ECD. Five different varieties of black soybeans (G. max) contained 249–405 μg/g dry wt of γ-tocopherol and 6.76–14.98 μg/g dry wt of lutein. Major polyphenols in black soybeans (G. max) were daidzein (193–288 μg/g dry wt) and genistein (145–223 μg/g dry wt), mainly present as glucosides and acetyl glucosides. No significant decrease was found in total phenols of black soybeans (G. max) stored at room temperature, 4 or −80 °C for 14 months. 【The purpose of the present study is to develop a simple, fast and sensitive LC/MS method for simultaneous separation and the determination of an active component in the oriental medicinal mushroom Cordyceps militaris. Based on this work, the contents of cordycepin in Cordyceps militaris fruiting cultivated on various media were determined and compared. And also, the nutritional components such as minerals and vitamins were determined in order to provide useful information to consumer as a food material. The analysis methods of nutritional components were chosen on the basis of AOAC. The optimum separation for cordycepin was achieved using a solvent gradient consisting of the mixture of 0.1% formic acid in methanol (solvent B) in a background of 0.1% formic acid in water (solvent A) as a mobile phase and a 3.0 ${\times}$ 150 Waters XTera column. Selective ion monitoring (SIR) mode ([M+H]+ at m/z 252) was used for quantitative analysis of cordycepin. The cultivated Cordyceps militaris on various media contained 1~14 /g of cordycepin, 0.65~1.08% of thiamine, 0.86~7.17% of riboflavin, and 3.01~5.26% of niacin. The content of mineral components varied on categories, especially contained 500~3500% of potassium as a major mineral. Cordycepin, niacin and potassium were found much higher in the fruiting cultivated with soy power media (gold 10) than other media.】

The fruit of the plum tree (Prunus salicina Lindl.) has been used as a traditional medicinal food in humans to enhance immunity against infectious agents and to treat cancers. However, limited information exists on the mechanisms responsible for its immune enhancing properties. In this study, the immunostimulatory effects of a methanol extract of plum fruit following methanol evaporation and dissolving in PBS were assessed by in vitro lymphocyte proliferation, tumor cell cytotoxicity, and nitric oxide (NO) production. The crude methanol extract stimulated spleen lymphocyte proliferation and NO production by cultured macrophages, and inhibited the viability of tumor cells, significantly greater than media controls. Sequential gel filtration chromatographic separation of the extract on Sephadex G-25 and Sephacryl S-200 gel filtration columns resulted in a more purified preparation that retained the ability to induce lymphoproliferation, tumor killing, and NO production. These results suggest that Prunus salicina contains immunostimulatory components that potentially may be useful in human and veterinary medicine.

Phytonutrients in Angelica keiskei, a dark green leafy vegetable, were characterised and their extraction efficiency by different compositions of water/ethanol as well as stability at different temperatures was determined. A range in the content of lutein (205–265 mg/kg dry wt), trans-β-carotene (103–130 mg/kg dry wt), and total phenols (8.6–9.7 g/kg) was observed amongst Angelica keiskei grown in three different conditions. LC-ESI-MS/MS analysis identified chlorogenic acid, chalcones and the glucosides of luteolin and quercetin as the major phenolic compounds in Angelica keiskei. Only 46% of lutein was extracted by 70% of ethanol and no carotenoid was detected in 40% ethanol and 100% water extracts of Angelica keiskei. Major phytonutrients in Angelica keiskei were stable at −80 °C up to 12 months whilst β-carotene was significantly degraded at room temperature and 4 °C within 2 months and lutein at room temperature within 12 months. The current study indicates that phytonutrients rich in Angelica keiskei can only be extracted partially using ethanol/water and substantial loss of certain phytonutrients such as β-carotene can occur during storage at either 4 °C or room temperature.

The process of biodegradation in lingo-cellulosic materials is critically relevant to biospheric carbon. The study of this natural process has largely involved laboratory investigations, focused primarily on the biodegradation and recycling of agricultural by-products, generally using basidiomycetes species. In order to collect super white rot fungi and evaluate its ability to degrade lingo-cellulosic material, 35 fungal strains, collected from forests, humus soil, livestock manure, and dead trees, were screened for enzyme activities and their potential to decolorize the commercially used Poly-R 478 dye. In the laccase enzymatic analysis chemical test, 33 white rot fungi and 2 brown rot fungi were identified. The degradation ability of polycyclic aromatic hydrocarbons (PAHs) according to the utilized environmental conditions was higher in the mushrooms grown in dead trees and fallen leaves than in the mushrooms grown in humus soil and livestock manure. Using Poly-R 478 dye to assess the PAH-degradation activity of the identified strains, four strains, including Agrocybe pediades, were selected. The activities of laccase, MnP, and Lip of the four strains with PAH-degrading ability were highest in Pleurotus incarnates. 87 fungal strains, collected from forests, humus soil, livestock manure, and dead trees, were screened for enzyme activities and their potential to decolorize the commercially used Poly-R 478 dye on solid media. Using Poly-R 478 dye to assess the PAHdegrading activity of the identified strains, it was determined that MKACC 51632 and 52492 strains evidenced superior activity in static and shaken liquid cultures. Subsequent screening on plates containing the polymeric dye poly R-478, the decolorization of which is correlated with lignin degradation, resulted in the selection of a strain of Coriolus versicolor, MKACC52492, for further study, primarily due to its rapid growth rate and profound ability to decolorize poly R-478 on solid media. Considering our findings using Poly-R 478 dye to evaluate the PAH-degrading activity of the identified strains, Coriolus versicolor, MKACC 52492 was selected as a favorable strain. Coriolus versicolor, which was collected from Mt. Yeogi in Suwon, was studied for the production of the lignin-modifying enzymes laccase, manganese-dependent peroxidase (MnP), and lignin peroxidase (LiP).

This study aimed to develop a fluorometric method to determine total antioxidant activity of plant foods. The antioxidant activities in plant foods were determined after extracting (1) hydrophilic components with acidified methanol (methanol:glacial acetate acid:water=50:3.7:46.3), (2) lipophilic components with methanol followed by tetrahydrofuran (THF), or (3) both hydrophilic and lipophilic components using sequential extraction of acidified methanol and THF together. This study was conducted to evaluate the effects of dietary safflower leaf on protective immunity against coccidiosis, the most economically important parasitic disease of poultry. White Leghorn chickens were fed a standard diet with or without safflower leaf and were either uninfected or orally infected with 5,000 sporulated oocysts of Eimeria acervulina. Protective immunity was assessed by body weight gain, fecal oocyst shedding, splenic lymphocytes proliferation, T lymphocyte subpopulations, and proinflammatory cytokine gene expression. We observed that the effect of safflower on experimental coccidiosis was dependant on the dose of the supplement used. A 0.1% (wt/wt) safflower-supplemented diet increased body weight gains of coccidia-infected chickens to a level identical to that of uninfected controls, and significantly reduced fecal oocyst shedding compared with animals that were given a non-supplemented standard diet. Furthermore, increased splenic lymphocyte proliferation as well as greater percentages of CD4+ T cells and decreased CD8+ cells were observed in animals fed a 0.1% safflower-supplemented diet. Finally, IFN-γ, IL-8, IL-15 and IL-17 transcripts in the 0.1% safflower-supplemented group were higher than the non-supplemented controls. These results indicate that safflower leaf when given as a dietary supplement possesses immune-enhancing properties that augment protective immunity against experimental coccidiosis.

A variety of fruits have traditionally been used in Asian cultures to enhance resistance to diseases and treat cancers. However, limited information exists on the underlying mechanisms responsible for these effects. The present investigation was conducted to examine the ability of three Korean indigenous fruits (persimmon, raspberry and tomato) to stimulate lymphocyte proliferation and macrophage nitric oxide production as parameters of innate immunity, and to inhibit tumor cell growth. In vitro co-culture of chicken spleen lymphocytes with methanol extracts of persimmon (Diospyros kaki) or tomato (Lycopersicon esculentum) induced greater cell proliferation compared with cells treated with the vehicle control. Stimulation of chicken macrophages with extracts of persimmon or raspberry (Rubus crataegifolius), but not tomato, stimulated robust nitric oxide production to levels similar to that produced by interferon-γ. All fruit extracts uniformly inhibited the growth chicken tumor cells in vitro. These results provide a rational basis for future studies investigating the effects of medicinal fruits on innate immunity and carcinogenesis in humans and animals.

This study examined the chemical composition of A. blasiliensis and the chemical structural properties of an immuno-stimulating polysaccharide. The amino acids, free sugars, and organic acids by HPLC and fatty acids by GC were analyzed. The immuno-stimulating substance from A. blasiliensis was extracted with hot water and purified by ethanol precipitation. It underwent ion exchange chromatography on DEAE-cellulose and gel filtration on Toyopearl HW 65F. Through GP-HPLC, the substance was found to be homogeneous. Its chemical structure was determined by (13)C-NMR. Fatty acids, organic acids, and sugar alcohol composition consisted exclusively of linoleic acid, fumaric acid and mannitol, respectively. The amino acids were mainly glutamic acid, glycine, and arginine. By (13)C-NMR analysis, the immuno-stimulating substance was identified as β-(1→3) (1→6)-glucan, composed of a backbone with (1→3)-linked D-glucopyranosyl residues branching a (1→6)-linked D-glucopyranosyl residue. The β-glucan from A. blasiliensis showed pronounced immuno-stimulating activity on the antibody-production ability of B-lymphocytes by the hemolytic suspension assay. In these results, A. blasiliensis was estimated to have potent pharmacological properties and potential nutritional values.

Safflower (Carthamus tinctorius) has been used as a traditional medicinal plant to enhance natural immunity and treat cancers. However, limited information exists on the mechanisms responsible for its immune enhancing properties. In this study, the immunostimulatory effects of a methanol extract of safflower leaf were assessed by in vitro lymphocyte proliferation, tumor cell cytotoxicity, and nitric oxide production by cultured chicken macrophages. A crude methanol extract of safflower leaf stimulated spleen lymphocyte proliferation and nitric oxide production, and inhibited the viability of tumor cells significantly greater than medium controls. Sequential gel filtration chromatographic separation of the methanol extract on Sephadex G-25 and Sephacryl S-200 columns resulted in a partially purified preparation that retained the ability to induce lymphoproliferation, tumor killing, and NO production. These results demonstrate for the first time that safflower leaf contains immunostimulatory and anti-tumor component(s) that may be potentially useful as a dietary immunomodulator for poultry.

This study describes the extraction and characterization of a novel inhibitor against β-hydroxy-β-methyl glutaryl (HMG) Coenzyme A (CoA) reductase from the mushroomPholiota adiposa. Methanol extracts ofP. adiposa PAD-022 fruiting body showed the highest HMG-CoA reductase-inhibitory activity of 55.8%. The HMG-CoA reductase-inhibitor,P. adipos a PAD-022, was maximally extracted when its fruiting body was treated with methanol at 30°C for 12 h. The HMG-CoA reductase-inhibitor was obtained by systematic solvent extraction methods followed by gel column chromatography, and RP-HPLC. The purified product was found to possess an activity of IC50 6.8 μg and a yield of 1.8%. The molecular weight of purified HMG-CoA reductase-inhibitor was deduced to be 412.7 Da. The inhibitor was identified as stigmasterol (C29H46O) by serial instrumental analyses, including LC-Mass. NMR, FT-IR, and UV spectrometry.

Aging is a multifactorial phenomenon. Several clinical conditions directly related to lipid metabolism that induces hypertriglycemia, hypercholesterolemia, and cardiovascular disease occur during aging. Attention has been focused on possible intervention strategies to control serum lipid levels. Phytic acid is a plant component in most grains that is the main source of energy intake for the aged, and its antioxidant and antinutrient effects have been reported. However, its effect on lipid metabolism in the aged model has not been evaluated. This study was carried out to investigate the effect of phytic acid on serum and hepatic lipid levels in aged mice. A total of 40 aged ICR male mice were fed purified diets supplemented with 0% (P0), 0.5% (P5), 1.0% (P10), and 1.5% (P15) sodium phytate for 12 weeks. There were no significant differences in food intake, body weight, and organ weight among the experimental groups. The concentrations of the serum low-density lipoprotein cholesterol, hepatic triacylglycerol, and total cholesterol, and the apparent absorption rates of total lipid and cholesterol were lower in the P15 group than in the P0 group. Serum high-density lipoprotein cholesterol levels of all groups fed phytate-containing diets were higher than that of the P0 group. The severity of fatty liver decreased as phytate percentage in the diet increased. The amounts of fecal total lipid, triacyglycerol, and total cholesterol were higher in the P10 and the P15 groups. These results suggested that phytate affect the serum and hepatic lipid levels in aged mice by increasing their fecal lipid content. Consuming phytate-rich foods may reduce serum and hepatic lipid levels in the aged.

Phytic acid, myo-inositol hexaphosphate, is a plant component existing in most grains and legumes. Although much attention has been focused on the biologic actions of phytic acid in human beings and animals, its effect on the blood glucose level in diabetic models has not been evaluated. This study was conducted to examine the supplementary effect of phytate on the blood glucose level in a diabetic rodent model. Thirty male diabetic KK mice were fed with purified diets supplemented with 0% (P0), 0.5% (P5), or 1.0% (P10) sodium phytate for 8 weeks. Diet intake, body and organ weights, and levels of fasting and random blood glucose, hemoglobin A 1c , as well as insulin were measured. A glucose tolerance test was conducted. There was no significant difference in diet intake, body weight, and organ weight among the experimental groups. Extracts from 63 kinds of Pholiota sp. fruiting bodies were prepared using water and methanol, and then their physiological functionalities were investigated. The methanol extracts from Pholiota adiposa PAD030 showed high fibrinolytic activity and those of P. adiposa ASI PAD-022 showed potential inhibitory activity of 76.8% against reductase. The highest antioxidant and tyrosinase inhibitory activities were found in the water extracts of Pholiota sp. PSP-015 (72.7%) and methanol extracts of P. nameko PNA-024 (69.5%), respectively. However, superoxide dismutase(SOD)-like activity and elastase inhibitory activity were low in almost of the extracts. The HMG-CoA reductase inhibitor from the fruiting body of P. adiposa PAD-022 which showed the highest functionality was extracted maximally when powder of the fruiting body was shaked at for 12 h by methanol and its HMG-CoA reductase inhibitory activity was 80.2%.

This study was designed to investigate the supplementary effects of Lentinus edodes which were harvested at different time period and part on acetylcholine content and its related enzyme activities in the brain of diabetic mouse model (KK mouse). We fed mice with standard diet (Control diet; CON) or 4 different kinds of experimental diets (DGC: on time harvested, cap of Dong Go; DGS: on time harvested, stipe of Dong Go; HSC: late harvested, cap of Hyang Sin: HSS: late harvested, stipe of Hyang Sin) to KK mouse for 8 weeks. Neurotransmitter such as acetylcholine contents, acetylcholinesterase activities, monoamine oxidase-B ac-tivities and lipid peroxide contents in the brain were measured. The results showed that acetylcholine content was significantly higher in DGC and HSC groups than CON group. The activities of acetylcholinesterase and monoamine oxidase-B enzyme were significantly inhibited in the brain of DGC and HSC groups compared with CON group. Lipid peroxide content was lower in DGC group than CON group. These results suggested that the cap of Lentinus edodes which were harvested on time and late time contain increased acetylcholine content and decreased acetylcholinesterase activities, monoamine oxidase-B activities and lipid peroxide contents. Thus the cap of Lentinus edodes which were harvested at different time periods may play an effective role in enhancing cognitive function.

Dandelions have been reported to have medicinal properties and bioactive components that impact human health. However, the precise biological properties of dandelions and the parts of the plants possessing bioactive components remain uncertain. In this study, we evaluated 3 different types of dandelions based on their cultivation origin (Songpa, Uiryung, and native Uiryung types) as well as their 4 different plant parts (leaf, flower, root, skin). Each sample was extracted with 80% methanol and then compared for the biological activities (anti-oxidative, immune cell proliferative and tumor cell growth inhibitory activities). All 3 types of dandelions possessed a degree of biological functions including the hydroxyl radical scavenger activity, immune cell proliferative activity and tumor cell growth inhibitory activity. However, there was no significant difference in these activities between the 3 dandelion types. Leaves of all three dandelion types showed the highest levels of all biological activities. To a lesser degree, the flower and root parts displayed biological activities. In the skin parts, anti-oxidative activity was also detected only at higher doses of dandelion extracts. Heating the dandelion leaf extract did not affect the biological activity, suggesting a heat-stable nature of the biological compounds. Taken together, these collective data suggest that dandelions, in particular their leaves, possess a high concentration of heat-resistant biological compounds, which are responsible for anti-oxidative, immune cell proliferative and tumor cell growth-inhibitory activities.

This study was performed to investigate the effect of natural plant extracts on acetylcholinesterase (AChE) activity and the free radical scavenging activity. The methanolic extracts of plants were tested for AChE inhibitory activity using Ellman`s colorimetric method in 96-welled microplates and antioxidant activity as the scavenging effect of 1, 1-diphenyl-2-picryl hydrazyl radical (DPPH). The results showed that AChE activities were inhibited (about 20-30%) in whole plant extract of Daucus carota var. sativa, Hypericum erectum and Fragaria yezoensis. AChE activities were inhibited (about 32-34%) in stems extract of Gingko biloba and leaves extract of Rhododendrondron yedoensa var. poukhanense. Fruit extract of Zanthoxylum schinifolium inhibited (about 18%) AChE activity. And the DPPH scavenging effects as antioxidant activity were similar to L-ascorbic acid in whole plant extract of Fragaria yezoensis and fruits extract of Comus officinalis.

The changes of microflora and chemical characteristics during fermentation process of cotton waste for oyster mushroom cultivation were investigated with 5 l bench-scale reactors placed in an incubator at different temperatures (). Cotton waste was wetted to 70% moisture, and air flow rates to the substrate were 50, 100 and 300 cc/min. In processing of composting, the mesophilic bacterial population decreased sharply but thermophilic bacterial population increased. In case of fungi, both mesophilic and thermophilic population decreased. The daily evolution showed little difference in all treatments, while dropped sharply after 3 days. The desirable composting temperature and air flow based on the mycelial growth of oyster mushroom were and 100 cc/min, respectively.

Effects of pH, sugar, ascorbic acid, organic acids and light sources on the stability of anthocyanin pigment extracted from Korean purple-fleshed potatoes (PL-6, PL-28, PL-31 and Jasim) were studied. The pH had remarkable influence on the color stability of anthocyanin pigment. With increasing pH, the color gradually fades as colorless pseudobases are formed. In acidic pH, anthocyanin was stable, but with increasing pH the color gradually changed to colorless. The addition of sugar decrease in color stability of the pigment during storage period. The most of organic acids, such as a tartaric, citric and succinic acids, were found to improve the stability of the pigment, while malic and malonic acid reduced the stability of the pigment. The addition of ascorbic acid considerable decreased in anthocyanin pigment stability, but the effect was not decreased by adding thiourea. The effect of light sources such as a darkroom, a fluorescent light, and sunlight, reduced gradually the stability of anthocyanin pigment. Therefore the pigment degradation could be minimized by shielding the light from the pigment.

In many organisms, trehalose has been Down as an energy source and a protectant against various environmental stresses such as desiccation, freezing, heat and osmotic pressure. Previously, we have isolated and characterized the genes encoding trehalose-6-phosphate synthase (ZrTPS1) and trehalose-6-phosphate phosphatase (ZrTPS2) from one of the most osmotolerant yeasts, Zygosaccharomyces rouxii. We have also generated transgenic plants by co-introduction of ZrTPS2 and ZrTPS2 into potato plant (ZrTPS2-2A-ZrTPS1 plant) in an attempt to metabolically engineer trehalose in the transgenic plant using the foot-and-mouth disease virus(FMDV) 2A system and to generate drought resistant crop plants. In this research, we assayed previously generated the ZrTPS2-2A-ZrTPS1 plant biofunctionally by drought treatment, and measured the amount of trehalose in the ZrTPS2-2A-ZrTPS1 transgenic plants. The ZrTPS2-2A-ZrTPS1 transgenic plant showed strong drought resistance in spite of little or no accumulation of transgenic in he transgenic plant compared with control plant.

γ-Oryzanol, a mixture of ferulic acid esters of triterpene alcohols and sterols, is a nutritionally important group of rice secondary metabolites. A library of 27 γ-oryzanol was assembled from existing data and used to assist identification and quantification of γ-oryzanol isolated from 16 Korean rice varieties (11 white and 5 pigmented). γ-Oryzanol was analyzed with liquid chromatography with diode array detection and electrospray ionization mass spectrometry. Nineteen different γ-oryzanol were observed and identified as stigmasterol, campesterol and sitosterol or common and hydroxylated triterpene alcohols. In the 16 varieties, the total γ-oryzanol content averaged 43.8 mg/100 g (range, 26.7-61.6 mg/100 g), which Josaengheugchal exhibited the highest level (61.6 mg/100 g). The Korean rice varieties were classified based on qualitative and quantitative γ-oryzanol data by multivariate statistical analysis. Clusters of specialty rice varieties exhibited higher γ-oryzanol levels than those of common rice varieties.

Subjects consumed 5 g dry Angelica keiskei powder encapsulated in gelatin capsules with a low flavonoid and carotenoid liquid meal. Plasma samples were collected at baseline, 0.5, 1, 2, 3, 4, 5, 6, 7, and 8 h. Samples were analyzed for flavonoids and carotenoids using HPLC systems with electrochemical and UV detection, respectively, and for total antioxidant performance by fluorometry.RESULTS: After ingestion of Angelica keiskei increases in plasma quercetin concentrations were observed at 1-3 and 6-8 hr in the healthy group and at all time points in the metabolic syndrome group compared to baseline (P < 0.05). Plasma lutein concentrations were significantly elevated in both the healthy and metabolic syndrome groups at 8 hr (P < 0.05). Significant increases in total antioxidant performance were also observed in both the healthy and the metabolic syndrome groups compared to baseline (P < 0.05).CONCLUSIONS: Findings of this study clearly demonstrate the bioavailability of phytonutrients of Angelica keiskei and their ability to increase antioxidant status in humans. To elucidate the composition of phytosterols in the fruit body of Pholiota spp. 6 species(P. adiposa, P. aurivella, P. highlandensis, P. nameko, P. squarrosa and Pholiota sp.) were analyzed with Gas chromatography(GC). Pholiota spp. were contained campesterol, stigmasterol, and -sitosterol as the major phytosterols.

A heterogalactan, fomitellan A, was isolated from a 0.9% sodium chloride extract of the fruit bodies of Fomitella fraxinea by a combination of fractionation procedures, including precipitation with ethanol and chromatography on columns of DEAE–cellulose and Toyopearl HW 65F. On employing gel permeation–high performance liquid chromatograpy, fomitellan A exhibited a single peak with a molecular weight of 15 kDa. The results of methylation, and 1 H and 13 C NMR spectroscopic analyses indicated that the heterogalactan fomitellan A was a repeating unit (pentasaccharide) composed of a backbone with (1→6)-linked d -galactopyranosyl residues. Its C-2 atom position was substituted with disaccharide units of 3-O- d -mannopyranosyl- l -fucopyranosyl residues. The fucomnnaogalactan (fomitellan A) from Fomitella fraxinea showed pronounced immunostimulating activity in an in vitro lymphocytes proliferation assay.
