Crystallisation of anthraquinone substituted p-tert-butylthiacalix[4]arene in the partial cone conformation from a range of solvents led to the isolation of two solvates, containing a mixture of chloroform and acetonitrile (PC1a) or toluene (PC1d) molecules, the structures of which were uncovered by SCXRD studies. Upon heating, the former solvate undergoes a single-crystal-to-single-crystal transformation, leading to removal of the acetonitrile molecules from the host and repositioning of the chloroform molecules (PC1b). This is connected with rearrangements of the non-covalent interactions, nicely reflected by a change in the hydrogen bond acceptor of the weak C–H⋯O hydrogen bonds. The transformation process is reversible, as confirmed by exposure of the crystal to acetonitrile vapour. Furthermore, the guest free phase of PC1 was obtained by melt crystallisation (PC1c). In the field of medicinal chemistry, the precise installation of a trideuteromethyl group is gaining ever‐increasing attention. Site‐selective incorporation of the deuterated "magic methyl" group can provide profound pharmacological benefits and can be considered an important tool for drug optimization and development. This review provides a structured overview, according to trideuteromethylation reagent, of currently established methods for site‐selective trideuteromethylation of carbon atoms. In addition to CD3, the selective introduction of CD2H and CDH2 groups is also considered. For all methods, the corresponding mechanism and scope are discussed whenever reported. As such, this review can be a starting point for synthetic chemists to further advance trideuteromethylation methodologies. At the same time, this review aims to be a guide for medicinal chemists, offering them the available C−CD3 formation strategies for the preparation of new or modified drugs. [ABSTRACT FROM AUTHOR]

With the resurgence of electrosynthesis in organic chemistry, there is a significant increase in the number of routes available for late-stage functionalization (LSF) of drugs. Electrosynthetic methods, which obviate the need for hazardous chemical oxidants or reductants, offer unprecedented control of reactions through the continuous variation of the applied potential and the possibility of combination with photochemical processes. This capability is a substantial advantage for performing electrochemical or photoelectrochemical LSF. Ultimately, these protocols are poised to become a vital component of the medicinal chemist's toolkit. In this review, we discuss electrochemical protocols that have been demonstrated to be applicable for the LSF of pharmaceutical drugs, their derivatives, and natural substrates.
