Abstract Plant-mediated synthesis of nanoparticles is a sustainable approach that has gained widespread scientific acceptance due to its numerous benefits and applications. In this study, a zinc oxide-doped activated carbon (ZAC) derived from palm kernel shells (PKS) was synthesized via a bioreduction route using a water-based extract of Nymphaea lotus leaves as a reducing agent. The synthesized ZAC nanocomposites were characterized using microscopic (TEM, SEM) and spectroscopic (FTIR, EDS, XRD, and UV–Vis) analyses. The adsorptive properties of ZAC and efficiency in scavenging a phenothiazine derivative (methylene blue) from an aqueous solution were investigated. Results reveal that nano-scale ZAC particles were crystalline, exhibited irregular shapes, with an average size of 45 nm, and were highly dispersed. The optimum quantity adsorbed was 248 mg/g at a methylene blue concentration of 140 mg/L for 60 min using 0.02 g/100 mL of ZAC. Adsorption and kinetics data closely aligned with the Freundlich isotherm and the pseudo-second-order model, respectively indicating heterogeneous surface adsorption and chemisorption as the dominant mechanisms. The regeneration study of ZAC shows that over five cycles, thermal regeneration maintained high adsorption capacity with minimal decline and chemical regeneration significantly led to reduction in the adsorption capacity but solvent washing offered a balance between efficacy and structural preservation.

This study evaluated catabolic gene expression as an index of hydrocarbon breakdown in ultisol treated with Bacillus altitudinis-Cropped Biofertilizers derived from Biosolid and Brewer Spent Grain (BSG) using phenotypic and molecular methods. We observed significant reductions in Total Petroleum Hydrocarbons (TPH) concentrations with both amendments, albeit the biosolid based amendment was markedly more effective. Concurrently, there was a substantial increase in hydrocarbon-degrading bacteria. Notably, the bulk of Operational Taxonomic Units (OTUs) in biosolid (95.77 %) and BSG (93.00 %) amended Ultisol were Proteobacteria, Acidobacteria, Actinobacteria, and Planctomycetes, which are key players in hydrocarbon bioremediation. We observed the presence of eleven hydrocarbon-degrading genes through M5nr analysis. These genes encompass essential catalysts for aliphatic hydrocarbon degradation and hydrocarbon desulfurization. Notably, these enzymes/genes include Bacterial Flavin-bound Monooxygenase (AlmA), Alkane Monooxygenase (AlkM), Alpha Ketoglutarate Dependent Dioxygenase (alkB), Propane Monooxygenase (PrM), Cytochrome P450 (cP450), Methane Monooxygenase/Ammonia Monooxygenase (MMO/AMO) subunits A, B, and C, Alkane Sulfonate Monooxygenase (ssuD), Alkane Sulfonates Transport System Permease Protein (ssuC), Dibenzothiophene Monooxygenase (dszC), Dibenzothiophene-Sulfone Monooxygenase (dszA), and Dibenzothiophene-5,5-Dioxide Monooxygenase (dszB). These genes play pivotal roles in the degradation of aliphatic hydrocarbons and hydrocarbon desulfurization. Interestingly, unique gene expression patterns were observed for each amendment, with Actinomycetales and Bulkhoderiales orders expressing the majority of identified genes. These findings have revealed the amendment-specific microbial and genetic alterations and, the diversity and potential of the annotated genes induced by the Bacillus altitudinis-Cropped Biofertilizers (biosolid and BSG amendments) for effective remediation of hydrocarbon-contaminated soils.

Human activities on land have grown significantly changing the entire landscape, while most of the changes have occurred in the tropics. The change has become a serious environmental concern at the local, regional and global scales. The intensity, speed, and degree of land use / land cover (LULC) changes are nowadays quicker compared to the past because of the development of society. Moreover, the rapid increase in population resulted in disturbing a large number of landscapes on the Earth. The main objective of this study was to detect historical (1990– 2020) and predicted (2020–2050) LULC changes in the Welmel River Watershed, which is located in the Genale-Dawa Basin, South Eastern Ethiopia. The dataset of 1990, 2005, and 2020 was generated from Landsat 5, Landsat 7 and Landsat 8 respectively to determine the historical LULC map. The result of this study revealed that agriculture/ settlement increased by 6.85 km 2∙y –1, while forestland declined by 9.16 km 2∙y –1 over the last 31 years between 1990 and 2020. In the coming 31 years (by 2050), if the existing trend of the LULC change continues, agriculture/settlement land is expected to increase from 290.64 km 2 in 2020 to 492.51 km 2 in 2050 at the rate of 6.73 km 2∙y –1, while forestland is expected to shrink from 690.48 km2 in 2020 to 427.01 km 2 in 2050 by a rate of 8.78 km 2∙y –1.

Abstract We investigated the impact of Coartem™ (COA) and aflatoxin B1 (AFB1) on rats’ hypothalamus, epididymis, and testis. Male rats were randomly grouped (n = 5 rats) and treated: control group (corn oil), AFB1 (70 µg/kg), COA (5 mg/kg), COA + AFB1 (5 + 0.035 mg/kg) and COA + AFB1 (5 + 0.07 mg/kg) for 28 days. Blood samples were collected for serum prolactin, testosterone, follicle-stimulating and luteinising hormones (FSH and LH) assay upon sacrifice. The semen, hypothalamus, epididymis, and testes were harvested for morphological, biochemical, and histopathology determination of oxidative, inflammation stress, genomic integrity, and pathological alterations. Exposure to the COA and AFB1 caused the cauda epididymal spermatozoa to display low motility, viability, and volume, with increased abnormalities. Hormonal disruption ensued in animals exposed to COA and AFB1 alone or together, exemplified by increased prolactin, and decreased testosterone, FSH and LH levels. Treatment-related reduction in biomarkers of testicular metabolism—acid and alkaline phosphatases, glucose-6-phosphate dehydrogenase, and lactate dehydrogenase—were observed. Also, COA and AFB1 treatment caused reductions in antioxidant (Glutathione and total thiols) levels and antioxidant enzyme (Catalase, superoxide dismutase, glutathione peroxidase, and glutathione-S-transferase) activities in the examined organs. At the same time, treatment-related increases in DNA damage (p53), oxidative stress (xanthine oxidase, reactive oxygen and nitrogen species and lipid peroxidation), inflammation (nitric oxide and tumour necrosis factor-alpha), and apoptosis (caspase-9, and -3) were observed. Chronic exposure to COA and AFB1 led to oxidative stress, inflammation, and DNA damage in male rats' hypothalamic-reproductive axis, which might potentiate infertility if not contained.

The main objective of this study was to evaluate the self-medication (SM) practice and associated factors with knowledge and attitude of undergraduate health science students at GAMBY Medical and Business College, Bahir Dar, Ethiopia.Methods: An institutional-based cross-sectional quantitative study was conducted using a self-administered questionnaire from May to July, 2022. A stratified random sampling method was applied to collect the data. The collected data were checked, and exported into SPSS 26. Descriptive statistics and regression analysis were performed to determine the results and the associated factors. Variables with p < 0.05 were regarded as significant.Results: 301 students (31 medicine, 163 pharmacy, and 107 medical laboratory students) responded (99% response rate). 68.1% of the respondents had practiced SM at least once within the last six months. 58.8% had good knowledge, while 55.5% have a positive attitude towards SM. Headache (33.7%) and cough (29.8%) were the primary disease conditions for SM. Analgesics (37.1%) and antimicrobials (29.8%) are most frequently self-consumed agents. Being a medicine student (AOR = 3.872; 95% CI: (1.263– 11.866); p = 0.018), not having health insurance (AOR = 2.431; 95% CI: (1.383– 4.274); p = 0.002), and not having a known medical illness (AOR = 2.241; 95% CI: (1.226– 4.127); p = 0.010) were independently associated with good knowledge. While, living in an urban area was significantly associated with a positive attitude (AOR = 3.593; 95% CI: (1.404– 9.197); p = 0.004).Conclusion: The SM rate in GAMBY is significantly higher. Besides, not more than half the students had acceptable knowledge and attitude towards SM. The college and the surrounding regulatory authorities should consider ways of controlling and recapitalizing SM practices by the students.Keywords: self-medication, health science students, knowledge, attitude, practice, Ethiopia

Abstract Inadvertent exposure to furan and Pb is associated with hepatorenal abnormalities in humans and animals. It is perceived that these two chemical species may work in synergy to orchestrate liver and kidney damage. Against this background, we investigated the combined effect of furan and incremental lead (Pb) exposure on hepatorenal dysfunction. Wistar rats (n = 30; 150 g) were treated for 28 days accordingly: Control; FUR (8 mg/kg), PbAc (100 µg/L), FUR + PbAc1 (8 mg/kg FUR + 1 µg/L PbAc); FUR + PbAc1 (8 mg/kg FUR + 10 µg/L PbAc), and FUR + PbAc1 (8 mg/kg FUR + 100 µg/L PbAc). Biomarkers of hepatorenal function, oxidative stress, inflammation, DNA damage, and apoptosis were examined. Furan and incrementally Pb exposure increased the levels of hepatorenal biomarkers and oxidative and pro-inflammatory mediators, including lipid peroxidation, reactive oxygen and nitrogen species, and interleukin-1 beta. Increased DNA damage, caspases- 9 and -3, and atypical histoarchitecture of the hepatorenal tissues exemplified furan and Pb treatment-related perturbations. Furthermore, the levels of antioxidants and IL-10 were also suppressed. Furan and Pb dose-dependently exacerbated hepatorenal derangements by altering the redox and inflammatory rheostats, worsened DNA damage, and related apoptotic onset that may potentiate hepatorenal disorders in humans and animals. The findings validate the synergistic effect of furan and Pb in the pathophysiology of kidney and liver disorders.

Metagenomics is the study of genetic information, including the sequences and genomes of microorganisms present in an environment. Since 1998, the full-scale application of this technique to environmental chemistry has brought significant advances in the characterization of the nature and chemical composition/distribution of contaminants present in environmental matrices of contaminated and/or remediated sites. This has been critical in the selection of microorganisms and has contributed significantly to the success of this biological treatment over the years. Metagenomics has gone through different phases of development, which ranges from initial sequencing strategies to next-generation sequencing (NGS), which is a recently developed technology to obtain more robust deoxyribonucleic acid (DNA) profile of microorganisms devoid of chimeric sequences which reduces the quality of metagenomic data. Therefore, the objective of this review is to evaluate the applications of metagenomics in the understanding of environmental dynamics of chemical contaminants during remediation studies. Also, this review presents the relationship between biological characteristics of microorganisms and chemical properties of chemical compounds, which forms the basis of bioremediation and could be useful in developing predictive models that could enhance remediation efficiency. In conclusion, metagenomic techniques have improved the characterisation of chemical contaminants in the environment and provides a correlation for useful prediction of the type of contaminant expected in various environmental matrices.

Abstract Sorghum bicolor plant has a high abundance of 3-deoxyanthocyanins, flavonoids and other polyphenol compounds that have been shown to offer numerous health benefits. Epidemiological studies have linked increased intake of S. bicolor to reduced risk of certain cancer types, including lung adenocarcinoma. S. bicolor extracts have shown beneficial effects in managing hepatorenal injuries. This study investigated the cytotoxic potential of three apigeninidin-rich extracts of S. bicolor (SBE-05, SBE-06 and SBE-07) against selected cancer cell lines and their ameliorative effect on aflatoxin B1 (AFB1)-mediated hepatorenal derangements in rats. We observed that, among the three potent extracts, SBE-06 more potently and selectively suppressed the growth of lung adenocarcinoma cell line (A549) (IC50 = 6.5 μg/mL). SBE-06 suppressed the expression of STAT3 but increased the expression of caspase 3. In addition, SBE-05, SBE-06 and SBE-07 inhibited oxidative and nitrosative stress, inflammation, and apoptosis and preserved the histoarchitectural networks of the liver and kidney of rats treated with AFB1. These in vitro and in vivo studies indicate the potential of these cheap and readily accessible extracts for cancer therapy and as chemo-preventive agents in preventing aflatoxin-related health issues.

Background. Vulvar schistosomiasis is a female genital schistosomiasis (FGS), which occurs because of the damage caused by the presence of schistosome ova within the vulva. FGS is mostly misdiagnosed as a sexually transmitted infection. There is no reported case of vulvar schistosomiasis from Schistosoma mansoni in an immunocompetent or immunocompromised person in Ghanaian medical literature; however, there is a reported case of S. haematobium in an immunocompromised person. This is the first case of vulvar schistosomiasis from S. mansoni infection in an immunocompromised person. This case report discusses the need to consider vulvar schistosomiasis in patients with itchiness of the vulva. Case Presentation. A sixty-nine-year-old married woman presents with a persistent vulvar itch that is unresponsive to treatment. A clinical diagnosis of vulvar lichen planus unresponsive to medical therapy was made. A histopathological diagnosis of vulvar schistosomiasis was, however, made. Ziehl–Neelsen stain revealed the ova of Schistosoma mansoni. Symptoms resolved on administration of oral praziquantel. Conclusion. Vulvar schistosomiasis must be considered in clinical history-taking and investigation of signs and symptoms related to itchiness of the vulva. Ziehl–Neelsen staining is a helpful histopathology armamentarium to determine the species of schistosome ova.

Introduction: Glaucoma, a disease of retinal ganglion cell (RGC) injury and potentially devastating vision loss, is associated with both ocular hypertension (OHT) and reduced ocular blood flow. However, the relationship between OHT and retinal capillary architecture is not well understood. In this project, we studied microvasculature damage in mice exposed to mild levels of induced OHT.Methods: Mild OHT was induced with the microbead model for 2 weeks. At this time point, some retinas were immunostained with CD31 (endothelium), Collagen IV (basement membrane), and RBPMS (RGCs) for z-stack confocal microscopy. We processed these confocal images to distinguish the three retinal capillary plexi (superficial, intermediate, and deep). We manually counted RGC density, analyzed vascular complexity, and identified topographical and spatial vascular features of the retinal capillaries using a combination of novel manual and automated workflows. Other retinas were dissociated and immunopanned to isolate RGCs and amacrine cells (ACs) for hypoxia gene array analysis.Results: RGC counts were normal but there was decreased overall retinal capillary complexity. This reduced complexity could be explained by abnormalities in the intermediate retinal capillary plexus (IRCP) that spared the other plexi. Capillary junction density, vessel length, and vascular area were all significantly reduced, and the number of acellular capillaries was dramatically increased. ACs, which share a neurovascular unit (NVU) with the IRCP, displayed a marked increase in the relative expression of many hypoxia-related genes compared to RGCs from the same preparations.Discussion: We have discovered a rapidly occurring, IRCP-specific, OHT-induced vascular phenotype that precedes RGC loss. AC/IRCP NVU dysfunction may be a mechanistic link for early vascular remodeling in glaucoma.

Aflatoxin B1 (AFB1) is a recalcitrant metabolite produced by fungi species, and due to its intoxications in animals and humans, it has been classified as a Group 1 carcinogen in humans. Preserving food products with Sorghum bicolor sheath can minimise the contamination of agricultural products and avert ill health occasioned by exposure to AFB1. The current study investigated the ameliorating effect of Sorghum bicolor sheath hydrophobic extract (SBE-HP) enriched in Apigenin (API) on the hepatorenal tissues of rats exposed to AFB1. The SBE-HP was characterised using TLC and LC-MS and was found to be enriched in Apigenin and its methylated analogues. The study used adult male rats divided into four experimental cohorts co-treated with AFB1 (50 µg/kg) and SBE-HP (5 and 10 mg/kg) for 28 days. Biochemical, enzyme-linked immunosorbent assays (ELISA) and histological staining were used to examine biomarkers of hepatorenal function, oxidative status, inflammation and apoptosis, and hepatorenal tissue histo-architectural alterations. Data were analysed using GraphPad Prism 8.3.0, an independent t-test, and a one-way analysis of variance. Co-treatment with SBE-HP ameliorated an upsurge in the biomarkers of hepatorenal functionality in the sera of rats, reduced the alterations in redox balance, resolved inflammation, inhibited apoptosis, and preserved the histological features of the liver and kidney of rats exposed to AFB1. SBE-HP-containing API is an excellent antioxidant regiment. It can amply prevent the induction of oxidative stress, inflammation, and apoptosis in the hepatorenal system of rats. Therefore, supplementing animal feeds and human foods with SBE-HP enriched in Apigenin may reduce the burden of AFB1 intoxication in developing countries with a shortage of effective antifungal agents.

This study compared the quality of life (QoL) and executive functioning in acromegaly patients vs healthy controls.Materials and methods: This was an observational case–control study on 38 subjects divided into 19 acromegaly patients and 19 matched controls. The groups were evaluated for QoL, attention, and executive function. All subjects completed Acromegaly Quality of Life Questionnaire (AcroQoL), Trail Making Test (parts A and B), Stroop, and phonemic fluency tests.Results: Acromegaly patients had an AcroQoL global score that was significantly lower than controls. The present study investigated the individual and combined impact of organophosphorus pesticide chlorpyrifos (CPF) and manganese (Mn), a naturally occurring trace metal, on hepatorenal function in adult rats. The four experimental groups namely control, CPF alone (5 mg/kg), Mn alone (10 mg/kg) and the co-exposure group consisted of eight rats which were orally gavage for 14 consecutive days. Following sacrifice, the biomarkers of hepatorenal damage, antioxidant enzyme activities, myeloperoxidase (MPO) activity as well as levels of nitric oxide, reactive oxygen and nitrogen (RONS) species and lipid peroxidation (LPO) were analysed spectrophotometrically. Balanced chromosomal translocations that generate chimeric oncoproteins are considered to be initiating lesions in the pathogenesis of acute myeloid leukemia. The most frequent is the t(15;17)(q22;q21), which fuses the PML and RARA genes, giving rise to acute promyelocytic leukemia (APL). An increasing proportion of APL cases are therapy-related (t-APL), which develop following exposure to radiotherapy and/or chemotherapeutic agents that target DNA topoisomerase II (topoII), particularly mitoxantrone and epirubicin. To gain insights into molecular mechanisms underlying the formation of the t(15;17) we mapped the translocation breakpoints in a series of t-APLs, which revealed significant clustering according to the nature of the drug exposure. Remarkably, in approximately half of t-APL cases arising following mitoxantrone treatment for breast cancer or multiple sclerosis, the chromosome 15 breakpoint fell within an 8-bp "hotspot" region in PML intron 6, which was confirmed to be a preferential site of topoII-mediated DNA cleavage induced by mitoxantrone. Chromosome 15 breakpoints falling outside the "hotspot", and the corresponding RARA breakpoints were also shown to be functional topoII cleavage sites. The observation that particular regions of the PML and RARA loci are susceptible to topoII-mediated DNA damage induced by epirubicin and mitoxantrone may underlie the propensity of these agents to cause APL.
