Early involvement of students in hands-on research experiences are known to demystify research and promote the pursuit of careers in science. But in large enrollment departments such opportunities for undergraduates to participate in research are rare. To counteract such lack of opportunities, inquiry-based laboratory module in plant tissue culture was implemented in an undergraduate botany course impacting approximately 140 Hispanic minority students per year. In this module, spread throughout the semester, the students used African violet to gain experience in plant tissue culture techniques. The objective was for the students to learn how to take part of the plant from "in vivo" to "in vitro" culture. This required the establishment of aseptic techniques and the use of different media components to multiply plants under "in vitro" conditions. In depth assessment of gain-of content knowledge and gain-of confidence revealed that our inquiry-based approach allowed the students to learn while increasing their self-perception of scientific methodology. In three semesters, the students reported a 2.5-fold overall increase in the post-module assessment for content knowledge compared to pre-module assessment. Similarly, approximately 85% of the students reported that they gained self-confidence in many aspects pertaining to conducting future research such as the use of primary literature, the design and performance of novel scientific experiments, and the formulation of a testable hypothesis. Though this lab module was solely in plant tissue culture, the inquiry-based nature of the exercise developed students' research skills and built confidence which is important in increasing retention of students in sciences.

Today, more minority students are entering undergraduate programs than ever before, but they earn only 6% of all science or engineering PhDs awarded in the United States. Many studies suggest that hands-on research activities enhance students' interest in pursuing a research career. In this paper, we present a model for the implementation of laboratory research in the undergraduate teaching laboratory using a culturally relevant approach to engage students. Laboratory modules were implemented in upper-division genetics and cell biology courses using cassava as the central theme. Students were asked to bring cassava samples from their respective towns, which allowed them to compare their field-collected samples against known lineages from agricultural stations at the end of the implementation. Assessment of content and learning perceptions revealed that our novel approach allowed students to learn while engaged in characterizing Puerto Rican cassava. In two semesters, based on the percentage of students who answered correctly in the premodule assessment for content knowledge, there was an overall improvement of 66% and 55% at the end in the genetics course and 24% and 15% in the cell biology course. Our proposed pedagogical model enhances students' professional competitiveness by providing students with valuable research skills as they work on a problem to which they can relate. Interclass hybridization between soft and hard wheat (Triticum aestivum L.) results in new genetic combinations of potential value. We investigated whether interclass hybridization could improve end-use quality of both classes. Our objectives were to analyze quality traits in a population of recombinant inbred lines (RILs) derived from a cross between the good quality soft white wheat NY6432-18 (NY18), and good quality hard white wheat Clark's Cream (CC), identify quantitative trait loci (QTLs) for those traits, and use linkage analysis to determine which parent was contributing favorable alleles at specific QTLs for a given trait. The population was assessed for milling, protein and dough mixing, hydration, cookie and loaf traits. Traits were measured in two to six environments grown over three seasons in Ithaca, NY. The molecular map for the population contains 370 molecular markers including restiction fragment length polymorphisms (RFLPs), microsatellites, and markers derived from known function genes in wheat. Linkage groups have been located to all the wheat chromosomes except for 7D. Pinb derived from the puroindoline b gene on chromosome 5DS was the major QTL for milling, hydration, and cookie baking traits. The major QTL for mixograph peak time was at the Glu-Dy1 marker, derived from Glu-D1-2 gene on chromosome 1DL. The Glu-Ax1 and Glu-By1 markers were QTLs for mixograph peak height and tolerance, respectively. QTLs for flour protein quantity were detected on chromosome 2B. With the exception of the hydration traits, multiple regression models included alleles from both parents. Summary: Storage roots of cassava (Manihot esculenta Crantz), a major subsistence crop of sub‐Saharan Africa, are calorie rich but deficient in essential micronutrients, including provitamin A β‐carotene. In this study, β‐carotene concentrations in cassava storage roots were enhanced by co‐expression of transgenes for deoxy‐ d‐xylulose‐5‐phosphate synthase (DXS) and bacterial phytoene synthase (crtB), mediated by the patatin‐type 1 promoter. Storage roots harvested from field‐grown plants accumulated carotenoids to ≤50 μg/g DW, 15‐ to 20‐fold increases relative to roots from nontransgenic plants. Approximately 85%–90% of these carotenoids accumulated as all‐trans‐β‐carotene, the most nutritionally efficacious carotenoid. β‐Carotene‐accumulating storage roots displayed delayed onset of postharvest physiological deterioration, a major constraint limiting utilization of cassava products. Large metabolite changes were detected in β‐carotene‐enhanced storage roots. Most significantly, an inverse correlation was observed between β‐carotene and dry matter content, with reductions of 50%–60% of dry matter content in the highest carotenoid‐accumulating storage roots of different cultivars. Further analysis confirmed a concomitant reduction in starch content and increased levels of total fatty acids, triacylglycerols, soluble sugars and abscisic acid. Potato engineered to co‐express DXS and crtB displayed a similar correlation between β‐carotene accumulation, reduced dry matter and starch content and elevated oil and soluble sugars in tubers. Transcriptome analyses revealed a reduced expression of genes involved in starch biosynthesis including ADP‐glucose pyrophosphorylase genes in transgenic, carotene‐accumulating cassava roots relative to nontransgenic roots. These findings highlight unintended metabolic consequences of provitamin A biofortification of starch‐rich organs and point to strategies for redirecting metabolic flux to restore starch production. Lima bean is a species cultivated broadly in the Americas and has been cultivated in the Caribbean for at least 500 years. In order to determine the genetic structure and diversity of Lima bean from the Caribbean, 50 landraces from Haiti, the Dominican Republic and Puerto Rico were collected and analyzed using 24 SSR markers. All landraces in this Caribbean collection were found to be of Middle-American descent. The genetic diversity (H E) was highest in the landraces from Puerto Rico and lowest in Haitian landraces. The observed proportion of heterozygotes (H O) was higher in the Haitian landraces and lowest in Puerto Rican landraces. Unweighted pair-group method with arithmetic averaging analysis showed that the landraces clustered into 3 clusters with all Haitian landraces grouping in one cluster. We also assessed the agro-morphological characteristics of the collection as well as the content of cyanogenic glucoside, linamarin, in leaves and dry seeds. Lima bean, which is a model crop for indirect plant defenses against herbivory, also possess linamarin as a source of direct plant defense. Upon tissue damage, linamarin is converted to toxic hydrogen cyanide. In our collection 44.6 % of the landraces had average seed HCN content ≤200 ppm which is the permitted level for Lima bean seed in the US. Our results also identified the landraces in this collection which have high linamarin levels in the leaves while having low levels in the seeds. Such landraces have the desirable combination of traits and will be the focus of our future plans for agronomic trait improvement though breeding. [ABSTRACT FROM AUTHOR]

One of the major constraints facing the large-scale production of cassava (Manihot esculenta) roots is the rapid postharvest physiological deterioration (PPD) that occurs within 72 h following harvest. One of the earliest recognized biochemical events during the initiation of PPD is a rapid burst of reactive oxygen species (ROS) accumulation. We have investigated the source of this oxidative burst to identify possible strategies to limit its extent and to extend cassava root shelf life. We provide evidence for a causal link between cyanogenesis and the onset of the oxidative burst that triggers PPD. By measuring ROS accumulation in transgenic low-cyanogen plants with and without cyanide complementation, we show that PPD is cyanide dependent, presumably resulting from a cyanide-dependent inhibition of respiration. To reduce cyanide-dependent ROS production in cassava root mitochondria, we generated transgenic plants expressing a codon-optimized Arabidopsis (Arabidopsis thaliana) mitochondrial alternative oxidase gene (AOX1A). Unlike cytochrome c oxidase, AOX is cyanide insensitive. Transgenic plants overexpressing AOX exhibited over a 10-fold reduction in ROS accumulation compared with wild-type plants. The reduction in ROS accumulation was associated with a delayed onset of PPD by 14 to 21 d after harvest of greenhouse-grown plants. The delay in PPD in transgenic plants was also observed under field conditions, but with a root biomass yield loss in the highest AOX-expressing lines. These data reveal a mechanism for PPD in cassava based on cyanideinduced oxidative stress as well as PPD control strategies involving inhibition of ROS production or its sequestration.
