Summary: Macrophages are critical for maintenance and repair of mucosal tissues. While functionally distinct subtypes of macrophage are known to have important roles in injury response and repair in the lungs, little is known about macrophages in the proximal conducting airways. Single-cell RNA sequencing and flow cytometry demonstrated murine tracheal macrophages are largely monocyte-derived and are phenotypically distinct from lung macrophages at homeostasis. Following sterile airway injury, monocyte-derived macrophages are recruited to the trachea and activate a pro-regenerative phenotype associated with wound healing. Animals lacking the chemokine receptor CCR2 have reduced numbers of circulating monocytes and tracheal macrophages, deficient pro-regenerative macrophage activation and defective epithelial repair. Together, these studies indicate that recruitment and activation of monocyte-derived tracheal macrophages is CCR2-dependent and is required for normal airway epithelial regeneration.

Higher education is considered an important tool for the overall development of any country, and it holds true in the context of Afghanistan as well. At the same time, a good eco-environment in terms of political will, leaders' farsighted vision, a fair budget, good infrastructure, and a good teaching community are some of the basic requirements for higher education to move in the direction of new and higher horizons. However, due to the ongoing war during the last couple of decades, the country's education system has become out of reach for a substantial part of the population due to poverty, lack of infrastructure, refugees and internally displaced, digital division, etc., critically affecting the education equity. This systematic review examines India's education diplomacy in addressing the inequities in Afghanistan's education system and making them more equitable. Education was further dilapidated with the outbreak of the COVID-19 pandemic. Afghanistan is caught between a war and a pandemic and suffers from a double whammy in losses. Subsequently, given their chilling effects, higher education becomes devoid of multiples equities, including education. However, because of their historical and geo-civilizational ties, India has focused on development diplomacy in general and education diplomacy (E.D) in particular to improve educational infrastructures.

Background: Bicortical engagement is the engagement of nasal cortical plate and basal cortical plate in maxillary and mandibular esthetic zone, respectively. These are longer implants which offer osseointegration along with high osseofixation. Increased length of implant provides increased bone–implant contact offering primary stability which permits immediate loading. Aim: The aim of the study was to clinicoradiographically evaluate immediate loading in bicortically engaged long implants in maxillary and mandibular esthetic region. Materials and Methods: Thirty patients with missing teeth in esthetic region were selected for the study. Implants were placed in both arches engaging nasal cortex and basal bone wherever required. One-time definitive abutment was placed and immediate loading was done. Results: No failure was observed in the study period of 9 months. The mean insertion and reverse torque was 66.67 ± 6.794 and 41.67 ± 3.101, respectively, indicating high primary stability. No pain, discomfort, infection, or implant mobility was observed. The mean crestal bone loss changes were statistically insignificant. Conclusion: Hundred percent implant survival was observed.

The ongoing debate on the conceptual underpinnings of constructivism and global health partnerships (GHPs) in global health studies has a dimension that deserves closer attention. This paper attempts to draw attention to a few aspects of the debate using Finnemore's constructivist analysis. According to this study, global actors need to rethink their paradoxical notions of pandemic crisis survival in light of the growing demand for mobilizing diverse global health agents and the necessity of constructing complex GHPs to address challenges of international significance. A global response based on solidarity and multilateralism is the only way to effectively combat this pandemic. Against this backdrop, the article analyses this development through an ideational ontological case study of the GAVI, the Vaccine Alliance. This article contributes to the debate by explaining how the GAVI Alliance fostered global collaboration and can serve as a template for future GHPs. [ABSTRACT FROM AUTHOR]

Background and Aim: The present study aimed to evaluate the knowledge and attitude of antimicrobial drugs prescribing by oral private health-care practitioners in the management of periodontal diseases across Mandi, Himachal Pradesh, India. Design: This was a descriptive cross-sectional percentage-based survey. Materials and Methods: A descriptive observational cross-sectional Study was conducted with the pretested and structured questionnaires which were sent physically to private oral health-care practitioners of Mandi district of Himachal Pradesh, India. The study was carried out for 2 months. A self-orchestrated questionnaire comprising 10 questions was sent by dental students to private practitioners of Mandi to assess their awareness along with a point of view concerning antimicrobial drugs prescribing for periodontal diseases. Facts entered in an Excel file were examined using SPSS version 22. Illustrative stats were carried out. Results: The survey was responded by 100 private practitioners providing oral health care who accomplish the questionnaire. Most of the private practitioners were in the age group of 20 and 60 years, and 63% were male and 37% were female. There was a statistically significant difference between knowledge on prescribing antimicrobial drugs by private oral health-care providers in Mandi, Himachal Pradesh, India. Amoxicillin Alone, amoxicillin with clavulanic acid and metronidazole was the most prescribing drug followed by clindamycin by private oral healthcare Practitioners in the management of periodontal diseases. Conclusion: Most of the private oral health-care providers practicing in Mandi, Himachal Pradesh, routinely prescribed prophylactic antimicrobial drugs, and the observational survey outcome shows that although the private practitioners do know about the antimicrobial drugs, they should be very careful while prescribing antimicrobial drugs in the management of periodontal diseases. Because of increasing microbial resistance, an immediate remedy is the need of the hour to tackle the devastating consequences of antimicrobial drugs resistance.

Aim and Objective: The objective of this study was to evaluate the knowledge of dental implants and peri-implantitis among various medical professionals across Himachal Pradesh. Materials and Methods: A cross-sectional survey was conducted among 100 medical professions residing in Himachal Pradesh using a self-explanatory questionnaire. The survey consisted of 11 self-explanatory questionnaires. The nature and purpose of the survey was explained to the professionals. Results: The present study was conducted to evaluate the knowledge of dental implants and peri-implantitis among 100 medical professionals. The results shows that although the medical practitioner do have some knowledge about dental implants, but still there is a scope of improvement about the knowledge of dental implants and peri-implant diseases. Conclusions: The results of this survey showed that majority of the medical professionals working in the state of Himachal Pradesh, were aware about dental implants as a treatment modality for the replacement of missing teeth. However, there is a deficit in the level of information regarding peri-implant diseases and the effect of systemic diseases and condition on dental implants, so further education is required.

Head and neck squamous cell carcinomas (HNSCC) include heterogeneous group of tumors, classified according to their anatomical site. It is the sixth most prevalent cancer globally. Among South Asian countries, India accounts for 40% of HNC malignancies with significant morbidity and mortality. In the present study, we have performed exome sequencing and analysis of 51 Head and Neck squamous cell carcinoma samples. Besides known mutations in the oncogenes and tumour suppressors, we have identified novel gene signatures differentiating buccal, alveolar, and tongue cancers. Around 50% of the patients showed mutation in tumour suppressor genes TP53 and TP63. Apart from the known mutations, we report novel mutations in the genes AKT1, SPECC1, and LRP1B, which are linked with tumour progression and patient survival. A highly curated process was developed to identify survival signatures. 36 survival-related genes were identified based on the correlation of functional impact of variants identified using exome-seq with gene expression from transcriptome data (GEPIA database) and survival. An independent LASSO regression analysis was also performed. Survival signatures common to both the methods led to identification of 4 dead and 3 alive gene signatures, the accuracy of which was confirmed by performing a ROC analysis (AUC=0.79 and 0.91, respectively). Also, machine learning-based driver gene prediction tool resulted in the identification of IRAK1 as the driver (p-value = 9.7 e-08) and also as an actionable mutation. Modelling of the IRAK1 mutation showed a decrease in its binding to known IRAK1 inhibitors.

Abstract Background The most widely used human genome reference assembly hg19 harbors minor alleles at 2.18 million positions as revealed by 1000 Genome Phase 3 dataset. Although this is less than 2% of the 89 million variants reported, it has been shown that the minor alleles can result in 30% false positives in individual genomes, thus misleading and burdening downstream interpretation. More alarming is the fact that, significant percentage of variants that are homozygous recessive for these minor alleles, with potential disease implications, are masked from reporting. Results We have demonstrated that the false positives (FP) and false negatives (FN) can be corrected for by simply replacing nucleotides at the minor allele positions in hg19 with corresponding major allele. Here, we have effectively replaced 2.18 million minor alleles Single Nucleotide Polymorphism (SNPs), Insertion and Deletions (INDELs), Multiple Nucleotide Polymorphism (MNPs) in hg19 with the corresponding major alleles to create an ethnically normalized reference genome called hg19KIndel. In doing so, hg19KIndel has both corrected for sequencing errors acknowledged to be present in hg19 and has improved read alignment near the minor alleles in hg19. Conclusion We have created and made available a new version human reference genome called hg19KIndel. It has been shown that variant calling using hg19KIndel, significantly reduces false positives calls, which in-turn reduces the burden from downstream analysis and validation. It also improved false negative variants call, which means that the variants which were getting missed due to the presence of minor alleles in hg19, will now be called using hg19KIndel. Using hg19KIndel, one even gets a better mapping percentage when compared to currently available human reference genome. hg19KIndel reference genome and its auxiliary datasets are available at https://doi.org/10.5281/zenodo.2638113

The tumor microenvironment of ovarian cancer is the peritoneal cavity wherein adipose tissue is a major component. The role of the adipose tissue in support of ovarian cancer progression has been elucidated in several studies from the past decades. The adipocytes, in particular, are a major source of factors, which regulate all facets of ovarian cancer progression such as acquisition of chemoresistance, enhanced metastatic potential, and metabolic reprogramming. In this review, we summarize the relevant studies, which highlight the role of adipocytes in ovarian cancer progression and offer insights into unanswered questions and possible future directions of research.
