Beta-adrenergic blocking agents, or beta-blockers, are primarily used to manage cardiovascular conditions such as hypertension, heart failure, and arrhythmias by blocking the effects of stress hormones on beta-adrenergic receptors, reducing heart rate and blood pressure. Recent research highlights their potential impact on cancer, including cutaneous and oral melanoma. Beta-adrenergic receptors, present on various cell types including tumor cells, influence cancer processes like cell proliferation, angiogenesis, metastasis, and immune response modulation. In melanoma, beta-blockers may alter the tumor microenvironment, affecting cancer progression and treatment outcomes. Studies suggest beta-blockers might reduce cutaneous melanoma progression by inhibiting angiogenesis and tumor cell proliferation, improving survival rates. In oral melanoma, beta-blockers may disrupt stress-induced signaling pathways, reducing tumor growth and metastasis while modulating immune responses. This emerging research indicates beta-blockers' potential as adjunct therapy in melanoma treatment, necessitating comprehensive clinical studies to confirm their efficacy and safety. Future investigations should focus on large-scale trials, exploring the effects of different beta-blockers, and their combination with existing therapies. Head and neck squamous cell carcinoma (HNSCC) encompass cancers originating in the head and neck mucosal surfaces, including the oral cavity, pharynx, and larynx. Standard treatments involve surgery, radiation therapy, chemotherapy, or combinations thereof. Radiation therapy, targeting pharyngeal cancer cells, is common but can damage critical structures, affecting quality of life (QoL). Dysphagia, xerostomia, speech difficulties, and pain are prevalent side effects. Research shows radiation dose correlates with severity of these effects; sparing healthy tissue reduces side effects. Advanced techniques like intensity-modulated radiation therapy (IMRT) and proton therapy offer precise tumor targeting, sparing normal tissues. Ongoing research aims to refine techniques for better long-term outcomes in HNSCC patients.

Locoregionally advanced mucosal melanoma, particularly when harboring the BRAF V600L mutation and situated in the hypopharynx, presents a therapeutic challenge due to limited treatment options and aggressive behavior. Traditional modalities often yield unsatisfactory outcomes, prompting the exploration of novel therapeutic approaches. A cutting-edge strategy involves the administration of combined BRAF and MEK inhibitors, targeting specific molecular pathways implicated in mucosal melanoma pathogenesis. BRAF inhibitors, such as vemurafenib and dabrafenib, block mutated BRAF activity, while MEK inhibitors, like trametinib and cobimetinib, target downstream components of the MAPK pathway. This synergistic combination overcomes resistance mechanisms and enhances treatment efficacy. Recent case studies and clinical trials have reported promising results, including improved response rates and prolonged progression-free survival in patients with locoregionally advanced, BRAF V600L–positive mucosal melanoma of the hypopharynx. These findings underscore the potential of combined BRAF and MEK inhibition as a novel therapeutic paradigm for this challenging disease subtype, warranting further investigation to optimize its integration into clinical practice.

The development of effective cancer treatments relies heavily on the successful execution of clinical trials, particularly Phase I trials, which are crucial for determining optimal drug dosages. In head and neck cancer, characterized by complex treatment needs and diverse patient responses, optimizing sample size in these trials is essential for ensuring both safety and efficacy. Traditional sample size planning often uses fixed statistical methods that lack flexibility. In contrast, a Bayesian approach offers a dynamic framework that integrates prior information and continuously updates probabilities with new data, enabling adaptive trial designs. This adaptability allows modifications based on interim results, enhancing patient safety without compromising trial integrity. Bayesian methods improve dose estimation precision, reduce required sample sizes, and facilitate informed decision-making throughout the trial. By continuously updating risk assessments, Bayesian approaches ensure ongoing patient safety and potentially reduce trial costs. This method's ability to provide deeper insights into patient-specific responses supports the development of more personalized therapeutic strategies. As oncology evolves toward precise and personalized treatments, Bayesian methods will play a crucial role in advancing clinical trial design, particularly in Phase I dose-finding trials for head and neck cancer, enhancing the development of next-generation cancer therapies.

Adenoid cystic carcinoma (ACC) presents a formidable challenge due to its slow growth, late-stage metastasis, and resistance to conventional therapies. Targeting the AXL receptor, implicated in aggressive tumor behavior and therapeutic resistance, offers a promising therapeutic avenue. Several studies demonstrate that AXL inhibition suppresses tumor cell proliferation, migration, and invasion, sensitizing ACC cells to chemotherapy and radiation therapy. Furthermore, AXL-targeted therapies modulate the tumor microenvironment, disrupting pro-tumorigenic signals. Despite challenges in clinical translation and tumor heterogeneity, early clinical findings show promise, suggesting AXL inhibition as a potential strategy to halt ACC progression and improve patient outcomes. The pursuit of precision-based treatments tailored to individual molecular profiles signifies a paradigm shift in ACC therapeutics, offering renewed hope for patients facing this relentless disease. Continued research into AXL biology and therapeutic strategies holds the potential to reshape the treatment landscape for metastatic, refractory ACC.

In recent years, targeted therapies have revolutionized cancer treatment, particularly for tumors with specific genetic alterations. High-grade salivary gland ductal carcinoma and non-small cell lung cancer (NSCLC) are two distinct malignancies characterized by unique challenges but share uncommon epidermal growth factor receptor (EGFR) mutations as potential therapeutic targets. High-grade salivary gland ductal carcinoma, rare and aggressive, presents significant treatment challenges, with limited efficacy from traditional modalities. Recent genomic profiling has identified EGFR amplification in a subset of cases, suggesting targeted therapy potential. Afatinib, an irreversible EGFR inhibitor, has emerged as a promising option for high-grade salivary gland ductal carcinoma. Preclinical studies demonstrate its effectiveness in inhibiting tumor growth and inducing apoptosis in EGFR-amplified cell lines. Clinical trials confirm significant tumor responses and improved progression-free survival with afatinib treatment. Similarly, in NSCLC, uncommon EGFR mutations historically resistant to conventional inhibitors respond favorably to afatinib, as evidenced by clinical trials and real-world data. The success of afatinib in both malignancies underscores the importance of personalized medicine. Targeting specific genetic alterations with afatinib offers tailored treatment strategies, potentially improving outcomes and quality of life. Ongoing research aims to optimize afatinib use in these contexts, refining treatment protocols and maximizing therapeutic efficacy.

The synthesis of a Ni-doped ZnO nanocomposite incorporating chitosan (CS/Ni-doped ZnO) was achieved via a precipitation method, followed by annealing at 250 °C. This study comprehensively examined the nanocomposite’s structural, functional, morphological, and porosity properties using various analytical techniques, including X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), high-resolution scanning electron microscopy (HR-SEM), transmission electron microscopy (TEM), and Brunauer–Emmett–Teller (BET) analysis. The presence of chitosan (CS) and nickel (Ni) within the nanocomposite, along with their influence on reducing the band gap of ZnO particles and enhancing the generation of electron-hole pairs, was confirmed using UV-visible near-infrared spectroscopy (UV-vis-NIR). The electrochemical properties of the CS/Ni-doped ZnO nanocomposite were investigated via electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV) by utilizing a phosphate buffer solution with a pH of 6, which closely resembled the typical pH of bacterial cell walls. Finally, the prepared CS/Ni-doped ZnO nanocomposite was evaluated for its antibacterial and anticancer activities. The results demonstrated the highest inhibition of bacterial growth in P. vulgaris, whereas the lowest inhibition was found in S. aureus across various concentrations, thus highlighting its potential in antimicrobial applications. The cytotoxicity of CS/Ni-doped ZnO nanocomposites demonstrated remarkable effects with a half-maximum inhibitory concentration of approximately 80 ± 0.23 µg mL−1 against MCF-7 breast cancer cell lines, following a dose-dependent manner.

Nanozymes represent a category of nano-biomaterial artificial enzymes distinguished by their remarkable catalytic potency, stability, cost-effectiveness, biocompatibility, and degradability. These attributes position them as premier biomaterials with extensive applicability across medical, industrial, technological, and biological domains. Following the discovery of ferromagnetic nanoparticles with peroxidase-mimicking capabilities, extensive research endeavors have been dedicated to advancing nanozyme utilization. Their capacity to emulate the functions of natural enzymes has captivated researchers, prompting in-depth investigations into their attributes and potential applications. This exploration has yielded insights and innovations in various areas, including detection mechanisms, biosensing techniques, and device development. Nanozymes exhibit diverse compositions, sizes, and forms, resembling molecular entities such as proteins and tissue-based glucose. Their rapid impact on the body necessitates a comprehensive understanding of their intricate interplay. As each day witnesses the emergence of novel methodologies and technologies, the integration of nanozymes continues to surge, promising enhanced comprehension in the times ahead. This review centers on the expansive deployment and advancement of nanozyme materials, encompassing biomedical, biotechnological, and environmental contexts.

Heat shock proteins (HSPs) encompass both extrinsic chaperones and stress proteins. These proteins, with molecular weights ranging from 14 to 120 kDa, are conserved across all living organisms and are expressed in response to stress. The upregulation of specific genes triggers the synthesis of HSPs, facilitated by the interaction between heat shock factors and gene promoter regions. Notably, HSPs function as chaperones or helper molecules in various cellular processes involving lipids and proteins, and their upregulation is not limited to heat-induced stress but also occurs in response to anoxia, acidosis, hypoxia, toxins, ischemia, protein breakdown, and microbial infection. HSPs play a vital role in regulating protein synthesis in cells. They assist in the folding and assembly of other cellular proteins, primarily through HSP families such as HSP70 and HSP90. Additionally, the process of the folding, translocation, and aggregation of proteins is governed by the dynamic partitioning facilitated by HSPs throughout the cell. Beyond their involvement in protein metabolism, HSPs also exert a significant influence on apoptosis, the immune system, and various characteristics of inflammation. The immunity of aquatic organisms, including shrimp, fish, and shellfish, relies heavily on the development of inflammation, as well as non-specific and specific immune responses to viral and bacterial infections. Recent advancements in aquatic research have demonstrated that the HSP levels in populations of fish, shrimp, and shellfish can be increased through non-traumatic means such as water or oral administration of HSP stimulants, exogenous HSPs, and heat induction. These methods have proven useful in reducing physical stress and trauma, while also facilitating sustainable husbandry practices such as vaccination and transportation, thereby offering health benefits. Hence, the present review discusses the importance of HSPs in different tissues in aquatic organisms (fish, shrimp), and their expression levels during pathogen invasion; this gives new insights into the significance of HSPs in invertebrates.

The present study aimed to analyze the enhancement of innate immune responses in juvenile-stage common carp (Cyprinus carpio L.), upon the administration of heat-killed Aeromonas hydrophila at a dosage of 1 × 107 CFU ml−1 through bio-encapsulation in the aquatic crustacean, Artemia salina. This work emphasizes the modulation of innate immune response when administered with the bio-encapsulated heat-killed antigen that acts as an inactivated vaccine against Motile Aeromonas Septicemia disease. Bio-encapsulated oral administration of antigens promotes innate immunity in juvenile-stage fishes. The optimization of effective bio-encapsulation of bacterin in Artemia salina nauplii was carried out and the best optimal conditions were chosen for immunization. The functional immune parameters such as myeloperoxidase, lysozyme, alkaline phosphatase, antiprotease and respiratory burst activity in serum, blood and intestinal tissue samples were analyzed along with blood differential leukocyte count and tissue histopathology studies. Both humoral and cellular immune responses analyzed were substantially induced or enhanced in the treatment groups in comparison with the control group. The results showed a significant variation in the bio-encapsulation group than the control group and also were comparable to the protection conferred with immersion route immunization under similar conditions. Thus, most of the innate non-specific immune responses are inducible, despite being constitutive of the fish immune system, to exhibit a basal level of protection and a road to better vaccination strategy in Cyprinus carpio L. aquaculture worldwide.
