Mycobacteroides abscessus (Mabc) is a rapidly growing nontuberculous mycobacterium that poses a considerable challenge as a multidrug-resistant pathogen causing chronic human infection. Effective therapeutics that enhance protective immune responses to Mabc are urgently needed. This study introduces trans-3,5,4′-trimethoxystilbene (V46), a novel resveratrol analogue with autophagy-activating properties and antimicrobial activity against Mabc infection, including multidrug-resistant strains. Among the resveratrol analogues tested, V46 significantly inhibited the growth of both rough and smooth Mabc strains, including multidrug-resistant strains, in macrophages and in the lungs of mice infected with Mabc. Additionally, V46 substantially reduced Mabc-induced levels of pro-inflammatory cytokines and chemokines in both macrophages and during in vivo infection. Mechanistic analysis showed that V46 suppressed the activation of the protein kinase B/Akt-mammalian target of rapamycin signaling pathway and enhanced adenosine monophosphate-activated protein kinase signaling in Mabc-infected cells. Notably, V46 activated autophagy and the nuclear translocation of transcription factor EB, which is crucial for antimicrobial host defenses against Mabc. Furthermore, V46 upregulated genes associated with autophagy and lysosomal biogenesis in Mabc-infected bone marrow-derived macrophages. The combination of V46 and rifabutin exerted a synergistic antimicrobial effect. These findings identify V46 as a candidate host-directed therapeutic for Mabc infection that activates autophagy and lysosomal function via transcription factor EB.

COVID-19 is an unprecedented pandemic that has impacted the whole world. The pandemic made researchers and educators realize the critical need to prepare for future disasters. This study explored a context-specific case for elementary online learning where we investigated how elementary school teachers transitioned to emergency remote teaching (ERT) from face-to-face to online learning during the pandemic. A case study approach was used to explore South Korean elementary teachers' ERT approaches and experiences during COVID-19. Using the CIPP (Context, Input, Process, and Product) framework, we sought to understand how the transition occurred from the perspectives of the teachers. The analysis uncovered several themes that fall under each category of the framework. In terms of context, limited technological aptitude and lack of training in online instructional design as well as policy issues and socio-economic differences were identified as key factors in assessing the current state of the ERT. In terms of input, instructors' efforts as well as support from in and out of school were discussed. Student interaction and engagement were identified as key factors in understanding the process of ERT. Lastly, learning outcomes, instructional strategies, and systemic transformation emerged as products of ERT.

Abstract The impact of COVID-19 pandemic on suicide remains unclear and might differ according to individuals’ socioeconomic characteristics. We aimed to investigate excess suicide attributable to COVID-19 in South Korea, stratified by the outbreak period and individual characteristics. We obtained daily time-series suicide mortality data for January 2017–December 2020 from the Korea National Statistics Office and performed a two-stage interrupted time-series analysis. We estimated excess suicide in 16 regions of Korea using a quasi-Poisson time-series regression model and pooled the region-specific estimates using a mixed-effects multivariate meta-analysis model in the first and second stages, respectively. From February 18 to December 31, 2020, suicide decreased by 9.5% [95% empirical confidence interval (eCI): 3.8%, 15.6%] compared to the number expected from the pre-pandemic period. The decrease in excess suicide risk from the initial pandemic was pronounced during the pandemic’s first and third waves. Further, we found that the decrease in suicide was more evident in individuals who were male [11.7% (95% eCI: 5.5%, 18.0%)], middle-aged [13.7% (95% eCI: 7.8%, 19.6%)], highly educated [12.6% (95% eCI: 6.4%, 19.4%)], and married [13.6% (95% eCI: 8.0%, 20.3%)] than in the general population, based on the point estimates. Our results provide timely evidence to establish public health policies for suicide prevention and suggest the prioritization of resource allocation for mental health of individuals based on individual characteristics.

Abstract Previous spectral analysis studies on insomnia have shown inconsistent results due to their heterogeneity and small sample sizes. We compared the difference of electroencephalogram (EEG) spectral power during sleep among participants without insomnia, insomniacs with no hypnotic use, hypnotic users with no insomnia complaints, and hypnotic users with insomnia complaints using the Sleep Heart Health Study data, which is large sample size and has good quality control. The fast Fourier transformation was used to calculate the EEG power spectrum for total sleep duration within contiguous 30-s epochs of sleep. Attention-deficit/hyperactivity disorder (ADHD) symptoms affect 40-60% of autistic children and have been linked to differences in adaptive behavior. It is unclear whether adaptive behavior in autistic youth is directly impacted by co-occurring ADHD symptoms or by another associated feature of both autism and ADHD, such as increased irritability. The current study examined relationships between irritability, ADHD symptoms, and adaptive behavior in 3- to 7-year-old autistic children. Results suggest that, after adjusting for co-occurring ADHD symptoms, higher levels of irritability are associated with differences in social adaptive behavior specifically. Understanding relationships between irritability, ADHD, and adaptive behavior in autistic children is critical because measures of adaptive behavior, such as the Vineland Scales of Adaptive Functioning, are often used as a proxy for global functioning, as well as for developing intervention plans and measuring outcomes as primary endpoints in clinical trials.

Objective We assessed the association between household decision-making and mental well-being among Asian immigrant women residing in Korea. We also investigated if the impact varies by the regional origin and examined potential factors for joint decision-making. Methods We conducted a cross-sectional study using the Korean National Survey of Multicultural Families 2015 and logistic regression. We analyzed data from 11,188 married immigrant women ages 20 to 59 who were originally from East Asia or Southeast/South Asia and co-living with their spouses. We defined households as joint-decision, wife-decision, or husband-decision based on who decides how living expenses are spent. Mental well-being encompassed a depressive mood for two consecutive weeks, and life and marital satisfaction. Results After controlling for covariates, we showed that immigrant women in husband-decision households were more likely to have depressive mood (odds ratio [OR] 1.26, 95% confidence interval [CI]: 1.14–1.38), poorer life satisfaction (OR 1.49, 95% CI: 1.24–1.78), and poorer marital satisfaction (OR 1.81, 95% CI: 1.47–2.22) than women in joint-decision households. Immigrant women in wife-decision households had a similar but slightly lower odds of poor mental well-being. This association was less prominent for Southeast/South Asian origin than East Asian origin, while the age-adjusted prevalence rates of poor mental well-being among them were higher than their East Asian counterparts. Factors that restricted the odds of husband-decision did not necessarily guarantee wife-decision. Conclusions This study suggests one-sided decision-making can be a risk factor for immigrant women’s poor mental well-being, while joint decision-making is protective. Differences across regional origins suggest domestic decision-making might be a less important predictor for mental well-being in immigrants more occupied with adapting to the host society. Factors for joint decision-making should be addressed to improve the mental well-being of immigrant women.

Recently, generative AI has been at the center of disruptive innovation in various settings, including educational sectors. This article investigates ChatGPT, which is one of the most prominent generative AI in the market, to explore its usefulness and potential for instructional design. Four researchers used a set of prompts to generate a course map for an online course that is aimed to teach the topic of makerspace and conducted SWOT analysis to identify strengths, weaknesses, opportunities, and threats of using generative AI for instructional design. The findings suggest that there is promise in using ChatGPT as an efficient and effective tool for creating course maps, yet it still requires the domain knowledge and instructional design expertise to warrant quality and reliability of the tool.

Abstract p16 Ink4a /CDKN2A is a tumor suppressor that critically regulates the cell cycle. Indeed, p16 Ink4a deficiency promotes tumor formation in various tissues. We now report that p16 Ink4a deficiency in female mice, but not male mice, induces leanness especially in old age, as indicated by lower body weight and smaller white adipose tissue, although other major organs are unaffected. Unexpectedly, the integrity, number, and sizes of adipocytes in white adipose tissue were unaffected, as was macrophage infiltration. Hence, hypermobility appeared to be accountable for the phenotype, since food consumption was not altered. Histological analysis of the cerebellum and deep cerebellar nuclei, a vital sensorimotor control center, revealed increased proliferation of neuronal cells and improved cerebellum integrity. Expression of estrogen receptor β (ERβ) and PCNA also increased in deep cerebellar nuclei, implying crosstalk between p16 Ink4a and ERβ. Furthermore, p16 Ink4a deficiency expands LC3B+ cells and GFAP+ astrocytes in response to estrogen. Collectively, the data suggest that loss of p16 INK4a induces sexually dimorphic leanness in female mice, which appears to be due to protection against cerebellar senescence by promoting neuronal proliferation and homeostasis via ERβ.

This study compared the prevalence of depressive symptoms and suicidal ideation in individuals living alone compared with those living with others and assessed the contribution of socio-demographic factors and physical health to these differences. We analyzed 2221 individuals living alone and 19,397 individuals living with others aged 20–80 years, drawn from the Korean National Health and Nutrition Examination Survey dataset in South Korea. The study group divided into three subgroups based on age to determine whether there were differences in mental health according to age. Depressive symptoms and suicidal ideation were evaluated by self-reported questionnaires. The sex- and age-adjusted prevalence rates of depressive symptoms and suicidal ideation were higher in those living alone than those living with others. The proportion of socio-economic status and physical health explaining the differences of depressive mood and suicidal ideation between the two groups was greater in the age group over 35 years old. Considering the difference in factors that explain depressive symptoms and suicidal ideation among individuals living alone in the age group over 35 years of age and younger groups under 34 years of age, policies should be developed that will address the mental health needs of each age group.

Within engineering education, informal, out-of-school making experiences and parent-child interactions within home environments are both considered as a promising context for the development of engineering discourse and practices. However, less is known about how parents support children's engagement in engineering learning, particularly when they are foregrounded with making that use materials and technologies that can introduce sources of uncertainty. To understand both the opportunities and uncertainties of centering making within parent--child engineering learning experiences, this study examines how parents' use of epistemic supports differ between engineering design tasks with technology and engineering design tasks without technology, and within the different phases in the engineering design process. The study further investigates how parents exhibit epistemic uncertainties differently between engineering design tasks. Building on the notion of guided participation to frame engineering learning and making as co-constructed through multiple situated interactions, this study demonstrates that: (a) parents are skilled knowledge practitioners for their children's engagement of engineering learning through the use of various epistemic supports; (b) the presence of technology in the engineering design tasks prompt different types of epistemic practices and engineering design phases; (c) opportunities and tensions co-emerge when parents experience epistemic uncertainty about STEM concepts or troubleshooting during engineering design tasks with technology. We discuss implications for the design of engineering design tasks within home environments that extend the use of parents' epistemic supports.

This study compares the mental health of single parents relative to partnered parents and assesses the contribution of the social and demographic factors to this difference, examining the gender difference in it.We analyzed 12,024 single and partnered subjects, aged 30-59 years, living with children, aged 0-19 years, drawn from the 4th, 5th, and 6th Korea National Health and Nutrition Examination Survey (KNHANES) dataset in South Korea conducted from 2007-2013. Mental health was evaluated by self-reported questionnaires including depressive mood for recent two weeks, presence of suicidal ideation, and the Korean version of the Alcohol Use Disorder Identification Test. Covariates included age, physical illness, socioeconomic status (family income, recipient of national basic livelihood guarantees, educational level, house ownership, job, and residential area), family structure, and support (co-residence of another adult). Multiple logistic regression was carried out and the explained fractions of each covariate was calculated.Single parents had significantly poorer mental health than their partnered counterparts, with odds ratio (OR) of 2.02 (95% confidence interval (CI) 1.56-2.63) for depressive symptoms, 1.69 (95% CI 1.27-2.25) for suicidal ideation, and 1.74 (95% CI 1.38-2.20) for any of the three mental health statuses (suspicious depression, suicidal ideation, and alcohol dependence) after controlling for the covariates. The odds of depressive symptoms (OR = 3.13, 95% CI 2.50-3.93) and suicidal ideation (OR = 2.50, 95% CI 1.97-3.17) among both single fathers and mothers were higher than partnered parents. However, the odds of alcohol dependence were 3.6 times higher among single mothers than partnered mothers (OR = 3.58, 95% CI 1.81-7.08) and were 1.4 times greater among single fathers than partnered fathers (OR = 1.35, 95% CI 0.81-2.25). Socio-economic status explained more than 50% (except for substance use disorders) of the poorer mental health in single parents. These results were more remarkable for single fathers than for single mothers except for alcohol dependence. However, physical illness, family structure, and support made only minor contributions to single parents' mental health.This study demonstrates that single parents have poorer mental health than partnered parents. Although lower SES is an important factor explaining poorer mental health in single parents, there are other factors we cannot explain about their poor mental health. Therefore, we should pay proper regard to identifying other factors affecting mental health and to establishing policies to support single parents.
