RNA terminal phosphorylase B (RTCB) has been shown to play a significant role in multiple physiological processes. However, the specific role of RTCB in the mouse colon remains unclear. In this study, we employ a conditional knockout mouse model to investigate the effects of RTCB depletion on the colon and the potential molecular mechanisms. We assess the efficiency and phenotype of Rtcb knockout using PCR, western blot analysis, histological staining, and immunohistochemistry. Compared with the control mice, the Rtcb-knockout mice exhibit compromised colonic barrier integrity and prominent inflammatory cell infiltration. In the colonic tissues of Rtcb-knockout mice, the protein levels of TNF-α, IL-8, and p-p65 are increased, whereas the levels of IKKβ and IκBα are decreased. Moreover, the level of GSK3β is increased, whereas the levels of Wnt3a, β-catenin, and LGR5 are decreased. Collectively, our findings unveil a close association between RTCB and colonic tissue homeostasis and demonstrate that RTCB deficiency can lead to dysregulation of both the NF-κB and Wnt/β-catenin signaling pathways in colonic cells.

Introduction: The grain supply-demand balance is a long-standing concern for many countries and is essential to guaranteeing social stability, maintaining economic development and ensuring national grain security. Methods: Based on the data of 65 countries along the "Belt and Road" (B&R) from 1993 to 2021, this paper analyzed the spatiotemporal evolution, the matching relationship, regional grain security situation and driving factors of grain supply-demand by measuring and classifying the grain self-sufficiency rate using methods such as geostatistical analysis and the GTWR model. Results: The results indicated the following: (1) The B&R region was still at the primary stage of "food-based and feed-supplemented". Grain supply and demand in the B&R region showed a steady upward trend, with grain yield contributing more to grain supply than sown area. (2) Overall, the B&R region has been largely self-sufficient since 2007, with grain supply meeting demand, but the level of grain self-sufficiency varied considerably between countries. (3) More than 58% of the countries were in grain insecurity, concentrated in West Asia-Middle East and South-East Asia. The gravity center of both grain supply and demand was near East Asia. (4) In terms of matching supply-demand, most countries fell into the category of high supply-high demand and low supply-low demand, with basically the same level of grain supply and demand. (5) Grain yield had the largest positive impact on grain supply-demand, GDP had the largest negative impact, and temperature change and precipitation change had a relatively small effect. The effects of fertilizer use, grain yield, and GDP on grain supply-demand fluctuated greatly over time. In DC microgrids, a large-capacity hybrid energy storage system (HESS) is introduced to eliminate variable fluctuations of distributed source powers and load powers. Aiming at improving disturbance immunity and decreasing adjustment time, this paper proposes active disturbance rejection control (ADRC) combined with improved MPC for n + 1 parallel converters of large-capacity hybrid energy storage systems. ADRC is utilized in outer voltage control loops, and improved MPC is employed in inner current control loops of n battery converters. Droop control is adopted to obtain power distribution between n battery converters, and a DC bus voltage compensator is used to compensate voltage deviations and maintain constant DC bus voltage. The low-pass filter (LPF) is adopted to obtain high-frequency power as the reference for the supercapacitor converter, ADRC is also utilized in the outer power control loop, and MPC is employed in the inner current control loop. Compared with traditional observers, the voltage expansion state observer of the proposed ADRC control is independent of the system model and parameters and consequently has strong disturbance immunity, and significantly reduces voltage overshoots during power fluctuations. The MPC-based inner current control loops of n + 1 converters accelerate current response speed and significantly decrease switching losses. Simulation and experimental results indicate that utilizing the proposed control strategies, large-capacity HESS has stronger anti-interference ability, shorter regulation time, smaller switching loss, and simultaneously maintains the stability of the DC bus voltage. Plant communities are complex systems shaped by a combination of deterministic and stochastic ecological processes. Bryophytes are an essential component of plant diversity in natural ecosystems, yet our understanding of their community ecology needs to catch up to that of other organisms. The unique geological history, alpine climatic conditions, and high habitat heterogeneity of Tibet provide suitable areas for bryophytes to survive in the alpine regions. Therefore, field surveys were conducted across 184 plots in forest, thicket, and herbaceous vegetation of Tibet to investigate the role of deterministic processes such as biological interactions and abiotic effects, along with stochastic processes, in shaping the distribution of bryophyte assemblages. We employed various analytical methods, including mixed effects models, partial least squares path modeling, null model analysis, and neutral community models. The study showed that bryophyte richness was highest in forests. Bryophyte assemblages showed greater segregation in forest and thicket environments compared to herbaceous vegetation. As the influence of stochastic processes increased, that of deterministic processes decreased from forests through thickets to herbaceous vegetation. Deterministic processes were the main driving forces for the bryophyte assemblage pattern. Soil properties and climatic factors, particularly pH played a key role in determining bryophyte patterns in Tibet. Investigating intraspecific trait variability is crucial for understanding plant adaptation to various environments, yet research on lithophytic mosses in extreme environments remains scarce. This study focuses on Indusiella thianschanica Broth. Hal., a unique lithophytic moss species in the extreme environments of the Tibetan Plateau, aiming to uncover its adaptation and response mechanisms to environmental changes. Specimens were collected from 26 sites across elevations ranging from 3642 m to 5528 m, and the relationships between 23 morphological traits and 15 environmental factors were analyzed. Results indicated that coefficients of variation (CV) ranged from 5.91% to 36.11%, with gametophyte height (GH) and basal cell transverse wall thickness (STW) showing the highest and lowest variations, respectively. Temperature, elevation, and potential evapo-transpiration (PET) emerged as primary environmental drivers. Leaf traits, especially those of the leaf sheath, exhibited a more pronounced response to the environment. The traits exhibited apparent covariation in response to environmental challenges and indicated flexible adaptive strategies. This study revealed the adaptation and response patterns of different morphological traits of I. thianschanica to environmental changes on the Tibetan Plateau, emphasizing the significant effect of temperature on trait variation.
