Abstract In the recent years, electroconductive scaffolds have shown promising capabilities in guided regeneration of electroactive tissues including nerve, heart muscle, bone, cartilage, and skin. Herein, the fabrication of a novel electroconductive poly (L‐lactic acid) (PLLA)/polyamidoamine (PAMAM) dendrimer nanofibrous scaffold containing graphene oxide (GO) nanosheets is described. The presence of PAMAM with amine terminal groups successfully aminolyzed PLLA. Interestingly, both PAMAM (5% w/w) and GO (0.5, 1, 2% w/w) not only contributed to reducing the fiber diameter, increasing the hydrophilicity and degradation rate, but also provided a nanocomposite scaffold with enhancement in electrical conductivity. By incorporating 1% w/w of GO, the nanocomposite scaffold exhibited optimized properties, including electrical conductivity (≈3.09 × 10−5 S m−1), crystallinity (≈ 47%), Young's modulus (≈16.95 MPa), as well as strength (≈1.58 MPa). This nanocomposite also demonstrated significant antibacterial activity of ≥ 90% against both gram‐positive and gram‐negative bacteria. Cellular assays confirmed acceptable cytocompatibility of the nanocomposite scaffolds containing GO and PAMAM, which can support the viability and proliferation of PC‐12 cells. In conclusion, the presence of GO nanosheets alongside PAMAM dendrimers can synergically promote the properties of the prepared nanofibrous mats which can be used as potential electroconductive scaffolds for guided tissue regeneration.

Abstract Rho GTPases play a key role in the spatio-temporal coordination of cytoskeletal dynamics during cell migration. Here, we directly investigate crosstalk between the major Rho GTPases Rho, Rac and Cdc42 by combining rapid activity perturbation with activity measurements in mammalian cells. These studies reveal that Rac stimulates Rho activity. Direct measurement of spatio-temporal activity patterns show that Rac activity is tightly and precisely coupled to local cell protrusions, followed by Rho activation during retraction. Furthermore, we find that the Rho-activating Lbc-type GEFs Arhgef11 and Arhgef12 are enriched at transient cell protrusions and retractions and recruited to the plasma membrane by active Rac. In addition, their depletion reduces activity crosstalk, cell protrusion-retraction dynamics and migration distance and increases migration directionality. Thus, our study shows that Arhgef11 and Arhgef12 facilitate exploratory cell migration by coordinating cell protrusion and retraction by coupling the activity of the associated regulators Rac and Rho.

Purpose: Pennyroyal is a species of the Lamiaceae family with potent anti-cancer and antioxidant properties. Combining this antioxidant with chemotherapeutic agents enhances the effectiveness of these agents by inducing more apoptosis in cancerous cells. Methods: Here, methotrexate (MTX) combined with pennyroyal oil based on PEGylated nanostructured lipid carriers (NLCs) was assessed. These nanoparticles were physiochemically characterized, and their anti-cancer effects and targeting efficiency were investigated on the folate receptor-positive human breast cancer cell line (MCF-7) and negative human alveolar basal epithelial cells (A549). Results: Results showed a mean size of 97.4±12.1 nm for non-targeted PEGylated NLCs and 220.4±11.4 nm for targeted PEGylated NLCs, with an almost small size distribution assessed by TEM imaging. Furthermore, in vitro molecular anti-cancer activity investigations showed that pennyroyal-NLCs and pennyroyal-NLCs/MTX activate the apoptosis and autophagy pathway by changing their related mRNA expression levels. This paper aims to review the State of the Art in metal self-expanding stents made from nitinol (NiTi), showing shape memory and superelastic behaviors, to identify the challenges and the opportunities for improving patient outcomes. A significant contribution of this paper is its extensive coverage of multidisciplinary aspects, including design, simulation, materials development, manufacturing, bio/hemocompatibility, biomechanics, biomimicry, patency, and testing methodologies. Additionally, the paper offers in-depth insights into the latest practices and emerging trends, with a special emphasis on the transformative potential of additive manufacturing techniques in the development of metal stents. By consolidating existing knowledge and highlighting areas for future innovation, this review provides a valuable roadmap for advancing nitinol stents.

Despite the importance of temperature distribution in spark plasma sintering of metallic glasses, its quantification has been experimentally laborious. This work proposes an experimental strategy to determine the sintering temperature by establishing a quantitative relationship between the temperature-thermal signal. We reproduced the thermal profiles of spark plasma sintering by isothermal annealing and found a correlation between annealing temperature and isothermal crystallization time. This strong correlation indicates the temperature-dependent structural evolution of glassy powders. Using isothermal crystallization time as the measuring gauge, we correlated the annealing temperature to the sintering temperature and obtained the sample temperature map. The sample temperature is at least 19 ​°C higher than the nominal temperature of 425 ​°C measured by the thermocouple. Meanwhile, the sample temperature shows a hump-shaped pattern closely correlated with the current density. The maximum temperature of 453 ​°C occurs on the sample/punches contact surfaces. Temperature heterogeneity within the sample induces diverse microstructures and porous structures. We elucidate that the temperature inhomogeneity is intrinsic, given the presence of contact interfaces. Contact resistances affect the current distribution and heat transfer, resulting in a larger temperature gradient than the traditional powder metallurgy process.

Abstract Over the past few years, the processing of motor imagery (MI) electroencephalography (EEG) signals has been attracted for developing brain-computer interface (BCI) applications, since feature extraction and classification of these signals are extremely difficult due to the inherent complexity and tendency to artifact properties of them. The BCI systems can provide a direct interaction pathway/channel between the brain and a peripheral device, hence the MI EEG-based BCI systems seem crucial to control external devices for patients suffering from motor disabilities. The current study presents a semi-supervised model based on three-stage feature extraction and machine learning algorithms for MI EEG signal classification in order to improve the classification accuracy with smaller number of deep features for distinguishing right- and left-hand MI tasks. Stockwell transform is employed at the first phase of the proposed feature extraction method to generate two-dimensional time–frequency maps (TFMs) from one-dimensional EEG signals. Next, the convolutional neural network (CNN) is applied to find deep feature sets from TFMs. Then, the semi-supervised discriminant analysis (SDA) is utilized to minimize the number of descriptors. Finally, the performance of five classifiers, including support vector machine, discriminant analysis, k-nearest neighbor, decision tree, random forest, and the fusion of them are compared. The hyperparameters of SDA and mentioned classifiers are optimized by Bayesian optimization to maximize the accuracy. The presented model is validated using BCI competition II dataset III and BCI competition IV dataset 2b. The performance metrics of the proposed method indicate its efficiency for classifying MI EEG signals.

Abstract An injectable hybrid hydrogel is synthesized, comprising silk fibroin (SF) and chondroitin sulfate (CS) through di‐tyrosine formation bond of SF chains. CS and SF are reported with excellent biocompatibility as tissue engineering scaffolds. Nonetheless, the rapid degradation rate of pure CS scaffolds presents a challenge to effectively recreate articular cartilage. As CS is one of the cartilage extracellular matrix (ECM) components, it has the potential to enhance the biological activity of SF‐based hydrogel in terms of cartilage repair. Therefore, altering the CS concentrations (i.e., 0 wt%, 0.25 wt%, 0.5 wt%, 1 wt%, and 2 wt%), which are interpenetrated between SF β‐sheets and chains, can potentially adjust the physical, chemical, and mechanical features of these hybrid hydrogels. The formation of β‐sheets by 30 days of immersion in de‐ionized (DI) water can improve the compression strength of the SF/CS hybrid hydrogels in comparison with the same SF/CS hybrid hydrogels in the dried state. Biological investigation and observation depicts proper cell attachment, proliferation and cell viability for C28/I2 cells. Gene expression of sex‐determining region YBox 9 (SOX9), Collagen II α1, and Aggrecan (AGG) exhibits positive C3H10T1/2 growth and expression of cartilage‐specific genes in the 0.25 wt% and 0.5 wt% SF/CS hydrogels.

Silk fibroin (SF)–chondroitin sulfate (CS)-hybrid hydrogels (HHGs) containing kartogenin (KGN) were synthesized by the induction of di-tyrosine bonds, using enzymatic crosslinking in SF/CS blend solution. Then, physical crosslinking was performed to optimize the HHG properties. The results showed that the increasing of the β-sheet content as side effect of physical crosslinking from 10% to 40% affects KGN accumulative release percentage almost three times. Biological investigations using C28/I2 human chondrocyte cell line showed that the presence of KGN, CS, and optimized β-sheets content improved cell supporting behavior of HHG. Finally, KGN loaded HHG might be a promising candidate for cartilage repair.
