Abstract The recent investigation targets to use adapted carbon paste (CP) with copper nanoparticles (CuNs) operating in a phosphate buffer (PBS) medium with a pH range of 5.0–8.0, to synthesize a novel, susceptible, and simple electrochemical sensor for the detection of one of the most important drugs, vitamin B6. Copper (Cu) is one of the most three common essential trace elements found in the bodies of both humans and animals, along with iron and zinc for all crucial physiological and biochemical functions. Its properties, which are assessed using a variety of methods including scanning electron microscopy (SEM), cyclic voltammetry (CV), differential pulse voltammetry (DPV), and electrochemical impedance spectroscopy (EIS), have also drawn a lot of attention recently. We considered the effects of pH, buffer, scan rate, interference, and calibration curve. The susceptible electrode's linear calibration curve encompassed concentration values between 8.88 and 1000.0 µM. The calculated limits of detection and quantification were 32.12 and 107.0 µM, respectively. Furthermore, this method was established in real human urine samples and drug validation which have been shown satisfactory results for vitamin B6 detection.

Abstract Endocrine cells in the fundic stomach of Clarias gariepinus were characterized in this work using transmission electron microscopy, immunohistochemistry, and histochemistry. Performic acid mixed with alcian blue pH2.5 and silver stain were among the histochemical stains used for endocrine cells. Endocrine cells can be found in the epithelium, lamina propria, submucosa, muscular layer, serosa, and the area between the stomach glands. Endocrine cells with one or more nuclei were found. Endocrine cells were studied using CD3, CD21, and CD68 in an immunohistochemistry analysis. The expression of the lymphocyte marker CD3 by endocrine cells is remarkable. In addition, they had a strong immunological response to CD21 and CD68, which are characteristics of phagocytic cells. Granules of varied sizes and electron densities are packed densely into the cytoplasm of the cells, as seen by transmission electron microscopy. We propose that endocrine cells play a crucial role in immune defense. The role of endocrine cells in the gut’s immune system is an area that needs further investigation.

Studying scute and fin morphology are advantageous approaches for phylogenetic identification and provide information on biological linkages and evolutionary history that are essential for deciphering the fossil record. Despite this, no prior research has precisely characterized the histological structures of scutes in the common pleco. Therefore, this research investigated the microstructure and organization of bone tissue within the dermal skeleton, including the scutes and fins, in the common pleco, using light microscopy, stereomicroscopy, and scanning electron microscopy. The dermal scutes were organized in a pentagonal shape with denticular coverage and were obliquely aligned with the caudal portion pointing dorsally. The dermal scutes consisted of three distinct portions: the central, preterminal, and terminal portions. Each portion comprised three layers: a superficial bony plate, a basal bony plate, and a mid-plate. Both the superficial and basal bony plates were composed of lamellar bone and lamellar zonal bone, whilst the mid-plate consisted of secondary osteons and woven bone. In the terminal portion, the superficial and basal bony plates became thinner. The pectoral fin consists of spines and rays composed of lepidotrichium (two symmetrical hemi-rays). The spine contained centrifugal and centripetal lamellar and trabecular bones. A centripetal fibrous bone was implanted between the lamellar bones. Besides being oriented in a V shape, the hemi-rays were also composed of thin centrifugal and centripetal lamellar bones and trabecular bones. A fibrous bone was identified between the centrifugal and centripetal bones. The trabecular bone and lamellar bone were made up of bone spicules.

Abstract Municipal wastewater treatment systems use the chemical oxygen demand test (COD) to identify organic contaminants in industrial effluents that impede treatment due to their high concentration. This study reduced the COD levels in tannery wastewater using a multistage treatment process that included Fenton oxidation, chemical coagulation, and nanotechnology based on a synthetic soluble COD standard solution. At an acidic pH of 5, Fenton oxidation reduces the COD concentration by approximately 79%. It achieves this by combining 10 mL/L of H2O2 and 0.1 g/L of FeCl2. Furthermore, the author selected the FeCl3 coagulant for the coagulation process based on the best results of comparisons between different coagulants. At pH 8.5, the coagulation dose of 0.15 g/L achieved the maximum COD removal efficiency of approximately 56.7%. Finally, nano bimetallic Fe/Cu was used to complete the degradation and adsorption of the remaining organic pollutants. The XRD, SEM, and EDX analyses proved the formation of Fe/Cu nanoparticles. A dose of 0.09 g/L Fe/Cu NPs, 30 min of contact time, and a stirring rate of 200 rpm achieve a maximum removal efficiency of about 93% of COD at pH 7.5. The kinetics studies were analyzed using pseudo-first-order P.F.O., pseudo-second-order P.S.O., and intraparticle diffusion models. The P.S.O. showed the best fit among the kinetic models, with an R2 of 0.998. Finally, the authors recommended that technique for highly contaminated industrial effluents treatment for agriculture or industrial purposes.

Abstract The current investigation aims to study the embryonic dermis formed in the early stages of development and identify the initial interstitial components of the dermis that serve as biological and structural scaffolds for the development of the dermal tissue. To investigate the dermal structure, the current study used morphological and immunological techniques. TCs identified by TEM. They had a cell body and unique podomeres and podoms. They formed a 3D network spread throughout the dermis. Homocellular contact established between them, as well as heterocellular contacts with other cells. Immunohistochemical techniques using specific markers for TCss CD34, CD117, and VEGF confirmed TC identification. TCs represent the major interstitial component in the dermal tissue. They established a 3D network, enclosing other cells and structures. Expression of VEGF by TC promotes angiogenesis. TCs establish cellular contact with sprouting endothelial cells. At the site of cell junction with TCs, cytoskeletal filaments identified and observed to form the pseudopodium core that projects from endothelial cells. TCs had proteolytic properties that expressed MMP-9, CD68, and CD21. Proteolytic activity aids in the removal of components of the extracellular matrix and the phagocytosis of degraded remnants to create spaces to facilitate the development of new dermal structures. In conclusion, TCs organized the scaffold for the development of future dermal structures, including fibrous components and skin appendages. Studying dermal TCs would be interested in the possibility of developing therapeutic strategies for treating different skin disorders and diseases.

ABSTRACT: The widespread use of antibiotics causes the development of antibiotic-resistant bacterial strains, which have a severe impact on poultry productivity and human health. As a result, research is continuing to develop safe natural antibiotic alternatives. In the current study, Bacillus pumilus SA388 was isolated from the chicken feces and confirmed to be a probiotic. The selected strain was tested for its antimutagenic and antioxidant capabilities before being employed as a probiotic food supplement and antibiotic alternative. The effect of B. pumilus SA388 impact on broiler chickens' growth performance, gut microbiome, blood biochemical markers, immunological response, and meat quality was also studied. B. pumilus SA388 showed significant bactericidal activity against Streptococcus pyogenes, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Salmonella typhi, and Klebsiella pneumonia. A total of 200 chickens were used in the present study, divided equally among four experimental groups (ten birds per group with 5 replicates): group 1 (control, G1) received a basal diet without B. pumilus SA388, group 2 (G2) received a basal diet supplemented with 0.4 mg/kg of B. pumilus SA388, group 3 (G3) received a basal diet supplemented with 0.8 mg/kg of B. pumilus SA388, and group 4 (G4) received a basal diet supplemented with 1.6 mg/kg of B. pumilus SA388. Over 35 d, the B. pumilus SA388-supplemented groups outperformed the G1 in terms of body weight gain, performance index, and feed conversion ratio, with a preference for the G4 treatment. The levels of alanine aminotransferase (ALT), aspartate transaminase (AST), low-density lipoprotein (LDL), and total cholesterol decreased significantly (P < 0.05) with increasing B. pumilus SA388 dosages compared to the control G1 group. Dietary supplementation of B. pumilus SA388 at 1.6 mg/kg (G4) significantly (P < 0.05) resulted in improved lipid profile, immunological response, thyroid function, and gut microbiota compared to the control group (G1). Compared to the broilers in the control treatment (G1), the addition of B. pumilus SA388 to broilers in G4 significantly (P < 0.05) enhanced juiciness, tenderness, aroma, and taste. Adding B. pumilus SA388 to chicken feed at different doses significantly (P < 0.05) decreased average feed intake while increasing economic and relative efficiency measures. In conclusion, B. pumilus SA388 has been proven to be an effective antibiotic and nutritional supplement.

Using semi-thin sections, the present investigation examined the microscopic characteristics of the lungs of the Nile monitor (Varanus niloticus). The lungs were composed of Intrapulmonary conducting airways and respiratory faveoli. Intrapulmonary airways originate from the terminal portion of the bronchus, which extends into the lung to create the bronchial tree. The bronchus was lined with pseudostratified ciliated epithelium composed of both ciliated and non-ciliated cells, and it was supported by plates of hyaline cartilage. The central lumen is surrounded by contractile fibers that contain smooth muscle cell bundles and are covered by ciliated and non-ciliated cells. The central lumen communicates with the faveoli. Separating adjacent faveoli are pulmonary trabeculae covered with various cell types, including type I pneumocytes, type II pneumocytes, and pulmonary macrophages. Some substantial pulmonary bronchi were also supported by small cartilage plate granules and lined with ciliated epithelium. Type I pneumocytes were flat cells, whereas type II pneumocytes had cuboidal cells with vacuolated cytoplasm. Surface irregularity and vacuolated cytoplasm were features of pulmonary macrophages. In addition, the connective tissue of the pulmonary septa contained immune cells, such as Mast and Eosinophils. In conclusion, the microstructure of the lung of the Nile monitor closely resembles that of other reptile species. However, the distinction between intrapulmonary cartilage palates and pulmonary septa raises the concept of species differentiation. In addition, the discovery of various types of pulmonary immune cells enhances the Nile monitor's ability to persist in a variety of environments by enhancing its pulmonary immunity.

ABSTRACT: This study explored the ability of formic acid (FA) to replace antibiotics in broiler chicken diets. It examined how FA affected the chickens' growth, carcass characteristics, blood chemistry, and gut bacteria. The experiment randomly assigned 300 one-day-old (Ross 308) broiler chicks to 5 groups, each divided into 6 replicates with 10 unsexed chicks. The following were the treatments: 1st group, negative control (NC): only received a basal diet; 2nd group, positive control (PC): received a basal diet supplemented with 0.5 grams of Colistin antibiotic per kilogram of diet; 3rd, 4th, and 5th groups (FA2, FA4, and FA6) these groups received a basal diet along with formic acid added at increasing levels: 2, 4, and 6 Cm3 per kilogram of diet, respectively. Results found no significant differences in live body weight (LBW) or body weight gain (BWG) between treatment groups, except for LBW at one week and BWG at 0 to 1 and 4 to 5 wk of age. No significant variations were found in feed intake (FI) and feed conversion ratio (FCR) among the treatment groups, excluding FI and FCR at 1 to 2 wk of age. The treatments significantly impacted carcass traits, dressing percentage, breast meat, thigh meat, spleen, giblets, blood levels of urea, creatinine, total protein, globulin, and albumin, as well as the activity of enzymes alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in chicks fed different diets compared to control groups. The addition of FA to the diet significantly impacted antioxidant levels. Also, the FA2 group had the highest total bacterial count (TBC). However, the FA6 group was the opposite; it had the lowest levels of harmful bacteria, such as E. coli and Coliform. Supplementing broiler diets with formic acid improves blood parameters, antioxidant activity, and gut bacteria counts, with 4.0 cm³ formic acid/kg diet supplementation promoting optimal broiler health and product quality.

Abstract Telocytes are a unique interstitial cell type that functions in adulthood and embryogenesis. They have characteristic immunohistochemical phenotypes while acquiring different immunohistochemical properties related to the organ microenvironment. The present study aims to investigate the immunohistochemical features of embryonic telocytes during myogenesis and describe their morphology using light microscopy and TEM. Telocytes represent a major cellular constituent in the interstitial elements. They had distinguished telopodes and podoms and formed a 3D interstitial network in the developing muscles. They formed heterocellular contact with myoblasts and nascent myotubes. Telocytes also had distinctive secretory activity. Telocytes identified by CD34. They also express CD68 and MMP-9 to facilitate the development of new tissues. Expression of CD21 by telocytes may reveal their function in immune defense. They also express VEGF, which regulates angiogenesis. In conclusion, the distribution and immunological properties of telocytes in the myogenic tissue indicate that telocytes provide biological and structural support in the development of the myogenic tissue architecture and organization.

Abstract The mechanism of interaction between magnesite mineral and phosphoric acid (0.001–0.5 M) in addition to the determination of the protective properties for Ti alloy (working electrode) in phosphoric acid both with and without an inhibitor have been investigated by electrochemical impedance spectroscopy (EIS) and potentiodynamic polarization measurements. Results of electrochemical tests show that the corrosion resistance of titanium alloy in phosphoric acid solution only increased and hydrogen production decreased by either decreasing acid concentration or increasing immersion time associated with the thickening of the oxide film formed on the alloy surface. On adding magnesite, the corrosion resistance of Ti alloy is enhanced by increasing the phosphoric acid concentration (0.001–0.5 M) due to the formation of sparingly soluble magnesium phosphate film on the alloy surface that inhibits the effect of increasing hydrogen evolution reaction due to the pH value decreases. The increasing adsorption behavior of the magnesite inhibitor and decreasing its diffusion were deduced from EIS measurements. Thus, the addition of 3% magnesite minimizes the corrosion by forming a new protective film (Mg3(PO4)2), which differs from the traditional passive film and prevents the effect of the increase of hydrogen evolution. The surface morphology and chemical composition of the tested alloy were determined using scanning electron microscopy (SEM), Fourier transform Infra-Red spectroscopy (FTIR), X-ray diffraction (XRD), X-ray Fluorescence (XRF) and In situ Raman spectroscopy.

Objective: This research assesses the utilization of sieved olive pulp (SOP) in the diet of growing rabbits through the use of an exogenous enzyme or dry yeast as a pretreatment. Materials and Methods: One hundred sixty-eight male V-Line rabbits aged 5 weeks (weighing 550 ± 25 gm) were randomly divided into seven groups with eight replicates each as follows: the control group was fed a basal diet without supplementation; while the other six groups were fed basal diets containing 20% and 25% of SOP and supplemented with 0.1 gm/kg Econase (E), 0.5 gm/kg dry yeast (Y), and a combination of both. The experiment lasted for 8 weeks. Results: The results indicated that supplementation of E, Y, and EY into rabbit diets containing SOP improved live body weight, body weight gain, feed conversion ratio, and nutrient digestibil¬ity. A higher dressing percentage was observed in the groups fed a 20% and 25% SOP diet sup¬plemented with EY (p < 0.05). The treated groups showed an increase in total protein, albumin, globulin, A/G ratio, total antioxidant capacity, superoxide dismutase, and glutathione peroxidase (p < 0.05), while there was a significant decrease in triglycerides, total cholesterol, and malondi¬aldehyde levels (p < 0.05) compared to the control. Rabbit groups fed an SOP diet supplemented with E, Y, or EY demonstrated higher (p < 0.05) economic efficiency compared to the control. Conclusion: Supplementing the diet of rabbits containing SOP with exogenous enzymes and/or dry yeast enhances the nutritional value of SOP while improving rabbit performance, nutrient digestibility, and antioxidant status. ABSTRACT: This work aimed to investigate the effects of dietary frankincense oil and ginger on the growth efficiency of growing Japanese quail, including live body weight, body weight gain, feed intake, feed conversion ratio, carcass traits, and physical characteristics of the meat. In total, 150 unsexed Japanese quail chicks that were 7 d old were utilized in the experiment. The chicks were randomly divided into 5 groups. Each group was divided into 3 replicates with ten birds in a completely randomized design. Group 1 received a basal diet without supplements and was used as a control group. Groups 2 and 3 received basal diets with 250 and 500 mg of ginger per kg of diet, respectively. Groups 4 and 5 received basal diets with 200 and 400 mg of frankincense oil per kg of diet, respectively. Results showed that BW of chicks received 500 mg of ginger and the 2 levels of frankincense oil at 5 wk of age, and 250 mg of ginger and 400 mg of frankincense oil at 6 wk significantly increased. BWG was significantly increased by using 500 mg of ginger and 2 levels of frankincense oil at 1 to 3 wk, 250 mg of ginger and 400 mg of frankincense oil at 3 to 6 wk, and 1 to 6 wk of age, in comparison with the control group. Treatments insignificantly influenced feed intake (FI), and feed conversion ratio (FCR) was improved considerably by using 250 mg of ginger and 400 mg of frankincense at 3 to 6 wk and 1 to 6 wk of age, respectively. Gizzard% was notably reduced with 200 mg of frankincense oil. The pH value of meat was significantly increased by having 2 levels of ginger. Still, water holding capacity and tenderness significantly decreased owing to 500 mg of ginger and 400 mg of frankincense oil. We can conclude that adding ginger and frankincense oil to Japanese quail diets may be beneficial.

Abstract Bio-diesel is used for engine as a replacement of diesel fuel which is characterized by lower emission, low pollution and renewable some of fuel. This study focus on how to enhance the production of bio-oil from Jatropha seeds by using microwave and ultrasonic as pre-treatments. To achieve that, the effects of extraction temperature (60, 80, 100 and 120 °C) and extraction screw speed (60, 90 and 120 rpm) on oil extraction yield and quality, extraction energy requirements and extraction time and were studied. Studying the effect of pretreatments by microwave and ultrasonic on the yield, energy and time of extraction were studied. The results most important indicate that the highest oil yield (25.1%) was recorded at 120 °C extraction temperature and 60 rpm screw speed. The energy required for extraction ranged from 8 to 11.5 W.h depending on temperature and speed of extraction. The results indicated that using both pretreatments improve the oil yield by 5.03% for microwave and by 6.75% for ultrasonic. Finally, the results concluded that to produce 1 kg of biodiesel you need 1.1 kg raw oil and consume from 2052.5 W.h energy requirement.

Background: Chronic liver diseases such as hepatic tumors can affect the brain through the liver–brain axis, leading to neurotransmitter dysregulation and behavioral changes. Cancer patients suffer from fatigue, which can be associated with sleep disturbances. Sleep is regulated via two interlocked mechanisms: homeostatic regulation and the circadian system. In mammals, the hypothalamic suprachiasmatic nucleus (SCN) is the key component of the circadian system. It generates circadian rhythms in physiology and behavior and controls their entrainment to the surrounding light/dark cycle. Neuron–glia interactions are crucial for the functional integrity of the SCN. Under pathological conditions, oxidative stress can compromise these interactions and thus circadian timekeeping and entrainment. To date, little is known about the impact of peripheral pathologies such as hepatocellular carcinoma (HCC) on SCN. Materials and Methods: In this study, HCC was induced in adult male mice. The key neuropeptides (vasoactive intestinal peptide: VIP, arginine vasopressin: AVP), an essential component of the molecular clockwork (Bmal1), markers for activity of neurons (c-Fos), astrocytes (GFAP), microglia (IBA1), as well as oxidative stress (8-OHdG) in the SCN were analyzed by immunohistochemistry at four different time points in HCC-bearing compared to control mice. Results: The immunoreactions for VIP, Bmal1, GFAP, IBA1, and 8-OHdG were increased in HCC mice compared to control mice, especially during the activity phase. In contrast, c-Fos was decreased in HCC mice, especially during the late inactive phase. Conclusions: Our data suggest that HCC affects the circadian system at the level of SCN. This involves an alteration of neuropeptides, neuronal activity, Bmal1, activation of glia cells, and oxidative stress in the SCN.

ABSTRACT: This study used 300 1-day-old, sexless, developing chicks of Japanese quail to estimate the ability of vitamin C and/or garlic to antagonize the venomous influence of cadmium (Cd) on the hematological, immunological, and performance characteristics of developing Japanese quail. The quail was separated into 5 similar groups of 60 chicks apiece, and 6 duplicates (10 each) were given to each sub-group. The control group received a basal diet without any supplements. The Cd group was nourished with a basal diet of + 80 mg cadmium chloride (CdCl2)/kg diet. The 3rd group was fed a basal diet + 80 mg CdCl2/kg diet and complemented with a 200 mg Vitamin C (Cd + C)/kg diet. The 4th group was nourished with a basal diet + 80 mg CdCl2/kg diet and complemented by a 500 mg dried garlic powder (Cd + G)/kg diet. The 5th group was fed a basal diet + 80 mg CdCl2/kg diet, complemented by a 200 mg vitamin C/kg diet + 500 mg dried garlic powder (Cd + CG)/kg diet. Results showed that in the 5th group in which cadmium was added together with Vit C + garlic, there was an improvement in both live weight gain (1–42 d) and feed consumption (1–21 and 1–42 d ) compared to the group in which Cd was added alone. The addition of Vit C alone and together with garlic seems to completely improve the cadmium-related increase in alkaline phosphatase (ALP), and Aspartate aminotransferase (AST), and Malondialdehyde (MDA) levels when compared to the control. Compared to cadmium-polluted diets, quail that got cadmium and feed additives significantly reduced cadmium residue. In addition, the cadmium group's serum immunoglobulin M (IgM) level decreased significantly. These data imply that dietary supplementation with (C) or (G) may be beneficial in retrogressing the drop in immunoglobulin G (IgG) and IgM caused by Cd and minimizing Cd's deleterious influence on immunity.

ABSTRACT: The investigation examined the use of willow leaf extract (WLE) on broiler chickens, examining carcass characteristics, cecal microbiota, antioxidants, and blood parameters. In 4 groups of 300 chicks, a basal diet was given for 5 wk, and the first treatment was basal diet (C). The diets for the remaining 3 treatments (WLE150, WLE300, and WLE450) contained 150, 300, and 450 mg of willow leaf extract /kg, respectively. The study found that birds fed willow leaf extract supplements had significantly greater body weight (BW), body weight gain (BWG), and enhanced feed conversion ratio (FCR) vs. the control group. Birds fed at 450 mg/kg food showed the greatest growth features, carcass weight, liver weight, lower abdominal fat, better low-density lipoprotein (LDL), and high-density lipoprotein (HDL) concentrations, and highest hematological characteristics. Chickens fed diets supplemented with varied doses of willow leaf extract showed significantly increased antioxidant enzyme activity, with higher amounts of glutathione peroxidase (GPx) activity, superoxide dismutase (SOD), total antioxidant capacity (TAC), and lower malondialdehyde (MDA). However, in the study, birds fed a diet supplemented with 450 mg of willow leaf extract per kg meal showed a significant drop of 13.02%, which found no significant variations in hazardous bacteria (Escherichia coli) across 2 treatments (WLE150 and WLE300). In addition, the study discovered that birds fed with varied doses of willow leaf extract had fewer cecum infections (Staphylococci aureus). We conclude that using willow at a level of 450 mg/kg diet can significantly enhance the BWG, FCR, antioxidant levels and beneficial bacteria activity besides the condition of broiler chicken's general health.

Papaya is a sparsely branched tree and grows in many tropical and subtropical areas. Herein, this study compared the effects of papaya at 0.75 % as seeds meal (PSM), leaves meal (PLM), latex powder (PLP), or a mixture of them (PMIX) on the performances of Japanese quails. Two hundred unsexed Japanese quail (1st week-old) with an initial body weight of 21.08 ± 0.32 g were allocated into five experimental groups in cages with free access to water and test diets. Abstract Background Several methods have emerged to predict the occurrence of early volume overload (VO) in pediatric patients with chronic kidney disease undergoing regular hemodialysis (HD). Nevertheless, achieving an accurate assessment remains challenging. Consequently, this study aimed to identify VO in pediatric HD patients using lung ultrasound (LUS). Additionally, the study sought to investigate the relationship between various clinical parameters employed to detect VO and the ultrasonographic B-line score. Methods This prospective observational cohort study was conducted on 30 pediatric patients with end-stage renal disease undergoing a maintenance HD program for 4 months. The clinical evaluation of the fluid status of pediatric patients involved using LUS pre-, intra, and post-HD. The study included the dry weight (DW) and non-DW groups; within these groups, the B-line scores were evaluated pre-, intra, and post-HD sessions. Tabulations were conducted to document the variations in body weight and B-line scores during pre-, intra-, and post-dialytic periods. Abstract Telocytes establish connections and communicate with various types of cells and structures. Few experimental studies have been performed on telocytes. In this study, we investigated the effect of salinity stress on telocytes in relation to osmoregulatory, immune, and stem cells. After exposing the common carp to 0.2 (control), 6, 10, or 14 ppt salinity, we extracted and fixed gill samples in glutaraldehyde, processed and embedded the samples in resin, and prepared semi-thin and ultrathin sections. Two types of telocytes were identified: intraepithelial and stromal telocytes. Intraepithelial telocytes were found to form part of the cellular lining of the lymphatic space and shed secretory vesicles into this space. Stromal telocytes were observed to shed their secretory vesicles into the secondary circulatory vessels. Both intraepithelial and stromal telocytes were enlarged and exhibited increased secretory activities as salinity increased. They exerted their effects via direct contact and paracrine signaling. The following changes were observed in samples from fish exposed to high salinity levels: chloride cells underwent hypertrophy, and their mitochondria became cigar-shaped; pavement cells were enlarged, and their micro-ridges became thin and elongated; stromal telocytes established contact with stem cells and skeletal myoblasts; skeletal muscle cells underwent hypertrophy; and macrophages and rodlet cells increased in number. In conclusion, our findings indicate that intraepithelial and stromal telocytes respond to salinity stress by activating cellular signaling and that they play major roles in osmoregulation, immunity, and regeneration.

Abstract In this research, Mg–3Zn–xSi (x = 0, 0.1, 0.3, and 0.6 wt%) biodegradable alloys were produced by simple stir casting method, their microstructure and phase changes were evaluated using X-ray diffraction (XRD), optical microscope (OM) and scanning electron microscope coupled with energy dispersive x-ray analysis (SEM–EDS). Potentiodynamic polarization was conducted to measure the alloys’ corrosion behavior in simulated body fluid (SBF). Tensile strength test and in-vitro biocompatibility evaluation regarding MTT cytotoxicity, ALP osseointegration assay and MG-63 cell growth pattern were conducted. Electrochemical investigations showed that Mg–3Zn alloys enclosing Si attained degradation rates suitable for structural support until bone healing, while the Mg–3Zn alloy without Si had a corrosion rate of 0.128 mm/year which is much lower than the required value. None of the inspected alloys exhibited a significant cytotoxic effect, meanwhile, Mg–Zn base alloy and the alloy with 0.3 wt% Si demonstrated the highest ALP level. The optimum cell growth pattern was demonstrated for Mg–Zn base alloy and the alloy with 0.1 wt% Si. Evidence of calcium phosphate precipitation was observed in the four investigated Mg–3Zn alloys. Therefore, based on the fore mentioned results, Mg–3Zn–xSi alloys were suggested as viable biodegradable materials due to their compatible degradation rates, proved cytocompatibility, high cell viability and excellent osseointegration potential.

Previous research divided long bone growth into intramembranous and endochondral ossification. The latter contributes to longitudinal bone growth through the addition of new bone tissue at the physeal growth plate. Bones, also, increase in thickness through appositional bone growth of the cortical bone. The aim of the present study was to investigate the age-related histomorphometrical changes that occur during the growth of the humerus in rats as a model of the long bone. Samples were collected for light microscopy from the right humerus at 3, 4, 6, 8, 10, 13, and 20 weeks (wks) of age. Histological investigation revealed that the distal physeal growth plate could be detected at 3 wks of age, while at 4 wks of age it was replaced by bone trabeculae. Morphometrically, differences were observed in the thickness of the proximal physeal growth plate, specifically its proliferative zone. The thickness of the proliferative zone initially decreased, then showed a significant increase around the age of puberty (6–8 wks), then decreased significantly from 8–20 wks of age. The midshaft cortical bone thickness showed a significant increase during the study. In conclusion, ageing affected both longitudinal and appositional growth, which both participated in the growth and development of the rat's humerus.

ABSTRACT: The search for a natural antimicrobial agent is ongoing and critical because of the rise and rapid proliferation of antibiotic-resistant pathogenic bacteria. The current study aims to examine the effect of Paenibacillus polymyxa AM20 as an alternative antibiotic and feed additive on Indian river broiler performance, digestive enzymes, thyroid hormones, lipid profile, hepatosomatic index, immunological response, gut bacteria, and antioxidant parameters. The bacterial isolate AM20 was identified at the gene level by isolating DNA and using PCR to detect genes. Based on 16S rRNA gene sequence analysis, the bacterial isolate was identified as Paenibacillus polymyxa. One hundred twenty Indian river broilers (1-day old) were randomly divided into 4 groups of 10 chicks each, with 3 replicates. The control group was fed a basal diet only, while the other 3 were administered control diets supplemented with P. polymyxa at 3 concentrations: 0.5, 1, and 1.5 mg/kg. The findings revealed that all groups that received graded amounts of P. polymyxa increased all growth parameters throughout the study. P. polymyxa treatment at 1.5 mg/kg increased body gain by 9% compared to the control due to increased feed intake (P = 0.0001), growth rate (P = 0.0001), and decreased feed conversion ratio. Compared to the control group, P. polymyxa (1.5 mg/kg) enhanced kidney functions in chickens by reducing uric acid and creatinine levels (P = 0.0451). Compared to the control group, alanine aminotransferase and aspartate transaminase levels in the liver were significantly reduced at all P. polymyxa doses. Liver function values were highest for P. polymyxa at 1.5 mg/kg. Compared to the control group, those whose diets included P. polymyxa had significantly better blood cholesterol levels, high-density lipoprotein, low-density lipoprotein, immunological response, thyroid function, and gut microbiota. In general, broiler chickens' economic efficiency was improved by including P. polymyxa in their diet, which also improved their growth performance, carcass dressing, specific blood biochemical levels and enzymes, and the composition of the gut microbiota.

Abstract Energy scarcity and conventional energy problems are the main reason of finding a renewable source of energy which is cheap and environmental friendly, therefore, biodiesel production is one of the most promising solutions of this problem. Also, Egyptian castor is one of the most important crops for oil production compared with other commonly used oil crops. The main aim of this study is to enhance the production of bio-oil from Egyptian castor seeds by using microwave and ultrasonic as pre-treatments. To achieve that, the effects of extraction screw speed (20, 40 and 60 rpm) and temperature (100, 150, 200 and 250 °C) on oil extraction yield and quality, extraction energy requirements and extraction time and were studied. Also, the effect of pretreatment conditions of microwave at three levels of power (Low, Med and High) and different times (1, 2 and 3 min) and pretreatment condition ultrasonic with different temperatures (40, 60 and 80 °C) and different times (15, 30 and 45 min) for castor seeds before extraction with the optimum condition of the screw press on oil extraction yield from castor seeds, extraction energy, extraction time and quality of the oil extracted. The results indicate that the optimum conditions oil extraction by screw press were 200 °C extraction temperature and 60 rpm screw speed. It could be seen that the extraction oil yield, extraction energy requirements and extraction time were 35.59%, 18.68 and 1.86 min, respectively. Microwave pretreatments had better on oil yield and energy required for extraction compared to ultrasonic pretreatments, where, microwave pretreatments recorded high oil yield and lower energy requirements compared to the ultrasonic pretreatments. Oil yield ranged from 32.67 to 37.41% compared to 13.29 to 39.83% in literature. The time required for extraction was ranged from 1.77 to 2.00 and 1.79 to 2.21 min for microwave and ultrasonic pretreatments, respectively. The pretreatment improved properties of the extracted oil.

MAD2L1BP-encoded p31comet mediates Trip13-dependent disassembly of Mad2- and Rev7-containing complexes and, through this antagonism, promotes timely spindle assembly checkpoint (SAC) silencing, faithful chromosome segregation, insulin signaling, and homology-directed repair (HDR) of DNA double-strand breaks. We identified a homozygous MAD2L1BP nonsense variant, R253*, in 2 siblings with microcephaly, epileptic encephalopathy, and juvenile granulosa cell tumors of ovary and testis. Patient-derived cells exhibited high-grade mosaic variegated aneuploidy, slowed-down proliferation, and instability of truncated p31comet mRNA and protein. Corresponding recombinant p31comet was defective in Trip13, Mad2, and Rev7 binding and unable to support SAC silencing or HDR. Furthermore, C-terminal truncation abrogated an identified interaction of p31comet with tp53. Another homozygous truncation, R227*, detected in an early-deceased patient with low-level aneuploidy, severe epileptic encephalopathy, and frequent blood glucose elevations, likely corresponds to complete loss of function, as in Mad2l1bp–/– mice. Thus, human mutations of p31comet are linked to aneuploidy and tumor predisposition.

ABSTRACT: The objective of the current study was to ascertain the effect of using ginger powder or frankincense oil at different levels on the production rate, biochemical properties of blood, and immune response of laying Japanese quail housed from 12 to 21 wk of age. Three hundred sexually mature quail (200 females and 100 males) were distributed among 5 groups at 12 wk of age in a completely randomized design. Group 1: received basal diet not including additives and acted as a control group. Groups 2 and 3: received a basal diet with 250 and 500 mg ginger/kg feed, respectively. Groups 4 and 5: received a basal diet with 200 and 400 mg frankincense oil/kg feed, respectively. Results revealed that egg production parameters of laying Japanese quail were not influenced by ginger or frankincense oil added in all groups tested during experimental periods except at the time (15–18 wks.). Moreover, egg weight was significantly increased only during the period (15–18 wks.) and total period (12–21 wk of age), and group 5 recorded the highest weight during the 2 periods studied. Feed additives did not impact feed consumption or feed conversion ratio (FCR). Except for WBCs, Hb, and monocytes, treatments impacted blood hematological parameters. Also, blood serum parameters were influenced by feed additives, except total protein, albumin, globulin, and creatinine were not influenced. Moreover, histological examination of the spleen was influenced by feed additives. It is concluded that utilizing frankincense oil or ginger powder in diets of laying Japanese quails enhanced blood serum properties and improved reproductive and productive performance.

Keratinolytic microorganisms are highly valuable for decomposition of poultry waste. This study aimed to isolate keratin-decomposing actinobacteria from poultry farm soils and examine their capacity to decompose feathers. Soil samples were placed in a basal medium with feather meal, which is a deposit of carbon and nitrogen. Nine actinobacterial strains were isolated. Actinobacteria were cultured in the media to show clear feather-decomposing potential. Actinobacterial strains were identified using 16S ribosomal RNA (rRNA) sequencing as being related to Streptomyces rochei AM8. Thus, the supernatant of S. rochei AM8 exhibited keratinolytic enzyme activity. Increased biodecomposition of feathers was recorded in a keratinase assay (0.782 U/mL) for separated cultures. The ability of the selected microorganisms to decompose feathers may be an effective biotechnological solution for managing feather waste from poultry.

Abstract Background While executive dysfunctions are present abundantly in children with psychiatric and developmental disorders, unfortunately, it is significantly underdiagnosed in Arab countries due to the lack of Arabic executive functions diagnostic scales. To our knowledge, there is no available Arabic rating scale for assessing executive functions in children and adolescents till this moment except for BRIEF-2 which excluded anyone with a mental, learning, developmental, or medical disorder constituting unfortunately 25% of the general population. Our paper describes the translation and validation of an Arabic version of the Barkley Deficits in Executive Functioning Scale - Children and Adolescents (BDEFS-CA) (long form). The translation was done using the forward-backward method and the study population for validation included 60 parents of children of age 6–13 years. Results Reliability of the scale domains was evident in Arabic showing high internal consistency (Cronbach’s alpha from 0.93 to 0.97 scores over the five domains). These values were close to the original English version which is 0.95 to 0.97. Conclusion The reliability of the Arabic version of BDEFS-CA was adequate making it a valuable instrument for executive function assessment in Arabic children and adolescents.

Objective: We herein describe two cases of de novo lymphoid blastic transformation in patients with no history of chronic-phase chronic myeloid leukemia (CP-CML), both of whom were labeled initially as Philadelphia positive B-Acute Lymphoblastic Leukemia (B-ALL).Methods: The first patient was an 18-year-old male who presented with subjective fever, intentional weight loss, generalized fatigue, and headache. Investigations showed leukocytosis (312 × 10^3/ul), thrombocytopenia and anemia. Flowcytometry was consistent with B-ALL, with aberrant expression of CD13 and CD33. He was found to be positive for BCR::ABL by FISH, and karyotype confirmed the presence of the Philadelphia chromosome. He received a pediatric-inspired regimen and achieved remission with negative measurable residual disease (MRD) by flowcytometry, however with persistent cytogenetic abnormality using FISH for BCR::ABL. FISH abnormality was confirmed to be in the myeloid compartment using myeloid segregated FISH, reclassifying the disease to de novo lymphoid blastic phase CML. The second patient was a 52-year-old male who presented with fever and shortness of breath. Bilateral cervical lymphadenopathy and hepatosplenomegaly were identified on examination, and investigations showed leukocytosis (371 × 10^3/ul), anemia, and thrombocytopenia. BCR::ABL rearrangement was identified by FISH, molecular testing, and confirmed with karyotype. He was treated with Mini-CVD and Ponatinib, achieved complete remission with negative MRD by flow cytometry, however molecular studies showed BCR-ABL1 level at 58% IS indicating a persistent cytogenetic abnormality.Results: De novo lymphoid blastic-phase CML can therefore be difficult to differentiate from Philadelphia positive B-ALL due to their overlapping clinical and laboratory picture, implying the need to do myeloid compartment evaluation at the time of diagnosis.Conclusion: With recent progress in the treatment of Philadelphia positive B-ALL, including the role of transplant with the use of novel agents, a better characterization of this disease entity in retrospective and prospective trials is warranted.

Abstract A novel environmental nano-catalyst based on zeolite (ZE) adjusted with carbon nanotube/silver nanoparticles (Ag/CNT) ornamented carbon paste electrode (CPE) is used for electrochemical oxidation of propylene glycol (PG) in 0.5 M H2SO4 solution. The techniques like cyclic voltammetry (CV), chronoamperometry (CA) and electrochemical impedance spectroscopy (EIS) are utilized to achieve the catalytic activity performance. Surface characteristics are achieved by means of scanning electron microscope (SEM) and Energy dispersive X-ray analysis (EDX) techniques. Enhancing the loading magnitude of CNT into catalyst's ingredient can meaningfully develop the catalytic activity of the electrocatalyst towards propylene oxidation. The impact of altering the concentration of propylene glycol and the scanning rate on the resulting electrocatalyst performance during the oxidation cycle is considered. Chronoamperograms present an amplify of the steady state oxidation current density values after addition of these nano-catalysts. A promising catalytic stability of nano-catalyst has been achieved in electing its use for propylene glycol electro-oxidation in fuel cells applications.

We investigated the effects of fucoxanthin isolated from the edible macroalga Dilophys fasciola on pathogenic microbes and probiotics in vitro and the antioxidant activity of fucoxanthin. The yield concentration of the obtained crude was 50.5% fucoxanthin. We found strong inhibition against Gram-positive Staphylococcus aureus and Listeria monocytogenes, and lower inhibition against Gram-negative bacteria and fungi. The probiotic strains progressed between 1.2 and 1.67 log cycles at a concentration of 30 μg/mL. The antioxidant activity ranged between 54.76% and 88.36% at a concentration of 40 μg/mL. The 50% lethal dose of algal fucoxanthin was shown to be more than 2511.88 mg/kg. The production of stirred yoghurt incorporated with 20 mg and 30 mg of fucoxanthin per kilogram of milk was evaluated through chemical, microbiological, and sensory analyses during storage for 21 days and compared with control samples. The maximum growth for probiotics (Bifidobacterium bifidum and Lacticaseibacillus casei) was found on day 14, but more viability counts were detected in the treatment with 30 mg/kg. All treatments were free from mould and yeast counts up to 7 days, and the small numbers of mould, yeast, and psychrotrophic counts appeared first in control samples. Also, the highest dry matter content was observed for treatments with 30 mg/kg. Moreover, the protein, fat, and ash content of all treatments increased with a progressive cold storage period. Greater reductions in the pH were found in treatments than in the control, and were consistent with the development of acidity. During storage, the amount of crude fucoxanthin had no significant impact on the flavour, colour, or appearance scores. Significance: • Fucoxanthin is a type of carotenoid that offers many benefits to human health. • The fucoxanthin of edible Dilophys fasciola had a strong antimicrobial effect against Gram-positive bacteria, Gram-negative bacteria, and fungi. • Stirred yoghurt fortified with crude fucoxanthin had good overall acceptability and the percentage of crude fucoxanthin had no noticeable effects on the flavour, colour, or appearance. Fucoxanthin, therefore, has potential benefit as a food additive.

Abstract Aloe vera possesses a great therapeutic importance in traditional medicine. It has attracted the attention of modern medical fields due to its wide pharmacological applications. The bioactive substances in Aloe vera proved to have antioxidant, anti-inflammatory, antibacterial, and antiviral properties. Taken into our consideration the long history of clinical applications of Aloe vera in traditional medicine, especially for promoting the healing of cutaneous wounds with rare adverse effects, it provides a cheap alternative to many expensive synthetic drugs. Recent techniques in tissue engineering created novel scaffolds based on Aloe gel extracts for wound healing applications. Nonetheless, further guided researche is required to foster the development of Aloe vera based scaffolds for the benefit of worldwide populations. Here, I systemically summarize the main events following wounding and the mechanism of action of Aloe vera in promoting the healing process. I hope to provide a solid piece of information that might be helpful for designing new research studies into this topic.

Abstract This study investigates whether a chronotherapeutic treatment of hepatocellular carcinoma (HCC) may improve treatment efficacy and mitigate side effects on non‐tumoral liver (NTL). HCC was induced in Per2::luc mice which were irradiated at four time points of the day. Proliferation and DNA‐double strand breaks were analyzed in irradiated and nonirradiated animals by detection of Ki67 and γ‐H2AX. Prior to whole animal experiments, organotypic slice cultures were investigated to determine the dosage to be used in whole animal experiments. Irradiation was most effective at the proliferation peaks in HCC at ZT02 (early inactivity phase) and ZT20 (late activity phase). Irradiation effects on NTL were minimal at ZT20. As compared with NTL, nonirradiated HCC revealed disruption in daily variation and downregulation of all investigated clock genes except Per1. Irradiation affected rhythmic clock gene expression in NTL and HCC at all ZTs except at ZT20 (late activity phase). Irradiation at ZT20 had no effect on total leukocyte numbers. Our results indicate ZT20 as the optimal time point for irradiation of HCC in mice at which the ratio between efficacy of tumor treatment and toxic side effects was maximal. Translational studies are now needed to evaluate whether the late activity phase is the optimal time point for irradiation of HCC in man.

Abstract Background The ovotestis gland (the hermaphrodite gland) is a reproductive system organ that affects fertility of land snails. Thus, the present study was conducted to evaluate the effect of three inorganic salts (sodium carbonate, sodium benzoate, and sodium nitrate) against the land snail, Massylaea vermiculata under laboratory and field conditions. Snails were treated with serial concentrations of the tested salts for seven days using thin-layer film technique. The LC50 of each salt was determined, and the effect of ½ LC50 of each was tested on egg-laying. In addition, LPO enzyme was determined, and the histological effect of the ovotestis gland was inspected. The field efficiency of sodium carbonate and sodium nitrate were evaluated as a spray for 21 days on pomegranate nursery trees at Giza Governorate. The effects of inorganic salts were compared with methomyl (the compound recommended by the Egyptian Ministry of Agriculture against land snails in the field). Results The laboratory results revealed that sodium carbonate and sodium nitrate were more toxic than sodium benzoate, whereas the LC50 were 2.4, 9.6, and 11.8% for the three inorganic salts, respectively. Sodium carbonate and sodium nitrate achieved complete inhibition for egg production, while sodium benzoate gave 37.7% compared to 96.8% for hatchability control. Sodium carbonate and sodium benzoate exhibited significant effect on LPO enzyme, while a non-significant effect was recorded by sodium nitrite compared with control. Regarding the histological effect, sodium carbonate and sodium nitrate had the same effect on ovotestis, as a necrobiotic change was observed in the ova, and diminishing in spermatozoal concentration, while sodium benzoate caused decreasing in spermatozoal concentration without any alteration in the ovarian compared with control. Regarding the field results, sodium carbonate and sodium nitrate achieved 78.4 and 79.0% reduction in land snail populations, respectively, compared with 75% for methomyl compound. Conclusions The tested inorganic salts have highly toxic effect against the land snail, M. vermiculata, and they can be used in field control programs, after conducting more studies on their effect on the soil and other environmental components.

The purpose of this study is to detect the impact of the covalent immobilization of β-glucosidase (β-Glu) from Thermatoga maritima on sodium alginate-based magnetically recyclable iron nanoparticles (SA-Fe3O4 MNPs) for boosting robustness, facile retrieval, and recycling of enzyme. We developed a novel green method to amend the transformation of lactose to galacto-oligosaccharides (GOS). The interaction of the Immobilized β-Glu and SA-Fe3O4 MNPs were characterized by SDS-PAGE, Transmission electron microscope (TEM) and FT-IR spectroscopy. After immobilization, the ideal pH maintained 7.0, the optimum temperature (temp.) increased to 85 °C and deceptive Km and Vmax raised by 65 and 41%, respectively. Immobilization provided low activation energy and enhanced pH and heat stability. The SA-Fe3O4 MNPs maintained the effectiveness of enzyme by 39.52% up to the 6th cycle of reuse and the hydrolysis power exceed 28.13% of lactose to GOS. Conclusively, thermal stability, improved pH, and excellent reusability over β-Glu make SA-Fe3O4 MNPs a viable option for long-term GOS production as well as other industrial uses.

Industrial pomaces are cheap sources of phenolic compounds and fibers but dumping them in landfills has negative environmental and health consequences. Therefore, valorizing these wastes in the food industry as additives significantly enhances the final product. In this study, the citrus pomaces, orange pomace (OP), mandarin pomace (MP), and lemon pomace (LP) were collected by a juice company and subjected to producing polyphenols and fiber-enriched fractions, which are included in functional yogurt; the pomace powder with different levels (1, 3, and 5%) was homogenized in cooled pasteurized milk with other ingredients (sugar and starter) before processing the yogurt fermentation. The HPLC phenolic profile showed higher phenolic content in OP extract, i.e., gallic acid (1,702.65), chlorogenic acid (1,256.22), naringenin (6,450.57), catechin (1,680.65), and propyl gallate (1,120.37) ppm with massive increases over MP (1.34–37 times) and LP (1.49–5 times). The OP extract successfully scavenged 87% of DPPH with a relative increase of about 16 and 32% over LP and MP, respectively. Additionally, it inhibits 77–90% of microbial growth at 5–8 μg/mL while killing them in the 9–14 μg/mL range. Furthermore, OP extract successfully reduced 77% of human breast carcinoma. Each of pomace powder sample (OP, MP, LP) was added to yogurt at three levels; 1, 3, and 5%, while the physiochemical, sensorial, and microbial changes were monitored during 21 days of cold storage. OP yogurt had the highest pH and lowest acidity, while LP yogurt recorded the reverse. High fat and total soluble solids (TSS) content are observed in OP yogurt because of the high fiber content in OP. The pH values of all yogurt samples decreased, while acidity, fat, and TSS increased at the end of the storage period. The OP yogurts 1 and 3% scored higher in color, flavor, and structure than other samples. By measuring the microbial load of yogurt samples, the OP (1 and 3%) contributes to the growth of probiotics (Lactobacillus spp) in yogurt samples and reduces harmful microbes. Using citrus pomace as a source of polyphenols and fiber in functional foods is recommended to enhance their physiochemical and sensory quality.

Air pollution is the atmospheric state in which specific elements concentrations has adverse impacts on human health as well as the environment including global warming, transportation, acidic rain, and ozone layer depletion. Currently, population around the world lives in urban areas coupled with population size in addition to the increasing levels of vehicles which led to deterioration in air quality. Additionally, the clean air is a critical and vital element for environment and health of any region. Therefore, the foci of this research are to detect the air pollution parameters and investigate the relationship with the meteorological parameters including temperature, and wind speed along Al-Shamal train pathway in Al-Qurayyat City in Saudi Arabia using satellite data. This study involved various field visits to distribute questionnaires to residents to gain information about their health status and get their opinions about air quality. The questionnaires analysis manifested poor air quality conditions and thermal pollution due to the rapid urbanization accompanied with the development of the industrial facilities. The main aim of the study was to shed light on using satellite data to detect the concentration of the primary air pollutants including NO2, SO2, and CO to constitute a pre-defensive step to control and improve the air quality in Saudi Arabia. The present study examined using Sentinel-5 Precursor, Moderate Resolution Imaging Spectroradiometer (MODIS), and The Modern-Era Retrospective analysis for Research and Applications, Version 2 (MERRA-2) satellite data to investigate and detect the air pollution parameters in the study area rather than using the traditional measurements from ground stations. The analysis of satellite images revealed an increase in the concentration of air pollution parameters compared to the standard limits of the WHO which can cause crucial negative impacts on the environment as well as human health. Pearson correlation coefficient was used to analyze the relation between various air pollutants with temperature and wind speed with values ranging between 0.0004 and 0.6604. The statistical analysis of air pollutants showed strong linear correlation that may by attributed to the same emission origin. CO and NO2 on roads have a strong correlation and are emitted by vehicles(R (Albugami et al., 2018) = 0.66). NO2 and CO do not seem to have strong correlation with either SO2 or AOD and their correlation coefficients are weaker, that indicating different emissions sources. The AOD in the study area originated mainly from the windblown dust, construction work, and suspension of particles and therefore do not emitted from traffic.

Abstract Background Primary healthcare centers (PHC) ensure that patients receive comprehensive care from promotion and prevention to treatment, rehabilitation, and palliative care in a familiar environment. It is designed to provide first-contact, continuous, comprehensive, and coordinated patient care that will help achieve equity in the specialty healthcare system. The healthcare in Saudi Arabia is undergoing transformation to Accountable Care Organizations (ACO) model. In order for the Kingdom of Saudi Arabia (KSA) to achieve its transformational goals in healthcare, the improvement of PHCs’ quality and utilization is crucial. An integral part of this service is the laboratory services. Methods This paper presents a pilot model for the laboratory services of PHC's in urban cities. The method was based on the FOCUS-PDCA quality improvement method focusing on the pre-analytical phase of the laboratory testing as well as the Saudi Central Board for Accreditation of Healthcare Institutes (CBAHI) gap analysis and readiness within the ten piloted primary healthcare centers. Results The Gap analysis, revealed in-consistency in the practice, lead to lower the quality of the service, which was seen in the low performance of the chosen key performance indicators (KPI's) (high rejection rates, lower turn-around times (TAT) for test results) and also in the competency of the staff. Following executing the interventions, and by using some of the ACO Laboratory strategies; the KPI rates were improved, and our results exceeded the targets that we have set to reach during the first year. Also introducing the electronic connectivity improved the TAT KPI and made many of the processes leaner. Conclusions Our results revealed that the centralization of PHC's laboratory service to an accredited reference laboratory and implementing the national accreditation standards improved the testing process and lowered the cost, for the mass majority of the routine laboratory testing. Moreover, the model shed the light on how crucial the pre-analytical phase for laboratory quality improvement process, its effect on cost reduction, and the importance of staff competency and utilization.

Abstract The current study focused on the histogenesis of the esophagus in quail embryos. Formation of the gut tube occurred on the 4th day of incubation. Development of the muscular layers occurred in a sequential manner; the inner circular layer on the 7th day, the outer longitudinal layer on the 8th day and the muscularis mucosae on the 9th day. Glandular development began on the 13th day of incubation. The epithelium was pseudostratified columnar that consisted of mucous cells, dendritic cells, and keratinocyte precursors. Epithelial stratification occurred on the 15th day of incubation. We used Mallory trichrome, Weigert-Van Gieson, and Gomori silver stains to visualize fibrous components. Scanned samples showed formation of endoderm and mesoderm on the 5th day of incubation. A layer of myoblasts developed on the 8th day of incubation. Formation of mucosal folds, which contained glandular openings, occurred on the 14th to 17th days of incubation. On the 5th to 8th days of incubation, CD34 and vascular endothelial growth factor (VEGF) positive-mesodermal cells, and telocytes (TCs) were detected. On the 15th day of incubation, CD34 and VEGF positive-telocytes, and fibroblasts, were identified. The current study described the correlations between functional morphology and evolutionary biology.

Abstract The current study investigated role of telocytes (TCs) in angiogenesis during embryonic development of quail using immunohistochemistry (IHC), transmission electron microscopy (TEM), and scanning electron microscopy (SEM). The angiogenic apparatus consisted of TCs, endothelial cells, and macrophages. TCs were identified morphologically by their telopodes and podoms using TEM and SEM and immunohistochemically using CD34, and vascular endothelial growth factor (VEGF). TCs also expressed CD68. TCs formed a three-dimensional network and established direct contact with blood vessels, sprouting endothelial cells, and active macrophages, while exerting their effect through paracrine signaling. VEGF was also expressed by endothelial cells and macrophages. Matrix metalloproteinase–9 (MMP-9) was expressed by TCs, endothelial cells, and macrophages. In conclusion, the expression of VEGF by TCs, endothelial cells, and macrophages is required for the proliferation and migration of endothelial cells and vascular growth. The expression of MMP-9 by TCs, endothelial cells, and macrophages is essential for the degradation of extracellular matrix (ECM) components during neoangiogenesis. Macrophages may facilitate phagocytosis and elimination of the degraded ECM components.

Abstract The current study investigated telocytes (TCs) in the intestinal bulb of Grass carp using light microscopy (LM), Transmission electron microscopy (TEM), scanning electron microscopy, and immunohistochemistry (IHC). By LM, TCs were distinguished by the typical morphological features that had a cell body and telopodes using HE, toluidine blue, methylene blue, Marsland silver stain, Grimelius’s silver nitrate, Giemsa, PAS, combined AB pH2,5/PAS, Crossmon’s and Mallory triple trichrome, Van Gieson stains, Verhoeff’s stain, Sudan black, osmic acid, performic acid with methylene blue and bromophenol blue. TCs were identified under the epithelium as an individual cell or formed a TCs sheath. They detected in the lamina propria, between muscle fibers, around the myenteric plexus and fibrous tissue. TCs acquired immunological features of endocrine cells that exhibited high affinity for silver stain, performic acid with methylene blue, Marsland stain, and immunohistochemical staining using chromogranin A. Sub epithelial TCs were closely related to the endocrine cells. TCs and their secretory activities were recognized using acridine orange. TCs were identified by IHC using CD34, CD117, S100-protein, desmin. TCs formed a3D network that established contact with macrophage, mast cells, dendritic cells, lymphocytes, smooth muscle fibers, fibroblast, Schwann cells and nerve fibers. In conclusion, the localization of TCs in relation to different types of immune cells indicated their potential role in the maintenance of intestinal immunity.

Traditional Arab culture hides male sexual abuse and considers it a shame. Instead of treating male victims of sexual assault, they are often stigmatized as homosexual individuals. Moreover, guardians of sexually abused boys are also negatively affected. They have to choose between reporting abuse to a legal authority with the risk of labeling their child or keeping a close eye on them to prevent further abuse. However, reporting abuse to healthcare authorities does take place. Disclosing sexual abuse in the privacy of a psychiatric clinic is often the only way for victims to be successfully treated for the psychological effects of such abuse.

Abstract Background The accurate non-invasive diagnosis of spontaneous bacterial peritonitis (SBP) in patients with decompensated liver cirrhosis has not been achieved yet. The aim of the study was to obtain an unmistakable diagnosis of SBP using a new simple serum bioscore, made by combined measurement of procalcitonin (PCT), erythrocyte sedimentation rate (ESR), and C-reactive protein (CRP), which we called the PEC index. This cross-sectional analytic study comprised 178 cirrhotic patients with ascites (60 patients with SBP and 118 patients with sterile ascites), after excluding non-SBP infection, during the period from March 2019 until September 2019. In all participants, serum levels of PCT, ESR, and CRP were measured, and PEC index was calculated [PEC index = PCT × (ESR + CRP)]. Results Patients with SBP (n = 60) had significantly higher serum PEC index than those with sterile ascites (n = 118) (41.0/31.2–93.0 vs. 9.9/5.9–15.0, P < 0.001). PEC index distinguished culture positive cases significantly (P < 0.001). Using receiver operating characteristic (ROC) statistics, the sensitivity and specificity of PCT, at a cutoff value of 0.590 ng/mL, for SBP diagnosis, were 81.67% and 93.33%, respectively (area under the curve [AUC] = 0.879; 95% confidence interval [CI] 0.809–0.948). The sensitivity and specificity of ESR, at a cutoff value of 27.0 mm/hour, were 73.33% and 61.67%, respectively (AUC = 0.679; 95% CI 0.581–0.776). The sensitivity and specificity of CRP, at a cutoff value of 21.0 mg/L, were 93.33% and 51.67%, respectively (AUC = 0.736; 95% CI 0.639–0.833). While, the sensitivity and specificity of PEC index, at a cutoff value of 20, were highest (98.33% and 96.67%, respectively, AUC = 0.977; 95% CI 0.940–0.996). Conclusion Serum PEC index makes an accurate noninvasive diagnosis of SBP, after excluding other infections.

Chronic liver diseases including hepatocellular carcinoma (HCC) create a state of chronic inflammation that affects the brain via the liver–brain axis leading to an alteration of neurotransmission and cognition. However, little is known about the effects of HCC on the hippocampus, the key brain region for learning and memory. Moreover, radiotherapy used to treat HCC has severe side effects that impair patients’ life quality. Thus, designing optimal strategies, such as chronotherapy, to enhance the efficacy and reduce the side effects of HCC treatment is critically important. We addressed the effects of HCC and the timed administration of radiotherapy in mice on the expression of pro-inflammatory cytokines, clock genes, markers for glial activation, oxidative stress, neuronal activity and proliferation in the hippocampal neurogenic niche. Our data showed that HCC induced the upregulation of genes encoding for pro-inflammatory cytokines, altered clock gene expressions and reduced proliferation in the hippocampus. Radiotherapy, in particular when applied during the light/inactive phase enhanced all these effects in addition to glial activation, increased oxidative stress, decreased neuronal activity and increased levels of phospho(p)-ERK. Our results suggested an interaction of the circadian molecular clockwork and the brain’s innate immune system as key players in liver–brain crosstalk in HCC and that radiotherapy when applied during the light/inactive phase induced the most profound alterations in the hippocampus.
