Background Technological communication methods such as telephone calls and video calls can help prevent social isolation and loneliness in frail older adults during confinement. Objective Our objectives were to determine which virtual communication method (ie, telephone call or video call) was preferred by confined older hospital patients and nursing home residents and the variables influencing this preference. Methods The TOVID (Telephony Or Videophony for Isolated elDerly) study was a cross-sectional study that was designed to examine the preference between telephone calls and video calls among frail older adults who were either hospitalized in a geriatric acute care unit or institutionalized in a long-term care and nursing home during the COVID-19 confinement period. Results A total of 132 older people were surveyed between March 25 and May 11, 2020 (mean age 88.2 years, SD 6.2); 79 (59.8%) were women. Patients hospitalized in the geriatric acute care unit were more able to establish communication independently than residents institutionalized in the long-term care and nursing home (P=.03) and were more satisfied with their communication experiences (P=.02). Overall, older people tended to favor telephone calls (73/132, 55.3%) over video calls (59/132, 44.7%); however, their satisfaction degree was similar regardless of the chosen method (P=.1), with no effect of age (P=.97) or gender (P=.2). In the geriatric acute care unit, the satisfaction degrees were similar for telephone calls (40/41, 98%) and video calls (33/38, 87%) in older patients (P=.10). Conversely, in the long-term care and nursing home, residents were more satisfied with the use of video calls to communicate with their relatives (14/15, 93%) versus the use of telephone calls (6/12, 50%; P=.02). Conclusions Older people confined to health care settings were able to complete telephone calls more independently than video calls, and they tended to use telephone calls more often than video calls. The satisfaction degrees were similar with both modalities and even greater with video calls among long-term care and nursing home residents when they were given assistance to establish communication. Dans le premier chapitre, nous tenterons de caractériser la situation actuelle en introduisant le secteur de la grande distribution en essayant de comprendre sa genèse et ses bases puis nous nous concentrerons sur l'étude approfondie des caractéristiques de la fonction Achats en grande distribution. Dans le deuxième chapitre, nous décrirons les mutations des attentes sociétales et des clients puis nous étudierons les nouveaux modèles économiques pour enfin se consacrer à la clarification du concept d'orientation clients, d'orientation marché et les liens avec la fonction Achats. Et enfin, dans le troisième et dernier chapitre, nous nous consacrerons à communiquer les données récoltées lors de notre enquête terrain et nous discuterons ces résultats.

The global context of energy savings and greenhouse gases emissions control led to significant efforts in France to boost the thermal insulation in buildings in order to reduce heating consumption. Nevertheless, the stated thermal performance before construction or refurbishment is rarely achieved in practice, for many reasons (calculation errors, defects in materials or workmanship, etc.). Yet, guaranteeing the real thermal performance of buildings on the spot is crucial to enhance the refurbishment market and the construction of energy efficient buildings. To do so, measurement techniques of the intrinsinc thermal insulation performance indicators are needed. Such techniques already exist worldwide, and consist in processing the measurement data from the indoor and outdoor thermal conditions and the heat consumption. Some of them have already proved themselves in the field, but are either binding or very imprecise. And above all, the related uncertainty calculations are often rough. The objective of this thesis funded by CSTB is to consolidate a novel measurement method of the thermal insulation quality of a whole building after reception of work (ISABELE method). In the first chapter, a state of the art of the existing methods allows to identify possible ways to pursue this goal from a comparative synthesis. The primary reflection is about the uncertainty calculation method (which is a central issue). The second chapter presents a global methodology to combine the propagation of random and systematic errors from bayesian and classical approaches. One of the most important uncertainty sources deals with the infiltration air flow evaluation during the test. The third chapter investigates the characterization of this uncertainty, as well as its impact on the final result, depending on the chosen experimental approach (rule of thumb, simplified aeraulic models, tracer gases). Lastly, an improvement of the inclusion of the bluiding thermal dynamics during the test will be proposed in the last chapter. The basis of this improvement is to adapt the inverse model according to the building type and the test conditions. To do so, the proposed algorithm selects a model form a variety of simplified greybox models based on statistical criteria and parcimony. All these contributions have been tested on a large serie of measurements on a same timber-framed building (OPTIMOB shed). The robustness and precision of the results have been slightly improved. The intial infiltration air flow calculation, neither too simple of too complicated, has also been validated. Finaly, a better ordrer of magnitude of the minimal test duration has been determined, depending on the building inertia., Dans un contexte d’économie d’énergie et de réduction des émissions de gaz à effet de serre, de nombreux efforts ont été réalisés en France pour renforcer l’isolation de l’enveloppe des bâtiments afin de contribuer à réduire les consommations de chauffage. Toutefois, il arrive souvent que la performance thermique calculée avant construction ou rénovation ne soit pas atteinte sur le terrain (erreur de calcul, défauts de mise en œuvre, etc.). Or, pour pouvoir généraliser la construction de bâtiments à basse consommation et la rénovation, il faut pouvoir garantir aux maîtres d'ouvrage une performance réelle de leur bâtiment après travaux. Le fait de mesurer in situ la performance intrinsèque d'isolation thermique de l'enveloppe permet de contribuer à cette garantie. Il existe à l’échelle internationale de nombreuses méthodes basées sur le suivi des consommations et des conditions thermiques intérieures et extérieures. Certaines ont déjà fait leurs preuves sur le terrain, mais sont souvent soit contraignantes, soit peu précises. Et surtout, les calculs d’incertitude associés sont souvent rudimentaires. L’objectif de ce travail financé par le CSTB est de consolider scientifiquement une nouvelle méthode de mesure de la qualité d’isolation globale d’un bâtiment à réception des travaux (méthode ISABELE). Dans le premier chapitre, un état de l'art sur les méthodes existantes a été réalisé afin de dégager des pistes d'amélioration sur la base d'une synthèse comparative. La piste prioritaire identifiée porte sur le calcul d'incertitude (un point central du problème). La propagation des erreurs aléatoires par un approche bayésienne ainsi que des erreurs systématiques par une approche plus classique feront l'objet de la méthodologie globale proposée dans le second chapitre. L'une des importantes sources d'incertitude porte sur l'évaluation du débit d'infiltration. La caractérisation de cette incertitude et de l'impact sur le résultat de mesure fera l'objet du troisième chapitre, avec un comparatif de différentes approches expérimentales (règle du pouce, modèles aérauliques, gaz traceur). Enfin, une amélioration de la prise en compte de la dynamique thermique du bâtiment au cours du test sera proposée dans le dernier chapitre. Son fondement repose sur l'adaptation du modèle thermique inverse en fonction du bâtiment et des conditions du test. Pour cela, une sélection parmi une banque de modèles simplifiés est réalisée sur la base de critères statistiques et du principe de parcimonie. Ces différentes dispositions ont été testés sur une large série de mesures menées sur un même bâtiment à ossature bois (chalet OPTIMOB). La robustesse et la précision du résultat de mesure ont ainsi pu être légèrement améliorées. La méthode de calcul du débit d'infiltration, ni trop simple ni trop complexe, a pu également être validée. Enfin, le temps de mesure minimal nécessaire a pu être déterminé en fonction de la classe d'inertie du bâtiment.

Both in clinical and environmental research, protein biomarkers are of growing interest. Although immunoassays are the gold standard for their quantification, recent advances in mass spectrometry (MS) make this technique a viable alternative to ELISA. This work provides some methodological elements to eliminate some limitations of the MS approach. First, two assays have been proposed. The first one achieved for G. fossarum represents the first example of quantification of vitellogenin in an invertebrate by LC-MS/MS. One of the challenges of the presented method is to specifically assay a protein in an organism whose genome is largely unknown. The second assay relates to methionine-containing peptides. A protocol was developped for total oxidation of methionines in order to overcome the bias due to their partial oxidation. This method was then applied to a protein involved in Alzheimer's disease (Apolipoprotein E4) in a cohort of 673 plasma samples. This assay is to date one of the largest study carried out by LC-MS/MS and shows all the robustness of this approach. Lastly, the influence of the mobile phase on the sensitivity of peptides assays was studied: first in reversed phase, where methanol is a good alternative to acetonitrile; then in HILIC, where the difficulties associated with the study of multicharged ions in a mainly organic content were discussed. Problems related to the carrying capacity of the columns were also raised.; En recherche clinique ou environnementale, les biomarqueurs protéiques présentent un intérêt croissant. Vitellogenin (Vg) is the precursor protein of vitellin that is the energy available for embryonic development in oviparous organisms. Vg is proposed as a relevant biomarker of endocrine disruptors in aquatic vertebrate species (e.g. fish, amphibian). Numerous strategies, such as enzyme-linked immunosorbent assays have been developed to characterize and quantify this protein in vertebrates. On the contrary, in invertebrates few methods are available in spite of their importance. This gap mainly comes from the low transferability of available antibodies across species. Recently, our laboratory developed a quantitative assay of Vg in a widespread amphipod, Gammarus fossarum, using liquid chromatography tandem mass spectrometry (LC-MS/MS) in selected reaction monitoring mode (SRM). In this context, the aim of this study was to assess the possibility to take advantage of peptidic motifs conservation to propose a transferable method across species. For this, our study focused on three model species for which sequence of egg yolk protein is well known: an amphipod (Gammarus fossarum), a cladocerean (Daphnia magna) and an insect (Drosophila melanogaster). In a first step, proteotypic peptides of Vg were identified for each species. Then we tried to find these proteotypic peptides in closely related species for which Vg sequence is unknown (such as G. pulex, G. wautierii, D. pulex). This study showed the high relevance of mass spectrometry to propose a specific methodology for Vg absolute quantification in organisms for which proteins sequences are unknown. Moreover, the proposed methodology is transferable from a species to another one.

communication par affiche; National audience; Cerebrospinal fluid and plasma apoE levels have been proposed as diagnostic biomarkers to discriminate patients with Alzheimer disease (AD) against normal ederly. However, controversy remains about the clinical significance of plasma apoE levels due to inconsistency across the correlations between those patients, which might arise from different ELISA protocols and/or size and demographic cohorts. Coupling between liquid chromatography and targeted mass spectrometry, in the so-called selected reaction-monitoring mode (LC-SRM), has been shown to be an effective and orthogonal alternative to immuno-enzymatic tests for absolute protein quantification in biofluids. In the present study, we have developed SRM-based assays of total ApoE and ApoE4 isoform in plasma to address the lack of consensus regarding the possible association between ApoE and ApoE4 genotype and AD. AQUA standardization was carried out with the heavy forms of proteotypic sequence LGPLVEQGR tracking total ApoE and of allele specific sequence LGADMEDVR monitoring ApoE4 isoform, respectively. Robustness and precision of the workflow have been extensively evaluated over a one-month period, with 12 technical replicates per weak carried out by two experimentalists. Statistics on the replicates show CV's better than 10% and a lower limit of quantification of 1 microgram/mL for the less sensitive peptide sequence LGADMEDVCGR specific of ApoE4 allele. The validated SRM-based assay was then applied to the absolute quantification of total ApoE and ApoE4 isoform across a clinical cohort of 800 patients.
