In order to understand better the survival and mutual interaction of epiphytic bacteria and fungi on apple plants, bacteria collected from these plants were cultivated on intact adaxial, stoma free cuticle membranes originally obtained from apple. The bacteria were labelled with luciferase genes fromVibrio harveyiin order to follow up their development and activity on the isolated cuticles. Our finding was that the epiphytic bacteria can have access to nutrients below the cuticle without causing damage to these cuticular membranes. Bacterial proteins may enable this nutrient mobilization and we found, indeed, that more than 46 proteins that must have been delivered by the bacteria in response to interaction with the cuticles as they could be found below the cuticle membrane. Eight major representatives of this group of external proteins have been sequenced with electron spray quadrupole time of flight mass spectrometry and subsequently identified by data base homology search as a flagellin, a porin type protein and proteins that are involved in amino acid recruitment and metabolism. Mitochondria from Orobanche were analysed for the activities of aconitate hydratase, isocitrate dehydrogenase, succinate dehydrogenase, fumarate hydratase, malate dehydrogenase, NADH oxidase, substrate-cytochrome c oxidoreductases, glutamate dehydrogenase, aminotransferases, ATPase and "malic" enzyme. The specific activities of isocitrate dehydrogenase, NADH oxidase, substrate-cytochrome c oxidoreductases and glutamate dehydrogenase in the mitochondrial fraction from parasite tissue compared favourably with those reported for most of the mitochondria from growing and storage tissues. Succinate dehydrogenase, fumarate hydratase and aspartate aminotransferase were of intermediate activity, while aconitate hydratase and malate dehydrogenase had rather low activity, and "malic" enzyme had very low activity in comparison with other preparations. The relevance of these findings in relation to mitochondrial metabolism in the parasite is discussed. No evidence was obtained to suggest any basic abnormality in the biochemical properties of the mitochondria from Orobanche cernua which may be correlated with its obligatorily parasitic existence. Mitochondria were isolated from the scapes of Orobanche cernua. On the basis of comparative studies with four media, a mannitol grinding medium was selected at a slightly alkaline pH and supplemented with EDTA, metabisulphite, BSA and soluble PVP. Various intermediates of the Krebs cycle, pyruvate, glutamate and NADH were oxidized by the isolated particles, though with different efficiencies. The rate of oxidation was steady for every intermediate tested over a 60 min period, except for NADH. In the majority of cases, the oxidation was accompanied by efficient coupling with phosphorylation. Antimycin A led to significant reduction in the oxygen uptake in the presence of succinate and complete suppression of phosphate‐esterification. The amount of protein recovered in the mitochondrial fraction was comparable with the reported recoveries from a number of plant tissues. The biochemical integrity of mitochondria in the particulate fraction appeared possible from: (a) the absence of respiratory response to added cytochrome c during the oxidation of succinate, (b) insignificant oxidation of exogenously added pyruvate in the absence of sparker and (c) the ability to satisfactorily couple phosphorylation to the oxidation of a number of substrates. The phothrespiratory nitrogen cycle proposed by Keys et al. (Nature 275: 741-743, 1978) involved formation of glycine by transamination of glyoxylate in the per-oxisomes utilizing glutamate. Subsequently, glycine is oxidized to ammonia, serine and CO2 in the mitochondria, The ammonia is reassimilated via the GS/GOGAT pathway generating glutamate. In this article, experimental evidence which suggests the occurrence of alternative mechanisms of glycolate and serine synthesis as well as of CO2 and ammonia evolution is. discussed. The problem of utilization of NADH coupled to ATP synthesis during photosynthesis is still unresolved, which complicates the glycine oxidation reaction in light. Further, factors are presented that determine the availability of amino donors in the peroxisomes and of amino acids viz., glycine, set-me and glutamate for the operation of the photorespiratory N cycle. Recent evidence regarding the role of formate arising, out of the reaction of glyoxylate with H2O2 in the regulation of photosynthetic electron flow in the Hill reaction, as well as of photorespiratory substrates functioning as carbon sources for the citric acid cycle in the light or for export to the growing, tissues, suggests that the role of photorespiration in plant metabolism needs to be reexamined.
