Monoacylglycerol lipase (MAGL) is the enzyme responsible for the metabolism of 2-arachidonoylglycerol in the brain and the hydrolysis of peripheral monoacylglycerols. Many studies demonstrated beneficial effects deriving from MAGL inhibition for neurodegenerative diseases, inflammatory pathologies, and cancer. MAGL expression is increased in invasive tumors, furnishing free fatty acids as pro-tumorigenic signals and for tumor cell growth. Here, a new class of benzylpiperidine-based MAGL inhibitors was synthesized, leading to the identification of 13, which showed potent reversible and selective MAGL inhibition. Associated with MAGL overexpression and the prognostic role in pancreatic cancer, derivative 13 showed antiproliferative activity and apoptosis induction, as well as the ability to reduce cell migration in primary pancreatic cancer cultures, and displayed a synergistic interaction with the chemotherapeutic drug gemcitabine. These results suggest that the class of benzylpiperidine-based MAGL inhibitors have potential as a new class of therapeutic agents and MAGL could play a role in pancreatic cancer.

Treballs Finals de Grau de Química, Facultat de Química, Universitat de Barcelona, Any: 2023, Tutor: Roc Matheu Montserrat, Lead halide perovskites are developed for light-absorbing applications (e.g., solar cells), but concerns about the toxicity of Pb have motivated the research of lead-free perovskites that might be equally as effective as the parent haloplumbates. One promising alternative family of materials consists of double perovskites in which two metals substitute the divalent Pb metal, for example with a monovalent metal and a trivalent metal. A prominent synthetic target are iodide double perovskites, which may display a bandgap much more suitable for solar-cell applications. The synthesis of iodide double perovskite is currently hindered by the crystallization of competing phases with low-dimensional structures with similar chemical composition as the double perovskites. This project proposes the hypothesis of moving away from standard solution methods by using microfluidic chips and thus avoiding competing phases. To do this, the chemical space of three iodide perovskites based on Cs and CH3NH3 as A-sites and Na, Ag, Bi, In and La as B/B’-sites (Cs2AgBiI6, (CH3NH3)2AgBiI6 Cs2AgInI6 and Cs2AgLaI6) was explored. Although it has not been possible to synthesize any iodide double perovskites, new compounds that have not yet been reported have been obtained such as (CH3NH3)InI4 and CsAgI2 as well as already reported ones such as Cs3Bi2I9, (CH3NH3) Bi2I9, CsInI4 and CsI3. It has also been observed that there is a tendency to obtain the same competitive phase for the cases of Bi and In in the B'-site (0D A3Bi2I9 and AInI4 respectively with A = Cs or CH3NH3). Moreover, it has also been observed that despite changing the elements of the B-site (Ag and Na), these did not interfere at all in the formation of the preferred phases in the cases of Bi and In in the B’-site but it did interfere when there was La in the B’-site. Finally, due to the high solubility of the starting reagents in the case of CH3NH3 in the A-site and La in the B’-site crystallization did not occur.

[ES] En el marco de una pandemia mundial producida por el SARS-CoV-2 se reafirma la necesidad imperiosa de continuar las líneas de investigación relacionadas con infecciones producidas por virus. En zonas de América Latina donde la prevalencia de enfermedades tropicales causadas por los virus Dengue-2 (DENV-2) y Zika (ZIKV) continúa presente, se manifiesta un sustancial problema de salud pública. El desarrollo de antivirales derivados de compuestos naturales de producción sostenible aparece como una alternativa para el tratamiento de dichas enfermedades. En el presente trabajo de fin de grado se presenta el desarrollo de la síntesis del antiviral 18-(phthalimid-2-yl) ferruginol como posible principio activo para el tratamiento antiviral en etapas post-infección. El proceso de escalado y validación de la síntesis se traslada a un laboratorio no especializado de las instalaciones de la Universidad de Antioquia, Colombia. The development of antivirals derived from natural compounds of sustainable production is postulated as a possible alternative for the treatment of these diseases. In the present final degree project, the development of the synthesis of the antiviral 18-(phthalimid-2-yl) ferruginol is presented as a possible active substance for antiviral treatment in post-infection stages. The process of scaling and validation of the synthesis is transferred to a non-specialized laboratory of the facilities of the University of Antioquia, Colombia. Cellular targets and non-structural viral proteins of Dengue serotype 2 and Zika viruses are explored, in which the target molecule 18-(phthalimid-2-yl) ferruginol with viral proteins and biological targets are studied by means of an IN-SILICO molecular docking study associated with post-infection stages to interrupt the translation and therefore replication of viral proteins.

Lo scopo di questo progetto di dottorato riguarda la sintesi di una nuova classe di molecole reticolanti per polimeri utilizzati nel mondo degli pneumatici. In particolare, queste molecole devono essere stabili a temperatura ambiente ma in grado reagire termicamente con la matrice polimerica a temperature maggiori di 100 °C, formando legami covalenti irreversibili. Questi funzionalizzanti devono essere anche semplici da sintetizzare, integrabili in un processo industriale, versatili, economici, relativamente poco tossici e in grado di interagire con il polimero senza però influenzarne significativamente le proprietà. In questo contesto è stato realizzato un sistema in grado di generare due specie reattive in due range di temperature diverse: la prima funzionalità genera un carbene, la seconda una specie 1,3-dipolare di tipo nitrilimminico. Entrambe le specie sono in grado di fornire cicloaddotti con sistemi olefinici. Dopo un’attenta ricerca bibliografica al fine di individuare i sistemi più adatti, sono stati sintetizzati sia composti noti in letteratura sia nuovi, cercando di modulare la temperatura di attivazione di queste strutture. Tutti i sistemi sintetizzati sono stati caratterizzati mediante analisi NMR e IR. La caratterizzazione è stata completata dall’analisi termogravimetrica (TGA), con la quale si è rilevata la temperatura di attivazione delle funzionalità presenti; nonchè la temperatura alla quale dovrebbe avvenire la reazione di cicloaddizione. Infine, sui sistemi più interessanti selezionati, sono state eseguite delle prove di reattività su sistemi polimerici modello e facilmente processabili su scala di laboratorio, al fine di dimostrare l’effettiva funzionalizzazione per via termica. Le molecole che presentavano caratteristiche tecnologiche più interessanti sono state preparate in quantità nell’ordine dei grammi necessari a condurre prove in mescola tramite un mini-mescolatore “Brabander”. Queste mescole sono state poi analizzate tramite RPA (Rubber Process Analyzer) per osservare eventuali effetti positivi dei polimeri funzionalizzati. Il sistema a due temperature di attivazione differenti è stato impiegato per studiare possibili effetti sulla morfologia delle fasi all’interno di blend polimerici: si sfrutta la diversa reattività delle componenti carbenica e nitrilimminica cercando di migliorare l’interazione dei due polimeri nonché le proprietà fisiche del materiale risultante. Tutti le mescole sono state caratterizzate anche tramite swelling, trazioni Dumbbell, DSC, DMA, frequency sweep e densità. The aim of this PhD project concerns the synthesis of a new class of cross-linking molecules for polymers used in the world of tires. In particular, these molecules must be stable at room temperature but capable of reacting thermally with the polymer matrix at temperatures greater than 100 ° C, forming irreversible covalent bonds. These functionalizers must also be simple to synthesize, integrable in an industrial process, versatile, economical, relatively low toxic and able to interact with the polymer without significantly affecting its properties. In this context, a system that is able to generate two reactive species in two different temperature ranges was created: the first functionality generates a carbene, the second a 1,3-dipolar species of the nitrilimino type. Both species are capable of providing cycloadducts with olefinic systems. After careful bibliographic research in order to identify the most suitable systems, both known and new compounds were synthesized, trying to modulate the activation temperature of these structures. All synthesized systems were characterized by NMR and IR analyzes. The characterization was completed by thermogravimetric analysis (TGA), with which the activation temperature of the functions present was detected; as well as the temperature at which the cycloaddition reaction should take place. Finally, on the most interesting selected systems, reactivity tests were performed on model polymeric systems that could be easily processed on a laboratory scale, in order to demonstrate the actual functionalization by thermal means. The molecules that presented the most interesting technological characteristics were prepared in quantities in the order of grams necessary to conduct compound tests using a "Brabander" mixer. These compounds were then analyzed by RPA (Rubber Process Analyzer) to observe any positive effects of the functionalized polymers. The system with two different activation temperatures was used to study possible effects on the morphology of the phases within polymeric blends: the different reactivity of the carbene and nitrilimine components is exploited, trying to improve the interaction of the two polymers as well as the physical properties of the resulting material. All compounds were also characterized by swelling, Dumbbell traction, DSC, DMA, frequency sweep and density.

Les transformacions catalitzades per or(I) han estat estudiades rigorosament com a tècniques per a la preparació de molècules complexes. Aquesta Tesi Doctoral cobreix tres temes relacionats amb la catàlisi homogènia d’or(I). Primerament, s’ha dissenyat i preparat una nova família de complexos d’or(I) amb lligands cavitands, quirals i aquirals, partint de precursors resorcin[4]arens. Els aquirals s’han empleat en la cicloisomerització de 1,6-dienins, donant lloc a una selectivitat inèdita. Paral·lelament, els complexos d’or(I) amb cavitands quirals s’han utilitzat en l’alcoxiciclació enantioselectiva de 1,6-enins. La utilitat d’aquest mètode ha estat demostrada en la seva aplicació en la síntesis total de la (+)-mafaicheenamina C. En segon lloc, nous precursors de carbens han estat desenvolupats i empleats en la corresponent reacció de decarbenació catalitzada per or(I). En aquesta, la fragmentació d’un grup ciclopropà dona lloc al corresponent carbè metàl·lic i està afavorida per l’alliberament d’una unitat poliaromàtica. Amb aquest mètode, diversos ciclopropans han estat obtinguts a partir de precursors ciclopropil dihidronaftalens o dihidrofenantrens. Finalment, en tercer lloc, l’habilitat dels complexos d’or(I) de construir arquitectures policícliques ha estat utilitzada per a la síntesis de decalines mitjançant una nova ciclació polienínica emprant éters d’enol tetrasubstituïts. Les decalines obtingudes han estat estudiades com a intermedis clau en la síntesi de diversos productes naturals. Las transformaciones orgánicas catalizadas por oro(I) han sido estudiadas ampliamente como herramientas poderosas para la construcción de moléculas complejas. Esta Tesis Doctoral cubre tres temas relacionados con la catálisis homogénea de oro(I). En primer lugar, se diseñó y preparó la síntesis de nuevas familias de complejos quirales y aquirales de oro(I) con estructuras tipo cavitando, a partir de precursores de resorcin[4]areno. Los complejos aquirales de oro(I) se aplicaron en la cicloisomerización de 1,6-dieninos lo que llevó al descubrimiento de nuevas selectividades. Además, los cavitandos quirales de oro(I) se investigaron en la alcoxiciclación enantioselectiva de 1,6-eninos. Este método se aplicó en la síntesis total de (+)-mafaicheenamina C. En segundo lugar, se desarrollaron nuevos precursores de carbenos no aceptores, que participan en reacciones de descarbenación catalizadas por oro(I) liberando unidades poliaromáticas a partir de ciclopropanos persistentes. Siguiendo esta metodología, se obtuvieron varios ciclopropanos liberando naftaleno o fenantreno como único subproducto. Finalmente, la capacidad de los complejos de oro(I) para construir estructuras policíclicas se demostró con el diseño y desarrollo de un nuevo enfoque para la síntesis de decalinas a través de la ciclación de polieninos de silil enol éteres tetrasustituidos. Las decalinas adquiridas son intermediarios clave en la síntesis de productos naturales. Gold(I)-catalyzed organic transformations have been widely developed as powerful tools for the assembly of complex molecular frameworks. This Doctoral Thesis covers three topics related to homogeneous gold(I) catalysis. First, new families of both chiral and achiral gold(I)-cavitand complexes were designed and prepared from resorcin[4]arene precursors. The achiral gold(I) complexes were applied in the cycloisomerization of 1,6-dienynes and led to uncovering of new selectivities. Furthermore, the chiral gold(I) cavitands were investigated in the enantioselective alkoxycyclization of 1,6-enynes. This method was applied to the total synthesis of (+)-mafaicheenamine C. Second, new types of non-acceptor carbene precursors were developed. These allowed to carry out gold(I)-catalyzed decarbenations of persistent cyclopropanes, releasing polyaromatic units as driving force. Following this methodology, a variety of cyclopropanes could be obtained upon release of naphthalene or phenanthrene. Finally, the ability of gold(I) complexes to construct polycyclic architectures was demonstrated by devising a novel approach for the synthesis of decalins from the polyenyne cyclization of tetrasubstituted silyl enol ethers. The acquired decalins are key intermediates in the synthesis of the natural products.

En aquest treball final de màster, es planifica el mòdul professional de síntesi del CFGM de Sistemes Microinformàtics i Xarxes amb la finalitat que l'alumnat d'aquest cicle formatiu esdevinguin el servei informàtic del centre. Aquesta planificació s'ha realitzant seguint la metodologia d'aprenentatge basat en projectes i la filosofia de l'aprenentatge-servei per tal d'aconseguir un aprenentatge amb sentit i, per tant, perdurable.

Durant les últimes dècades, el potencial de la catàlisis amb or(I) per la síntesi de productes naturals ha estat àmpliament explorat. Aquesta Tesi Doctoral recull els nostres esforços en l'ampliació de l'ús de les cicloaddicions catalitzades per or(I) aplicades en el camp de la síntesi total. Primerament, hem desenvolupat un nou mètode per activar acetilè, en què la selectivitat de la reacció és controlada pel lligand emprat en el catalitzador d'or(I). Així, (Z,Z)-1,3-diens o bis-ciclopropans han sigut sintetitzats de manera selectiva, partint dels mateixos substrats. Aquesta reacció ha estat implementada en la síntesi de la waitziacuminona, en un sol pas i de manera diastereoespecífica. En segon lloc, hem establert una nova cicloisomerització seguida per cicloaddició, en una reacció en cascada que dóna lloc a productes hidroazulens o divinilciclopropans, a partir de diens i enins. El mecanisme ha estat estudiat experimetalment i computacionalment, suggerint una forta dependència del resultat de la reacció en les característiques del diè i en el catalitzador d'or(I) utilitzat. Finalment, aquesta estratègia ha estat aplicada a una ruta síntetica comuna cap als productes: àcid apsterric, penigrisacid A i schisanwilsonene A. Els esquelets de carboni dels tres compostos naturals han sigut sintetitzats en diversos intermedis i caracteritzats per difracció de rajos x. Durante las últimas décadas, el potencial de la catálisis con oro(I) para la síntesis de productos naturales ha sido ámpliament explorado. Esta Tesis Doctoral recoje nuestros esfuerzos en la ampliación del uso de las cicloaddiciones catalitzados por oro(I) aplicadas en el campo de la síntesis total. Primeramente, hemos desarrollado un nuevo método para activar acetileno, en qué la selectividad de la reacción es controlada por el ligando emprado en el catalitzador de oro(I). Así, (Z,Z)-1,3-dienos o bis-ciclopropanos han sido sintetitzados de manera selectiva, partiendo de los mismos sustratos. Esta reacción ha sido implementada en la síntesis de la waitziacuminona, en un solo paso y de manera diastereoespecífica. En segudo lugar, hemos establecido una nueva cicloisomeritzación seguida por cicloaddición, en una reacción en cascada que da lugar a productos hidroazulenos o divinilciclopropanos, a partir de dienos y eninos. El mecanismo ha sido estudiado experimetal y computacionalmente, sugiriendo una fuerte dependencia del resultado de la reacción en las características del dieno y en el catalitzador de oro(I) utilitzado. Finalmente, esta estrategia ha sido aplicada a una ruta síntetica comuna hacia los productos: ácido apstérrico, penigrisacid A y schisanwilsonene A. Los esqueletos de carbono de los tres compuestos naturales han sido sintetitzados en varios intermedios y caracteritzados por difracción de rayos x. The potential of gold(I) catalysis on the synthesis of natural products has been widely explored in the last decades. This Doctoral Thesis presents our efforts to expand the applications of gold(I)-catalyzed cycloadditions on total synthesis. First, we have developed a new methodology for acetylene activation by means of gold(I) catalysis, in which the ligand on the gold(I) catalyst can tune the reaction outcome, either for the formation of (Z,Z)-1,3-dienes or towards bis-cyclopropanes. This method was applied to the one-step total synthesis of waitziacuminone. In second place, we have established a novel cycloisomerization/cycloaddition cascade which allows for the rapid and efficient construction of hydroazulenes and divinylcyclopropanes, from enynes and dienes. The mechanism was studied experimentally and computationally, finding a strong dependence on the nature of the diene and on the gold(I) catalyst employed, for the reaction outcome. Finally, this strategy was applied to a common synthetic route towards aspterric acid, penigrisacid A and schisanwilsonene A. The carbon-skeletons of the three natural products were synthesized and characterized by x-ray diffraction.

Il vaccino BNT162b2 è composto da un RNA avente 4284 nucleotidi, suddivisi in 6 sezioni, che portano le informazioni per creare una fabbrica di proteine S Spike, quelle usate dal Sars-CoV-2 (Covid-19) per infettare il soggetto. Dopodiché, queste proteine vengono dirette al di fuori della cellula, facendo scattare la reazione immunitaria e la produzione di anticorpi. Il problema è la pesante alterazione dell'mRNA: l'Uracile viene sostituito con Ψ (Pseudouridina)per ingannare il sistema immunitario, le lettere di tutte le triplette di codoni vengono sostituite da una C o una G, per aumentare la velocità di produzione delle proteine all'estremo, sostituzione di alcuni aminoacidi con la Prolina, l'aggiunta della sequenza (3'-UTR) con sconosciute alterazioni. Queste modificazioni potrebbero causare forti dubbi circa la presenza di errori nell’uso dei codoni. Un’eventuale errata traduzione ha conseguenze nella fisiopatologia di una varietà di malattie. Inoltre, l’mRNA iniettato è un pre-mRNA, che può portare a multipli mRNA maturi; queste sono le anomalie dello splicing alternativo, sorgente diretta di danni a lungo termine sulla salute umana. In sostanza, ciò che verrà creato potrebbe non essere identico alla proteina S Spike: basta un errore di traduzione, lettura errata dei codoni, produzione errata di aminoacidi, quindi di proteine, per provocare gravi danni a lungo termine alla salute umana, nonostante il DNA non venga modificato, essendo invece nel nucleo cellulare e non nel citoplasma, ove arriva l'mRNA modificato. Tuttavia, in questo caso, la correlazione tra velocità di sintesi ed espressione proteica, oltre al meccanismo che potrebbe inficiare la traduzione della sequenza restano oscuri, in quanto molti trials non sono ancora stati effettuati., FINAL version con domande comuni

El Análisis y la Síntesis son las dos últimas fases de una revisión sistematizada que sigue el Framework SALSA, parte a su vez del (meta)framework ReSiste-CSH que estamos presentando en esta serie de tres capítulos, de la cual este es el último. El Análisis se ocupa de proveer un medio de examen de los documentos que debe ser aplicado a todos por igual, con el objetivo de tener una base de comparación y, sobre todo, de Síntesis. La Síntesis, entonces es la última de las fases en el mencionado Framework SALSA. En un marco más amplio, tendríamos aún una fase posterior de presentación de la revisión o del estado de la cuestión, según el destino concreto de nuestra revisión. The Analysis and the Synthesis are the last two phases of a systematized review that follows the SALSA Framework, part of the ReSiste-CSH (meta) framework that we are presenting in this series of three chapters, of which this is the last one. The Analysis is concerned with providing a means of examining the documents that must be applied to all equally, with the aim of having a base of comparison and, above all, of Sinthesis. The Synthesis, then is the last of the phases in the mentioned SALSA Framework. In a broader framework, we would still have a subsequent phase of presentation of the review or the state/s of the art, according to the specific destination of our review. L’Anàlisi i la Síntesi són les dues últimes fases d’una revisió sistematitzada que segueix el Framework SALSA, part al seu torn de l’(meta) framework resisteix-CSH que estem presentant en aquesta sèrie de tres capítols, de la qual aquest és l’últim. L’Anàlisi s’ocupa de proveir un mitjà d’examen dels documents que ha de ser aplicat a tots per igual, amb l’objectiu de tenir una base de comparació i, sobretot, de Síntesi. La Síntesi, llavors és l’última de les fases en l’esmentat Framework SALSA. En un marc més ampli, tindríem encara una fase posterior de presentació de la revisió o de l’estat de la qüestió, segons la destinació concreta de la nostra revisió.

Any expression of rationality is based upon premises, many of which cannot be ultimately justified. The role of these presuppositions becomes particularly important in the domains of the social sciences and the humanities. A philosophical reflection on the founda-tion methodologies of these disciplines can shed light on how to overcome the rigidity of many present conceptual systems, so as to fully grasp the richness and complexity of human action. Ogni espressione della razionalità si basa su delle premesse, molte delle quali non sono suscettibili di giustificazione in senso assoluto. Il ruolo di questi presupposti diventa particolarmente importante nelle scienze sociali e nelle discipline umanistiche. Una riflessione filosofica sulle metodologie basilari di queste discipline può gettare luce sul modo di superare la rigidità di molti sistemi concettuali moderni, per poi potere comprendere appieno la ricchezza e la complessità dell’agire umano.

[ES] Dada la actual necesidad de innovación a nivel arquitectural, es necesario contar con herramientas y metodologías eficientes para validar y evaluar nuevos diseños. En el ámbito de este TFM se ha diseñado e implementado un procesador RISC-V totalmente parametrizado en un intervalo de tiempo muy reducido. Esto se ha logrado incorporando y desarrollando a lo largo del flujo de trabajo herramientas que han facilitado este proceso, combinando la simplicidad y eficiencia de la simulación con la precisión y utilidad de contar con una implementación hardware. El procesador en sí soporta la ejecución fuera de orden (mediante renombrado de registros explícito y colas de issue), decodificación/lanzamiento/commit superescalar de ancho variable, y genericidad en la mayoría de las estructuras de datos. En cuanto al juego de instrucciones, soporta las principales del ISA RISC-V y varias de sus extensiones. La implementación ha sido validada ejecutando programas de ejemplo sobre múltiples configuraciones. El código se ha planteado para que sea flexible y de fácil expansión: siguiendo nuestra metodología y con las herramientas desarrolladas, futuros proyectos de investigación que consistan en la modificación de una arquitectura podrán ser validados e implementados rápidamente, incluso en un entorno que, al igual que muchos grupos de investigación, esté formado por gente con bases y conocimientos distintos. En esa línea, este trabajo ya ha allanado el terreno para experimentar con ideas más innovadoras, y se proporciona un ejemplo de esto: el desarrollo de una extensión sobre nuestro núcleo que añade funcionalidades propias de un sistólico., [EN] Given the dire need for architectural innovation, efficient tools and methodologies are required to validate, test, and evaluate new designs. For this Master's Thesis we designed and implemented a fully parameterized RISC-V core in a very restricted timeframe. We achieved this by means of a toolflow that combines the simplicity and efficiency of simulation with the accuracy and general usefulness of an actual hardware implementation. The core itself supports Out-of-Order execution (by means of explicit register renaming and Issue Queues), superscalar instruction decode/issue/commit with variable width, and most of its data structures are generic. As for the instruction set, it can handle the main instructions of the base RISC-V ISA and several of its extensions. Its correctness has been validated with sample programs running on multiple configurations. The code has been engineered with flexibility and expandability in mind: aided by the rest of said toolflow, future research that involves modifying the architecture can be validated and implemented as soon as possible, even in an environment that, as many research groups, is formed by people of many different backgrounds. In that regard, it has already laid the groundwork to test more innovative ideas, and an example of this is provided in the form of a systolic extension to our base core., [CA] Amb l’actual necessitat d’innovació a nivell arquitectural, és necessari comptar amb ferramentes i metodologies eficients per a validar i avaluar nous dissenys. En l’àmbit d’este TFM s’ha dissenyat i implementat un processador RISC-V totalment parametritzat en un interval de temps molt reduït. Açò s’ha aconseguit incorporant i desenvolupant al llarg del flux de treball ferramentes que han facilitat este procés, combinant la simplicitat i eficiència de la simulació amb la precisió i utilitat de comptar amb una implementació hardware. El processador en si suporta l’execució fora d’orde (amb register renaming explícit i cues d’issue) , decodificaci ó/llançament/commit superescalar d’ample variable, i genericitat en la majoria de les estructures de dades. Quant al joc d’instruccions, suporta les principals de l’ISA RISC-V i diverses de les seues extensions. La implementació ha sigut validada executant programes d’exemple sobre múltiples configuracions. El codi s’ha plantejat perquè siga flexible i de fàcil expansió: seguint la nostra metodologia i amb les ferramentes desenvolupades, futurs projectes d’investigació que consistisquen en la modificació d’una arquitectura podran ser validats i implementats ràpidament, inclús en un entorn que, igual que molts grups d’investigació, estiga format per gent amb bases i coneixements distints. En eixa línia, este treball ja ha aplanat el terreny per a experimentar amb idees més innovadores, i es proporciona un exemple d’açò: el desenvolupament d’una extensió sobre el nostre nucli que afig funcionalitats pròpies d’un sistólic.

Els greixos vegetals són una important matèria primera renovable per a la indústria química. Dins d'aquest grup destaquen els àcids grassos homoalílics hidroxilats com l’àcid ricinoleic o l’àcid lesqueròlic, els quals donen lloc a una gran quantitat de derivats amb múltiples aplicacions tant industrials com farmacèutiques. Aquesta tesi es va centrar, d’una banda, en l’obtenció d’àcids grassos carbonil α,β-insaturats mitjançant oxidacions més selectives i en un sol pas del grup hidroxil d’aquests àcids. D’altra banda, es va procedir a l’obtenció de l’àcid sebàcic, un àcid dicarboxílic amb gran interès industrial, a partir dels àcids grassos carbonil α,β-insaturats anteriorment sintetitzats. Finalment, es van dissenyar i sintetitzar noves N-vanillil acilamides de cadena llarga a partir dels àcids ricinoleic i lesqueròlic amb l’objectiu de desenvolupar una nova família de molècules que presentessin activitat biològica dual davant de receptors del dolor com són el receptor TRPV1 i l’enzim FAAH. Las grasas vegetales son una importante materia prima renovable para la industria química. Dentro de este grupo destacan los ácidos grasos homoalílicos hidroxilados como el ácido ricinoléico o el ácido lesquerólico, los cuales dan lugar a una gran cantidad de derivados con múltiples aplicaciones tanto industriales como farmacéuticas. Esta tesis se centró, por un lado, en la obtención de ácidos grasos carbonilo α,β-insaturados a través de oxidaciones más selectivas y en un solo paso del grupo hidroxilo de dichos ácidos. Por otro lado, se procedió a la obtención del ácido sebácico, un ácido dicarboxílico con gran interés industrial, a partir de los ácidos grasos carbonilo α,β-insaturados anteriormente sintetizados. Por último, se diseñaron y sintetizaron nuevas N-vanillil acilamidas de cadena larga (LCNVAs) a partir de los ácidos ricinoléico y lesquerólico con el objetivo de desarrollar una nueva familia de moléculas que presentaran actividad biológica dual frente a receptores del dolor como son el receptor TRPV1 y la enzima FAAH. Vegetable oils are an important renewable raw material for the chemical industry. Homoallyl hydroxy fatty acids such as ricinoleic or lesquerolic acid are highlighted within this oils. They lead to a large number of derivatives with multiple applications, both industrial and pharmaceutical. This work was focused on obtaining and using α,β-unsaturated carbonyl fatty acids through selective processes. First at all, an one-step oxidations of the hydroxy group of these acids were studied. Subsequently, sebacic acid was obtained from the α,β-unsaturated carbonyl fatty acids previously synthesized. This dicarboxylic acid has a great industrial interest. Finally, new long-chain N-vanillyl acylamides (LCNVAs) were designed and synthesized from ricinoleic and lesquerolic acid with the aim of developing a new family of molecules exhibiting dual biological activity against pain receptors such as the TRPV1 receptor and the FAAH enzyme.

Esta tesis aborda la síntesis, caracterización y funcionalización de nanoestructuras magnéticas biocompatibles y anisótropas de óxido de hierro (Fe3O4) para su aplicación en diagnóstico biomédico mediante imagen de resonancia magnética (MRI) y uso terapéutico en dos modalidades de hipertermia: magnética y fototérmica. Para ello, se escogieron dos tipos de estructuras: nanocubos y nanorods. Para sintetizar los nanocubos, se probaron varios métodos ya publicados. Sin embargo, ninguno de ellos proporcionó resultados completamente satisfactorios en cuanto a monodispersión de tamaños, reproducibilidad, pureza de fase, alta cristalinidad y definición de forma. Por ello, desarrollamos una estrategia nueva basada en la introducción de oleato de sodio y una mezcla de disolventes que permitían el control de la temperatura de reflujo y la polaridad del medio, lo que además mejoró la estabilidad química del entorno en el que tenía lugar el crecimiento, dando lugar a una síntesis más reproducible. Estos resultados mostraron el éxito a la hora de producir partículas cúbicas en un rango de tamaños muy amplio, con unas excelentes propiedades y reproducibilidad. En cuanto a los nanorods, la síntesis fue especialmente complicada, ya que la estructura cúbica del Fe3O4 dificulta la formación de morfologías tan alargadas. De entre todos los procedimientos probados, solo la síntesis solvotermal dio buenos resultados. Para tener un mejor control sobre el tamaño y la relación de aspecto, se desarrollaron nuevas estrategias basadas en el ajuste de la presión y del ratio entre surfactantes. La superficie de las partículas sintetizadas es hidrófoba y por tanto fue necesario modificarla para que éstas pudieran dispersarse en medios biológicos. Además, el recubrimiento de las partículas debería proporcionar grupos funcionales para conjugar biomoléculas y así dirigirlas contra células malignas. Se probaron varias estrategias y los resultados mostraron que, a pesar de que la repulsión electrostática puede ser suficiente para estabilizar nanopartículas pequeñas o no magnéticas, en nuestro caso era necesario combinarla con impedimento estérico para evitar la agregación irreversible. Con este fin, se desarrolló un nuevo procedimiento de encapsulación basado en la formación de bicapas lipídicas que, a pesar de dar resultados prometedores, fue descartado finalmente al tener en cuenta el tiempo que se necesitaría para optimizar completamente todo el protocolo. En su lugar, se usó un procedimiento basado en la encapsulación con copolímeros anfipáticos, que también dio unos resultados excelentes, garantizando la estabilidad coloidal en entornos biológicos. El potencial biomédico de las partículas se evaluó primero como herramienta diagnóstica midiendo el contraste T2 para resonancia magnética de partículas de diferentes tamaños y formas, resaltando el mayor contraste de las nanopartículas anisótropas respecto a las isótropas (esferas). En cuanto al uso terapéutico, se evaluó también el potencial de las partículas en hipertermia magnética. Los resultados mostraron una buena capacidad de calentamiento a pesar de las suaves condiciones que usamos en nuestro estudio. Además, gracias a un amplio estudio espectroscópico teórico y experimental, se vio que las nanopartículas de Fe3O4 son adecuadas para fototermia, sobre todo en la segunda ventana biológica del infrarrojo cercano (1000-1350 nm). Esta región espectral es especialmente interesante porque permite la aplicación de mayores potencias de irradiación y tiene una mayor penetración en los tejidos humanos. A 1064 nm se consiguieron eficiencias de calentamiento óptico similares a los mejores agentes fototérmicos. Además, se aprovecharon las anisotropías magnética y óptica para medir la temperatura local en tiempo real mediante un método relativamente nuevo. Los experimentos in vitro usando células tumorales HeLa demostraron que las nanopartículas son internalizadas fácilmente y que no son tóxicas para concentraciones inferiores a 4 mM de hierro y que la fototermia usando nanocubos de Fe3O4 es una terapia excelente para destruir células tumorales. This thesis tackles the synthesis, characterisation and functionalisation of biocompatible anisotropic iron oxide (Fe3O4) magnetic nanostructures for their application in biomedical diagnosis by means of magnetic resonance imaging (MRI) and therapy by two different modalities of hyperthermal therapy: magnetic fluid hyperthermia and photothermia. Two different types of structures were chosen for these purposes: nanocubes and nanorods. Several approaches published in literature were tested to synthesize the nanocubes. However, none of them rendered fully satisfactory results in size monodispersity, reproducibility, phase purity, high crystallinity and well-defined shape. Thus, we developed a new strategy based on the introduction of sodium oleate and a solvent mixture enabling the control of the reflux temperature and the polarity of the medium, which also resulted in an improvement of the chemical stability of the growth environment, leading to a more reproducible synthesis. The results demonstrate the successful synthesis of highly cubic particles in a very broad size range, with excellent properties and reproducibility. Concerning the nanorods, their synthesis was particularly challenging since the cubic crystal structure of Fe3O4 complicates the formation of such elongated morphologies. Among all the tested procedures, only the solvothermal synthesis provided good results. To have a better control on the size and aspect ratio new approaches based on adjusting the pressure and surfactants have been developed. The surface of the freshly synthesized particles is hydrophobic and therefore it was necessary to modify the surface to make them dispersible in biological media. In addition, the coating should provide functional groups to attach biomolecules for targeting malignant cells. Several approaches were tested and the results showed that, despite electrostatic repulsion can be enough to stabilize smaller or non-magnetic nanoparticles, in our case it was necessary to combine it also with steric hindrance to avoid irreversible aggregation. For this purpose, a novel procedure based on the formation of a lipid bilayer coating was developed which, despite providing promising results, was eventually discarded considering the time that would be required to fully optimise the protocol. Instead, a procedure based on the coating with amphiphilic copolymers was used, which also provided excellent results, ensuring colloidal stability in biological environments. The biomedical potential of the particles was evaluated first as a diagnostic tool by measuring the MRI T2 contrast of particles of different sizes and shapes, evidencing the enhanced contrast of anisotropic nanoparticles with respect to isotropic ones (spheres). In terms of therapy, the potential of the particles in terms of magnetic hyperthermia was also evaluated. The results showed the good heating capacity of the particles despite the mild conditions used in our study. In addition, thanks to a comprehensive theoretical and experimental spectroscopic study, it was established that Fe3O4 nanoparticles are suitable for photothermia, particularly in the near infrared second biological window (1000-1350 nm). This spectral range is especially appealing because it allows the application of higher powers and has a deeper penetration in human tissues. At 1064 nm were measured some heating efficiencies similar to the best photothermal agents. In addition, the magnetic and optic anisotropies were exploited for a relatively new approach for in situ local temperature sensing. The in vitro experiments using HeLa cancerous cells demonstrated that the nanoparticles are easily internalized and are not toxic for concentrations below 4 mM Fe and that photothermia using Fe3O4 nanocubes at 1064 nm is an excellent therapy for destroying cancerous cells.

La present tesi descriu la preparació d’una nova família de fosfines carboxíliques-trifluorometilades. Aquests lligands han sigut sintetitzats mitjançant la reacció d’hidròlisi del grup trifluorometil en triarilfosfines trifluorometilades amb àcid sulfuric fumant i àcid bòric. Aquesta reacció ofereix una nova aproximació per a l’obtenció de fosfines carboxíliques fent servir un procediment relativament simple. L’abast i l’aplicabilitat de la reacció s’ha estudiat en un grup de vuit monofosfines aríliques trifluorometilades, tant homolèptiques com heterolèptiques, i dues difosfines aríliques trifluorometilades. Amb les fosfines homolèptiques s’ha trobat que la velocitat de la reacció d’hidròlisi del trifluorometil depèn del patró de substitució respecte a l’àtom de fòsfor, amb velocitats majors quan aquests grups són en posició meta que quan es troben en para. La fosfina orto-substituïda mostra velocitats molt més lentes que les dues anteriors i la formació de compostos de tipus oxifosforans espirocíclics. Els grups trifluorometil situats en posicions 3,5 també mostren velocitats de reacció molt lentes amb la conseqüent oxidació de la fosfina. Les fosfines heterolèptiques estudiades van mostrar que la velocitat de la reacció disminueix en augmentar el nombre de grups trifluorometil en les fosfines. Amb aquest procediment s’han preparat, purificat i caracteritzat completament vuit noves fosfines carboxíliques-trifluorometilades. Per contra, la reacció d’hidròlisi del trifluorometil no va permetre obtenir fosfines carboxíliques a partir de les difosfines trifluorometilades. La reacció d’aquestes amb òleum dóna lloc als corresponents òxids de fosfina. Addicionalment s’ha analitzat en detall la reacció d’hidròlisi del trifluorometil. Aquest anàlisi inclou l’estudi de les espècies implicades a la reacció i el seu paper en la mateixa, així com el mecanisme de formació del grup àcid carboxílic a partir del grup trifluorometil. En aquest context, s’ha proposat també un possible mecanisme de formació dels compostos oxifosforats espirocíclics. Les monofosfines carboxíliques-trifluorometilades s’han fet servir en la síntesi de difosfines quelatants mitjançant la reacció de dues fosfines amb una diamina, amb la formació d’enllaços amida. A l’estructura d’aquestes difosfines s’ha introduït un grup vinil amb la intenció d’obtenir difosfines suportades a partir de la copolimerització d’aquestes amb partícules de làtex. Les difosfines sintetitzades s’han assajat com a lligands a la reacció d’hidroformilació del 1-octè catalitzada per Rh, mostrant la seva capacitat per actuar com a lligands quelat. S’han observat regioselectivitats de fins al 97 % d’aldehid lineal front el total d’aldehids formats a la reacció. The present thesis describes the preparation of a new family of carboxylic-trifluoromethylated phosphines. These ligands have been synthesised by applying the reaction of hydrolysis of the trifluoromethyl group in trifluoromethylated triarylphosphines using fuming sulfuric acid and boric acid. This reaction offers a new approach to obtain carboxylic phosphines by applying a relatively simple synthetic procedure. The scope and the applicability of the reaction have been studied in a set of eight trifluoromethylated aryl-monophosphines, both homoleptic and heteroleptic, and two trifluoromethylated aryl-diphosphines. In the homoleptic aryl-monophosphines, the rate of the reaction of hydrolysis of the trifluoromethyl was found to depend on the pattern of substitution with respect to the phosphorus atom, it being faster in meta-position than in para. The ortho-substituted phosphine showed much slower rates than the meta and para-compounds together with the formation of spirocyclic oxyphosphoranes. Slow rates were also observed in the reaction of hydrolysis in trfluoromethyl groups in 3,5-position with the resulting oxidation of the phosphine. The heteroleptic phosphines showed that the reaction rate was slower when increasing the number of trifluoromethyl groups in the phosphines. With this procedure, eight new carboxylic-trifluoromethylated phosphines were prepared, purified and fully characterised. In contrast, no carboxylic phosphines could be prepared from the hydrolysis of the trifluoromethyls in the trifluoromethylated diphosphines. Difficulties were found in the protection of these phosphines which in contact with oleum yielded the corresponding phosphine oxides. Moreover, the reaction of hydrolysis of the trifluoromethyl group was analysed in detail, including the study of the species involved in the reaction and their role as well as the mechanism of formation of the carboxylic acid from the trifluoromethyl group. In this context, the mechanism of formation of the spirocyclic oxyphosphoranes was proposed. The carboxylic-trifluoromethylated monophosphines were used in the preparation of chelating diphosphines by reaction with a diamine leading to amide linkages. Vinyl moiety was included in the structure of the diamine in order to eventually form a polymer-supported diphosphine by co-polymerisation with latex particles. The study of these ligands in the Rh-catalysed hydroformylation of 1-octene showed the chelating behaviour of the phosphines with regioselectivities up to 97 %, expressed as the percentage of linear aldehyde relative to total aldehydes obtained.

Materiales orgánicos basados en moléculas pequeñas con propiedades optoelectrónicas son particularmente atractivos en los campos de celdas solares orgánicas y en el campo de la electrónica molecular. Porfirinas y curcuminoides (CCMoids) son moléculas que suscitan interés, siendo buenas candidatas en los campos mencionados, debido a que presentan estructuras químicas modificables y excelentes propiedades electrónicas. En esta tesis, se han estudiado la preparación y diseño de moléculas pertenecientes a las dos familias aquí indicadas, la capacidad de autoensamblaje de dichas moléculas conjuntamente con sus posibles aplicaciones. En el capítulo II, se han sintetizado dos metaloporfirinas (Zn(4R-PPP) y Zn(PPP)) ligandos largos que contienen centros quirales o aquirales conjutantemente con grupos carbonilos en las cuatro posiciones meso de sus respectivos núcleos de porfirina. Posteriorment, se ha sintetizado un estudio completo relacionado con las interacciones intermoleculares no covalentes de dichas multiporfirinas y experimentos iniciales con la creación de celdas solares orgánicas de hetero-unión (BHJ-OCs) donde este tipo de porfirinas han sido procesadas en solución para el estudio de su comportamiento como sistemas donadores en dichas celdas. Dicho estudio se extendió en el capítulo III a dos nuevas porfirinas (TEP y Zn (TEP)) con sustituyentes carbonilos pero más cortos donde se investigó el efecto de las dimensiones de los ligandos en las interacciones intermoleculares finales, y cómo esto afecta el rendimiento de OSCs. En el capítulo IV se explica como con los mismos centros quirales en las posiciones meso, se obtuvo una nueva porfirina, Zn(4R-CPP), que contiene grupos carboxílicos. También se muestran los estudios con dicho sistema y porfirinas adicionales con la formación de sistemas de autoensamblaje binarios basados en interacciones no covalentes o de autoensamblaje iónico para su posible aplicación como nanomateriales. En el Capítulo V, se sintetizaron CCMoides que contienen grupos quirales, de forma similar al capítulo II, así como el estudio del efecto de la quiralidad de dichos materiales. Además, la investigación hace énfasis en el logro de grupos ácidos terminales a partir de la hidrolización de los grupos ésteres (como se muestra en el capítulo IV) que permitió investigar la posible creación de sistemas con diferente dimensionalidades. Después, en el Capítulo VI, se exploró la síntesis de CPs /redes mediante el uso de CCMoides que contienen grupos piridínicos en sus terminaciones (3Py-CCM). El último capítulo está dedicado al diseño de nuevos derivados de porfirina que contienen grupos de anclaje basados en azufre para su aplicación en electrónica molecular conjuntamente con el estudio de sus propiedades electrónicas en solución y en estado sólido. Además, una familia de CCMoids también se ha analizado de manera similar a los derivados de porfirina, con el objetivo de recopilar información para mejorar su diseño molecular para aplicaciones electrónicas. Organic small molecules materials with optoelectronic properties are particularly attractive in the fields of organic solar cells and molecular electronics. Porphyrins and curcuminoids (CCMoids) are prospective candidates in these fields due to their modifiable chemical structures and outstanding properties. In this thesis, the design and preparation of these two families of molecules, together with their self-assembly abilities and potential applications have been studied. In chapter II, two metalloporphyrins (Zn(4R-PPP) and Zn(PPP)) containing long chiral or achiral moieties with carbonyl substituents in the four meso-positions of their porphyrin cores have been synthesized. Then, a complete study related to non-covalent multiporphyrin assemblies has been performed, and initial solution-processed bulk heterojunction organic solar cell experiments were presented. To extend the above study, in chapter III, new porphyrins (TEP and Zn(TEP)) with shorter carbonyl substituents have been investigated and the effect of the length of the ligands in intermolecular interactions was studied, searching how this factor affects as well the OSCs performance. With the same chiral centres in the meso-positions, a porphyrin Zn(4R-CPP) involving carboxylic groups was obtained in chapter IV. And the binary self-assembling systems based on its derivations were achieved through non-covalent interaction or ionic self-assembly towards their potential application as active components in nanomaterials. In Chapter V, CCMoids containing chiral groups, in a similar manner as chapter II, were synthesized. In addition, research towards the achievement of terminal acid groups from the hydrolyzation of the ester groups (as chapter IV shows) allowed the investigation of the possible creation of systems with different dimensionalities. Then in Chapter VI, the synthesis of CPs/networks was explored by the use of a CCMoid containing pyridine moieties at its endings (3Py-CCM). The last chapter is devoted to the design of new porphyrin derivatives containing sulphur-based anchoring groups for their application in single molecular electronics together with the study of their electronic properties in solution and solid state. In addition, a family of CCMoids has also been analysed in a similar manner as the porphyrin derivatives, with the aim of gathering information to improve their molecular design for electronic applications.

The development of novel materials for energy harvesting applications or strain sensing has generated great interest towards zinc oxide (ZnO) nanostructures, and in particular towards the synthesis of ZnO nanowires or nanorods with well controlled morphology and properties. The high-yield mass production of such nanostructures by catalyst-free methods is a crucial aspect to enable a cost-effective large-scale development of new ZnO-based piezoelectric devices and materials. In the present work, we propose a method for the mass-production of high-purity ZnO-nanorods with a uniform size distribution, based on the combination of thermal decomposition of zinc acetate dihydrate and probe sonication in acetone. The quality of the produced ZnO nanorods is assessed through multi-technique characterization using field-emission scanning electron microscopy (FE-SEM), X-ray diffraction (XRD), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), and photo-luminescence spectroscopy (PL). The adopted synthesis method is simple, cost effective and feasible for large-scale production. Various process parameters such as precursor amount and growth time have been found to play an important role in controlling the formation of the as grown nanostructures with high uniformity in size and morphology. Size distribution curves were employed to depict the effect of various process parameters for tailoring the morphology, homogeneity and aspect ratio of the nanorods. Our results reveal that the high crystallographic quality of ZnO nanorods grown by a long-time thermal decomposition method is not affected by probe sonication, which is proposed as a post-synthesis step necessary to produce ZnO nanorod powder with a uniform distribution of diameters and lengths. © 2015 The Royal Society of Chemistry.

La caracterización de arreglos de micro hilos de óxido de Zinc (ZnO) se ha realizado con la finalidad de diseñar una batería experimental que utiliza la energía de deformación como fuente de energía. El principio de almacenamiento está basado en la compresión simple de un muelle cuya fuerza de compresión se encuentra asociada a la energía que es cosechada. Mediante el mismo principio se propone la compresión de un arreglo de nano hilos de ZnO crecidos de forma alineada perpendiculares al sustrato de crecimiento, funcionando como muelles elásticos que almacenen energía en un medio enteramente mecánico. El ZnO se ha elegido por ser un material en el que se pueden crecer nano estructuras con forma de hilos, barras y filamentos de manera sencilla y de bajo coste a través de un método de síntesis llamado método hidrotérmico. Tomando en cuenta las mejores muestras alineadas de micro hilos de ZnO, se realizó un proceso para su caracterización mecánica en dos fases principales. La primera fase consiste en el estudio estadístico para dimensionar su morfología y tamaños así como su densidad de crecimiento media por toda la muestra. En segundo lugar se llevó a cabo la realización de la aplicación de la teoría lineal elástica que después fue corroborada y replicada con simulaciones de elementos finitos (FEM). Las condiciones de contorno consideradas como comportamientos posibles en los micro hilos de ZnO fueron libre-empotrado y articulado-empotrado. Finalmente los micro hilos se sometieron a pruebas de compresión simple utilizando un nanoindentador comercial, para llevar a cabo el análisis experimental puntual de las propiedades del material del que se conforman y así determinar la discrepancia con el modelo teórico así como su capacidad de almacenamiento de energía real., The synthesis and characterization for a ZnO array have been done in this research, with the aim of design an experimental mechanical battery, which uses the strain energy as energy source. The storage principle is based on a common spring, which is loaded by the force associated to the energy to be harvested. By following the same principle our approach is based on the compression of a ZnO fine wires array (ZnOfws), grown vertically on a substrate surface, working as regular springs and storing energy entirely on the mechanical domain. The ZnO has been chosen as a “spring-like” material, due to the fact the fine wires growth is cheap and simple, using an aqueous synthesis called “hydrothermal method”. By taking the best aligned ZnO array obtained, the dimensions measurement was carried out through a statistically method, based on the SEM images analysis. From this study the average growth density, diameters and lengths were obtained. Afterwards, a mechanical study for a single fine wire was done, applying the linear elastic theory and FEM simulations. In order to, analyze the most likely mechanical behaviors for a single ZnOfw. Finally, the fine wires samples were submitted to a mechanical characterization through the nanoindentation technique, with the objective of see the real deformation levels and, mechanical energy storage capability that they have. As an alternative to this measurement a customized compression set-up was fabricated, in order to, analyze the mechanical behavior of a ZnOfws array at a larger scale.

L’architettura scavata è letteralmente il risultato di un processo di sottrazione di materia, ma può essere anche intesa come l’esito di uno scavare messo in atto solo in quanto operazione concettuale. Per definizione massiva e pesante, l’architettura scavata contempla incredibilmente tra i suoi attributi principali la leggerezza, che si manifesta nella supremazia dello spazio vuoto sottratto rispetto al pieno. Questa prima intuizione di ambivalenza suggerisce il riferimento proprio all’idea di ambiguità come filo conduttore di un’indagine che consenta di schiudere i significati più autentici di tale atteggiamento progettuale. Il discorso prende l’avvio dalla considerazione dell’antefatto della caverna, luogo archetipico contemporaneamente partecipe dei significati simbolici dell’alto e del basso; prosegue, poi, nella riflessione sull’architettura scavata, espressione della stereotomia, nella quale l’uniformità materiale dello spazio è sottilmente negata dallo strutturarsi materico della luce e dell’aria; e termina, infine, nell’analisi di quell’architettura in cui gli scavi sono solo concettuali e rinsaldano il primato dell’interno, che però inevitabilmente si dispiega solo nel rapporto di opposizione con il limite che lo racchiude. Le contraddizioni si sublimano nella definizione dello spazio, che rappresenta un momento di sintesi. Parallelamente alla trattazione teorica si sviluppa il racconto di un progetto per la città di Sines: la volontà di collegarne le parti alta e bassa, separate da una falesia, porta alla composizione di un baluardo che si contrappone alla roccia, disegna una piazza alla quota elevata, e si conforma come una porta simbolica di accesso dal mare alla città e viceversa. I percorsi di collegamento verticale, insieme ad uno spazio per performance artistiche, sono scavati all’interno di tale volume.

El KRN7000 (α-GalCer) és un glicoesfingolipid sintètic que exhibeix una potent activitat antitumoral estimulant una forta resposta immune. El mecanisme bioquímic del KRN7000 per a aquest efecte terapèutic passa per la formació d'un complex ternari entre la proteïna CD1d, el mateix KRN7000 i el receptor TCR de les cèl•lules NKT. Després d'aquesta complexació, s'alliberen diferents tipus de citoquines donant lloc a diferents respostes biològiques. Es creu que tant la potència com la naturalesa del resultat biològic final depenen de la força de les interaccions entre els components del complex ternari. En aquest sentit, recentment s'ha demostrat que les cadenes perfluorades produeixen interaccions més fortes amb les proteïnes que els seus homòlegs hidrogenats, degut a la seva major hidrofòbia. En aquest context, la recerca descrita en aquesta tesi pretén investigar la síntesi d'anàlegs del KRN7000 en els quals les cadenes lipídiques hidrocarbonades es reemplacen per cadenes polifluorades per determinar el resultat biològic d'aquesta modificació estructural. Per assolir tal propòsit, s'han estudiat diferents mètodes de fluoroalquilació donant lloc a la preparació d'una bateria d'anàlegs del KRN7000 que difereixen tant en la posició de les cadenes fluorades com del grau de fluoració del compost. A més, durant el desenvolupament d'aquesta tesi, va sorgir un objectiu addicional: la síntesi d'anàlegs del KRN7000 restringits conformacionalment. En aquesta tesi també es presenta l'avaluació biològica preliminar dels anàlegs sintetitzats que evidencien l'alt impacte de la substitució H / F en l'activitat del compost. A més, també s'han dut a terme estudis de dinàmiques moleculars, que proporcionen informació útil per entendre, a nivell molecular, el comportament de les cadenes fluorades en el sistema biològic. En este contexto, la investigación descrita en esta tesis pretende investigar la síntesis de análogos del KRN7000 en los que las cadenas lipídicas hidrocarbonadas se reemplazan por cadenas polifluoradas para determinar el resultado biológico de esta modificación estructural. Para alcanzar tal propósito, se han estudiado diferentes métodos de fluoroalquilación dando lugar a la preparación de una batería de análogos del KRN7000 que difieren tanto en la posición de las cadenas fluoradas como en el grado de fluoración del compuesto. Además, durante el desarrollo de esta tesis, surgió un objetivo adicional: la síntesis de análogos del KRN7000 restringidos conformacionalmente. En esta tesis también se presenta la evaluación biológica preliminar de los análogos sintetizados que evidencian el alto impacto de la sustitución H/F en la actividad del compuesto. Además, también se han llevado a cabo estudios de dinámicas moleculares, que proporcionan información útil para entender, a nivel molecular, el comportamiento de las cadenas fluoradas en el sistema biológico. KRN7000 (α-GalCer) is a synthetic glycosphingolipid which exhibits potent anti-tumor activity by stimulating a strong immune response. The KRN7000 biochemical mechanism for such therapeutical effect goes through the formation of a ternary complex between CD1d protein, KRN7000 and TCR receptor of NKTcells. Upon complexation, different types of cytokines are released promoting different biological responses. It is believed that both the potency and the nature of the final biological outcome depend on the strength of the interactions between the components of the ternary complex. In this sense, it has been recently demonstrated that perfluorinated chains produce stronger interactions with proteins than its hydrocarbon counterparts, due to their higher hydrophobicity. Within this context, the research described in this thesis aims to investigate the synthesis of KRN7000 analogues in which lipid hydrocarbon chains are replaced by fluoroalkyl ones in order to determine the biological outcome of such structural modification. For this purpose, different fluoroalkylation methods have been tested leading to the successful preparation of a battery of KRN7000 analogues differing on both the position of fluoroalkyl chains and the compound fluorination degree. In addition, during the development of this thesis, an additional objective aroused: the synthesis of coformationally restricted KRN7000 analogues. Preliminar biological evaluation of the synthesized analogues is also presented herein evidencing the high impact of H/F replacement on the compound activity. In addition, molecular dynamic studies have been performed, providing useful information for the understanding, at a molecular level, of the behaviour of the fluoroalkyl chains in the biological system.

[EN] Prototips Mínims is the title for a series of artworks, the result of a theoretical and practical research. The technique used is silkscreen on paper. The study has focused on the search for formal minimization by reducing the color and form. Everything that happens in the support is separated from its context and symbolic implications, and the results become autonomous objects with their own language and rules. The works are prototypes, not original or final products of an artistic process; they are, instead, starting points open to collaborations, leaving aside the myth of the individualistic artwork, unique and inimitable., [CA] Prototips mínims és el títol que dóna nom a una sèrie de treballs plàstics fruit d’una investigació teòrica-pràctica. La tècnica utilitzada és la serigrafia sobre paper. L’estudi s’ha centrat en la cerca de la minimització formal mitjançant la reducció del color i la forma. Tot allò que succeeix al suport es separa de les seues implicacions contextuals i simbòliques, i els resultats es converteixen en objectes autònoms amb unes regles i un llenguatge propi. Els treballs són prototips, ni originals ni productes finals d’un procés artístic; són, al contrari, punts de partida oberts a col·laboracions, deixant de costat el mite de l’obra d’art individualista, única i inimitable.

El receptor de quimiocina CXCR4 ha resultat ser una diana terapèutica de principal importància des del descobriment de la seva funció en l’etapa d’unió del virus del VIH amb la cèl·lula hoste en el seu cicle de replicació i el seu paper en una gran varietat de processos tumorals en la seva expansió en l’organisme mitjançant metàstasi. Des del descobriment, l’any 1990, del biciclam AMD3100 (Plerixafor, EC50= 0.002 µM), la primera molècula petita inhibidora de CXCR4, s’han dedicat grans esforços en trobar-ne anàlegs més potents, selectius i menys tòxics. El Grup d’Enginyeria Molecular (GEM) d’IQS ha treballat prèviament amb anàlegs d’AMD3100 dissenyant i sintetitzant diverses quimioteques de tetraamines potencialment inhibidores de CXCR4. D’elles n’han sorgit diversos compostos amb resultats anti-VIH prometedors. En el present treball s’ha optat per realitzar diverses modificacions en aquests caps de sèrie del grup amb l’objectiu d’ampliar l’espai químic al seu voltant i millorar aquests resultats. En primer lloc, es dissenyen i es sintetitzen dues noves famílies que contenen dos espaiadors diferents als utilitzats fins al moment. Un primer espaiador fenílic, com el que conté l’AMD3100 però no simètric, amb una cadena d’una sola baula i una de dues baules metilèniques entre l’anell fenílic i el primer nitrogen de banda i banda de l’anell. El segon és un espaiador simètric amb un anell naftalènic central. En segon lloc, s’amplia la diversitat molecular en dissenyar noves tetraamines amb l’espaiador p-fenilbismetilènic que implementen nous substituents en busca d’una millora en la capacitat d’inhibició de CXCR4. En aquesta cerca, es dissenya una nova ruta sintètica que permet incloure la funcionalitat èster en l’anell heterocíclic dels extrems de la tetraamina, no accessible amb les metodologies usades fins al moment. Finalment, es realitzen assaigs d’inhibició del receptor CXCR4, proliferació cel·lular i anti-VIH així com estudis computacionals per estudiar el mode d’acció d’algun d’aquests compostos front el receptor CXCR4 i com actuen en diverses malalties amb les que està relacionat, tals com la infecció del VIH, el càncer de pàncrees o la leucèmia., El receptor de quimosina CXCR4 ha resultado ser una diana terapéutica de principal importancia desde el descubrimiento de su función en la etapa de unión del virus del VIH con la célula huésped durante el ciclo de replicación y su papel en una gran variedad de procesos tumorales en su expansión en el organismo a través de la metástasis. El Grup d’Enginyeria Molecular (GEM) de IQS ha trabajado previamente con análogos de AMD3100 diseñando y sintetizando diversas quimiotecas de tetraaminas potencialmente inhibidoras de CXCR4. De ellas han surgido diversos compuestos con resultados anti-VIH prometedores. En el presente trabajo se ha optado por realizar diversas modificaciones en estos cabezas de serie del grupo con el objetivo de ampliar el espacio químico a su alrededor y mejorar estos resultados. En primer lugar, se diseñan y se sintetizan dos nuevas familias que contienen dos espaciadores diferentes a los utilizados hasta el momento. Un primer espaciador fenílico, como el que contiene AMD3100 pero no simétrico, con una cadena de un solo eslabón y una de dos eslabones metilénicos entre el anillo fenílico y el primer nitrógeno a cada lado del anillo. El segundo es un espaciador simétrico con un anillo naftalénico central. En segundo lugar, se amplía la diversidad molecular en diseñar nuevas tetraaminas con el espaciador p-fenilbismetilénico que implementan nuevos sustituyentes en busca de una mejora en la capacidad de inhibición de CXCR4. En esta búsqueda, se diseña una nueva ruta sintética que permita incluir la funcionalidad éster en el anillo heterocíclico de los extremos de la tetraamina, no accesible con las metodologías usadas hasta el momento. Finalmente, se realizan ensayos de inhibición del receptor CXCR4, proliferación celular y anti VIH a la vez que estudios computacionales para estudiar el modo de acción de alguno de estos compuestos frente el receptor CXCR4 y cómo actúan en diversas enfermedades con las que está relacionado, tales como la infección del VIH, el cáncer de páncreas o la leucemia., The chemokine receptor CXCR4, has proved to be a therapeutic target of major importance since the discovery of its role in the binding and fusion steps of the HIV virus with the host cell during the replication cycle and its role in a variety of tumor processes in its expansion in the organism through the metastasis. The Grup d'Enginyeria Molecular (GEM) of IQS has previously worked with AMD3100 analogues designing and synthesizing various libraries of tetramines as potential CXCR4 inhibitors. Among them various compounds with promising anti-HIV results have emerged. In the present work, we carried out various modifications of these structures with the objective of expanding the chemical space around them and improve these results. First, we designed and synthetized two new families of compounds containing different cores to those previously. The first core presents a central phenyl ring, the same included in AMD3100 but not symmetrical, with a methylene and two methylene carbons between the phenyl ring and the first nitrogen of the substituents on each side. The second is a symmetrical core with a central naphthalene ring. Secondly, the molecular diversity was increased by designing new tetramines with a p-xylene core implementing new substituents to improve the inhibition CXCR4. In this search, a new synthetic route was designed, which allowed to introduce the ester in the heterocyclic ring present in the ends of the tetramine, functionality not accessible with the methodologies used so far. Finally, inhibition assays of the CXCR4 receptor, cell proliferation assays and anti HIV tests were carried out together with computational studies to cast light on the mode of action of these compounds against the CXCR4 receptor and how they act in various related diseases such as HIV infection, pancreatic cancer or leukemia.

A suitable deterministic partitioning of large planar arrays in two kinds of properly shaped subarrays is shown to improve performances in terms of side-lobes peaks, directivity and aperture efficiency with respect to other existing solutions. The application of this simple idea to the case of large planar arrays with a reduced number of control points, useful in satellite applications, is demonstrated.
