Objective To compare the disinfection effect of hydrogen peroxide low temperature plasma sterilization and traditional lens disinfection on rigid gas permeable contact lens (RGP), so as to improve the clinical management procedure of RGP trial lens. Methods A total of 90 RGP trial lenses used by patients in our hospital from April to August 2020 were collected and randomly divided into 3 groups. Group A was treated with modified hydrogen peroxide low temperature plasma sterilizing method (n=30); group B was disinfected with conventional conditioning solution (n=30); and group C was sterilized with anti-bacterial case made of polymer injection material (n=30). The preserving solution of the 3 groups was respectively cultured for 5 d and then tested with the automatic microbial culture system of American Simmer TREK 528, and a comparison of the sterilization effect was made among the groups. The test was performed once a month for 3 months. Moreover, patients' ocular surface condition was examined with a slit lamp, and the comfort level after wearing was evaluated through questionnaire survey. Results The bacteria culture of group A was negative with the sterilization effect of 100.0%, while abnormal bacteria were detected 2 cases in group B and 1 case in group C, with the disinfection rates of 97.7% and 98.9%, respectively, though there was no statistical difference among the 3 disinfection methods. In addition, the results of corneal examination and comfort questionnaire survey showed no statistical difference among the groups as well. Conclusion Hydrogen peroxide low-temperature plasma sterilization has a thorough disinfection effect with a safe and simple procedure, and is worth popularizing.

Abstract Background The prognosis of human glioma is poor, and the highly invasive nature of the disease represents a major impediment to current therapeutic modalities. The oncoprotein B-cell-specific Moloney murine leukemia virus integration site 1 protein (Bmi-1) has been linked to the development and progression of glioma; however, the biological role of Bmi-1 in the invasion of glioma remains unclear. Methods A172 and LN229 glioma cells were engineered to overexpress Bmi-1 via stable transfection or to be silenced for Bmi-1 expression using RNA interfering method. Migration and invasiveness of the engineered cells were assessed using wound healing assay, Transwell migration assay, Transwell matrix penetration assay and 3-D spheroid invasion assay. MMP-9 expression and activity were measured using real-time PCR, ELISA and the gelatin zymography methods. Expression of NF-kappaB target genes was quantified using real-time PCR. NF-kappaB transcriptional activity was assessed using an NF-kappaB luciferase reporter system. Expression of Bmi-1 and MMP-9 in clinical specimens was analyzed using immunohistochemical assay. Results Ectopic overexpression of Bmi-1 dramatically increased, whereas knockdown of endogenous Bmi-1 reduced, the invasiveness and migration of glioma cells. NF-kappaB transcriptional activity and MMP-9 expression and activity were significantly increased in Bmi-1-overexpressing but reduced in Bmi-1-silenced cells. The reporter luciferase activity driven by MMP-9 promoter in Bmi-1-overexpressing cells was dependent on the presence of a functional NF-kappaB binding site, and blockade of NF-kappaB signaling inhibited the upregulation of MMP-9 in Bmi-1 overexpressing cells. Furthermore, expression of Bmi-1 correlated with NF-kappaB nuclear translocation as well as MMP-9 expression in clinical glioma samples. Conclusions Bmi-1 may play an important role in the development of aggressive phenotype of glioma via activating the NF-kappaB/MMP-9 pathway and therefore might represent a novel therapeutic target for glioma.

Abstract Background Overexpression of sphingosine kinase-1 (SPHK1) has been demonstrated to be associated with the development and progression in various types of human cancers. The current study was to characterize the expression of SPHK1 in salivary gland carcinomas (SGC) and to investigate the association between SPHK1 expression and progression of SGC. Methods The expression of SPHK1 was examined in 2 normal salivary gland tissues, 8 SGC tissues of various clinical stages, and 5 pairs of primary SGC and adjacent salivary gland tissues from the same patient, using real-time PCR and western blot analysis. Furthermore, the SPHK1 protein expression was analyzed in 159 clinicopathologically characterized SGC cases by immunohistochemistry. Statistical analyses were performed to determine the prognostic and diagnostic associations. Results SPHK1 expression was found to be markedly upregulated in SGC tissues than that in the normal salivary gland tissues and paired adjacent salivary gland tissues, at both mRNA and protein levels. Statistical analysis revealed a significant correlation of SPHK1 expression with the clinical stage (P = 0.005), T classification (P = 0.017), N classification (P = 0.009), M classification (P = 0.002), and pathological differentiation (P = 0.013). Patients with higher SPHK1 expression had shorter overall survival time, whereas patients with lower SPHK1 expression had better survival. Importantly, patients in the group without adjuvant therapy who exhibited high SPHK1 expression had significantly lower overall survival rates compared with those with low SPHK1 expression. Moreover, multivariate analysis suggested that SPHK1 expression might be an independent prognostic indicator for the survival of SGC patients. Conclusions Our results suggest that SPHK1 expression is associated with SGC progression, and might represent as a novel and valuable predictor for adjuvant therapy to SGC patients.

To address residual plastic film in fields in which mulched drip irrigation technology is applied, a sprayable degradable film (consisting of 1–5% clay, 5–20% humic acid, 0.5–5% crosslinker, and 0.5–5% auxiliary and water) can be used as a superior alternative to the plastic film applied in drip-irrigated fields. A field experiment was conducted in Xinjiang, Northwest China, to test the impacts of five different mulching treatments (SF1, sprayable degradable film applied at 1900 kg ha−1; SF2, sprayable degradable film applied at 1900 kg ha−1; SF3, sprayable degradable film applied at 2500 kg ha−1; PF, plastic film; and NF, no film mulching) on cotton growth and development, yield, and water use efficiency. The results showed that, compared to the NF treatment, sprayable degradable film mulching (SF1, SF2, and SF3) positively impacted the soil hydrothermal environment, promoted root growth, significantly increased plant height and leaf area, and enhanced physiological characteristics, which, in turn, increased yield and water use efficiency by 11.79–15.00% and 21.88–30.21%, respectively. The maximum yield and water use efficiency were observed in the PF treatment, amounting to 5345 kg ha−1 and 1.28 kg m−3, respectively, and they had no significant differences from those in the SF3 treatment. In general, applying moderate amounts of sprayable degradable film at a rate of 2500 kg ha−1 represents an effective agronomic strategy for managing residual film contamination while maintaining stable cotton yields. In order to study the deformation and failure laws of roadway surrounding rock under the condition of layered rock, the one orbit lane of Shandong Yuncheng Coal Mine was taken as the research background, and PFC2D discrete element software was adopted to study the stress, deformation and crack development of surrounding rock during excavation. Meanwhile, the influence of rock thickness variation on deformation and failure of surrounding rock was analyzed. The results show that: after the excavation of layered rock roadway, the radial displacement of surrounding rock roof will have a non-smooth transition in shallow of mudstone about 1 m, while the surrounding rock floor in the mudstone will overall uplift and appear phenomenon of separation; the surrounding rock crack of straight wall arch roadway will first appear at the vault and wall foot on both sides, with the increase of the excavation time, the crack of the roadway will gradually extend to the top and two sides, which causes a roof fall accident; with the increase of mudstone layer thickness, the displacement and failure area of the roadway surrounding rock will both decrease first and then increase.
