The reactions occurred at excited states are promising and flourished. Many strategies have been discovered reactions initiated by visible light and without transition metals and organic dyes. Diboron ester (B2cat2) has special activity towards unsaturated bonds at excited states. In this work, azobenzene and derivatives are reduced to hydrazo with high yield and high selectivity by a very convenient and rapid reaction. Only three factors, B2cat2, water and 400 nm lighting or sunlight are necessary. Density functional theory (DFT) studies and NMR results show that azobenzene is excited and a trans to cis isomerization occurs under 400 nm and B2cat2 reacts with cis azobenzene through B−B bond cleavage rather than with ground state trans azobenzene. Hydrazo compounds are obtained after hydrolysis with water. Gram level hydrazobenzenes are obtained under sunlight with moderate yields. This strategy is benefit to sustainable chemistry. The green synthesis of medicines with an indazole skeleton has gained significant attention in recent years. Sustainable and environmentally friendly approaches for the synthesis of this important class of heterocyclic compounds from azobenzenes are attractive but rare. In this work, a series of neat, fast, and efficient reactions are well developed as a notable methodology for the photo-organic synthesis of indazoles. Metal and hydrogen source-free direct deoxygenative cyclization of o-carbonyl azobenzene under visible light is achieved through a new mechanism with huge potential in reduction cyclization reactions. Most examples are shown to give the corresponding 2H-indazole products in excellent yields, demonstrating that the present reaction has a wide substrate scope and good functional group tolerance. Construction of spirocyclic pyrrolidines bearing a spiro quaternary stereocenter is an enormous challenge in synthetic organic chemistry. In this report, we introduce a Ag(I)/CAAA-amidphos-catalyzed enantioselective 1,3-dipolar cycloaddition between azomethine ylides and α-methylene-γ-butyrolactone as an effective strategy for the construction of excellent endo-selective spiro-[butyrolactone-pyrrolidine] derivatives. Meanwhile, the catalytic system was also successfully applied in the three-component one-pot reaction of azomethine ylides formed in situ under the action of N,N′-diisopropylcarbodiimide serving as a dehydrator. Under the optimal conditions, endo-pyrrolidine derivatives bearing a spiro quaternary stereocenter were obtained with high to excellent yields (up to 95% yield) and enantioselectivities (up to 93% ee). The persistence of hepatitis B virus (HBV) covalently closed circular DNA (cccDNA) is a key obstacle for HBV cure. This study aims to comprehensively assess the effect of interferon (IFN) and small-interfering RNA (siRNA) combination on the cccDNA minichromosome. Utilizing both cell and mouse cccDNA models, we compared the inhibitory effects of IFNα, siRNA, and their combination on cccDNA activity and assessed its epigenetic state. IFNα2 treatment alone reduced HBV RNAs, HBeAg, and HBsAg levels by approximately 50%, accompanied by a low-level reconstitution of SMC5/6-a chromatin modulator that restricts cccDNA transcription. HBx-targeting siRNA (siHBx) achieved significant suppression of viral antigens and reconstitution of SMC5/6, but this effect could be reversed by the deacetylase inhibitor Belinostat. The combination of IFN with siHBx resulted in over 95% suppression of virological markers, reduction in epigenetic activation modifications (H3Ac and H4Ac) on cccDNA, and further reduced cccDNA accessibility, with the effect not reversible by Belinostat. In an extracellular humanized IFNAR C57BL/6 mouse model harboring recombinant cccDNA, the effect of combination of clinically used pegylated IFNα2 and GalNac-siHBx was further clarified, indicating a higher and more durable suppression of cccDNA activity compared to either therapy alone. In conclusion, the combination of IFNα and siRNA achieves a more potent and durable epigenetic inhibition of cccDNA activity in cell and mouse models, compared to monotherapy. The specific stacking mode of D/A blocks is often considered to largely determine the physicochemical properties of cocrystals. However, this rule may fail when encountering a large degree of (integer or near-integer) charge transfer situations. Herein, we explore the extensive correlations between the possible smallest structural units, stacking modes, and near-infrared photothermal conversion (NIR-PTC) properties of F4TCNQ-based cocrystals with typical features of integer-charge-transfer. Surprisingly, these cocrystals with distinct stacking modes display analogous D-A interactions, broad red-shift absorption, ultrafast (1-3 ps) relaxation dynamics of excited states, and excellent NIR-PTC properties. This supports that the resulting "D + A - " ion pairs from integer-charge-transfer may serve as the primary structural units beneath the secondary stacking modes to dominate the property of cocrystals. The stacking modes play an important but only secondary role. This work provides new insights into the structure-dynamics-property correlations and modular design of organic cocrystals for PTC and other applications.
