Objective To exploring the effect of human amniotic mesenchymal stem cells(hAMSCs) on the repair of rat uterine scars. Methods The hAMSCs were isolated and cultured,Female SPF grade SD rats were selected for full-thickness incision of uterine wall and then implanted with hAMSCs. On the 30th day after operation, the uterine incision was examined histologically.ImageJ image analysis software was used to analyze and to compare the thickness of uterine myometrium and the percentage of fibrotic area in each group.Immunohistochemical method was used for detecting the percentage of positive areas of α-SMA, TGF-β1, and Ki-67. Results Compared with the PBS group, the hAMSCs group showed significant thickening of the uterine muscle layer and fibrotic area was decreased, The positive expression of α-SMA, and Ki-67 significantly increased(P＜0.05), while the expression of TGF-β1 was significantly reduced(P＜0.05). Conclusions The hAMSCs may promote the repair of uterine incision scars by reducing the formation of scar fibrosis and promoting the proliferation of smooth muscle cells in uterine scars.

Purposes In order to obtain additional microstructural information on the ConMoS(n＝1-5) cluster and establish a theoretical foundation for subsequent investigations of its electronic properties and catalytic performance, the atomic interaction relationships and frontier orbital compositions of the cluster are investigated. Methods To accomplish this, density functional theory with the B3LYP functional and def2-TZVP basis set is adopted to optimize the cluster configuration and perform theoretical analysis under various spin multiplicities with the Gaussian09 software package. Findings The results demonstrate the existence of 21 stable configurations. AIM theory, electron localization function, and deformation density are analyzed to obtain information on the degree of internal interaction and electron density transfer trends of the cluster. The findings indicate that covalent bonding interactions between Co, Mo, and S atoms are strong in the internal atom interactions of the cluster, with S atoms exhibiting high localization and low probability of electron escape from the domain. In contrast, Co and Mo atoms exhibit strong delocalization and are likely to exchange electrons with the outside electrons. Through Hirshfeld analysis of atomic contribution rates, it can be observed that metal atoms are the primary contributors to the HOMO and LUMO orbitals, with Mo and Co atoms dominating the frontier orbitals. Overall, covalent bonding is the primary atomic interaction within the cluster, but the strength of these interactions varies. Mo and Co atoms should be the focus in chemical reactions of the cluster.

The Late Ediacaran Dengying Formation, located in the Sichuan Basin of the northwestern Yangtze Craton, is of significant interest in oil and gas exploration due to its abundant pores and vugs within microbial mound-shoal complexes. However, there is still uncertainty regarding the spatiotemporal distribution and controlling factors of the platform margin. This study comprehensively analyzes the retrogradation pattern of the Dengying Formation platform margin using seismic data, well logs, field outcrops, and petrological characteristics. Our findings reveal that the Dengying Formation strata surrounding the rift basin at the northwestern of the Yangtze Craton can be divided into three main depositional facies: basin facies, slope facies, and platform margin facies. Additionally, based on the integration of lithological, log, and seismic characteristics, the Dengying Formation is subdivided into four third-order sequences, with five sequence boundaries and three seismic facies identified. Supported by sequence stratigraphy and geophysical data, we have reconstructed the tectono-sedimentary evolution of the multiple platform margins on the eastern side of the Deyang-Anyue rift in the Sichuan Basin during the late Ediacaran. Our findings indicate that the platform underwent two phases of retrogradation. The second-stage platform margin underwent retrogradation towards the interior, spanning a distance between 10 and 80 km, based on the initial configuration established by the first-stage platform margin. The main controls for progradation and retrogradation of carbonate platforms are eustatic sea-level changes and tectonic activity. Eustatic sea-level changes can be divided into constructive and destructive phases. Constructive phases are commonly observed in highstand systems tracts, while destructive phases are often associated with transgressive systems tracts and are related to platform retrogradation processes. However, sea-level changes alone cannot fully control the process of platform retrogradation. The thermal subsidence following mantle plume events likely played a significant role in the retrogradation of the platform in the study area. During this period, tectonic processes controlled the geometry of the platform and the deposition of carbonates in the platform margin-slope-basin environment. Additionally, karst-related mound-shoal complexes developed extensively along the platform margin of the Dengying Formation in the northwestern Yangtze Craton. The Lower Cambrian dark shales represent high-quality hydrocarbon source rocks, while the Dengying Formation exhibits an optimal source-reservoir configuration.

ABSTRACT: Nutritional therapy, which may have advantages over medication, is being investigated as a novel treatment for pregnancy-induced hypertension. Several studies have shown that probiotic yogurt supplementation during pregnancy has beneficial effects on maternal and fetal health. In this study, fermented buffalo milk was produced with yogurt culture and Lactobacillus plantarum B, a probiotic isolated from healthy breast milk with high angiotensin-converting enzyme inhibitory activity. The fermentation conditions under which the angiotensin-converting enzyme (ACE) inhibitory activity reached 84.51% were optimized by the response surface method as follows: 2 × 106 cfu/mL of L. plantarum B, yogurt culture 2.5 × 105 cfu/mL, and 8 h at 37°C. The distribution of ACE inhibitory peptides from fermented buffalo milk and fermented cow milk were further analyzed by liquid chromatography-mass spectrometry. By searching according to the structural features of ACE inhibitory peptides, 29 and 11 peptides containing ACE inhibitory peptide features were found in fermented buffalo milk and fermented cow milk, respectively. To investigate the in vivo antihypertensive activity of fermented buffalo milk, 18 pregnant rats were divided into 3 groups (n = 6 in each group) and administered 10 mL of normal saline, yogurt (20 mg/kg), or labetalol hydrochloride (4 mg/kg) daily from the beginning of pregnancy to parturition. To induce hypertension, methyl nitrosoarginine (125 mg/kg) was injected subcutaneously every day from d 15 of pregnancy to the day of delivery. Blood pressure was not significantly changed in the yogurt and labetalol groups after induction of hypertension and was lower compared with the normal saline group, but there was no difference between the yogurt and labetalol groups. This implied that the buffalo yogurt had a preventive and antihypertensive effect in the pregnancy-induced hypertensive rat model. Further studies to determine the mechanism of action, as well as a randomized control trial, are warranted.

In this study, we enhanced the CaffeNet network for recognizing students’ facial expressions in a music classroom and extracted emotional features from their expressions. Additionally, students’ speech signals were processed through filters to identify emotional characteristics. Using the LRLR fusion strategy, these expression and speech-based emotional features were combined to derive multimodal fusion emotion results. Subsequently, a music teaching model incorporating this multimodal emotion recognition was developed. Our analysis indicates a mere 6.03% discrepancy between the model’s emotion recognition results and manual emotional assessments, underscoring its effectiveness. Implementation of this model in a music teaching context led to a noticeable increase in positive emotional responses—happy and surprised emotions peaked at 30.04% and 27.36%, respectively, during the fourth week. Furthermore, 70% of students displayed a positive learning status, demonstrating a significant boost in engagement and motivation for music learning. This approach markedly enhances student interest in learning and provides a solid basis for improving educational outcomes in music classes.

ObjectiveTo explore the absorption， distribution， accumulation， and excretion after oral exposure to lanthanum and the effects of lanthanum on the blood element spectrum of rats.Methods48 female SPF-grade rats were randomly divided into 8 groups with a one-time gavage of 700 mg/kg·BW lanthanum nitrate solution using single oral exposure experiment. And the blood， heart， liver， spleen， lung， kidneys， brain， femur， and fecal of rats were collected and measured by ICP-MS after gavage for 1， 2， 4， 8， 16， 32， 64， and 128 h. Another 48 SPF-grade rats were randomly divided into 4 groups （half male and female）， each group with a control and an experimental group was observed in experimental time-point in repeat oral exposure experiment. The experimental group repeatedly gavage lanthanum nitrate solution containing lanthanum 66 mg/kg·BW， and the control group gavage an equal amount of physiological saline. Then， on the 7th， 14th， 21st and 28th days， the rats （including the control group and the experimental group） were taken to determine the contents of 44 elements in blood and the contents of lanthanum in liver， kidney， femur， feces and urine by ICP-MS.ResultsAfter single oral exposure to lanthanum， lanthanum peaked in blood for 2 h， the elimination half-life was 69.315 h， and the overall clearance rate was 1.733 L/h/kg. Lanthanum was widely distributed in the heart， liver， spleen， lung， kidneys， brain， and femur with blood， and the level in the femur was the highest （P0.05）.ConclusionIn this experiment， lanthanum can be rapidly absorbed into the blood after oral exposure and is widely distributed in the heart， liver， spleen， lung， kidneys， brain， and femur. After repeat oral exposure to lanthanum， lanthanum is easily deposited in the liver and femur， feces excretion is the main route of elimination， and no effect of lanthanum on the blood element spectrum of rats has been observed.

Labeled data is widely used in various classification tasks. However, there is a huge challenge that labels are often added artificially. Wrong labels added by malicious users will affect the training effect of the model. The unreliability of labeled data has hindered the research. In order to solve the above problems, we propose a framework of Label Noise Filtering and Missing Label Supplement (LNFS). And we take location labels in Location-Based Social Networks (LBSN) as an example to implement our framework. For the problem of label noise filtering, we first use FastText to transform the restaurant's labels into vectors, and then based on the assumption that the label most similar to all other labels in the location is most representative. We use cosine similarity to judge and select the most representative label. For the problem of label missing, we use simple common word similarity to judge the similarity of users' comments, and then use the label of the similar restaurant to supplement the missing labels. To optimize the performance of the model, we introduce game theory into our model to simulate the game between the malicious users and the model to improve the reliability of the model. Finally, a case study is given to illustrate the effectiveness and reliability of LNFS.

Currently, China is in the period of rapid urbanization and gradual improvement of the rail transportation network. With the increasing population in towns, the construction of many cities has become blind and disorderly to meet people’s housing needs, resulting in traffic congestion, uneven distribution of resources, and other “big city diseases” more serious. From the urban construction experience of developed countries, the mode that urban rail transportation and urban development complement each other and develop together, that is, the regional-wide TOD construction, is an effective way to promote sustainable urban development. This paper focuses on how to select the stations of the rail transit for priority development of TOD projects (TOD development advance sites) within the whole area of the town. When selecting stations, the investment potential and regional driving ability of each station are taken into consideration to make a comprehensive evaluation of each rail transit station, so that the TOD development advance sites can drive the gradual development of its surrounding areas and finally form an urban renewal pattern of interoperability and integrated development of the whole area. With this method, we can effectively solve the current situation of blind and disorderly construction of TOD and various problems in urban development in China, and help to build efficient, green, and livable cities in various places.

Rationale: Acute exacerbation of idiopathic pulmonary fibrosis (AE-IPF) has a poor prognosis and high mortality. However, there is limited information regarding the mechanisms of AE-IPF. Aims: We aimed to explore the function of macrophage-inducible C-type lectin (Mincle) in AE-IPF. Methods: In the present study, Mincle was detected in the lung tissues of AE-IPF patients. Mincle-deficient (Mincle-/-) mice and wild-type C57BL/6 mice were administered bleomycin (BLM), followed by HSV1 viral infection to establish the AE-IPF model. Results: Mincle was increased in the lung tissues of AE-IPF patients compared with those with stable IPF (P = 0.04) and healthy controls (P = 0.009). The survival rate of the Mincle-/-+BLM+HSV group was higher than that of the WT+BLM+HSV group. The mice in the Mincle-/-+BLM+HSV group exhibited milder inflammation and lower acute lung injury scores (P = 0.008). Mincle was expressed on inflammatory monocytes and neutrophils (CD11b+Gr1 +F4/80-) and monocyte-derived macrophages (Mo-AMs, CD11b+Gr1 +F4/80 +) in the BALF of AE-IPF mice. With the completion of Chinese BeiDou Navigation Satellite System (BDS), the world has begun to enjoy the Positioning, Navigation, and Timing (PNT) services of four Global Navigation Satellite Systems (GNSS). In order to improve the GNSS performance and expand its applications, Low Earth Orbit (LEO) Enhanced Global Navigation Satellite System (LeGNSS) is being vigorously advocated. Combined with high-, medium-, and low- earth orbit satellites, it can improve GNSS performance in terms of orbit determination, Precise Point Positioning (PPP) convergence time, etc. This paper comprehensively reviews the current status of LeGNSS, focusing on analyzing its advantages and challenges for precise orbit and clock determination, PPP convergence, earth rotation parameter estimation, and global ionosphere modeling. Thanks to the fast geometric change brought by LEO satellites, LeGNSS is expected to fundamentally solve the problem of the long convergence time of PPP without any augmentation. The convergence time can be shortened within 1 minute if appropriate LEO constellations are deployed. However, there are still some issues to overcome, such as the optimization of LEO constellation as well as the real time LEO precise orbit and clock determination.

This paper aims to classify the information on insecticidal proteins from microorganism over the world, and to provide fresh opinions for the application of insecticidal proteins in agriculture, forestry and other industries in China.

With the development of global positioning and radar technology,more and more trajectory data can be collected.In particular,trajectories generated by aircrafts,ships,migratory birds are complicated and varied,and free from any constraints on the ground.For helping identifying the behaviors and intention of the flying objects,the recognition of the type of the aircraft tra-jectories has important value.Specifically,on the basis of identifying frequent route patterns,the paper proposes a new method,consisting of a frequent route patterns extracting algorithm and a convolution neural network model.The extracting algorithm first gets key points from the compressed trajectory,next finds the closed routes through the self-intersecting points of the trajectory,then discovers frequent patterns in the closed routes and treats them as the basis of classification.Further,the model recognizes the trajectory type via image analyses.This paper conducts extensive experiments on the real aircraft trajectory data disclosed on the FlightRadar24 website as well as the simulated data.The experimental results show that our method can effectively identify complex trajectory types.Compared with LeNet-5 CNN classification without trajectory extraction,our method has the superior performance,achieving an average accuracy of more than 95% for trajectory classification.

MRI is the most clear and accurate imaging technique for detecting muscle involvement. In recent years, MRI has been widely used in diagnosis and follow⁃up of muscle disorders. A variety of muscle disorders have specific selective muscle involvement patterns. There is accumulating evidence that patterns of involved muscles shown by muscle MRI can help delineate the possible diagnoses. The aim of this paper is to state MRI manifestations of hereditary myopathies, so as to improve neurologist's understanding of muscle MRI and promote application of muscle MRI in diagnosis of hereditary myopathies.

Circular RNA (CircRNA) is a newly identified special class of non-coding RNA (ncRNA) that plays an important regulatory role in the progression of certain diseases. Herein, our results indicate that CircMEG3 is downregulated expression and negatively correlated with the expression of telomerase-related gene Cbf5 in human liver cancer. Moreover, CircMEG3 inhibits the growth of human liver cancer stem cells in vivo and in vitro. CircMEG3 inhibits the expression of m6A methyltransferase METTL3 dependent on HULC. Moreover, CircMEG3 inhibits the expression of Cbf5, a component of telomere synthetase H/ACA ribonucleoprotein (RNP; catalyst RNA pseudouracil modification) through METTL3 dependent on HULC. Thereby, CircMEG3 inhibits telomerase activity and shortens telomere lifespan dependent on HULC and Cbf5 in human liver cancer stem cell. Strikingly, increased Cbf5 abrogates the ability of CircMEG3 to inhibit malignant differentiation of human liver cancer stem cells. In summary, these observations provide important basic information for finding effective liver cancer therapeutic targets.

Objective To summarize the sociodemographic information, injury data, treatment methods and prognosis of patients with traumatic brain injury (TBI) in Guizhou Province, and to explore the treatment ideas of TBI in this area. Methods A total of 1931 patients with TBI who were diagnosed and treated in Department of Neurosurgery, The Affiliated Hospital of Guizhou Medical University from January 2016 to December 2018 were recruited by self⁃designed data questionnaire. The clinical data included gender, age, occupation, area of injury, cause of injury, type of injury, location of injury, severity of injury, whether the midline deviated, location of hematoma, combined injury, whether pre⁃hospital first aid, whether surgical treatment, operation time window, postoperative complications and infection during hospitalization. Glasgow Outcome Scale (GOS) was used to evaluate the prognosis of patients. Results According to GOS score, there were 1488 cases (77.06%) with good recovery, 134 cases (6.94%) with mild disability, 101 cases (5.23%) with severe disability, 134 cases (6.94%) with vegetative survival and 74 cases (3.83%) with death. 1) Social demographic data: there were significant differences in GOS scores between different ages (F = 6.411, P = 0.000) and occupations (F = 5.446, P = 0.000). The prognosis of 46-55 years old patients was worse than that of 5-15 years old patients (t = 5.047, P = 0.000). The prognosis of self ⁃ employed workers was worse than that of preschool children (t = ⁃ 3.570, P = 0.021), students (t = ⁃ 5.050, P = 0.000) and cooks (t = ⁃ 3.763, P = 0.013). There was no significant difference in the prognosis between different genders (t = 0.123, P = 0.902). 2) Injury data: there were significant differences in the prognosis among different injury causes (F = 2.585, P = 0.017), injury sites (F = 5.314, P = 0.000) and injury severity (F = 238.321, P = 0.000). The prognosis of traffic injury was worse than that of strike injury (t = ⁃ 3.731, P = 0.004). The prognosis of basal ganglia injury was worse than that of cerebellar injury (t = ⁃ 3.340, P = 0.002). The prognosis of severe (t = 15.983, P = 0.000) and medium (t = 5.711, P = 0.000) patients was worse than that of mild, and severe patients was worse than that of the medium (t = 9.130, P = 0.000). The prognosis of no⁃Guiyang patients was worse than that of patients in Guiyang (t = ⁃ 2.231, P = 0.026). The prognosis of open injury patients was worse than that of closed injury patients (t = ⁃ 3.069, P = 0.002), subdural hematoma patients was worse than that of epidural hematoma patients (t = 4.559, P = 0.000), and the prognosis of patients with midline shift was worse than that of patients without midline shift (t = ⁃ 17.781, P = 0.000). The prognosis of patients with combined injury was worse than that of patients without combined injury (t = ⁃ 4.725, P = 0.000). 3) Treatment and complications: the prognosis of patients with pre⁃hospital first aid was worse than that without pre⁃hospital first aid (t = 4.343, P = 0.000). The prognosis of patients with surgical treatment was worse than that of patients with conservative treatment (t = ⁃ 5.506, P = 0.000). The prognosis of patients with postoperative complications was worse than that of patients without complications (t = ⁃ 15.845, P = 0.000), and the prognosis of patients with concurrent infection during hospitalization was worse than that of patients without infection (t = ⁃ 14.645, P = 0.000). While there was no significant difference in prognosis between CT examination time (t = ⁃ 1.942, P = 0.052) and operation time window (t = ⁃ 1.483, P = 0.140). Conclusions The disability rate of TBI in Guizhou Province is high. Strengthening the propaganda and education of traffic and production safety knowledge can prevent and reduce TBI. Strengthening the professional training of neurosurgeons in county⁃level hospitals can make TBI patients get timely, standardized and homogeneous treatment, and improve the prognosis. doi：10.3969/j.issn.1672⁃6731.2021.03.012

Shale gas is one of the hot spots of energy development. Due to the strong heterogeneity and low physical properties of shale gas reservoirs, and the complex influencing factors of pore development, it is necessary to conduct a comprehensive analysis of the control factors of high-quality reservoirs. The sedimentary characteristics, mineral composition, pore structure and controlling factors of high quality reservoir development are studied on the basis of thin section, scanning electron microscopy (SEM) and QEMSCAN analysis, X-ray diffraction (XRD), total organic carbon (TOC), Nano-CT scanning and Low-pressure N2 adsorption (N2GA) analysis on shale samples from the Longmaxi Formation in South Sichuan Basin. The results show the following: (1) According to lithology, sedimentary structure, organic carbon content and mineral composition, six sedimentary microfacies can be divided. (2) Organic matter pores are developed in organic-rich siliceous shale and organic-rich silty shale at the bottom of the first member of the Longmaxi formation, with average porosity of more than 5% and permeability of more than 2 × 10−3 μm2, which is conducive to the formation of high-quality shale gas reservoirs. (3) The contents of siliceous and TOC are positively correlated with porosity and specific surface area, while the contents of carbonate and clay minerals are negatively correlated with reservoir quality. (4) In the first member of the Longmaxi formation, the sedimentary water depth becomes shallower from bottom to top, and the sedimentary environment changes from a reduction to an oxidation environment. The contents of siliceous and organic matter decrease, while the contents of clay minerals and carbonate minerals show the opposite trend. The difference in sedimentary microfacies affects the distribution of mineral and organic matter and controls the heterogeneity of the shale reservoir.

T-2 toxin is a secondary metabolite produced by Fusarium spp., and belongs to the type A mycotoxins in the cephalosporins family. T-2 toxin is highly toxic, and it pollutes not only crops in the field,but also granary which leads the post-harvest lost of grain, furthermore, it causes huge damage to human and animal health. Therefore, it becomes world widely the target to be prevented and controlled in the food industry and livestock industry in recent years. At present, the research on the biological toxicity of T-2 toxin is more indepth, but it still lacks adequate study on detoxification of T-2 toxin, especially the biological detoxification. The harm, mechanism and physical, chemical and biological methods of detoxification of T-2 toxins were reviewed. Focusing on the acquisition of efficient detoxification enzymes and the development of detoxification processes are conducive to the healthy development of multiple industries in China, such as grain industry, livestock industry, and by-product deep processing.

Food consumption during the rest phase promotes circadian desynchrony, which is corrected with harmful physiological and mental disorders. Previously, we found that circadian desynchrony was involved in isoflurane-induced cognitive impairment. Here, we scheduled food access to modulate daily rhythm to examine its impact on isoflurane-induced cognitive impairments. Mice were randomly transferred to restricted feeding (RF) time groups: Control group (Zeitgeber time (ZT) 0–ZT24, ad libitum feeding), Day-Feeding group (ZT0–ZT12, misaligned feeding), and Night-Feeding group (ZT12–ZT24, aligned feeding). Then, some of them were subjected to 5 h of 1.3% isoflurane anaesthesia from ZT14 to ZT19 and were divided into the Control + Anes group, the Day-Feeding + Anes group, and the Night-Feeding + Anes group. Mini-Mitter was used to monitor the daily rhythm. Fear conditioning system was conducted to assess cognition of mice. We observed that the Night-Feeding group adapted to RF gradually, whereas the Day-Feeding group exhibited a disturbed daily rhythm. The Night-Feeding + Anes group exhibited a partially enhanced daily rhythm, whereas the Day-Feeding + Anes group exhibited sustained phase advances and diurnality score increase 7 days after isoflurane anaesthesia. Notably, in tests of hippocampus-dependent contextual memory, the Night-Feeding + Anes group demonstrated decreased deficits; the Day-Feeding + Anes group showed prolonged post-anaesthetic deficits 14 days after isoflurane anaesthesia. However, amygdala-dependent cued-fear conditioning post-anaesthesia was not altered by the RF schedule. In conclusion, we demonstrated that misaligned feeding disturbed the daily rhythm and led to persistent post-anaesthetic cognitive dysfunction. Aligned feeding enhanced the daily rhythm partially and improved post-anaesthetic cognitive dysfunction.

miR-155 is associated with the promotion of tumorigenesis. Herein, we indicate that abnormal miR-155 was negatively correlated with the expression of P21WAF1/Cip1. Our results suggest that miR-155 alters the transcriptome and inhibits the expression of H3F3A in liver cancer cells. Therefore, miR-155 inhibits the methylation modification of histone H3 on the 27th lysine. Notably, on the one hand, miR-155-dependent CTCF loops cause the CDK2 interacting with cyclin E in liver cancer cells; on the other hand, miR-155 promotes the phosphorylation modification of CDK2 by inhibiting H3F3A. Subsequently, miR-155 competitively blocks the binding of RNA polymerase II (RNA Pol II) to the P21WAF1/CIP1 promoter by increasing the phosphorylation of CDK2, inhibiting the transcription and translation of P21WAF1/CIP1. Strikingly, excessive P21WAF1/CIP1 abolishes the cancerous function of miR-155. In conclusion, miR-155 can play a positive role in the development of liver cancer and influence a series of gene expression through epigenetic regulation.

Objective To investigate the mechanism of sulfhydrated Ras related protein 7a (Rab7a) in HepG2.2.15 cells and its effect on hepatitis B virus (HBV) replication. Methods The sulfhydrated Rab7a and sulfhydrated sites of Rab7a were verified by biotin switch method. The effect of the sulfhydrated Rab7a on the level of HBV replication markers was detected by enzyme-linked immunosorbent assay, RT-qPCR and Western blot. Results In HepG2.2.15 cells, H2S enhanced the sulfhydration level of endogenous Rab7a (P＜0.01). NaHS (H2S rapid release donor) enhanced the expression of sulfhydration of exogenous Rab7a, but the mutation of sulfhydration site was not affected by NaHS (P＜0.01). In HepG2.2.15 cells, H2S enhanced the sulfhydrylation level of endogenous and exogenous Rab7a; the sulfhydrylated Rab7a inhibited the expression of HBsAg, HBeAg and HBV-DNA in supernatant, as well as HBV-cccDNA, 3.5RNA, total RNA and HBcAg protein in HepG2.2.15 cells (P＜0.05). Conclusions Sulfhydrated Rab7a inhibites the replication of HBV in HBV positive cell line HepG2.2.15.

PTFE/Al/MoO3 composites with different PTFE concentrations were prepared by mold pressing and sintering. The materials were characterized by scanning electron microscopy (SEM) and X-ray diffraction (XRD), in addition, the properties of the composites under quasi-static compression and dynamic impact were also studied. The results indicate that, the strength of PTFE/Al/MoO3 composites is increased first and then decreased with the increase of PTFE, reaches the maximum of 80MPa when the volume fraction of PTFE is 70%. Meanwhile, these specimens can ignite under quasi-static compression conditions, with a huge explosion and bright flames, while no ignition phenomenon was observed in the other specimens under the same conditions. Under the dynamic impact conditions, all of the PTFE/Al/MoO3 composites can ignite, but with the increase of PTFE, the degree of intensity is decreased gradually. The reaction products are AlF3, Al2O3, Mo and C, indicating that the redox reactions of Al and PTFE, Al and MoO3have both been triggered. Under the dual influence of material strength and composition, the energy required for ignition of PTFE/Al/MoO3 composites tends to drop first and then increase with the increase of PTFE.

Background and Aims. Recent evidences reveal that epithelial to mesenchymal transition (EMT) exacerbates the process of intestinal fibrosis. Tumor necrosis factor-like ligand 1A (TL1A) is a member of the tumor necrosis family (TNF), which can take part in the development of colonic inflammation and fibrosis by regulating immune response or inflammatory factors. The purpose of this study was to elucidate the possible contribution of TL1A in onset and progression of intestinal inflammation and fibrosis through EMT. Methods. Colonic specimens were obtained from patients with inflammatory bowel disease (IBD) and control individuals. The expression levels of TL1A and EMT-related markers in intestinal tissues were evaluated. Furthermore, the human colorectal adenocarcinoma cell line, HT-29, was stimulated with TL1A, anti-TL1A antibody, or BMP-7 to assess EMT process. In addition, transgenic mice expressing high levels of TL1A in lymphoid cells were used to further investigate the mechanism of TL1A in intestinal fibrosis. Results. High levels of TL1A expression were detected in the intestinal specimens of patients with ulcerative colitis and Crohn’s disease and were negatively associated with the expression of an epithelial marker (E-cadherin), while it was positively associated with the expression of interstitial markers (FSP1 and α-SMA). Transgenic mice with high expression of TL1A were more sensitive to dextran sodium sulfate and exhibited severe intestinal inflammation and fibrosis. Additionally, the TGF-β1/Smad3 pathway may be involved in TL1A-induced EMT, and the expression of IL-13 and EMT-related transcriptional molecules (e.g., ZEB1 and Snail1) was increased in the intestinal specimens of the transgenic mice. Furthermore, TL1A-induced EMT can be influenced by anti-TL1A antibody or BMP-7 in vitro. Conclusions. TL1A participates in the formation and process of EMT in intestinal fibrosis. This new knowledge enables us to better understand the pathogenesis of intestinal fibrosis and identify new therapeutic targets for its treatment.

Background: MicroRNAs (miRNAs) have an increasing functional role in some neurodegenerative diseases. Autophagy, the degradation of bulk protein in the cytoplasm, is the quality control function of protein and has a protective role in the survival of neural cells. miR-433 may play a regulatory role in neurodegenerative diseases. Many aspects underlying the mechanism of miR-433 in neural development and neurodegeneration are not clear.Methods: In this study, we established stable cell lines expressing miR-433 by infecting mouse hippocampal neural cell line (HT-22) cells with rLV-miR-433 and the control rLV-miR. Pre-miR-433 expression was analyzed using polymerase chain reaction (PCR). Mature miR-433 expression was measured using quantitative PCR (qPCR). The effect of miR-433 overexpression on cell proliferation was determined using a CCK-8 assay and flow cytometry. RNA interference was used to analyze the function of Cdk12 in mediating the effect of miR-433 on cell proliferation. The effect of miR-433 overexpression on cell apoptosis was determined by flow cytometry. Autophagy-related genes Atg4a, LC3B, and Beclin-1 were determined using qPCR, Western blot, or immunofluorescence. In addition, RNA interference was used to analyze the effect of Atg4a on the induction of autophagy. TargetScan 7.2 was used to predict the target genes of miR-433, and Smad9 was determined using qPCR.Results: Our results indicated that miR-433 increased the expression of Atg4a and induced autophagy by increasing the expression of LC3B-Ⅱ and Beclin-1 in an Atg4a-dependent manner. In addition, miR-433 upregulated the expression of Cdk12 and inhibited cell proliferation in a Cdk12-dependent manner and promoted apoptosis in HT-22 cells under the treatment of 10-hydroxycamptothecin.Conclusion: The results of our study suggest that miR-433 may regulate neuronal growth by promoting autophagy and attenuating cell proliferation. This might be a potential therapeutic intervention in neurodegenerative diseases.

MicroRNAs are known to be involved in carcinogenesis. Recently, microRNA-372 (miR372) has been proven to play a substantial role in several human cancers, but its functions in liver cancer remain unclear. Herein, our results demonstrate that miR372 accelerates growth of liver cancer cells in vitro and in vivo. Mechanistically, miR372 enhances expression of Y-box-binding protein 1 (YB-1) by targeting for phosphatase and tensin homolog (PTEN) directly and consequently promotes phosphorylation of YB-1 via HULC looping dependent on ERK1/2 and PTEN. In particular, HULC knockdown or PTEN overexpression abrogated this miR372 action. Moreover, miR372 inhibits the degradation of β-catenin dependent on phosphorylation of YB-1 and then enhances the expression and activity of pyruvate kinase M2 isoform (PKM2) by β-catenin-LEF/TCF4 pathway. Furthermore, the loading of LEF/TCF4 on PKM2 promoter region was significantly increased in miR372 overexpressing Hep3B, and thus, glycolytic proton efflux rate (glycoPER) was significantly increased in rLV-miR372 group compared to the rLV group. Moreover, β-catenin knockdown abrogates this function of miR372. Ultimately, miR372 promotes the expression of erbB-2 through PKM2-pH3T11-acetylation on histone H3 lysine 9 (H3K9Ac) pathway. Of significance, both YB-1 knockdown and erbB-2 knockdown abrogate oncogenic action of miR372. Our observations suggest that miR372 promotes liver cancer cell cycle progress by activating cyclin-dependent kinase 2 (CDK2)-cyclin E-P21/Cip1 complex through miR372-YB-1-β-catenin-LEF/TCF4-PKM2-erbB-2 axis. This study elucidates a novel mechanism for miR372 in liver cancer cells and suggests that miR372 can be used as a novel therapeutic target of liver cancer.

Inflammatory and autophagy-related gene P62 is highly expressed in most human tumor tissues. Herein, we demonstrate that P62 promotes human mesenchymal stem cells’ malignant transformation via the cascade of P62-tumor necrosis factor alpha (TNF-α)-CUDR-CTCF-insulin growth factor II (IGFII)-H-Ras signaling. Mechanistically, we reveal P62 enhances IGFII transcriptional activity through forming IGFII promoter-enhancer chromatin loop and increasing METTL3 occupancy on IGFII 3′ UTR and enhances H-Ras overexpression by harboring inflammation-related factors, e.g., TNFR1, CLYD, EGR1, NFκB, TLR4, and PPARγ. Furthermore, the P62 cooperates with TNF-α to promote malignant transformation of mesenchymal stem cells. These findings, for the first time, provide insight into the positive role that P62 plays in malignant transformation of mesenchymal stem cells and reveal a novel link between P62 and the inflammation factors in mesenchymal stem cells.

Abstract Background Long noncoding RNA HULC is highly up-regulation in human hepatocellular carcinoma (HCC). However, the functions of HULC in hepatocarcinogenesis remains unclear. Methods RT-PCR, Western blotting, Chromatin immunoprecipitation (CHIP) assay, RNA Immunoprecipitation (RIP) and tumorignesis test in vitro and in vivo were performed. Results HULC is negatively associated with expression of PTEN or miR15a in patients of human liver cancer. Moreover, HULC accelerates malignant progression of liver cancer cells in vitro and in vivo. Mechanistically, HULC increasesthe expression of P62 via decreasing mature miR15a. On the other hand, excessive HULC increases the expression of LC3 on the level of transcription and then activates LC3 through Sirt1 (a deacetylase). Notably, HULC enhanced the interplay between LC3 and ATG3. Furthermore, HULC also increases the expression of becline-1(autophagy related gene). Therefore, HULC increases the cellular autophagy by increasing LC3II dependent on Sirt1.Noteworthy, excessive HULC reduces the expression of PTEN, β-catenin and enhances the expression of SAPK/JUNK, PKM2, CDK2, NOTCH1, C-Jun in liver cancer cells. Of significance, our observations also revealed that HULC inhibited PTEN through ubiquitin–proteasome system mediated by autophagy-P62.Ultimately,HULC activates AKT-PI3K-mTOR pathway through inhibiting PTEN in human liver cancer cells. Conclusions This study elucidates a novel mechanism that lncRNA HULC produces a vital function during hepatocarcinogenesis.

In the article the author analyses the mechanism of the impact of global financial crisis on China’s banking sector. The increasing interaction between global macroeconomic disproportions in the last decade has contributed to the crisis around the world, including China. The analysis of the effects of the 2008-2009 global financial crisis revealed key problems of the Chinese banking system: the dependence on Western financial corporations, inconsistency of government policy to regulate macroeconomic problems. The author shows the prerequisites of the emerging crisis in modern China and identifies the conditions which determine the possibility of ensuring stable development of the Chinese banking system.

The laboratory practice "Primary culture and directional differentiation of rat bone marrow mesenchymal stem cells (BMSCs)" is part of a required course for sophomore medical students at Tongji university, which has been conducted since 2012. Blended learning has been widely applied in medical courses. Based on a student-centered teaching philosophy, we reconstructed a comprehensive stem cell laboratory module with blended learning in 2021, aiming to facilitate students in enhancing their understanding of the multi-lineage differentiation potential of stem cells and improve their experimental skills, self-directed learning ability, and innovative thinking. First, we constructed in-depth online study resources, including videos demonstrating laboratory procedures, a PowerPoint slide deck, and published literature on student self-learning before class. In class, students performed a primary culture of BMSCs, freely chose among adipogenic, osteogenic, or chondrogenic differentiation, and used cytochemical or immunofluorescence staining for identification. After class, the extracurricular part involved performing quantitative polymerase chain reaction to examine the expression of multi-lineage differentiation marker genes, which was designed as an elective. After 2 years of practice, positive feedback was obtained from both students and faculty members who achieved, the learning goal as expected. The reconstructed stem cell laboratory module provides comprehensive practice opportunities for students. Students have a better understanding of BMSC at the molecular, cellular, and functional levels and have improved their experimental skills, which forms a basis for scientific research for medical students. Introducing blended learning into other medical laboratory practices thus seems valuable.

Summary: Spatial sorting to discrete quality control sites in the cell is a process harnessing the toxicity of aberrant proteins. We show that the yeast t-snare phosphoprotein syntaxin5 (Sed5) acts as a key factor in mitigating proteotoxicity and the spatial deposition and clearance of IPOD (insoluble protein deposit) inclusions associates with the disaggregase Hsp104. Sed5 phosphorylation promotes dynamic movement of COPII-associated Hsp104 and boosts disaggregation by favoring anterograde ER-to-Golgi trafficking. Hsp104-associated aggregates co-localize with Sed5 as well as components of the ER, trans Golgi network, and endocytic vesicles, transiently during proteostatic stress, explaining mechanistically how misfolded and aggregated proteins formed at the vicinity of the ER can hitchhike toward vacuolar IPOD sites. Many inclusions become associated with mitochondria in a HOPS/vCLAMP-dependent manner and co-localize with Vps39 (HOPS/vCLAMP) and Vps13, which are proteins providing contacts between vacuole and mitochondria. Both Vps39 and Vps13 are required also for efficient Sed5-dependent clearance of aggregates. : Babazadeh et al. show that Sed5, the yeast t-snare syntaxin-5, and COPII-dependent trafficking direct misfolded proteins toward the vacuole and mitochondria. Popularisation and application of Public Private Partnerships (PPP) model is an important measure used to deepen the investment and financing reform of China. The transportation industry actively explores and promotes the wide application of PPP models as they have injected new impetus into the construction and development of transportation investment. For PPP projects of expressways that are subsidised by government in the construction or operation period, since the traffic flow in the project construction period or the early operation period is low, the government subsidy is more concentrated and the payments are high, which aggravates pressure on the fiscal purse in the current period and is not conducive to the realisation of smooth fiscal expenditure in the project cooperation period. In this regard, aiming at the features of the investment revenue of expressway projects, a whole life-cycle government optimisation of subsidy mode is proposed through the analysis of the problems existing in the general subsidy mode of the PPP model in the present work. The results show that the proposed model can not only smooth government expenditure in the project’s cooperation period, but also fully exploit the advantages of higher income in the middle and later stages of project operation and maximise the growth potential of government revenue. The results of the research can provide decision-making reference for the development of innovative PPP models for transportation infrastructure in the new Era.
