Abstract The aim of the current study was to investigate the effects of ingesting different dosages of caffeine (CAF) prior to plyometric jump training (PJT) on sport-related performance and physiological parameters in male basketball players. Twenty-four young athletes were randomly divided into 3 groups and performed 6 weeks of PJT while consuming 3 mg·kg–1 of body mass caffeine (CAF3, n = 8), 6 mg·kg–1 body mass caffeine (CAF6, n = 8) or placebo (PL; n = 8) one hour prior to each training session. Before and after the 6-week PJT, the players were evaluated for field-based basketball-specific performance measures (vertical jump, 20-m sprint, Illinois change of direction speed [CODS], and maximal strength) and lab-based physiological (aerobic capacity and anaerobic power) parameters. CAF3, CAF6, and PL groups demonstrated significant improvements in vertical jump (ES = 1.07, 1.45, and 1.1, respectively), 20-m sprint (ES = – 0.50, – 0.61, and – 0.36), change of direction performance (ES = – 1.22, – 1.26, and – 1.09), maximal strength (ES = 1.68, 2.29, and 1.17), maximum oxygen uptake (V̇O2max) (ES = 1.09, 1.59, and 0.92), and peak (ES = 1.82, 1.85, and 0.82) and average power output (ES = 1.39, 1.32, and 1.07) after 6 weeks of training. Comparative analysis of individual adaptive responses to training indicated that the CAF6 led to insignificantly greater effects in vertical jump (ES = 1.45), maximal strength (ES = 2.29), and V̇O2max (ES = 1.59) with lower residuals in individual changes and lower coefficient of variations (CV) in mean group changes. Regarding sprint and CODS performance, both experimental groups indicated similar changes, residuals in individual changes, and CVs in mean group changes. Overall, consuming 6 mg·kg–1 body mass caffeine induces superior adaptations in aerobic fitness, anaerobic power, and sport-specific performance measures, with lower inter-individual variability in the adaptations and more homogenized changes over the training period.

The Sichuan-Xizang Railway is a global challenge, surpassing other known railway projects in terms of geological and topographical complexity. This paper presents an approach for rapidly profiling rock mass quality underneath tunnel face for the ongoing construction of the Sichuan-Xizang Railway. It adopts the time-series method and carries out the quantitative analysis of the rock mass quality using the depth-series measurement-while-drilling (MWD) data associated with drilling of blastholes. A tunnel face with 15 blastholes is examined for illustration. The results include identification of the boundary of homogeneous geomaterial by plotting the blasthole depth against the net drilling time, as well as quantification of rock mass quality through the recalculation of the new specific energy. The new specific energy profile is compared and highly consistent with laboratory test, manual logging and tunnel seismic prediction results. This consistency can enhance the blasthole pattern design and facilitate the dynamic determination of charge placement and amount. This paper highlights the importance of digital monitoring during blasthole drilling for rapidly profiling rock mass quality underneath and ahead of tunnel face. It upgrades the MWD technique for rapid profiling rock mass quality in drilling and blasting tunnels.

Rock and geotechnical engineering investigations involve drilling holes in ground with or without retrieving soil and rock samples to construct the subsurface ground profile. On the basis of an actual soil nailing drilling for a slope stability project in Hong Kong, this paper further develops the drilling process monitoring (DPM) method for digitally profiling the subsurface geomaterials of weathered granitic rocks using a compressed airflow driven percussive-rotary drilling machine with down-the-hole (DTH) hammer. Seven transducers are installed on the drilling machine and record the chuck displacement, DTH rotational speed, and five pressures from five compressed airflows in real-time series. The mechanism and operations of the drilling machine are elaborated in detail, which is essential for understanding and evaluating the drilling data. A MATLAB program is developed to automatically filter the recorded drilling data in time series and classify them into different drilling processes in sub-time series. These processes include penetration, push-in with or without rod, pull-back with or without rod, rod-tightening and rod-untightening. The drilling data are further reconstructed to plot the curve of drill-bit depth versus the net drilling time along each of the six drillholes. Each curve is found to contain multiple linear segments with a constant penetration rate, which implies a zone of homogenous geomaterial with different weathering grades. The effect from fluctuation of the applied pressures is evaluated quantitatively. Detailed analyses are presented for accurately assess and verify the underground profiling and strength in weathered granitic rock, which provided the basis of using DPM method to confidently assess drilling measurements to interpret the subsurface profile in real time.

Abstract Saturn Kilometric Radiation (SKR), being the dominant radio emission at Saturn, has been extensively investigated. The low‐frequency extension of SKR is of particular interest due to its strong association with Saturn's magnetospheric dynamics. However, the highly anisotropic beaming of SKR poses challenges for observations. In most cases, the propagation of SKR is assumed to follow straight‐line paths. We explore the propagation characteristics of SKR across different frequencies in this study. An extended equatorial shadow region for low‐frequency SKR is identified, resulting from the merging of the Enceladus plasma torus and the previously known equatorial shadow zone. Ray‐tracing simulations reveal that low‐frequency (≲100 kHz) SKR is unable to enter the shadow region and is instead reflected toward high latitudes. In contrast, high‐frequency SKR (≳100 kHz) generally propagates without hindrance. Observations suggest that some low‐frequency SKR can enter the shadow region through reflection by the magnetosheath or leakage from the plasma torus.

Periodontitis, a common chronic inflammatory disease, is characterized by T cell responses leading to gradual alveolar bone loss and eventual tooth mobility or loss. Herein, polylysine-derived carbon quantum dots (PLL-CQDs) are synthesized via the pyrolysis method to facilitate the recovery of periodontal bone loss induced by periodontitis by modulating T cell phenotype. The PLL-CQDs, characterized by small particle size (2.31 ± 0.70 nm) and fluorescence properties, preserved the positive charge of PLL, affirming successful preparation. In vitro studies revealed that PLL-CQDs significantly increased T cell proliferation in a concentration-dependent manner, while effectively suppressing differentiation into CD4+ phenotype. Furthermore, they promoted alkaline phosphatase expression and calcium nodule formation in MC3T3-E1 cells, indicating potential for periodontal bone regeneration. In vivo experiments demonstrated that PLL-CQDs mitigated alveolar bone resorption and modulated periodontal tissue inflammation. Overall, PLL-CQDs exhibited the ability to regulate T cell differentiation and expedite periodontal bone healing, offering promising implications for periodontitis treatment and suggesting novel strategies for addressing immune-associated bone diseases using CQDs.

Abstract Using the high‐time‐resolution data from the Magnetospheric Multiscale mission, precursor waves upstream of foreshock transient (FT) shocks are statistically investigated using the four‐spacecraft timing method. The wave frequencies and wave vectors determined in the plasma rest frame (PRF) are shown to follow the cold plasma dispersion relation for whistler waves. Combining with the feature of the right‐hand polarization in the PRF, the precursors are identified as whistler‐mode waves around the lower hybrid frequency. The occurrence of whistler precursors is independent of the Alfvén Mach number and the FT shock normal angle. More importantly, all the whistler precursors have group velocities pointing upstream in the shock frame, suggesting the dispersive radiation to be a possible generation mechanism. The study improves the understanding of not only the whistler precursors but also the overall FT shock dynamics.

Abstract The emergence of antibacterial resistance (ABR) is an urgent and complex public health challenge worldwide. Antibiotic resistant genes (ARGs) are considered as a new pollutant by the WHO because of their wide distribution and emerging prevalence. The role of environmental factors in developing ARGs in bacterial populations is still poorly understood. Therefore, the relationship between environmental factors and bacteria should be explored to combat ABR and propose more tailored solutions in a specific region. Here, we collected and analyzed surface water samples from Yangtze Delta, China during 2021, and assessed the nonlinear association of environmental factors with ARGs through a sigmoid model. A high abundance of ARGs was detected. Amoxicillin, phosphorus (P), chromium (Cr), manganese (Mn), calcium (Ca), and strontium (Sr) were found to be strongly associated with ARGs and identified as potential key contributors to ARG detection. Our findings suggest that the suppression of ARGs may be achieved by decreasing the concentration of phosphorus in surface water. Additionally, Group 2A light metals (e.g., magnesium and calcium) may be candidates for the development of eco-friendly reagents for controlling antibiotic resistance in the future.

Abstract Patients with chronic obstructive pulmonary disease (COPD) who exhibit elevated blood eosinophil levels often experience worsened lung function and more severe emphysema. This implies the potential involvement of eosinophils in the development of emphysema. However, the precise mechanisms underlying the development of eosinophil-mediated emphysema remain unclear. In this study, we employed single-cell RNA sequencing to identify eosinophil subgroups in mouse models of asthma and emphysema, followed by functional analyses of these subgroups. Assessment of accumulated eosinophils unveiled distinct transcriptomes in the lungs of mice with elastase-induced emphysema and ovalbumin-induced asthma. Depletion of eosinophils through the use of anti-interleukin-5 antibodies ameliorated elastase-induced emphysema. A particularly noteworthy discovery is that eosinophil-derived cathepsin L contributed to the degradation of the extracellular matrix, thereby leading to emphysema in pulmonary tissue. Inhibition of cathepsin L resulted in a reduction of elastase-induced emphysema in a mouse model. Importantly, eosinophil levels correlated positively with serum cathepsin L levels, which were higher in emphysema patients than those without emphysema. Expression of cathepsin L in eosinophils demonstrated a direct association with lung emphysema in COPD patients. Collectively, these findings underscore the significant role of eosinophil-derived cathepsin L in extracellular matrix degradation and remodeling, and its relevance to emphysema in COPD patients. Consequently, targeting eosinophil-derived cathepsin L could potentially offer a therapeutic avenue for emphysema patients. Further investigations are warranted to explore therapeutic strategies targeting cathepsin L in emphysema patients.

Random variables introduced in modelling of seismic engineering are often the result of cognitive limitations and the unpredictability of structures, leading to uncertainties in the field. A practical method for dealing with them is to develop sensitivity analysis in the framework of data and probability statistics. Of existing non-structural components, cable tray systems are characterized by a number of uncertainties which may influence their bearing capacity drastically. In this research, the main characteristics of material, geometry, member layout along with the connection stiffness in cable tray are considered as random variables using global sensitivity analysis, with their results relative importance of these potential uncertainties on the seismic performance of cable tray. The sensitivity analysis method developed especially for cable tray under seismic excitation is constructed based on modal analysis and equivalent inertia force method combined with the Latin hypercube sampling method. The final results demonstrate the need to consider the effects of random variables in modeling assumption in seismic performance analyses of cable tray and can be further used in optimization design.

Objective Using transcriptome sequencing, we screened differentially expressed transcription factors (TFs) in keratinocyte (KC) with overexpression of the epidermal growth factor receptor (EGFR) induced by recombinant human parathyroid hormone (1-34) in order to investigate the mechanism of rhPTH(1-34) in the treatment of psoriasis. Methods Overexpression of EGFR in HaCaT cells was achieved by lentiviral transfection. Cells in rhPTH (1-34) group were incubated with 0.01 μmol/L of rhPTH (1-34), while the control group were treated with DMEM solution alone. After extraction of RNA, the quantity and quality of RNA were assessed using UV absorbance assay and denaturing agarose gel electrophoresis, respectively. The cDNA was synthesized and amplified by real-time quantitative PCR for accurate quantification of the effective concentration of the library. Abstract Inorganic sulfide solid-state electrolytes, especially Li6PS5X (X = Cl, Br, I), are considered viable materials for developing all-solid-state batteries because of their high ionic conductivity and low cost. However, this class of solid-state electrolytes suffers from structural and chemical instability in humid air environments and a lack of compatibility with layered oxide positive electrode active materials. To circumvent these issues, here, we propose Li6+xMxAs1-xS5I (M=Si, Sn) as sulfide solid electrolytes. When the Li6+xSixAs1-xS5I (x = 0.8) is tested in combination with a Li-In negative electrode and Ti2S-based positive electrode at 30 °C and 30 MPa, the Li-ion lab-scale Swagelok cells demonstrate long cycle life of almost 62500 cycles at 2.44 mA cm−2, decent power performance (up to 24.45 mA cm−2) and areal capacity of 9.26 mAh cm−2 at 0.53 mA cm−2.

Underwater imagery plays a vital role in ocean development and conservation efforts. However, underwater images often suffer from chromatic aberration and low contrast due to the attenuation and scattering of visible light in the complex medium of water. To address these issues, we propose an underwater image enhancement network called CM-Net, which utilizes color mapping techniques to remove noise and restore the natural brightness and colors of underwater images. Specifically, CM-Net consists of a three-step solution: adaptive color mapping (ACM), local enhancement (LE), and global generation (GG). Inspired by the principles of color gamut mapping, the ACM enhances the network’s adaptive response to regions with severe color attenuation. ACM enables the correction of the blue-green cast in underwater images by combining color constancy theory with the power of convolutional neural networks. To account for inconsistent attenuation in different channels and spatial regions, we designed a multi-head reinforcement module (MHR) in the LE step. The MHR enhances the network’s attention to channels and spatial regions with more pronounced attenuation, further improving contrast and saturation. Compared to the best candidate models on the EUVP and UIEB datasets, CM-Net improves PSNR by 18.1% and 6.5% and SSIM by 5.9% and 13.3%, respectively. At the same time, CIEDE2000 decreased by 25.6% and 1.3%.

Fibrosis, a pathological alteration of the repair response, involves continuous organ damage, scar formation, and eventual functional failure in various chronic inflammatory disorders. Unfortunately, clinical practice offers limited treatment strategies, leading to high mortality rates in chronic diseases. As part of investigations into gaseous mediators, or gasotransmitters, including nitric oxide (NO), carbon monoxide (CO), and hydrogen sulfide (H2S), numerous studies have confirmed their beneficial roles in attenuating fibrosis. Their therapeutic mechanisms, which involve inhibiting oxidative stress, inflammation, apoptosis, and proliferation, have been increasingly elucidated. Additionally, novel gasotransmitters like hydrogen (H2) and sulfur dioxide (SO2) have emerged as promising options for fibrosis treatment. In this review, we primarily demonstrate and summarize the protective and therapeutic effects of gaseous mediators in the process of fibrosis, with a focus on elucidating the underlying molecular mechanisms involved in combating fibrosis.

Abstract Background Tooth development, as one of the major mineralized tissues in the body, require fine-tuning of mineralization micro-environment. The interaction between dental epithelium and mesenchyme plays a decisive role in this process. With epithelium–mesenchyme dissociation study, we found interesting expression pattern of insulin-like growth factor binding protein 3 (IGFBP3) in response to disruption of dental epithelium–mesenchyme interaction. Its action and related mechanisms as regulator of mineralization micro-environment during tooth development are investigated. Results Expressions of osteogenic markers at early stage of tooth development are significantly lower than those at later stage. BMP2 treatment further confirmed a high mineralization micro-environment is disruptive at early stage, but beneficial at later stage of tooth development. In contrast, IGFBP3's expression increased gradually from E14.5, peaked at P5, and decreased afterwards, demonstrating an inverse correlation with osteogenic markers. RNA-Seq and Co-immunoprecipitation showed that IGFBP3 regulates the Wnt/beta-catenin signaling pathway activity by enhancing DKK1 expression and direct protein–protein interaction. The suppression of the mineralization microenvironment effectuated by IGFBP3 could be reversed by the DKK1 inhibitor WAY-262611, further demonstrating that IGFBP3 exerted its influence via DKK1. Conclusion A deeper understanding of tooth development mechanisms is essential for tooth regeneration, which have great implications for dental care. The current study demonstrated that the IGFBP3 expression is regulated in accordance with the needs of the mineralization microenvironment during tooth development, and IGFBP3 exerts its modulating action on osteogenic/odontogenic differentiation of hDPSCs by DKK1-Wnt/ beta-catenin axis.

Abstract The features of electron pitch angle distributions (PADs) often imply different physical mechanisms in planetary magnetospheres. We report a simultaneous equatorial electrostatic electron cyclotron harmonic (ECH) wave event with butterfly PADs of electrons at L ∼ 7.6–9 observed by the Cassini spacecraft. Via calculating the bounce‐averaged electron diffusion rates, we found that Saturnian ECH waves can resonate with ∼10 eV to several keV electrons at

Single-cell sequencing technologies have revolutionised the life sciences and biomedical research. Single-cell sequencing provides high-resolution data on cell heterogeneity, allowing high-fidelity cell type identification, and lineage tracking. Computational algorithms and mathematical models have been developed to make sense of the data, compensate for errors and simulate the biological processes, which has led to breakthroughs in our understanding of cell differentiation, cell-fate determination and tissue cell composition. The development of long-read (a.k.a. third-generation) sequencing technologies has produced powerful tools for investigating alternative splicing, isoform expression (at the RNA level), genome assembly and the detection of complex structural variants (at the DNA level).In this review, we provide an overview of the recent advancements in single-cell and long-read sequencing technologies, with a particular focus on the computational algorithms that help in correcting, analysing, and interpreting the resulting data. Additionally, we review some mathematical models that use single-cell and long-read sequencing data to study cell-fate determination and alternative splicing, respectively. Moreover, we highlight the emerging opportunities in modelling cell-fate determination that result from the combination of single-cell and long-read sequencing technologies.

Abstract Objective: This study aimed to develop a pro-angiogenic hydrogel with in situ gelation ability for alveolar bone defects repair. Methodology: Silk fibroin was chemically modified by Glycidyl Methacrylate (GMA), which was evaluated by proton nuclear magnetic resonance (1H-NMR). Then, the photo-crosslinking ability of the modified silk fibroin was assessed. Scratch and transwell-based migration assays were conducted to investigate the effect of the photo-crosslinked silk fibroin hydrogel on the migration of human umbilical vein endothelial cells (HUVECs). In vitro angiogenesis was conducted to examine whether the photo-crosslinked silk fibroin hydrogel would affect the tube formation ability of HUVECs. Finally, subcutaneous implantation experiments were conducted to further examine the pro-angiogenic ability of the photo-crosslinked silk fibroin hydrogel, in which the CD31 and α-smooth muscle actin (α-SMA) were stained to assess neovascularization. The tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) were also stained to evaluate inflammatory responses after implantation. Results: GMA successfully modified the silk fibroin, which we verified by our 1H-NMR and in vitro photo-crosslinking experiment. Scratch and transwell-based migration assays proved that the photo-crosslinked silk fibroin hydrogel promoted HUVEC migration. The hydrogel also enhanced the tube formation of HUVECs in similar rates to Matrigel®. After subcutaneous implantation in rats for one week, the hydrogel enhanced neovascularization without triggering inflammatory responses. Conclusion: This study found that photo-crosslinked silk fibroin hydrogel showed pro-angiogenic and inflammation inhibitory abilities. Its photo-crosslinking ability makes it suitable for matching irregular alveolar bone defects. Thus, the photo-crosslinkable silk fibroin-derived hydrogel is a potential candidate for constructing scaffolds for alveolar bone regeneration.

Background and purposeBlood markers have important value in the diagnosis of depressive disorders. Serum alkaline phosphatase (ALP) not only predicts stroke recurrence and poor functional prognosis in cerebrovascular disease (CVD) patients but also increases significantly in middle-aged women with depression. Thus, it has not been reported whether serum ALP is associated with the development of depression and/or vascular depression (VDe) in CVD patients.MethodsThis was a cross-sectional study of 353 CVD patients (stroke patients, n = 291; cerebral small vessel disease (CSVD) patients, n = 62). Baseline demographic information, fasting blood markers (such as blood counts, liver function, kidney function and lipids), and brain CT/MRI scans were collected. CVD patients were divided into non-depression, suspected vascular depression (SVD), and positive vascular depression (PVD) groups according to their Hamilton Rating Scale for Depression (HAMD) scores. Univariate analysis of baseline data, blood markers, and the prevalence of lesions (> 1.5 cm) was performed. Subsequently, the diagnostic performance of the univariate and combined variables for SVD and PVD was analyzed using binary logistic regression. The diagnostic value of the multivariate model for VDe was analyzed by ordinal logistic regression.Results(1) Serum ALP (p = 0.003) and hypersensitive C-reactive protein (hs-CRP, p = 0.001) concentrations increased as HAMD scores increased, and the prevalence of brain atrophy (p = 0.016) and lesions in the basal ganglia (p = 0.001) and parietal (p = 0.001), temporal (p = 0.002), and frontal lobes (p = 0.003) also increased, whereas the concentrations of hemoglobin (Hb, p = 0.003), cholinesterase (ChE, p = 0.001), and high-density lipoprotein cholesterol (HDL-C, p = 0.005) declined. Objective To assess the clinical efficacy and safety of sequential therapy for nodular cystic acne. Methods Forty three patients with nodular cystic acne were divided into sequential and control groups. In the sequential group, 22 patients were treated with ALA-PDT every two weeks for 3 times, and oral antibiotics in the first stage, followed by chemical peeling once every two weeks for 3 times, and oral isotretinoin in the second stage. Finally, oral isotretinoin alone was given in the last stage. While 21 patients in the control group were treated with isotretinoin alone. The treatments lasted for 16 weeks. After each treatment course, the efficacy and adverse effect were compared between the two therapies. The patients in both groups were followed up for 6 months, and the relapse rate and adverse reactions were recorded. Results In the first stage of treatment, effective rates after the second and the third treatment course were 9.09% and 22.73%, respectively, in the sequential group, and 0 and 4.76%, respectively, in the controls. In the second stage, the effective rates after each treatment course were higher in sequential groups than in the controls (1st treatment course: 50.00% vs. 23.81%; 2nd treatment course: 90.90% vs. 42.86%, and the 3rd treatment course: 100% vs. 66.67%. Fast ion conductor materials screening based on high-throughput calculations involves enormous computing tasks. The process usually includes structural optimization, energy calculation, charge analysis and ionic migration performance estimation. The first one involves looking for the equilibrium atomic positions in huge amount of candidate compounds or derivative structures, and the computational cost is always high because of the task-intensive features. The last one relates to the kinetic problems, for which the time-consuming transition state theory and the molecular dynamics are the main simulation methods. In this work, two predictive models, ionic migration activation energy model and structural optimization model, are developed based on machine learning (ML) techniques to accelerate the process of estimating activation energy and relaxing the doped crystal structures, respectively. By training 3136 energy barrier data calculated by bond valence (BV) method, an ionic migration activation energy model (Ea model) with mean absolute error (MAE) of 0.26 eV on testing data set is obtained. We apply this model and filter LiBiOS as a promising fast Li+ conductor from 49 Li-containing hetero-anionic compounds. Although the model-predicted result shows relatively low energy barrier, further analysis indicates that the high carrier formation energy restricts the ionic transportability. Therefore, we substitute fractional Li+ with Mg2+ in LiBiOS to relieve the large difficulty of forming carriers in the structure. In order to fast explore the optimal doping scheme, we develop the structural optimization model (E-f model) containing the ML-based energy and force prediction to accelerate the structural optimization under various LiMg ratio and doping configurations. Decent doping scheme Li1-2xMgxBiOS (x = 0.1875) shows much better Li+ migration performance compared with LiBiOS without substitution. This method of screening fast ion conductor materials and finding optimal doping scheme will extremely accelerate materials explorations.

ABSTRACT: The response of basilar arteries to noradrenaline varies among many animal species, but remains little studied in poultry. Accordingly, we aimed to characterize the adrenergic receptor (AR) subtypes that modulate vascular response in basilar arteries in the chicken, with isometric recording of arterial ring tension using an organ bath. We demonstrated the presence of both alpha and beta (α and β) receptor subtypes through evaluating the response to noradrenaline, with and without a range of β-AR and α-AR antagonists. The concentration-dependent relaxations then induced by a range of β-AR agonists indicated a potency ranking of isoproterenol > noradrenaline > adrenaline > procaterol. We then investigated the effects of β-AR antagonists that attenuate the effect of isoproterenol (propranolol for β1,2,3-ARs, atenolol for β1-ARs, butoxamine for β2-ARs, and SR 59230A for β3-ARs), with Schild regression analysis, ascertaining multiple β-AR subtypes, with neither the β1-AR nor the β2-AR as the dominant subtype. SR 59230A was the only antagonist to yield a pA2 value (7.52) close to the reported equivalent for the relevant receptor subtype. Furthermore, treatment with SR 58611 (a β3-AR agonist) induced relaxation, which was inhibited (P < 0.01) by L-NNA and SR 59230A. Additionally, treating basilar arterial strips (containing endothelium) with SR 58611 induced nitric oxide (NO) production, which was inhibited (P < 0.01) by L-NNA and SR 59230A. Based on this first characterization of AR subtypes in chicken basilar arteries (to our knowledge), we suggest that α- and β-ARs are involved in contraction and relaxation, and that β3-ARs, especially those on the endothelium, may play an important role in vasodilation via NO release.

Lung cancer is one of the most commonly diagnosed cancers worldwide. Although cisplatin-based chemotherapy regimens serve a pivotal role in non-small cell lung cancer (NSCLC) treatment, drug resistance and serious side effects limited its further clinical application. Regorafenib, a small-molecule multi-kinase inhibitor, was demonstrated to have promising anti-tumor activity in various solid tumors. In the present study, we found that regorafenib markedly enhanced cisplatin-induced cytotoxicity in lung cancer cells by activating reactive oxygen species (ROS)-mediated endoplasmic reticulum stress (ER Stress), c-Jun N-terminal kinase (JNK) and p38 mitogen-activated protein kinase (MAPK) signaling pathways. Regorafenib increased ROS generation by promoting NADPH oxidase 5 (NOX5) expression, and knocking down NOX5 attenuated ROS-mediated cytotoxicity of regorafenib in lung cancer cells. Additionally, mice xenograft model validated that synergistic anti-tumor effects of combined treatment with regorafenib and cisplatin. Our results suggested that combination therapy with regorafenib and cisplatin may serve as a potential therapeutic strategy for some NSCLC patients.

Vasoreactivity is relatively well documented in terrestrial snakes but has previously been investigated in only one semi-arboreal snake species. Consequently, the extent to which vasoreactivity is common across snake taxa or varies by habitat is unclear. The Tokara habu (Protobothrops tokarensis) is a semi-arboreal snake endemic to only two small adjacent Japanese islands, and hence a useful species for further investigation of vasoreactivity. We evaluated responses to known vasoactive substances in thoracic aortas isolated from Tokara habu. Under resting tension, noradrenaline and angiotensin II induced concentration-dependent contraction, but acetylcholine, serotonin (5-hydroxytriptamine; 5-HT), and isoproterenol induced relaxation followed by contraction. Histamine and rattlesnake bradykinin had no effect. Experiments with receptor-specific antagonists suggest that M1 and M3 receptors are involved in the acetylcholine-induced response; 5-HT1, 5-HT2, and 5-HT7 receptors in the serotonin-induced response; and β1 and β2 adrenoceptors in isoproterenol-induced relaxation. This is the first report on such response patterns in snakes (including serotonin- and isoproterenol-induced relaxation). Nitric oxide may be involved in acetylcholine-induced relaxation but not in the responses to serotonin or isoproterenol. In contrast to the uniform vasoreactivity observed in terrestrial snakes, the vasoreactivity of semi-arboreal snakes may be governed by diverse regulatory mechanisms.

Processing with heat treatment has been reported to alter several therapeutic effects of turmeric. In Vietnamese traditional medicine, turmeric has been long used for bacterial infections, and roasting techniques are sometimes applied with this material. However, there have been no studies investigating the effects of these thermal processes on the plant’s antibacterial properties. Our study was therefore performed to examine the changes that roasting produced on this material. Slices of dried turmeric were further subjected to light-roasting (80 °C in 20 min) or dark-roasting (160 °C in 20 min) processes. Broth dilution and agar-well diffusion methods were applied to examine and compare the effects of ethanol extracts obtained from non-roasted, light-roasted and dark-roasted samples, on a set of 6 gram-positive and gram-negative bacteria. In both investigations, dark-roasted turmeric was significantly less antibacterial than non-roasted and light-roasted materials, as evident by the higher values of minimum inhibitory concentrations and the smaller diameters of induced inhibitory zones. In addition, dark-roasting was also found to clearly reduce curcumin contents, total polyphenol values and antioxidant activities of the extracts. These results suggest that non-roasting or light-roasting might be more suitable for the processing of turmeric materials that are aimed to be applied for bacterial infections.

Abstract Based on Cassini observations, we report representative electrostatic electron cyclotron harmonic (ECH) wave events observed in Saturn's magnetosphere within remote plasma injections. Unlike local injections, remote injections are “older” injection events that have evolved to form a dispersed signature in particle energy spectrum. We show that Saturnian ECH waves within remote injections present a strong fundamental band and much weaker high harmonic bands. By calculating the linear wave growth rates based on the measured electron velocity distributions, we indicate that Saturnian ECH waves can be excited by the loss cone distribution of remotely injected, hot electrons of ∼100 eV to several keV. We find that ECH waves tend to intensify with increased fluxes of injected hot electrons but weakening with increased evolution time of the flux tubes, which is consistent with the results of wave growth rates and improves the current understanding of the generation of ECH waves at Saturn.

Background The endoplasmic reticulum stress (ERS) and unfolded protein response (UPR) pathways play an essential role in the pathophysiology of hepatocellular carcinoma (HCC), and activation of the UPR pathway is strongly associated with tumor growth. However, the function of ERS-associated long non-coding RNAs (lncRNAs) in HCC is less recognized. Methods We have used TCGA (The Cancer Genome Atlas) to obtain clinical and transcriptome data for HCC patients and the GSEA (Gene Set Enrichment Analysis) molecular signature database to get the ERS gene. ERS-associated prognostic lncRNA was determined using univariate Cox regression study. Then, least absolute shrinkage and selection operator and multivariate Cox regression study were used to construct ERS-associated lncRNAs risk model. Next, we use Kaplan-Meier (KM) survival study, time-dependent receiver operating characteristic (ROC) curve, univariate and multivariate Cox regression study to validate and evaluate the risk model. GSEA reveals the underlying molecular mechanism of the risk model. In addition, differences in Immune cell Infiltration Study, half-maximal inhibitory concentration (IC50) and immune checkpoints blockade (ICB) treatment between high and low risk groups were analyzed. Results We constructed a risk model consisting of 6 ERS-associated lncRNAS (containingMKLN1-AS, LINC01224, AL590705.3, AC008622.2, AC145207.5, and AC026412.3). The KM survival study showed that the prognosis of HCC patients in low-risk group was better than that in high-risk group. ROC study, univariate and multivariate Cox regression study showed that the risk model had good predictive power for HCC patients. Our verification sample verified the aforesaid findings. GSEA suggests that several tumor- and metabolism-related signaling pathways are associated with risk groups. Simultaneously, we discovered that the risk models may help in the treatment of ICB and the selection of chemotherapeutic drugs. Conclusions In this article, we created an ERS-associated lncRNAs risk model to help prognostic diagnosis and personalized therapy in HCC.

Abstract Intercalation provides to the host materials a means for controlled variation of many physical/chemical properties and dominates the reactions in metal‐ion batteries. Of particular interest is the graphite intercalation compounds with intriguing staging structures, which however are still unclear, especially in their nanostructure and dynamic transition mechanism. Herein, the nature of the staging structure and evolution of the lithium (Li)‐intercalated graphite was revealed by cryogenic‐transmission electron microscopy and other methods at the nanoscale. The intercalated Li‐ions distribute unevenly, generating local stress and dislocations in the graphitic structure. Each staging compound is found macroscopically ordered but microscopically inhomogeneous, exhibiting a localized‐domains structural model. Our findings uncover the correlation between the long‐range ordered structure and short‐range domains, refresh the insights on the staging structure and transition of Li‐intercalated/deintercalated graphite, and provide effective ways to enhance the reaction kinetic in rechargeable batteries by defect engineering.

Abstract Although multistability is an important dynamic property of a wide range of complex systems, it is still a challenge to develop mathematical models for realising high order multistability using realistic regulatory mechanisms. To address this issue, we propose a robust method to develop multistable mathematical models by embedding bistable models together. Using the GATA1-GATA2-PU.1 module in hematopoiesis as the test system, we first develop a tristable model based on two bistable models without any high cooperative coefficients, and then modify the tristable model based on experimentally determined mechanisms. The modified model successfully realises four stable steady states and accurately reflects a recent experimental observation showing four transcriptional states. In addition, we develop a stochastic model, and stochastic simulations successfully realise the experimental observations in single cells. These results suggest that the proposed method is a general approach to develop mathematical models for realising multistability and heterogeneity in complex systems.

Abstract Background Methotrexate (MTX) has a protective effect against cardiovascular diseases (CVD), but the mechanism is unclear. Objective To investigate the effect of MTX on lipid profiles and the difference between psoriasis without arthritis (PsO) and psoriatic arthritis (PsA). Methods In this prospective study, we recruited 288 psoriatic patients (136 PsA and 152 PsO) who completed 12 weeks of MTX treatment. Total cholesterol (TC), triglycerides (TG), lipoprotein A [LP(a)], high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein (LDL), apolipoprotein A1 (ApoA1), and ApoB were measured. Results Compared with sex- and age-matched healthy controls, psoriatic patients had significantly (p < 0.0001) higher levels of proatherogenic lipids and lower levels of anti-atherogenic lipids. PsA patients had a higher ApoB/ApoA1 ratio than PsO patients (p < 0.05). Stepwise regression analysis found a positive correlation between the inflammatory marker hCRP and the Psoriasis Area Severity Index (PASI), ApoB/ApoA1 ratio, BMI, and smoking. ApoB was positively associated with concomitant arthritis, diabetes, and hypertension. MTX decreased the levels of pro-atherogenic and anti-atherogenic lipids. However, a significant reduction of the ApoB/ApoA1 ratio by MTX was only observed in male patients. Conclusion PsA patients had a significantly higher percentage of concomitant disease than PsO. The decrease of MTX on CVD might be related with sex. Trial registration ChiCTR2000036192

The basilar arterial endothelium mediates blood vessel relaxation partly through the release of nitric oxide (NO). Apoptosis of cerebrovascular endothelial cells is linked to a high mortality rate in chickens infected with the highly pathogenic avian influenza virus, but interestingly, ducks exhibit a greater resistance to this virus. In this study, we examined the responsiveness of duck basilar arteries (BAs) to various vasoactive substances, including 5-hydroxytryptamine (5-HT), histamine (His), angiotensin (Ang) II, noradrenaline (NA), acetylcholine (ACh), and avian bradykinin ornithokinin (OK), aiming to characterize the receptor subtypes involved and the role of endothelial NO in vitro. Our findings suggest that arterial contraction is mediated with 5-HT1 and H1 receptors, while relaxation is induced with β3-adrenergic and M3 receptors. Additionally, OK elicited a biphasic response in duck BAs, and Ang II had no effect. Endothelial NO appears to be crucial in relaxation mediated with M3 and OK receptors but not β3-adrenergic receptors in the duck BA. The reduced endothelial NO involvement in the receptor-mediated relaxation response in duck BAs represents a clear difference from the corresponding response reported in chicken BAs. This physiological difference may explain the differences in lethality between ducks and chickens when vascular endothelial cells are infected with the virus.

Satellite sensors often capture remote sensing images that contain various types of stripe noise. The presence of these stripes significantly reduces the quality of the remote images and severely affects their subsequent applications in other fields. Despite the existence of many stripe noise removal methods in the research, they often result in the loss of fine details during the destriping process, and some methods even generate artifacts. In this paper, we proposed a new unidirectional variational model to remove horizontal stripe noise. The proposed model fully considered the directional characteristics and structural sparsity of the stripe noise, as well as the prior features of the underlying image, to design different sparse constraints, and the ℓp quasinorm was introduced in these constraints to better describe these sparse characteristics, thus achieving a more excellent destriping effect. Moreover, we employed the fast alternating direction method of multipliers (ADMM) to solve the proposed non-convex model. This significantly improved the efficiency and robustness of the proposed method. The qualitative and quantitative results from simulated and real data experiments confirm that our method outperforms existing destriping approaches in terms of stripe noise removal and preservation of image details.

Abstract Crowd understanding and analysis have received increasing attention for couples of decades, and development of human behaviour recognition strongly supports the application of crowd understanding and analysis. Human behaviour recognition usually seeks to automatically analyse ongoing movements and actions in different camera views by using various machine learning methodologies in unknown video clips or image sequences. Compared to other data modalities such as documents and images, processing video data demands much higher computational and storage resources. The idea of using middle level semantic concepts to represent human actions from videos is explored and it is argued that these semantic attributes enable the construction of more descriptive methods for human action recognition. The mid‐level attributes, initialized by a cluster processing, are built upon low level features and fully utilize the discrepancies in different action classes, which can capture the importance of each attribute for each action class. In this way, the representation is constructed to be semantically rich and capable of highly discriminative performance even paired with simple linear classifiers. The method is verified on three challenging datasets (KTH, UCF50 and HMDB51), and the experimental results demonstrate that our method achieves better results than the baseline methods on human action recognition.

Necrotizing enterocolitis (NEC) is a life-threatening disease of the digestive system that occurs in the neonatal period. NEC is difficult to diagnose early and the prognosis is poor. Previous studies have reported that abnormalities can be detected before the presentation of clinical symptoms. Based on an analysis of literature related to the early prediction of NEC, we provide a detailed review on the early prediction and diagnosis methods of NEC, including ultrasound, near-infrared spectroscopy, biomarkers, and intestinal microbiota. This review aimed to provide a reference for further research and clinical practice.

Hyperspectral Anomaly Detection (HAD) aims to detect the pixel or target whose spectral characteristics are significantly different from the surrounding pixels or targets. The effectiveness of reconstructing the background model is an essential element affecting the improvement of the HAD performance. This paper proposes a Hyperspectral Anomaly Detection method based on Attention-aware Spectral Difference Representation (HAD-ASDR) to reconstruct more accurate background models by using the generated noise distribution matchable to the background as input. The proposed HAD-ASDR mainly includes three modules: Attention-aware Spectral Difference Representation Module (ASDRM), Convolutional Auto-Encoder based Background Reconstruction Module (CAE-BRM) and Joint Spectrum Intensity and Angle based Anomaly Detection Module (JSIA-ADM). First, inspired by Generative Adversarial Network (GAN), ASDRM is proposed to generate a noise distribution that better matches the background by the attention mechanism and the different operation. Then, CAE-BRM is employed to reconstruct the accurate background using the generated noise distribution as input and the convolutional auto-encoder with skip connections. Finally, JSIA-ADM is presented to detect anomalies more accurately by calculating the reconstructed errors from both spectral intensity and angle perspectives. The proposed HAD-ASDR has been verified on five data sets and achieves better or comparable HAD results compared to six other comparison methods. The average AUC of HAD-ASDR on these five data sets is 0.9817 higher than that of the comparison methods, resulting in an improvement of 0.0253. The experimental results demonstrate its superior performance and stability.

As a traditional Chinese herbal formula, Xiasangju (XSJ) is widely used in China for antipyresis and influenza treatment. However, XSJ still fails to have a comprehensive summary of the research progress in the last decade. This review summarizes the advanced research on the extraction process, phytochemistry, pharmacological activity, and quality control of XSJ. Current research mainly focuses on quality control and the pharmacological effects of single herbs and active ingredients, but many pharmacological mechanisms of the formula are unclear. The development of active ingredients reflects the active characteristics of triterpenes, phenolic acids and flavonoids, but the hepatotoxicity of Prunella vulgaris L. has not been taken into account. XSJ has extensive historical practical experiences, while systematic clinical trials remain lacking. Therefore, it is necessary to study the active ingredients and define the mechanisms of XSJ to develop multiple applications, and further studies on the dose range between its hepatoprotective activity and hepatotoxicity are necessary to improve the safety of the clinical application. In this review, the current problems are discussed to facilitate the reference basis for the subsequent research on the development of XSJ and future application directions.

Objective: To explore the repair effect of the prepared drug-loaded AM1241 poly(ethylene glycol)–dithiothreitol (PEG-DTT) hydrogel on cranial bone defects in SD rats.Methods: The PEG-DTT hydrogel under borax catalysis was quickly prepared, and the characterization of the material was observed by a scanning electron microscope. The effect of AM1241 on cell activity and bone tissue differentiation was tested. The SD rat model of cranial bone defect was established, and the defect was repaired by injecting the prepared hydrogel into the defect. The defect was divided into four groups, namely, sham group, blank group, PEG-DTT group, and PEG-DTT + AM1241 group. The rats were euthanized, and whole cranial bone was taken out for micro-CT and histological observation.Results: The prepared hydrogel is porous; it is liquid when heated to 80°C and a hydrogel when cooled to 25°C. 5–10 μM AM1241 increased osteoblast activity. A moderate amount of AM1241 can promote osteogenic differentiation. Both the PEG-DTT group and PEG-DTT + AM1241 group showed obvious new bone tissue formation, but the PEG-DTT + AM1241 group had a better effect. In addition, the new bone tissue in the PEG-DTT + AM1241 group was significantly more than that in the other groups.Conclusion: The prepared AM1241-loaded PEG-DTT hydrogel showed a good repair effect on SD rats with cranial bone defects. It can be used as materials for cranial bone repair in SD rats with cranial bone defects, but the repair effect is weaker than that of normal bone. These results provide a theoretical and practical basis for its further clinical application.

This study aims to propose an approach for determining key Performance Shaping Factors (PSFs) to promote human reliability management during LNG ship offloading process. Offloading LNG from ship to onshore terminal is a high-risk and human-related operation; a small human error may trigger catastrophic consequences such as fire, explosion, and even fatality. Therefore, ensuring high human reliability level is necessary. It is widely acknowledged that human reliability is mainly influenced by plenty of PSFs. If some top important PSFs can be identified, then it will be helpful to human reliability assurance and targeted management for avoiding human errors in the shipping LNG offloading work. Determining key PSFs is a decision-making system, but there is always lack of historical data of PSF. Namely, this decision-making system has strong characteristic of grey, which is an obstacle for finding the significant PSFs. Due to this condition, the grey theory-based Grey Relational Analysis (GRA) method is a choice and should be selected for handling the insufficient PSF data and grey characteristics. Apart from GRA, the definition of risk (frequency products consequence) is utilised as the basis for reasonably explaining the ranking order of each involved PSF. In one word, GRA is firstly conducted from the view of frequency and the view of consequence, then combining the results together to identify key PSFs. The proposed method is applied to a real shipping LNG offloading case. The final result indicates that the proposed method provides a reasonable and effective way to find key PSFs for ensuring human reliability in shipping LNG offloading work.

The influence of sediment media on the blade pressure and cavitation of a tubular turbine was investigated in this study. The Zwart–Geber–Belamri cavitation model and the shear stress transport k–w turbulence model were applied to numerically simulate and experimentally validate the full flow path of the tubular turbine under combined conditions for sediment particle sizes of 0.01 and 0.05 mm and concentrations of 1%, 1.5%, and 2%. The results show that the pressure of the blade increases with the sediment concentration. Cavitation mainly occurs between the blade shroud and the leading edge. The higher the sediment concentration, the lower the vapor volume fraction at the same sediment particle size. The presence of sediments inhibits further cavitation development, and the inhibition effect is significant. At the same concentration, the larger the particle size of the sediment, the lower the vapor volume fraction. Thus, the increase in the particle size inhibits cavitation, but the inhibition effect is not significant.

People who drink naturally hardened water may experience longevity-enhancing effects. In this study, we investigated water hardness and longevity from both geological and epidemiological perspectives in Japan’s Amami islands, where drinking water is drawn from coralline or non-coralline bedrock. We investigated drinking water hardness, limestone bedrock occupancy, and the centenarian rate (number per 10,000 population) by municipality across four adjacent islands (Amami-Oshima (non-coralline), Tokunoshima, Okinoerabu, and Yoron (predominantly coralline)). Limestone was strongly correlated with water hardness (r = 0.99; p < 0.01), occupying more than 80% of the bedrock where the water was the hardest (Tokunoshima’s Isen municipality: 86.5%; Yoron: 82.9%) and being scarcely detectable in Amami-Oshima (0.0 to 0.2%), where the water was the least hard. The centenarian rate was also strongly correlated with water hardness (r = 0.84, p < 0.01), with the highest figures in Yoron (29.7) and Isen (29.2), and the lowest in Amami-Oshima (0.0 to 12.2). Therefore, we hypothesize a potentially beneficial effect of hard water on longevity when that water is drawn from coralline limestone. Water hardness is determined by the water content of calcium and magnesium and may plausibly influence life expectancy through a preventative effect against cardiovascular disease. Our findings are of interest to current debates about future global access to drinking water and its quality.

To lay the foundation for environmentally friendly energetic polymer composites, GAP (glycidyl azide polymer) adhesive-based polymer films with different curing parameter R (mol ratio of hydroxyl/isocyanate) and energetic polymer composites with different RDX contents were studied. GAP/TDI (toluene diisocyanate)/GLY(glycerol) was selected as the adhesive system. The tensile strength and elongation at the break of the polymer film with R = 2.2, was 14.34 MPa and 176.86%, respectively, as observed by an AGS-J electronic universal testing machine. A relatively complete cross-linking network and high hydrogen bonding interaction were observed by LF-NMR (low-field nuclear magnetic resonance, where the cross-linking density was 11.06 × 10−4 mol/cm3) and FT-IR (fourier transform infrared spectroscopy, where the carbonyl bonding ratio was 64.84%). Forty percent RDX(hexogen) was added into the adhesive system. The tensile strength was 4.65 MPa, and the elongation at the break was 78.49%; meanwhile, the heat of the explosive was 2.87 MJ/kg, and the residue carbon rate was only 2.47%. The tensile cross-sections of energetic polymer composites were observed by SEM (Scanning electron microscopy).

The mechanism of systemic osteoporosis caused by chronic infection is not completely clear, and there is a lack of reasonable interventions for this disease. In this study, heat-killed S. aureus (HKSA) was applied to simulate the inflammation caused by the typical clinical pathogen and to explore the mechanism of systemic bone loss caused by it. In this study, we found that the systemic application of HKSA caused bone loss in mice. Further exploration found that HKSA caused cellular senescence, telomere length shortening, and telomere dysfunction-induced foci (TIF) in limb bones. As a well-known telomerase activator, cycloastragenol (CAG) significantly alleviated HKSA-induced telomere erosion and bone loss. These results suggested that telomere erosion in bone marrow cells is a possible mechanism of HKSA-induced bone loss. CAG may protect against HKSA-induced bone loss by alleviating telomere erosion in bone marrow cells.

Epigenetic DNA modification by 5-hydroxymethylcytosine (5hmC), generated by the Ten-eleven translocation (TET) dioxygenases, regulates diverse biological functions in many organ tissues, including the mammalian eye. For example, 5hmC has been shown to be involved in epigenetic regulation of retinal gene expression. However, a functional role of 5hmC in corneal differentiation has not been investigated to date. Here, we examined 5hmC and TET function in the human cornea. We found 5hmC highly expressed in MUC16-positive terminally differentiated cells that also co-expressed the 5hmC-generating enzyme TET2. TET2 knockdown (KD) in cultured corneal epithelial cells led to significant reductions of 5hmC peak distributions and resulted in transcriptional repression of molecular pathways involved in corneal differentiation, as evidenced by downregulation of MUC4, MUC16, and Keratin 12. Additionally, integrated TET2 KD RNA-seq and genome-wide Reduced Representation Hydroxymethylation Profiling revealed novel epigenetically regulated genes expressed by terminally differentiated cells, including KRT78, MYEOV, and MAL. In aggregate, our findings reveal a novel function of TET2 in the epigenetic regulation of corneal epithelial gene expression and identify novel TET2-controlled genes expressed in differentiated corneal epithelial cells. These results point to potential roles for TET2 induction strategies to enhance treatment of corneal diseases associated with abnormal epithelial maturation.

Background and Objectives: Rho GTPase-activating protein (RhoGAP) is a negative regulatory element of Rho GTPases and participates in tumorigenesis. Rho GTPase-activating protein 21 (ARHGAP21) is one of the RhoGAPs and its role in cholangiocarcinoma (CCA) has never been disclosed in any publications. Materials and Methods: The bioinformatics public datasets were utilized to investigate the expression patterns and mutations of ARHGAP21 as well as its prognostic significance in CCA. The biological functions of ARHGAP21 in CCA cells (RBE and Hccc9810 cell) were evaluated by scratch assay, cell counting kit-8 assay (CCK8) assay, and transwell migration assay. In addition, the underlying mechanism of ARHGAP21 involved in CCA was investigated by the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis, and the most significant signaling pathway was identified through gene set enrichment analysis (GSEA) and the Western blot method. The ssGSEA algorithm was further used to explore the immune-related mechanism of ARHGAP21 in CCA. Results: The ARHGAP21 expression in CCA tissue was higher than it was in normal tissue, and missense mutation was the main alteration of ARHGAP21 in CCA. Moreover, the expression of ARHGAP21 had obvious differences in patients with different clinical characteristics and it had great prognostic significance. Based on cell experiments, we further observed that the proliferation ability and migration ability of the ARHGAP21-knockdown group was reduced in CCA cells. Several pathological signaling pathways correlated with proliferation and migration were determined by GO and KEGG analysis. Furthermore, the PI3K/Akt signaling pathway was the most significant one. GSEA analysis further verified that ARHGAP21 was highly enriched in PI3K/Akt signaling pathway, and the results of Western blot suggested that the phosphorylated PI3K and Akt were decreased in the ARHGAP21-knockdown group. The drug susceptibility of the PI3K/Akt signaling pathway targeted drugs were positively correlated with ARHGAP21 expression. Moreover, we also discovered that ARHGAP21 was correlated with neutrophil, pDC, and mast cell infiltration as well as immune-related genes in CCA. Conclusions: ARHGAP21 could promote the proliferation and migration of CCA cells by activating the PI3K/Akt signaling pathway, and ARHGAP21 may participate in the immune modulating function of the tumor microenvironment.

Background and Purpose: Hemoglobin is one of the main proteins in erythrocytes. There are significant correlations between low hemoglobin and white matter hyperintensities (WMH) and cognitive impairment. This study explored whether erythrocytopenia has predictive value for vascular cognitive impairment (VCI) in patients with WMH.Method: We conducted a cross-sectional study of 302 patients, including 62 with cerebral small vessel disease and 240 with stroke. Basic demographic data and fasting blood were collected. First, all patients were divided into normal cognition (NC), mild VCI (mVCI), and severe VCI (sVCI) groups (subgroups later) based on cognitive behavior scores. Second, all patients were divided into mild WMH (mWMH) and severe WMH (sWMH) groups based on Fazekas scores. The differences in blood markers between different groups or subgroups with different cognitive levels were analyzed by univariate analysis. Then, binary logistic regression was used to analyze the diagnostic value of erythrocyte counts for VCI in the sWMH group, and ordinal logistic regression was used to analyze the predictive value of multiple variables for different cognitive levels.Results: Univariate analysis showed that erythrocytes, hemoglobin, high-sensitivity C-reactive protein, retinol binding protein and prealbumin were potential blood markers for different cognitive levels in sWMH patients. Among them, erythrocytopenia has good predictive value for the diagnosis of mVCI (AUC = 0.685, P = 0.008) or sVCI (AUC = 0.699, P = 0.003) in patients with sWMH. Multivariate joint analysis showed that erythrocytes were an independent protective factor reducing the occurrence of VCI in patients with sWMH (OR = 0.633, P = 0.045). Even after adjusting for age, there was still a significant difference (P = 0.047).Conclusion: Erythrocytes are an independent protective factor for VCI in patients with sWMH. Promoting hematopoietic function may have potential value for prevention of cognitive decline in patients with cerebrovascular disease.

For the petrochemical industry, the spontaneous burning of iron sulfide compounds has been a major issue. In this study, XRD characterization of samples of iron sulfide compounds with spontaneous combustion tendency revealed that amorphous FeS was the primary constituent of the samples. A molecular simulation was used to build an amorphous FeS cluster model, and the density functional theory was used to examine the adsorption and reactivity characteristics of Fe4S4 clusters with O2. Different adsorption structures are generated by considering different adsorption sites and the electronic characteristics of each adsorption structure are evaluated. The results show that O2 prefers to adsorb around Fe atoms and has repulsion with S atoms, and the adsorption energy is maximum when two O atoms are co-adsorbed around Fe atoms, which is 198.13 kJ/mol. After adsorption charge, oxygen is in the superoxide state. The calculation of the reaction path divides the reaction process into different stages and considers different reaction routes. A thorough evaluation of the energy barriers and reaction energies of the two exothermic reactions leads to the conclusion that reaction path 1 is the optimal reaction path, and the reaction can release a total of 582.76 kJ/mol of heat. According to calculations, dimeric sulfur S2 must absorb a large part amount of energy in order to conduct the oxidation process. However, because S2 is present in the Fe4S4 reaction system, it may start the oxidation reaction by absorbing heat from the system and releasing 470.94 kJ/mol of heat. As a result, we conclude that this spontaneous exothermic reaction is a major cause of iron sulfide compounds spontaneous combustion. The thermal oxidation of the dimeric sulfur S2 generated in the reaction system releases heat that aggregates with the heat from the Fe4S4 cluster’s oxidation reaction system, eventually causing spontaneous combustion as a result of the heat’s continual buildup. In this study, we explore the reason for the extremely easy oxidation and spontaneous burning of iron sulfide compounds from a microscopic perspective to provide a theoretical foundation for the prevention and control of iron sulfide compound spontaneous combustion in the petrochemical sector.

Recently, the prevention and control of industrial pollution has attracted extensive attention. Many studies based on emission data have achieved great success, but there are relatively few studies on the emission data acquisition. To this end, we propose a data acquisition and processing system (DAPS) for online monitoring of industrial pollutants. The system receives real-time emission data through the RS485, automatically generates periodic data, and saves it in the SQL Server database. Real-time Ethernet (RTE) is used to transmit control signals between the system and programmable logic controllers (PLCs). Emission data is transmitted to local industrial control systems and remote monitoring centers (RMCs) via a wireless network. We tested the acquisition function and communication function of the system through experiments and evaluated the overall performance of the system by observing the memory and CPU usage. The system is characterized by simple implementation and versatility and can be applied to continuous monitoring of various industrial pollutants.

Bombyx mori silk fibroin (SF) is widely used in the field of biomaterials due to its excellent biocompatibility and mechanical properties. However, SF cannot be used directly in many applications and needs to be dissolved first. Lithium bromide (LiBr) is a traditional solvent which is usually used to dissolve SF. However, LiBr has several limitations, e.g., it is expensive, it is toxic to organisms, and it is environmentally unfriendly. Herein, we investigate the possibility of developing a ternary reagent system that is inexpensive, non-toxic to organisms, and environmentally friendly as an alternative for silk fibroin solubilization. The results confirm that regenerated silk fibroin (RSF) prepared using a ternary reagent has the same morphology and amino acid composition as that prepared using LiBr, but the RSF prepared using a ternary reagent still had a small amount of calcium residue even after long-term dialysis. Further research found that the residual calcium does not cause significant differences in the structure and biological performance of the RSF, such as its cytotoxicity, blood compatibility, and antibacterial properties. Therefore, we believe that ternary reagents are an ideal alternative solvent for dissolving SF.

As an indispensable task for traffic management department, road maintenance has attracted much attention during the last decade due to the rapid development of traffic network. As is known, crack is the early form of many road damages, and repair it in time can significantly save the maintenance cost. In this case, how to detect crack regions quickly and accurately becomes a huge demand. Actually, many image processing technique based methods have been proposed for crack detection, but their performances can not meet our expectations. The reason is that, most of these methods use bottom features such as color and texture to detect the cracks, which are easily influenced by the varied conditions such as light and shadow. Inspired by the great successes of machine learning and artificial intelligence, this paper presents a sample and structure guided network for detecting road cracks. Specifically, the proposed network is based on U-Net architecture, which remains the details from input to output by using skip connection strategy. Then, because the scale of crack samples is much smaller than that of non-crack ones, directly using the conventional cross entropy loss can not optimize the network effectively. In this case, the Focal loss is utilized to address the model optimization problem. Additionally, we incorporate the self-attention strategy into the proposed network, which enhances its stability by encoding the 2-order information among different local regions into the final features. Finally, we test the proposed method on four datasets, three public ones with labels and a photographed one without labels, to validate its effectiveness. It is noteworthy that, for the photographed dataset, we design a series of image processing strategies such as contrast enhancement to improve the generalization capability of the proposed method.
