5. Enhanced Catalytic Synthesis of Flavonoid by UV-B Radiation in Artemisia argyi.

Abstract

Enzymatic synthesis of specific active substances is an important foundation for biological adaptations to various stresses. In this study, we investigated the metabolic response of the medicinal herb Artemisia argyi to UV-B radiation through transcriptome and metabolome analysis. In all tested samples, there were 544 shared differentially expressed genes, most of which were linked to the metabolism of flavonoids and fatty acids. A total of 283 differential metabolites were identified and classified into 10 categories, with flavonoids being the largest category. Through an integrated analysis of genes and metabolites involved in flavonoid biosynthesis, flavonoids were predicted to be critical for the adaptation of A. argyi to UV radiation. The increased plant hormones methyl jasmonate and salicylic acid were considered as key regulatory approaches for catalyzing the large-scale synthesis of flavonoids. We explored this by investigating the flavonoid production of A. argyi grown at different altitudes. It showed that total flavonoid content of A. argyi planted in high-altitude areas was 45% higher than that in low-altitude areas. These findings not only deepen our understanding of flavonoid anabolism and its regulation but also provide a reliable strategy for improving flavonoid content in the genus Artemisia. Abstract

Summary: The conquest of land by plants was concomitant with, and possibly enabled by, the evolution of three‐dimensional (3D) growth. The moss Physcomitrium patens provides a model system for elucidating molecular mechanisms in the initiation of 3D growth. Here, we investigate whether the phytohormone ethylene, which is believed to have been a signal before land plant emergence, plays a role in 3D growth regulation in P. patens.We report ethylene controls 3D gametophore formation, based on results from exogenously applied ethylene and genetic manipulation of PpEIN2, which is a central component in the ethylene signaling pathway.Overexpression (OE) of PpEIN2 activates ethylene responses and leads to earlier formation of gametophores with fewer gametophores produced thereafter, phenocopying ethylene‐treated wild‐type. Conversely, Ppein2 knockout mutants, which are ethylene insensitive, show initially delayed gametophore formation with more gametophores produced later. Furthermore, pharmacological and biochemical analyses reveal auxin levels are decreased in the OE lines but increased in the knockout mutants.Our results suggest that evolutionarily, ethylene and auxin molecular networks were recruited to build the plant body plan in ancestral land plants. Abstract

Abstract: Phosphoinositide-specific phospholipase C (PI-PLC) is an important enzyme, which is a key player involved in eukaryotic signal transduction pathways. In plants, it plays a key role in growth and development as well as environmental stress. However, little is known about its roles in signal transduction during sexual reproduction process. In this study, we cloned and characterized a gene of full-length PI-PLC from ovules of Torenia fournieri, designated as TfPLC1. It was 2,171 bp in length, including an open reading frame encoding a polypeptide of 583 amino acids with molecular mass of 66.02 kDa. The amino acid sequence deduced from the cDNA sequence shows 40–76% similarity to other plant PI-PLCs and contains the characteristic X, Y and C2 domains. Northern blot analysis demonstrated it was predominantly expressed in ovules and flowers. Furthermore, TfPLC1 promoter::GUS transgenic analysis indicated it specifically expressed in ovule, stigma and mature pollen grain. Immunohistochemical staining showed that, in mature stigma, TfPLC1 protein was principally localized in the cells of stigmatic receptive surface. Together, our data suggest that TfPLC1 may play an important role in plant sexual reproduction.

32. Four targeted genes for predicting the prognosis of colorectal cancer: A bioinformatics analysis case

Abstract

The molecular mechanisms underlying the development and progression of colorectal cancer (CRC) have not been clarified. The purpose of the present study was to identify key genes that may serve as novel therapeutic targets or prognostic predictors in patients with CRC using bioinformatics analysis. Four gene expression datasets were downloaded from the Gene Expression Omnibus database, which revealed 19 upregulated and 34 downregulated differentially expressed genes (DEGs). The downregulated DEGs were significantly enriched in eight pathways according to Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis. A protein-protein interaction network was constructed with 52 DEGs and 458 edges. Ten key genes were identified according to the degree value, betweenness centrality and closeness centrality. Survival analysis revealed that low expression of four of the ten genes, carcinoembryonic antigen related cell adhesion molecule 7 (CEACAM7), solute carrier family 4 member 4 (SLC4A4), glucagon (GCG) and chloride channel accessory 1 (CLCA1) genes, were associated with unfavorable prognosis in CRC. Furthermore, gene set enrichment analysis revealed that two pathways were significantly enriched in the CEACAM7 low-expression group. Thus, CEACAM7, SLC4A4, GCG and CLCA1 may be prognostic markers or therapeutic targets of CRC. Low CEACAM7 expression may be associated with the activation of glycosaminoglycan biosynthesis-chondroitin sulfate and extracellular matrix receptor interaction pathways and may affect the prognosis of CRC.

37. Effects of Long-term K Fertilizer Application on the Crop Yield and K Distribution of Soil Aggregates in a Paddy - Upland Rotation System.

Abstract

[Objective] A long-term paddy - upland experiment was conducted to evaluate the effects of the recommended amount of potassium (K) fertilizer on crop yield, distribution of soil aggregate, and soil available K content. It was mainly to ascertain the distribution of K in the soil aggregate components in the paddy --upland rotation system, and then to provide theoretical basis for soil structure improvement, K pool management, and reasonable application of K fertilizer. [Method] There were 2 treatments selected, namely, NP (-K) treatment and NPK (+K) treatment in this study. Then the effects of K fertilizer application on the yield, available K content, and aggregate distribution in the middle rice-winter rape rotation system were analyzed. [Result] The results showed that the output of the crop rotation was affected by the year and fertilization. Compared with NP (--K), the average yield increases of rice and winter rape after application of K fertilizer were 0.51 and 0.33 t/hm2 , with the increments of 7.5% and 14.1% respectively. The long-term application of K fertilizer (7 a) had no significant effects on the distribution of soil aggregates but could significantly increase the water-soluble K content and available K content at depths of 20 -30 and 30 -40 cm, and available K content of aggregates in each particle size. In addition, the balance of available K at the 10 -20 and 30 - 40 cm of soil layers was the most significant through calculation of surplus-deficit value of available K in agglomerates of different soil layers. [Conclusion] Compared with the initial available K content in the farmland in 2011, the current K fertilizer application could lead to that farmland K content continues to decline. Therefore, we should pay attention to straw returned and supplement of organic K fertilizer, to maintain crop rotation system's productivity and soil K balance. [ABSTRACT FROM AUTHOR]

Abstract

Dracaena, a remarkably long-lived and slowly maturing species of plant, is world famous for its ability to produce dragon’s blood, a precious traditional medicine used by different cultures since ancient times. However, there is no detailed and high-quality genome available for this species at present; thus, the molecular mechanisms that underlie its important traits are largely unknown. These factors seriously limit the protection and regeneration of this rare and endangered plant resource. Here, we sequenced and assembled the genome of Dracaena cochinchinensisat the chromosome level. The D. cochinchinensisgenome covers 1.21 Gb with a scaffold N50 of 50.06 Mb and encodes 31 619 predicted protein-coding genes. Analysis showed that D. cochinchinensishas undergone two whole-genome duplications and two bursts of long terminal repeat insertions. The expansion of two gene classes, cis-zeatin O-glucosyltransferase and small auxin upregulated RNA, were found to account for its longevity and slow growth. Two transcription factors (bHLH and MYB) were found to be core regulators of the flavonoid biosynthesis pathway, and reactive oxygen species were identified as the specific signaling molecules responsible for the injury-induced formation of dragon’s blood. Our study provides high-quality genomic information relating to D. cochinchinensisand significant insight into the molecular mechanisms responsible for its longevity and formation of dragon’s blood. These findings will facilitate resource protection and sustainable utilization of Dracaena.
