FACEPE A epiisopiloturina (EPI), um alcalóide extraído do resíduo de produção da pilocarpina, tem demonstrado promissora atividade anti-leishmaniose e esquistossomicida in vitro (doenças negligenciadas). Entretanto, por se tratar de um protótipo à fármaco, suas características físico-químicas ainda são pouco esclarecidas. A literatura indica ainda que EPI apresenta baixa solubilidade aquosa, condição que se apresenta como um entrave na dissolução deste protótipo, podendo repercutir na sua absorção e biodisponibilidade. Uma das alternativas tecnológicas para incrementar tal propriedade é a complexação com ciclodextrinas (CDs), polímero capaz de interagir com o fármaco hidrofóbico, aumentando a solubilidade. Assim, este trabalho tem como objetivo realizar a caracterização físico-química da EPI, verificar sua compatibilidade físico-química frente a excipientes farmacêuticos e incrementar seu perfil de dissolução por meio da obtenção de complexos de inclusão (CI) com as seguintes CDs: βCD, sulfobutiléter-βCD e hidroxipropilmetil- βCD, por liofilização. A análise por Infravermelho e o perfil térmico da EPI obtido estão de acordo com a literatura, confirmando a identificação da molécula. A caracterização por Difração de Raios-X e Microscopia Eletrônica de Varredura evidenciou o comportamento cristalino da EPI. A obtenção da cinética de degradação não-isotérmica revelou uma reação de ordem 1, com energia de ativação de 88,95 kJ.mol-1. O pKa da EPI, detectado por meio do Sirius T3®, foi de 7,29, demonstrando que a molécula se encontra em sua maior parte ionizada no meio durante toda a extensão do trato gastrintestinal. No perfil de dissolução, aos 15 minutos, a EPI apresentou 97,34% de dissolução em pH 1,2 e 48,1% em meio pH 6,8. No estudo de compatibilidade, verificou-se que EPI é compatível com todos os excipientes testados (lactose monohidratada; Amido de milho; Celulose Microcristalina; Polivinilpirrolidona K-30 e Hidroxipropil metil celulose. A formação de CI da EPI com as CDs foi confirmada pelas mesmas técnicas de caracterização, que evidenciaram indícios de fortes interações entre o protótipo e todas as CDs testadas, repercutindo em incremento do perfil de dissolução da EPI em pH 6,8 (1,8 vezes). Observou-se também a interessante associação da EPI com as CDs na forma de mistura física, sem o processamento das amostras. O contato destes materiais foi favorável a molhabilidade da EPI o que proporcionou aumento na taxa de dissolução da EPI. Assim, é possível concluir que a caracterização do protótipo forneceu subsídios para compreensão de suas limitações e vantagens, e que a EPI se mostrou compatível frente aos excipientes empregados na obtenção de formas farmacêuticas sólidas. A complexação com CDs incrementou de forma satisfatória o perfil de dissolução do protótipo, sendo estes potenciais excipientes para o ajuste desta propriedade. The Epiisopiloturine (EPI), an alkaloid extracted from the residue production of pilocarpine, has shown promising anti-leishmaniasis activity in vitro and schistosomicidal (neglected diseases). However, in the case of a prototype the drug, its physical and chemical characteristics are still unclear. The literature also indicates that EPI has low aqueous solubility, a condition that appears as a barrier for the dissolution of this prototype and can pass on their absorption and bioavailability. One of the technological alternatives to increase the property is complexation with cyclodextrins (CDs) polymer capable of interacting with the hydrophobic drug by increasing the solubility. Thereby, this study aims to define the physical-chemical characterization of EPI, check their physical and chemical compatibility against pharmaceutical excipients and increase its dissolution profile by obtaining inclusion complexes (IC) with the following CDs: βCD, sulfobutylether-βCD and hydroxypropyl-βCD by lyophilization. The infrared (IR) analysis and the thermal profile of the EPI obtained are according to the literature, confirming the identification of the molecule. The characterization by X-ray diffraction and scanning electron microscopy showed the crystalline behavior of EPI. Obtaining the non-isothermal degradation kinetics revealed a reaction order 1, with activation energy of 88.95 kJ.mol-1. The pKa of EPI detected by Sirius T3® was 7.29, showing that the molecule is mostly ionized in the medium for the entire length of the gastrointestinal tract. In the dissolution profile, after 15 minutes, The EPI presented 97.34% dissolution at pH 1.2 and 48.1% in medium pH 6.8. In compatibility study, it was found that EPI is compatible with all the tested excipients (lactose monohydrate, maize starch, microcrystalline cellulose, Polyvinylpyrrolidone K-30, hydroxypropyl methyl cellulose. The IC formation of EPI with DCs was confirmed by the same characterization techniques, which showed evidence of strong interactions between the prototype and all tested CDs, reflecting the increase in EPI the dissolution profile at pH 6.8 (1.8 times). Also noted up the interesting combination of EPI with the CDs in the form of physical mixture, without the sample processing. The contact of these materials was favorable wettability of EPI which provided increased dissolution rate of EPI. Thus, we conclude that the characterization of the prototype provided information for understanding its limitations and advantages, and that EPI proved compatible front of the excipients employed in producing solid dosage forms. The complexation with the CD satisfactorily increased dissolution profile of the prototype, with these potential excipients for adjusting this property.
