Abstract

We report on a search for millicharged particles (mCPs) produced in cosmic ray proton atmospheric interactions using data collected during the first science run of the LUX-ZEPLIN experiment. The mCPs produced by two processes -- meson decay and proton bremsstrahlung -- are considered in this study. This search utilized a novel signature unique to liquid xenon (LXe) time projection chambers (TPCs), allowing sensitivity to mCPs with masses ranging from 10 to 1000 MeV/c$^2$ and fractional charges between 0.001 and 0.02 of the electron charge e. With an exposure of 60 live days and a 5.5 tonne fiducial mass, we observed no significant excess over background. This represents the first experimental search for atmospheric mCPs and the first search for mCPs using an underground LXe experiment.

Abstract

The XLZD collaboration is developing a two-phase xenon time projection chamber with an active mass of 60 to 80 t capable of probing the remaining WIMP-nucleon interaction parameter space down to the so-called neutrino fog. In this work we show that, based on the performance of currently operating detectors using the same technology and a realistic reduction of radioactivity in detector materials, such an experiment will also be able to competitively search for neutrinoless double beta decay in $^{136}$Xe using a natural-abundance xenon target. XLZD can reach a 3$\sigma$ discovery potential half-life of 5.7$\times$10$^{27}$ yr (and a 90% CL exclusion of 1.3$\times$10$^{28}$ yr) with 10 years of data taking, corresponding to a Majorana mass range of 7.3-31.3 meV (4.8-20.5 meV). XLZD will thus exclude the inverted neutrino mass ordering parameter space and will start to probe the normal ordering region for most of the nuclear matrix elements commonly considered by the community., Comment: 29 pages, 7 figures

Abstract

This report describes the experimental strategy and technologies for a next-generation xenon observatory sensitive to dark matter and neutrino physics. The detector will have an active liquid xenon target mass of 60-80 tonnes and is proposed by the XENON-LUX-ZEPLIN-DARWIN (XLZD) collaboration. The design is based on the mature liquid xenon time projection chamber technology of the current-generation experiments, LZ and XENONnT. A baseline design and opportunities for further optimization of the individual detector components are discussed. The experiment envisaged here has the capability to explore parameter space for Weakly Interacting Massive Particle (WIMP) dark matter down to the neutrino fog, with a 3$\sigma$ evidence potential for the spin-independent WIMP-nucleon cross sections as low as $3\times10^{-49}\rm cm^2$ (at 40 GeV/c$^2$ WIMP mass). The observatory is also projected to have a 3$\sigma$ observation potential of neutrinoless double-beta decay of $^{136}$Xe at a half-life of up to $5.7\times 10^{27}$ years. Abstract

We report results of a search for nuclear recoils induced by weakly interacting massive particle (WIMP) dark matter using the LUX-ZEPLIN (LZ) two-phase xenon time projection chamber. This analysis uses a total exposure of $4.2\pm0.1$ tonne-years from 280 live days of LZ operation, of which $3.3\pm0.1$ tonne-years and 220 live days are new. A technique to actively tag background electronic recoils from $^{214}$Pb $\beta$ decays is featured for the first time. Enhanced electron-ion recombination is observed in two-neutrino double electron capture decays of $^{124}$Xe, representing a noteworthy new background. After removal of artificial signal-like events injected into the data set to mitigate analyzer bias, we find no evidence for an excess over expected backgrounds. World-leading constraints are placed on spin-independent (SI) and spin-dependent WIMP-nucleon cross sections for masses $\geq$9 GeV/$c^2$. The strongest SI exclusion set is $2.1\times10^{-48}$ cm$^{2}$ at the 90% confidence level at a mass of 36 GeV/$c^2$, and the best SI median sensitivity achieved is $5.0\times10^{-48}$ cm$^{2}$ for a mass of 40 GeV/$c^2$., Comment: 9 pages, 7 figures. Abstract

The broad physics reach of the LUX-ZEPLIN (LZ) experiment covers rare phenomena beyond the direct detection of dark matter. We report precise measurements of the extremely rare decay of $^{124}$Xe through the process of two-neutrino double electron capture (2$\nu$2EC), utilizing a $1.39\,\mathrm{kg} \times \mathrm{yr}$ isotopic exposure from the first LZ science run. A half-life of $T_{1/2}^{2\nu2\mathrm{EC}} = (1.09 \pm 0.14_{\text{stat}} \pm 0.05_{\text{sys}}) \times 10^{22}\,\mathrm{yr}$ is observed with a statistical significance of $8.3\,\sigma$, in agreement with literature. First empirical measurements of the KK capture fraction relative to other K-shell modes were conducted, and demonstrate consistency with respect to recent signal models at the $1.4\,\sigma$ level., Comment: 15 pages, 3 figures

Abstract

Weakly interacting massive particles (WIMPs) may interact with a virtual pion that is exchanged between nucleons. This interaction channel is important to consider in models where the spin-independent isoscalar channel is suppressed. Using data from the first science run of the LUX-ZEPLIN dark matter experiment, containing 60 live days of data in a 5.5~tonne fiducial mass of liquid xenon, we report the results on a search for WIMP-pion interactions. We observe no significant excess and set an upper limit of $1.5\times10^{-46}$~cm$^2$ at a 90\% confidence level for a WIMP mass of 33~GeV/c$^2$ for this interaction.

Abstract

The Data Acquisition System (DAQ) for the LUX-ZEPLIN (LZ) dark matter detector is described. The signals from 745 PMTs, distributed across three subsystems, are sampled with 100-MHz 32-channel digitizers (DDC-32s). A basic waveform analysis is carried out on the on-board Field Programmable Gate Arrays (FPGAs) to extract information about the observed scintillation and electroluminescence signals. This information is used to determine if the digitized waveforms should be preserved for offline analysis. The system is designed around the Kintex-7 FPGA. In addition to digitizing the PMT signals and providing basic event selection in real time, the flexibility provided by the use of FPGAs allows us to monitor the performance of the detector and the DAQ in parallel to normal data acquisition. The hardware and software/firmware of this FPGA-based Architecture for Data acquisition and Realtime monitoring (FADR) are discussed and performance measurements are described., Comment: 18 pages, 24 figures

Abstract

LUX-ZEPLIN (LZ) is a tonne-scale experiment searching for direct dark matter interactions and other rare events. It is located at the Sanford Underground Research Facility (SURF) in Lead, South Dakota, USA. The core of the LZ detector is a dual-phase xenon time projection chamber (TPC), designed with the primary goal of detecting Weakly Interacting Massive Particles (WIMPs) via their induced low energy nuclear recoils. Surrounding the TPC, two veto detectors immersed in an ultra-pure water tank enable reducing background events to enhance the discovery potential. Intricate calibration systems are purposely designed to precisely understand the responses of these three detector volumes to various types of particle interactions and to demonstrate LZ's ability to discriminate between signals and backgrounds. In this paper, we present a comprehensive discussion of the key features, requirements, and performance of the LZ calibration systems, which play a crucial role in enabling LZ's WIMP-search and its broad science program. The thorough description of these calibration systems, with an emphasis on their novel aspects, is valuable for future calibration efforts in direct dark matter and other rare-event search experiments.

Abstract

The first science run of the LUX-ZEPLIN (LZ) experiment, a dual-phase xenon time project chamber operating in the Sanford Underground Research Facility in South Dakota, USA, has reported leading limits on spin-independent WIMP-nucleon interactions and interactions described from a non-relativistic effective field theory (NREFT). Using the same 5.5~t fiducial mass and 60 live days of exposure we report on the results of a relativistic extension to the NREFT. We present constraints on couplings from covariant interactions arising from the coupling of vector, axial currents, and electric dipole moments of the nucleon to the magnetic and electric dipole moments of the WIMP which cannot be described by recasting previous results described by an NREFT. Using a profile-likelihood ratio analysis, in an energy region between 0~keV$_\text{nr}$ to 270~keV$_\text{nr}$, we report 90% confidence level exclusion limits on the coupling strength of five interactions in both the isoscalar and isovector bases., Comment: 7 pages, 4 figures

Abstract The most prevalent genetic form of inherited arrhythmogenic cardiomyopathy (ACM) is caused by mutations in desmosomal plakophilin-2 (PKP2). By studying pathogenic deletion mutations in the desmosomal protein PKP2, here we identify a general mechanism by which PKP2 delocalization restricts actomyosin network organization and cardiac sarcomeric contraction in this untreatable disease. Computational modeling of PKP2 variants reveals that the carboxy-terminal (CT) domain is required for N-terminal domain stabilization, which determines PKP2 cortical localization and function. In mutant PKP2 cells the expression of the interacting protein MYH10 rescues actomyosin disorganization. Conversely, dominant-negative MYH10 mutant expression mimics the pathogenic CT–deletion PKP2 mutant causing actin network abnormalities and right ventricle systolic dysfunction. A chemical activator of non-muscle myosins, 4-hydroxyacetophenone (4-HAP), also restores normal contractility. Our findings demonstrate that activation of MYH10 corrects the deleterious effect of PKP2 mutant over systolic cardiac contraction, with potential implications for ACM therapy.

Abstract

Lowland meadows represent aboveground and belowground biodiversity reservoirs in intensive agricultural areas, improving water retention and filtration, ensuring forage production, contrasting erosion and contributing to soil fertility and carbon sequestration. Besides such major ecosystem services, the presence of functionally different plant species improves forage quality, nutritional value and productivity, also limiting the establishment of weeds and alien species. Here, we tested the effectiveness of a commercial seed mixture in restoring a lowland mixed meadow in the presence or absence of inoculation with arbuscular mycorrhizal (AM) fungi and biostimulation of symbiosis development with the addition of short chain chito-oligosaccharides (CO). Plant community composition, phenology and productivity were regularly monitored alongside AM colonization in control, inoculated and CO-treated inoculated plots. Our analyses revealed that the CO treatment accelerated symbiosis development significantly increasing root colonization by AM fungi. Moreover, the combination of AM fungal inoculation and CO treatment improved plant species evenness and productivity with more balanced composition in forage species. Altogether, our study presented a successful and scalable strategy for the reintroduction of mixed meadows as valuable sources of forage biomass; demonstrated the positive impact of CO treatment on AM development in an agronomic context, extending previous observations developed under controlled laboratory conditions and leading the way to the application in sustainable agricultural practices.

Pollen grain classification has a remarkable role in many fields from medicine to biology and agronomy. Indeed, automatic pollen grain classification is an important task for all related applications and areas. This work presents the first large-scale pollen grain image dataset, including more than 13 thousands objects. After an introduction to the problem of pollen grain classification and its motivations, the paper focuses on the employed data acquisition steps, which include aerobiological sampling, microscope image acquisition, object detection, segmentation and labelling. Abstract

The word “secretome” was first used to describe the proteins that cells secrete under different circumstances; however, recent studies have proven the existence of other molecules such as RNA and chemical compounds in the secretome. The study of secretome has significance for the diagnosis and treatment of disease as it provides insight into cellular functions, including immune responses, development, and homeostasis. By halting cell division, cellular senescence plays a role in both cancer defense and aging by secreting substances known as senescence-associated secretory phenotypes (SASP). A variety of techniques could be used to analyze the secretome: protein-based approaches like mass spectrometry and protein microarrays, nucleic acid-based methods like RNA sequencing, microarrays, and in silico prediction. Each method offers unique advantages and limitations in characterizing secreted molecules. Top-down and bottom-up strategies for thorough secretome analysis are became possible by mass spectrometry. Understanding cellular function, disease causes, and proper treatment targets is aided by these methodologies. Their approaches, benefits, and drawbacks will all be discussed in this review.

Abstract

Microfluidic devices may provide significant technological advancement in the production of micro and nanoparticles of interest in the pharmaceutical sector, for instance, particles used as drug carriers (Yeh et al., 2013). Indeed, microfluidics enables their production with precise control of the dimensional distribution, not achievable with conventional beaker methods. The particle size is crucial for the optimal operation, thus representing a key target along with the generation frequency. The device geometry, properties, and flow rates of the fluids are the main parameters that affect the generation process. In this work, we investigate the production of alginate droplets in an X-shaped microdevice (Tomasi Masoni et al., 2022). More specifically, we feed an alginate solution in sunflower oil working in the dripping regime. Experimental visualizations are carried out for different flow rates and concentrations of the surfactant to comprehend their effect on droplet formation. Interestingly, we identify operating conditions that prevent the formation of satellite droplets to preserve a narrow dimensional distribution.

Abstract

Developmental signaling inputs are fundamental for shaping cell fates and behavior. However, traditional fluorescent-based signaling reporters have limitations in scalability and molecular resolution of cell types. We present SABER-seq, a CRISPR-Cas molecular recorder that stores transient developmental signaling cues as permanent mutations in cellular genomes for deconstruction at later stages via single-cell transcriptomics. We applied SABER-seq to record Notch signaling in developing zebrafish brains. SABER-seq has two components: a signaling sensor and a barcode recorder. The sensor activates Cas9 in a Notch-dependent manner with inducible control, while the recorder obtains mutations in ancestral cells where Notch is active. We combine SABER-seq with an expanded juvenile brain atlas to identify cell types derived from Notch-active founders. Our data reveal rare examples where differential Notch activities in ancestral progenitors are detected in terminally differentiated neuronal subtypes. Abstract

Neuropsychiatric manifestations of viral infections (both per se and secondary to the neuroinflammatory reaction of the host) are mainly attributed to immunological reactions, so many aspects of their pathogenesis are still nuclear. Some novel therapeutic strategies are progressively emerging in which a vaccination may be having a particular impact on recovery and reduction of death. In this context, it is accepted that the SARS-CoV-2 virus is profoundly neurotropic and neuroinvasive, with various effects on the nervous system, although there is no complete understanding of the mechanism of neuroinvasion, brain injury, or short- or long-term neuropsychiatric sequelae. Therefore, it is necessary to understand the post-infectious manifestations of COVID-19 to guide the management of neuropsychiatric diseases. Thus, based on different research groups focused on this field, in this manuscript we summarize papers on COVID-19 and the nervous system (NS) published in a series of articles by Cuban authors. This review focuses on cognitive and affective emotional states, pathogenesis, biomarkers, clinical manifestations, and intervention strategies.

In the context of the global COVID-19 pandemic, the world of cultural heritage has had to cope with the closures of museums and dedicated structures. In this dramatic context, the digitisation of assets represented a partial solution to guarantee the fruition of the world’s cultural heritage. Even in the context of teaching, the pandemic represented a challenging moment. This contribution presents three different university teaching experiences focused on lighting for cultural heritage. The workshops described were held before, on horseback, and amid the pandemic. The differences are contextualised in the Italian regulatory and methodological framework. Attention is also paid to the digitisation of assets regarding the lighting design verification of the students’ project proposals. The teaching approach, the procedures, the material presented by the students, and the revision methodology are described. The comparison between the three editions allows for evaluations of the main problems encountered in the different contexts and when the used procedures were instead strong points. The use of three-dimensional simulation, initially designed exclusively for lighting verification, has also proved crucial in the communication of students’ works, demonstrating once more that in the future, the information technologies will be able to help those who deal with cultural assets to expand the catchment area and to enhance the communication of sites and museum structures.

Abstract

Congenital myopathies (CMs) are a group of diseases that primarily affect the muscle fiber, especially the contractile apparatus and the different components that condition its normal functioning. They present as muscle weakness and hypotonia at birth or during the first year of life. Centronuclear CM is characterized by a high incidence of nuclei located centrally and internally in muscle fibers. Clinical case: a 22-year-old male patient with symptoms of muscle weakness since early childhood, with difficulty in performing physical activity according to his age, with the presence of a long face, a waddling gait, and a global decrease in muscle mass. Electromyography was performed, showing a neurogenic pattern and not the expected myopathic one, neuroconduction with reduced amplitude of the motor potential of the peroneal nerve and axonal and myelin damage of the posterior tibial nerves. The microscopic study of the studied striated muscle fragments stained with hematoxylin–eosin and Masson’s trichrome showed the presence of fibers with central nuclei, diagnosing CM. The patient meets most of the description for CM, with involvement of all striated muscles, although it is important to note the neurogenic pattern present in this case, due to the denervation of damaged muscle fibers, which contain terminal axonal segments. Neuroconduction shows the involvement of motor nerves, but with normal sensory studies, axonal polyneuropathy is unlikely, due to normal sensory potentials. Different pathological findings have been described depending on the mutated gene in this disease, but all coincide with the presence of fibers with central nuclei for diagnosis by this means, which is so important in institutions where it is not possible to carry out genetic studies, and allowing early specific treatment, according to the stage through which the patient passes.

44. Cumulative Spatial and Temporal Analysis of Anthropogenic Impacts in the Protected Area of the Gran Paradiso National Park in the NW Alps, Italy

Abstract

Anthropogenic threats are responsible for habitat degradation and biodiversity decline. The mapping of the distribution and intensity of threats to biodiversity can be useful for informing efficient planning in protected areas. In this study, we propose a cumulative spatial and temporal analysis of anthropogenic impacts insisting on an alpine protected area, the Gran Paradiso National Park. The applied methodology starts with the construction of a spatial and temporal dataset of anthropogenic impacts and normalization based on relative intensity. The impacts analyzed include overgrazing, helicopter flights, road networks, built-up areas, worksites, derivations and discharges, sports activities, and dams and hydroelectric power plants. Each impact was assigned a weight based on its temporal persistence. Threats maps obtained from the collected, normalized, and weighted geodata are thus obtained. Finally, the risk map is calculated by combining the impact map with the vulnerability map, estimated through the methodology outlined in the Green Guidelines of the Metropolitan City of Turin. The risk map obtained was cross-referenced with the Park’s cartography to highlight any critical issues to specific habitats. Results show that most of the territory falls in low-risk (63%) or no-risk (35%) areas. However, there are some habitats that are totally or nearly totally affected by some degree of risk, although different to zero, such as the “Lentic waters with aquatic vegetation [incl. cod. 3130]”, the “Lentic waters partially buried”, the “Mountain pine forests (Pinus uncinata) [cod. 9430]”, and the “Mixed hygrophilous woods of broad-leaved trees [incl. cod. 91E0]”. This study highlights both the potential of these analyses, which enable informed management and planning of the fruition of protected areas, and the limitations of such approaches, which require in-depth knowledge of the territory and ecosystems and how they respond to threats in order to refine the model and obtain realistic maps.

Abstract

Summary: The increasing prevalence of food allergy and related pathologies in recent years has underscored the need to understand the factors affecting adverse reactions to food. Food allergy is caused when food‐specific IgE triggers the release of histamine from mast cells. However, other food‐specific antibody isotypes exist as well, including IgG and IgA. IgA is the main antibody isotype in the gut and mediates noninflammatory reactions to toxins, commensal bacteria, and food antigens. It has also been thought to induce tolerance to food, thus antagonizing the role of food‐specific IgE. However, this has remained unclear as food‐specific IgA generation is poorly understood. Particularly, the location of IgA induction, the role of T cell help, and the fates of food‐specific B cells remain elusive. In this review, we outline what is known about food‐specific IgA induction and highlight areas requiring further study. We also explore how knowledge of food‐specific IgA induction can be informed by and subsequently contribute to our overall knowledge of gut immunity. [ABSTRACT FROM AUTHOR]

Abstract

The word "secretome" was first used to describe the proteins that cells secrete under different circumstances; however, recent studies have proven the existence of other molecules such as RNA and chemical compounds in the secretome. The study of secretome has significance for the diagnosis and treatment of disease as it provides insight into cellular functions, including immune responses, development, and homeostasis. By halting cell division, cellular senescence plays a role in both cancer defense and aging by secreting substances known as senescence-associated secretory phenotypes (SASP). A variety of techniques could be used to analyze the secretome: protein-based approaches like mass spectrometry and protein microarrays, nucleic acid-based methods like RNA sequencing, microarrays, and in silico prediction. Each method offers unique advantages and limitations in characterizing secreted molecules. Top-down and bottom-up strategies for thorough secretome analysis are became possible by mass spectrometry. Understanding cellular function, disease causes, and proper treatment targets is aided by these methodologies. Their approaches, benefits, and drawbacks will all be discussed in this review.
