1. Remembering 'the scariest movie of all time' : a grounded audience study of 'The Exorcist'

Abstract

This study of the forty-six-year history of The Exorcist (Friedkin, 1973) uses grounded theory methodology to investigate how audiences have engaged with and now remember what is still repeatedly voted as “the scariest movie of all time” (Bailey, 2018). Original data from 746 survey respondents and 32 interviewees form the basis for new theories of the importance and meaning of physical and social place in cinema-going, processes of parental regulation over children’s viewing habits and their associated meanings, and the relationship between memories of film experiences and intra-family dynamics. Since its inception in 1965 in the field of medicine and patient care (Gla ser and Strauss, 1965), grounded theory methodology has provided an insightful, flexible, and participant-driven method of producing research which rejects extant concepts and theories to produce findings which are thus “grounded” in original data. This thesis represents the result of the first study of media audiences to fully employ grounded theory methodology, taking Kathy Charmaz’s (2014) constructivist iteration as its basis. Its findings are contextualised within theorisations of place, censorship, and memory from New Cinema History (Jancovich, Faire, and Stubbings, 2003; Kuhn, 2002; Biltereyst, Maltby, and Meers 2011), censorship studies (Kuhn, 1988; Smith, 2005; Barber, 2016; Smith, 2019), and oral history (Portelli, 1981; Thompson, 2000; Thomson, 2006). The overarching theme of this study to emerge from participants’ accounts is how memories of The Exorcist are defined not by the film itself, nor by any factor which can be measured with demographic information, but by participants’ relationships with other people. This particularly includes their families and, often, their past selves. In adapting grounded theory for audience studies, this thesis conceives of and provides an outline for grounded audience studies as an alternative approach to researching audiences which can better reflect the complexity of everyday life.

2. Dynamic structures of rare conformations involved in amyloid formation

Abstract

In work described in this thesis the conformational landscape of a recently identified ß2m variant, D76N, was examined with NMR and state of the art molecular dynamics simulations. The D76N ß2m variant was recently identified as the first naturally occurring amyloidogenic variant of ß2m, a 99 residue, seven stranded Ig domain protein. The D76N variant of ß2m causes a hereditary, late onset and fatal systemic amyloid disease. Affected individuals are heterozygous for the D76N mutation and thus also express wild-type (WT) ß2m in the serum. Co-aggregation of D76N and WT ß2m is not observed in vivo, and D76N amyloidosis follows a distinctive disease progression from the WT ß2m associated disease, Dialysis-Related Amyloidosis, which occurs in patients undergoing long-term haemodialysis. This work aims to understand the mechanism by which the single amino acid substitution, D76N, results in a dramatically more aggregation-prone protein while maintaining the same overall native structure as WT ß2m. In this work, the structure, stability and dynamics of the D76N variant are studied with NMR spectroscopy and a range of complementary biophysical techniques. The folding intermediates of D76N and WT ß2m are also probed with real-time NMR and circular dichroism spectroscopy, revealing structural, dynamic and kinetic insights into the folding intermediates present within the D76N and WT ß2m folding pathways and their roles in aggregation. The study of the folding intermediates demonstrated that the IT -state, which is a known precursor for ß2m aggregation, is not responsible for the rapid aggregation of the D76N variant. Instead, a low-populated state, unique to D76N, in equilibrium with the native state must be responsible for D76N aggregation, consistent with the different disease progression associated with D76N. The NMR and data-guided molecular dynamics simulations presented in this thesis reveal the effects of the D76N substitution on the conformational landscape of the D76N variant and detail the structures of the low-populated states in equilibrium with the native state. Additionally, saturation mutagenesis of position 76 in D76N, carried out with a split-ßlactamase assay have demonstrated the exquisite specificity for an aspartate residue at position 76 in the ß2m structural ensemble.

6. On the design of finite-state type systems

Abstract

Practical computers have only finite amounts of memory. However, the programs that run on them are often written in languages that effectively assume (via providing constructs such as general recursion) that infinite memory is available, meaning that an implementation of those programs is necessarily an approximation. The main focus of this thesis is on the use of contraction: the ability to use a function parameter more than once in the body of that function (or more generally, to mention a free variable more than once in a term). Unrestricted contraction is a common reason for a language to require unbounded amounts of memory to implement. This thesis looks at a range of type systems, both existing and new, that restrict the use of contraction so that they can be implemented with finite amounts of state, identifying common themes, and explaining and suggesting solutions for common deficiencies. In particular, different restrictions on contraction are seen to correspond to different features of the language’s implementation.

9. Changes in acetate availability : effect upon substrate utilisation at rest and during exercise

Abstract

The aim of the present series of studies was to investigate the effect of sodium acetate ingestion on energy metabolism with particular emphasis upon changes in fat and carbohydrate utilisation at rest and during exercise. Ingestion of sodium acetate at rest reduced fat oxidation by 30% over a 90 min period post-ingestion assuming that 90% of the ingested organic acid was oxidised during this time. The decrease in fat utilisation was found to be almost entirely accounted for by oxidation of the exogenous acetate such that carbohydrate utilisation was unaffected.  Plasma glycerol concentration was unchanged following ingestion of the sodium salt, suggesting that the suppression in fat metabolism following acetate ingestion was not due to reduced lipid availability and may have been located intracellularly.  Through the use of stable isotope techniques it was subsequently shown that 80% of the ingested bolus was oxidised over 180 min post-ingestion when sodium acetate was ingested at a dose of 2 mmol.kg-1 body mass.  Fat utilisation was found to be significantly lower over this period compared to ingestion of a similar dose of sodium bicarbonate (18.43 ± 7.12 vs. 13.85 ± 4.69 g in the bicarbonate and acetate trials, respectively; P < 0.05), with no difference in carbohydrate utilisation between trials. Despite acetate being shown to have no effect upon carbohydrate utilisation, it was observed in a separate study that acetate ingestion improved glucose tolerance during a 75 g oral glucose tolerance test. Blood glucose and plasma insulin concentration were significantly elevated 15 min after consumption of a glucose load compared with a trial matched on fluid volume, sodium ingested and changes in acid-base balance (bicarbonate ingestion).  Glucose and insulin then decreased at a quicker rate between 30 and 120 min post-ingestion of the glucose load following sodium acetate ingestion.  In a second series of studies the effect of perturbing substrate utilisation at rest on metabolism during subsequent prolonged exercise was examined. Ingestion of sodium acetate at a dose of 4 mmol.kg-1 body mass was shown to suppress fat oxidation at rest (0.11 ± 0.03 vs. 0.06 ± 0.02 g.min-1 in the bicarbonate and acetate trials, respectively; P < 0.05) with no effect upon carbohydrate utilisation thus supporting the previous findings.

10. Modelling diagenesis in archaeological bone

Abstract

This thesis presents a series of papers intended to further our knowledge and understanding of bone taphonomy, with special attention given to the survival of organic molecules (collagen and DNA). An integrated modelling and measuring approach has been taken. Initially, the rate of chemical degradation of bone collagen was estimated from data derived from high temperature laboratory experiments. These rates were then extrapolated to realistic archaeological burial temperatures using palaeoclimatic reconstructions. The model was tested using radiocarbon databases using data on location and collagen yield. Despite the simplicity of the approach and the large errors involved, it does appear that the model may be useful for predicting the limits of collagen survival, although most bones have less collagen than the model predicts. A similar model was produced for the depurination of DNA. The models which are based on rates of chemical deterioration, highlight the role that temperature may play in biomolecular preservation, however they fail to address the complexity of bone taphonomy. A study of bones from a site in southern Italy (Apigliano) identified bone with no evidence of microbial attack, but otherwise severe diagenesis (including little or no collagen) - so called `pre-fossilised' bone. In a wider study of 200+ archaeological bones excavated form 40 sites across Europe this `pre-fossilisation' was identified as one of four possible diagenetic states, the others being, `microbial attack', `complete dissolution' and `good preservation'. These states of preservation are defined by characteristic sets of diagenetic parameters, although the environmental and taphonomic conditions that control these states of preservation are less well defined. In addition to the standard diagenetic parameters measured, the preservation of osteocalcin (a potential proxy for DNA) was investigated using immunological techniques.

11. Modern language pedagogy, spiritual development and Christian faith : a study of their interrelationships

Abstract

This thesis sets out to investigate the relationship between the design of modern language pedagogy, spiritual development in the school context, and Christian faith. It does so in the context of recent discussions of spiritual development across the curriculum, and draws upon literature on faith-learning integration emerging from the evangelical and Reformed Christian traditions. Part one of the thesis develops a framework within which the interconnections between these areas can be examined. Negatively, a range of objections to establishing a relationship between Christian faith on the one hand and spiritual development and modern language teaching methodology on the other are countered. Positively, the structure of teaching 'methodology' and the roles of control beliefs, spiritual virtues and metaphor in shaping an approach to teaching are elucidated. Five 'significant themes' in Christian reflection on the nature of the human person are discussed in terms of their theological centrality, pedagogical relevance and distinctive contribution to a view of the learner as a spiritual being. Hospitality to the alien is proposed as a metaphor which could fruitfully inform modern language learning. Part two of the thesis examines three modern language pedagogies using the framework developed in part one. The version of communicative language teaching implicit in the National Curriculum for Modern Foreign Languages, the humanistic pedagogy of Charles A. Curran (Community Language Learning), and the critical pedagogy of Claire Kramsch are explored in turn for points of connection or tension with spiritual development and Christian faith. The thesis concludes that a Christian conception of the learner's spiritual development does lead to identifiable modifications in the design of modern language pedagogy. The thesis has implications for how modern language pedagogy is understood, for how spiritual development is approached across the curriculum, and for discussions of the relationship between religion and education in school settings.

Abstract

Introduction Anxiety disorders have a high lifetime prevalence, early-onset and long duration or chronicity. Exposure therapy is considered one of the most effective elements in cognitive behavioural therapy (CBT) for anxiety, but in vivo exposure can be challenging to access and control, and is sometimes rejected by patients because they consider it too aversive. Virtual reality allows flexible and controlled exposure to challenging situations in an immersive and protected environment. Aim The SoREAL-trial aims to investigate the effect of group cognitive behavioural therapy (CBT-in vivo) versus group CBT with virtual reality exposure (CBT-in virtuo) for patients diagnosed with social anxiety disorder and/or agoraphobia, in mixed groups. Methods and analysis The design is an investigator-initiated randomised, assessor-blinded, parallel-group and superiority-designed clinical trial. Three hundred two patients diagnosed with social anxiety disorder and/or agoraphobia will be included from the regional mental health centres of Copenhagen and North Sealand and the Northern Region of Denmark. All patients will be offered a manual-based 14-week cognitive behavioural group treatment programme, including eight sessions with exposure therapy. Therapy groups will be centrally randomised with concealed allocation sequence to either CBT-in virtuo or CBT-in vivo. Patients will be assessed at baseline, post-treatment and 1-year follow-up by treatment blinded researchers and research assistants. The primary outcome will be diagnosis-specific symptoms measured with the Liebowitz Social Anxiety Scale for patients with social anxiety disorder and the Mobility Inventory for Agoraphobia for patients with agoraphobia. Secondary outcome measures will include depression symptoms, social functioning and patient satisfaction. Exploratory outcomes will be substance and alcohol use, working alliance and quality of life. 41. Endopeptidases 3.4.24.15 and 24.16 in endothelial cells: potential role in vasoactive peptide metabolism

Abstract

The closely related metalloendopeptidases EC 3.4.24.15 (EP24.15; thimet oligopeptidase) and 24.16 (EP24.16; neurolysin) cleave a number of vasoactive peptides such as bradykinin and neurotensin in vitro. We have previously shown that hypotensive responses to bradykinin are potentiated by an inhibitor of EP24.15 and EP24.16 (26), suggesting a role for one or both enzymes in bradykinin metabolism in vivo. In this study, we have used selective inhibitors that can distinguish between EP24.15 and EP24.16 to determine their activity in cultured endothelial cells (the transformed human umbilical vein endothelial hybrid cell line EA.hy926 or ovine aortic endothelial cells). Endopeptidase activity was assessed using a specific quenched fluorescent substrate [7-methoxycoumarin-4-acetyl-Pro-Leu-Gly-D-Lys(2,4-dinitrophenyl)], as well as the peptide substrates bradykinin and neurotensin (assessed by high-performance liquid chromatography with mass spectroscopic detection). Our results indicate that both peptidases are present in endothelial cells; however, EP24.16 contributes significantly more to substrate cleavage by both cytosolic and membrane preparations, as well as intact cells, than EP24.15. These findings, when coupled with previous observations in vivo, suggest that EP24.16 activity in vascular endothelial cells may play an important role in the degradation of bradykinin and/or other peptides in the circulation. peptidase; bradykinin; inhibitor; endothelium

Abstract

Bradykinin is a vasoactive peptide that has been shown to increase the permeability of the cerebral microvasculature to blood-borne macromolecules. The two zinc metalloendopeptidases EC 3.4.24.15 (EP 24.15) and EC 3.4.24.16 (EP 24.16) degrade bradykinin in vitro and are highly expressed in the brain. However, the role that these enzymes play in bradykinin metabolism in vivo remains unclear. In the present study, we investigated the role of EP 24.15 and EP 24.16 in the regulation of bradykinin-induced alterations in microvascular permeability. Permeability of the cerebral microvasculature was assessed in anesthetized Sprague-Dawley rats by measuring the clearance of 70-kDa FITC dextran from the brain. Inhibition of EP 24.15 and EP 24.16 by the specific inhibitor N-[1-(R,S)-carboxy-3-phenylpropyl]-Ala-Aib-Tyr-p-aminobenzoate (JA-2) resulted in the potentiation of bradykinin-induced increases in cerebral microvessel permeability. The level of potentiation was comparable to that achieved by the inhibition of angiotensin-converting enzyme. These findings provide the first evidence of an in vivo role for EP 24.15/EP 24.16 in brain function, specifically in regulating alterations in microvessel permeability induced by exogenous bradykinin. Abstract

Results show that incorporation of beta-amino acids into the design of inhibitors of metalloendopeptidase EP 24.15 and EP 24.16 decreases the inhibitory rate to a low micromolar range. Data show that the inhibitors are resistant to cleavage by peptidase, suggesting the role of beta-amino acids in stabilizing enzyme inhibitors.
