The Community Land Model (CLM) is the land component of the Community Earth System Model (CESM) and is used in several global and regional modeling systems. In this paper, we introduce model developments included in CLM version 5 (CLM5), which is the default land component for CESM2. We assess an ensemble of simulations, including prescribed and prognostic vegetation state, multiple forcing data sets, and CLM4, CLM4.5, and CLM5, against a range of metrics including from the International Land Model Benchmarking (ILAMBv2) package. CLM5 includes new and updated processes and parameterizations: (1) dynamic land units, (2) updated parameterizations and structure for hydrology and snow (spatially explicit soil depth, dry surface layer, revised groundwater scheme, revised canopy interception and canopy snow processes, updated fresh snow density, simple firn model, and Model for Scale Adaptive River Transport), (3) plant hydraulics and hydraulic redistribution, (4) revised nitrogen cycling (flexible leaf stoichiometry, leaf N optimization for photosynthesis, and carbon costs for plant nitrogen uptake), (5) global crop model with six crop types and time-evolving irrigated areas and fertilization rates, (6) updated urban building energy, (7) carbon isotopes, and (8) updated stomatal physiology. New optional features include demographically structured dynamic vegetation model (Functionally Assembled Terrestrial Ecosystem Simulator), ozone damage to plants, and fire trace gas emissions coupling to the atmosphere. Conclusive establishment of improvement or degradation of individual variables or metrics is challenged by forcing uncertainty, parametric uncertainty, and model structural complexity, but the multivariate metrics presented here suggest a general broad improvement from CLM4 to CLM5.

7. An Overlap of Anticoagulant-Related and IgA Nephropathy: A Case Report

- Abstract
Introduction:Anticoagulant-related nephropathy (ARN) is an increasingly recognized cause of acute kidney injury (AKI), initially associated with warfarin use. Supratherapeutic warfarin levels were implicated in kidney toxicity. With the widespread adoption of direct oral anticoagulants (DOACs), it becomes imperative to understand their potential risk for ARN and its clinical presentation. Case Presentation:We report a case of a 64-year-old male prescribed DOAC for paroxysmal atrial fibrillation management, presenting with heart failure and worsening AKI. Hematuria and mild proteinuria were also observed. Despite management attempts, AKI persisted, prompting a kidney biopsy. Histopathological examination revealed acute tubular injury with numerous intratubular red blood cell casts consistent with ARN. Additionally, findings indicative of IgA nephropathy (IgAN), including mesangial hypercellularity and IgA dominant deposition, were noted. Conclusion:This case underscores the emerging risk of ARN associated with DOACs and emphasizes the potential exacerbation of ARN in the presence of underlying glomerular diseases such as IgAN. Clinicians should maintain a high index of suspicion for ARN in patients on anticoagulation therapy, particularly DOACs, who present with AKI and urinary abnormalities, as early recognition and intervention are crucial in preventing further renal damage.

White blood cells play diverse roles in innate and adaptive immunity. Genetic association analyses of phenotypic variation in circulating white blood cell (WBC) counts from large samples of otherwise healthy individuals can provide insights into genes and biologic pathways involved in production, differentiation, or clearance of particular WBC lineages (myeloid, lymphoid) and also potentially inform the genetic basis of autoimmune, allergic, and blood diseases. We performed an exome array-based meta-analysis of total WBC and subtype counts (neutrophils, monocytes, lymphocytes, basophils, and eosinophils) in a multi-ancestry discovery and replication sample of ∼157,622 individuals from 25 studies. We identified 16 common variants (8 of which were coding variants) associated with one or more WBC traits, the majority of which are pleiotropically associated with autoimmune diseases. Based on functional annotation, these loci included genes encoding surface markers of myeloid, lymphoid, or hematopoietic stem cell differentiation (CD69, CD33, CD87), transcription factors regulating lineage specification during hematopoiesis (ASXL1, IRF8, IKZF1, JMJD1C, ETS2-PSMG1), and molecules involved in neutrophil clearance/apoptosis (C10orf54, LTA), adhesion (TNXB), or centrosome and microtubule structure/function (KIF9, TUBD1). Together with recent reports of somatic ASXL1 mutations among individuals with idiopathic cytopenias or clonal hematopoiesis of undetermined significance, the identification of a common regulatory 3' UTR variant of ASXL1 suggests that both germline and somatic ASXL1 mutations contribute to lower blood counts in otherwise asymptomatic individuals. These association results shed light on genetic mechanisms that regulate circulating WBC counts and suggest a prominent shared genetic architecture with inflammatory and autoimmune diseases.

Platelet production, maintenance, and clearance are tightly controlled processes indicative of platelets' important roles in hemostasis and thrombosis. Platelets are common targets for primary and secondary prevention of several conditions. They are monitored clinically by complete blood counts, specifically with measurements of platelet count (PLT) and mean platelet volume (MPV). Identifying genetic effects on PLT and MPV can provide mechanistic insights into platelet biology and their role in disease. Therefore, we formed the Blood Cell Consortium (BCX) to perform a large-scale meta-analysis of Exomechip association results for PLT and MPV in 157,293 and 57,617 individuals, respectively. Using the low-frequency/rare coding variant-enriched Exomechip genotyping array, we sought to identify genetic variants associated with PLT and MPV. In addition to confirming 47 known PLT and 20 known MPV associations, we identified 32 PLT and 18 MPV associations not previously observed in the literature across the allele frequency spectrum, including rare large effect (FCER1A), low-frequency (IQGAP2, MAP1A, LY75), and common (ZMIZ2, SMG6, PEAR1, ARFGAP3/PACSIN2) variants. Several variants associated with PLT/MPV (PEAR1, MRVI1, PTGES3) were also associated with platelet reactivity. In concurrent BCX analyses, there was overlap of platelet-associated variants with red (MAP1A, TMPRSS6, ZMIZ2) and white (PEAR1, ZMIZ2, LY75) blood cell traits, suggesting common regulatory pathways with shared genetic architecture among these hematopoietic lineages. Our large-scale Exomechip analyses identified previously undocumented associations with platelet traits and further indicate that several complex quantitative hematological, lipid, and cardiovascular traits share genetic factors.

Red blood cell (RBC) traits are important heritable clinical biomarkers and modifiers of disease severity. To identify coding genetic variants associated with these traits, we conducted meta-analyses of seven RBC phenotypes in 130,273 multi-ethnic individuals from studies genotyped on an exome array. After conditional analyses and replication in 27,480 independent individuals, we identified 16 new RBC variants. We found low-frequency missense variants in MAP1A (rs55707100, minor allele frequency [MAF] = 3.3%, p = 2 × 10(-10) for hemoglobin [HGB]) and HNF4A (rs1800961, MAF = 2.4%, p < 3 × 10(-8) for hematocrit [HCT] and HGB). In African Americans, we identified a nonsense variant in CD36 associated with higher RBC distribution width (rs3211938, MAF = 8.7%, p = 7 × 10(-11)) and showed that it is associated with lower CD36 expression and strong allelic imbalance in ex vivo differentiated human erythroblasts. We also identified a rare missense variant in ALAS2 (rs201062903, MAF = 0.2%) associated with lower mean corpuscular volume and mean corpuscular hemoglobin (p < 8 × 10(-9)). Mendelian mutations in ALAS2 are a cause of sideroblastic anemia and erythropoietic protoporphyria. Gene-based testing highlighted three rare missense variants in PKLR, a gene mutated in Mendelian non-spherocytic hemolytic anemia, associated with HGB and HCT (SKAT p < 8 × 10(-7)). These rare, low-frequency, and common RBC variants showed pleiotropy, being also associated with platelet, white blood cell, and lipid traits. Our association results and functional annotation suggest the involvement of new genes in human erythropoiesis. We also confirm that rare and low-frequency variants play a role in the architecture of complex human traits, although their phenotypic effect is generally smaller than originally anticipated.

The Project for the Intercomparison of Land-Surface Parameterization Schemes (PILPS) aims to improve understanding and modeling of land surface processes. PILPS phase 2(d) uses a set of meteorological and hydrological data spanning 18 yr (1966-83) from a grassland catchment at the Valdai water-balance research site in Russia. A suite of stand-alone simulations is performed by 21 land surface schemes (LSSs) to explore the LSSs' sensitivity to downward longwave radiative forcing, timescales of simulated hydrologic variability, and biases resulting from single-year simulations that use recursive spinup. These simulations are the first in PILPS to investigate the performance of LSSs at a site with a well-defined seasonal snow cover and frozen soil. Considerable model scatter for the control simulations exists. However, nearly all the LSS scatter in simulated root-zone soil moisture is contained within the spatial variability observed inside the catchment. In addition, all models show a considerable sensitivity to longwave forcing for the simulation of the snowpack, which during the spring melt affects runoff, meltwater infiltration, and subsequent evapotranspiration. A greater sensitivity of the ablation, compared to the accumulation, of the winter snowpaek to the choice of snow parameterization is found. Sensitivity simulations starting at prescribed conditions with no spinup demonstrate that the treatment of frozen soil (moisture) processes can affect the long-term variability of the models. The single-year recursive runs show large biases, compared to the corresponding year of the control run, that can persist through the entire year and underscore the importance of performing multiyear simulations.

Bullying has been a source of disquiet, if not moral panic, in recent years. Yet pupil experience outside the classroom has rarely been given the attention it deserves in educational research devoted to the problem. This study examines the social relations between pupils in the lower years of a mixed, multicultural, inner city school. It is based upon long term participant observation as a teacher researcher and aims to develop a sociological appreciation of aggression and bullying during school-day free-time. Part One explains the origins of the research. Recent studies of 'cussing' (verbal abuse), fighting (a topic which has hitherto received very little attention), and bullying are then examined in detail. The research seeks to identify the links, if any, between hostile social relations in school and broader social inequalities at a societal level. Further, it aims to tease out ways in which micro level divisions of power within the pupils' social world shape, and are used by children within interactions. Close attention is therefore given to the meaning, or meanings, of the term 'power'. Models of relative power which inform research focusing upon pupil experience are also identified. In Part Two, both the research site, City School, and the research techniques used are described. Cussing, fighting and bullying, forms of aggressive interaction which distress pupils and obstruct the achievement of curricular goals, are then examined closely. Consideration of gender, 'race' and age grading provides a sharper awareness of underlying power divisions and of how these constrain opportunities for the relatively weak. In Part Three, ways of improving the quality of experience available for pupils during school-day free-time are identified. Whilst the complexity of this task is acknowledged, the study concludes with a renewed sense of optimism about what may be achieved when teachers are more effectively equipped with the skills to understand and, where necessary, make sensitive interventions.

26. The use of the axiophyte species concept to describe the ecological network of the Birmingham and Black Country of the UK West Midlands

The UK conurbation of Birmingham and the Black Country has recently been surveyed for a new Flora, on the basis of a 1 km square grid. The present paper uses the data to describe the ecological network of the conurbation. The total number of taxa per 1 km squares is shown to be moderately but significantly correlated, and the number of native taxa more strongly correlated, with the area of the previously-established network of protected sites. Nevertheless coincidence maps of total numbers or numbers of native species per 1 km square give only poor representations of the ecological network compared with maps of protected sites. Axiophytes are defined as plant species 90% restricted to conservation habitats and recorded in fewer than 25% of 2km × 2km squares in a county. Applying the concept to 1 km squares in Birmingham and the Black Country creates a list of 256 axiophytes. Numbers of axiophytes are shown to be more strongly correlated with areas of protected sites than total taxa or native taxa and a coincidence map of the axiophytes is found to provide a useful quantitative assessment of the ecological network. Maps of axiophytes are used to divide the network into core and linking areas and their use in consolidating and improving the botanical ecological network is explored.

Land models require evaluation in order to understand results and guide future development. Examining functional relationships between model variables can provide insight into the ability of models to capture fundamental processes and aid in minimizing uncertainties or deficiencies in model forcing. This study quantifies the proficiency of land models to appropriately transfer heat from the soil through a snowpack to the atmosphere during the cooling season (Northern Hemisphere: October-March). Using the basic physics of heat diffusion, we investigate the relationship between seasonal amplitudes of soil versus air temperatures due to insulation from seasonal snow. Observations demonstrate the anticipated exponential relationship of attenuated soil temperature amplitude with increasing snow depth and indicate that the marginal influence of snow insulation diminishes beyond an "effective snow depth" of about 50 cm. A snow and heat transfer metric (SHTM) is developed to quantify model skill compared to observations. Land models within the CMIP5 experiment vary widely in SHTM scores, and deficiencies can often be traced to model structural weaknesses. The SHTM value for individual models is stable over 150 years of climate, 1850-2005, indicating that the metric is insensitive to climate forcing and can be used to evaluate each model's representation of the insulation process.

As earth system models (ESMs) become increasingly complex, there is a growing need for comprehensive and multi-faceted evaluation of model projections. To advance understanding of terrestrial biogeochemical processes and their interactions with hydrology and climate under conditions of increasing atmospheric carbon dioxide, new analysis methods are required that use observations to constrain model predictions, inform model development, and identify needed measurements and field experiments. Better representations of biogeochemistryclimate feedbacks and ecosystem processes in these models are essential for reducing the acknowledged substantial uncertainties in 21st century climate change projections.

The author shares his experience of using the Khmer Empire as a substantive content to teach Year 7 students the importance of history. The author argues that historical significance should be explicitly tackled in Key Stage 3 and proposes using the structure of historical research as a framework for curriculum design.
