12. Topographic and faults control of hydrothermal circulation along dormant faults in an orogen

Abstract

Hydrothermal systems involving dormant faults within orogenic belts are rarely targeted for geothermal exploration, partly because of the complexity of the 3‐D topography, the unknown permeability of the fault zones and the basement lithology, and the lack of deep‐level data. This study brings together various types of surface information (spring features, geological data, topography, and hydrochemistry) to explain the alignment of 29 hot springs (29–73 °C) along the dormant Têt fault (Eastern Pyrénées, France). Water ion concentrations, stable water isotopes, and lithium isotopic ratios indicate that (i) fluids originating from meteoric water infiltrate above an altitude of 2,000 m, (ii) the rocks interacting with the fluids are similar for all the springs, and (iii) the maximum fluid temperatures at depth show similar variations along the fault and at the surface. A 3‐D numerical model of the system, assembled from field structural data and from a digital elevation model, explores the permeability combinations for the basement and for a three‐fault network. The models indicate that for a relatively permeable basement (10−16 m2), fluids are topography‐driven down to thousands of meters (until −3,700 m) before being captured by the more permeable Têt fault. Hot spring temperatures can be numerically reproduced when fault permeability is around 10−14 m2, a value slightly lower than the critical permeability for which free convection would occur within the Têt fault. Our study shows that thermal anomalies are possible along dormant faults close to elevated topography in the core of an orogenic belt, thereby opening new perspectives for geothermal exploration.

Abstract

International audience; We analyze the magnitudes and distributions of displacements on normal faults that are contained within a five layer marly-limestone/silty-clay sedimentary sequence. Observations of bedding plane exposures of the wellexposed mechanically isolated faults reveal a systematic strong decrease in the displacement gradient related to the horizontal lengthening of faults growing at constant height. Because mechanically heterogeneous sequences produce populations of vertically restricted faults having a large range of aspect ratios (length/downdip height), much of the scatter on a displacement-length diagram can be attributed to nonlinear growth paths. Our results demonstrate the significant influence of layering on fault scaling relations, growth, and maximum dimensions.

Abstract

3D seismic data located in the Gjallar Ridge (Vøring Basin, offshore Norway) reveals a closely-spaced polygonal fault system affecting more than 800 m of homogeneous mud-dominated Quaternary and Tertiary sequences. As some faults reach the modern seafloor, they represent an active polygonal fault system at present day. Even if the processes remain unclear and are still under debate, it is generally agreed that the initiation of polygonal faults is the result of shallow burial dewatering of fine-grained unconsolidated sediments by volumetric compaction. 3D seismic data are commonly interpreted by propagating horizons automatically and by picking faults manually. However, in the case of polygonal fault intervals, this approach is time consuming due to the huge number of faults and because automatic propagation can be misleading. In this study, we applied a new technique of 3D seismic interpretation based on a sequential stratigraphy analysis, using the new PaleoScan© software (Eliis Company). It allowed us to build a 3D geological model computing more than 300 horizons within the faulted intervals. We then used the coherency attribute, depicting anomalies in the shape of seismic waveform like faults, in order to constrain a possible link between fault distribution and stratigraphic levels. Our approach allows fault throws to be calculated in milliseconds on any polygonal fault plane. The result shows that fault segments have been reactivated by dip-linkage. Distribution of faults depends on mechanical units, intervals characterized by different petrophysical properties, which are independent from lithological and diagenetic changes. According to these results, we propose a model showing the evolution of polygonal fault intervals in which faulting stages are separated by a quiescence phase during burial. Abstract

Quantum mechanics, molecular dynamics and statistical mechanics methods are used to analyze the importance of neutral Hoogsteen-wobble G.C pairing in the stabilization of triple helices based on the poly-(G.C-C) trio at neutral pH and low ionic strength. In spite of the existence of a single hydrogen bond, the Hoogsteen-wobble G.C pair is found to be quite stable both in gas phase and solvated DNA. Molecular dynamics simulations of different triplexes based on the d(G.C-C) trio leads to stable structures if the neutral d(G.C-C) steps stabilized by Hoogsteen-wobble pairs are mixed with d(G.C-C+) steps. Finally, high level ab initio calculations and thermodynamic integration techniques are used to determine the relative stability of G.C wobble and G.C imino pairings. It is found that triplexes containing the imino pairing are slightly more stable structures than those with the wobble one, due mainly to a better stacking.

Abstract

The synthesis and the pharmacological activity of a series of 1,5-diarylimidazoles developed as potent and selective cyclooxygenase-2 (COX-2) inhibitors are described. The new compounds were evaluated both in vitro (COX-1 and COX-2 inhibition in human whole blood) and in vivo (carrageenan-induced paw edema, air-pouch, and hyperalgesia tests). Modification of all the positions of two regioisomeric imidazole cores led to the identification of 4-[4-chloro-5-(3-fluoro-4-methoxyphenyl)imidazol-1-yl]benzenesulfonamide (UR-8880, 51f) as the best candidate, which is now undergoing Phase I clinical trials.

43. Theoretical Studies on the Inhibition Mechanism of Cyclooxygenase-2. Is There a Unique Recognition Site?

Abstract

The mechanism of binding of different nonsteroidal anti-inflammatory drugs to the cyclooxygenase active site of cyclooxygenase-2 (COX-2) has been studied by means of a wide range of theoretical techniques including molecular dynamics and free energy calculations. It is found that theoretical methods predict accurately the binding of different drugs based on different scaffolds. Calculations allow us to describe in detail the key recognition sites and to analyze how these recognition sites change depending on the scaffold of the drug. It is concluded that the recognition site of COX-2 is very flexible and can adapt its structure to very subtle structural changes in the drug.

The major tautomeric forms in the gas phase and aqueous solution have been determined. The ability of oxanosine to recognize thymine and cytosine in the gas phase and in the DNA environment has been compared with that of guanine. A physicochemical explanation for the mutagenic properties of oxanosine is suggested.

47. The GAGA factor of Drosophila binds triple-stranded DNA.

Abstract

The Drosophila GAGA factor binds specifically to simple repeating d(GA.TC)n DNA sequences. These sequences are known to be capable of forming triple-stranded DNA as well as other non-B-DNA conformations. Here, it is shown that GAGA binds to a d[CT(GA.TC)]22 intermolecular triplex with similar specificity and affinity as to a regular double-stranded B-form d(GA.TC)22 sequence. The interaction of GAGA with triplex DNA cannot, however, stimulate transcription in vitro. The affinity of GAGA for triplexes of the purine motif, such as a d[AG(GA.TC)]22 intermolecular triplex, is significantly lower. The DNA binding domain of GAGA is sufficient for efficient binding to triplex DNA. Based on the reported solution structure of the complex of GAGA-DNA binding domain with double-stranded DNA, a model for its interaction with triplex DNA is proposed in which most of the protein-DNA contacts observed in duplex DNA are maintained, especially those occurring through the minor groove. The higher negative charge of the triplex is likely to have also an important contribution to both the specificity and affinity of the interaction.

48. Grain Deformation Processes in Porous Quartz Sandstones – Insight from the Clusters of Cataclastic Deformation Bands

Abstract

Porous sandstones represent classical reservoirs for water or hydrocarbons. Deformation in such granular material is due to tectonic events and occurs through the process of cataclasis, implying the crushing of the grains to a diversity of smaller clasts. Cataclasis is generally accompanied by porosity and permeability decrease. Although it is known that cataclastic deformation localizes to form individual bands and clusters of bands, the parameters controlling the distribution of this deformation were not well understood until recently. We used scanline measurements to show a favoured localization and clustering of the deformation on the case of normal-fault stress regime and potentially in strike-slip fault regime. The reverse regime favours the formation of distributed networks of conjugate deformation bands. At the scale of a cluster, field data reveals that the minimum modal grain size value of the host sandstone(s) controls the band density. Finally, microscopic cathodoluminescence analysis reveals enhanced quartz cementation for high degree of cataclasis. Hence, because band clustering, high degree of cataclasis and band cementation are favoured in normal-fault stress regime, tectonic extension appears to be favourable conditions for the formation of efficient barriers to fluid-flow in porous sandstone reservoirs.

Abstract

SETD8 is a methyltransferase that is overexpressed in several cancers, which monomethylates H4K20 as well as other non-histone targets such as PCNA or p53. We here report novel SETD8 inhibitors, which were discovered while trying to identify chemicals that prevent 53BP1 foci formation, an event mediated by H4K20 methylation. Consistent with previous reports, SETD8 inhibitors induce p53 expression, although they are equally toxic for p53 proficient or deficient cells. Thermal stability proteomics revealed that the compounds had a particular impact on nucleoli, which was confirmed by fluorescent and electron microscopy. Similarly, Setd8 deletion generated nucleolar stress and impaired ribosome biogenesis, supporting that this was an on-target effect of SETD8 inhibitors. Furthermore, a genome-wide CRISPR screen identified an enrichment of nucleolar factors among those modulating the toxicity of SETD8 inhibitors. Accordingly, the toxicity of SETD8 inhibition correlated with MYC or mTOR activity, key regulators of ribosome biogenesis. Abstract

G protein-coupled receptors (GPCRs) are sophisticated signaling machines able to simultaneously elicit multiple intracellular signaling pathways upon activation. Complete (in)activation of all pathways can be counterproductive for specific therapeutic applications. This is the case for the serotonin 2 A receptor (5-HT 2A R), a prominent target for the treatment of schizophrenia. In this study, we elucidate the complex 5-HT 2A R coupling signature in response to different signaling probes, and its physiological consequences by combining computational modeling, in vitro and in vivo experiments with human postmortem brain studies. We show how chemical modification of the endogenous agonist serotonin dramatically impacts the G protein coupling profile of the 5-HT 2A R and the associated behavioral responses. Importantly, among these responses, we demonstrate that memory deficits are regulated by G αq protein activation, whereas psychosis-related behavior is modulated through G αi1 stimulation. These findings emphasize the complexity of GPCR pharmacology and physiology and open the path to designing improved therapeutics for the treatment of stchizophrenia., (© 2024. The Author(s).)
