Gliomas are the most common primary tumours affecting the adult central nervous system and respond poorly to standard therapy. Myc is causally implicated in most human tumours and the majority of glioblastomas have elevated Myc levels. Using the Myc dominant negative Omomyc, we previously showed that Myc inhibition is a promising strategy for cancer therapy. Here, we preclinically validate Myc inhibition as a therapeutic strategy in mouse and human glioma, using a mouse model of spontaneous multifocal invasive astrocytoma and its derived neuroprogenitors, human glioblastoma cell lines, and patient-derived tumours both in vitro and in orthotopic xenografts. Across all these experimental models we find that Myc inhibition reduces proliferation, increases apoptosis and remarkably, elicits the formation of multinucleated cells that then arrest or die by mitotic catastrophe, revealing a new role for Myc in the proficient division of glioma cells.

La mitosi è un processo biologico complesso, che assicura nelle cellule in divisione la ripartizione uguale dell’informazione genetica nelle due cellule figlie. La corretta esecuzione della mitosi è necessaria per il normale sviluppo di organismi multicellulari e per prevenire l’insorgenza di tumori. Per questo motivo, un obiettivo molto importante della ricerca biologica attuale, è la comprensione dei meccanismi e l’identificazione dei geni/proteine coinvolti nel processo mitotico. La tecnica dell’RNA interference (RNAi) ha avuto un notevole impatto sugli studi di genomica funzionale, e sono state pubblicate numerose ricerche nelle quali questa tecnica è stata usata per assegnare funzioni a geni ignoti. Nel laboratorio dove sto svolgendo l’attività di ricerca abbiamo condotto esperimenti di RNAi su cellule in coltura S2 di Drosophila melanogaster, al fine di identificare geni coinvolti nella divisione cellulare. Questo lavoro ha portato all’identificazione di 155 geni mitotici, 70 dei quali non erano mai stati precedentemente implicati in questo processo. Sorprendentemente, questi studi mostrano che l’inattivazione mediante RNAi di diversi fattori di splicing provoca evidenti difetti nella segregazione dei cromosomi. Il mio lavoro di tesi è consistito nella caratterizzazione funzionale di due di questi fattori. In Drosophila queste proteine sono codificati dai geni CG6876 e CG10754, che risultano omologhi rispettivamente ai geni PRPF31 e SF3A2 presenti in uomo. Le proteine PRPF31 e SF3A2 sono componenti di due snRNP (“small nuclear ribonucleoprotein”) coinvolti nel processo di “splicing”. Per definire il ruolo mitotico di questi fattori, ho confrontato il fenotipo ottenuto trattando le cellule in coltura S2 con dsRNA per questi geni con il fenotipo ottenuto dall’inattivazione del gene Ndc80/Hec1, che codifica per una componente del cinetocore in grado di interagire con i microtubuli del fuso mitotico. Queste analisi hanno rivelato che i geni CG6876, CG10754 e Ndc80/Hec1 hanno, durante la mitosi, un ruolo molto simile. Infatti sia i fattori di “splicing” che Hec1 sono richiesti per la funzione del cinetocore di formazione delle fibre cinetocoriche. In assenza di queste fibre, i fusi che si formano sono fortemente alterati ed i cromosomi non segregano correttamente. Complessivamente, questi risultati suggeriscono un ruolo inaspettato per i fattori di “splicing” nella formazione del fuso mitotico e nella segregazione cromosomica. Saranno necessari ulteriori studi per chiarire il ruolo di queste proteine nella struttura e funzione del cinetocore. Mitosis is the evolutionarily conserved process that enables a dividing cell to equally partition its genetic material between the two daughter cells. The fidelity of mitotic division is crucial for normal development of multicellular organisms and to prevent cancer or birth defects. Understanding the molecular mechanisms of mitosis requires the identification of genes involved in this process. Previous studies have shown that such genes can be readily identified by RNA interference (RNAi) in Drosophila tissue culture cells. Exploiting the powers of bioinformatics and RNAi technology, our laboratory has recently performed a large screen aimed at detection of genes involved in the Drosophila mitotic process. This screen has led to the identification of 155 mitotic genes, 70 of which have not been previously implicated in cell division. Surprisingly, these studies showed that RNAi-mediated inactivation of several highly conserved splicing factors results in strong defects in chromosome segregation. My thesis project was focused on the functional characterization of two of these factors. They are encoded by the CG6876 and CG10754 Drosophila genes, and are highly homologous respectively to the PRPF31 and SF3A2 human genes. PRPF31 and SF3A2 are component of two snRNPs involved in splicing regulation. To define the mitotic role of these splicing factors I compared the phenotypes elicited by their depletion with that caused by RNAimediated knockout of Ndc80/Hec1, which encodes a kinetochore protein that is thought to interact with spindle microtubules. These analyses revealed that CG6876, CG10754 and Ndc80/Hec1 have very similar mitotic roles. Both the splicing factors and the Ndc80/Hec1 protein are required for the formation of kinetochore-driven kinetochore fibers. In the absence of these fibers spindle assembly is highly abnormal and the chromosomes fail to segregate. Collectively, these results highlight an unanticipated role of splicing factors in spindle assembly and chromosome segregation.
