The delivery of pathogens to lysosomes for degradation provides an important defense against infection. Degradation is enhanced when LC3 is conjugated to endosomes and phagosomes containing pathogens to facilitate fusion with lysosomes. In phagocytic cells, TLR signaling and Rubicon activate LC3-associated phagocytosis (LAP) where stabilization of the NADPH oxidase leads to sustained ROS production and raised vacuolar pH. Raised pH triggers the assembly of the vacuolar ATPase on the vacuole membrane where it binds ATG16L1 to recruit the core LC3 conjugation complex (ATG16L1:ATG5-12). This V-ATPase-ATG16L1 axis is also activated in nonphagocytic cells to conjugate LC3 to endosomes containing extracellular microbes. Pathogens provide additional signals for recruitment of LC3 when they raise vacuolar pH with pore-forming toxins and proteins, phospholipases, or specialized secretion systems. Many microbes secrete virulence factors to inhibit ROS production and/or the V-ATPase-ATG16L1 axis to slow LC3 recruitment and avoid degradation in lysosomes.

The cynomolgus macaque, Macaca fascicularis, is a non-human primate (NHP) widely used in biomedical research as it shares behavioural, genetic, immunological and physiological similarities with humans. These similarities may extend to the enteric microbiome, with some microbial taxa common to both humans and NHPs. However, to date, the majority of these microbial surveys have focused on the prokaryome, and have largely ignored or overlooked the NHP gut mycobiome. To address this shortfall, we have undertaken a region-by-region taxonomic survey of the cynomolgus intestinal mycobiota, from duodenum to distal colon, of ten captive animals of differing age. Using a high-throughput ITS1 amplicon sequencing-based approach, we found that fungi from the Ascomycota phylum dominate the cynomolgus enteric mycobiota. The budding yeast genus Kazachstania was most abundant, with K. pintolopesii and K. telluris highly prevalent, and the predominant species in many of the intestinal samples. However, while K. pintolopesii was present throughout the primate GI tract, K. telluris was found mainly in the small intestine. In this study, K. pintolopesii was identified as the dominant enteric fungus in captive cynomolgus macaques. This contrasts with humans, where Candida albicans is a common member of the intestinal microbiota. To our knowledge, this is the first time K. pintolopesii has been identified as a primate gut commensal.

To determine whether signaling via CD122 (interleukin-2 [IL-2]/IL-15 receptor beta-chain) plays a role in regulating the expansion and differentiation of lymphocyte precursors, we have characterized its expression and evaluated its ability to influence the activity of developing lymphoid cells. A significant fraction of Sca1+Lin- hematopoietic stem cells in day 12 fetal liver were found to be CD122+. CD122-mRNA+ and IL-2-mRNA+ cells were also localized in embryo sections within pharyngeal blood vessels adjacent to and surrounding the thymic analgen. This distribution is consistent with the migration of CD122+ progenitor cells from the liver to the developing thymus where a majority of Sca1+ intrathymic T-cell progenitors were CD122+. Analysis of CD122 expression in the day 12 fetal liver revealed that the majority of B220+ cells were CD122+. Furthermore, CD122 expression was restricted to the earliest B220+ cells (CD43+CD24-; prepro B cells; fraction A) that proliferate vigorously to IL-2 in the absence of any stromal cells, but not to IL-15. Consistent with a role for the IL-2/IL- 2R pathway in lymphocyte development is the progressive loss of B cells seen in IL-2-deficient mice. Together, these observations suggest that CD122 plays a role in regulating normal lymphocyte development in vivo.

gamma delta T-cell hybrid clones were obtained from intestinal intraepithelial lymphocytes (i-IEL) by fusion with the BW5147 thymoma line. Four clones which expressed different V gamma/V delta genes were selected for further study. All of the gamma delta clones secreted interleukin-2 (IL-2) in the presence of the BALB/c-derived B-lymphoma line, A20. No alpha beta T-cell hybrid clones derived from spleen or i-IEL responded to A20. We obtained several pieces of evidence which strongly suggest that these responses are mediated by the gamma delta T-cell receptor (TcR). Class II major histocompatibility complex (MHC), FcR and surface Ig expressed on A20 are not involved in the response. Native i-IEL derived from BALB/c selectively survive in culture in the presence of A20 cells. The ligand may be a superantigen-like molecule because all our gamma delta T-cell clones responded to A20 in spite of their different combinations of V gamma/V delta gene segments.

A specific subset of gamma delta T lymphocytes bearing a V gamma 6V delta 1-encoded TCR is known to populate the normal nonpregnant murine uterine and vaginal epithelium. However, gamma delta T lymphocytes residing at the maternal-fetal interface during pregnancy have not yet been investigated. Using mAb and cytofluorographic analysis, we analyzed gamma delta TCR-bearing lymphocytes obtained from placental/decidual tissues of allogeneic (C57B1/10 X BALB/c and BALB/c X C57B1/10) pregnancies. Gestations were analyzed at several time points during the second half of pregnancy, with lymphocytes from both maternal spleen and nonpregnant C57B1/10 uteri analyzed as controls. We found that all gamma delta T lymphocytes at the maternal-fetal interface are maternally derived. Relative to the total T lymphocyte population, the percentage of gamma delta TCR-bearing T lymphocytes at the maternal-fetal interface is enriched three- to four-fold compared with maternal spleen, and twofold compared with nonpregnant uteri. Although one-third of gamma delta T lymphocytes from pregnant animals express a cell-surface marker associated with activation (IL-2R), gamma delta cells from uteri of nonpregnant mice fail to express IL-2R. In terms of absolute numbers, we estimate that reproductive-tract gamma delta T cells are increased nearly 100-fold in pregnant animals compared with nonpregnant animals. To characterize the TCR-gamma delta repertoire in the placenta/decidua, we generated 21 TCR-gamma delta-bearing hybridomas from lymphocytes in this tissue. Analysis of these hybridomas revealed at least six distinct gamma delta receptor types expressed at the maternal-fetal interface, with V gamma 6V delta 1 encoded TCR representing the predominant population. As specific resident constituents of the reproductive tract, gamma delta T lymphocytes may be involved in regulating a variety of physiologic and pathophysiologic events in reproductive biology.

The relationship between c-myc expression and thymocyte activation was studied in freshly isolated human thymocytes and in thymocytes activated with various inducing agents. In freshly isolated thymocytes c-myc mRNA is expressed at low levels, while thymocytes activated with Concanavalin A (Con A), the tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA), Con A in combination with TPA or interleukin 2 (IL-2) are induced to express higher levels of c-myc mRNA. The expression of c-myc is increased within 3 h of stimulation with these inducing agents; the amount of c-myc mRNA which is accumulated is not correlated with the rate of thymocyte proliferation. Dexamethasone and Cyclosporin A (CsA) which inhibit early events of T cell activation and the expression of the interleukin-2 (IL-2) and gamma-interferon (gamma-IFN) genes also markedly suppress the expression of c-myc mRNA in Con A, and Con A + TPA-activated thymocytes. We conclude that activation of c-myc gene expression is an early event observed in activated human thymocytes. The level of c-myc expression is dependent on the mode of thymocyte activation rather than on the rate of thymocyte proliferation. Since freshly isolated thymocytes express low levels of c-myc mRNA it is possible that IL-2 induces c-myc expression at least in a responsive subpopulation of thymocytes during ontogeny.

BACKGROUND: Although gamma delta T cells are a major component of the human intestinal mucosa, it is not clear what role they play in mucosal immunity or if they are involved in the disease process of inflammatory bowel disease (IBD). MATERIALS AND METHODS: Flow cytometry and reverse transcriptase-polymerase chain reaction (RT-PCR) assays were used to identify quantitative and qualitative changes in the repertoire of gamma delta T cells present in surgical and/or biopsy samples or normal and inflamed colon from individual patients with ulcerative colitis (UC) or Crohn's disease (CD). Cytokine production and the ability to adhere to and interact with colonic fibroblasts were used to compare the functional properties of gamma delta T cells isolated from the normal and diseased colonic mucosa. RESULTS: Increased numbers of gamma delta T cells localized in areas of inflammation and tissue injury were found in the majority of patients, irrespective of the type of IBD present. This expansion was attributable to an increase in V delta 1+ cells expressing a V delta 1-(D delta 3)-J delta 1-encoded T cell receptor and was seen in patients with severe disease as well as those with newly diagnosed or less severe forms of IBD. Among T cells present in the inflamed mucosa of patients with CD, gamma delta T cells, particularly V delta 1+ cells, were a major source of the proinflammatory cytokine interferon-gamma and could interact with colonic fibroblasts. CONCLUSIONS: Our results demonstrate that the chronic inflammatory immune response characteristic of IBD is associated with distinct changes in the number, distribution, composition, and function of mucosal gamma delta T cells. Through the production of cytokines and physical interaction with other cells, gamma delta T cells can perform an immunoregulatory function and contribute to the pathophysiology of IBDs.

The liver of mice contains elevated percentages of gamma delta T cells when compared with peripheral lymphoid organs. We have now analyzed these cells clonally, by generating a random collection of liver gamma delta T cell hybridomas and sequencing the productively rearranged TCR-gamma and -delta genes in each hybridoma clone. Examining C57BL/10 mice of various ages, we have found that over half of their normal gamma delta T cells are one of two types, V delta 4+ or V delta 6.3+. gamma delta T cell hybridomas generated from mouse liver contain clones that are "spontaneously" reactive, and respond to purified protein derivative from mycobacteria and to a 17-amino acid peptide from mycobacterial heat shock protein-60 (HSP-60). Like similar cells found in newborn thymus or adult spleen, all of the cells showing this HSP-60 reactivity pattern were found to express V gamma 1-J gamma 4-C gamma 4, most in conjunction with V delta 6-J delta 1-C delta, particularly with V delta 6.3. However, the gamma and delta junctional sequences of the V gamma 1/V delta 6+ cells isolated from adult liver differed from those found in adult spleen; being less diverse, their receptors instead resemble those of similar cells from newborn thymus. These data suggest that HSP-60-reactive gamma delta cells in adult murine liver and spleen are independent of each other and may be resident in their respective sites.

Mice lacking interleukin-2 (IL-2) developed a severe hematopoietic disorder characterized by the abnormal development of myeloid cells and neutropenia. Analysis of the bone marrow of IL-2-deficient (IL-2(-/-)) mice showed that the number of mature polymorphonuclear cells was decreased by 65% to 75%, and granulocyte/macrophage precursor cells were reduced by 50%. Bone marrow cells from IL-2(-/-) mice were unable to sustain myelopoiesis in lethally irradiated mice and in long-term bone marrow cultures (LTBMC). The addition of exogenous IL-2 to LTBMC of IL-2(-/-) cells partially restored hematopoietic progenitor activity. In the bone marrow of wild-type mice, immature (Mac-1(lo)) myeloid cells, including myeloblasts and promyelocytes, constitutively expressed the beta-chain of the IL-2R, and the number of Mac-1(lo)IL-2Rbeta+ cells was increased by twofold to threefold in IL-2(-/-) mice. During culture in the presence of IL-2 and the absence of stromal cells, Mac-1(lo)IL-2Rbeta+ immature myeloid cells proliferated and gave rise to mature granulocytes and macrophages. Collectively, these observations indicate that defective myelopoiesis in IL-2(-/-) mice is at least in part a consequence of their direct dependency on IL-2, and by regulating the growth of immature myeloid cells, IL-2 plays an important role in the homeostatic regulation of myelocytic cell generation.

Although gammadelta T cells are known to contain the highest frequency of mycobacteria-reactive cells in humans and numerous studies have suggested that they play an important role in the initial immune response to Mycobacterium tuberculosis (Mtb), very few studies have attempted to analyze these cells in patients with active pulmonary tuberculosis. The aim of the present study was therefore to evaluate the consequences of infection on the number and activity of mycobacteria-reactive gammadelta T cells. Three-color flow cytometric analysis of blood and bronchoalveolar lavage gammadelta T cells of patients diagnosed with active pulmonary tuberculosis showed that compared with normal healthy subjects and patients with the unrelated pulmonary granulomatous diseases sarcoidosis and berylliosis the size of the mycobacteria-reactive Vgamma9+/Vdelta2+ gammadelta T cell subset in both the blood and lung was dramatically reduced. In addition, the Vgamma9+/Vdelta2+ cells left intact in patients with tuberculosis were refractory to in vitro stimulation by Mtb Ags, which are potent stimuli for these cells in normal subjects. Our results demonstrate for the first time a strong correlation between the absence or loss of the major Vgamma9+/Vdelta2+ Mtb-reactive subset of gammadelta T cells and manifestations of disease, consistent with the hypothesis that these gammadelta T cells play a role in the protective immune response to Mtb infection.

The nature of how human gammadelta T cells are normally generated is not clear. We have used an RT-PCR assay and DNA sequencing to identify and compare delta-encoded TCRs (TCRDs) that are generated de novo in the fetal gut, liver, and thymus and to determine when, where, and how the TCRD repertoire is established during normal embryonic development. Rearranged TCRDV genes are first expressed outside of the thymus in the liver and primitive gut between 6 and 9 wk gestation. Although DV1Rs and/or DV2Rs predominated, differences in the pattern of TCRDV gene rearrangement and transcription in each tissue during ontogeny were identified. Specific, DV2-encoded TCRs are highly conserved throughout ontogeny in the tissues from the same and between genetically distinct donors. Although the thymic and intestinal gammadelta T cell repertoires partially overlap early in development, they diverge and become nonoverlapping during the second trimester, and the generation of the intestinal gammadelta T cell repertoire is characterized by differences in the processing of DV1Rs and DV2Rs. Whereas the structural diversity of DV1Rs progressively increases during gut development up to birth, DV2Rs have limited structural diversity throughout ontogeny. Together, our findings provide evidence for the ability of different fetal tissues to support the development of gammadelta T cells.

This study shows that unfractionated thymocytes can be activated to proliferate in response to activation by the CD 2 pathway, to express interleukin 2 receptors, and to synthesize interleukin 2 and interferon-gamma. Less mature, T3- thymocytes, isolated by negative selection are activated to a lesser extent than are unfractionated thymocytes; activation by the CD 2 pathway, induces proliferation, the expression of the interleukin 2 gene and interferon-gamma synthesis. 12-O-Tetradecanoyl phorbol 13-acetate in combination with the anti-CD 2 antibodies T11(2) + T11(3) increases the response of both unfractionated and T3- thymocytes. In addition we demonstrate that tetradecanoyl phorbol acetate in combination with T11(3) can replace the requirement for T11(2) for thymocyte activation and induce the expression of T11(3).

The role that gamma delta-T lymphocytes play in virus infections is yet to be defined. The TCR-gamma delta + cell population found late in the course of influenza pneumonia has been analyzed for ligand-dependent lytic function. These gamma delta-T cells are not constitutively cytotoxic when recovered directly from the site of virus-induced damage in the respiratory tract, although the TCR-alpha beta + population that is present concurrently contains such lytic effectors. Both sets of lymphocytes mediate cytotoxic activity after further in vitro stimulation in the presence of mAb to CD3 and low concentrations of rIL-2. Secondary stimulation in vivo with a cross-reactive influenza A virus does not lead to the emergence of a cytotoxic gamma delta-T cell population, although substantial numbers of these gamma delta-T cells express mRNA for a variety of lymphokines and cytokines. Analysis of DNA content indicates that many of the gamma delta-T cells isolated directly from the pneumonic lung are cycling. This could reflect continuing stimulation by a specific ligand, perhaps a self-component expressed at abnormally high levels in the site of virus-induced pathology. However, we could find no evidence to indicate that the gamma delta-T cells are acting to eliminate redundant components of the host response. The percentage of inflammatory macrophages and nonphagocytic cells expressing mRNA for a 65-kDa heat-shock protein (the proposed target for at least a subset of these gamma delta-T cells) is not reduced during the time that lymphocytes with mRNA for the TCR-gamma delta are present in greatest numbers. Possible alternative functions for the gamma delta-T cells are discussed.

B7-CD28 costimulation is essential for the activation of CD4+ T helper cells, mainly by regulating IL-2 and other cytokine production. The requirement for this costimulatory pathway in the activation of CD8+ T cells, however, is still poorly understood. Here we analyzed the role of B7-CD28 costimulation in the differentiation of Ag-specific CTL precursor. We found that the activation of not only IL-2 production but also cytotoxic function in CD8 T cells requires B7 costimulation. The costimulatory signal, which cannot be replaced by exogenous IL-2, is directly implicated in the activation of the lytic machinery in CD8 T cells. Moreover, B7-CD28 costimulation appears to play a critical role in the accumulation of mRNA encoding at least one of the granzymes required for cytolytic function, granzyme B or CTLA-1. The production of IFN-gamma by CD8 T cells, however, does not appear to require costimulation.

We have recently demonstrated that human thymocytes can be induced to express interleukin 2 (IL-2) receptors and to synthesize IL-2. The present study shows that relatively immature T6+ human thymocytes as well as the more mature T3+ thymocytes could be induced to express functional IL-2 receptors when activated with either Concanavalin A (Con A), Con A and 12-O-tetradecanoylphorbol 13-acetate (TPA) or IL-2 in combination with Con A or TPA. The phenotype of the common, immature thymocyte was identified by the binding of either fluorescein isothiocyanate (FITC)- or phycoerythrin (PE)-conjugated monoclonal anti-T6 (OKT 6) antibody and of mature thymocytes by the binding of monoclonal anti-T3 (OKT 3) antibody. We also observed that the expression of the T3 antigen on thymocytes, freshly isolated from thymic specimens obtained in the course of cardiac surgery of infants and children, was greater than 40% in 14 of 18 donors and that thymocytes co-expressed the T3 and the T6 antigen as determined by dual colour cytofluorometry. In thymocytes activated in vitro the expression of IL-2 receptors, determined by dual colour cytofluorometry with the PE-conjugated monoclonal anti-human IL-2 receptor antibody (PE anti-IL-2 R), was detected by the second day of induction in both immature T6+ and mature T3+ thymocytes. T6+ thymocytes proliferated in response to IL-2 and persisted in cultures for the duration of the study (18 days) and continued to express IL-2 receptors.

Cytokine production and mRNA profiles have been analyzed at the single cell level for bronchoalveolar lavage (BAL) populations from mice infected with an influenza A virus in the presence or absence of the CD4+ and CD8+ T cell subsets. Phagocytes were identified by their capacity to engulf latex particles, but the cellular elements of this inflammatory process were otherwise not characterized. BAL preparations from undepleted mice contained numerous IL-2, IL-4 and IFN-gamma-producing cells, with many fewer secreting TNF or IL-10. The frequency of mRNA+ cells detected by in situ hybridization was, in general, much higher than that for protein-secreting cells determined by ELISPOT analysis. In addition to IL-2, IL-4, and IFN-gamma, large numbers of cells were found to contain IL-10 and TNF-beta transcripts. Depletion of CD4+ and CD8+ cells caused significant reduction in the frequency of IL-2 and IL-4-producing cells, but even simultaneous elimination of both T cell subsets failed to totally remove all cells producing these cytokines. Similarly, a residual population of IFN-gamma-producing cells remained after depletion of the CD4+ and CD8+ subsets. Likely sources of these cytokines (apart from NK cells) are the CD4-8- alpha beta and gamma delta T cells found previously in BAL populations from doubly-depleted mice infected with this virus. Somewhat surprisingly, mRNA for IFN-gamma, IL-5, and TNF beta was prevalent in cells that had engulfed latex particles, though mRNA for IL-2 and IL-4 was never detected in macrophages.(ABSTRACT TRUNCATED AT 250 WORDS)

T cells expressing the cell surface differentiation antigen CD4 are involved in most immune responses. Our studies address two issues about CD4 T cell responses to antigen: first, how does the T cell receptor come together with its ligand to generate an immune response, and what is the role of the CD4 molecule in this response? Second, are all CD4 T cells identical in their functional activity, and how does the activating signal determine the functional outcome of a response? Our studies outlined below suggest that the T cell receptor and its peptide: class II major histocompatibility complex (MHC) molecule ligand come together in a defined orientation determined in part by the binding of CD4 to both the T cell receptor and its ligand. Our studies suggest that the V beta chain is involved directly in MHC antigen recognition, binding self MHC with low affinity and non-self MHC with high affinity. The selective effect of the Mls locus on V beta expression is believed to reflect the binding of the Mls protein directly to the V beta region. CD4 is described as a co-receptor, forming an inducible part of the T cell receptor and binding to the same class II MHC molecule as that receptor. Studies with both cloned lines and normal CD4 T cell populations suggest the existence of two separable subsets with definable function. One set appears to be specialized for the activation of the humoral immune response, while the other drives the cell-mediated immune responses, particularly those involving the activation of macrophages. These two subsets of CD4 T cells have differential activation requirements, seen particularly in the requirement for interleukin 1 (IL-1) in the activation and clonal expansion of CD4 T cells involved in humoral immunity. This requirement for IL-1 may also be observed in the priming of this subset of CD4 T cells. These studies demonstrate that the optimal activation of CD4+ T cells involves recognition of peptide fragments presented by class II MHC molecules and accessory signals derived from the antigen presenting cells.
