Objectives: Data sharing has become a requirement of many funding bodies and is becoming a scientific standard in many disciplines. In medical research, however, data sharing can conflict with clinicians’ obligation to protect patients’ privacy. General recommendations on data sharing exist also for clinical research, but so far lack practical and Swiss-specific aspects. The objective of this document is to provide practical recommendations for all relevant aspects of data sharing in agreement with legislation in Switzerland. Methods: This document was written by members of the Swiss CTU Network, a network of academic clinical trial units. The process did not follow a formalized Delphi process. After an internal consensus round, this report is now published as pre-print for external review. A second version will incorporate external comments. We plan to publish this document as a text in progress, as we expect relevant changes in related fields such as the development of further dedicated medical repositories or methodological advances in anonymization techniques. Results: We developed principles and practical recommendations with respect to informed consent, data management plan, anonymization, data structure and format, coding of variables, metadata and documentation, version control, selection of repository, requesting and use of data. We also provide a summary of legal aspects relevant for the Swiss context. Conclusions: The intension to share data has an impact not only after a clinical trial or an observational study is completed, but also during the planning period, the conduct and the analysis phase. Clinical researchers need to be aware at the beginning of a study on how to inform patients and at least the amount of work related to preparing data for sharing, metadata, and any further documentation. This report provides details of aspects to be considered, suggests decision criteria, and provides examples and checklists, in order to support data sharing in practice.

BACKGROUND Poor recruitment of patients is the predominant reason for early termination of randomized clinical trials (RCTs). Systematic empirical investigations and validation studies of existing recruitment models, however, are lacking. We aim to provide evidence-based guidance on how to predict and monitor recruitment of patients into RCTs. Our specific objectives are the following: (1) to establish a large sample of RCTs (target n = 300) with individual patient recruitment data from a large variety of RCTs, (2) to investigate participant recruitment patterns and study site recruitment patterns and their association with the overall recruitment process, (3) to investigate the validity of a freely available recruitment model, and (4) to develop a user-friendly tool to assist trial investigators in the planning and monitoring of the recruitment process. METHODS Eligible RCTs need to have completed the recruitment process, used a parallel group design, and investigated any healthcare intervention where participants had the free choice to participate. To establish the planned sample of RCTs, we will use our contacts to national and international RCT networks, clinical trial units, and individual trial investigators. From included RCTs, we will collect patient-level information (date of randomization), site-level information (date of trial site activation), and trial-level information (target sample size). We will examine recruitment patterns using recruitment trajectories and stratifications by RCT characteristics. We will investigate associations of early recruitment patterns with overall recruitment by correlation and multivariable regression. To examine the validity of a freely available Bayesian prediction model, we will compare model predictions to collected empirical data of included RCTs. Finally, we will user-test any promising tool using qualitative methods for further tool improvement. DISCUSSION This research will contribute to a better understanding of participant recruitment to RCTs, which could enhance efficiency and reduce the waste of resources in clinical research with a comprehensive, concerted, international effort.

The recent observations of the leptonic mixing angle \theta^PMNS_13 are consistent with \theta^PMNS_13 = \theta_C / \sqrt2 (with \theta_C being the Cabibbo angle \theta^CKM_12). We discuss how this relation can emerge in Grand Unified Theories (GUTs) via charged lepton corrections. The key ingredient is that in GUTs the down-type quark Yukawa matrix and the charged lepton Yukawa matrix are generated from the same set of GUT operators, which implies that the resulting entries are linked and differ only by group theoretical Clebsch factors. This allows a link \theta^e_12 = \theta_C to be established, which can induce \theta^PMNS_13 = \theta_C / \sqrt2 provided that the 1-3 mixing in the neutrino mass matrix is much smaller than \theta_C. We find simple conditions under which \theta^PMNS_13 = \theta_C / \sqrt2 can arise via this link in SU(5) GUTs and Pati-Salam models. We also discuss possible corrections to this relation. Grand Unified Theories (GUTs) offer an attractive framework for flavour models, since they feature relations between quarks and leptons. Combining them with Supersymmetry (SUSY) and flavour symmetries, we derive predictions for the flavour and SUSY flavour structure from various GUT models and discuss how the double missing partner mechanism (DMPM) solution to the doublet-triplet splitting problem can be combined with predictions for GUT scale quark-lepton Yukawa coupling relations. We construct two predictive SUSY SU(5) GUT models with an A4 flavour symmetry, that feature realistic quark-lepton Yukawa coupling ratios and mixing angle relations. These GUT scale predictions arise after GUT symmetry breaking from a novel combination of group theoretical Clebsch-Gordan factors, and we carefully construct additional shaping symmetries and renormalisable messenger sectors to protect the models' predictions from dangerous corrections. The major difference between both models are their respective predictions of a normal and inverse neutrino mass ordering. We perform Markov Chain Monte Carlo analyses, fit to experimental data, and discuss how the models can be tested by present and future experiments. To combine predictive GUT scale quark-lepton Yukawa coupling ratios with the DMPM in SUSY SU(5), we introduce a second GUT breaking Higgs field in the adjoint representation. Two explicit flavour models with different predictions for the GUT scale Yukawa sector are presented, including shaping symmetries and renormalisable messenger sectors, and combined with the DMPM. We calculate the effective masses of the colour triplets mediating proton decay and find that they can be made sufficiently heavy. In SUSY theories, the one-loop SUSY threshold corrections are of particular importance in investigating GUT scale quark-lepton mass relations and thus link a given GUT flavour model to the sparticle spectrum. We calculate the one-loop SUSY threshold corrections of the full MSSM Yukawa coupling matrices in the electroweak-unbroken phase and introduce a new software tool SusyTC as a major extension to the Mathematica package REAP. Finally we find predictions for the CMSSM parameters and sparticle masses from the GUT scale Yukawa coupling ratios used in the flavour models of this thesis.
