8. Balancing the ideals of public participation: Discursive legitimation strategies of a disputed practice.

Local authorities in western societies increasingly initiate public participation processes despite criticism of these methods. To understand this development calls for in-depth studies of how the goals and values of public participation are articulated in practice. This paper analyzes the discursive legitimation strategies deployed by civil servants in twelve planning meetings for a participatory process aimed at mitigating violence in a Swedish suburb. The study draws on previous research on discursive legitimation, and presents the context-specific strategies of authorization, de-legitimation, cosmology and addressing of values. The analysis highlights that a core discursive tension in this practice arises from its need to seek mandate in the municipal structure, while the value discourses used to legitimize public participation build on a break from traditional bureaucracy. The study contributes to the broader literature on discursive legitimation in contemporary political contexts, with detailed empirical accounts of how a disputed practice is legitimized in interaction. Denna uppsats har undersökt könsstereotypisering och representationen av kvinnor och män i läromedel i italienska på gymnasieskolan. De analyserade läromedlen till antalet fyra (Prego 1, Ciao 1, Comunicare 1 och Amici 1). De är utgivna av Sveriges största läromedelsförlag och utgår från att läsaren talar svenska (i exempelvis inledningar, grammatikavsnitt, gloslistor och anvisningar). Läromedel har i allmänhet ett stort inflytande att forma elevers föreställningar om vad som är kvinnligt och manligt. Den svenska läroplanen för gymnasieskolan fäster stor vikt vid värden om jämställdhet och möjligheten att eleven själv ska få skapa sin identitet utan att vara bunden till traditionella könsmönster. Studiens frågeställningar syftar till att utforska synligheten av kvinnor och män, representationen av deras givna yrkesroller och hur karaktärerna framställs utifrån genusstereotyper. Metoden som används är både kvantitativ och kvalitativ för att kunna besvara på uppsatsens frågor på ett lämpligt sätt. Resultaten visar att skillnaden mellan manliga och kvinnliga karaktärer vad gäller synlighet är marginell, men männen är alltid i majoritet. Det stämmer även gällande yrken: Män tilldelas fler yrkesroller än kvinnor. Trots det finns det något enstaka normbrytande exempel. Slutligen visar analysen på att läromedlen till en viss del ger uttryck för genusstereotyper.

I kommunen X skolplan står det skrivet att Vägledande samspel är ett arbetsverktyg för ökad måluppfyllelse bland eleverna. Vägledande samspel är ett program vars mål är att öka lärarnas medvetenhet om sina egna roller och om det betydelsefulla samspelet med barnen. Syftet med den här studien var att ta reda på hur lärare och rektorer på två F-6 skolor upplevde Vägledande samspel. Via åtta kvalitativa intervjuer har vi fått svar på hur lärarna och rektorerna upplever Vägledande samspel, hur Vägledande samspel kan tillämpas under skoldagen och vilka begränsningar som upplevs med Vägledande samspel. Respondenterna är alla positivt inställda till Vägledande samspel. De anser att Vägledande samspel genomsyrar hela skoldagen och att det är en hjälp i att stärka barnen i deras utveckling. Samtliga respondenter var överens om att deras förhållningssätt gentemot barnen hade förändrats positivt efter att de genomgått programmet Vägledande samspel.

Vår avsikt med detta arbete har varit att utforma ett arbetsmaterial, en Storyline som vi har kallat ”This is it”, ett komplement till de läromedel som finns i skolan idag. Storyline är en redan beprövad metod i en del skolor och ligger som underlag för detta utvecklingsarbete.För att få insikt i hur erfarna lärare i Storyline ser på detta arbetssätt har vi som vetenskaplig metod valt att genomföra en kvalitativ intervju i kombination med enkäter. Dessa har, tillsammans med teoretiska belägg mynnat ut i ”This is it”. Vi ville undersöka om Storyline som undervisningsmetod skapar förutsättningar för utveckling av ”speciella” kompetenser hos eleverna, kompetenser som vi anser innefattas av eller är en grund till utvecklingen av handlingskompetens. Förmågorna som avses är: kritisk granskning, empati, respekt, tillit, samarbetsförmåga, problemlösning och ansvarstagande. - Abstract
Background: Artemisinin-based combinations currently recommended for treatment of uncomplicated Plasmodium falciparum malaria in many countries of sub-Saharan Africa are substrates of CYP enzymes. The cytochrome enzyme system is responsible for metabolism of about 80-90% of clinically used drugs. It is, therefore, important to obtain the pharmacogenetics of the population in the region with respect to these combinations and thereby enable practitioners to predict treatment outcomes. The aim of this study was to detect and determine allelic frequencies of CYP2C8*2, CYP2C8*3, CYP3A4*1B, CYP3A5*3 and CYP2B6*6 variant alleles in a Tanzanian indigenous population. Methods: Genomic DNA extraction from blood obtained from 256 participants who escorted patients at Karume Health Centre in Mwanza Tanzania, was carried out using the Gene JET (TM) Genomic DNA purification kit (Thermo Scientific). Genotyping for the cytochrome P450 variant alleles was performed using predesigned primers. Amplification was done by PCR while differentiation between alleles was done by restriction fragment length polymorphism (PCR-RFLP) (for CYP2C8*2, CYP2C8*3) and sequencing (for CYP2B6*6, CYP3A5*3 and CYP3A4*1B). Results: CYP2C8*2, CYP2C8*3, CYP3A5*3, CYP3A4*1B and CYP2B6*6 variant allelic frequencies were found to be 19,10,16,78 and 36% respectively. Conclusion: Prevalence of CYP2C8*2, CYP3A5*3, CYP3A4*1B and CYP2B6*6 mutations in a Tanzanian population/ subjects are common. The impact of these point mutations on the metabolism of anti-malarial drugs, particularly artemisinin-based combinations, and their potential drug-drug interactions (DDIs) needs to be further evaluated.

Denna studie behandlar hur frågor kring kusterosion hanterats inom förvaltning i Ystadskommun. Den behandlar även turerna kring den juridiska process som uppstod då Ystadkommun sökte tillstånd att utvinna sand vid Sandhammarens bank för strandfodring viderosionsdrabbade områden. Syftet har varit att utreda vilka mål de olika aktörerna i frågan harhaft, vilka konflikter som uppkommit och hur de har hanterats. Genomförandet av studien hari första hand bestått av en textstudie av material från tillståndsprocessen samt kvalitativaintervjuer med representanter från Länsstyrelsen i Skåne, Ystad kommun och Lunds TekniskaHögskola.Resultaten från studien visar att de olika aktörernas mål är förenliga men ofta otydligtpreciserade. Tydligare mål och bättre samverkan mellan olika aktörer skulle förhindrakonflikter och leda till en effektivare förvaltning av erosionsdrabbade kuster. This study investigates how coastal erosion is being managed at the local government ofYstad. It also evolves the legal process that arose when the local government of Ystad appliedfor permission to extract sediment at the Sandhammaren bank for beach nourishment at thedamaged areas. The purpose of the study has been to unravel the aims of the differentparticipants and how those aims have been managed. The method of the study has mainlybeen textual studies of material in the legal process but has also included qualitativeinterviews with informants representing the county administrative board of Scania, TheMunicipality of Ystad and the Faculty of Engineering at Lund University.The result from the study shows how the different administrative authorities’ aims arecompatible but often unclearly worded. More elaborative aims and better integration amongstthe administrative authorities could lead up to more efficient coastal management.

HIV/AIDS förekommer globalt men få studier fokuserar på hur patienter i Europa och Nordamerika upplever mötet med hälso- och sjukvården. Syftet med denna studie var att belysa hur patienter med HIV/AIDS uppfattar att de blir bemötta inom hälso- och sjukvården i Europa och Nordamerika. En systematisk litteraturgenomgång genomfördes. Det framkom att HIV/AIDS-patienter i dessa delar av världen upplevde stigmatisering i mötet med vården. Detta ledde till ett minskat välbefinnande och lägre grad av tillfredsställelse med hälso- och sjukvården. Fem teman utkristalliserades relaterade till bemötande från hälso- och sjukvårdspersonalen: oprofessionellt bemötande, särbehandling, försummelse, rädsla relaterat till kunskapsbrist samt specialiserade kliniker. Insatser behövs för att säkerställa att dessa patienter får ett respektfullt bemötande och vård på lika villkor. Detta kan förverkligas genom utbyggnad av specialistvården. Vidare forskning inom ämnet kan ta avstamp i denna litteraturstudie.

18. Den gröna staden från koncept till praktik – om hur urban grönstruktur kan bidra till en ekologiskt hållbar stadsutveckling

- Abstract
The establishment of green areas in urban environment for ecological sustainability is a strategy recognized in academics as well as in politics and governance. This thesis investigates how and to what extent various measures and interventions of increased urban green areas and their ecological benefits may contribute to ecological sustainability and improve urban life quality. In addition to a literature review of topic this thesis also contains an applied study of the new construction project “Södra Nyhamnen” in Malmö. The study shows that measures and interventions to increase urban green areas can contribute to locally enhanced biodiversity, possible better air quality, infiltration of storm water and retainment of storm water contaminations in soil instead of polluting of receiving waters. Shading, less heat radiation and wind protection, will also regulate the local climate. The study further shows that measures for urban green areas contributes to improvement of local urban environment, although it should primarily be described as environmental health improvement and only secondly as ecologically sustainable, as the measures consist of reducing or remediating measures with environmental health focus.

Problem: Dagens fragmentariska samhälle har ökat vikten och samtidigt försvårat kommunikationen av organisationsidentitet. Uppsatsen tar ett brett grepp om den problematiken genom att sammanföra förståelsen av tre fält, kommunikation, berättelser och identitet då dessa är tydligt interrelaterade men inte tillräckligt belysta i förhållande till varandra, och särskilt i relation till kommunikationen av organisationsidentitet. Syfte: Att belysa organisationsidentitet ur ett narrativt och kommunikativt perspektiv för att bringa insikter kring kommunikationen av identitet i organisationer och därigenom öka kunskapen om hur berättelser kan användas för att analysera kommunikationen och konstruktion av identitet. Metod: En fallstudie av Svenska institutet, inkluderande kvalitativ dokumentanalys och kvalitativa intervjuer med sju organisationsmedlemmar. Slutsatser: Ett fokus på berättelser ger komplexa insikter kring konstruktionen och kommunikationen av organisationsidentitet och visar hur plurala meningsskapande processer kan belysas på flera nivåer och ur olika perspektiv.

- Abstract
Thyroid hormone (TH) deficiency during development causes severe and permanent neuronal damage, but the primary insult at the tissue level has remained unsolved. We have defined locomotor deficiencies in mice caused by a mutant thyroid hormone receptor alpha 1 (TR alpha 1) with potent aporeceptor activity attributable to reduced affinity to TH. This allowed identification of distinct functions that required either maternal supply of TH during early embryonic development or sufficient innate levels of hormone during late fetal development. In both instances, continued exposure to high levels of TH after birth and throughout life was needed. The hormonal dependencies correlated with severely delayed appearance of parvalbumin-immunoreactive GABAergic interneurons and increased numbers of calretinin-immunoreactive cells in the neocortex. This resulted in reduced numbers of fast spiking interneurons and defects in cortical network activity. The identification of locomotor deficiencies caused by insufficient supply of TH during fetal/perinatal development and their correlation with subtype-specific interneurons suggest a previously unknown basis for the neuronal consequences of endemic cretinism and untreated congenital hypothyroidism, and specifies TR alpha 1 as the receptor isoform mediating these effects.

21. Arbetstagarinflytande vid bolagisering

- Abstract
Denna uppsats behandlar det legala arbetstagarinflytandet i svenska privat-ägda bolag vid en bolagisering. De rättsregler som normalt är tillämpliga vad gäller inflytande i relationen mellan arbetsgivare och arbetstagare presenteras, samt deras tillämplighet i och inför en bolagiseringssituation. Även möjligheterna att göra dessa tillämpliga genom ett mer flexibelt arbetsgivarbegrepp än det formella arbetsgivarbegreppet behandlas. I syfte att ge en något mer nyanserad bild av förutsättningarna för arbetstagarinflytandet ges även exempel på regler som återfinns i folkrättsliga överenskommelser och medbestämmandeavtal. Regler som i vissa fall ligger utanför området av gällande svensk arbetsrätt. De svenska lagar och regler som ger arbetstagare rätt till inflytande grundar sig i de flesta fall på rättigheter och skyldigheter för den formella arbetsgivaren å ena sidan och arbetstagarorganisation med vilken denne har tecknat kollektivavtal å den andra. Arbetsgivare är den fysiska eller juridiska person hos vilken en arbetstagare är anställd, vilket ska anges i ingånget anställningsavtal. Om det i praktiken är ett annat bolag som fattar besluten finns det i princip ingen rätt att förhandla med detta. Det finns dock exempel på fall då ett moderbolag gjorts ansvarig efter att ha fattat beslut om att utlämna arbete, för sin egen räkning, via ett kommissionärsföretag utan att först förhandla med sina anställda. Ett undantag till att rätten för inflytande och information stannar vid bolagsgränsen finns i rätten till styrelserepresentation, vilken kan ge arbetstagarna rätt till representation i moderbolagets styrelse. Detta förutsätter dock att moderbolaget är svenskt. Arbetstagarrepresentanter omfattas av tystnadsplikt i känsliga frågor, en tystnadsplikt som även gäller gentemot arbetstagarorganisationen de representerar. 22. Hypothyroidism caused by a mutant thyroid hormone receptor alpha1 : consequences for development and physiology

- Abstract
Lack of sufficient thyroid hormone during pregnancy and early postnatal development results in profound mental retardation and motor deficits whereas altered thyroid status in the adult is associated with disturbed metabolic homeostasis and impaired cardiac function. Thyroid hormone mediates its effects through the distinct thyroid hormone receptors (TR) TRalpha1 and TRbeta1-2, which are ligand-modulated transcription factors that regulate gene expression both in the presence and absence of hormone. Mutations in the TRbeta gene that decrease affinity to ligand are associated with the well known syndrome Resistance to thyroid hormone (RTH), which is characterized by elevated circulating levels of thyroid hormone and a mixed hyper- and hypothyroid phenotype. However, no patient with a corresponding mutation in the TRalpha gene has been found. In this thesis, I describe the developmental and physiological consequences of a point mutation introduced into the mouse TRalpha1 gene. The mutation, originally identified in TRbeta (R438C) of RTH patients, reduces affinity for T3 10-fold, and causes the receptor to act as an aporeceptor unless challenged with high levels of thyroid hormone. We report that mice heterozygous for this mutation (TRalpha1+m mice) exhibit locomotor dysfunctions caused by insufficient supply of thyroid hormone during fetal/postnatal development. Furthermore, treatment with thyroid hormone during specific stages of development ameliorated these deficiencies. The locomotor dysfunctions correlated with delayed or perturbed development of several brain regions. Notably, we report that specific GABAergic cells in the neocortex were affected: the appearance of parvalbumin-immunoreactive GABAergic interneurons was severely delayed whereas the numbers of calretinin-immunoreactive cells were increased. Thyroid hormone (TH) deficiency during development causes severe and permanent neuronal damage, but the primary insult at the tissue level has remained unsolved. We have defined locomotor deficiencies in mice caused by a mutant thyroid hormone receptor alpha1 (TR alpha1) with potent aporeceptor activity attributable to reduced affinity to TH. This allowed identification of distinct functions that required either maternal supply of TH during early embryonic development or sufficient innate levels of hormone during late fetal development. In both instances, continued exposure to high levels of TH after birth and throughout life was needed. The hormonal dependencies correlated with severely delayed appearance of parvalbumin-immunoreactive GABAergic interneurons and increased numbers of calretinin-immunoreactive cells in the neocortex. This resulted in reduced numbers of fast spiking interneurons and defects in cortical network activity. The identification of locomotor deficiencies caused by insufficient supply of TH during fetal/perinatal development and their correlation with subtype-specific interneurons suggest a previously unknown basis for the neuronal consequences of endemic cretinism and untreated congenital hypothyroidism, and specifies TR alpha1 as the receptor isoform mediating these effects.

- Abstract
In this study, a significant increase by 50% in intracellular free calcium concentration ([Ca(2+)](i)) was observed in differentiated human neuroblastoma (SH-SY5Y) cells after exposure to 0.25microM of the fungal metabolite gliotoxin for 72h. Further, the involvement of caspases and calpains was demonstrated to underlie the gliotoxin-induced cytotoxic and neurite degenerative effects. The caspase inhibitor Z-VAD-fmk almost completely reduced the neurite degeneration from 40% degeneration of neurites to 5% as compared to control. Inhibition of calpains with calpeptin significantly attenuated gliotoxin-induced cytotoxicity, determined as reduction in total cellular protein content, from 43% to 14% as compared to control cells. Western blot analyses of alphaII-spectrin breakdown fragments confirmed activity of the proteases, and that alphaII-spectrin was cleaved by caspases in gliotoxin-exposed cells. These results show that calpains and caspases have a role in the toxicity of gliotoxin in differentiated SH-SY5Y cells and that the process may be Ca(2+)-mediated.

Abstract: The profound effects of thyroid hormone (TH) on heart development and function are mediated by the thyroid hormone receptors (TR) α1 and β1. While numerous patients with TRβ1 mutations have been identified, patients with similar mutations in TRα1 are yet to be discovered. Recently generated heterozygous mice with a dominant negative mutation in TRα1 (TRα1+/m mice) have normal TH levels, which may have hampered the discovery of patients with such mutations. We now measure intracellular Ca2+ and contraction in cardiomyocytes isolated from TRα1+/m mice and wildtype littermates (WT). TRα1+/m cardiomyocytes showed a phenotype similar to that in hypothyroidism with significant slowing of voltage-activated Ca2+ transients and contractions. Increased stimulation frequency (from 0.5 to 3 Hz) or β-adrenergic stimulation reduced the differences between TRα1+/m and WT cardiomyocytes. However, in TRα1+/m cells stimulation at 3 Hz gave a marked increase in diastolic Ca2+ and β-adrenergic stimulation triggered spontaneous Ca2+ release events during relaxation. Both TRα1+/m and WT cardiomyocytes responded to TH treatment by displaying a “hyperthyroid” phenotype with faster and larger Ca2+ transients and contractions. Excised TRα1+/m hearts showed an increased expression of phospholamban (PLB). In conclusion, isolated TRα1+/m cardiomyocytes display major dysfunctions with marked slowing of the Ca2+ transients and contractions. [Copyright &y& Elsevier]

- Abstract
Thyroid hormone, via its nuclear receptors TRalpha and TRbeta, controls metabolism by acting locally in peripheral tissues and centrally by regulating sympathetic signaling. We have defined aporeceptor regulation of metabolism by using mice heterozygous for a mutant TRalpha1 with low affinity to T3. The animals were hypermetabolic, showing strongly reduced fat depots, hyperphagia and resistance to diet-induced obesity accompanied by induction of genes involved in glucose handling and fatty acid metabolism in liver and adipose tissues. Increased lipid mobilization and beta-oxidation occurred in adipose tissues, whereas blockade of sympathetic signaling to brown adipose tissue normalized the metabolic phenotype despite a continued perturbed hormone signaling in this cell type. The results define a novel and important role for the TRalpha1 aporeceptor in governing metabolic homeostasis. Furthermore, the data demonstrate that a nuclear hormone receptor affecting sympathetic signaling can override its autonomous effects in peripheral tissues.
