Abstract Marine protists are globally distributed and sensitive to environmental conditions, which makes them a focal group when studying the effects of climate change on biodiversity and ocean health. However, they are a highly diverse group with varying evolutionary histories and morphologies and widely variable preservation potential in the fossil record. Thus, their past diversity and composition are poorly known. Paleogenetics, which relies, among other approaches, on DNA metabarcoding of sedimentary ancient DNA (sedaDNA), provides a promising avenue to explore the past history and responses of marine protists to global change. Choosing the right marker for sedaDNA studies is critical, striking a balance between marker length and taxonomic resolution. While marker guides exist for modern environmental DNA surveys, a thorough assessment of existing short markers for sedaDNA studies targeting protists is lacking. In this study, we report on a comparison of in silico PCR for eight short 18S rDNA markers, including one from the Tara Oceans initiative and a longer marker commonly used in modern marine eDNA studies. We analyze their taxonomic coverage and resolution, taxonomic overlap and uniqueness between markers, co‐amplification of non‐protist taxa, and amplicon size differences across taxonomic groups. Additionally, we provide a detailed analysis of diatoms, dinoflagellates, haptophytes, and chlorophytes. Our study is aimed at supporting project‐specific marker choices for characterizing protist composition and diversity. While we focus on marine protists, our results are applicable to other aquatic and terrestrial environments.

Abstract Sea ice is a critical component of the Earth’s Climate System and a unique habitat. Sea-ice changes prior to the satellite era are poorly documented, and proxy methods are needed to constrain its past variability. Here, we demonstrate the potential of sedimentary DNA from Polarella glacialis, a sea-ice microalga, for tracing past sea-ice conditions. We quantified P. glacialis DNA (targeting the nuclear ribosomal ITS1 region) in Arctic marine and fjord surface sediments and a sediment core from northern Baffin Bay spanning 12,000 years. Sea ice and sediment trap samples confirmed that cysts of P. glacialis are common in first-year sea ice and sinking particulate matter following sea-ice melt. Its detection is more efficient with our molecular approach than standard micropaleontological methods. Given that the species inhabits coastal and marine environments in the Arctic and Antarctic, P. glacialis DNA has the potential to become a useful tool for circum-polar sea-ice reconstructions.

Cells sense extracellular matrix‐induced biophysical signals, which are transduced into intracellular signaling cascades, and trigger a series of cell responses, including adhesion, migration, and lineage commitment. Traditionally, in in vitro context, monofactorial approaches are employed to control cell fate, despite the fact that in vivo cells are exposed simultaneously to a diverse range of signals. Herein, an overview of key mechanotransduction pathways is first provided. Conventional single‐factor and contemporary multifactorial methodologies, based on substrate rigidity and surface topography, are then reviewed to recapitulate in vitro the in vivo niche, in an attempt to elucidate the underlying mechanisms involved in human mesenchymal stromal cell‐material interactions.

Tissue engineering strategies for tendon repair and regeneration rely heavily on the use of tendon derived cells. However, these cells frequently undergo phenotypic drift in vitro, which compromises their therapeutic potential. In order to maintain the phenotype of tendon derived cells in vitro, microenvironmental cues (biophysical, biochemical and/or biological in origin) have been used to better imitate the complex tendon microenvironment. Herein, the influence of planar and grooved (groove width of ∼1.0 µm, groove depth of ∼1.4 µm and distance between groves of ∼1.7 µm) poly(glycolide-co-ε-caprolactone) substrates with elastic modulus of 7 kPa and poly(lactide-co-trimethylene carbonate) substrates with elastic modulus of 12 kPa on human tendon derived cell response was assessed, using planar tissue culture plastic substrates of 3 GPa elastic modulus as control, in both basal and tenogenic media. At day 17, the grooved 12 kPa poly(lactide-co-trimethylene carbonate) substrate induced the highest deposition and alignment of collagen type I in tenogenic media. At day 17, the grooved 12 kPa poly(lactide-co-trimethylene carbonate) substrate and the tissue culture plastic induced the highest deposition and the tissue culture plastic and the planar 7 kPa poly(glycolide-co-ε-caprolactone) induced the lowest alignment of tenascin C in tenogenic media. Also at day 17 in tenogenic media, the grooved 12 kPa poly(lactide-co-trimethylene carbonate) substrate induced the upregulation of most tenogenic genes (COL1A1, COL3A1, MKX, TNMD). Our data further support the notion of multifactorial tissue engineering for effective control over cell fate in vitro setting.

Abstract Background The past years have witnessed dramatic changes in the population admitted to the intensive care unit (ICU). Older and sicker patients are now commonly treated in this setting due to the newly available sophisticated life support. However, the short- and long-term benefit of this strategy is scarcely studied. Methods The Critically Ill patients’ mortality by age: Long-Term follow-up (CIMbA-LT) was a multicentric, nationwide, retrospective, observational study addressing short- and long-term prognosis of patients admitted to Portuguese multipurpose ICUs, during 4 years, according to their age and disease severity. Patients were followed for two years after ICU admission. The standardized hospital mortality ratio (SMR) was calculated according to the Simplified Acute Physiology Score (SAPS) II and the follow-up risk, for patients discharged alive from the hospital, according to official demographic national data for age and gender. Survival curves were plotted according to age group. Results We included 37.118 patients, including 15.8% over 80 years old. The mean SAPS II score was 42.8 ± 19.4. The ICU all-cause mortality was 16.1% and 76% of all patients survive until hospital discharge. The SAPS II score overestimated hospital mortality [SMR at hospital discharge 0.7; 95% confidence interval (CI) 0.63–0.76] but accurately predicted one-year all-cause mortality [1-year SMR 1.01; (95% CI 0.98–1.08)]. Survival curves showed a peak in mortality, during the first 30 days, followed by a much slower survival decline thereafter. Older patients had higher short- and long-term mortality and their hospital SMR was also slightly higher (0.76 vs. 0.69). Patients discharged alive from the hospital had a 1-year relative mortality risk of 6.3; [95% CI 5.8–6.7]. This increased risk was higher for younger patients [21.1; (95% CI 15.1–39.6) vs. 2.4; (95% CI 2.2–2.7) for older patients]. Conclusions Critically ill patients’ mortality peaked in the first 30 days after ICU admission. Older critically ill patients had higher all-cause mortality, including a higher hospital SMR. A long-term increased relative mortality risk was noted in patients discharged alive from the hospital, but this was more noticeable in younger patients.

ABSTRACT Concerns about colistin-resistant bacteria in animal food-environmental-human ecosystems prompted the poultry sector to implement colistin restrictions and explore alternative trace metals/copper feed supplementation. The impact of these strategies on the selection and persistence of colistin-resistant Klebsiella pneumoniae in the whole poultry production chain needs clarification. We assessed colistin-resistant and copper-tolerant K. pneumoniae occurrence in chickens raised with inorganic and organic copper formulas from 1-day-old chicks to meat (7 farms from 2019 to 2020), after long-term colistin withdrawal (>2 years). Clonal diversity and K. pneumoniae adaptive features were characterized by cultural, molecular, and whole-genome-sequencing (WGS) approaches. Most chicken flocks (75%) carried K. pneumoniae at early and preslaughter stages, with a significant decrease (P 50%) of colistin-resistant/mcr-negative K. pneumoniae were observed among fecal samples, independently of feed. Most samples carried multidrug-resistant (90%) and copper-tolerant (81%; silA and pcoD positive and with a MICCuSO4 of ≥16 mM) isolates. WGS revealed accumulation of colistin resistance-associated mutations and F type multireplicon plasmids carrying antibiotic resistance and metal/copper tolerance genes. The K. pneumoniae population was polyclonal, with various lineages dispersed throughout poultry production. ST15-KL19, ST15-KL146, and ST392-KL27 and IncF plasmids were similar to those from global human clinical isolates, suggesting chicken production as a reservoir/source of clinically relevant K. pneumoniae lineages and genes with potential risk to humans through food and/or environmental exposure. Despite the limited mcr spread due to the long-term colistin ban, this action was ineffective in controlling colistin-resistant/mcr-negative K. pneumoniae, regardless of feed. This study provides crucial insights into the persistence of clinically relevant K. pneumoniae in the poultry production chain and highlights the need for continued surveillance and proactive food safety actions within a One Health perspective. IMPORTANCE The spread of bacteria resistant to last-resort antibiotics such as colistin throughout the food chain is a serious concern for public health. The poultry sector has responded by restricting colistin use and exploring alternative trace metals/copper feed supplements. However, it is unclear how and to which extent these changes impact the selection and persistence of clinically relevant Klebsiella pneumoniae throughout the poultry chain. We found a high occurrence of copper-tolerant and colistin-resistant/mcr-negative K. pneumoniae in chicken flocks, regardless of inorganic and organic copper formulas use and a long-term colistin ban. Despite the high K. pneumoniae isolate diversity, the occurrence of identical lineages and plasmids across samples and/or clinical isolates suggests poultry as a potential source of human K. pneumoniae exposure. This study highlights the need for continued surveillance and proactive farm-to-fork actions to mitigate the risks to public health, relevant for stakeholders involved in the food industry and policymakers tasked with regulating food safety.

We present the most recent Intergovernmental Panel on Climate Change Sixth Assessment Report (AR6) sea-level projections for four Danish cities (Aarhus, Copenhagen, Esbjerg and Hirtshals) under the Shared Socioeconomic Pathway (SSP) family of climate scenarios. These sea-level changes projected over the next century are up to an order of magnitude larger than those observed over the previous century. At these cities, year 2150 sea-level changes of between 29 and 55 cm are projected under the very low emissions scenario (SSP1-1.9), whilst changes of between 99 and 123 cm are projected under the very high emissions scenario (SSP5-8.5). These differences highlight the potentially significant impact of remaining opportunities for climate change mitigation. Due to this increase in mean sea level, the mean recurrence time between historically extreme events is expected to decrease. Under the very high emissions scenario, the historical 100-year storm flood event will become a 1- to 5-year event at most Danish harbours by 2100. There is considerable uncertainty associated with these sea-level projections, primarily driven by uncertainty in the future evolution of the Antarctic ice sheet and future sterodynamic changes in ocean volume. The AR6 characterises collapse of the West Antarctic ice sheet as a low-probability but high-impact event that could cause several metres of sea-level rise around Denmark by 2150. In climate adaptation policy, the scientific landscape is shifting fast. There has been a tremendous proliferation of diverse sea-level projections in recent years, with the most relevant planning target for Denmark increasing c. 50 cm in the past two decades. Translating sea-level rise projections into planning targets requires value judgments about acceptable sea-level risk that depend on local geography, planning timeline and climate pathway. This highlights the need for an overarching national sea-level adaptation plan to ensure municipal plans conform to risk and action standards.

Abstract The combined effect of surface topography and substrate rigidity in stem cell cultures is still under‐investigated, especially when biodegradable polymers are used. Herein, we assessed human bone marrow stem cell response on aliphatic polyester substrates as a function of anisotropic grooved topography and rigidity (7 and 12 kPa). Planar tissue culture plastic (TCP, 3 GPa) and aliphatic polyester substrates were used as controls. Cell morphology analysis revealed that grooved substrates caused nuclei orientation/alignment in the direction of the grooves. After 21 days in osteogenic and chondrogenic media, the 3 GPa TCP and the grooved 12 kPa substrate induced significantly higher calcium deposition and alkaline phosphatase (ALP) activity and glycosaminoglycan (GAG) deposition, respectively, than the other groups. After 14 days in tenogenic media, the 3 GPa TCP upregulated four and downregulated four genes; the planar 7 kPa substrate upregulated seven genes and downregulated one gene; and the grooved 12 kPa substrate upregulated seven genes and downregulated one gene. After 21 days in adipogenic media, the softest (7 kPa) substrates induced significantly higher oil droplet deposition than the other substrates and the grooved substrate induced significantly higher droplet deposition than the planar. Our data pave the way for more rational design of bioinspired constructs.

Enthesis repair remains a challenging clinical indication. Herein, a three-layer scaffold composed of a tendon-like layer of collagen type I, a fibrocartilage-like layer of collagen type II and a bone-like layer of collagen type I and hydroxyapatite, was designed to recapitulate the matrix composition of the enthesis. To aid tenogenic and fibrochondrogenic differentiation, bioactive molecules were loaded in the tendon-like layer or the fibrocartilage-like layer and their effect was assessed in in vitro setting using human bone marrow derived mesenchymal stromal cells and in an ex vivo model. Seeded human bone marrow mesenchymal stromal cells infiltrated and homogeneously spread throughout the scaffold. As a response to the composition of the scaffold, cells differentiated in a localised manner towards the osteogenic lineage and, in combination with differentiation medium, towards the fibrocartilage lineage. Whilst functionalisation of the tendon-like layer did not improve tenogenic cell commitment within the time frame of this work, relevant fibrochondrogenic markers were detected in the fibrocartilage-like layer when scaffolds were functionalised with bone morphogenetic protein 2 or non-functionalised at all, in vitro and ex vivo, respectively. Altogether, our data advocate the use of compartmentalised scaffolds for the repair and regeneration of interfacial tissues, such as enthesis.

Neighbourhood and health research often relies on personal location data (e.g., home address, daily itineraries), despite the risks of geoprivacy breaches. Thus, geoprivacy is an important emerging topic, contemplated in international regulations such as the General Data Protection Regulation. In this mini-review, we briefly assess the potential risks associated with the usage of personal location data and provide geoprivacy-preserving recommendations to be considered in epidemiological research. Risks include inference of personal information that the individual does not wish to disclose, reverse-identification and security breaches. Various measures should be implemented at different stages of a project (pre-data collection, data processing, data analysis/publication and data sharing) such as informed consent, pseudo-anonymization and geographical methods.

The North Water polynya is a unique but vulnerable ecosystem, home to Indigenous people and Arctic keystone species. New palaeoecological records from Greenland suggest human abandonment c. 2200–1200 cal yrs BP occurred during climate-forced polynya instability, foreshadowing future ecosystem declines.

Abstract Baffin Bay hosts the largest and most productive of the Arctic polynyas: the North Water (NOW). Despite its significance and active role in water mass formation, the history of the NOW beyond the observational era remains poorly known. We reconcile the previously unassessed relationship between long-term NOW dynamics and ocean conditions by applying a multiproxy approach to two marine sediment cores from the region that, together, span the Holocene. Declining influence of Atlantic Water in the NOW is coeval with regional records that indicate the inception of a strong and recurrent polynya from ~ 4400 yrs BP, in line with Neoglacial cooling. During warmer Holocene intervals such as the Roman Warm Period, a weaker NOW is evident, and its reduced capacity to influence bottom ocean conditions facilitated northward penetration of Atlantic Water. Future warming in the Arctic may have negative consequences for this vital biological oasis, with the potential knock-on effect of warm water penetration further north and intensified melt of the marine-terminating glaciers that flank the coast of northwest Greenland.

Introdução: A Paralisia Cerebral (PC) é uma lesão do Sistema Nervoso Central com forte impacto na funcionalidade. As alterações neuromusculoesqueléticas, sensoriopercetivas e cognitivas originam frequentemente necessidades complexas de comunicação, cuja intervenção multidisciplinar passa amiudadamente pela implementação de Sistemas Aumentativos e Alternativos de Comunicação (SAAC). O terapeuta ocupacional pode colaborar para esta intervenção, sobretudo quando são envolvidas Tecnologias de Apoio (TA). Objetivo: Descrever o processo da Terapia Ocupacional (TO) na implementação de SAAC, em crianças com PC, verificando a influência nas atividades, cuja participação está restringida por défices na comunicação. Metodologia: Scoping Review desenvolvida com base no guia orientador Preferred Reporting Items for Systematic Reviews and Meta-Analyses extension for Scoping Reviews (PRISMA-ScR) e de acordo com o protocolo do Joanna Briggs Institute (JBI), encetada com pesquisa na b-on. Foi realizada uma análise categorial temática dos artigos com recurso ao software webQDA. Resultados: Foram selecionados 10 artigos, dos 353 estudos inicialmente encontrados, de acordo com os critérios de inclusão, demonstrando a contribuição do processo da TO, a importância de uma equipa multidisciplinar, bem como, a identificação de lacunas, que sugerem investimento em investigações futuras. Conclusão: A TO tem um papel pertinente na implementação de SAAC na: avaliação, com identificação das capacidades e necessidades a diferentes níveis, incluindo o posicionamento, tipo de acesso mais adequado e contextos; intervenção, através do posicionamento, da adaptação e treino de utilização de SAAC, de formação, consultadoria e advocacy das partes interessadas. A implementação de SAAC é uma área que ainda carece de investimento em investigação e formação.

Polysaccharides constitute an important carbon pool in marine systems, but much is still unknown about the fate and degradation of these compounds. They are derived partly from production in situ, and in coastal areas, they are partly terrestrially derived, originating from freshwater runoff from land. The aim of this study was to test the applicability of high-throughput polysaccharide profiling for plant and algal cell-wall compounds in dated sediment cores from a coastal marine environment, to examine the preservation of cell-wall polysaccharides and explore their potential as proxies for temporal environmental changes. Preserved compounds and remains of organisms are routinely used as paleoenvironmental proxies as the amount and composition of different compounds that can provide insight into past environmental conditions, and novel means for reporting environmental changes are highly sought.

A comunicação é um elemento essencial nas relações entre as pessoas e as organizações, quer sejam privadas ou públicas, desempenhando um papel de facilitador para a democratização do acesso à informação. É comum referir-se à acessibilidade como “aquilo que pode ser alcançado”. Contudo, o presente artigo visa a reflexão sobre a comunicação acessível também como “aquilo que é fácil de entender”. Com efeito, para que a comunicação seja eficaz, é essencial que consiga derrubar as barreiras que impeçam as pessoas de perceber e lembrar uma mensagem. Como abordagem metodológica, apoiámo-nos no caso de estudo do programa Edifícios + Sustentáveis, recorrendo à análise qualitativa e quantitativa da acessibilidade do website, redes sociais e materiais digitais de apoio.

Matrix stiffness is a crucial regulator of cell fate in both in vitro and in vivo setting. Although studies with non-degradable polymers have contributed to our understanding of the influence of rigidity on cell response, very little work has been conducted with biodegradable polymers that constitute the building blocks of implantable devices. Herein, we investigated human bone marrow stem cell response as a function of rigidity (7 kPa, 10 kPa, 12 kPa, 22 kPa, 15 MPa elastic modulus values) that induced by five different aliphatic polyesters (tissue culture plastic with ~ 3 GPa rigidity was used as control). Cell morphology analysis revealed that stiff substrates stimulated a large cell area with defined stress fibres, whilst soft substrates prompted a small cell area without evident stress fibres. Immunocytochemistry analysis made apparent that YAP was accumulated at the nuclei when the cells were seeded on stiff substrates and at the cytoskeleton on soft substrates. ABSTRACT Objective: Dornase alfa (rhDNase) reduces the viscosity of purulent sputum in the lungs. The use in patients with cystic fibrosis (CF) is proven. However, the evidence of its applicability to other conditions is limited. This study aims to present the authors’ experience with the use of rhDNase in non-CF patients admitted to the Pediatric Intensive Care Unit (PICU). At the study center, rhDNase was used during flexible bronchoscopies in 24 cases, of which 20 (83%) had atelectasis and seven (29%) were admitted to PICU. Four patients (57%) were on invasive mechanical ventilation (MV). Case description: Two cases of daily rhDNase administration at PICU are presented: patient A was an 8-year-old boy admitted with septic shock and acute respiratory distress syndrome (ARDS). The patient required mechanical ventilation with aggressive settings and experienced several clinical complications. On D50, he started rhDNase treatment with an improvement in FiO2, PaCO2 and PaO2/FiO2 ratio according to radiologic findings. He was extubated on D23 of treatment. Patient B was a 17-month-old girl admitted with a convulsive status epilepticus who experienced respiratory complications (infectious and barotrauma) with ARDS, requiring aggressive ventilation. She initiated rhDNase treatment on D60. During the treatment an improvement in FiO2, PaO2/FiO2 ratio and a tendency of PaCO2 decrease were found. She had radiological improvement. No complications were described. Comments: RhDNase may be a helpful and safe tool to use in PICU prolonged intubated patients with ventilator-induced lung injury. Further studies are needed to assess and propose valid indications.

Abstract Background The existing knowledge on the interplay between reproductive and sexual health, migration and acculturation is recent and inconsistent, particularly on the sociocultural motives and constraints regarding fertility. Therefore, sexual and reproductive health (SRH) surveys are needed to provide accurate and comparable indicators to identify and address SRH inequalities, with specific focus on under researched aspects, such as the interrelation between migration and gender. FEMINA (FErtility, MIgratioN and Acculturation) aims to investigate intersectional SRH inequalities among Cape Verdean immigrant and Portuguese native families and how they impact on fertility in Portugal. This study will use a comprehensive approach exploring simultaneously the components of SRH, namely regarding identities, perceptions and practices of both women and men among lay people and relevant experts and stakeholders. The project has three main goals: 1) to identify social determinants of SRH among Cape Verdean immigrant and Portuguese native men and women of reproductive age; 2) to gain understanding of the diversity of the sexual and reproductive experiences and expectations of Cape Verdean immigrant and Portuguese native men and women of reproductive age, considering the singularities of their migratory, social and family dynamics; and 3) to produce recommendations for policy makers, employers and service providers on how to better address the SRH needs of Portuguese-born and immigrant populations. Methods The study will address these goals using a mixed methods approach, including: a cross-sectional telephone survey with a probabilistic sample of 600 Cape Verdean immigrant and 600 Portuguese native women and men (women aged 18 to 49 and men aged 18 to 54), residents of the Greater Lisbon Area; a qualitative research through in-depth interviews with a subsample of 30 Cape Verdean immigrants and 30 Portuguese native men and women; and a Delphi technique for finding consensus on good practices in SRH for the entire population with a special emphasis on immigrants, namely extra-EU migrants. Discussion Data will be used to produce a comprehensive set of indicators to monitor SRH in Portugal, to foster a greater understanding of its specificities and challenges to policy and decision makers, and to provide targeted recommendations to promote inclusive and migrant sensitive SRH services.

Human–wildlife coexistence may increase the potential risk of direct transmission of emergent or re-emergent zoonotic pathogens to humans. Intending to assess the occurrence of three important foodborne pathogens in wild animals of two wildlife conservation centers in Portugal, we investigated 132 fecal samples for the presence of Escherichia coli (Shiga toxin-producing E. coli (STEC) and non-STEC), Salmonella spp. and Campylobacter spp. A genotypic search for genes having virulence and antimicrobial resistance (AMR) was performed by means of PCR and Whole-Genome Sequencing (WGS) and phenotypic (serotyping and AMR profiles) characterization. Overall, 62 samples tested positive for at least one of these species: 27.3% for STEC, 11.4% for non-STEC, 3.0% for Salmonella spp. and 6.8% for Campylobacter spp. AMR was detected in four E. coli isolates and the only Campylobacter coli isolated in this study. WGS analysis revealed that 57.7% (30/52) of pathogenic E. coli integrated genetic clusters of highly closely related isolates (often involving different animal species), supporting the circulation and transmission of different pathogenic E. coli strains in the studied areas. These results support the idea that the health of humans, animals and ecosystems are interconnected, reinforcing the importance of a One Health approach to better monitor and control public health threats.

The increasing demand for more aesthetic/comfortable orthodontic alternatives fostered the utilization of clear aligners in recent years. However, the efficacy of clear aligners for treating complex malocclusions is often treated with scepticism. This case series aims to evaluate the predictability of the Invisalign® First system in moderate and severe cases requiring interceptive orthodontic treatments in mixed dentition. A total of 23 patients with 102 interceptive orthodontic malocclusion traits were selected for orthodontic treatment with Invisalign® First and were examined over 18 months (Phase 1). Clinical assessments included ClinCheck® predictions, cephalometric measurements, and measuring tools commonly used to quantify tooth movement. Measurements taken at the beginning and end of the treatment were compared. The complexity degree of each case was established based on the set of problems presented by each patient. All treatment objectives were achieved within 18 months, except for two Class II cases, with 69% of them solved with the first set of aligners. Additional aligners were used in the remaining cases. Even though these 23 cases suggest that the Invisalign® First (Phase 1) may be effective in most interceptive problems, controlled randomized clinical trials are required to evaluate movement predictability and how this relates to the problem complexity and additional aligners required.

Four marine sediment cores from two sites in the Independence Fjord system near the Wandel Sea in eastern North Greenland were analyzed for their dinoflagellate cyst (dinocyst) and foraminiferal fossil content to gain insight into the water mass properties and evolution of the outer fjord system over the Holocene and Last Glacial Period. While regarded as a climate-sensitive region, the climatic history of the area remains largely unknown and has been documented through the study of two composite marine sediment cores only once before. The results presented here reveal that Atlantic waters entered the Independence Fjord episodically during the studied interval. High concentrations of dinocysts and foraminifers in the upper few centimeters of the cores are in line with oceanographic measurements clearly illustrating that Atlantic-sourced waters make up the lower part of the water column in the area in modern times. Radiocarbon dating of foraminiferal tests and increasing microfossil concentrations and diversity toward the top of the cores suggest that this inflow has been occurring for at least 2,000 years and intensified toward recent times. The core sections below the upper few centimeters are devoid of (Quaternary) dinocysts and calcareous foraminifera with the exception of the lowermost segments of the longer cores. While low foraminiferal test quantities in these lowermost core sections prevent precise age determination, their radiocarbon ages reveal that they were deposited prior to 30,000 years ago, indicating the existence of a pathway for the occasional intrusion of Atlantic-sourced waters into, and thus relatively small local ice caps around, the fjord system prior to the Last Glacial Maximum. The previously documented early Holocene inflow of Atlantic-sourced waters was not detected in our records, likely suggesting a strong topographical and deglacial control on the routing of these water masses during the early Holocene.

Although it is important to communicate scientific knowledge, it seems that this assumption is even more evident when it comes to environment-related themes, which have gained more relevance in the public sphere in the last decades. This article evaluates the spectacularization techniques of technology and science displayed as if they were the only solution to the environmental crisis. Firstly, from a literature perspective, this article shows the evolution of science communication and the relationship between society and science over the last centuries. After that, it presents a critical view on the excessive optimism around technology and scientific advancement, arguing that the possible solutions to the environmental crisis cannot come exclusively through technology substitution. The article thus shows that there is today a clear distinction between science communication and the communication of science as a mass product, which is promoted today by different stakeholders to manipulate public opinion for different reasons. Finally, the article identifies some common elements of this phenomenon that we call technowashing, insofar as it aims at a laundering of responsibilities and harmful impacts of business and political decisions.

Between 1850 and 2006 global mean sea level rose by 24 ± 18 cm. It is projected to rise a further 52 ± 21 cm under the Representative Concentration Pathway (RCP) 4.5 scenario, which approximates the carbon emissions reductions of the ‘Paris Agreement’ climate pathway. It is projected to rise 74 ± 28 cm under the RCP8.5 scenario, which represents a ‘business-as-usual’ climate pathway (Box & Colgan 2017). These rates of recent and future sea-level rise are faster than those reconstructed for previous warm intervals, such as the Medieval Climatic Optimum (c. 1000 to 1400 CE) and the Holocene Thermal Maximum (c. 7000 to 3000 BCE) (Gehrels & Shennan 2015). Moreover, palaeo reconstructions indicate a global sea-level sensitivity of two metres per degree of warming (Levermann et al. 2013). The forces driving global sea-level change are complex. The global sea-level budget includes the transfer of land ice into the ocean, thermal expansion of seawater, changes in land water storage, and changes in ocean basin volume (Church et al. 2013). At the local scale, the evolving planetary gravity due to shifting water and ice masses, shifting oceanic and atmospheric currents and persistent tectonic and glacial isostatic adjustment processes can also be important. Sea-level changes around the globe are therefore far from uniform (Jevrejeva et al. 2016). Here, we highlight the value of combining palaeo reconstructions of sea level, the measured tide gauge record, and projections of future sea level. This allows us to understand local sea-level changes from the recent past in the context of global projections for the near future (0 to 2100 CE). We explore the strong differences in local sea-level histories and future projections at three Danish cities: Skagen and Esbjerg, as they have contrasting glacio-isostatic adjustment histories, and Copenhagen, where we also compare local and global drivers of present-day sea-level rise based on previously published research.

Introduction: Pre-exposure prophylaxis is defined as the use of antiretroviral drugs to prevent HIV acquisition in uninfected individuals. Recognizing the increasing use of informal pre-exposure prophylaxis in Portugal, CheckpointLX, a community clinic targeted to men who have sex with men in Lisbon, Portugal, began offering counselling and follow-up services prior to formal introduction. This study aims to characterize pre-exposure prophylaxis users attending CheckpointLX before formal pre-exposure prophylaxis introduction in Portugal, and those who were referred to pre-exposure prophylaxis in the National Health Service following formal approval of pre-exposure prophylaxis. Material and Methods: Data was collected by peer counsellors between May 2015 and September 2018 and inserted in a database. Medical care followed the European AIDS Clinical Society recommendations for pre-exposure prophylaxis eligibility, initiation and follow-up. For formal pre-exposure prophylaxis, the General-Directorate for Health’s Pre-exposure Prophylaxis guidelines checklist was used. Results: Until the end of May 2018, CheckpointLX had a total of 90 appointments for wild pre-exposure prophylaxis, of which 64 (71%) were first time visits. As for the 380 service users referred to the National Health Service, most were Portuguese (n = 318, 84%), and the mean age was 31 (8.9) years old. Condomless sex in the last six months with partners of unknown HIV status was the most common eligibility criteria (n = 59, 83%). Discussion: Pre-exposure prophylaxis delivery should be complemented with effective information on the importance of immunization and education on safer practices of drug administration, in the scope of broader preventive sexual health care. Conclusion: Much remains to be done in Portugal to ensure that pre-exposure prophylaxis is available to those who need it the most. Offering pre-exposure prophylaxis at community clinics could be a first step.

Abstract Mean body size decreases with increasing temperature in a variety of organisms. This size–temperature relationship has generally been tested through space but rarely through time. We analyzed the sedimentary archive of dinoflagellate cysts in a sediment record taken from the West Greenland shelf and show that mean cell size decreased at both intra‐ and interspecific scales in a period of relatively warm temperatures, compared with a period of relatively cold temperatures. We further show that intraspecific changes accounted for more than 70% of the change in community mean size, whereas shifts in species composition only accounted for about 30% of the observed change. Literature values on size ranges and midpoints for individual taxa were in several cases not representative for the measured sizes, although changes in community mean size, calculated from literature values, did capture the direction of change. While the results show that intraspecific variation is necessary to accurately estimate the magnitude of change in protist community mean size, it may be possible to investigate general patterns, that is relative size differences, using interspecific‐level estimates.

Tobacco is responsible for the death of 6 million people every year globally, of whom 700 000 are in Europe. Effective policies for tobacco control exist; however, the status of their implementation varies across the World Health Organization (WHO) European Region. In order to tackle the tobacco epidemic, action has been taken though the implementation of both legally binding and non-legally binding measures. This article aims to present the achievements and challenges of tobacco control in Europe, focussing on the available legally binding instruments such as the WHO Framework Convention on Tobacco Control and the revision of the Tobacco Products Directive at the European Union level. Tobacco still faces heavy lobbying of the tobacco industry, which has systematically contrasted policies to achieve public health objectives. The legal instruments for tobacco control in Europe presented here are not always adequately enforced in all the countries and there is certainly room for improving their implementation. Finally, the need for a strong political commitment towards the end-game of the tobacco epidemic is emphasised.

Poultry products are still an important cause of Salmonella infections worldwide, with an increasingly reported expansion of less-frequent serotypes or atypical strains that are frequently multidrug-resistant. Nevertheless, the ability of Salmonella to survive antimicrobials promoted in the context of antibiotic reducing/replacing and farming rethinking (e.g., organic acids and copper in feed/biocides) has been scarcely explored. We investigated Salmonella occurrence (conventional and molecular assays) among chicken meat at the processing stage (n = 53 batches/29 farms) and characterized their tolerance to diverse stress factors (antibiotics, copper, acid pH, and peracetic acid). Whole-genome sequencing was used to assess adaptive features and to perform comparative analysis. We found a low Salmonella occurrence (4%) and identified S. Enteritidis/ST11 plus atypical non-H2S-producing S. 1,4,[5],12:i:-/ST3478. The ST3478 presented the ability to grow under diverse stresses (antibiotics, copper, and acid-pH). Comparative genomics among ST3478 isolates showed similar antibiotic/metal resistance gene repertoires and identical nonsense phsA thiosulfate reductase mutations (related to H2S-negative phenotype), besides their close phylogenetic relationship by cgMLST and SNPs. This study alerts for the ongoing national and international spread of an emerging monophasic Salmonella Typhimurium clonal lineage with an enlarged ability to survive to antimicrobials/biocides commonly used in poultry production, being unnoticed by conventional Salmonella detection approaches due to an atypical non-H2S-producing phenotype.

Mediterranean wetlands are critical strongholds for biodiversity and the provision of ecosystem functions and services; yet, they are being severely degraded by a number of socio-economic drivers and pressures, including climate change. Moreover, we still lack comprehensive understanding of the extent to which biodiversity loss in Mediterranean wetlands will accelerate change in ecosystem processes. Here, we evaluate how changes in biodiversity can alter the ecosystem of the Camargue (southern France). We collected data on species presence/absence, trends and abundance over a 40-year period by combining observations from the scholarly literature with insights derived from expert knowledge. In total, we gathered more than 1500 estimates of presence/absence, over 1400 estimates of species abundance, and about 1400 estimates of species trends for eight taxonomic groups, i.e. amphibians, reptiles, breeding birds, fish, mammals, dragonflies (odonates), orthopterans and vascular plants. Furthermore, we used information on recently arrived species and invasive species to identify compositional changes across multiple taxa. Complementing targeted literature searches with expert knowledge allowed filling important gaps regarding the status and trends of biodiversity in the Camargue. Species trend data revealed sharp population declines in amphibians, odonates and orthopterans, while birds and plants experienced an average increase in abundance between the 1970s and the 2010s. The general increasing trends of novel and invasive species is suggested as an explanation for the changing abundance of birds and plants. While the observed declines in certain taxa reflect the relative failure of the protection measures established in the Camargue, the increasing exposure to novel and invasive species reveal major changes in the community structure of the different taxonomic groups. This study is the first attempt to assess changes in biodiversity in the Camargue using an expert knowledge approach, and can help manage the uncertainties and complexities associated with rapid social-ecological change in other Mediterranean wetlands.

We present a theoretical description of the coupling between longitudinal optical phonons and collective excitations of a two-dimensional electron gas. By diagonalizing the Hamiltonian of the system, including Coulomb electron−electron and Fröhlich interactions, we observe the formation of multisubband polarons, mixed states partially phonon and partially multisubband plasmon, characterized by a coupling energy which is a significant fraction, up to ∼ 40 % , of the phonon energy. We demonstrate that multisubband plasmons and longitudinal optical phonons are in the ultra-strong coupling regime in several III−V and II−VI material systems.

The biomarker IP25 and fossil diatom assemblages preserved in seafloor sediments are commonly used as proxies for paleo Arctic sea-ice reconstructions, but how their production varies over the seasons and is exported to the sediment remains unclear. We analyzed IP25 concentrations and diatom assemblages from a 5-week consecutive series of sea-ice cores and compared the results with sediment trap and surface sediment samples collected at the same site in the Young Sound fjord, Northeast Greenland. Our aim was to investigate the dynamics of diatom colonization of the spring sea ice and the in situ production of IP25. Additionally, selected diatom taxa observed in the sea-ice samples were isolated from in-ice assemblages and their lipid composition was analyzed via gas chromatography-mass spectrometry. We confirm that Haslea spicula (and not the closely related species H. crucigeroides) is an IP25-producer. All three known IP25-producing taxa (Haslea spicula, H. kjellmanii, and Pleurosigma stuxbergii var. rhomboides) were present in Young Sound sea-ice and the low IP25 concentrations measured in the sea-ice (0.44–0.72 pg mL−1) were consistent with the low abundance of these source species (0.21–9.66 valves mL−1). Total sympagic diatom production also remained very low (21–985 valves mL−1), suggesting that the fjord's sea ice did not provide an optimal physical-chemical environment for diatoms to thrive. Temporal changes in the sympagic diatom community were also observed, with an early presence of the pelagic Thalassiosira hyperborea and subsequent dominance of pennate taxa, including Nitzschia and Navicula species, Fossula arctica and Stauronella arctica. The assemblages observed during and after the seasonal ice melt consisted primarily of Fossula arctica, Fragilariopsis oceanica, Thalassiosira antarctica var. borealis (resting spores), and Chaetoceros spp. (vegetative cells and resting spores). The seafloor sediment assemblages largely reflected the melt and post-melt planktic production and were dominated by the resting spores of the centric Chaetoceros spp. and Thalassiosira antarctica var. borealis, and the pennate Fragilariopsis oceanica, Fossula arctica, and Fragilariopsis reginae-jahniae. This study documents that IP25 is produced in Young Sound, and that the weak fingerprint of sea ice in the sediment appears to be primarily due to the limited sea-ice diatom biomass.

Há um novo elemento entre os media e as instituições políticas, os spin doctors. A crescente influência que estes elementos têm na cobertura mediática das campanhas politicas e a crescente necessidade que os políticos têm em recorrer a especialistas da comunicação política, torna urgente delimitar o conceito e regular deontológica e eticamente a sua ação. Só deste modo se garante o princípio de transparência das sociedades democráticas. O presente texto pretende evidenciar as diferenças de ação em alguns países por parte dos spin doctors e chamar a atenção para a necessidade de regular, especialmente dentro do espaço europeu, esta terceira força.

O presente artigo tem como objetivo analisar em que medida os grupos dos amigos dos museus podem ser um vetor estratégico para os museus.?O espaço museológico encontra-se em profunda transformação e tem hoje novos desafios. Em muitos casos, os museus descobrem nas associações de afiliação cultural a possibilidade de revitalização. Dirigimos a atenção para os grupos dos amigos dos museus (GAM), pois torna-se essencial potenciar uma estrutura que deve ser considerada um vetor estratégico para o museu – um pivô indispensável para os museus conseguirem dar resposta às expectativas e necessidades do público e dos stakeholders. Os grupos de amigos têm hoje novos desafios: às tarefas tradicionais de angariação de novas peças para os acervos, do restauro de obras ou da angariação de fundos e mecenas, junta-se a difícil tarefa de atrair público, que é cada vez mais diversificado. A afiliação cultural pode assim ter um papel fundamental na democratização do acesso à cultura por vários grupos do tecido social.

Marine protist species have been used for several decades as environmental indicators under the assumption that their ecological requirements have remained more or less stable through time. However, a growing body of evidence suggests that marine protists, including several phytoplankton species, are in fact highly diverse and may quickly respond to changes in the environment. Predicting how future climate will impact phytoplankton populations is important, but this task has been challenged by a lack of time-series of ecophysiological parameters at time-scales relevant for climate studies (i.e. at least decadal). Here, we report on ecophysiological variability in a marine dinoflagellate over a 100-year period of well-documented environmental change, by using the sedimentary archive of living cysts from a Scandinavian fjord (Koljö Fjord, Sweden). During the past century, Koljö Fjord has experienced important changes in salinity linked to the North Atlantic Oscillation (NAO). We revived resting cysts of Pentapharsodinium dalei preserved in the fjord sediments and determined growth rates for 18 strains obtained from 3 sediment core layers at salinity 15 and 30, which represent extreme sea-surface conditions during periods of predominantly negative and positive NAO phases, respectively. Upper pH tolerance limits for growth were also tested. In general, P. dalei grew at a higher rate in salinity 30 than 15 for all layers, but there were significant differences among strains. When accounting for inter-strain variability, cyst age had no effect on growth performance or upper pH tolerance limits for this species, indicating a stable growth response over the 100-year period in spite of environmental fluctuations. Our findings give some support for the use of morphospecies in environmental studies, particularly at decadal to century scales. Furthermore, the high intra-specific variability found down to sediment layers dated as ca. 50 years-old indicates that cyst-beds of P. dalei are repositories of ecophysiological diversity.

The osteolytic nature of bone metastasis results from a tumor-driven increased bone resorption. Bone remodeling is orchestrated by the molecular triad RANK-RANKL-OPG. This process is dysregulated in bone metastases, mostly via induction of RANKL by tumor-derived factors. These factors increase expression of RANKL, which induce osteoclast formation, function, and survival, thereby increasing bone resorption. RANK is unexpectedly expressed by cancer cells, and the activation of RANKL-RANK pathway correlates with an increased invasive phenotype. To investigate the interaction between RANK expression in human breast and prostate cancer cells and their pro-metastatic phenotype we analyzed the activation of RANKL-RANK pathway and its effects on cell migration, invasion, gene expression in vitro, and osteolysis-inducing ability in vivo. RANKL activates kinase signaling pathways, stimulates cell migration, increases cell invasion, and up-regulates MMP-1 expression. In vivo, MMP-1 knockdown resulted in smaller x-ray osteolytic lesions and osteoclastogenesis, and decreased tumor burden. Therefore, RANKL inhibition in bone metastatic disease may decrease the levels of the osteoclastogenesis inducer MMP-1, contributing to a better clinical outcome.
