Abstract

MDCK/Caco-2 assays serve as essential in vitro tools for evaluating membrane permeability and active transport, especially mediated by P-glycoprotein (P-gp). Despite their utility, challenges remain in quantifying active transport and using the efflux ratio (ER) to determine intrinsic values for active efflux. Such an intrinsic value for P-gp facilitated efflux necessitates knowing whether this transporter transports the neutral or ionic species of a compound. Utilising MDCK-MDR1 assays, we investigate a method for determining transporter substrate fraction preference by studying ER pH-dependence for basic, acidic and non-dissociating compounds. These results are compared with model fits based on various assumptions of transporter species preference. As an unexpected consequence of these assays, we also give evidence for an additional influx transporter at the basolateral membrane, and further extend our model to incorporate this transport. The combined influences of paracellular transport, the previously unaccounted for basolateral influx transporter, as well as potential pH effects on the transporter impedes the extraction of intrinsic values for active transport from the ER. Furthermore, we determined that using inhibitor affects the measurement of paracellular transport. While clear indications of transporter species preference remain elusive, this study enhances understanding of the MDCK system.

Abstract

Le projet présenté dans le cadre du FSPI relève en premier lieu de la valorisation et de la transmission culturelle. Il s'appuie sur une connaissance anthropologique très approfondie des populations moken, qui constituent une poche culturelle d'environ deux mille personnes localisées au coeur de l'archipel de Mergui, au sud du Myanmar. Les Moken sont des nomades maritimes, partiellement sédentarisés (saisonnalité) qui possèdent une culture unique notamment manifestée par un langage et une cosmologie propres, ainsi qu'une organisation sociale singulière. Dans le monde d'aujourd'hui, ce peuple fait figure d'exception tant par le faible nombre de ses représentants que par son ethos de non-violence et de non-accumulation. Une exception que les porteurs du projet ont choisi de placer au centre de leur préoccupations pour ce qu'elle représente en termes de diversité et d'humanité. L'objectif ultime de ce projet et d'établir un musée vivant, très innovant et fonctionnant d'une façon plus proche d'un centre culturel que d'un 'musée vitrine ' comme cela se voit parfois. Le musée se dit 'éclaté ' car il existera sur deux pans principaux, l'un réel sur un lieu choisi dans l'archipel par les moken et l'autre virtuel, via son site Internet et les réseaux sociaux. Ses caractéristiques combinent interactivité, oralité, cartographie évolutive de l'espace social, implication forte des populations locales, communication locale et internationale et formation adaptée. Le projet financé par le FSPI a pour vocation de faciliter et d'encourager la création du Moken Alive Museum, tout en visant à promouvoir un tourisme de qualité. L'un des points forts du projet Moken est le socle ethnographique et anthropologique sur lequel il repose. Les porteurs du projet effectuent depuis de nombreuses années des travaux de recherche auprès des moken, qui ont nourri le projet de façon remarquable. Il ne regroupe pas moins que les meilleurs spécialistes mondiaux sur le sujet, tous français. A cela s'ajoute la qualité de l'expertise des membres réunis en muséologie, ainsi qu'en programmation et design informatique. C'est sur cette excellente base que le projet soumis au FSPI s'est développé. Ce développement inclus des partenariats robustes avec les autorités locales comme avec des institutions françaises (MNHN, CNRS par exemple) et étrangères (Metalaw) spécialisées. Sa plus grande force est l'ancrage du projet dans la société moken et l'implication de ce peuple dans son opérationnalisation. Il convient aussi de relever la démarche générale du projet fondée sur le comparatisme et la recherche d'excellence afin de le distinguer d'initiatives similaires conduites ailleurs. Du point de vue des évaluateurs, le projet Moken fait figure d'exemple dans le domaine qui est le sien. Et cela est d'autant plus vrai lorsque l'on prend la mesure des contraintes exceptionnelles qui ont pesé sur ce projet. Sur la période concernée par le financement, entre mars 2019 et novembre 2021, le Myanmar a connu la crise sanitaire du Covid-19 puis un coup d'état qui a meurtri le pays et vu les institutions gouvernementales s'effondrer. Ce cadre a constitué un frein pour certaines actions du projet, comme la formation muséographique dans une certaine mesure, mais il n'a pas été rédhibitoire. Lors de l'évaluation, nous avons constaté que la quasi intégralité des actions proposées dans le cadre logique initial avaient été mises en place et que les indicateurs de succès avaient été vérifiés. Parmi les quelques points faibles relevés figure la question de la traduction automatique des matériaux présentés au public, ainsi que l'irrégularité des publications sur les réseaux sociaux qui, rappelons-le, constituent une partie importante du musée 'éclaté '. Ces écueils peuvent être très aisément corrigés et demeurent mineurs en tout état de cause. Le projet Moken montre également une grande cohérence avec les politiques françaises et birmanes. Le calibrage entre le budget alloué et les résultats obtenus nous parait tout à fait juste.

Abstract

Fuel-air mixing in a direct injection spark ignition (DISI) engine occurs in a highly unsteady, turbulent and three-dimensional flow. As a result, any cycle-to-cycle unsteady variation in the mixing process can directly impact the performance of the DISI engine. To study the unsteady process in these engines, we have developed and implemented a large-eddy simulation (LES) approach with an innovative subgrid scalar mixing model based on the linear-eddy mixing (LEM) model into a commercial IC engine code (KIVA-3V). Time-averaged results of the simulations using the new LES version (KIVALES) are compared to the steady-state predictions of the original KIVA-3V. Significantly different in-cylinder turbulent fuel-air mixing is predicted by these two methods. Analysis shows that KIVALES resolves spatial features larger than the grid and that the subgrid kinetic energy adjusts to the LES resolution. As a result, KIVALES captures a highly unsteady, anisotropic fuel-air mixing process whereas a more diffused mixed field is predicted by the original KIVA-3V. This ability of KIVALES is attributed to the subgrid closure which scales the subgrid dissipation with the local grid size and thus, decreases the overall dissipation in the flow. Abstract

WMB is a SBR/carbon black masterbatch made by the wet-dispersion coagulation of carbon black, SBR latex and extender oil. Tests of tire-tread compounds indicate that WMB-mix compound produces better carbon black dispersion and a stronger carbon black-polymer interaction than dry-mix compound. WMB-mix also has a higher dynamic modulus and loss tangent than dry-mix.

Abstract

The efflux ratio (ER), determined by Caco-2/MDCK assays, is the standard in vitro metric to establish qualitatively whether a compound is a substrate of an efflux transporter. However, others have also enabled the utilisation of this metric quantitatively by deriving a relationship that expresses the ER as a function of the intrinsic membrane permeability of the membrane (P0) as well as the permeability of carrier-mediated efflux (Ppgp). As of yet, Ppgp cannot be measured directly from transport experiments or otherwise, but the ER relationship provides easy access to this value if P0 is known. However, previous derivations of this relationship failed to consider the influence of additional transport resistances such as the aqueous boundary layers (ABLs) and the filter on which the monolayer is grown. Since single fluxes in either direction can be heavily affected by these experimental artefacts, it is crucial to consider the potential impact on the ER. We present a model that includes these factors and show both mathematically and experimentally that this simple ER relationship also holds for the more realistic scenario that does not neglect the ABLs/filter. Furthermore, we also show mathematically how paracellular transport affects the ER, and we experimentally confirm that paracellular dominance reduces the ER to unity and can mask potential efflux.

Abstract

We report here the efficacy of amantadine hydrochloride for two patients with alternating hemiplegia of childhood (AHC) that did not respond to flunarizine. Amantadine was administered to one patient at age one year and seven months and to the other at age 25 years. The frequencies and duration of the hemiplegic attacks significantly improved in both patients. However, the attacks gradually returned to the previous level after a significant reduction in seizures for three years in the younger patient with ongoing AHC. Our therapeutic results further support the hypothesis that glutamate and NMDA receptors are involved in inducing alternating hemiplegic attacks, because amantadine as well as its derivative, memantine, are clinically available non-competitive N-methyl-D-aspartate (NMDA) receptor antagonists, with neuroprotective effects. Amantadine is worth trying when treating patients with AHC as a first trial or a substitute for flunarizine once the latter agent looses effect.
