Retinoic acid receptor gamma (RARG) belongs to the nuclear receptor superfamily and has 90% homology to RAR alpha (RARA) and RAR beta. The promyelocytic leukemia (PML)–RARA fusion gene has been implicated in acute promyelocytic leukemia (APL). RARG gene rearrangement has been identified in a rare subtype of acute myeloid leukemia (AML) that resembles APL. To date, only 10 cases of gene rearrangements involving RARG (nucleoporin [NUP]98–RARG, promyelocytic leukemia protein–RARG, cleavage and polyadenylation-specific factor 6–RARG, or nucleophosmin [NPM]1–RARG–NPM1) have been reported. These patients show characteristics similar to APL, including bone marrow morphology, coagulation abnormality, and immunophenotype; however, they are resistant to all-trans retinoic acid and arsenic trioxide treatment. Moreover, there is no optimal therapeutic regimen for this subtype of AML. In this study, we report the clinical presentation and experimental findings of a case of AML with NUP98–RARG gene fusion similar to APL and review other cases of RARG gene rearrangement described in the literature.

Cabbage (Brassica oleracea L. var. capitata) accounts for a critical vegetable crop belonging to Brassicaceae family, and it has been extensively planted worldwide. Simple sequence repeats (SSRs), the markers with high polymorphism and co-dominance degrees, offer a crucial genetic research resource. The current work identified totally 64,546 perfect and 93,724 imperfect SSR motifs in the genome of the cabbage ‘TO1000.’ Then, we divided SSRs based on the respective overall length and repeat number into different linkage groups. Later, we characterized cabbage genomes from the perspectives of motif length, motif-type classified and SSR level, and compared them across cruciferous genomes. Furthermore, a large set of 64,546 primer pairs were successfully identified, which generated altogether 1,113 SSR primers, including 916 (82.3%) exhibiting repeated and stable amplification. In addition, there were 32 informative SSR markers screened, which might decide 32 cabbage genotypes for their genetic diversity, with level of polymorphism information of 0.14–0.88. Cultivars were efficiently identified by the new strategy designating manual diagram for identifying cultivars. Lastly, 32 cabbage accessions were clearly separately by five Bol-SSR markers. Besides, we verified whether such SSRs were available and transferable in 10 Brassicaceae relatives. Based on the above findings, those genomic SSR markers identified in the present work may facilitate cabbage research, which lay a certain foundation for further gene tagging and genetic linkage analyses, like marker-assisted selection, genetic mapping, as well as comparative genomic analysis.

Cabbage (Brassica oleracea var. capitata) is one of the most important cruciferous leafy vegetable crops and is widely cultivated all over the world. Its yield and quality are often affected by diseases such as cabbage black rot. 2R is a cabbage line that is newly resistant to black rot, which was created by radiation mutagenesis and backcross transfer. However, the underlying molecular bases and mechanisms of early-phase response of different resistant cabbage lines against black rot infections remain unknown. Here, we completed a comprehensive transcriptome profile analysis between resistant (2R) and susceptible (2T) cabbage lines after black rot inoculations. The results showed that the typical V-shaped lesions were found in inoculated plants after 15 days, and the symptoms in the susceptible cabbage lines (2T) were significant severe than that of the resistant 2R line. A total of 10,030 differentially expressed genes (DEGs) were identified, of which 384 DEGs were found to overlap in resistant and susceptible cabbage lines after black rot infections, suggesting those DEGs may play more important roles in cabbage early responses to black rot infections. We ranked the expression levels of DEGs among the four comparison sets of resistant and susceptible cabbage lines and, interestingly, found the top ten differential expression genes contained NBS-LRR genes, protein kinase genes and expansin genes. These findings provide a comprehensive differential transcriptome profile between resistant and susceptible cabbage lines and indicate some genes play key roles in the regulation of early response to black rot infections, which will help to understand the molecular resistance of cabbage against these infections. SUMMARYMicrospore embryogenesis in plants is a process by which microspores can switch their developmental pathway from gametophytic to embryogenic via specific inductive stress treatments, which results in the formation of haploid embryos. However, only a small portion of the cultured cells can undergo the entire process to produce embryos, and most die at different developmental stages during culture. The objective of this study was to improve the induction of embryogenesis in an isolated microspore culture (IMC) of white cabbage (Brassica oleracea L. var. capitata) by focussing on the regulation of dead microspores. The results indicated that microspores died rapidly after 24 h at a heat shock temperature of 32.5°C, and that 80–90% were dead after 3 d in culture. Cell purification performed after 3 d in culture, eliminated most of the dead microspores and had a beneficial effect on embryo yield. Paroxysmal nocturnal hemoglobinuria (PNH) is an acquired hematopoietic stem cell (HSC) disorder arising from a somatic mutation of the X-linked phosphatidylinositol glycan complementation class A gene (PIG-A) which leads to partial or complete deficiency of glycosyl-phosphatidylinositol (GPI)-linked membrane proteins and causes intravascular hemolysis. Its pathophysiological links with aplastic anemia (AA) and myelodysplastic syndrome (MDS) have been described frequently, and few acute leukemia are proved to be derived from PNH. However, PNH with transformation to chronic myeloid leukemia (CML) has never been reported. Here, we report a patient initially diagnosed with PNH while 11 years later, Ph chromosome and BCR/ABL fusion gene were detected and the patient was eventually confirmed the diagnosis of CML. Here, the diagnosis and management of the interesting case, as well as questions regarding pathogenesis, are discussed.
