Conventional laser panel displays are developed through the mass integration of electrically pumped lasers or through the incorporation of a beam steering system with an array of optically pumped lasers. Here a novel configuration of a laser panel display consisting of a non-steered pumping beam and an array of electrically Q-switchable lasers is reported. The laser oscillator consists of a robust, self-standing, and deformable minute droplet that emits laser through Whispering-Gallery Mode resonance when optically pumped. The laser oscillation is electrically switchable during optical pumping by applying a vertical electric field to the droplet. Electromagnetic and fluid dynamics simulations reveal the deformation of the droplet into a prolate spheroid under the electric field and associated attenuation of quality factor (Q-factor), leading to the halt of the laser oscillation. A 2 × 3 array of droplets is fabricated by inkjet printing as a prototype of a laser panel display, and it successfully achieves the pixel-selective switching of the oscillation., (© 2024 The Author(s). Cardiac regenerative therapy using human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) has been applied in clinical settings. Herein, we aimed to investigate the in vivo metabolic profiles of hiPSC-CM grafts. RNA sequencing and imaging mass spectrometry were performed in the present study, which revealed that hiPSC-CM grafts matured metabolically over time after transplantation. Glycolysis, which was active in the hiPSC-CM grafts immediately after transplantation, shifted to fatty acid oxidation. Additionally, we examined the metabolic profile of teratomas that may form when non-CMs, including undifferentiated human induced pluripotent stem cells (hiPSCs), remain in transplanted cells. The upregulated gene expression of amino acid transporters and the high accumulation of amino acids, such as methionine and aromatic amino acids, were observed in the teratomas. We show that subcutaneous teratomas derived from undifferentiated hiPSCs can be detected in vivo using positron emission tomography with [ 18 F]fluorophenylalanine ([ 18 F]fPhe). We describe herein a facile method to access 2,3-epoxyesters with fluorine-containing substituents at their 3-position starting from the corresponding enoates by utilization of the low-costed and easy-to-handle reagent, NaOCl·5H 2 O. Because very little has been disclosed about the reactivity of such 2,3-epoxyesters, their epoxy ring opening by a variety of nucleophiles was carried out and we succeeded in clarifying these chemo- as well as regioselective processes proceeding via the S N 2 mechanism to mainly afford 2-substituted 3-hydroxyesters usually in a highly anti selective manner., (Copyright © 2024, Miyashita et al.)

Transmembrane protein 135 (TMEM135) is a 52 kDa protein with five predicted transmembrane domains that is highly conserved across species. Previous studies have shown that TMEM135 is involved in mitochondrial dynamics, thermogenesis, and lipid metabolism in multiple tissues; however, its role in the inner ear or the auditory system is unknown. We investigated the function of TMEM135 in hearing using wild-type (WT) and Tmem135 FUN025/FUN025 ( FUN025 ) mutant mice on a CBA/CaJ background, a normal-hearing mouse strain. Although FUN025 mice displayed normal auditory brainstem response (ABR) at 1 month, we observed significantly elevated ABR thresholds at 8, 16, and 64 kHz by 3 months, which progressed to profound hearing loss by 12 months. Consistent with our auditory testing, 13-month-old FUN025 mice exhibited a severe loss of outer hair cells and spiral ganglion neurons in the cochlea. Our results using BaseScope in situ hybridization indicate that TMEM135 is expressed in the inner hair cells, outer hair cells, and supporting cells. Together, these results demonstrate that the FUN025 mutation in Tmem135 causes progressive sensorineural hearing loss, and suggest that TMEM135 is crucial for maintaining key cochlear cell types and normal sensory function in the aging cochlea.

Although estrogen affects the structure and function of the nervous system and brain and has a number of effects on cognition, its roles in the auditory and vestibular systems remain unclear. The actions of estrogen are mediated predominately through two classical nuclear estrogen receptors, estrogen receptor 1 (ESR1) and estrogen receptor 2 (ESR2). In the current study, we investigated the roles of ESR1 in normal auditory function and balance performance using 3-month-old wild-type (WT) and Esr1 knockout (KO) mice on a CBA/CaJ background, a normal-hearing strain. As expected, body weight of Esr1 KO females was lower than that of Esr1 KO males. Body weight of Esr1 KO females was higher than that of WT females, while there was no difference in body weight between WT and Esr1 KO males. Similarly, head diameter was higher in Esr1 KO vs. WT females. Contrary to our expectations, there were no differences in auditory brainstem response (ABR) thresholds, ABR waves I-V amplitudes and ABR waves I-V latencies at 8, 16, 32, and 48 kHz, distortion product otoacoustic emission (DPOAE) thresholds and amplitudes at 8, 16, and 32 kHz, and rotarod balance performance (latency to fall) between WT and Esr1 KO mice. Furthermore, there were no sex differences in ABRs, DPOAEs, and rotarod balance performance in Esr1 KO mice. Taken together, our findings show that Esr1 deficiency does not affect auditory function or balance performance in normal hearing mice, and suggest that loss of Esr1 is likely compensated by ESR2 or other estrogen receptors to maintain the structure and function of the auditory and vestibular systems under normal physiological conditions.

We propose generalized equilibria of a three-dimensional color-gradient lattice Boltzmann model for two-component two-phase flows using higher-order Hermite polynomials. Although the resulting equilibrium distribution function, which includes a sixth-order term on the velocity, is computationally cumbersome, its equilibrium central moments (CMs) are velocity-independent and have a simplified form. Numerical experiments show that our approach, as in Wen et al. [Phys. Rev. E 100, 023301 (2019)2470-004510.1103/PhysRevE.100.023301] who consider terms up to third order, improves the Galilean invariance compared to that of the conventional approach. Dynamic problems can be solved with high accuracy at a density ratio of 10; however, the accuracy is still limited to a density ratio of 1000. For lower density ratios, the generalized equilibria benefit from the CM-based multiple-relaxation-time model, especially at very high Reynolds numbers, significantly improving the numerical stability.

Background: Person-specific computational models can estimate transcranial direct current stimulation (tDCS) current dose delivered to the brain and predict treatment response. Artificially created electrode models derived from virtual 10-20 EEG measurements are typically included in these models as current injection and removal sites. The present study directly compares current flow models generated via artificially placed electrodes ("artificial" electrode models) against those generated using real electrodes acquired from structural MRI scans ("real" electrode models) of older adults., Methods: A total of 16 individualized head models were derived from cognitively healthy older adults (mean age = 71.8 years) who participated in an in-scanner tDCS study with an F3-F4 montage. Visible tDCS electrodes captured within the MRI scans were segmented to create the "real" electrode model. In contrast, the "artificial" electrodes were generated in ROAST. Percentage differences in current density were computed in selected regions of interest (ROIs) as examples of stimulation targets within an F3-F4 montage., Main Results: We found significant inverse correlations ( p < 0.001) between median current density values and brain atrophy in both electrode pipelines with slightly larger correlations found in the artificial pipeline. The percent difference (PD) of the electrode distances between the two models predicted the median current density values computed in the ROIs, gray, and white matter, with significant correlation between electrode distance PDs and current density. The correlation between PD of the contact areas and the computed median current densities in the brain was found to be non-significant., Conclusions: This study demonstrates potential discrepancies in generated current density models using real versus artificial electrode placement when applying tDCS to an older adult cohort. Our findings strongly suggest that future tDCS clinical work should consider closely monitoring and rigorously documenting electrode location during stimulation to model tDCS montages as closely as possible to actual placement. Hearing loss is the third most prevalent chronic health condition affecting older adults and age-related hearing loss (ARHL) is the most common form of hearing impairment. Significant sex differences in hearing have been documented in humans and rodents. In general, the results of these studies show that men lose their hearing more rapidly than women. However, the cellular mechanism underlying sex differences in hearing or hearing loss remains largely unknown, and to our knowledge, there is no well-established animal model for studying sex differences in hearing. In the current study, we examined sex differences in body composition, voluntary wheel running activity, balance performance, auditory function, and cochlear histology in young, middle-age, and old CBA/CaJ mice, a model of age-related hearing loss. As expected, body weight of young females was lower than that of males. Similarly, lean mass and total water mass of young, middle-age, and old females were lower than those of males. Young females showed higher voluntary wheel running activity during the dark cycle, an indicator of mobility, physical activity, and balance status, compared to males. Young females also displayed higher auditory brainstem response (ABR) wave I amplitudes at 8 kHz, wave II, III, V amplitudes at 8 and 48 kHz, and wave IV/I and V/I amplitude ratios at 48 kHz compared to males.
