Abstract

We address a biophysical network dynamical model to study how the modulation of dopamine (DA) activity and related N-methyl-d-aspartate (NMDA) glutamate receptor activity as well as the emerging Pre-Frontal Cortex (PFC) functional connectivity network (FCN) affect inhibitory cognitive function in schizophrenia in an antisaccade task. The values of the model parameters and the topology of the PFC-FCN were estimated by minimizing the differences between simulations and the observed distributions of reaction times (RT) during the performance of the antisaccade task in 30 patients with schizophrenia and 30 healthy controls. We show that the proposed model approximates remarkably well the predicted prefrontal cortical DA hypo-activity and the related NMDA receptor hypo-function as well as the FCN dysconnection pattern that are considered as the major etio-pathological hypotheses to explain cognitive deficits in schizophrenia.

Abstract

Digital phenotyping is a nascent multidisciplinary field that has the potential to revolutionize psychiatry and its clinical practice. In this paper, we present a rigorous statistical analysis of short-time features extracted from wearable data, during long-term continuous monitoring of patients with psychotic disorders and healthy control counterparts. Our novel analysis identifies features that fluctuate significantly between the two groups, and offers insights on several factors that differentiate them, which could be leveraged in the future for relapse prevention and individualized assistance.

Abstract

Broad-based cognitive deficits are an enduring and disabling symptom for many patients with severe mental illness, and these impairments are inadequately addressed by current medications. While novel drug targets for schizophrenia and depression have emerged from recent large-scale genome-wide association studies (GWAS) of these psychiatric disorders, GWAS of general cognitive ability can suggest potential targets for nootropic drug repurposing. Here, we (1) meta-analyze results from two recent cognitive GWAS to further enhance power for locus discovery; (2) employ several complementary transcriptomic methods to identify genes in these loci that are credibly associated with cognition; and (3) further annotate the resulting genes using multiple chemoinformatic databases to identify "druggable" targets. Using our meta-analytic data set (N = 373,617), we identified 241 independent cognition-associated loci (29 novel), and 76 genes were identified by 2 or more methods of gene identification. Actin and chromatin binding gene sets were identified as novel pathways that could be targeted via drug repurposing. Leveraging our transcriptomic and chemoinformatic databases, we identified 16 putative genes targeted by existing drugs potentially available for cognitive repurposing.

Abstract

Susceptibility to schizophrenia is inversely correlated with general cognitive ability at both the phenotypic and the genetic level. Paradoxically, a modest but consistent positive genetic correlation has been reported between schizophrenia and educational attainment, despite the strong positive genetic correlation between cognitive ability and educational attainment. Here we leverage published genome-wide association studies (GWASs) in cognitive ability, education, and schizophrenia to parse biological mechanisms underlying these results. Association analysis based on subsets (ASSET), a pleiotropic meta-analytic technique, allowed jointly associated loci to be identified and characterized. Specifically, we identified subsets of variants associated in the expected ("concordant") direction across all three phenotypes (i.e., greater risk for schizophrenia, lower cognitive ability, and lower educational attainment); these were contrasted with variants that demonstrated the counterintuitive ("discordant") relationship between education and schizophrenia (i.e., greater risk for schizophrenia and higher educational attainment). ASSET analysis revealed 235 independent loci associated with cognitive ability, education, and/or schizophrenia at p < 5 × 10-8. Pleiotropic analysis successfully identified more than 100 loci that were not significant in the input GWASs. Many of these have been validated by larger, more recent single-phenotype GWASs. Leveraging the joint genetic correlations of cognitive ability, education, and schizophrenia, we were able to dissociate two distinct biological mechanisms-early neurodevelopmental pathways that characterize concordant allelic variation and adulthood synaptic pruning pathways-that were linked to the paradoxical positive genetic association between education and schizophrenia. Furthermore, genetic correlation analyses revealed that these mechanisms contribute not only to the etiopathogenesis of schizophrenia but also to the broader biological dimensions implicated in both general health outcomes and psychiatric illness.

Abstract

Hill (Twin Research and Human Genetics, Vol. 21, 2018, 84-88) presented a critique of our recently published paper in Cell Reports entitled 'Large-Scale Cognitive GWAS Meta-Analysis Reveals Tissue-Specific Neural Expression and Potential Nootropic Drug Targets' (Lam et al., Cell Reports, Vol. 21, 2017, 2597-2613). Specifically, Hill offered several interrelated comments suggesting potential problems with our use of a new analytic method called Multi-Trait Analysis of GWAS (MTAG) (Turley et al., Nature Genetics, Vol. 50, 2018, 229-237). In this brief article, we respond to each of these concerns. Using empirical data, we conclude that our MTAG results do not suffer from 'inflation in the FDR [false discovery rate]', as suggested by Hill (Twin Research and Human Genetics, Vol. 21, 2018, 84-88), and are not 'more relevant to the genetic contributions to education than they are to the genetic contributions to intelligence'.

Abstract

Background: Deficits in Multisensory Integration (MSI) in ASD have been reported repeatedly and have been suggested to be caused by altered longrange connectivity. Here we investigate behavioral and ERP correlates of MSI in ASD using ecologically valid videos of emotional expressions. Methods: In the present study, we set out to investigate the electrophysiological correlates of audiovisual MSI in young autistic and neurotypical adolescents. We employed dynamic stimuli of high ecological validity (500 ms clips produced by actors) that depicted fear or disgust in unimodal (visual and auditory), and bimodal (audiovisual) conditions. Results: We report robust MSI effects at both the behavioral and electrophysiological levels and pronounced differences between autistic and neurotypical participants. Specifically, neurotypical controls showed robust behavioral MSI for both emotions as seen through a significant speed-up of bimodal response time (RT), confirmed by Miller's Race Model Inequality (RMI), with greater MSI effects for fear than disgust. Adolescents with ASD, by contrast, showed behavioral MSI only for fear. At the electrophysiological level, the bimodal condition as compared to the unimodal conditions reduced the amplitudes of the visual P100 and auditory P200 and increased the amplitude of the visual N170 regardless of group. Furthermore, a cluster-based analysis across all electrodes revealed that adolescents with ASD showed an overall delayed and spatially constrained MSI effect compared to controls. Conclusion: Given that the variables we measured reflect attention, our findings suggest that MSI can be modulated by the differential effects on attention that fear and disgust produce. We also argue that the MSI deficits seen in autistic individuals can be compensated for at later processing stages by (a) the attention-orienting effects of fear, at the behavioral level, and (b) at the electrophysiological level via increased attentional effort. Abstract

Intelligence is highly heritable1 and a major determinant of human health and well-being2. Recent genome-wide meta-analyses have identified 24 genomic loci linked to variation in intelligence3-7, but much about its genetic underpinnings remains to be discovered. Here, we present a large-scale genetic association study of intelligence (n = 269,867), identifying 205 associated genomic loci (190 new) and 1,016 genes (939 new) via positional mapping, expression quantitative trait locus (eQTL) mapping, chromatin interaction mapping, and gene-based association analysis. We find enrichment of genetic effects in conserved and coding regions and associations with 146 nonsynonymous exonic variants. Associated genes are strongly expressed in the brain, specifically in striatal medium spiny neurons and hippocampal pyramidal neurons. Gene set analyses implicate pathways related to nervous system development and synaptic structure. We confirm previous strong genetic correlations with multiple health-related outcomes, and Mendelian randomization analysis results suggest protective effects of intelligence for Alzheimer's disease and ADHD and bidirectional causation with pleiotropic effects for schizophrenia. These results are a major step forward in understanding the neurobiology of cognitive function as well as genetically related neurological and psychiatric disorders.

Abstract

Here, we present a large (n = 107,207) genome-wide association study (GWAS) of general cognitive ability ("g"), further enhanced by combining results with a large-scale GWAS of educational attainment. We identified 70 independent genomic loci associated with general cognitive ability. Results showed significant enrichment for genes causing Mendelian disorders with an intellectual disability phenotype. Competitive pathway analysis implicated the biological processes of neurogenesis and synaptic regulation, as well as the gene targets of two pharmacologic agents: cinnarizine, a T-type calcium channel blocker, and LY97241, a potassium channel inhibitor. Transcriptome-wide and epigenome-wide analysis revealed that the implicated loci were enriched for genes expressed across all brain regions (most strongly in the cerebellum). Enrichment was exclusive to genes expressed in neurons but not oligodendrocytes or astrocytes. Finally, we report genetic correlations between cognitive ability and disparate phenotypes including psychiatric disorders, several autoimmune disorders, longevity, and maternal age at first birth.

Abstract

Although magnetic resonance spectroscopy (MRS) has provided in vivo measurements of brain chemical profiles in bipolar disorder (BD), there are no data on clinically and therapeutically important onset polarity (OP) and predominant polarity (PP). We conducted a proton MRS study in BD polarity subphenotypes, focusing on emotion regulation brain regions. Forty-one euthymic BD patients stratified according to OP and PP and sixteen healthy controls (HC) were compared. 1H-MRS spectra of the anterior and posterior cingulate cortex (ACC, PCC), left and right hippocampus (LHIPPO, RHIPPO) were acquired at 3.0T to determine metabolite concentrations. We found significant main effects of OP in ACC mI, mI/tNAA, mI/tCr, mI/tCho, PCC tCho, and RHIPPO tNAA/tCho and tCho/tCr. Although PP had no significant main effects, several medium and large effect sizes emerged. Compared to HC, manic subphenotypes (i.e., manic-OP, manic-PP) showed greater differences in RHIPPO and PCC, whereas depressive suphenotypes (i.e., depressive-OP, depressive-PP) in ACC. Effect sizes were consistent between OP and PP as high intraclass correlation coefficients (ICC) were confirmed. Our findings support the utility of MRS in the study of the neurobiological underpinnings of OP and PP, highlighting that the regional specificity of metabolite changes within the emotion regulation network consistently marks both polarity subphenotypes. Abstract

Individuals with autism spectrum disorder (ASD) show altered processing of sensory stimuli as compared to typically developed (TD) individuals, and this difference is maintained during their development (e.g., Greenspan & Wieder, 1997). This atypical sensory processing has also been reported regarding their ability to integrate multisensory stimuli (e.g., Foss-Feig et al., 2010), and more specifically, for the integration of audiovisual speech stimuli (e.g., Giraud et al., 2012). A multisensory illusion commonly utilized to examine multisensory integration (MI) in individuals with ASD is the McGurk illusion (i.e., the combination of an auditory /ba/ with the simultaneous visual presentation of a person uttering the syllable /ga/, leads to the percept of a third syllable, usually /da/, /ta/, or /tha/; McGurk & McDonald, 1976). Studies utilizing the McGurk illusion have demonstrated that children with ASD exhibit lower rates of the illusion as compared to TD matched children, indicating weaker MI abilities (e.g., Bebko, Schroeder, & Weiss, 2013; Irwin, Tornatore, Brancazio, & Whalen, 2011; Mongillo et al., 2008; Stevenson et al., 2014d; Taylor, Isaac, & Milne, 2010; Williams et al., 2004; Woynaroski et al., 2013), or wider TBWs indicating lower levels of multisensory binding selectivity (e.g., Feldman et al., 2019; Woynaroski et al., 2013). Additionally, the "sound-induced flash illusion" (SIFI) is commonly used to examine MI abilities of ASD and TD individuals, whereby the presentation of a single visual flash with multiple auditory tones leads to the illusory percept that multiple flashes were presented (or “fission” illusion; Shams, Kamitani, & Shimojo, 2000), while the presentation of two visual flashes accompanied by a single auditory tone leads to the illusory percept that a unique flash (or “fusion” illusion) was presented. It has been found that ASD and TD children exhibit similar levels of “fission” illusion, but individuals with ASD experience the “fusion” illusion to a greater extent, and therefore, exhibit reduced selectivity regarding the integration of multisensory stimuli (e.g., Bao et al., 2017). It has also been shown that children with ASD experience the SIFI illusion for greater TBWs than TD children (e.g., Foss-Feig et al., 2010), and that children with ASD are less sensitive in experiencing this illusion, indicating an impaired MI ability (e.g., Stevenson et al., 2014e). These findings highlight the MI differences between individuals with ASD and TD with the use of both simple and complex audiovisual stimuli. In the present study, in order to obtain a better understanding of MI in ASD, we aim to: a) investigate how MI maturates during development (from toddlers to adolescents) in both ASD and TD participants, b) obtain a detailed participants’ clinical assessment, in order to associate MI abilities with ASD related symptomatology, and c) examine and compare ASD individuals’ performance for both social complex AV stimuli and simple AV stimuli through MI illusions. Specifically, the McGurk paradigm will be utilized so that ASD and TD matched toddlers, young children and adolescents will perform a simple detection task in synchronous or asynchronous stimulus pairings (Experiment 1). The same individuals will also participate in a set of experiments utilizing the SIFI, as it will allow for the comparison of the MI processes of ASD and TD children between complex, social and simple, non-social AV stimuli. Specifically, children and adolescents with ASD and their TD matched individuals will be asked to identify the number of the presented flashes in each trial (Experiment 2). Furthermore, for illusory trials containing a single flash and two beeps, various SOAs will be introduced between the onset of the flash and the second beep (as the first beep’s onset will always coincide with that of the flash’s), so that we will be able to investigate the temporal window within which MI occurs (i.e., the illusion is experienced within that window).

Abstract

Objective: Apathy, a frequent neuropsychiatric symptom in aging neurocognitive disorders, has been associated with cognitive decline and functional disability. Therefore, timely provision of pharmacological interventions for apathy is greatly needed. Design: A systematical literature review of existing studies was conducted up to 30 May 2023 in several databases (PubMed, PsychInfo, Cochrane, Google Scholar, etc.) that included randomized controlled trials (RCTs) and meta-analyses assessing pharmacological treatments for apathy in aging neurocognitive disorders. The quality of the studies was appraised. Results: In patients with Alzheimer's Disease (AD), donepezil, galantamine, rivastigmine, methylphenidate, and gingko biloba were proven efficacious for apathy, while rivastigmine, cognitive enhancer IRL752 and piribedil were found to be beneficial in patients with Parkinson's Disease (PD) and agomelatine in patients with Frontotemporal Dementia (FD). The extensive proportion of RCTs in which apathy was used as a secondary outcome measure, along with the considerable methodological heterogeneity, did not allow the evaluation of group effects. Conclusions: Pharmacological interventions for apathy in aging neurocognitive disorders are complex and under-investigated. The continuation of systematic research efforts and the provision of individualized treatment for patients suffering from these disorders is vital. [ABSTRACT FROM AUTHOR]
