2. Relationship between Feline Knee Joint Osteoarthritis and Bone Mineral Density Quantified Using Computed Tomography and Computed Digital Absorptiometry.

Abstract

Simple Summary: Osteoarthritis (OA) is a common and painful condition affecting 91% of cats, with knee joint OA present in 50% of cases. Diagnosing feline knee joint OA typically involves clinical examination and radiographic imaging, which also provides information about bone health, including bone mineral density (BMD). While the link between knee joint OA and BMD is well-established in humans, it has not been studied in cats. Understanding this relationship could shed light on how bone changes contribute to OA. This study aims to measure the BMD of cat knee joints, compare different BMD measurement methods, and explore how BMD changes with various severities of OA. For this purpose, 46 cat knee joints were analyzed using two techniques: computed tomography (CT) for the volumetric BMD (vBMD) and conventional radiography with a method called computed digital absorptiometry (CDA) for relative BMD (rBMD). This study found a positive correlation between vBMD and rBMD in key areas of the knee, such as the distal femur, patella, and proximal tibia. Additionally, this study adjusted rBMD for bone size differences (corrected rBMD) because variations in bone width were observed. All measures—vBMD, rBMD, and corrected rBMD—differed between normal knee joint and knee joint with mild to severe OA. Furthermore, all measures showed a linear increase as OA severity worsened. This suggests that there is a relationship between OA and BMD in the feline knee joint, which can be preliminarily confirmed by this study. Osteoarthritis (OA), including knee joint OA, is a common chronic condition in cats. In both cats and humans, knee joint OA is characterized radiographically by the presence of osteophytes, enthesiophytes, subchondral sclerosis, and joint space narrowing. However, only in humans have these radiographic signs been reported to increase bone mineral density (BMD). Therefore, this study aims to quantify the volumetric (vBMD) and relative (rBMD) BMD measures of the feline knee joint and compare BMD measures between various severities of OA to test the hypothesized OA–BMD relationship in the knee joint in cats. The 46 feline knee joints were imaged using computed tomography (CT) and conventional radiography supported by the computed digital absorptiometry (CDA) method to obtain vBMD and rBMD, respectively. Both BMD measures were assessed in three regions of interest (ROIs): the distal femur (ROI 1), patella (ROI 2), and proximal tibia (ROI 3). In all locations, vBMD and rBMD showed moderate (ROI 2: r = 0.67, p < 0.0001) to strong (ROI 1: ρ = 0.96, p < 0.0001; ROI 3: r = 0.89, p < 0.0001) positive correlations. Due to differences (p < 0.0001) in the width of the distal femur (17.9 ± 1.21 mm), patella (8.2 ± 0.82 mm), and proximal tibia (19.3 ± 1.16 mm), the rBMD was corrected (corr rBMD) using the thickness coefficient of 0.46 ± 0.04 for ROI 2 and 1.08 ± 0.03 for ROI 3. Regardless of the quantification method used, BMD measures increased linearly from a normal knee joint to severe OA, with differences in BMD between normal and mild to severe knee joint OA. 6. Nazi 'black' Propaganda to Britain: Secret Radio Stations and British Renegades.

Abstract

While many will be familiar with the British traitor William Joyce who, as Lord Haw-Haw, became infamous for his radio broadcasts to Britain on behalf of Nazi Germany during the Second World War, much less widely known and studied is the work of Büro Concordia, an organisation created by Joseph Goebbels to broadcast 'black' propaganda from Berlin to Britain. Between 1940 and 1945 five secret radio stations posed as the voice of British dissident, anti-war organisations operating within the United Kingdom, broadcasting a stream of propaganda which aimed to demoralise the British people and undermine its support for the war. While they can be credited with some limited success, they fell far short of what Goebbels had hoped and expected from them. These stations were launched by the Reich Propaganda Ministry, but the two dozen writers and speakers who produced these broadcasts from studios in Berlin were British citizens. This study evaluates these 'British renegades' and the system that identified, recruited and trained them, and concludes that whatever potential there might have been to influence the course of the war, the employment of people so wholly unsuited to the work ensured that Concordia was bound to fail from its inception. 8. Radio Caledonia: Scottish Nationalism and Nazi Radio Propaganda, 1940–1942.

Abstract

In June 1940, from somewhere in Scotland, a radio station named Radio Caledonia began to broadcast Scottish nationalist, anti-government and anti-war propaganda to Britain, on behalf of an organisation called the Scottish Peace Front. Its objective was to bring about Scotland's exit from the war, an end to the capitalist system, and the creation of an independent, socialist Scotland. The speaker and scriptwriter behind Radio Caledonia was Donald Grant, a Scot, since dubbed Scotland's Lord Haw-Haw. But Radio Caledonia was only superficially a Scottish nationalist station. In reality, the Scottish Peace Front was an entirely fictitious group, and Radio Caledonia's programmes were made in and broadcast from Berlin under the direction of Joseph Goebbels, as one element of his propaganda campaign against Britain. Radio Caledonia, like other similar 'secret stations' commissioned by Goebbels, was conceived of by the Nazi Propaganda Ministry, and its broadcasts written and produced by British renegades. This paper looks at Grant's background and political beliefs, the Nazi propaganda organisation within which he worked in Berlin, and the broadcast output of Radio Caledonia. It concludes that far from being an authentically nationalist or socialist enterprise, Radio Caledonia was devised by German Nazis and British fascists as a tool to achieve their shared aim of bringing about an end to Britain's involvement in the war. 10. ‘Loathsome people’: British informers in the Nazi-occupied Channel Islands.

Abstract

For those who have speculated about the behaviour of the British people under Nazi rule, the Channel Islands have sometimes been used as a proxy, from which evidence can be selected to hypothesise about a Nazi-occupied Britain. In the area of collaboration, in particular, the historiography of the Channel Islands has been a victim of this flawed Anglo-centric approach. Looking at the evidence for the Channel Islands in their own right, this article looks at informing, as one of the most damaging and betterdocumented forms of collaboration, and asks: what kind of people were informers, what were their motives, and were they an integral part of a self-policing Nazi terror state? How did British Intelligence respond to informers on both professional and personal levels, and with what success did they investigate collaboration after the liberation of the Islands in May 1945? Abstract

The hormone FGF21 regulates carbohydrate and lipid homeostasis as well as body weight, and increasing FGF21 improves metabolic abnormalities associated with obesity and diabetes. FGF21 is thought to act on its target tissues, including liver and adipose tissue, to improve insulin sensitivity and reduce adiposity. Here, we used mice with selective hepatic inactivation of the IR (LIRKO) to determine whether insulin sensitization in liver mediates FGF21 metabolic actions. Remarkably, hyperglycemia was completely normalized following FGF21 treatment in LIRKO mice, even though FGF21 did not reduce gluconeogenesis in these animals. Improvements in blood sugar were due in part to increased glucose uptake in brown fat, browning of white fat, and overall increased energy expenditure. These effects were preserved even after removal of the main interscapular brown fat pad. In contrast to its retained effects on reducing glucose levels, the effects of FGF21 on reducing circulating cholesterol and hepatic triglycerides and regulating the expression of key genes involved in cholesterol and lipid metabolism in liver were disrupted in LIRKO mice. 30. ESCAPE to Reality, Post-Trial Outcomes in an ESCAPE Centre: A Retrospective Case-Control Study.

Abstract

Introduction The Royal Victoria Hospital, Belfast provides the regional neuroendovascular service for Northern Ireland and was an enrolling centre for the ESCAPE endovascular stroke trial. Our aim was to assess outcomes for patients presenting with acute stroke following discontinuation of trial enrolment at our centre. Methods We collected data on all patients presenting with acute stoke between Nov-1st-2014 and Oct-31st-2015 who received endovascular treatment or received IV thrombolysis (IV-tPA) alone. ESCAPE eligibility of each patient was assessed. Primary outcome was modified Rankin Score (mRS) at 3 months. Results 129 patients presented with acute stoke symptoms during the time period; 56/129 (43.4%) patients in the intervention group and 73/129 (56.5%) patients in the control group. In the interventional group, 42/56 (75%) were considered ESCAPE eligible and 14/56 (25%) were considered ESCAPE ineligible. 30/42 (71.4%) ESCAPE eligible patients had a positive functional outcome at 3 months compared to 9/14 (64.2%) ESCAPE ineligible patients. In the control group, 37 (50.7%) had identifiable thrombotic occlusion and 13/37 (35.1%) were considered eligible for intervention. 4/13 (30.8%) achieved functional independence (mRS<3) at 3 months. There was a statistically significant difference in functional independence in those who underwent endovascular therapy compared to the control group (p= 0.04). Conclusion ESCAPE eligible patients in our centre had favourable outcome rates superior to the published trial data. ESCAPE ineligible patients tended to do slightly less well, but still better than the favourable outcome rates achieved with IVtPA alone. There is potentially a wide discordance between the threshold for futility and trial eligibility criteria when considering endovascular treatment for acute ischaemic stroke. [ABSTRACT FROM AUTHOR]

Targets are frequently counter-productive because organisations concentrate on the target rather than the work it is supposed to promote. In the case of youth crime, there have been apparently impressive reductions in the number of young people entering the criminal justice system as first offenders. Whether this is primarily the result of prevention of offending or diversion from the system is unclear. This uncertainty is discussed, along with contradictory policy directions in the response to youth crime which suggest the importance of prevention along with trends which may lead key organisations to turn against it. Abstract

Infrared thermography (IRT) was applied as a potentially useful tool in the detection of pregnancy in equids, especially native or wildlife. IRT measures heat emission from the body surface, which increases with the progression of pregnancy as blood flow and metabolic activity in the uterine and fetal tissues increase. Conventional IRT imaging is promising; however, with specific limitations considered, this study aimed to develop novel digital processing methods for thermal images of pregnant mares to detect pregnancy earlier with higher accuracy. In the current study, 40 mares were divided into non-pregnant and pregnant groups and imaged using IRT. Thermal images were transformed into four color models (RGB, YUV, YIQ, HSB) and 10 color components were separated. From each color component, features of image texture were obtained using Histogram Statistics and Grey-Level Run-Length Matrix algorithms. The most informative color/feature combinations were selected for further investigation, and the accuracy of pregnancy detection was calculated. The image texture features in the RGB and YIQ color models reflecting increased heterogeneity of image texture seem to be applicable as potential indicators of pregnancy. Their application in IRT-based pregnancy detection in mares allows for earlier recognition of pregnant mares with higher accuracy than the conventional IRT imaging technique. [ABSTRACT FROM AUTHOR]

47. Fgf8 is required for pharyngeal arch and cardiovascular development in the mouse

Abstract

We present here an analysis of cardiovascular and pharyngeal arch development in mouse embryos hypomorphic for Fgf8. Previously, we have described the generation of Fgf8 compound heterozygous (Fgf8neo/–) embryos. Although early analysis demonstrated that some of these embryos have abnormal left-right (LR) axis specification and cardiac looping reversals, the number and type of cardiac defects present at term suggested an additional role for Fgf8 in cardiovascular development. Most Fgf8neo/– mutant embryos survive to term with abnormal cardiovascular patterning, including outflow tract, arch artery and intracardiac defects. In addition, these mutants have hypoplastic pharyngeal arches, small or absent thymus and abnormal craniofacial development. Neural crest cells (NCCs) populate the pharyngeal arches and contribute to many structures of the face, neck and cardiovascular system, suggesting that Fgf8 may be required for NCC development. Fgf8 is expressed within the developing pharyngeal arch ectoderm and endoderm during NCC migration through the arches. Analysis of NCC development in Fgf8neo/– mutant embryos demonstrates that NCCs are specified and migrate, but undergo cell death in areas both adjacent and distal to where Fgf8 is normally expressed. This study defines the cardiovascular defects present in Fgf8 mutants and supports a role for Fgf8 in development of all the pharyngeal arches and in NCC survival.

Abstract

In order to understand how secreted signals regulate complex morphogenetic events, it is crucial to identify their cellular targets. By conditional inactivation of Fgfr1 and Fgfr2 and overexpression of the FGF antagonist sprouty 2 in different cell types, we have dissected the role of FGF signaling during heart outflow tract development in mouse. Contrary to expectation, cardiac neural crest and endothelial cells are not primary paracrine targets. FGF signaling within second heart field mesoderm is required for remodeling of the outflow tract: when disrupted, outflow myocardium fails to produce extracellular matrix and TGF? and BMP signals essential for endothelial cell transformation and invasion of cardiac neural crest. We conclude that an autocrine regulatory loop, initiated by the reception of FGF signals by the mesoderm, regulates correct morphogenesis at the arterial pole of the heart. 49. Relationship between Feline Knee Joint Osteoarthritis and Bone Mineral Density Quantified Using Computed Tomography and Computed Digital Absorptiometry.

Abstract

Osteoarthritis (OA), including knee joint OA, is a common chronic condition in cats. In both cats and humans, knee joint OA is characterized radiographically by the presence of osteophytes, enthesiophytes, subchondral sclerosis, and joint space narrowing. However, only in humans have these radiographic signs been reported to increase bone mineral density (BMD). Therefore, this study aims to quantify the volumetric (vBMD) and relative (rBMD) BMD measures of the feline knee joint and compare BMD measures between various severities of OA to test the hypothesized OA-BMD relationship in the knee joint in cats. The 46 feline knee joints were imaged using computed tomography (CT) and conventional radiography supported by the computed digital absorptiometry (CDA) method to obtain vBMD and rBMD, respectively. Both BMD measures were assessed in three regions of interest (ROIs): the distal femur (ROI 1), patella (ROI 2), and proximal tibia (ROI 3). In all locations, vBMD and rBMD showed moderate (ROI 2: r = 0.67, p < 0.0001) to strong (ROI 1: ρ = 0.96, p < 0.0001; ROI 3: r = 0.89, p < 0.0001) positive correlations. Due to differences ( p < 0.0001) in the width of the distal femur (17.9 ± 1.21 mm), patella (8.2 ± 0.82 mm), and proximal tibia (19.3 ± 1.16 mm), the rBMD was corrected (corr rBMD) using the thickness coefficient of 0.46 ± 0.04 for ROI 2 and 1.08 ± 0.03 for ROI 3. Regardless of the quantification method used, BMD measures increased linearly from a normal knee joint to severe OA, with differences in BMD between normal and mild to severe knee joint OA. The OA-BMD relationship in the feline knee joint can be preliminarily confirmed.

Abstract

A patient in his mid-40s presented with acute basilar artery thrombosis 7 hours postsymptom onset. Initial attempts to perform mechanical thrombectomy (MT) via the femoral and radial arterial approaches were unsuccessful as the left vertebral artery (VA) was occluded at its origin and the right VA terminated in the posterior inferior cerebellar artery territory, without contribution to the basilar system. MT was thus performed following ultrasound-guided direct arterial puncture of the left VA in its V3 segment, with antegrade advancement of a 4 French radial access sheath. First pass thrombolyisis in cerebral infarction (TICI) 3 recanalisation achieved with a 6 mm Solitaire stent retriever and concurrent aspiration on the 4 French sheath.
