Abstract Background Dwarf rootstocks have important practical significance for high-density planting in pear orchards. The shoots of ‘Cuiguan’ grafted onto the dwarf rootstock were shorter than those grafted onto the vigorous rootstock. However, the mechanism of shorter shoot formation is not clear. Results In this study, the current-year shoot transcriptomes and phytohormone contents of ‘CG‒QA’ (‘Cuiguan’ was grafted onto ‘Quince A’, and ‘Hardy’ was used as interstock) and ‘CG‒DL’ (‘Cuiguan’ was grafted onto ‘Duli’, and ‘Hardy’ was used as interstock) were compared. The transcriptome results showed that a total of 452 differentially expressed genes (DEGs) were identified, including 248 downregulated genes and 204 upregulated genes; the plant hormone signal transduction and zeatin biosynthesis pathways were significantly enriched in the top 20 KEGG enrichment terms. Abscisic acid (ABA) was the most abundant hormone in ‘CG‒QA’ and ‘CG‒DL’; auxin and cytokinin (CTK) were the most diverse hormones; additionally, the contents of ABA, auxin, and CTK in ‘CG‒DL’ were higher than those in ‘CG‒QA’, while the fresh shoot of ‘CG‒QA’ accumulated more gibberellin (GA) and salicylic acid (SA). Metabolome and transcriptome co-analysis identified three key hormone-related DEGs, of which two (Aldehyde dehydrogenase gene ALDH3F1 and YUCCA2) were upregulated and one (Cytokinin oxidase/dehydrogenase gene CKX3) was downregulated. Conclusions Based on the results of transcriptomic and metabolomic analysis, we found that auxin and CTK mainly regulated the shoot differences of ‘CG–QA’ and ‘CG–DL’, and other hormones such as ABA, GA, and SA synergistically regulated this process. Three hormone-related genes ALDH3F1, YUCCA2, and CKX3 were the key genes contributing to the difference in shoot growth between ‘CG–QA’ and ‘CG–DL’ pear. This research provides new insight into the molecular mechanism underlying shoot shortening after grafted onto dwarf rootstocks.

Unlike traditional ridging, mulching broad ridges with a woven polypropylene fabric (WPF) can reduce soil evaporation during the drought season and avoid long saturation time in the root zone of pear trees during the rainy season. In this study, field experiments were conducted from 2017 to 2020 in a pear orchard in the North China Plain to investigate the effects of mulching broad ridges (0.3 m in height and 2 m in width) with WPF on soil temperature and moisture, nitrogen leaching, vegetative and reproductive growth of young pear trees (Pyrus bretschneideri Rehd. ‘Yuluxiang’). The experiments involved two treatments, namely, control (traditional no-ridge planting without mulching) and mulching broad ridges with WPF (RM treatment). The results showed that the RM treatment increased soil moisture and temperature and decreased nitrogen leaching, resulting in vigorous growth of the young pear trees. Moreover, the RM treatment increased the tree trunk cross-sectional area and height of the young pear trees by 37% and 8% in 2020, respectively. The nitrate nitrogen content at the soil layer depth of 0–30 cm was significantly higher in the RM than that in control. Furthermore, the RM treatment significantly increased the fruit yield due to larger tree size. In addition, compared with control, significantly higher fruit soluble solid content of RM treatment was detected in 2020. High precipitation (423 mm) occurred during fruit enlargement stage in 2020, RM treatment decreased the rainfall infiltration in the ridge and the soil moisture in root region, resulting in the improvement of fruit quality, compared with control. Therefore, mulching broad ridges with WPF can be implemented to increase soil moisture during drought season, soil temperature, and nitrate nitrogen content, thereby improving the growth and fruit yield of young pear trees. Additionally, it can reduce soil moisture in the root zone during the rainy season and improve the fruit quality of the trees. Finally, it can reduce nitrate nitrogen leaching, thereby reducing environmental pollution.

Abstract The data described in this paper were collected from the Circum-Bohai Bay and Loess Plateau regions of northern China. Soil, leaf and fruit nutrients from 225 typical pear orchards in these regions were measured. Soil data included pH, organic matter, total N, alkaline hydrolysable N, available P and available K concentrations of 3 different soil layers, 0–20 cm, 20–40 cm and 40–60 cm, from different orchards. Leaf and fruit data included N, P, K, Ca, Fe, Mn, Cu, Zn and B concentrations of pear trees from different orchards. These data can be used to assess the soil nutrient supply and leaf and fruit nutrient status of pear orchards in two major producing areas, Circum-Bohai Bay and Loess Plateau. Additionally, this dataset provides data to support the development of regionalized and standardized soil nutrient management programs for pear orchards, as well as regionalized layouts of the main varieties in the two producing areas.

IntroductionQuince A (Cydonia oblonga Mill.), a typical dwarfing rootstock in pear cultivation, is susceptible to iron (Fe) deficiency in calcareous soils. The aim of this study was to compare the strategies in Fe uptake and utilization in dwarfing rootstock quince A (low Fe efficiency) versus a typical vigorous rootstock Pyrus betulifolia (PB) with high Fe efficiency.MethodsQuince A and PB were grown in nutrient solution (pH 6.3) for 4 weeks followed by three pH treatments: pH6.3, pH8.3a (adjusted with hydroxide) and pH8.3b (adjusted with bicarbonate). The Fe uptake and utilization indicators of the rootstocks were assessed at the onset of chlorosis symptoms (after 58 days of treatments).Results and discussionIn contrast to PB, quince A exhibited Fe deficiency chlorosis under bicarbonate (pH8.3b). Bicarbonate stimulated the root proton secretion, inhibited root growth and ferric chelate reductase (FCR) activity in both PB and quince A, whereas high pH without bicarbonate (pH8.3a) stimulated only root proton release. Both species accumulated more Fe in roots under high pH treatments than under pH6.3, resulting in Fe sufficiency in leaves. Both high pH treatments increased the activity of leaf FCR in PB and quince A. However, extractable Fe(II) concentration in leaves was increased by high pH treatments in PB only. This study demonstrated that depressed Fe(III) reduction in leaves caused by bicarbonate rather than high pH explained Fe deficiency in quince A grown in bicarbonate-containing medium.

Determination of Occidental pear (Pyrus communis) ripening is difficult because the appearance of Occidental pears does not change significantly during the ripening process. Occidental pears at different ripening stages release different volatile organic compounds (VOCs), which can be used to determine fruit ripeness non-destructively and rapidly. In this study, VOCs were detected using proton-transfer-reaction mass spectrometry (PTR-MS). Notably, data were acquired within 1 min. Occidental pears harvested at five separate times were divided into three ripening stages: unripe, ripe, and overripe. The results showed that the composition of VOCs differed depending on the ripening stage. In particular, the concentrations of esters and terpenes significantly increased during the overripe stage. Three ripening stages were clearly discriminated by heatmap clustering and principal component analysis (PCA). This study provided a rapid and non-destructive method to evaluate the ripening stages of Occidental pears. The result can help fruit farmers to decide the optimum harvest time and hence reduce their economic losses.

Owing to iron chlorosis, pear trees are some of the most severely impacted by iron deficiency, and they suffer significant losses every year. While it is possible to determine the iron content of leaves using laboratory-standard analytical techniques, the sampling and analysis process is time-consuming and labor-intensive, and it does not quickly and accurately identify the physiological state of iron-deficient leaves. Therefore, it is crucial to find a precise and quick visualization approach for metabolites linked to leaf iron to comprehend the mechanism of iron deficiency and create management strategies for pear-tree planting. In this paper, we propose a micro-Raman spectral imaging method for non-destructive, rapid, and precise visual characterization of iron-deficiency-related metabolites in pear leaves. According to our findings, iron deficiency significantly decreased the Raman peak intensities of chlorophylls and lipids in leaves. The spatial distributions of chlorophylls and lipids in the leaves changed significantly as the symptoms of iron insufficiency worsened. The technique offers a new, prospective tool for rapid recognition of iron deficiency in pear trees because it is capable of visual detection of plant physiological metabolites induced by iron deficiency.

To understand the soil nutrient status of pear orchards in Beijing, we investigated their fertilization situation, including the fertilizer type, amount, and period. Furthermore, soil samples were collected at a depth of 0 to 40 cm to determine the contents of soil nitrogen, phosphorus, and potassium. The status of nutrient profits and losses was analyzed. The results showed that 50% of the pear orchards received organic fertilizer as a single nutrient source, and 35.7% of the pear orchards received a combined application of organic fertilizer and chemical fertilizer. Most pear orchards received organic fertilizer in autumn, but the application of chemical fertilizer occurred mainly before germination and during fruit expansion. The average nutrient input to the investigated pear orchards was 569.6 kg/ha for N, 855.0 kg/ha for P2O5, and 448.1 kg/ha for K2O, and the corresponding proportion of organic fertilizer was 76.9%, 88.0%, and 85.8%, respectively. However, the pear orchards had surpluses of nitrogen, phosphorus, and potassium, with average surplus amounts as high as 445.5, 794.3, and 321.4 kg/ha, respectively. Among all pear orchards surveyed, 93.33% faced environmental risks and 37.04% faced leaching risks. The average content of soil phosphorus was 2.23 times its critical value, and 64.29% of the studied pear orchards exceeded the critical value. Most pear orchards had surplus potassium, with 26.92% exceeding 500 kg/ha. This study provides a basis for soil improvement, high-quality production of fruits, and efficient utilization of pear orchards in Beijing.

Nitrogen is one of the essential elements for fruit tree growth. Therefore, a 15N tracing experiment was conducted to investigate the characteristics of nitrogen uptake and distribution in young pear trees. The experiment included: groups A and B, fertilized with 15 g unlabeled and 5 g labeled N per tree in the spring of 2017; and group C, fertilized with 20 g unlabeled N per tree in the spring of 2017 and 25 g unlabeled and 5 g labeled N per tree in the spring of 2018. Results indicated that new organs grew vigorously before June, and about 50% of the fertilizer nitrogen was concentrated in leaves. From June to November, the growth rate of storage organs was faster than that of new organs, and fertilizer nitrogen was mainly stored in the shoots, trunk, and roots. During the defoliation period, about 84% of the leaf fertilizer nitrogen was mobilized to the shoots, trunk, and roots. There was significant correlation between current fertilizer nitrogen and the dry mass weight of each organ after the completion of the nitrogen cycling of a whole year; the correlation coefficient was 0.98. Fertilizer nitrogen use efficiency increased with the ages of the pear trees. Therefore, nitrogen fertilizer rate should be formulated according to the tree growth characteristics and increased with the ages of the young pear trees; topdressing fertilizer should be applied in summer to ensure trunk and root development.

A field experiment was conducted to evaluate the effects of regulated deficit irrigation (RDI) on the fine root redistribution of mature pear trees in 2009 and 2010. The experiment consisted of four RDI treatments: MRDI-1 and SRDI-1, in which the trees received irrigation replacing 60% and 40% of pan evaporation (Ep) during Stage 1 (cell division stage), and MRDI-1+2 and SRDI-1+2, in which the trees received irrigation replacing 60% and 40% of Ep during Stage 1+2 (cell division and slow shoot growth stage). All the RDI-treated trees received irrigation replacing 80% of Ep (full irrigation) in other stages, and the control trees were fully irrigated during the whole growth season. The results showed that the fine root length density (RLD) of mature pear trees was reduced by water stress. The resumption of full irrigation boosted fine root growth. The RLD of the SRDI-1-treated trees in the irrigated zones recovered in early July, they maintained water and nutrient absorption during the fruit enlargement stage, and the final fruit yield was significantly improved. The RLD of trees in the irrigated zones with MRDI-1 and MRDI-1+2 recovered in July and September, respectively, but there were no significant differences in fruit yield between the MRDI-1, MRDI-1+2, and the control. This indicates that the fruit yield was not negatively or positively impacted by the redistribution of moderate water stress applied during either Stage 1 or Stage 1+2.

In North China, the high-quality pear cultivar “Huangguan” (Pyrus bretschneideri Rehd. cv), which is grafted onto dwarf quince A (Cydonia oblonga Mill. cv) rootstock and grown in calcareous soil, experiences severe iron (Fe) deficiency; this deficiency greatly constrains tree growth as well as fruit yield and quality. Therefore, we evaluated the effects of six practices for alleviating chlorosis caused by Fe deficiency in “Huangguan” grafted onto quince A (HG-QA). The practices included ridging with landscape fabric mulching as a control, flattening with landscape fabric mulching (FM), ridging without landscape fabric mulching (R), flattening without landscape fabric mulching (F), Fe fertilizer application in soil (SFe), foliar Fe application (FFe), and manure application (M). The results showed that the leaf Fe concentration increased by 356% under FFe, compared to that under the control, but the practice failed to alleviate Fe deficiency chlorosis. In contrast, an increase in leaf Fe concentration and chlorosis alleviation were observed under F. F alleviated chlorosis mainly by increasing the root ferric-chelate reductase activity. These results indicate that Fe uptake and utilization in leaves are independent biochemical processes and soil aeration improvement have positive effect on increasing Fe uptake. M improved both the soil active Fe concentration and leaf Fe utilization. Thus, manure application should be the first choice for alleviating Fe deficiency chlorosis in HG-QA grown in calcareous soils. Combining manure application with other practices that increase Fe uptake would likely be an effective way to address the problem of Fe deficiency chlorosis.

The remobilization of storage nitrogen (N) is affected by growth characteristics of young pear trees. Aboveground parts of young pear trees grafted on P. betulifolia grew more vigorously than that on dwarfing rootstocks. Therefore, the remobilization of storage N within young pear trees on vigorous rootstocks may be different from that on dwarfing rootstocks. A 15N tracing experiment, including six groups of one-year-old pear trees grafted on vigorous rootstocks in 2016, was conducted to investigate the mobilization of storage N from 2016 to 2018. Results indicated that about 44%, 31.4% and 24.6% of storage N remobilized in new growth was derived from the trunk, shoots and roots, respectively. Most of storage N remobilized in new organs were supplied by trunks and shoots. About 82.2% of storage N withdrawn from senescent leaves were recovered in the trunk and shoots during autumn. The aboveground parts played a more important role than roots in the cycling process of storage N in young pear trees. However, as compared with young pear trees on dwarfing rootstocks, more storage N recovered in new organs were supplied by roots of that on vigorous rootstocks, due to vigorous growth and more nutrient requirement of aboveground parts.

We used DNA-based pyrosequencing to characterize the bacterial community structure of the sandy soil of an apple orchard with different manure ratios. Five manure percentages (5%, 10%, 15%, 20% and 25%) were examined. More than 10,000 valid reads were obtained for each replicate. The communities were composed of five dominant groups (Proteobacteria, Actinobacteria, Chloroflexi, Acidobacteria and Bacteroidetes), of which Proteobacteria content gradually decreased from 41.38% to 37.29% as manure ratio increased from 0% to 25%, respectively. Redundancy analysis showed that 37 classes were highly correlated with manure ratio, 18 of which were positively correlated. Clustering revealed that the rhizosphere samples were grouped into three components: low manure (control, 5%) treatment, medium manure (10%, 15%) treatment and high manure (20%, 25%) treatment. Venn analysis of species types of these three groups revealed that the bacteria community difference was primarily reflected by quantity ratio rather than species variety. Although greater manure content led to higher soil organic matter content, the medium manure improved soil showed the highest urease activity and saccharase activity, while 5% to 20% manure ratio improvement also resulted in higher bacteria diversity than control and 25% manure ratio treatment. Our experimental results suggest that the use of a proper manure ratio results in significantly higher soil enzyme activity and different bacteria community patterns, whereas the use of excessive manure amounts has negative effect on soil quality.
