Campylobacter, one of the most prevalent bacteria responsible for gastroenteritis in human worldwide. Campylobacter infections result from ingestion of contaminated food and water depending upon susceptibility of individuals and virulence of infecting strain. The prevalence of these bacteria is highly diverse and present worldwide. Increase in antibiotic resistance among these bacteria is recognized as emerging public health problem. Current study is designed to analyze the risk factors related with Campylobacteriosis, prevalence in meat (chicken, beef and mutton) and antimicrobial resistance profiles of these bacteria from district Okara, Pakistan. The time span for lab work was from October 2020 to March 2021including sampling and testing. A total of 300 swab samples were collected from poultry, beef and mutton. The ISO 10272-1:217(E) method for isolation through biochemical and molecular identification carried out. Disc diffusion method used for AMR calculation. The overall prevalence of campylobacter was only 2%, detected only in poultry meat out of 300 samples. Among the isolated strains, Campylobacter indicated highest resistance against Tetracycline 100%, followed by Cefotaxime, Tylocine 83%, Enrofloxacin 66%, Clindamycin and Nalidixic acid 50%, lowest resistance against Streptomycin and Doxycycline only 33% and 16% respectively. The significance relation was obtained for two variables that include eating during dealing meat and contact of excreta with meat. Prevalence of Campylobacter is very low as compared to study conducted earlier in Pakistan. The drug of choice for Campylobacter related infections is doxycycline and streptomycin. The results indicate that prevalence of Campylobacter is very low on meat in Okara region but higher microbial resistance.

Newcastle disease virus (NDV), an avian orthoavulavirus, is a causative agent of Newcastle disease named (NDV), and can cause even the epidemics when disease is not treated. Previously several vaccines based on attenuated and inactivated viruses have been reported which are rendered useless with the passage of time due to versatile changes in viral genome. Therefore, we aimed to develop an effective multi-epitope vaccine against the haemagglutinin neuraminidase (HN) protein of 26 NDV strains from Pakistan through a modern immunoinformatic approaches. As a result, a vaccine chimaera was constructed by combining T-cell and B-cell epitopes with the appropriate linkers and adjuvant. The designed vaccine was highly immunogenic, non-allergen and antigenic; therefore, the potential 3D-structureof multi epitope vaccine was constructed, refined and validated. The study was designed to determine the prevalence and epidemiology of SARS-CoV-2 variants during the fifth wave of COVID-19 in selected districts of Punjab, Pakistan. The nasal swab samples (n=708)1 of suspected patients from various districts across Punjab were collected between December 2021 to April 2022. The genome extraction was done using an auto-extractor (Uni-medica) in the BSL-3 facility. Reverse-transcription real-time polymerase chain reaction (RT-PCR) was employed for viral detection and quantification. For the identification of SARS-CoV-2 variants, various mutations of spike protein were targeted using Multiplex PCR. The study’s results revealed the Omicron variant of concern (VOC) as the prevalent lineage of SARS-CoV-2 circulating in the selected regions of Punjab at the time of sampling. The VOC accounted for 90.01% of COVID-19 cases, followed by Delta (6.81%) and wild variant (3.80%). Prevalence of Omicron (VOC) was recorded higher in men (47.96%) as compared to women (42.05%). In addition, the highest percentage of VOC was observed in adults (47.39%) as compared to older people (32.07%) and young people (10.55%). This study highlighted the circulation of the Omicron variant during 5 wave of COVID-19 in Punjab Province, Pakistan.

PurposeIn the modern era, libraries confront significant service challenges. Some challenges are linked to information resource management which includes direct availability of information for immediate decision making. The Internet of Things (IoT) is a recent technological shift that library personnel should be aware of because it has the potential to enhance information resource management. The purpose of the research is to highlight the willingness to adopt IoT technology in libraries.Design/methodology/approachThis study uses a quantitative research design in which a survey of public sector universities in Nanjing, China, is conducted to investigate the determinants of IoT adoption intention in libraries. A total of 389 responses were captured from experienced library personnel. The literature on technology adoption is then used to formulate quantitative theories. For data analysis, partial least squares structural equation modeling using SmartPLS.FindingsThe research highlights the various success factors which support the IoT service adoption process. It is concluded that IoT augmented services in academic libraries must be supported through robust management practices and effective utilization of technological resources. Many libraries have made substantial modifications to their structure in terms of technology and design to satisfy the demands of patrons.Originality/valueThis is an empirical paper that looks at IoT adoption intention in libraries using a quantitative approach through surveying library personnel. The library personnel can aid in the understanding of the motivations behind technology adoption in libraries, particularly of IoT services that may bring about advances in the libraries' capability to provide information access services.

The causative agent of Newcastle disease (ND) is Newcastle disease virus. It belongs to avian species of Orthoavulavirus, Avulavirinae subfamily and if left untreated it may cause epidemic in poultry. Many vaccines have been made against Newcastle disease based on inactivated and attenuated viruses but become useless due to the genetic changes in the virus. We have recently reported epitope based vaccine by using immunoinformatics approaches. The vaccine was previously constructed against Hemagglutunin neuraminidase protein of Newcastle disease virus. Here we extended our work to develop several chimera of the proposed vaccine to design a new multi-epitope vaccine by shuffling the cytotoxic T lymphocytes (CTL) segments of the vaccine. Total 5040 constructs have been analyzed by shuffling 7 CTL epitopes. Highest antigenic multi-epitope construct was selected for the further study. Our new multi-epitope vaccine (MEV) construct contains 259 amino acids and is immunogenic, more antigenic and non-allergen. The refinement of the structure of MEV construct was performed. Molecular docking analyses showed its maximum binding with avian Toll-like 4 receptor. Subsequently, immune simulations showed its predicted ability to induce the host primary and secondary responses. Study suggests that our new multi-epitope vaccine chimera is more effective and stable protein against Newcastle disease virus strains in Pakistan. However, further studies are required to validate the vaccine through In vitro and In vivo studies.

Since the emergence of COVID-19 pandemic in China in late 2019, scientists are striving hard to explore non-toxic, viable anti-SARS-CoV-2 compounds or medicines. We determined In vitro anti-SARS-CoV-2 activity of oral formulations (syrup and capsule)of an Iodine-complex (Renessans). First, cell cytotoxicity of Renessans on the Vero cells was determined using MTT assay. Afterwards, the antiviral activity of Renessans was determined using viral inhibition assays and TCID50. For this, nontoxic concentrations of the Renessans were used. The results showed that Renessans is nontoxic to the cells up to 50 µg/mL. At 1.5 µg/mL concentration, SARS-CoV-2 production was significantly reduced to 101.43 TCID50 and 101.58 TCID50 for the syrup and capsule, respectively, as compare to virus infected control cells 106.08 TCID50 and we found the dose dependent inhibition of virus replication in the presence of Renessans. Renessans inhibited SARS-CoV-2 with an EC50 value of 0.425 µg/mL and 0.505 µg/mL for syrup and capsule, respectively. Furthermore, there was no virus detected at concentration of 50 µg/mL of Renessans. This study indicates that Renessans, containing iodine, have potential activity against SARS-CoV-2 which needs to be further investigated in human clinical trials.

The novel coronavirus (SARS-CoV-2) is a third member of its group that has introduced public health catastrophes around the globe. Since, its emergence in Wuhan, China by December 2019, SARS-CoV-2 infected millions of human population along with many casualties globally. The transmission potential of SARS-CoV-2 between humans has already been studied. Despite this transmission in human population, the primary origin of SARS-CoV-2 has been linked with bats by the help of an intermediate (secondary) host. This study was assumed to investigate the possible secondary or intermediate host to shuttle down the SARS-CoV-2 transmission and further to mitigate future pandemics. The antigenic surface/spike (S) protein was used for the structural and genomic analysis through currently available computer assisted technology. For the In-silico analysis, 43 sequences of S-protein of coronaviruses originated in various species were retrieved from nucleotide database of NCBI. These sequences were matched to find any similarities/differences by employing pairwise and multiple sequence alignment. The phylogenetic analysis was conducted to observe the relation among different species through MEGA software. Finally, comparative analysis for structures of S-protein (superimposition) was done with reference structure by using UCSF Chimera software. The results of this study expressed maximum match of S-protein sequences of human coronavirus with Bat and with Pangolin sequences respectively. The Phylogenetic analysis between Bat and Pangolin showed that SARS-CoV-2 transmitted from bats to humans possibly through the intermediate host of Pangolin. © 2021 Friends Science Publishers

In exchange rate determination models, the purchasing power parity (PPP) acts as a fundamental theory, which provides the basis for exchange rate determination across countries respectively. The main objective of this study is to investigate if there exists long-run purchasing power parity in the long run between the Asian economies and developed economies. This will help to understand the long-run exchange rate determination in these countries and helps to make effective monetary policies. This study tests the long-run PPP relationships between Asian economies (China, Japan, Singapore) with the UK and the US respectively. It is important to observe whether PPP holds between Asian economies and the economies of the UK and US by using time-series analysis respectively. Johansen Cointegration test is used to examine the relative PPP among these countries on full sample and sub-samples before and after the global financial crisis respectively. In full sample and subsamples, the Cointegration results show that PPP holds Asian economies (China, Japan, and Singapore) with the UK and the US respectively. This study finds significant evidence to accept that PPP holds which implies that the domestic prices of Asian economies are influenced by the US and UK prices and it explains a significant amount of variation in their domestic prices and inflation.

This paper attempts to find out the question of whether the benchmark bond yields affect thestock index in the US. To study the impact of the US bond market on the US stock indices, this study employs the Ordinary Least Square (OLS) technique, where the stock index of S&P 500, NYSE composite, and Dow Jones indices are used as the dependent variable, and US 10Yr Bond yield as the main independent variable. The time series used in this analysis were found nonstationary and the first difference is taken to convert them into stationary series. The Augmented Dickey Fuller(ADF) test statistics show that the series becomes stationary at first difference. Three separate regressions on S&P 500, NYSE Composite, and Dow Jones at first difference order are performed to analyze the effect of bond yield on stock indices with a reasonable set of control variables such as inflation, unemployment, crime rates, real effective exchange rate, etc. The statistical results show that the benchmark bond yield has a positive significant impact on the stock index of S&P 500, NYSE Composite, and Dow Jones. The economic implication of the results is that the benchmark bond yield predicts the stock index significantly and there is a moderate amount of correlation between the estimates and the time series as shown by the Variance Inflation Factor (VIF) respectively

Purpose The purpose of this paper is to describe the application of design and behavior science collectively in refining the government to citizens’ (G2C) cognitive-communication by using artificial intelligence (AI) artifacts. It investigates the citizens’ behavior intention to use AI cognitive-communication channels in e-government services. Design/methodology/approach This study reveals the potential of unification in design and behavior science paradigms when integrated into AI artifacts in measuring the constructive outcome of proposed architecture that embraces the perceived risk and trust in citizens’ usage behavior intentions. To evaluate the practical relevance of design science paradigm, action research was incorporated by developing an app as an exemplified scenario. Findings The weaknesses of traditional G2C channel communication obliterated by AI virtual agents. This study proposes a five-layer architecture model created with citizens-centered design highlighting the improved relevance for G2C cognitive-communication. The behavior model supports citizens’ usage behavior intentions through trust as a positive attribute and negative attribute as a perceived risk. Research limitations/implications This study contributes to the knowledge of design science paradigm particularly to AI-based cognitive G2C communication. The architecture serves for deploying virtual agents to automate the public officials for decision-making processes. The authors have constructed a cognitive-communication model and have examined the relationships among the extents of cognitive-communication model. This channel has the potential to significantly transform G2C cognitive and intelligent communication. Practical implications This cognitive-communication channel has the potential to significantly transform the communication between government and citizens. Such a model can be an interest of developers, researchers and government policymakers in a holistic approach drawn from theory and methods of both design and behavior science to create a successful communication scenario. Social implications Advances in technology are not without social implications. The study results can dramatically improve the efficiencies of government workplaces and can augment the human working. It will help to reduce the government services discriminations among the citizens and contribute to society through social inclusion. Originality/value This study addresses the duality in information system research by first defining the theoretical framework for G2C cognitive-communication architecture and then developing a research model drawn upon a citizens’ service usage behavioral theory. It will also contribute to providing important perceptions of AI practices in the public sector.

Purpose This study aims to fulfill the research gap by suggesting an appropriate and adaptable e-government (e-gov) maturity model for Pakistan not just in the abstract pattern relatively but also in a practical solution assured by the industry experts. Design/methodology/approach The qualitative research approach using key informants from the public sector domain; furthermore, e-gov performance artifacts were verified by the citizens through the process of focus group interviews. Findings The major finding of the study is the development of e-gov maturity model with implementations artifacts in proposed stages as follows: availability, interaction, integration, transactions and public participation. Research limitations/implications This study contributes a qualitative meta-synthesis in the field of e-gov maturity models and could support researchers who are in a quest of knowledge and references to develop new maturity models for their specific countries by providing them with useful resources for further investigation and study. Practical implications This maturity model will strengthen the argument that the e-gov services are necessary for the acceptance behaviour of the citizens and the prosperous public administration by the Government in Pakistan. This research strengthens the science-policy interface that has prevented governments from delivering changes on the ground to the public, and it will also identify diversified opportunities for the e-gov sector that can reverse the lack of investment in this domain. Social implications The study intends to provide directions to policymakers for the development of e-gov services for the citizens. Additionally, the public value of e-gov can be better understood in the form of citizens’ expectations from the government and this model will integrate public participation in the development of e-gov service. Originality/value The public value of e-gov can be better understood in the form citizens’ expectations from the government through this maturity model, furthermore, it can be recommended that the government can improve the relationship between the citizens and the state through the use of information and communication technologies which will strengthen the democratic process in Pakistan.

Due to fast transmission and various circulating SARS-CoV-2 variants, a significant increase of coronavirus 2019 infection cases with acute respiratory symptoms has prompted worries about the efficiency of current vaccines. The possible evasion from vaccine immunity urged scientists to identify novel therapeutic targets for developing improved vaccines to manage worldwide COVID-19 infections. Our study sequenced pooled peripheral blood mononuclear cells transcriptomes of SARS-CoV-2 patients with moderate and critical clinical outcomes to identify novel potential host receptors and biomarkers that can assist in developing new translational nanomedicines and vaccine therapies. The dysregulated signatures were associated with humoral immune responses in moderate and critical patients, including B-cell activation, cell cycle perturbations, plasmablast antibody processing, adaptive immune responses, cytokinesis, and interleukin signaling pathway. The comparative and longitudinal analysis of moderate and critically infected groups elucidated diversity in regulatory pathways and biological processes. Several immunoglobin genes (IGLV9-49, IGHV7-4, IGHV3-64, IGHV1-24, IGKV1D-12, and IGKV2-29), ribosomal proteins (RPL29, RPL4P2, RPL5, and RPL14), inflammatory response related cytokines including Tumor Necrosis Factor (TNF, TNFRSF17, and TNFRSF13B), C-C motif chemokine ligands (CCL3, CCL25, CCL4L2, CCL22, and CCL4), C-X-C motif chemokine ligands (CXCL2, CXCL10, and CXCL11) and genes related to cell cycle process and DNA proliferation (MYBL2, CDC20, KIFC1, and UHCL1) were significantly upregulated among SARS-CoV-2 infected patients. 60S Ribosomal protein L29 (RPL29) was a highly expressed gene among all COVID-19 infected groups. Our study suggested that identifying differentially expressed genes (DEGs) based on disease severity and onset can be a powerful approach for identifying potential therapeutic targets to develop effective drug delivery systems against SARS-CoV-2 infections. As a result, potential therapeutic targets, such as the RPL29 protein, can be tested in vivo and in vitro to develop future mRNA-based translational nanomedicines and therapies to combat SARS-CoV-2 infections.

The COVID-19 pandemic is striking the world with serious public health and socioeconomic complications. The pandemic has influenced all forms of daily life, including educational institutions. Therefore, this cross-sectional survey was conducted to understand knowledge, attitudes, and practices related to COVID-19 among the students of the University of Veterinary and Animal Sciences, Lahore. The data was collected using an online self-directed questionnaire. The survey form includes six items about sociodemographic characteristics, 14 knowledge-based questions, seven questions on attitude, and eight questions on practices. The sample number was calculated using the Raosoft sample size calculator. A total number of 3,854 students, including 1,823 men and 2,031 women, were engaged in this survey, having student representation from all the provinces in the country. The data were analyzed using a chi-square test. A total of 97% of the students knew that the etiological agent of COVID-19 is a virus and that it is a disease of the respiratory system (94%). Many students kept visiting their relatives during the lockdown (45%), and their relatives kept visiting them at home (59%). The responses from the students varied a lot on specific questions about the transmission of the virus. Women tended to have less information regarding precautionary travel measures (p < 0.01), but supplemental knowledge of prevention of disease transmission from positive patients (p < 0.01). Conclusively, the majority of the university students surveyed had imperative knowledge, a good attitude, and active practice in response to the COVID-19 outbreak. Moreover, the KAP scores have varied by demography, gender, and the number of family members. Therefore, continuous awareness of preventative behaviors should be disseminated regularly in emergencies.

Numerous studies have examined the role of social media as an open-learning (OL) tool in the field of education, but the empirical evidence necessary to validate such OL tools is scant, specifically in terms of student academic performance (AP). In today’s digital age, social media platforms are most popular among the student community, and they provide opportunities for OL where they can easily communicate, interact, and collaborate with each other. The authors of this study aimed to minimize the literature gap among student communities who adopt social media for OL, which has positive impacts on their AP in Chinese higher education. We adopted social constructivism theory (SCT) and the technology acceptance model (TAM) to formulate a conceptual framework. Primary data containing 233 questionnaires of international medical students in China were collected in January 2021 through the survey method. The gathered data were analyzed through structural equation modeling techniques with SmartPLS 3. The results revealed that perceived usefulness, perceived ease of use, and interactions with peers have positive and significant influence on OL. In addition, OL was found to have positive and significant influence on students’ AP and engagement. Lastly, engagement showed a positive impact on students’ AP. Thus, this study shows that social media serves as a dynamic tool to expedite the development of OL settings by encouraging collaboration, group discussion, and the exchange of ideas between students that reinforce their learning behavior and performance.

Foot and Mouth disease (FMD) is economically devastating, highly contagious transboundry viral disease of livestock with 100% morbidity, rapid spread and severe production losses in animals. The FMDV has seven different serotypes. There is no vaccine that can protect animals from all serotypes. Hence, it is need of the day to develop a vaccine that protects animals from hetrologous challenge. In this study, we used immunoinformatics approach to find T and B-cell epitopes that will help to construct a universal vaccine for FMDV. For this purpose, first we constructed a consensus sequence for four structural proteins (VP1, VP2, VP3 and VP4) of aphthovirus for seven serotypes (A, O, C, Asia1, SAT1, SAT2 and SAT3). Various computational tools were used to perform multiple sequence alignment to identify the conserved regions, generation of consensus sequence through conserved regions, structures prediction and finally prediction of B and T cell epitopes. We predicted 5 B cell and 18 T cell epitopes. Finally a GPGPG spacer was used to join these epitopes to decrease binding affinity around the core binding regions. Hence, our study identified the epitopes which can be used to develop cross protective vaccines against all the fatal strains of Aphthovirus which can easily protect all the serotypes. Though, successful In vivo and In vitro studies are required to determine the genuine strength of our predicted epitopes against the fatal strains of Aphthovirus.

Purpose The public sector use of the internet of Things (IoT) technology is an emerging field with a potential for a variety of institutional applications in the fields of information gathering and policy implementation to establish resilience for modern society. In this context, we aim to investigate the success factors which can positively influence the continuous usage intention of IoT service with the mediation effect of perceived public value. Design/methodology/approach A conceptual model based on the strong literature and theoretical background is tailored to provide factors for IoT services success in the public sector. A quantitative survey was conducted through 381 citizens using IoT services to validate the proposed conceptual model using structural equation modeling approach in order to examine the influence of identified constructs on the continuous usage intention. Findings The results highlighted that the proposed model successfully accounted for about 59% perceived variance in public value creation and 63% perceived variance in citizens' continuous usage intention. IoT services success is best dignified by the perceptions of citizens who use the services, therefore, this study identifies the positive role of digital society affinity in context of social sustainability and it climaxes that the real value of IoT in public sector comes from establishment of services on top of the service delivery and that's where the public value is going to be created. Practical implications The use of IoT technology in public services will fetch much benefit to the citizens as well as to the government. The study findings are significant in identifying good practices for generating public value in digital society affinity. Originality/value The integrated conceptual model explores the driving factors of citizens' inclination towards IoT technology in a social context. Through this study, we can analyze the role of perceived public value in enhancing the citizens' engagement with IoT services. The government policymakers and ICT managers take help from this study to designing IoT public services with making the government more responsive to citizens.
