3. Imaging stress and magnetism at high pressures using a nanoscale quantum sensor

Abstract

Pressure alters the physical, chemical and electronic properties of matter. The development of the diamond anvil cell (DAC) enables tabletop experiments to investigate a diverse landscape of high-pressure phenomena ranging from the properties of planetary interiors to transitions between quantum mechanical phases. In this work, we introduce and utilize a novel nanoscale sensing platform, which integrates nitrogen-vacancy (NV) color centers directly into the culet (tip) of diamond anvils. We demonstrate the versatility of this platform by performing diffraction-limited imaging (~600 nm) of both stress fields and magnetism, up to pressures ~30 GPa and for temperatures ranging from 25-340 K. For the former, we quantify all six (normal and shear) stress components with accuracy $<0.01$ GPa, offering unique new capabilities for characterizing the strength and effective viscosity of solids and fluids under pressure. For the latter, we demonstrate vector magnetic field imaging with dipole accuracy $<10^{-11}$ emu, enabling us to measure the pressure-driven $\alpha\leftrightarrow\epsilon$ phase transition in iron as well as the complex pressure-temperature phase diagram of gadolinium. In addition to DC vector magnetometry, we highlight a complementary NV-sensing modality using T1 noise spectroscopy; crucially, this demonstrates our ability to characterize phase transitions even in the absence of static magnetic signatures. 4. Robust block magnetism in the spin ladder compound BaFe2Se3 under hydrostatic pressure

Abstract

The majority of the iron-based superconductors (FeSCs) exhibit a two-dimensional square lattice structure. Recent reports of pressure-induced superconductivity in the spin-ladder system, BaFe2X3(X=S, Se), introduce a quasi-one-dimensional prototype and an insulating parent compound to the FeSCs. Here we report x-ray, neutron diffraction, and muon spin relaxation experiments on BaFe2Se3 under hydrostatic pressure to investigate its magnetic and structural properties across the pressure-temperature phase diagram. A structural phase transition was found at a pressure of 3.7(3) GPa. Neutron diffraction measurements at 6.8(3) GPa and 120 K show that the block magnetism persists even at these high pressures. A steady increase and then fast drop of the magnetic transition temperature TN and greatly reduced moment above the pressure Ps indicate potentially rich and competing phases close to the superconducting phase in this ladder system.

17. The Impact of Immersive Exhibit Design on Visitor Behaviour and Learning at Chester Zoo, UK

Abstract

As extinction rates accelerate, zoos have evolved from places for public entertainment to centres of conservation, research and education. Zoo-based learning is inherently ‘free-choice’, meaning it is subject to visitors’ experiences, baseline knowledge and visit agendas. Naturalistic, ‘immersive’ exhibits are commonplace in modern zoos and should provide the sense of discovery that maximises free-choice learning. Chester Zoo is developing Grasslands, a multi-species, immersive exhibit based on African savannahs, due to open in 2023. To assess the educational potential of Grasslands, this study uses a mixed-methods approach combining quantitative visitor observation data with qualitative surveys to compare visitor learning at three Chester Zoo exhibits housing charismatic grassland megafauna. The Giraffe House is a traditional, ‘second-generation’ exhibit lacking naturalistic features, whereas Mkomazi Painted Dog Reserve is an immersive, ‘third-generation’ exhibit. Tsavo Black Rhino Reserve is a functional but aesthetically pleasing exhibit, treated as an intermediate. While visitor ‘dwell time’ was almost five times higher at the Giraffe House than Mkomazi, visitors were more than twice as likely to engage with interpretation at Mkomazi than the Giraffe House, and spent significantly more time doing so at Mkomazi than any other exhibit. Survey data revealed occasional, modest increases in knowledge at Mkomazi and Tsavo, while there was no evidence of increased knowledge at the Giraffe House. While overall dwell time did not influence learning, a tentative link between engagement with interpretation and learning outcomes can be drawn. Providing attractive and engaging interpretive elements should therefore be central to the design of Grasslands.

Abstract

Motivation: The BioTIME database contains raw data on species identities and abundances in ecological assemblages through time. These data enable users to calculate temporal trends in biodiversity within and amongst assemblages using a broad range of metrics. BioTIME is being developed as a community‐led open‐source database of biodiversity time series. Our goal is to accelerate and facilitate quantitative analysis of temporal patterns of biodiversity in the Anthropocene. Main types of variables included: The database contains 8,777,413 species abundance records, from assemblages consistently sampled for a minimum of 2 years, which need not necessarily be consecutive. In addition, the database contains metadata relating to sampling methodology and contextual information about each record. Spatial location and grain: BioTIME is a global database of 547,161 unique sampling locations spanning the marine, freshwater and terrestrial realms. Grain size varies across datasets from 0.0000000158 km² (158 cm²) to 100 km² (1,000,000,000,000 cm²). Time period and grain: BioTIME records span from 1874 to 2016. The minimal temporal grain across all datasets in BioTIME is a year. Major taxa and level of measurement: BioTIME includes data from 44,440 species across the plant and animal kingdoms, ranging from plants, plankton and terrestrial invertebrates to small and large vertebrates. Abstract

Motivation: The BioTIME database contains raw data on species identities and abundances in ecological assemblages through time. These data enable users to calculate temporal trends in biodiversity within and amongst assemblages using a broad range of metrics. BioTIME is being developed as a community-led open-source database of biodiversity time series. Our goal is to accelerate and facilitate quantitative analysis of temporal patterns of biodiversity in the Anthropocene. Main types of variables included: The database contains 8,777,413 species abundance records, from assemblages consistently sampled for a minimum of 2 years, which need not necessarily be consecutive. In addition, the database contains metadata relating to sampling methodology and contextual information about each record. Spatial location and grain: BioTIME is a global database of 547,161 unique sampling locations spanning the marine, freshwater and terrestrial realms. Grain size varies across datasets from 0.0000000158 km2 (158 cm2) to 100 km2 (1,000,000,000,000 cm2). Time period and grain: BioTIME records span from 1874 to 2016. The minimal temporal grain across all datasets in BioTIME is a year. Major taxa and level of measurement: BioTIME includes data from 44,440 species across the plant and animal kingdoms, ranging from plants, plankton and terrestrial invertebrates to small and large vertebrates. 47. Investigation of ʟ-amino acid hydroxylases

Abstract

Ferrous-iron/2-oxoglutarate(2-OG)-dependent oxygenases have emerged as a widely distributed family of oxygenases. They have applications both in biocatalysis and as pharmaceutical/agrochemical targets. One of the first subfamilies of 2-OG oxygenases to be studied in detail were the prolyl hydroxylases. The work described in my thesis focused on substrate analogue studies on the proline hydroxylases and on a highly unusual type of 2-OG oxygenase that catalyses the production of ethylene from 2-OG. The work involved the development of assays, in particular mass spectrometry and nuclear magnetic resonance spectroscopy, to screen a panel of substrate analogues. The results identified the need for an α-amino acid for productive catalysis, and were consistent with previous studies showing that the proline hydroxylases can accept different ring sizes, and can catalyse production of dihydroxylated products. New substrates for the proline hydroxylases including bicyclic compounds, which undergo a bifurcating pathway to give singly hydroxylated products, were also identified. In a separate line of investigation, a novel ethylene forming enzyme was studied. Previous work had identified an arginine-oxidising 2-OG oxygenase from Pseudomonas spp. that also produced ethylene. This observation is interesting given the key role of ethylene, as produced by aminocyclopropanecarboxylate (ACC) oxidase, in plant biology. ACC oxidase is structurally related to the 2-OG oxygenases but does not employ 2-OG during catalysis. Methods for the production of the recombinant ethylene-forming enzyme from Pseudomonas spp were developed as were assays for monitoring catalysis. The results reveal that the enzyme produces ethylene in an arginine-dependent manner and coproduces pyrroline-5-carboxylic acid (ʟ-P5C) (the cyclic form of glutamate-5-semialdehyde). Crystal structures were obtained for the ethylene forming enzyme in the presence of a 2-OG analogue with and without arginine. The combined biochemical and structural studies imply that the enzyme has bifurcating pathway, with ethylene production arising from 2-OG and requiring arginine, but which is not oxidised; alternatively arginine can be oxidised to give ∆1-pyrroline-5-carboxylic acid (ʟ-P5C) in a 'normal' 2-OG oxygenase type reaction with concomitant production of succinate. The results open up new possibilities for ethylene production in organisms ranging from bacteria to animals.

50. Statistical aspects of aquaculture research: one-and two-season cross-over designs

Abstract

One-and two-season cross-over designs are compared with completely randomized designs for their potential use in pond aquaculture experiments. Cross-over designs allow treatment comparisons to be made within the same pond, removing much of the between-pond variability. Different treatments are applied consecutively to the same pond. The one-season design involves splitting the season into two periods and applying different treatments in each period. The fish growth in each of the periods is compared taking account of both pond and period differences. The two-season design compares the harvest weight or growth over a whole season in consecutive seasons. Each of the designs is explained further. Data from uniformity trials are used to make these comparisons. Trials on the use of buffalo manure supplemented with urea and triple sulphate, which were completed in consecutive seasons, are used to verify the conclusions. The advantages and disadvantages of each design are discussed.
