Abstract

Habitability has been generally defined as the capability of an environment to support life. Ecologists have been using Habitat Suitability Models (HSMs) for more than four decades to study the habitability of Earth from local to global scales. Astrobiologists have been proposing different habitability models for some time, with little integration and consistency among them, being different in function to those used by ecologists. Habitability models are not only used to determine if environments are habitable or not, but they also are used to characterize what key factors are responsible for the gradual transition from low to high habitability states. Here we review and compare some of the different models used by ecologists and astrobiologists and suggest how they could be integrated into new habitability standards. Such standards will help to improve the comparison and characterization of potentially habitable environments, prioritize target selections, and study correlations between habitability and biosignatures. Abstract

Habitability has been generally defined as the capability of an environment to support life. Ecologists have been using Habitat Suitability Models (HSMs) for more than four decades to study the habitability of Earth from local to global scales. Astrobiologists have been proposing different habitability models for some time, with little integration and consistency between them and different in function to those used by ecologists. In this white paper, we suggest a mass-energy habitability model as an example of how to adapt and expand the models used by ecologists to the astrobiology field. We propose to implement these models into a NASA Habitability Standard (NHS) to standardize the habitability objectives of planetary missions. These standards will help to compare and characterize potentially habitable environments, prioritize target selections, and study correlations between habitability and biosignatures. 11. Targeting SNARE proteins and the trafficking of sodium channels in inflammatory pain

Abstract

Dorsal root ganglion (DRG) neurones perceive and discriminate diverse types of sensations. Nociceptors are a subgroup of DRG neurones specialised in translating noxious pain stimuli to the spinal cord and higher brain centres. Following a noxious insult, nociceptors are known to have enhanced excitability and peptide secretion both of which, are likely to be a consequence of increased membrane trafficking and vesicle fusion with the plasma membrane. Vesicle-associated membrane proteins (VAMPs) are vesicular SNARE proteins (v-SNAREs) which complex together with cognate target SNARE proteins (t-SNAREs) found on 'acceptor' compartments. Together they regulate membrane trafficking and vesicle fusion; while much is known about the v- and t-SNAREs involved in the fusion of neurotransmitter and neuropeptide-containing vesicles with the plasma membrane, to date the identity of SNAREs involved in ion channel trafficking or secretion in nociceptors is limited. To explore the role of SNAREs on DRG neurones' secretion and excitability, an in vitro inflammation model was established. An inflammatory soup containing ATP, bradykinin, prostaglandin E2, histamine, noradrenaline, nerve growth factor, and serotonin was added for 22 h. This incubation induced hyperexcitability. In voltage-clamp, sodium currents resistant to tetrodotoxin, Nav1.9 and Nav1.8 currents, were increased. However, a decrease in the expression of Nav1.7 (TTX-sensitive) and Nav1.9 (TTX-resistant) at the plasma membrane was observed, which likely reflects changes in subcellular location of these channels induced by the inflammatory model. This study also identified the expression of seven vesicle membrane-associated proteins in DRG neurones (VAMP1-5, 7 and 8). This expression was found across all soma diameters. VAMP1/2/7 were observed in the neurites. The potential of the botulinum chimaeras, tetbot A and tetbot B, in reducing CGRP (calcitonin gene-related peptide) release and preventing excitability induced by an inflammatory soup was also explored. These chimaeras are designed to target isolectin B4-negative DRG neurones and cleave SNAP25 (t-SNARE) and VAMP1/2/3 (v-SNAREs). Tetbot A cleaved SNAP25 and significantly reduced CGRP release elicited by 60 mM KCl. The interpretation of the electrophysiology results is problematic as the detergent used in the chimaera preparations altered baseline properties of the DRG neurones.

39. Penicillium spp. strains as a possible weapon to fight microbial infections

Abstract

Bacteria are becoming increasingly resistant to antibiotics, leading to untreatable infections and constituting a major public health hazard (Lewis, 2013). This problem is further increased by the reduction in the number of effective antibiotics to tackle resistant strains (Penesyan et al. 2015), so the search for new compounds is seen as a vital priority. Our study consisted in the analysis of four different environmental strains of Penicillium spp., with screening for their extracellular metabolites for potential antimicrobial activity. Several methods were combined and tested for metabolite production and extraction. The strains were grown on: solid media (I-potato dextrose agar and II-malt extract agar), broth (malt extract broth with III-0 and IV-5% NaCl), and V-tapwater; extractions were performed using three different solvents: A-methanol, B-butanol and C-ethyl acetate. All extracts were tested on three different model organisms: Micrococcus luteus, Escherichia coli and Mycobacterium smegmatis. These are models for Gram-positive and Gram-negative bacteria, and for Tuberculosis. The extracts were compared and analysed in order to determine minimal inhibitory concentrations for each of the microorganisms tested. This research presents preliminary results on the development of potential new chemical compounds to help us circumvent the problem of drug resistance. Lewis, K., 2013. 41. Application of microbial fuel cell technology for vinasse treatment and bioelectricity generation

Abstract

Objective Our study evaluated the performance of different two-chambered microbial fuel cell (MFC) prototypes, operated with variable distance between electrodes and Nafion membrane and specific inoculum concentration, applied for vinasse treatment. Results The performance of the developed MFC resulted in a maximum current density of 1200 mA m−2 and power density of 800 mW m−2 in a period of 61 days. MFC performed a chemical oxygen demand removal at a rate ranging from 51 to 60%. Conclusions Taking our preliminary results into consideration, we concluded that the MFC technology presents itself as highly promising for the treatment of vinasse.

Abstract

O Brasil é um dos maiores produtores de etanol no mundo, no entanto, durante o processo de obtenção deste combustível, elevadas quantidades de vinhaça são geradas, em média, para cada litro de etanol produzido são gerados 13 litros deste resíduo. A vinhaça possui propriedades físicas e químicas que podem desencadear efeitos nocivos à biota, deste modo, existe um interesse crescente em identificar novas tecnologias para o seu tratamento. A utilização de células a combustível microbianas (CCMs) apresenta um enorme potencial, uma vez que, pode promover simultaneamente o tratamento deste resíduo e a biogeração de energia elétrica, pela conversão da energia química disponível nesses substratos biodegradáveis diretamente em eletricidade, por meio da ação catalítica de bactérias eletrogênicas aderidas ao eletrodo. Neste contexto, o presente estudo avaliou a utilização de uma CCM de câmara dupla no tratamento de vinhaça sintética. Durante a operação deste reator foi detectada uma máxima densidade de potência de 330 mW·m-2 e uma redução na demanda química de oxigênio (DQO) em 58,8% após 28 dias. Diante destes resultados preliminares, a tecnologia CCM pode ser considerada promissora no tratamento da vinhaça, contudo, estudos a respeito da configuração, tipo de inóculo e formas de operação da CCM são necessários para que amplie sua eficiência.

Abstract

This paper analyses the relationship between productive specialisation and economic growth in the 15 older European Union member states between 1970 and 2005. The sectoral taxonomy proposed by (Peneder, 2007) is used to classify the different sectors of activity according to the educational levels of the respective workforce and establish a comparison between the manufacturing and the services sector, based on their potential contribution to productivity improvements. The empirical model corresponds to a growth regression where the employment share of the different sectors is the main explanatory variable taken alongside other control variables identified in the empirical growth literature as robust growth determinants and is estimated with the fixed effects method. The results indicate that a higher weight of manufacturing activities that use mostly very low and low educated workers presents a negative association with growth. Services activities that require low educated workers make a negative growth contribution. Manufacturing activities with high and medium-high educational requirements have a positive growth influence, while in the case of services only activities that require highly-educated workers show a positive correlation with growth. The policy advice that can be extrapolated from this study contemplates the design of industrial policies that promote manufacturing activities such as chemicals, telecommunications and transports equipment, and services such as financial intermediation, audit, tax consulting, engineering and legal activities, to promote growth. Abstract

Light is the principal synchronizing environmental factor for the biological clock. Light quantity (intensity), and light quality (type of light source) can have different effects. The aim of this study was to determine the effects of the type of light experienced from the time of birth on mouse growth, circadian behaviour and stress levels. We raised pigmented and albino mice under 24 h light–dark cycles of either fluorescent or white light-emitting diode (LED) light source during the suckling stage, and the animals were then exposed to various light environments after weaning and their growth rate, locomotor activity and plasma corticosterone concentration were measured. We found that the type of light the animals were exposed to did not affect the animals’ growth rates or stress levels. However, we observed significant effects on the expression of the locomotor activity rhythm under low contrast light–dark cycles in pigmented mice, and under constant light in both albino and pigmented mice. These results highlight the importance of environmental light quality (light source) on circadian behavioural rhythms, and the need for close monitoring of light environments in animal facilities.
